RESUMO

This study aimed to investigate the effects of chronic ß-alanine (ßA) plus acute sodium bicarbonate (SB) co-supplementation on neuromuscular fatigue during high-intensity intermittent efforts in swimming. Eleven regional and national competitive-level young swimmers performed a neuromuscular fatigue assessment before and immediately after two 20 × 25-m front crawl maximal efforts every 90 s, performed at pre- and post-4-week co-supplementation. Neuromuscular fatigue was evaluated by percutaneous electrical stimuli through the twitch interpolation technique on the triceps brachii and quadriceps femoris. Performance was defined by the mean time of the 20 efforts and blood samples to lactate concentrations were collected every four efforts. Participants supplemented 3.2-6.4 g·day-1 of chronic ßA or placebo (PL) during four weeks, and acute 0.3 g·kg-1 of SB or PL 60 min before the second assessment (allowing ßA+SB and PL+PL groups). No statistical changes were found in neuromuscular fatigue of triceps brachii. In the quadriceps femoris, a main effect of time was found in potentiated twitch delta values in pooled groups, showing a statistical increase of 19.01% after four weeks (Δ = 13.05 [0.35-25.75] N; p = 0.044), without time × group interactions. No statistical difference was found in the swimming performance. Blood lactate increased by 25.06% only in the ßA+SB group (Δ = 6.40 [4.62-8.18] mM; p Bonf < 0.001) after the supplementation period. In conclusion, 4-week ßA and SB co-supplementation were not able to reduce neuromuscular fatigue levels and improve performance in highintensity intermittent efforts, but statistically increased blood lactate levels.

RESUMO

Hypersecretion of gastric acid damages the stomach lining, causing the formation of peptic ulcers. Mucilage from medicinal plants offers a relaxing and soothing effect to the endodermal lining of the gut and has antacid properties, which can protect the mucosal lining from gastric acidity. This is the first report aimed to evaluate the physicochemical characteristics, acid-neutralizing, and cytotoxicity properties of traditionally used aqueous mucilage from Asparagus exuvialis and Sesamum capense. The physicochemical properties were determined by biochemical methods. Acid neutralizing and buffering capacities were determined by titration methods. Normal mouse embryonic fibroblast cells were used for cytotoxicity evaluation by MTT assay. The physicochemical characterization confirmed the presence of carbohydrates, alkaloids, saponins, proteins, tannins, flavonoids, and glycosides. Sesamum capense mucilage exhibited the most potent artificial gastric juice neutralizing capacity pH of 4.62±0.01, 8.0±0.00 acid neutralization capacity per gram of acid, and 30 minutes duration of acid neutralization. The aqueous mucilage from S. capense did not cause any significant cytotoxicity to 3T3 cell lines showing an IC50 value of 91.5 ± 0.06 µg mL-1, confirming the safe nature of the mucilage. These findings revealed that S. capense has the potential to neutralize gastric acid responsible for ulceration and can be safely consumed.(AU)

RESUMO

The aim of this study was to evaluate the effect of inclusion of two acidifiers blend (with different protection levels) on the acidification potential in vitro, apparent total tract digestibility of diet nutrients, blood metabolites, and average daily gain of nursery piglets. Two commercial acidifiers blend additives (AC1 and AC2) were evaluated. The acidification potential of the two acidifiers blend added to the diet were tested by buffering capacity and initial pH in vitro. For the evaluation of apparent digestibility of diet nutrients, blood metabolites and average daily gain, two experiments were carried out. The AC2 acidifier had a greater effect on the initial reduction in pH and a lower linear buffering rate of the diet. The use of acidifiers did not improve the nutrient digestibility of the diets and had no effect on the blood metabolite concentrations of urea, total protein, albumin, globulin, and lactate. Although the inclusion of acidifiers in the diets did not affect the apparent total tract digestibility of the diet nutrients the inclusion of the AC2 acidifier at different levels showed a negative quadratic effect on the average daily gain, with the best inclusion level being 0.26%.

In the present study, the modification of branched polyethyleneimine (b-PEI) was carried out using mesquite gum (MG) to improve its hemocompatibility to be used in biomedical applications. In the copolymer synthesis process (carboxymethylated mesquite gum grafted polyethyleneimine copolymer (CBX-MG-PEI), an MG carboxymethylation reaction was initially carried out (carboxymethylated mesquite gum (CBX-MG). Subsequently, the functionalization between CBX-MG and b-PEI was carried out using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and N-hydroxysuccinimide (NHS) as crosslinking agents. The synthesis products were characterized using Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), and thermogravimetric analysis (TGA). Thermogravimetric analysis showed that CBX-MG and CBX-MG-PEI presented a lower decomposition temperature than MG. The CBX-MG-PEI has a high buffer capacity in the pH range of 4 to 7, similar to the b-PEI. In addition, the CBX-MG-PEI showed an improvement in hemocompatibility in comparison with the b-PEI. The results showed a non-hemolytic property at doses lower than 0.1 µg/mL (CBX-MG-PEI). These results allow us to propose that this copolymer be used in transfection, polymeric nanoparticles, and biomaterials due to its physicochemical and hemocompatibility properties.

RESUMO

Ensiling sorghum with tropical forages has been shown to be a promising alternative for ruminant feed production, as this approach improves the quality of the sorghum silage. In this context, the goal of this study was to evaluate the fermentation characteristics and nutritive value of sweet sorghum silage made with different levels of Paiaguas palisadegrass and Ipypora grass. The experiment was conducted in a completely randomized 2 x 6 factorial design with four replications. Two forages (Paiaguas palisadegrass and Ipypora grass) were added to sorghum at six levels (0, 20, 40, 60, 80 and 100%), for a total of 48 experimental silos. After 50 days of fermentation, the silos were opened, and the fermentation profile and chemical composition of the silage were analysed. The results showed that the addition of Paiaguas palisade grass and Ipyporagrass to the sorghum silage increased the pH, buffering capacity and NH3-NT of the silage and reduced its dry matter content but did not affect its fermentation characteristics, which remained within an appropriate range. The addition of increasing levels of Paiaguas palisadegrass and Ipypora grass to the sorghum silage increased the crude protein and in vitro dry matter digestibility (IVDMD) of the silage and decreased the fibre fraction (neutral detergent fibre, acid detergent fibre and lignin) and ether extract contents. Adding tropical forages to sorghum silage material at levels above 40% is recommended. Among forage crops, addition of Paiaguas palisadegrass to sorghum silage resulted in higher content of crude protein and IVDMD. Mixed silages can be an effective alternative to improve the quality of forage sorghum silage.

The study investigated the influence of ß-alanine supplementation during a high-intensity interval training (HIIT) program on repeated sprint ability (RSA) performance. This study was randomized, double-blinded, and placebo controlled. Eighteen men performed an incremental running test until exhaustion (TINC) at baseline and followed by 4-wk HIIT (10 × 1-min runs 90% maximal TINC velocity [1-min recovery]). Then, participants were randomized into two groups and performed a 6-wk HIIT associated with supplementation of 6.4 g/day of ß-alanine (Gß) or dextrose (placebo group; GP). Pre- and post-6-wk HIIT + supplementation, participants performed the following tests: 1) TINC; 2) supramaximal running test; and 3) 2 × 6 × 35-m sprints (RSA). Before and immediately after RSA, neuromuscular function was assessed by vertical jumps, maximal isometric voluntary contractions of knee extension, and neuromuscular electrical stimulations. Muscle biopsies were performed to determine muscle carnosine content, muscle buffering capacity in vitro (ßmin vitro), and content of phosphofructokinase (PFK), monocarboxylate transporter 4 (MCT4), and hypoxia-inducible factor-1α (HIF-1α). Both groups showed a significant time effect for maximal oxygen uptake (Gß: 6.2 ± 3.6% and GP: 6.5 ± 4.2%; P > 0.01); only Gß showed a time effect for total (-3.0 ± 2.0%; P = 0.001) and best (-3.3 ± 3.0%; P = 0.03) RSA times. A group-by-time interaction was shown after HIIT + Supplementation for muscle carnosine (Gß: 34.4 ± 2.3 mmol·kg-1·dm-1 and GP: 20.7 ± 3.0 mmol·kg-1·dm-1; P = 0.003) and neuromuscular voluntary activation after RSA (Gß: 87.2 ± 3.3% and GP: 78.9 ± 12.4%; P = 0.02). No time effect or group-by-time interaction was shown for supramaximal running test performance, ßm, and content of PFK, MCT4, and HIF-1α. In summary, ß-alanine supplementation during HIIT increased muscle carnosine and attenuated neuromuscular fatigue, which may contribute to an enhancement of RSA performance.NEW & NOTEWORTHY ß-Alanine supplementation during a high-intensity interval training program increased repeated sprint performance. The improvement of muscle carnosine content induced by ß-alanine supplementation may have contributed to an attenuation of central fatigue during repeated sprint. Overall, ß-alanine supplementation may be a useful dietary intervention to prevent fatigue.

RESUMO

Hydrological and hydrochemical processes occurring within riparian zones in temperate mid-latitudes flatland areas have significant implications for water management by controlling nutrient transfer between the watershed and the stream system. The riparian zone in a high-order flatland stream located within a 7063-km2 agricultural watershed in Argentina was investigated to study its hydrological connectivity to upland zones, interactions with the stream, and their implications for groundwater hydrochemistry. The analysis was based on 9-year-long time series of groundwater/stream water levels collected along a 220-m-long transect comprising six piezometers, a river stage sensor, and hydrochemical information from 37 groundwater/stream water sampling campaigns. Samples were analyzed for electrical conductivity (EC), Cl-, SO4+2, (Ca+2 + Mg+2), pH, and redox potential (ORP). Data were interpreted using descriptive statistics, statistical tests, groundwater flux calculations, and identification of hydrological patterns and associated hydrochemical responses. The system was hydrologically controlled by shallow groundwater. Three representative landscape hydrological patterns were identified: disconnected, incipient-weakly connected, and fully connected. Groundwater hydrochemistry was closely linked to hydrological connectivity, which played an important role in the mobilization and fluxes of solutes. Overall, groundwater EC, Cl-, SO4+2, and (Ca+2 + Mg+2) concentrations decreased from upland to lowland. For full connectivity, Cl- concentrations reduced 33%, while SO4+2 reduced 42%, demonstrating the system's buffering capacity. This investigation constitutes the first attempt to formulate the riparian zone functioning in this agricultural region and has contributed to the understanding on the complex interactions between hydrologic regimes of large flatland-high-order streams and shallow groundwater systems in fine-texture sediments.

RESUMO

ABSTRACT Humic acid (HA) may reduce adsorption and increase soil P availability, however, the magnitude of this effect is different when Ca2+ prevails over Mg2+ in limed soils. The objective of this study was to evaluate the effects of HA rates and carbonate sources on the adsorption, phosphate maximum buffering capacity (PMBC), and P availability in two contrasting soils. Oxisol and Entisol samples were firstly incubated with the following HA rates: 0, 20, 50, 100, 200 and 400 mg kg-1, combined with CaCO3 or MgCO3, to evaluate P adsorption. In sequence, soil samples were newly incubated with P (400 mg kg-1) to evaluate P availability. The least P adsorption was found when 296 mg kg-1 of HA was added to Oxisol. Applying HA rates decreased maximum adsorption capacity, increased P binding energy to soil colloids and did not alter PMBC of Entisol. Available P contents in Oxisol increased with HA rates, but it did not change in Entisol. Choosing the right HA rate can decrease PMBC up to 40% and increase the Oxisol P availability by 17%. Application of MgCO3 instead of CaCO3 decreased P adsorption in both soils. Thus, a positive correlation between Ca2+ content and PMBC was verified. Optimum rate of HA and the preponderance of Mg2+ instead of Ca2+ in soil volume fertilized with P are effective practices to reduce adsorption and increase P availability, especially in clayey Oxisol.

Since there is conflicting data on the buffering and ergogenic properties of calcium lactate (CL), we investigated the effect of chronic CL supplementation on blood pH, bicarbonate, and high-intensity intermittent exercise performance. Sodium bicarbonate (SB) was used as a positive control. Eighteen athletes participated in this double-blind, placebo-controlled, crossover, fully counterbalanced study. All participants underwent three different treatments: placebo (PL), CL, and SB. The dose was identical in all conditions: 500 mg/kg BM divided into four daily individual doses of 125 mg/kg BM, for five consecutive days, followed by a 2-7-day washout period. On the fifth day of supplementation, individuals undertook four 30-s Wingate bouts for upper body with 3-min recovery between bouts. Total mechanical work (TMW) for the overall protocol and for the initial (1st+2nd) and final (3rd+4th) bouts was determined at each session. Blood pH, bicarbonate, and lactate levels were determined at rest, immediately and 5 min after exercise. CL supplementation did not affect performance (P > 0.05 for the overall TMW as well for initial and final bouts), nor did it affect blood bicarbonate and pH prior to exercise. SB supplementation improved performance by 2.9% for overall TMW (P = 0.02) and 5.9% in the 3rd+4th bouts (P = 0001). Compared to the control session, SB also promoted higher increases in blood bicarbonate than CL and PL (+0.03 ± 0.04 vs +0.009 ± 0.02 and +0.01 ± 0.03, respectively). CL supplementation was not capable of enhancing high-intensity intermittent performance or changing extracellular buffering capacity challenging the notion that this dietary supplement is an effective buffering agent.

RESUMO

Wine is an acidic beverage; its pH (2.9-3.8) is critically important to its organoleptic properties. During degustation, wine interacts with <1 mL of mouth saliva, the pH of which is near 7.0. This is buffered predominantly by the carbonate/bicarbonate pair (pKa = 6.1). Few data are available on whether the buffering capacity of saliva may alter the pH of wine and thus its sensorial properties. In this study both in vitro and in vivo approaches were conducted to measure pH in mixtures of representative red and white wines with human saliva. Continuous additions of microvolumes of either wine to a definite volume (3 mL) of saliva in vitro resulted in a progressive and steep decline in the pH of the wine/saliva mixture. Thus, a few microliters of either wine (<0.27 mL) was sufficient to reduce the pH of saliva by 1 pH unit. Further additions of wine to saliva lowered the pH to that of the corresponding wine. In the in vivo assay, definite volumes (1.5-18 mL) of either wine were mixed for 15 s with the mouth saliva of individual healthy subjects before pH determination in the expectorated wine/saliva mixtures. Compared to saliva, pronounced decreases in pH were observed, thus approaching the pH of wine even with the smallest volume of wine in the assay. Altogether, these results demonstrate that the buffering capacity of wine prevails over that of saliva and that during degustation the pH of the wine/saliva mixture in the mouth is, at least temporarily, that of the corresponding wine.

Health care services show interest in establishing rehabilitation protocols with mandibular overdentures retained on two implants, but they require scientific evidence evaluated with reliable instruments. Objective: To evaluate masticatory performance, retention and therapeutic satisfaction of patientswith a model rehabilitation protocol for implant-retained overdentures. Materials and methods: This study was conducted between the Faculty of Dentistry, University of Concepción (FAO UDEC) and Bío Bío Health Service(SS-Bío Bio). Fourteen patients, selected users of Bío Bío Health Service, Los Angeles, received implant-overdentures using a protocol of the Oral Rehabilitation Prosthesis Program (ROMPE FAO UDEC). Two Neodent™ implants (3.75 × 11 and 13 mm) were installed in each patient and connected with early loading. Before and after installing and connecting the implants, (0, 30 and 60 days), therapeutic satisfaction was assessed using the ROMPE FAO UDEC graduate program questionnaire. Retention was measured with a Force Gauge Dynamometer, and masticatory performance was quantified with a peanut screening test. Data were processed with SPSS 16.0 program and Kolmogorov-Smirnov test and t- test were used. Significance was set at P < .05. Results: Masticatory efficiency significantly increases after connectingthe implants. Therapeutic satisfaction with the prosthesis and the tensile strength are highly significant. Conclusions: These results coincide with the vast majority of published studies. It can be scientifically argued that the ROMPE FAO UDEC, SS Bío Bío overdenture protocol is a reliable alternative.

OBJECTIVE To determine, using in vitro pH measurements, the buffering capacity of saliva when added to different energy drinks sold in Chile. METHOD The participation of 3 young and systemically healthy patients, with no diseases of the saliva glands, was obtained. Samples of stimulated saliva where obtained from each patient and then mixed and stored as one sample. The study used 13 energy drinks sold nationwide, with 5 mL of each one being distributed into 4 Falcon tubes. The pH of each of the energy drinks, the saliva sample, and drinking water was measured. 1 mL of drinking water was added into Falcon tube number 1, and 1 mL of saliva into the 3 remaining every 3 min until completing 13 mL of solution in each one (38% vol./vol.). The pH measurements where performed upon adding each 1 mL (saliva/water) to allow the pH meter to correctly register the data. RESULTS The pH levels for energy drinks range between pH 2.42 ± 0.008 (Battery Gingered®), to pH 3.44 ± 0.005 (Battery Sugar Free®). The saliva had a mean pH value of 7.99, and 7.05 for drinkable water. The energy drink that achieved a higher increase in pH level was Speed®, reaching a value of pH 4.38, while the energy drink that increased the pH level the least was Quick Energy®, only reaching a pH of 3.37. CONCLUSION The buffering capacity of saliva managed to increase the pH level of energy drinks analyzed in this study between 17 and 54%. However, saliva was unable to neutralize further than 4.38 the low levels of pH in these drinks, thus being critical to dental structure.

RESUMO

The forage conservation as silage is a way to provide good quality forage during grazing scarcity periods, in this way, the objective was to evaluate the effect of pre-wilting and nitrogen fertilization on the fermentation characteristics and nutritive value of white oat silage, in two harvest stages. The experiment was conducted in 2011 at UTFPR, Dois Vizinhos-PR, Brazil. The experimental design was in a randomized blocks with two factorial arrangement and three replicates. The main treatments were six nitrogen doses: 0, 40, 80, 120, 160 and 200 kg ha-1, and secondary the stages of cutting for silage: early flowering with and without pre-wilting, and at blooming without pre-wilting. It was observed that nitrogen fertilization promoted a reduction in pH and mineral content of the silage, reducing fiber content and increasing crude protein content (CP), when made at oat early flowering. The effect of pre-wilting in early flowering inhibits the production of effluents and has higher levels of CP, without any effect on the fermentation and nutritional value. The silage at blooming stage features better standards of fermentation, with lower pH and buffering capacity, but with lower nutritional value.(AU)

The purpose of this study was to evaluate the salivary pH and buffering capacity of reproductive age women, without hormonal therapy, with [contraceptive] (CC) and menopausal. Samples of stimulated (SS) and unstimulated (USS) saliva were collected in 9 women between 33 and 56 years old, which were divided into three groups; Group 1, consisting of women of reproductive age without hormonal therapy (control group), group 2, consisting of women of reproductive age with hormone therapy [contraceptive] (CC) and group 3, consisting of menopausal women. Salivary buffering capacity was determined by acid-base titration using the method described by Singer et al. (1983). The pH was measured with an Orion potentiometer, model 710A and a Fisher Scientific Accumet electrode. The buffering capacity in SNE, was (7.8±2.0) in the control group, (38.1±22.4) in women with AC and (12.6±2.4) in the group of menopausal women. When samples were evaluated SE, the behavior was similar and the observed results were (37.56±3.2) in group 2 over control group (13,8±3,2). No changes in the pH of both the SE as SNE evaluated in groups 2 , group except where the pH was significantly lower (p<0.05). The results seem to indicate that use of hormonal contraceptive therapy (estrogens) causes a change in both the pH and buffering capacity of saliva...

Aiming to evaluate the effect of increasing levels of soybean hulls (0, 7, 14, 21 and 28%) in horses concentrate, two experiments were conducted in the Equideocultura Sector of Faculdade de Zootecnia e Engenharia de Alimentos City Hall. The first experiment was the palatability test, composed by the joint analysis of variables: first action (action performed by the animal as soon as it came in contacted with the concentrates: smelled or ate), first choice (first concentrate effectively ingested by the animal), and ratio intake (amount of each one concentrate ingested in the total consumed, over a period of 3 minutes). Thus, ten horses were used in a completely randomized test. Effect was observed (p0.05), demonstrating that the preference and consumption of each one of the concentrates were similar. The second experiment conducted was the nutrient digestibility test and physicochemical characteristics of the feaces. Five mares were used, arranged in 5x5 Latin Square design. The diet had 60% roughage (Coast Cross hay) and 40% concentrate, to meet the nutritional requirements of the animal category. [...] Soybean hulls can be included in the horses diet at levels up to 28% of the concentrate, without causing deleterious effect on the nutrients digestibility and the concentrates palatability, and there wasn/'t deleterious effect on the feaces physicochemical characteristics, suggesting maintenance of the digestive tract healthy.

RESUMO

Experimental mini-silos were prepared, in a completely randomized design distributed in a 2 x 5 factorial arrangement, with two silages (orange peel and corn) and five treatments (without protein source; with urea; soybean meal; cotton seed meal and sunflower meal). The protein additives were added to each silage to achieve about 10% protein. The silos were opened 90 days after the make silages. The following determinations were accomplished: pH, lactic acid (LA), buffering capacity (BC), dry matter (DM), crude protein (CP), neutral detergent fiber (NDF), calcium (Ca), total carbohydrates (TC), non fibrous carbohydrates (NFC) and in vitro digestibility (IVD). The protein additives increased the DM averages of the silages. The pH of silages varied from 3.41 to 3.95, being the lactic acid and in vitro digestibility higher to the orange peel silage when compared to the corn silage. It can be concluded that the orange peel silage to be preserved in silage form with protein source.

Objective: Demonstrate the association between salivary flow and buffer capacity with caries experience in children with low and high Decayed, Missing and Filled Teeth Index (DMFT). Methods: A cross-sectional study was conducted with 60 6-to-11-year-old children of both sexes, who attended the Dental School of Universidad Autónoma de Baja California, Mexicali campus. The patients were assigned to two groups according to the DMFT score, group 1 (score 0-3) and group 2 (score 4 or more). A clinical record including patient’s general information and dental evaluation was filled out. Cavity experience evaluation was carried out following WHO parameters. Also, salivary flow and buffer capacity were measured in each individual. Results: A weak negative correlation between salivary flow and DMFT score was observed (r=–0.221); however, this trend was not statistically significant (p>0.05). Regarding the buffer capacity, statistically significant differences between this salivary property and DMFT were found (p<0.05). Conclusion: The buffer capacity of saliva was associated with lower caries experience. On the other side, stimulated salivary flow show was not found to be related with the DMFT score.
