RESUMO

In alpacas (Vicugna pacos), the high cost of in vitro embryo production is also a consequence of the use of several substances in the culture medium. In addition, embryo production rates in this species are still considered low. Thus, in attempt to reduce the cost and to improve the in vitro embryo production rates, this study evaluates the effect of adding follicular fluid (FF) in the in vitro maturation medium on oocyte maturation and subsequent embryo production. After ovary collection at the local slaughterhouse, the oocytes were recovered, selected, and allocated in experimental groups: standard maturation medium (G1) and simplified medium added by 10% FF (G2). The FF was acquired from follicles between 7- and 12-mm diameter. The cumulus cell expansion and the embryo production rates were analyzed by chi-square with p < 0.05. No differences (p > 0.05) were observed in maturation rate between G1 (66.36%) and G2 (63.12%) groups. Likewise, no significant difference (p > 0.05) was verified between G1 and G2 for morula (40.85 vs 38.45%), blastocyst (7.01 vs 6.93%), and total number of embryos (47.87 vs 45.38%). In conclusion, it was possible to simplify the medium used for in vitro maturation of alpaca oocytes resulting in embryo production rates similar to the standard medium.

The epidermal growth factor receptor (EGFR) signaling pathway is one of the main pathways responsible for propagating the luteinizing hormone (LH) signal throughout the cumulus cells and the oocyte. Recently, we have proposed the C-C motif chemokine receptor 2 (CCR2) and its main ligand (monocyte chemoattractant protein-1, MCP1) as novel mediators of the ovulatory cascade. Our previous results demonstrate that the gonadotropins (GNT), amphiregulin (AREG), and prostaglandin E2 (PGE2) stimulation of periovulatory gene mRNA levels occurs, at least in part, through the CCR2/MCP1 pathway, proposing the CCR2 receptor as a novel mediator of the ovulatory cascade in a feline model. For that purpose, feline cumulus-oocyte complexes (COCs) were cultured in the presence or absence of an EGFR inhibitor, recombinant chemokine MCP1, and gonadotropins [as an inducer of cumulus-oocyte expansion (C-OE), and oocyte maturation] to further assess the mRNA expression of periovulatory key genes, C-OE, oocyte nuclear maturation, and steroid hormone production. We observed that MCP1 was able to revert the inhibition of AREG mRNA expression by an EGFR inhibitor within the feline COC. In accordance, the confocal analysis showed that the GNT-stimulated hyaluronic acid (HA) synthesis, blocked by the EGFR inhibitor, was recovered by the addition of recombinant MCP1 in the C-OE culture media. Also, MCP1 was able to revert the inhibition of progesterone (P4) production by EGFR inhibitor in the C-OE culture media. Regarding oocyte nuclear maturation, recombinant MCP1 could also revert the inhibition triggered by the EGFR inhibitor, leading to a recovery in the percentage of metaphase II (MII)-stage oocytes. In conclusion, our results confirm the chemokine receptor CCR2 as a novel intermediate in the ovulatory cascade and demonstrate that the EGFR/AREG and the CCR2/MCP1 signaling pathways play critical roles in regulating feline C-OE and oocyte nuclear maturation, with CCR2/MCP1 signaling pathway being downstream EGFR/AREG pathway within the ovulatory cascade.

Aquaporins (AQPs) are proteins with various functions related to proper cell function and early development in mammals. The aim of this study was to evaluate the presence of AQPs and determine their mRNA levels in the cumulus oocyte complex (COC) of four bovine breeds and in blastocysts of five bovine crosses. Grade I, II and III COCs were collected by ovum pick up from non-lactating heifers of the Brahaman, Holstein, Gir and Romosinuano breeds. Embryos were produced in vitro up to the blastocyst stage of the bovine ♀Gir × â™‚Holstein, ♀Holstein × â™‚Gir, ♀Brahman × â™‚Holstein, ♀Holstein × â™‚Brahman, and ♀Romosinuano × â™‚Holstein crosses. mRNA expression of AQP1-AQP12b was estimated in COC and embryos by real-time-PCR. The presence of the twelve AQPs in the COCs and bovine embryos was established. Additionally, significant differences were determined in the expression of AQP6 and AQP12b in COCs, as well as in transcripts levels of AQP4, AQP8 and AQP9 from bovine embryos. Gene expression of AQPs in COCs and bovine embryos is consistent with the previously described biological functions. This is the first report of AQPs in COC of Gir, Brahman, Holstein and Romosinuano and embryos of five crossbreeds between Bos indicus and B. taurus.

An acute heat stress event after the LH surge increased interleukin 6 (IL6) levels in the follicular fluid of the ovulatory follicle in hyperthermic cows. To examine direct consequences of a physiologically-relevant elevated temperature (41.0°C) on the cumulus-oocyte complex (COC), IL6 transcript abundance and related receptor components were evaluated throughout in vitro maturation. Heat-induced increases in IL6 were first noted at 4 hours of in vitro maturation (hIVM); peak levels occurred at 4.67 versus 6.44 hIVM for 41.0 and 38.5°C COCs, respectively (SEM = 0.23; P < 0.001). Peak IL6ST levels occurred at 6.95 versus 8.29 hIVM for 41.0 and 38.5°C, respectively (SEM = 0.23; P < 0.01). Transcript for LIF differed over time (P < 0.0001) but was not affected by 41.0°C exposure. Blastocyst development after performing IVF was not affected by 41.0°C exposure for 4 or 6 h. When limiting analysis to when IL6 was temporally produced, progesterone levels were only impacted by time and temperature (no interaction). Heat-induced shift in the temporal production of IL6 and IL6ST along with its impact on progesterone likely cooperate in heat-induced hastening of meiotic progression described by others.

The aim of this study was to evaluate the genotoxic and cytotoxic effects of amitraz (AMZ) on the primary culture of bovine cumulus cells (CC) and oocyte nuclear maturation. Cytotoxicity was evaluated by assessing mitochondrial activity with the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. Genotoxicity was estimated using the alkaline single cell gel electrophoresis (SCGE) assay. Apoptosis was detected with the Annexin V-affinity assay. The in vitro maturation test was performed in bovine oocytes. To understand AMZ action, glutathione content, superoxide dismutase enzyme activity, and lipid peroxidation were evaluated in CC. Results showed that AMZ lethal concentration (LC 5024h) for bovine CC was 32.55 µg/mL (MTT assay). A 25 µg/mL induced late apoptosis and necrotic cells (p < 0.05); however, DNA damage was decreased at the same concentration (SCGE assay; p < 0.05). A decrease in metaphase II was observed at 25 µg/mL, and degenerate oocytes were observed at 15 and 25 µg/mL (p < 0.05). None of the oxidative stress parameters evaluated showed significant differences. This study contributes to a better understanding of AMZ in this model, suggesting its potential cytotoxicity and impact on bovine reproduction.

In this report, we present an unusual case of a couple who achieved a successful pregnancy by ICSI with a single oocyte collected. The cumulus-oocyte-complex (COC) was retrieved at 36.5 hours post trigger, and was found to be at metaphase II, when sperm injection was performed at around 39 hours post trigger. At 18 hours post injection, the single oocyte was fertilized, developed to four-cell embryo on day 2, and 8-cell grade on day 3, when it was relocated in the uterine cavity. The pregnancy yielded a positive ß-hCG result. The scan performed at seven weeks, revealed the presence of one amniotic sac with a fetal heartbeat. The ongoing pregnancy has been eventless at 24 weeks of gestation.

Expression of immune function genes within follicle cells has been reported in ovaries from many species. Recent work from our laboratory showed a direct effect of the monocyte chemoattractant protein 1/C-C motif chemokine receptor 2 system within the feline cumulus oocyte complex, by increasing the mRNA levels of key genes involved in the ovulatory cascade in vitro. Studies were designed to evaluate if C-C motif chemokine receptor 2 acts as a novel mediator of the ovulatory cascade in vitro. Therefore, feline cumulus oocyte complexes were cultured in the presence or absence of a highly selective C-C motif chemokine receptor 2 antagonist together with known inducers of cumulus-oocyte expansion and/or oocyte maturation to assess mRNA expression of key genes related to periovulatory events in other species as well as oocyte maturation. Also, the effects of recombinant monocyte chemoattractant protein 1 on spontaneous or gonadotrophin-induced oocyte maturation were assessed. This is an in vitro system using isolated cumulus oocyte complexes from feline ovaries. The present study reveals the modulation of several key ovulatory genes by a highly selective C-C motif chemokine receptor 2 antagonist. However, this antagonist was not enough to block the oocyte maturation induced by gonadotropins or amphiregulin. Nonetheless, recombinant monocyte chemoattractant protein 1 had a significant effect on spontaneous oocyte maturation, increasing the percentage of metaphase II stage oocytes in comparison to the control. This is the first study in any species to establish C-C motif chemokine receptor 2 as a mediator of some actions of the mid-cycle gonadotrophin surge.

Zinc (Zn) is a vital trace element for the body and its bioavailability influences numerous reproductive events. However, the mechanisms that regulate Zn homeostasis in the cumulus-oocyte complex (COC) are yet to be elucidated. The aim of this study was to investigate the role of estradiol 17-beta (E2), FSH and LH in Zn homeostasis regulation in bovine COC matured in vitro and Zn transporters gene expression. For this purpose, intracellular Zn levels in oocytes and cumulus cells (CC) were assessed using a Zn-specific fluorescent indicator. In addition, gene expression and sequencing of six Zn transporters (Slc39a6, Slc39a8, Slc39a14, Slc30a3, Slc30a7 and Slc30a9) were assessed. Our results demonstrated that the simultaneous presence of E2, FSH, and LH during oocyte maturation altered intracellular zinc levels and transporters expression in both oocytes and CC. Transporter's gene expression was different in oocytes and CC, possibly due to cell-specific changes in Zn levels during maturation. The interaction effects of Zn with hormonal treatments influenced the results. This study emphasizes that Slc39a6 is highly sensitive to hormone induction. Overall, the hormonal modulation of Zn homeostasis in the COC was evidenced. Also, a preponderant role of FSH as a modulator of Zn intracellular levels and transporter gene expression is suggested.

Studies were designed to (a) evaluate the mRNA expression of the C-C motif chemokine receptor 2 (CCR2) and its chemokine ligands, as well as genes related to periovulatory events, within the cumulus oocyte complex (COC) and follicle wall after a luteinizing hormone (LH) stimulus in cultured feline antral follicles; (b) assess the immunolocalization of CCR2 and its main ligand (monocyte chemoattractant protein 1, MCP1) within the feline COC; and (c) examine the direct effects of exogenous recombinant MCP1 on mRNA expression of the CCR2 receptor and MCP1 as well as key periovulatory genes in the COC, using a feline COC culture system. Both culture systems were developed by our laboratory and exhibit physiological response to gonadotropin stimuli. In summary, this study demonstrated mRNA expression of CCR2 receptor and its assessed ligands (MCP1, MCP2, MCP3, and MCP4) within the feline COC and follicle antral wall, and a significant increase in CCR2 mRNA by LH within the COC. Also, CCR2 and MCP1 immunoreactivity was observed in the oocyte and cumulus cells of the feline COC. Remarkably, this is the first report, in any species, describing a direct effect of the recombinant MCP1 in the CCR2/MCP1 system within the COC, by increasing the mRNA levels of key genes involved in the ovulatory cascade, as well as its own receptor CCR2. Together, these data suggest that CCR2 receptor signaling in the COC may regulate events critical for promoting cumulus oocyte expansion and/or oocyte maturation.

PURPOSE: We hypothesized that the chemokine SDF1/CXCR4 system was present in feline cumulus-oocyte complexes (COCs) and that COCs cultured with SDF1 would directly upregulate gene expression in the ovulatory cascade. METHODS: Ovaries (n = 50) were obtained from adult domestic cats during the breeding season and COCs were recovered from antral follicles. Because IVM media triggers cumulus-oocyte expansion, culture conditions needed to be optimized to study periovulatory genes. After optimization, the effects of 25 ng/ml SDF1 and the CXCR4 inhibitor were examined in a COC culture for 3, 12, and 24 h. RESULTS: MEM-hepes with 1% of charcoal stripped-FBS was the optimized culture medium, assessed by the expansion of COCs at 24 h in the gonadotropin (GNT) group but not in the media with serum alone. The mRNA expression of HAS2, TNFAIP6, PTX3, and AREG peaked at 3 h in GNT group as compared to all other groups (p < 0.05). COCs cultured with SDF1 showed increased HAS2 and TNFAIP6 mRNA expression at 3 h compared to negative controls and to the CXCR4 inhibitor group. CXCR4 and SDF1 immunostaining was present in both cumulus cells and the oocyte. CONCLUSIONS: These results demonstrate that GNT stimulation upregulates key periovulatory genes and expansion in feline COCs from antral follicles, and support the use of this culture system to examine molecular processes within the COC. In addition, SDF1 directly promotes key periovulatory genes in feline COCs, suggesting that the SDF1-CXCR4 pathway may extend its function beyond a chemoattractant, and may play a direct role within the COC.

In this study, goats were subjected to ovarian stimulation protocols to evaluate possible differences in ovarian follicular responses and oocyte production. Two experiments were conducted to assess the effects of hormonal protocol duration (seven or twelve days) and number of follicle stimulating hormone (FSH) applications (one or five doses). All animals received intravaginal sponges saturated with 60 mg medroxyprogesterone acetate and an application of 125 µg cloprostenol 72 h before the sponges were removed. For ovarian stimulation, 120 mg FSH was applied in a single dose 36 h before laparoscopic follicular aspiration (LOPU) or in five doses (30, 30, 20, 20, and 20 mg) at 12 h intervals, with the last dose applied 36 h before LOPU. In the first experimental phase, ultrasonography was performed to monitor follicular number and diameter, and in the second phase, the animals received LOPU to count the follicles and cumulus-oocyte complexes (COCs) and for morphological classification. There was no significant effect (P > 0.05) of any variable or combination of variables on follicle number on Day (D) 0 or D3/D8 (day of LOPU). However, evaluation at D6/D11 revealed an effect (P < 0.05) from the protocol duration with the highest number of small follicles resulting from the short protocol. There was also an effect (P < 0.05) of FSH dose number on the resulting number of medium and large follicles, with more medium follicles recovered after a single dose and more large follicles after multiple doses. There was no effect of any variable (P > 0.05) on the diameters of the large, medium, and small follicles, except for the D4/D9 evaluation, which showed a combination effect for large follicles (P < 0.05). In the second phase, no variable had an effect on the number of follicles visualized or aspirated, number of COCs recovered, recovery rate, morphological quality of COCs in grades 1, 2, 3, and 4, or rate of viable COCs. Thus, all four protocols efficiently stimulated the ovarian response and oocyte production of goats, although the short protocol (7 days) with a single dose of FSH was most efficacious based upon the greater number of medium-sized follicles available for aspiration, the shorter time, and greater practicality of execution.

Abstract The morphological selection of cumulus-oocyte complexes (COCs) is an important step for in vitro embryo production. It has been suggested that COC showing signs of atresia have the ability to generate embryos. The objective of this work was to evaluate the effect of COC morphology from Bos indicus animals with signs of early atresia versus no signs of atresia on in vitro embryo production. COC were classified in: Type I (TI): homogeneous ooplasm with ≥ 4 layers of compact cumulus cells (CC) and Type II (TII): granular ooplasm and ≥ 4 layers of CC slightly expanded. The COC were matured in vitro for 24 hours in TCM199 medium and subsequently fertilized in vitro for 18 h. The suspected zygotes were cultured in vitro for seven days in modified SOFaa medium. Embryonic quality was determined by blastomeric count following staining with Hoechst 33342. Student test was used to determine statistical differences for cleavage, blastocyst rate and blastomeric counts between types of COC. The cleavage rate for TI (n = 220) and TII (n = 161) was 88 ± 4% and 89 ± 8% respectively (p> 0.05); embryo development rate was 36 ± 7% and 33 ± 8% (p>0.05) respectively. The blastomeric count for both groups was 101 and 104 cells for TI and TII respectively (n = 10), (p>0.05). These results demonstrate that there is no difference in the quantity and quality of embryos produced in vitro using COC type I or type II, suggesting that both types could be used for bovine in vitro embryo production in Bos indicus cows.

Se ha sugerido que COC que muestran signos de atresia poseen capacidad de generar embriones. El objetivo de este trabajo fue evaluar el efecto de la morfología de los COC provenientes de animales Bos indicus con signos de atresia temprana y sin signos de atresia sobre la producción de embriones in vitro. Se clasificaron COC obtenidos de ovarios de faenado en dos grupos: Tipo I (TI): ooplasma homogéneo con ≥ 4 capas de células del cumulo (CC) compactas y Tipo II (TII): ooplasma granular y ≥ 4 capas de CC ligeramente expandidas. Los COC fueron madurados por 24 horas en medio TCM199 y posteriormente fueron fertilizados in vitro durante 18 h. Los presuntos cigotos se cultivaron in vitro por siete días en medio SOF modificado. La calidad embrionaria se determinó por conteo de blastómeras posterior a tinción con Hoechst 33342. Se usó la prueba t para determinar diferencias estadísticas. La tasa de clivaje para los COC, TI (n=220) y TII (n=161), fue 88 ± 4% y 89 ± 8% respectivamente (p>0,05); la tasa de desarrollo embrionario fue 36 ± 7% y 33 ± 8% (p>0,05) respectivamente. El conteo de blastómeras para ambos grupos fue de (TI:101,TII:104) (n=10), (p>0,05). Los resultados de este trabajo permiten concluir que no hay diferencia en la cantidad y calidad de embriones producidos in vitro utilizando COC tipo I o tipo II, sugiriendo que ambas calidades podrían ser usadas en la producción de embriones in vitro a partir de animales Bos indicus.


Resumo A seleção morfológica dos complexos cumulus-oócito (COC) é crucial para a produção in vitro de embriões. Relata-se que COC que apresentam sinais de atresia têm a capacidade de gerar embriões. O objetivo deste estudo foi avaliar o efeito morfológico dos COC de animais Bos indicus com sinais de atresia precoce ou sim sinais de atresia sobre a produção in vitro de embriões. COC obtidos de ovários de animais abatidos foram classificados em dois grupos: tipo I (TI): ooplasma homogéneo com ≥ 4 camadas de células do cúmulos (CC) compactos e Tipo II (TII): ooplasma granular e ≥ 4 camadas CC levemente expandidas. Os COC foram submetidos à maturação por 24 horas em meio TCM199 e depois fertilizados in vitro por 18 h. Os prováveis zigotos foram cultivados in vitro por sete dias em meio SOFaa modificado. A qualidade embrionária foi determinada pela contagem dos blastômeros após coloração com Hoechst 33342. O teste t foi utilizado para determinar diferenças estatísticas significativas. A taxa de clivagem para os COC TI (n = 220) e TII (n = 161) foi de 88 ± 4% e 89 ± 8%, respectivamente (p>0,05); a taxa de desenvolvimento embrionário foi de 36 ±7% e 33 ±8% (p>0,05), respectivamente. E a contagem de blastômeros para ambos os grupos foi (TI: 101, TII: 104) (n = 10) (p>0,05). Com base nos resultados deste trabalho conclui-se que não existe diferença na quantidade e qualidade dos embriões produzidos in vitro, utilizando COC tipo I ou tipo II, sugerindo que ambas as qualidades podem ser utilizadas na produção in vitro de embriões em animaes Bos indicus.

OBJECTIVE: Hyaluronidase enzyme is an extremely important factor for the process of oocyte denudation, but little is known about its negative effects. METHODS: This prospective randomized study analyzed the results of using different concentrations of hyaluronidase (Diluted: 8IU/mL and Normal: 80IU/mL) used for denudation of sibling-oocytes for 22 women undergoing treatment for assisted reproduction by ICSI. A total of 192 oocytes were injected, being 104 for group I (diluted) and 88 for group II (normal). We analyzed fertilization rate, cleavage, embryo quality at 48 and 72 hours and number of transferred embryos in each group. RESULTS: The diluted enzyme group showed better results in fertilization rates (92.3% vs. 80.6%), mean cleavage (4.18 ± 2.57 vs. 3.09 ± 1.90), in 48-hour embryos A and A + B (60.9% vs. 44.1% and 90.2% vs. 82.3%) and at 72 hours (45.6% vs. 36.8% and 77.1% vs 66.2%), and number of embryos selected for transfer (61.8% vs. 38.1%). The overall pregnancy rate was 59.1%. CONCLUSION: This study demonstrates that the use of 8 IU/mL of hyaluronidase, according to the following protocol, is beneficial and can be successfully used for oocyte denudation, and it is also economically advantageous to the laboratory.

A produção in vitro de embriões (PIVE) é uma biotecnologia que possibilita principalmente o rápido aumento no número de descendentes com mérito genético superior. Atualmente o Brasil é líder mundial na PIVE e grande parte desse sucesso deve-se às inúmeras pesquisas realizadas nessa área. A seleção de oócitos de boa qualidade para essa técnica é determinante para obtenção de embriões de qualidade e satisfatórios índices de prenhez. Desta forma, esse trabalho teve como objetivo avaliar oócitos, obtidos de ovários com e sem a presença de corpo lúteo (CL), de acordo com a classificação morfológica e nuclear, afim de identificar a possível influência do CL na viabilidade dos oócitos. Foram coletados ovários de abatedouro e separados em dois grupos, com e sem CL. Os folículos foram aspirados, e os oócitos obtidos foram selecionados, classificados e em seguida corados com Hoechst 33342 e avaliados sob microscópio de epifluorescência, para avaliação nuclear. Realizou-se análise descritiva de cada tratamento com relação a classificação morfológica e nuclear. Os resultados obtidos foram analisados através de testes não-paramétricos, sendo o efeito do corpo lúteo verificado por meio do teste Qui-quadrado (2) (R core Team, 2013). Currently, Brazil is the world leader in IVP and much of this success is due to the extensive research carried out in that area. The selection of good quality oocytes for this technique is critical to obtaining quality embryos and satisfactory pregnancy rates. Thus, this study aimed to assess oocytes obtained from ovaries with and without the presence of corpus luteum (CL), according to the morphological and nuclear classification, in order to identify the possible influence of CL on the viability of oocytes. They were collected from slaughterhouse ovaries and separated into two groups, with and without CL. Follicles were aspirated and the oocytes were selected, ranked, and then stained with Hoechst 33342 and evaluated by epifluorescence microscope for nuclear evaluation. A descriptive analysis of each treatment with respect to morphological and nuclear classification. The results were analyzed using non-parametric tests, with the effect of the corpus luteum verified using the chisquare test (χ2) (R Core Team, 2013). It was concluded that the presence of CL does not influence the quality of immature oocytes, from the quantitative and morphological nuclear standpoint.(AU)

In this study, the polymerase chain reaction (PCR) was used to evaluate the presence of viral DNA in ovarian tissue, in the cumulus-oocyte complex (COC), follicular liquid, and blood of animals naturally infected with bovine herpesvirus-1 (BoHV-1). The serum profile of the sampled animals was also evaluated. Samples of serum, blood, ovarian tissue, follicular liquid, and COC were collected from 147 slaughterhouse animals that were not vaccinated against BoHV-1. Contaminated or insufficient samples were disregarded. Serological tests allowed the identification of serum-positive animals with neutralizing antibodies against BoHV-1. Analysis of samples by PCR revealed the presence of viral DNA in 0.9% (1/115) of the COC samples, in 4.3% (5/117) of the ovarian tissue samples, and in 2.8% (3/108) of the blood samples. Viral DNA was not detected in any of the follicular liquid samples. In serological samples, a positivity of 83.6% (117/140) was observed for BoHV-1. All PCR-positive animals, regardless of the samples analyzed, showed positivity in the serum neutralization test for the detection of BoHV-1-specific antibodies. According to these results, a high prevalence of antibodies against BoHV-1 was detected in naturally infected animals from different herds, and the molecular tests revealed the presence of viral DNA in bovine ovarian tissue, providing evidence that this might be...(AU)


Neste trabalho foi avaliada a presença do DNA viral, por meio da Reação em Cadeia da Polimerase (PCR), no tecido ovariano, nos oócitos, líquido folicular e sangue de vacas naturalmente infectadas. Também foi avaliado o perfil sorológico dos animais amostrados. Foram coletadas amostras de soro, sangue, tecido ovariano, líquido folicular e complexo cumulus-oócitos de 147 animais abatidos em frigorífico não vacinados contra o herpesvirus bovino 1 (BoHV-1). Amostras tóxicas ou insuficientes foram descartadas. Os testes sorológicos foram realizados permitindo a identificação dos animais soropositivos para anticorpos neutralizantes contra o BoHV-1. Foram realizadas as PCRs onde foi observada a presença do DNA viral em 0,9% (1/115) dos oócitos, em 4,3% (5/117) do tecido ovariano e em 2,8% (3/108) do sangue. Em nenhuma das amostras de líquido folicular foi detectado o DNA viral. Nas amostras sorológicas observou-se 83,6% (117/140) de positividade para o BoHV-1. Para estudar os fatores anatomofisiológicos que interferem na qualidade de complexos cumulus-oócitos (CCOs) bovinos, foram obtidas 396 ovários após abate de 198 fêmeas Bos indicus em frigorífico. Os ovários foram separados por categorias, sendo distribuídos em nulípara vs multípara e com progesterona (P4 - presença de corpo lúteo em um dos ovários) vs sem progesterona (NP4 - ausência de corpo lúteo). Todos os folículos foram mensurados e categorizados em pequenos (<6mm), médios (6 a 9mm) ou grandes (>9mm). Em seguida todos os folículos foram puncionados e os CCOs recuperados e avaliados morfologicamente. Não houve diferença na taxa de recuperação nem na qualidade dos CCOs de fêmeas nulíparas vs multíparas. O percentual de CCOs desnudos/degenerados foi maior no grupo NP4 e os CCOs expandidos foram superiores no grupo P4. A taxa de recuperação e o percentual de CCOs selecionados para PIV (graus I e II) foram similares nos grupos P4 vs NP4. Folículos pequenos apresentam menor taxa de recuperação em comparação aos de tamanho médio e grande, porém o percentual de CCOs de grau I foi superior em folículos pequenos e médios. Diante dos resultados aqui encontrados conclui-se que a categoria da doadora e a progesterona não influenciaram a qualidade de CCOs selecionados para PIV e que folículos menores apresentam de CCOs de melhor qualidade.(AU)


To study the anatomical and physiological factors that affect the bovine cumulus-oocyte complexes (COCs) quality, were examined 396 ovaries obtained after slaughter of 198 Bos indicus females. Ovaries were categorized in nulliparous vs multiparous and progesterone (P4 - a corpus luteum at least one ovary) vs without progesterone (NP4 - absence of corpus luteum). All follicles were measured and categorized into small (<6mm), medium (6 to 9mm) or large (>9mm). Then all follicles were punctured and COCs recovered and assessed morphologically. There was no difference in COCs recovery rate or quality from nulliparous vs multiparous females. The rate of denuded or degenerated oocytes was higher in NP4 and expanded COCs were higher in P4. The COCs recovery and grades I and II rates were similar in P4 vs NP4 groups. Small follicles have lower COCs recovery rate than medium or large follicles, but the grade I oocytes rates was higher in small and medium follicles. In conclusion, the category of donor female and progesterone did not affect the quality of COCs selected for PIV and smaller follicles present best quality of COCs.(AU)
