INTRODUCTION: Various strategies have been researched to enhance the susceptibility of biofilms, given their tolerance to antibiotics. This study evaluated the effect of the anti-microbial peptide nisin in association with antibiotics used in regenerative endodontics, exploring different treatment times and biofilm growth conditions. METHODS: A mixture of 10 bacterial species was cultivated on dentin specimens anaerobically for 21 days. Biofilms were treated with 1 mL of high-purity nisin Z (nisin ZP, 200 µg/mL) and a triple antibiotic mixture (TAP: ciprofloxacin + metronidazole + minocycline, 5 mg/mL), alone or in combination. The effectiveness of antimicrobial agents was assessed after 1 and 7 days. During the 7-day period, biofilms were treated under 2 conditions: a single dose in a nutrient-depleted setting (ie, no replenishment of growth medium) and multiple doses in a nutrient-rich environment (ie, renewal of medium and antimicrobial agents every 48 h). After treatments, biofilm cells were dispersed, and total colony-forming units were counted. RESULTS: After 1 d-treatment, nisin ZP + TAP resulted in 2-log cell reduction compared to TAP alone (P < .05). After 7 d-treatment with a single dose, nisin ZP + TAP and TAP reduced bacteria to nonculturable levels (P < .05), whereas repeated antimicrobial doses did not eliminate bacteria in a nutrient-rich environment. No bacterial reduction was observed with nisin ZP alone in any treatment time. CONCLUSIONS: The additional use of nisin improved the TAP activity only after a short exposure time. Longer exposure to TAP or nisin + TAP in a nutrient-deprived environment effectively eliminated biofilms.

Abstract This in vitro study aimed to determine the efficacy of dentin bonding agents in preventing color changes following Regenerative Endodontic Procedures. One hundred twenty bovine incisors were endodontically prepared and randomly assigned to a two main factors design: application of a dentin bonding agent (Scotchbond Adper, 3M ESPE, St Paul, MN, USA) in the pulp chamber (Group 1, n=60) versus no bonding intervention (Group 2, n=60), and five levels of intracanal medication (n=12/subgroup): Triple antibiotic paste (TAP), double antibiotic paste (DAB), calcium hydroxide (CH), modified triple antibiotic paste (TAPM), and Control (CTL). Color changes were measured over 28 days at multiple time points (1, 3, 7, 14, 21, and 28 days) using the CIEDE2000 formula to calculate the color difference (ΔE00) from baseline (T0). The ΔE00 quantifies the perceptible color difference between the initial and final tooth color, with lower values indicating less discoloration. The results were analyzed using repeated measures ANOVA-2 and post-hoc Holm-Sidak tests. The TAP subgroups, both with and without the bonding agent, exhibited the highest color variation. However, a pulp chamber seal with a bonding agent showed a protective effect against discoloration compared to no seal, even though complete prevention was not achieved. All groups demonstrated ΔE00 values beyond acceptable interpretation thresholds for clinical application, primarily driven by a reduction in lightness (L*) and a decrease in redness (a* value, shifting towards green). In conclusion, while the pulp chamber seal with a bonding agent mitigated TAP-induced discoloration, it did not eliminate it.


Resumo Este estudo in vitro avaliou adesivos dentinários na prevenção de alterações de cor após procedimentos endodônticos regenerativos. Cento e vinte incisivos bovinos foram preparados endodonticamente e aleatoriamente designados para um desenho com dois fatores principais: aplicação de agente adesivo (Scotchbond Adper, 3M ESPE, St Paul, MN, EUA) na câmara pulpar (Grupo 1, n=60) versus não intervenção adesiva (Grupo 2, n=60), e cinco níveis de medicação intracanal (n=12/subgrupo): pasta de triantibiótica (TAP), pasta diantibiótica (DAB), hidróxido de cálcio (CH), pasta triantibiótica modificada (TAPM) e Controle (CTL). Alterações cromáticas foram monitoradas por 28 dias em intervalos (1, 3, 7, 14, 21, e 28 dias), usando CIEDE2000 para calcular a diferença de cor (ΔE00) em relação a cor inicial. O ΔE00 quantifica a diferença entre a cor inicial e final do dente, com valores menores indicando menos descoloração. Os resultados foram analisados usando ANOVA-2 de medidas repetidas e teste posthoc de Holm-Sidak. Os grupos utilizando medicação TAP, com ou sem adesivo, possuíram as maiores variações cromática. Contudo, o uso do adesivo na câmara pulpar mostrou um efeito protetor contra descoloração, em comparação com a ausência do selamento adesivo, embora a prevenção completa não tenha sido alcançada. Todos os grupos demonstraram valores de ΔE00 além dos limiares aceitáveis para aplicação clínica, principalmente devido à redução na luminosidade (L*) e redução no vermelho (a*, deslocando-se em direção ao verde). Em conclusão, enquanto o selamento da câmara pulpar com um agente adesivo mitigou a descoloração induzida pelo TAP, mas não a eliminou completamente.

The antimicrobial peptide LL-37 and D-amino acids (D-AAs) have been proposed as antibiofilm agents. Therefore, this study aimed to test the antimicrobial effect of antibiofilm agents associated with antibiotics used in regenerative endodontic procedures (the triple antibiotic paste­TAP: ciprofloxacin + metronidazole + minocycline). An endodontic-like biofilm model grown on bovine dentin discs was used in this study. After 21-day growth, the biofilms were treated with 1 mg/mL TAP, 10 µM LL-37, an association of LL-37 + TAP, 40 mM D-AAs solution, an association of D-AAs + TAP, and phosphate-buffered saline (negative control). Colony forming unit (CFU) data were analyzed by two-way ANOVA and Tukey's multiple comparison test (p < 0.05). LL-37 + TAP showed the best antibacterial activity (7-log10 CFU/mL ± 0.5), reaching a 1 log reduction of cells in relation to the negative control (8-log10 CFU/mL ± 0.7) (p < 0.05). In turn, no significant reduction in bacterial cells was observed with TAP, LL-37, D-AAs, and D-AAs + TAP compared to the negative control. In conclusion, the combination of antibiotics and LL-37 peptide showed mild antibacterial activity, while the combination of antibiotics and D-AAs showed no activity against complex biofilms.

Objective: This study aimed to evaluate the effect of calcium hydroxide and triple antibiotic paste as intracanal medication on the interappointment pain at 8, 24, and 48 hours postoperatively in patients with symptomatic apical periodontitis undergoing multiple visit root canal treatment. Material and Methods: Two hundred and seven systemically healthy patients under the age group of 18-45 years with mandibular molars presenting with symptomatic apical periodontitis which require root canal treatments were included in this study. After access cavity preparation, cleaning and shaping was done till ISO 25 size file, and the patients were randomized into three groups (each group of 69 samples). Group I: no medicament, group II: calcium hydroxide and group III: triple antibiotic paste (TAP). Postoperative pain was evaluated at 8 hours, 24 hours and 48 hours. Results: The results showed that at 8 hours, 24hours and 48hours, there was a statistical difference between I and III (p < 0.05); and Group III and Group II (p < 0.05). Within the group, there was a statistical difference at all time points IN Group I and II (p < 0.05) except between 24 hours and 48 hours in the Group III (p > 0.05). Conclusion: Within the limitations of this study, TAP was more effective than calcium hydroxide in relieving pain and reducing the analgesic intake at the first 24hours.(AU)


Objetivo: Este estudo teve como objetivo avaliar o efeito do hidróxido de cálcio e da pasta tripla de antibiótico como medicação intracanal na dor na interconsulta às 8, 24 e 48 horas de pós-operatório em pacientes com periodontite apical sintomática submetidos ao tratamento endodôntico por múltiplas visitas. Material e Métodos: Duzentos e sete pacientes sistemicamente saudáveis com idade inferior a 18-45 anos com molares inferiores apresentando periodontite apical sintomática que requerem tratamento de canal radicular foram incluídos neste estudo. Após o preparo da cavidade de acesso, a limpeza e modelagem foram feitas até arquivo ISO 25, e os pacientes foram randomizados em três grupos (cada grupo de 69 amostras). Grupo I: sem medicamento, grupo II: hidróxido de cálcio e grupo III: pasta tripla de antibiótico (TAP). A dor pós-operatória foi avaliada em 8 horas, 24 horas e 48 horas. Resultados: Os resultados mostraram que às 8 horas, 24 horas e 48 horas, houve diferença estatística entre I e III (p <0,05); e Grupo III e Grupo II (p <0,05). Dentro do grupo, houve diferença estatística em todos os momentos do Grupo I e II (p <0,05), exceto entre 24 horas e 48 horas no Grupo III (p>0,05). Conclusão: Dentro das limitações deste estudo, o TAP foi mais eficaz do que o hidróxido de cálcio no alívio da dor e na redução da ingestão de analgésicos nas primeiras 24 horas. (AU)

INTRODUCTION: This study aimed to assess the effectiveness of antibacterial activity of medications used in regenerative endodontic treatment. METHODS: Sixty-seven dentin cylinders of single-rooted teeth were contaminated with a culture of Enterococcus faecalis (ATCC 29212; American Type Culture Collection, Manassas, VA) for 5 days. Samples were divided into 1 control group and the following experimental groups according to the medication applied: traditional triple antibiotic paste (TAP), clindamycin-modified TAP (mTAP), triple antibiotic medication with macrogol (3Mix-MP), clindamycin-modified 3Mix-MP (m3Mix-MP), calcium hydroxide (CH), and ethanol extract of propolis (EEP). After 14 days, the medications were removed, and the samples were submitted to confocal laser scanning microscopic analysis to quantify the percentage of viable bacteria. The distribution of data was confirmed by the Shapiro-Wilk test. The Kruskal-Wallis and Dunn tests were used for intergroup comparisons, and the Wilcoxon test was used for comparison between superficial and deep antibacterial efficacy for the same medication. The level of significance was set at P < .05. RESULTS: 3Mix-MP and m3Mix-MP presented significantly higher antibacterial efficacy compared with the other tested medications (P < .05), except for mTAP. mTAP was more effective than TAP (P < .05). The antibacterial efficacy of EEP and CH did not differ significantly from TAP and mTAP (P > .05). All medications showed effective antibacterial action compared with the control group (P < .05). CONCLUSIONS: 3Mix-MP and m3Mix-MP, which present extremely high concentrations of antibiotics (1500 mg/mL), were not more effective than mTAP at the concentration recommended by the American Association of Endodontists (5 mg/mL). Moreover, CH and EEP were as effective as TAP and mTAP.

AIM: To evaluate the antimicrobial and immunomodulatory activity of double antibiotic paste (DAP) in an in vitro infection model. METHODOLOGY: The minimum inhibitory and bactericidal concentrations (MIC and MBC) and the antibiofilm activities (TTC assay) of DAP and its components (ciprofloxacin and metronidazole) were evaluated against Staphylococcus aureus and Enterococcus faecalis compared with triple antibiotic paste (TAP). The cellular viability of RAW 264.7 macrophages (24 and 72 h) and L929 fibroblasts (48 and 72 h) was evaluated by MTT. Furthermore, the production of TNF-α, IL-12, IL-6, IL-1α, IL-10 and NO (on RAW 264.7), besides IL-6, TGF-ß and NO (on L929), stimulated with DAP in baseline and associated with heat-killed microbial-antigen conditions was measured by ELISA and Griess reaction. Data were analysed using the one-way ANOVA test with Bonferroni's corrections. RESULTS: The MBC of pharmacopoeia DAP was similar to TAP for E. faecalis (0.25 µg.  mL-1 ) and lower for S. aureus (DAP 1 µg. mL-1 and TAP 2 µg. mL-1 ; p < .001). Ciprofloxacin was the most effective antibiofilm drug from the pastes (35% of reduction for E. faecalis and S. aureus; p < .0001), and both pastes had a similar antibiofilm eradication against both biofilm species (29% and 35% for S. aureus and 76% and 85% for E. faecalis; p < .0001). DAP was cytotoxic against the tested cells. DAP significantly upregulated IL-1α (p < .001), IL-6 (p < .0001), TNF-α (p < .01) and IL-12 (p < .05; in the absence of antigens) and significantly reduced IL-6 (p < .0001; in the presence of HK-S. aureus) and IL-10 (p < .05; in the presence of both antigens) on macrophages. Furthermore, DAP upregulated IL-6 (p < .001) and NO (p < .05; in the absence of antigens), IL-6 (p < .001; in the presence of HK-S. aureus) and reduced NO (p < .001; in the presence of HK-S. aureus). CONCLUSIONS: Double antibiotic paste and TAP had similar antimicrobial activity against S. aureus and E. faecalis. DAP upregulated pro-inflammatory cytokines mainly in the absence of antigens and had pro- and anti-inflammatory activity in RAW 264.7 macrophages and L929 fibroblasts in the presence of antigens involved in pulp infections.

This study aimed to evaluate the antibacterial efficacy and discolouration potential of antibiotic pastes containing macrogol. Seventy single-rooted teeth were contaminated with a culture of Enterococcus faecalis for 30 days. Double (DAP) and triple antibiotic pastes (TAP) containing propylene glycol or macrogol as vehicles were applied. After 15 days, bacterial viability was compared. Change in teeth coronal colour was measured by a spectrophotometer. Statistical analysis was performed using anova and Tukey's test (P < 0.05). TAP containing clindamycin and propylene glycol was the only medicament that led to the presence of contamination, while the same combination of antibiotics containing macrogol did not result in bacterial growth (P < 0.05). Only the pastes containing minocycline in formulation resulted in coronal discolouration (P < 0.05). Both DAPs evaluated and the TAP containing clindamycin and macrogol showed to be the preferable options for regenerative endodontic therapy, as they were effective against E. faecalis biofilm and did not promote tooth discolouration.

Regenerative therapies such as dental pulpal revascularization appear as an option for traumatized immature permanent teeth. However, the triple antibiotic paste - TAP (metronidazole, minocycline, and ciprofloxacin), used for these therapies, can generate cytotoxicity and dentin discoloration. In contrast, host defense peptides (HDPs) are promising antimicrobial and immunomodulatory biomolecules for dentistry. This study aimed to evaluate in vitro the antimicrobial activity (against Staphylococcus aureus and Enterococcus faecalis) and the immunomodulatory potential (by the evaluation of IL-1α, IL-6, IL-12, IL-10, TNF-α and NO, in RAW 264.7 macrophages and IL-6, TGF-ß and NO, in L929 fibroblast) of synthetic peptides (DJK-6, IDR-1018, and IDR-1002), compared to TAP in an in vitro infection model containing heat-killed antigens from E. faecalis and S. aureus. Furthermore, the synergistic potential of ciprofloxacin and IDR-1002 was evaluated by checkerboard. Ciprofloxacin was the best antimicrobial of TAP, besides acting in synergism with IDR-1002. TAP was pro-inflammatory (p < 0.05), while the association of ciprofloxacin and IDR-1002 presented an anti-inflammatory profile mainly in the presence of both heat-killed antigens (p < 0.05). Based on these results, ciprofloxacin associated with IDR-1002 may demonstrate an efficient antimicrobial and immunomodulatory action in this in vitro model. Further in vivo studies may determine the real potential of this combination.

Objetivo: Presentar la terapia regenerativa como una al- ternativa para la resolución de un caso de traumatismo denta- rio en un diente permanente incompletamente desarrollado. Caso clínico: Se realizó el tratamiento de un incisivo central superior con mortificación pulpar y periodontitis api- cal aguda subsecuente a trauma dental en un paciente de 8 años de edad. Se aplicó el protocolo de regeneración pulpar recomendado por la Asociación Americana de Endodoncia. Se estimuló la formación de un coágulo en el interior del con- ducto a partir de los tejidos periapicales, previa desinfección con la pasta triple antibiótica, y finalmente se colocó mineral trióxido agregado coronal a este. Se obtuvo así una matriz es- teril que permitió el crecimiento de nuevo tejido y se realiza- ron controles periódicos durante 4 años. Se constató silencio clínico. Radiográficamente, se observó la formación de tejido sobre las paredes del conducto y el cierre apical. Conclusión: La terapia regenerativa como alternativa de tratamiento, en este caso, permitió la disminución de la luz del conducto por el depósito de tejidos calcificados y el cierre del foramen apical, mejorando el pronóstico de la pieza dentaria (AU)


Aim: To present pulp regeneration therapy as an alter- native to resolve dental trauma in immature permanent teeth. Clinical case: We report a clinical case of an immature central superior incisor with pulp mortification and acute api- cal periodontitis subsequent to dental trauma, in an 8 year old patient. The pulp Regeneration protocol recommended by the American Endodontics Association was applied. We stimulated a clot formation inside the duct from periapical tissues and after disinfection with a mixture of three antibi- otics mineral trioxide aggregated was finally place coronal to the clot. Thus a sterile matrix was obtained that allowed new tissue's growth. Periodic check-up visits were carried out over a 4 years period. Clinical silence was observed. Tissue formation on duct walls and apical closure were radiograph- ically detected (AU) Conclusion: Regenerative therapy is an alternative for the treatment of immature permanent teeth, in ths clinical case it allowed the reduction of the width of the duct by the opposition of hard tissues and the closure of the apical fora- men improving the forecast of these teeth.

OBJECTIVES: This study aimed to quantitatively assess the ability of two single-step restorative materials to avoid crown darkening caused by the use of minocycline as an intracanal medicament. MATERIALS AND METHODS: After coronal access and instrumentation, 120 maxillary incisors were divided into four groups (n = 30). Two experimental groups according to the restorative material applied to the inner walls of the access cavity: OB, OptiBond All-In-One + minocycline intracanal paste; U200, RelyX U200 + minocycline intracanal paste. Two control groups without restorative materials: MIN, minocycline intracanal paste and SL, saline intracanal. Color determination was performed using a spectrophotometer at five time points, immediately after materials were applied (baseline), and at 7, 14, 21, and 28 days from the baseline. RESULTS: Decrease in the mean values of L* (luminosity) was observed after insertion of minocycline paste in all groups at all time points. Statistically significant differences were absent between the time points (P > .05). After 28 days, MIN showed significantly more darkening (ΔL*) (- 10.6 ± 7.3) than OB (- 5.4 ± 6.2), U200 (- 5.8 ± 3.9) and SL (- 2.3 ± 1.2) (P < .05). CONCLUSIONS: Crown darkening can be minimized by the previous application of RelyX U200 or OptiBond All-In-One to the inner walls of the access cavity before a minocycline-containing paste is applied as an intracanal medication. CLINICAL RELEVANCE: The American Association of Endodontists Clinical Considerations for Regenerative Procedures in necrotic immature teeth suggests the triple antibiotic paste as an intracanal medication (2018). However, discoloration and crown darkening are common unfavorable outcomes. The clinical protocol suggested in this paper has shown to be able to minimize crown darkening, predictably leading to a better patient-centered clinical success.

Introduction: The objective of this study was to evaluate the effects of short term intracanal medicaments on the fracture resistance of root in simulated necrotic immature teeth in pulp revitalization procedure. Methods and Materials: Bovine teeth (n=180) were selected, sectioned coronally and apically and then, internally fragilized. Intracanal medicament groups were arranged as follows: "Triple Antibiotic Paste" (TAP) group (n=60), "Calcium Hydroxide" (CH) group (n=60) and the control group (n=60). No medication was used in the control group. Fracture resistance tests were performed after 7, 14, and 21 days. At allocated intervals, 20 teeth from each group had fractured. The Kruskal-Wallis and Mann-Whitney tests were performed to verify the effects of the employed medicaments at each time point. Friedman test and Wilcoxon signed-rank test were also performed to verify the association between time and fracture resistance. The level of significance was set at 0.05. Results: After 7 days, there was no statistical difference between groups (P=0.376). Intragroup analysis revealed that, after 21 days, the TAP group (P=0.015) and the CH group (P=0.006) presented a statically significant reduction in fracture resistance comparison with 7 days. Statistical difference was not verified for the control group after 7, 14 and 21 days (P=0.25). There was no statistical difference between CH group and the control group after 7, 14 and 21 days (P>0.05). The reduction was significant for TAP after 14 and 21 days (P=0.018, P=0.033 Respectively). Conclusions: This in vitro study showed that the duration at which TAP and CH remained in the root canal influenced the fracture resistance of bovine teeth with simulated incomplete root formation.

Abstract This systematic review aimed to evaluate whether alternative materials to conventional triple antibiotic paste (TAP - metronidazole, ciprofloxacin, and minocycline) and grey mineral trioxide aggregate (GMTA) could avoid tooth discoloration in teeth submitted to Regenerative Endodontic Procedure (REP). It was also investigated if dental bleaching is able to reverse the color of darkened teeth due to REP. The search was conducted in four databases (Medline via PubMed, Scopus, ISI Web of Science and BVS - Virtual health library), following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses. The retrieved papers were uploaded in the software EndNoteTM and two reviewers independently selected the studies and extracted the data. Only studies in humans (case reports, case series, clinical trials) were included in the review. From 1,122 potentially eligible studies, 83 were selected for full-text analysis, and 38 were included in the review. The included studies were mainly case reports (76.3 %). The studies described a total of 189 teeth submitted to REP. From these, about 54% of teeth presented some degree of discoloration. Most teeth presenting color alteration were treated with TAP, especially when combined with GMTA. Only three studies performed dental bleaching to restore the color of teeth and neither bleaching technique was able to restore the original color of the crowns. The use of alternative materials to TAP and GMTA, such as double antibiotic paste or Ca(OH)2 pastes and white mineral trioxide aggregate or BiodentineTM, reduces the occurrence of tooth discoloration.


Resumo Esta revisão sistemática teve como objetivo avaliar se materiais alternativos à pasta tri-antibiótica convencional (TAP, em Inglês - metronidazol, ciprofloxacina e minociclina) e ao agregado trióxido mineral cinza (MTA cinza) poderiam evitar a descoloração dentária em dentes submetidos ao procedimento endodôntico regenerativo (REP, em Inglês). Também foi investigado se o clareamento dental é capaz de reverter a cor dos dentes escurecidos devido ao REP. A busca foi realizada em quatro bases de dados (Medline via PubMed, Scopus, ISI Web of Science e BVS - Biblioteca Virtual de Saúde), seguindo os Principais Itens para Relatar Revisões Sistemáticas e Meta-análises. Os artigos obtidos foram carregados no software EndNoteTMe dois revisores independentes selecionaram os estudos e extraíram os dados. Apenas estudos em humanos (relatos de casos, séries de casos, ensaios clínicos) foram incluídos na revisão. De 1.122 estudos potencialmente elegíveis, 83 foram selecionados para análise do artigo completo, e 38 foram incluídos na revisão. Os estudos incluídos foram principalmente relatos de casos (76,3%). Os estudos descreveram um total de 189 dentes submetidos ao REP. Destes, cerca de 54% dos dentes apresentaram algum grau de descoloração. A maioria dos dentes com alteração de cor foi tratada com TAP, principalmente quando combinada com MTA-cinza. Apenas três estudos realizaram o clareamento dental para restaurar a cor dos dentes e nem a técnica de clareamento conseguiu restaurar a cor original das coroas. O uso de materiais alternativos à TAP e ao MTA cinza, como a pasta bi-antibiótica, pasta de Ca(OH)2e MTA branco ou BiodentineTM, reduz a ocorrência de descoloração dentária.

Abstract Regenerative endodontic procedure (REP) has been proposed as a new approach to treat immature permanent teeth. However, materials used in REP for root canal disinfection or cervical sealing may induce tooth discoloration. Objectives To assess tooth crown’s color after intracanal treatment with triple antibiotic paste (TAP) or calcium hydroxide (CH); cervical sealing with glass ionomer cement (GIC) or mineral trioxide aggregate (MTA); and bleaching with carbamide peroxide. Material and Methods After pulp removal and color spectrophotometer measurement, 50 bovine incisors were divided into 4 experimental groups and one control (untreated). Experiments were performed in phases (Ph). Ph1: TAP (ciprofloxacin, metronidazole, minocycline), TAPM (ciprofloxacin, metronidazole, amoxicillin), DAP (ciprofloxacin, metronidazole), or CH treatment groups. After 1 and 3 days (d); 1, 2, 3 weeks (w); and 1, 2, 3 and 4 months (m), color was measured and medications were removed. Ph2: GIC or MTA cervical sealing, each using half of the specimens from each group. Color was assessed after 1d, 3d; 1w, 2w, 3w; 1m and 2m. Ph3: Two bleaching sessions, each followed by color measurement. Data were analyzed with ANOVA and post-hoc Holm-Sidak method. Results Ph1: Specimens of TAP group presented higher color alteration (ΔE) mean than those of TAPM group. No significant difference was found among TAP or TAPM and CH, DAP or Control groups. Ph2: cervical sealing materials showed no influence on color alteration. Ph3: Different ΔE means (from different groups), prior to bleaching, became equivalent after one bleaching session. Conclusions TAP induces higher color alteration than TAPM; color alteration increases over time; cervical sealing material has no influence on color alteration; and, dental bleaching was able to recover, at least partially, the tooth crown’s color.

To compare the degree of pigmentation in coronal extracted premolars after applying bi-antibiotic and tri-antibiotic pastes. Method: Intentional non-probability sampling. A total of 30 healthy premolars which were extracted for orthodontic reasons and met the inclusion criteria were divided into 2 experimental groups and a control group of 10 teeth each. Group I (G1) was treated with bi-antibiotic paste, group II (G2) with tri- antibiotic paste; and group III or control (G3) was treated with saline solution. Two observers previously calibrated took the initial color sample from each tooth by giving them a value corresponding to the shade guide. After access opening and instrumentation, all teeth received the appropriate medication and color records were taken under the same initial light conditions for 30 days after the procedure. The obtained data were compiled into Excel for further descriptive and inferential analysis with SPSS 19.0. ANOVA test was used with a confidence level of 95 percent. Results: The degree of pigmentation was 1+/-0.9 in G1, 12.2+/-1.2 in G2 and 0.9+/-0.8 in G3. No statistically significant differences were found between G1 and G3 (p=0.8086), but there was between G1 and G2 (p<0.001), and G2 and G3 (p<0.001). Conclusion: The degree of pigmentation with tri-antibiotic paste was higher than the degree of pigmentation with bi-antibiotic paste used as temporary medication. No difference was found between the degree of pigmentation in the group treated with bi-antibiotic and the control group treated with saline solution. Bi-antibiotic paste did not show pigmentation in the crown of the extracted premolars...


Comparar el grado de pigmentación coronal en premolares extraídos después de aplicar pastas Bi-antibióticay Tri-antibiótica. Método: Muestreo no probabilístico e intencional. Se seleccionaron 30 premolares sanos extraídos por motivos ortodónticos, que cumplieran los criterios de inclusión, distribuidos en 2 grupos experimentales y un grupo control de 10 dientes cada uno. El grupo I (G1) fue tratado con pasta Bi-antibiótica, el grupo II (G2) con pasta Tri- antibiótica; y el grupo III o control (G3) fue tratado consuero fisiológico. Dos observadores previamente calibrados tomaron el color inicial a cada muestra dándoles un valor correspondiente a la guía de colores. Luego de la apertura e instrumentación, todos los dientes recibieron los medicamentos correspondientes por 30 días, tomándose registros de color con las mismas condiciones iniciales de luz. Los datos obtenidos se recopilaron en Excel para analizarlos de formadescriptiva e inferencial con el programa SPSS 19.0, se utilizó el test de ANOVA con un nivel de confianza del 95 por ciento. Resultados: El grado de pigmentación fue de 1+/-0,9 en G1,12,2 +/-1,2 en G2 y 0,9 +/-0,8 en G3. No se hallaron diferencias estadísticamente significativas entre G1 y G3 (p=0,8086), si las hubo entre G1 y G2 (p<0,001), y G2 y G3 (p<0,001).Conclusión: El grado de pigmentación con la pasta Tri-antibiótica fue superior al grado de pigmentación de la pastaBi-antibiótica utilizada como medicación temporal. No se observó diferencia entre el grado de pigmentación presentede la pasta Bi-antibiótica y el grupo control tratado con suero fisiológico. INTRODUCTION: Pulp revascularization may be considered a promising alternative for traumatized necrotic immature teeth. The aim of this study was to evaluate traumatized immature teeth treated with 2 protocols of pulp revascularization. METHODS: Twenty-three teeth of young patients (7-17 years old) with necrotic upper incisors caused by dental trauma were divided into 2 groups; one group was treated with triple antibiotic paste (metronidazole, ciprofloxacin, and minocycline) (TAP) (n = 12), and the other was medicated with combination of calcium hydroxide and 2% chlorhexidine gel (CHP) (n = 11). Patients were treated and followed up for a period from 9-19 months in 2 dental institutions for evaluation of clinical and radiographic data. RESULTS: Most of the teeth were affected by lateral luxation (47.8%). Clinical evaluation in group TAP showed significant reduction in spontaneous pain (P = .01), pain on horizontal percussion (P = .007), and pain on palpation (P = .03), whereas group CHP showed significant reduction in pain on vertical percussion (P = .03). Crown discoloration was observed significantly more in teeth of group TAP (83.3%) (P < .002). On radiographic exam, periapical repair was found in all TAP-treated teeth (P = .03). Similarly, the same findings were found for all teeth treated with CHP with exception of 1 tooth (P = .21). Apical closure was significantly observed in both groups (P < .05). Increase in root length was demonstrated in 5 teeth (41.7%) and 3 teeth (27.3%) of groups TAP and CHP, respectively. Thickening of lateral dentinal walls was observed in 5 teeth of each group. CONCLUSIONS: Revascularization outcomes for traumatized patients treated with the tested protocols presented similar clinical and radiographic data. However, TAP caused esthetic problem leading to tooth discoloration, which can be considered a disadvantage when compared with CHP.

INTRODUCTION: The aim of this study was to characterize the response of mouse subcutaneous tissue to triple antibiotic paste (TAP) using conventional light microscopy and real-time PCR (qRT-PCR). METHODS: Polyethylene tubes containing TAP or calcium hydroxide (CH) (ie, the control group) were implanted in mouse subcutaneous tissue. Animals that received empty tubes or no tubes were used as additional controls. After periods of 7, 21, and 63 days postimplantation, the specimens were removed and subjected to histologic processing. The number of inflammatory cells and vessels, vessel areas, vascular density, and relative percentage of collagen were evaluated. Gene expression of proinflammatory (interleukin-1 beta, tumor necrosis factor alpha, and interleukin 17) and anti-inflammatory (transforming growth factor beta) cytokines and angiogenic factors (vascular endothelial growth factor and hypoxia-inducible factor-1 alpha) was quantified by 7 and 21 days postimplantation. Results were analyzed using the Student t test, analysis of variance, and the Tukey test (α = 0.05). RESULTS: TAP induced an exuberant inflammatory and angiogenic response, with higher numbers of inflammatory cells, higher vascular area and density, and lower relative percentage of collagen compared with CH. In general, the expression of genes involved in inflammation and angiogenesis was higher in the TAP group compared with animals that received CH or empty tubes. CONCLUSIONS: The response of mouse subcutaneous tissue to TAP was characterized by exuberant and persistent inflammatory and angiogenic responses with no repair and high gene expression of biomarkers associated with inflammation and angiogenesis.
