Monitoring soil moisture content is especially important as it provides relevant information for making informed decisions regarding irrigation, fertigation, and water stress management. This study aims to validate a model for estimating soil water content using diffuse reflectance spectroscopy in the near-infrared range. The evaluated soils come from the municipalities of Puerto Gaitán (Meta), Espinal (Tolima), and Mosquera (Cundinamarca). In the first two municipalities, rigid networks were established to select sampling points, with two depths considered for each case (0-10 and 10-30 cm; 0-10 and 10-25 cm, respectively). For the third municipality, 77 soil pits were described, and samples were taken at depths of 0-10 and 10-35 cm. Subsequently, moisture content was evaluated at 0, 15, and 30 % moisture levels. The obtained data were analyzed using descriptive statistics. Cross-validation and external validation were applied to each model, and a general model was developed based on the data from all three sites. The obtained models for each sampling site and the general model demonstrated good predictive capacity. Based on the results, it is affirmed that near-infrared diffuse reflectance spectroscopy is an excellent option for determining soil water content. Similarly, principal component analysis identified differentiation between water contents of the studied soils.

RESUMO

An operando DRIFT-MS system (Diffuse Reflectance Infrared Fourier Transform Spectroscopy coupled with Mass Spectrometry) was designed and set up to study the oxidative steam reforming of ethanol reaction (OSRE). This reaction involves the mixture of water, ethanol and oxygen to produce mainly hydrogen, which is a rather attractive energy carrier. Spectroscopic monitoring of the process is a key tool to contribute to the understanding of: i) the dynamics on the catalyst surface, ii) the reaction mechanism and iii) the effect of the solid's properties on the catalytic process. In this sense, this document sets forth the experimental design that allows to carry out the study under operando DRIFT-MS conditions through time for the OSRE reaction. Selection criteria for parameters, materials, configuration, and experimental conditions are included, particularly optimizing the parameters of particle size and the dilution factor with KBr as well as the temperature and flow conditions for carrying out the reaction.•Clear signals of the adsorbed species in IR that do not present interference by water in the reaction atmosphere.•Simple assembly and online product detection by MS that allow to follow the change in the products of the OSRE reaction according to the temperature.•Controlled entry of gases and quantification by loop injection.

RESUMO

The nanoencapsulation of thyme essential oil has been greatly important in food science, given its remarkable antioxidant and antimicrobial capacity. However, its analysis in storage has not been established in terms of physical stability, antioxidant capacity, and release studies. In this paper, chitosan-thyme oil nanocapsules were prepared by the ionic gelation method. These were characterized for differential calorimetry, release kinetic, and infrared spectroscopy. The chitosan-thyme oil nanocapsules were stored at 4 and 25 °C for 5 weeks, the changes in particle size, zeta potential, stability (diffuse reflectance), and antioxidant capacity were analyzed and associated with nanocapsules' functionality. The results show that the storage time and temperature significantly modify the particle size (keeping the nano-size throughout the storage), the release of the bioactive was Fickian with t0.193 according to Korsmery & Peppas and best described by Higuchi model associated with changes in the zeta potential from 8 mV to -11 mV at 4 °C. The differential scanning calorimetry and infrared spectroscopy results confirm the good integration of the components. The antioxidant capacity revealed a direct relationship with residual oil concentration with a decrease in the ABTS test of 15% at 4 °C and 37% at 25 °C. The residual bioactive content was 77% at 4 °C and 62% at 25 °C, confirming nanoencapsulation effectiveness. The present investigation provides helpful information so that these systems can be applied in food conservation.

RESUMO

Cutaneous leishmaniasis (CL) is a parasitic disease that produces chronic skin ulcers. Although it has a worldwide presence, it is a neglected disease that still requires novel tools for its management. In order to study the use of optical tools in CL, this article presents a preliminary study of the correlation between CL histopathological and optical parameters. Optical parameters correspond to absorption and scattering coefficients obtained from diffuse reflectance spectra of treated CL in golden hamsters. Independently, histopathological data were collected from the same hamsters. As a result, after Spearman correlation and the Kruskal-Wallis test, inverse correlation was found between absorption/scattering optical parameters and inflammatory histopathological values, such as the scattering parameter related to the diameter of fibroblasts with the histopathological parameters of fibrosis, polymorphonuclear neutrophils, lymphocytes, plasmocytes, hyperplasia, and Leishmania, and the absorption parameter oxygen saturation showed a relation with the granulation tissue histopathological parameter. These correlations agree with the expected behavior of tissue composition during the healing process in CL. The results correspond to a proof of concept that shows that optical diffuse reflectance-based tools and methods could be considered as an alternative to assist in CL diagnosis and treatment follow-up.

RESUMO

Pyrimidine-2-thione (HSpym) reacts with lead(II) thiocyanate and lead(II) bromide in N,N-dimethylformamide (DMF) to form poly[(µ-isothiocyanato-κ2N:S)(µ4-pyrimidine-2-thiolato-κ6N1,S:S:S:S,N3)lead(II)], [Pb(C4H3N2S)(NCS)]n or [Pb(Spym)(NCS)]n, (I), and the polymeric one-dimensional (1D) compound catena-poly[[µ4-bromido-di-µ-bromido-(µ-pyrimidine-2-thiolato-κ3N1,S:S)(µ-pyrimidine-2-thione-κ3N1,S:S)dilead(II)] N,N-dimethylformamide monosolvate], {[Pb2Br3(C4H3N2S)(C4H4N2S)]·C3H7NO}n or {[Pb2Br3(Spym)(HSpym)]·DMF}n, (IIa), respectively. Poly[µ4-bromido-di-µ3-bromido-(µ-pyrimidine-2-thiolato-κ3N1,S:S)(µ-pyrimidine-2-thione-κ3N1,S:S)dilead(II)], [Pb2Br3(C4H3N2S)(C4H4N2S)]n or [Pb2Br3(Spym)(HSpym)]n, (IIb), could be obtained as a mixture with (IIa) when using a lesser amount of solvent. In the crystal structures of the pseudohalide/halide PbII stable compounds, coordination of anionic and neutral HSpym has been observed. Both Spym- (in the thiolate tautomeric form) and NCS- ligands were responsible for the two-dimensional (2D) arrangement in (I). The Br- ligands establish the 1D polymeric arrangement in (IIa). Eight-coordinated metal centres have been observed in both compounds, when considering the Pb...S and Pb...Br interactions. Both compounds were characterized by FT-IR and diffuse reflectance spectroscopies, as well as by powder X-ray diffraction. Compound (IIa) and its desolvated version (IIb) represent the first structurally characterized PbII compounds containing neutral HSpym and anionic Spym- ligands. After a prolonged time in solution, (IIa) is converted to another compound due to complete deprotonation of HSpym. The structural characterization of (I) and (II) suggests HSpym as a good candidate for the removal of PbII ions from solutions containing thiocyanate or bromide ions.

RESUMO

The present work shows the synergistic application of reflectance spectroscopy and core-shell molecularly imprinted polymer (MIP) for rapid quantification of tartrazine in soft drinks. Studies evaluated the performance of the MIPs synthesized in the presence of silica nanoparticles unfunctionalized and functionalized with [3-(methacryloyloxy)propyl]trimethoxysilane. Although the use of functionalized silica nanoparticles promoted the highest adsorption capability of tartrazine, the material was found to be less selective when it was applied in real samples. Interestingly, the most accurate results were obtained via the application of the MIP synthesized in the presence of unfunctionalized silica nanoparticles (SiO2@MIP). The optimized core-shell MIP was also characterized by Raman spectroscopy and scanning electron microscopy. The use of direct reflectance spectroscopy in the analyte detection strategy after the template extraction from the MIPs resulted in faster and more accurate results than conventional UV-Visible spectroscopy. With regard to the analysis of the soft drink samples, no significant differences were found between the results obtained from the proposed reflectance spectroscopy-based technique and those obtained from the comparative high-performance liquid chromatography technique. Under optimized conditions, this method displayed a linear range of 1.0-12.5 µmol L-1 with LOD and LOQ values of 0.303 and 1.0 µmol L-1, respectively. The selectivity factor (ß) ranged between 1.4 up to 264 showed better recognition of tartrazine in front of other dyes. Based on the results obtained, the proposed method is found to be suitable for rapid determination of tartrazine in food samples with complex matrices without the need of applying tedious sample preparation and cost-demanding instruments.

RESUMO

BACKGROUND AND OBJECTIVE: Due to the existing prevalence of nonalcoholic fatty liver disease (NAFLD) and its relation to the epidemic of obesity in the general population, it is imperative to develop detection and evaluation methods of the early stages of the disease with improved efficacy over the current diagnostic approaches. We aimed to obtain an improved diagnosis, combining methods of optical spectroscopy -diffuse reflectance and fluorescence- with statistical data analysis applied to detect early stages of NAFLD. METHODS: Statistical analysis scheme based on quadratic discriminant analysis followed by canonical discriminant analysis were applied to the diffuse reflectance data combined with endogenous fluorescence spectral data excited at one of these wavelengths: 330, 365, 385, 405 or 415 nm. The statistical scheme was also applied to the combinations of fluorescence spectrum (405 nm) with each one of the other fluorescence spectra. Details of the developed software, including the application of machine learning algorithms to the combination of spectral data followed by classification statistical schemes, are discussed. RESULTS: Steatosis progression was differentiated with little classification error (≤1.3%) by using diffuse reflectance and endogenous fluorescence at different wavelengths. Similar results were obtained using fluorescence at 405 nm and one of the other fluorescence spectra (classification error ≤1.0%). Adding the corresponding areas under the curves to the above combinations of spectra diminished errors to 0.6% and 0.3% or less, respectively. The best results for the compounded reflectance-plus-fluorescence spectra were obtained with fluorescence spectra excited at 415 nm with a total classification error of 0.2%; for the combination of the 405nm-excited fluorescence spectrum with another fluorescence spectrum, the best results were achieved for 385 nm, for which total relative classification error amounted 0.4%. The consideration of the area under the spectral curves further improved both classifiers, reducing the error to 0.0% in both cases. CONCLUSION: Spectrometric techniques combined with statistical processing are a promising tool to improve steatosis classification through a label free approach. However, statistical schemes here applied, might result complex for the everyday medical practice, the designed software including machine learning algorithms is able to render automatic classification of samples according to their steatosis grade with low error.

RESUMO

Fractal and polarization analysis of diffusively scattered light is applied to determine the complex relationship between fractal dimension of structural morphology and concentration of chemically active ingredients in two pharmaceutical mixture systems including a series of binary mixtures of acetaminophen in lactose and three multicomponent blends with a proprietary active ingredient. A robust approach is proposed to identify and filter out multiple- and single-scattering components of scattering indicatrix. The fractal dimension extracted from scattering field reveals complex structural details of the sample, showing strong dependence on low-dose drug concentration in the blend. Low-angle diffraction shows optical "halo" patterns near the angle of specular reflection caused by light refraction in microcrystalline aggregates. Angular measurements of diffuse reflection demonstrate noticeable dependence of Brewster's angle on drug concentration. It is shown that the acetaminophen microcrystals produce scattered light depolarization due to their optical birefringence. The light scattering measurement protocol developed for diffusively scattered light by microcrystalline pharmaceutical compositions provides a novel approach for the pattern recognition, analysis and classification of materials with a low concentration of active chemical ingredients.

RESUMO

Vis-NIR-SWIR reflectance spectra of leaf samples, collected in the laboratory, allow the calibration of predictive models to quantify their physicochemical attributes in a practical manner and without producing chemical residues. This technique should enable the development of management strategies for intensification of pasture use. However, spectral analysis performed in the laboratory may be affected by the deterioration of plant material during transport from the field to the lab, so storage methods are necessary. This research aimed to evaluate the effects of different storage methods on the spectral response of Mombasa grass leaves. Three methods were evaluated: (i) artificially refrigerated environment, (ii) humid environment, and (iii) without microenvironment control. These methods were tested in five different storage times: 2 hours, 4 hours, 8 hours, 24 hours and 48 hours. The spectral behavior of the leaves still inserted in the plant was used as a quality reference. Results showed notable changes at the earliest storage time for the treatment without microenvironment control. Both methods with microenvironment control stabilized the occurrence of spectral changes over 48 hours of the samples storage, thus both were suggested for this species.(AU)

RESUMO

Thin films are a type of coating that have a very wide spectrum of applications. They may be used as single layers or composed in multilayer stacks, which significantly extend their applications. One of the most commonly used material for thin films is silicon dioxide, SiO2. Although there are other tools that can be used to measure the thickness of SiO2 films, these tools are very complex and sophisticated. In this article, we propose the use of an exponential two-layer light-material interaction model, throughout its diffuse reflectance spectra, as an alternative for the measurement of the thickness of evaporated SiO2 on Si wafers. The proposed model is evaluated experimentally by means of a 980-nm-thick SiO2 layer evaporated on a Si wafer. The results show that the proposed model has a strong correlation with the thickness measurements obtained using commercial equipment.

RESUMO

Bee pollen consumption has increased in the last years, mainly due to its nutritional value and therapeutic applications. The quantification of mineral constituents is of great importance in order to evaluate both, the toxicity and the beneficial effect of essential elements. The purpose of this work was to quantify the essential elements, Ca, Mg, Zn, P and K, by diffuse reflectance spectra in the near infrared region (NIR) combined with partial least squares regression (PLS), which is a clean and fast method. Reference method used was ICP OES. The determination coefficients for calibration models (R2) were above 0.87 and the mean percent calibration error varied from 5 to 10%. For external validation R2 values were higher than 0.76. The results indicated that NIR spectroscopy can be useful for an approximate quantification of these minerals in bee pollen samples and can be used as a faster alternative to the standard methodologies.

RESUMO

Nonalcoholic fatty liver disease (NAFLD) ranges from steatosis to nonalcoholic steatohepatitis and cirrhosis. Liver biopsy, considered the gold standard to diagnose NAFLD, shows significantly high rates of interobserver variability. Thus there is a need to develop tools that accurately categorize mild and advanced grades of steatosis in order to identify patients at higher risk of developing chronic liver disease. Diffuse reflectance spectroscopy (DRS) has proved to be useful in grading liver fibrosis and cirrhosis, without having been implemented for steatosis. We aim to categorize early and advanced stages of liver steatosis in a methionine-choline deficient (MCD) mouse model. C57bl/6 mice are fed either methionine-choline control or MCD diet during 2 or 8 weeks to induce mild and advanced steatosis. Liver samples are obtained and steatosis is evaluated by oil red O staining. Diffuse reflectance spectra are directly measured on ex vivo liver specimens, in a wavelength range of 400 to 800 nm. DRS is able to discriminate between early or advanced steatosis and healthy hepatic tissue with negligible error while showing high average sensitivity and specificity (0.94 and 0.95, respectively). Our results suggest that liver steatosis can be accurately evaluated by DRS, highlighting the importance of applied spectroscopic methods in assessing NAFLD.

RESUMO

Global demands for an increased food production have prompted scientists to create technological innovations in various segments of the agricultural and environmental sectors. The use of diffuse reflectance spectroscopy in soil color determination to estimate its attributes is an example of these advances. Thus, the aim of this research was to identify the potential of the three parameters of the Munsell color system (Hue, Value, and Chroma) in estimating different chemical and particle size attributes by pedotransfer functions in a Typic Hapludox in the Cerrado of Piauí State, Brazil. This soil showed a yellowish color due to the goethite and low contents of bases and organic carbon. Among the calibrated pedotransfer functions, we observed that phosphorus, aluminum, hydrogen, potential acidity, potential cation exchange capacity, and base saturation could be estimated by Hue, while clay content by Value, and Chroma used to estimate hydrogen, potential acidity, and cation exchange capacity. We conclude that the parameter color could be used to estimate soil attributes.(AU)

A simple, fast, low-cost, portable, and eco-friendly method using a spot test on a paper platform, together with diffuse reflectance spectroscopy, was developed and validated for the quantification of aluminum hydrochloride, a potential neurotoxic agent, in antiperspirant samples. The determination of aluminum hydrochloride was performed at a wavelength of 615 nm, by measuring consumption of the purple colorimetric reagent Alizarin S, due to reaction with aluminum. The linear range was from 10.0 to 125.0 mg L-1 and could be described by the equation: AR = 0.4479 - 0.002543 CAl (R = 0.999). The limits of detection (LOD) and quantification (LOQ) were 3.06 and 10.2 mg L-1, respectively. The method was specific, accurate, and repeatable, with relative standard deviation (RSD) <5.0%. The recovery was between 92.2 and 103.4%. The method was successfully used for the determination of aluminum hydrochloride in commercial antiperspirant samples, revealing concentrations below the maximum permitted by current legislation.

RESUMO

Identification of hot spots of land degradation is strongly related with the selection of soil tracers for sediment pathways. This research proposes the complementary and integrated application of two analytical techniques to select the most suitable fingerprint tracers for identifying the main sources of sediments in an agricultural catchment located in Central Argentina with erosive loess soils. Diffuse reflectance Fourier transformed in the mid-infrared range (DRIFT-MIR) spectroscopy and energy-dispersive X-ray fluorescence (EDXRF) were used for a suitable fingerprint selection. For using DRIFT-MIR spectroscopy as fingerprinting technique, calibration through quantitative parameters is needed to link and correlate DRIFT-MIR spectra with soil tracers. EDXRF was used in this context for determining the concentrations of geochemical elements in soil samples. The selected tracers were confirmed using two artificial mixtures composed of known proportions of soil collected in different sites with distinctive soil uses. These fingerprint elements were used as parameters to build a predictive model with the whole set of DRIFT-MIR spectra. Fingerprint elements such as phosphorus, iron, calcium, barium, and titanium were identified for obtaining a suitable reconstruction of the source proportions in the artificial mixtures. Mid-infrared spectra produced successful prediction models (R2 = 0.91) for Fe content and moderate useful prediction (R2 = 0.72) for Ti content. For Ca, P, and Ba, the R2 were 0.44, 0.58, and 0.59 respectively.

This study describes changes observed in the near-infrared (NIR) diffuse reflectance (DR) spectra of pharmaceutical tablets after these tablets were subjected to different levels of strain (exposure to shear) during the mixing process. Powder shearing is important in the mixing of powders that are cohesive. Shear stress is created in a system by moving one surface over another causing displacements in the direction of the moving surface and is part of the mixing dynamics of particulates in many industries including the pharmaceutical industry. In continuous mixing, shear strain is developed within the process when powder particles are in constant movement and can affect the quality attributes of the final product such as dissolution. These changes in the NIR spectra could affect results obtained from NIR calibration models. The aim of the study was to understand changes in the NIR diffuse reflectance spectra that can be associated with different levels of strain developed during blend shearing of laboratory samples. Shear was applied using a Couette cell and tablets were produced using a tablet press emulator. Tablets with different shear levels were measured using NIR spectroscopy in the diffuse reflectance mode. The NIR spectra were baseline corrected to maintain the scattering effect associated with the physical properties of the tablet surface. Principal component analysis was used to establish the principal sources of variation within the samples. The angular dependence of elastic light scattering shows that the shear treatment reduces the size of particles and produces their uniform and highly isotropic distribution. Tablet compaction further reduces the diffuse component of scattering due to realignment of particles.

RESUMO

The current study presents an application of Diffuse Reflectance Infrared Fourier Transform Spectroscopy for detection and quantification of fraudulent addition of commonly employed adulterants (spent coffee grounds, coffee husks, roasted corn and roasted barley) to roasted and ground coffee. Roasted coffee samples were intentionally blended with the adulterants (pure and mixed), with total adulteration levels ranging from 1% to 66% w/w. Partial Least Squares Regression (PLS) was used to relate the processed spectra to the mass fraction of adulterants and the model obtained provided reliable predictions of adulterations at levels as low as 1% w/w. A robust methodology was implemented that included the detection of outliers. High correlation coefficients (0.99 for calibration; 0.98 for validation) coupled with low degrees of error (1.23% for calibration; 2.67% for validation) confirmed that DRIFTS can be a valuable analytical tool for detection and quantification of adulteration in ground, roasted coffee.

In order to differentiate between benign and malignant lesions in the human skin using diffuse reflection spectra, different classification algorithms were tested using the WEKA data mining software. In addition, due to the high dimensionality of the spectral signal, an attribute selection technique was applied to determine the variables that contribute with more information. The spectral signal classification was tested using support vector machines, neural networks and random forests, their performance was measured using the k-fold cross-validation percentages of the Kappa statistic, area under the ROC curve, specificity and sensitivity. Finally it is shown that the one layer neural network with 6 neurons and the parameters momentum and learning rate in 0.6 and 0.3 respectively, is best suited to the problem of pattern recognition, achieving correctly classify 89.89% of the cases.
