BACKGROUND: The aim of this study was to evaluate cytotoxic, mutagenic and genotoxic effects on buccal mucosa and peripheral blood cells from marijuana and tobacco smokers. METHODS: For this purpose, a total of 45 volunteers were distributed into four groups: CTRL group (control): individuals who did not smoke marijuana or tobacco (n = 11); Group M: Marijuana smokers (n = 13); Group T: Tobacco smokers (n = 13); Group M + T: Smokers of both marijuana and tobacco (n = 08). RESULTS: Smokers of both marijuana and tobacco led an increase of micronucleated cells on buccal mucosa when compared to control group. The occurrence of karyolysis showed significant changes in this group as well. The comet assay data revealed genetic damage in peripheral blood cells for all groups of smokers. CONCLUSION: In summary, our results showed that marijuana and /or tobacco are able to induce genetic damage and cytotoxicity in oral and peripheral blood cells.

BACKGROUND/AIM: Since street sweepers comprises a group of workers who are in daily contact with rubbish, dust and air pollution, the aim of this study was to evaluate potential cytotoxic and mutagenic effects in buccal mucosa cells of street sweepers. MATERIALS AND METHODS: A total of 20 male street sweepers aged from 22 to 56 years were included in the experimental group. A total of 20 men matched by age were used as the control group. Cytotoxicity and mutagenicity were analyzed by micronucleus test in buccal mucosal cells. RESULTS: Statistically significant differences (p<0.05) in the frequency of micronuclei was detected in the street sweepers when compared to the control group. No remarkable differences were found to other metanuclear alterations indicative for cytotoxicity such as pyknosis, karyolysis, and karryorhexis when compared to matched controls. CONCLUSION: Taken together, our results indicate that street sweepers comprise an at-risk group as a result of increased mutagenicity found to buccal mucosa cells.

Neurodegenerative diseases are characterized by the presence of abnormal aggregates of proteins in brain tissue. Among them, the presence of aggregates of phosphorylated Tau protein (p-Tau) is the hallmark of Alzheimer's disease (AD) and other major neurodegenerative disorders such as corticobasal degeneration and frontotemporal dementia among others. Although Tau protein has previously been assumed to be exclusive to the central nervous system, it is also found in peripheral tissues. The purpose of this study was to determine whether there is a differential Tau expression in oral mucosa cells according to cognitive impairment. Eighty-one subjects were enrolled in the study and classified per Mini-Mental State Examination test score into control, mild cognitive impairment (MCI), and severe cognitive impairment (SCI) groups. Immunocytochemistry and immunofluorescence revealed the presence of Tau and four p-Tau forms in the cytoplasm and nucleus of oral mucosa cells. More positivity was present in subjects with cognitive impairment than in control subjects, both in the nucleus and cytoplasm, in a speckle pattern. The mRNA expression of Tau by quantitative real-time polymerase chain reaction was higher in SCI as compared with the control group (P < 0.01). A significantly higher percentage of immunopositive cells in the SCI group was found via flow cytometry in comparison to controls and the MCI group (P < 0.01). These findings demonstrate the higher presence of p-Tau and Tau transcript in the oral mucosa of cognitively impaired subjects when compared with healthy subjects. The feasibility of p-Tau quantification by flow cytometry supports the prospective analysis of oral mucosa as a support tool for screening of proteinopathies in cognitively impaired patients.

Waste collectors collect, transport, and process the garbage produced by people living in the city. Nowadays, this activity requires special attention due to the environmental impact of garbage and its potential consequences on human health. The aim of this study was to evaluate potential cytotoxic and mutagenic effects of garbage collection on waste collectors. For this purpose, a total of 47 male waste collectors aged from 24 to 53 years were included in the experimental group. A total of 30 men matched by age were used as the control group. Cytotoxicity and mutagenicity were analyzed by micronucleus test in buccal mucosaI cells. No statistically significant difference (p>0.05) in the frequency of micronuclei was detected in the waste collectors when compared to controls. Nevertheless, higher frequencies of karyolysis and pyknosis (p<0.05) were detected in buccal mucosaI cells from waste collectors when compared to matched controls. Taken together, our results indicate that waste collectors comprise an at-risk group as a result of increased cytotoxicity apparent from buccal mucosa cells.

In addition to surgery, one of the most widely applied treatments for breast cancer is chemotherapy. Chemotherapy is currently considered efficient in curing this disease; however, the therapy may induce damage to the patient's genetic material. Thus, the aim of this study was to evaluate putative cytotoxic and mutagenic effects induced by chemotherapy in women diagnosed with breast cancer. For this purpose, a cross-sectional study was carried out in 42 women, aged 18 to 70 years, allocated according to the diagnosis and stage of breast cancer treatment: control group (healthy) (n=15), chemotherapy group (n=11) and post-chemotherapy group (n=16). Cytotoxicity and mutagenicity were analyzed by the micronucleus test in buccal mucosa cells. A higher frequency (p<0.05) of micronucleated cells was detected in the chemotherapy and post-chemotherapy groups when compared to the control. A higher frequency (p<0.05) of karyorrhexis and pyknosis in the chemotherapy group was also noted. Taken together, our results indicate that chemotherapy induces mutagenicity and cytotoxicity in buccal mucosa cells of women diagnosed with breast cancer, being persistent after finishing their treatment.

BACKGROUND: Thousands of chemical compounds are used in paint products, like pigments, extenders, binders, additives, and solvents (toluene, xylene, ketones, alcohols, esters, and glycol ethers). Paint manufacture workers are potentially exposed to the chemicals present in paint products although the patterns and levels of exposure to individual agents may differ from those of painters. The aim of the present study was to evaluate genome damage induced in peripheral blood lymphocytes and oral mucosa cells of paint industry workers. MATERIALS AND METHODS: Genotoxicity was evaluated using the alkaline Comet assay in blood lymphocytes and oral mucosa cells, and the Micronucleus test in oral mucosa cells. For the micronucleus test in exfoliated buccal cells, no significant difference was detected between the control and paint industry workers. RESULTS: The Comet assay in epithelia buccal cells showed that the damage index (DI) and damage frequency (DF) observed in the exposed group were significantly higher relative to the control group (P≤0.05). In the same way, the Comet assay data in peripheral blood leukocytes showed that both analysis parameters (DI and DF) were significantly greater than that for the control group (P≤0.05). CONCLUSIONS: Chronic occupational exposure to paints may lead to a slightly increased risk of genetic damage among paint industry workers.
