RESUMO

Fear memory expression can be attenuated by updating the footshock perception during the plastic state induced by retrieval, from a strong unconditioned stimulus to a very weak one through deconditioning. In this process, the original fear association of the conditioned stimulus with the footshock is substituted by an innocuous stimulus and the animals no longer express a fear response. In the present study, we explore the boundaries of this deconditioning-update strategy by the characterization of this phenomenon. We found that there is an optimal mismatch between the footshock intensity delivered in the training and in the reactivation. Likewise, we characterized the temporal window that the protocol is efficient in hindering fear response. Our findings contribute to a better understanding of the limits in which deconditioning acts in attenuating fear memory, so that an optimized protocol using this strategy can be planned in order to deal with emotional disorders.

BACKGROUND: Pretreatments of drying can represent an alternative to minimize the negative aspect process on the dry samples. Thus, the influence of ethanol (ET) and freezing (FG) as drying pretreatments was analyzed for slices of papaya. The slices (5 mm) were submitted for drying kinetics (60 °C and 1.5 m s-1 ). Drying kinetics experimental data were fitted using Page's model and a diffusive model with boundary condition of types I and III. Also the thermophysical properties (thermal conductivity and specific heat) were determined and finally, the kinetics of ultrasound-assisted (40 kHz and 132 W) extraction of total phenolic compounds (TPCs) were realized. RESULTS: The combined method (ET + FG) was more efficient in reducing the drying time of papaya slices (48%) and the model 2 simulating the boundary condition of the third type (type III) showed the best fit to the experimental data. Effective diffusivity and convective coefficient were higher for ET + FG, where the maximum reduction in water content was 91% compared to fresh slices. The pretreatments did not influence the thermal conductivity, however, they were significant in the specific heat and in the extraction of TPCs, being higher in the time of 180 min. CONCLUSION: It was confirmed in the results presented that the combined pretreatment ET + FG is the most viable for drying papaya slices. Furthermore, it was found to be the most efficient in minimizing the loss of TPCs. Therefore, this pretreatment has great potential for application in the development of high value-added foods. © 2022 Society of Chemical Industry.

In the last decades, the numerical studies on hemodynamics have become a valuable explorative scientific tool. The very first studies were done over idealized geometries, but as numerical methods and the power of computers have become more affordable, the studies tend to be patient specific. We apply the study to the numerical analysis of tumor-targeting during liver radioembolization (RE). RE is a treatment for liver cancer, and is performed by injecting radiolabeled microspheres via a catheter placed in the hepatic artery. The objective of the procedure is to maximize the release of radiolabeled microspheres into the tumor and avoid a healthy tissue damage. Idealized virtual arteries can serve as a generalist approach that permits to separately analyze the effect of a variable in the microsphere distribution with respect to others. However, it is important to use proper physiological boundary conditions (BCs). It is not obvious, the need to account for the effect of tortuosity when using an idealized virtual artery. We study the use of idealized geometry of a hepatic artery as a valid research tool, exploring the importance of using realistic spiral-flow inflow BC. By using a literature-based cancer scenario, we vary two parameters to analyze the microsphere distribution through the outlets of the geometry. The parameters varied are the type of microspheres injected and the microsphere injection velocity. The results with realistic inlet velocity profile showed that the particle distribution in the liver segments is not affected by the analyzed injection velocity values neither by the particle density. NOVELTY STATEMENT: In this article, we assessed the use of idealized geometries as a valid research tool and applied the use of an idealized geometry to the case of an idealized hepatic artery to study the particle-hemodynamics during radioembolization (RE). We studied three different inflow boundary conditions (BCs) to assess the usefulness of the geometry, two types of particle injection velocities and two types of commercially available microspheres for RE treatment. In recent years, the advent in computational resources allowed for more detailed patient-specific geometry generation and discretization and hemodynamics simulations. However, general studies based on idealized geometries can be performed in order to provide medical doctors with some basic and general guidelines when using a given catheter for a given cancer scenario. Moreover, using an idealized geometry can be a reasonable approach which allows us to isolate a given parameter and control other parameters, so that parameters can be independently assessed. Even though an idealized geometry does not match any patient's geometry, the use of an idealized geometry can be valid when drawing general conclusions that may be useful in patient-specific cases. However, we believe that even if an idealized hepatic artery geometry is used for the study, it is necessary to account for the upstream and downstream tortuosity of vessels through the BCs. In this work, we highlighted the need of modeling the tortuosity of upstream and downstream vasculatures through the BCs.

Metropolitan areas may suffer with increase of air pollution due to the growth of urbanization, transportation, and industrial sectors. The Metropolitan Area of Vitória (MAV) in Brazil is facing air pollution problems, especially because of the urbanization of past years and of having many industries inside the metropolitan area. Developing air quality system is crucial to understand the air pollution mechanism over these areas. However, having a good input dataset for applying on photochemical models is hard and requires quite of research. One input file for air quality modeling which can play a key role on results is the lateral boundary conditions (LBC). This study aimed to investigate the influence of LBC over CMAQ simulation for particulate matter and ozone over MAV by applying four different methods as LBC during August 2010. The first scenario (M1) is based on a fixed, time-independent boundary conditions with zero concentrations for all pollutants; the second scenario (M2) used a fixed, time-independent concentration values, with average values from local monitoring stations; the third CMAQ nesting scenario (M3) used the nested boundary conditions varying with time from a previous simulation with CMAQ over a larger modeling domain, centered on MAV; and finally, the fourth GEOS-Chem scenario (M4) used the boundary conditions varying with time from simulations of global model GEOS-Chem. All scenarios runs are based on the same meteorology conditions and pollutant emissions. The air quality simulations were made over a domain 61 × 79 km centered on coordinates - 20.25° S, - 40.28° W with a resolution of 1 km. The results were evaluated with the measured data from the local monitoring stations. Overall, significant differences on concentrations and number of chemical species between the LBC scenarios are shown across all LBC scenarios. The M3 and M4 dynamic LBC scenarios showed the best performances over ozone estimates while M1 and M2 had poor performance. Although no LBC scenarios do not seem to have a great influence on total PM10 and PM2.5 concentrations, individual PM2.5 species like Na, NO3-, and NH4+concentrations are influenced by the dynamic LBC approach, since those hourly individual PM2.5 species from CMAQ nesting approach (M3) and GEOS-Chem model (M4) were used as an input to LBC.

Analysis of the human locomotor system using rigid-body musculoskeletal models has increased in the biomechanical community with the objective of studying muscle activations of different movements. Simultaneously, the finite element method has emerged as a complementary approach for analyzing the mechanical behavior of tissues. This study presents an integrative biomechanical framework for gait analysis by linking a musculoskeletal model and a subject-specific finite element model of the pelvis. To investigate its performance, a convergence study was performed and its sensitivity to the use of non-subject-specific material properties was studied. The total hip joint force estimated by the rigid musculoskeletal model and by the finite element model showed good agreement, suggesting that the integrative approach estimates adequately (in shape and magnitude) the hip total contact force. Previous studies found movements of up to 1.4 mm in the anterior-posterior direction, for single leg stance. These results are comparable with the displacement values found in this study: 0-0.5 mm in the sagittal axis. Maximum von Mises stress values of approximately 17 MPa were found in the pelvic bone. Comparing this results with a previous study of our group, the new findings show that the introduction of muscular boundary conditions and the flexion-extension movement of the hip reduce the regions of high stress and distributes more uniformly the stress across the pelvic bone. Thus, it is thought that muscle force has a relevant impact in reducing stresses in pelvic bone during walking of the finite element model proposed in this study. Future work will focus on including other deformable structures, such as the femur and the tibia, and subject-specific material properties.

The reconsolidation process is the mechanism by which strength and/or content of consolidated memories are updated. Prediction error (PE) is the difference between the prediction made and current events. It is proposed as a necessary condition to trigger the reconsolidation process. Here we analyzed deeply the role of the PE in the associative memory reconsolidation in the crab Neohelice granulata. An incongruence between the learned temporal relationship between conditioned and unconditioned stimuli (CS-US) was enough to trigger the reconsolidation process. Moreover, after a partial reinforced training, a PE of 50% opened the possibility to labilize the consolidated memory with a reminder which included or not the US. Further, during an extinction training a small PE in the first interval between CSs was enough to trigger reconsolidation. Overall, we highlighted the relation between training history and different reactivation possibilities to recruit the process responsible of memory updating.

The reconsolidation process is the mechanism by which the strength and/or content of consolidated memories are updated. This process is triggered by the presentation of a reminder (training cues). It is not always possible to trigger the reconsolidation process. For example, memory age and strength are boundary conditions for the reconsolidation process. Here, we investigated the dynamic changes in these conditions. We propose that the boundary conditions of the reconsolidation process are not fixed and vary as a consequence of the interaction between memory features and reminder characteristics. To modify memory properties, participants received a threatening social protocol that improves memory acquisition or a control condition (fake, without social interaction) prior to learning pairs of meaningless syllables. To determine whether a strong young or old declarative memory undergoes the reconsolidation process, we used an interference task (a second list of pairs of meaningless syllables) to disrupt memory re-stabilization. To assess whether the older memory could be strengthened, we repeated the triggering of reconsolidation. Strong young or old memories modulated by a threatening experience could be interfered during reconsolidation and updated (strengthened) by reconsolidation. Rather than being fixed, boundary conditions vary according to the memory features (strong memory), which indicates the dynamic nature of the reconsolidation process. Our findings demonstrate that it is possible to modify these limits by recruiting the reconsolidation process and making it functionally operative again. This novel scenario opens the possibility to new therapeutically approaches that take into account the reconsolidation process.

Following the presentation of a reminder, consolidated memories become reactivated followed by a process of re-stabilization, which is referred to as reconsolidation. The most common behavioral tool used to reveal this process is interference produced by new learning shortly after memory reactivation. Memory interference is defined as a decrease in memory retrieval, the effect is generated when new information impairs an acquired memory. In general, the target memory and the interference task used are the same. Here we investigated how different memory systems and/or their valence could produce memory reconsolidation interference. We showed that a reactivated neutral declarative memory could be interfered by new learning of a different neutral declarative memory. Then, we revealed that an aversive implicit memory could be interfered by the presentation of a reminder followed by a threatening social event. Finally, we showed that the reconsolidation of a neutral declarative memory is unaffected by the acquisition of an aversive implicit memory and conversely, this memory remains intact when the neutral declarative memory is used as interference. These results suggest that the interference of memory reconsolidation is effective when two task rely on the same memory system or both evoke negative valence.

ABSTRACT: In this article, we propose the application of a modified Taylor series method (MTSM) for the approximation of nonlinear problems described on finite intervals. The issue of Taylor series method with mixed boundary conditions is circumvented using shooting constants and extra derivatives of the problem. In order to show the benefits of this proposal, three different kinds of problems are solved: three-point boundary valued problem (BVP) of third-order with a hyperbolic sine nonlinearity, two-point BVP for a second-order nonlinear differential equation with an exponential nonlinearity, and a two-point BVP for a third-order nonlinear differential equation with a radical nonlinearity. The result shows that the MTSM method is capable to generate easily computable and highly accurate approximations for nonlinear equations. AMS SUBJECT CLASSIFICATION: 34L30.
