Pesticide use in crops is a severe problem in some countries. Each country has its legislation for use, but they differ in the degree of tolerance for these broadly toxic products. Several synthetic pesticides can cause air, soil, and water pollution, contaminating the human food chain and other living beings. In addition, some of them can accumulate in the environment for an indeterminate amount of time. The agriculture sector must guarantee healthy food with sustainable production using environmentally friendly methods. In this context, biological biopesticides from microbes and plants are a growing green solution for this segment. Several pests attack crops worldwide, including weeds, insects, nematodes, and microorganisms such as fungi, bacteria, and viruses, causing diseases and economic losses. The use of bioproducts from microorganisms, such as microbial biopesticides (MBPs) or microorganisms alone, is a practice and is growing due to the intense research in the world. Mainly, bacteria, fungi, and baculoviruses have been used as sources of biomolecules and secondary metabolites for biopesticide use. Different methods, such as direct soil application, spraying techniques with microorganisms, endotherapy, and seed treatment, are used. Adjuvants like surfactants, protective agents, and carriers improve the system in different formulations. In addition, microorganisms are a tool for the bioremediation of pesticides in the environment. This review summarizes these topics, focusing on the biopesticides of microbial origin.

Endophytic fungi are associated with plant health and represent a remarkable source of potential of enzymes and bioactive compounds, but the diversity of endophytes remains uncertain and poorly explored, especially in Cactaceae, one of the most species-rich families adapted to growing in arid and semi-arid regions. The aim of this study was to conduct a systematic review on the diversity and bioprospecting of endophytic fungi from Cactaceae. We analysed peer-reviewed articles from seven databases using PRISMA guidelines. The results showed that the Cactaceae family is a source of new taxa, but the diversity of endophytic fungi of Cactaceae is little explored, mainly the diversity among tissues and by metagenomics. Bioprospecting studies have shown that these microorganisms can be used in the production of enzymes and larvicidal and antifungal compounds. Our results are relevant as a starting point for researchers to develop studies that expand the knowledge of plant mycobiota in arid and semi-arid ecosystems, as well as comprising a remarkable source of fungal compounds with several biotechnological applications.

BACKGROUND: Tick control is a worldwide challenge due to its resistance to acaricides. Essential oils (EOs) and isolated compounds (EOCs) are potential alternatives for tick control technologies. METHODS: A review with EOs and EOCs, under field and semi-field conditions, was performed based on Scopus, Web of Science and PubMed databases. Thirty-one studies published between 1991 and 2022 were selected. The search was performed using the following keywords: "essential oil" combined with "tick," "Ixodes," "Argas," "Rhipicephalus," "Amblyomma," "Hyalomma," "Dermacentor," "Haemaphysalis" and "Ornithodoros." The words "essential oil" and "tick" were searched in the singular and plural. RESULTS: The number of studies increased over the years. Brazil stands out with the largest number (51.6%) of publications. The most studied tick species were Rhipicephalus microplus (48.4%), Ixodes scapularis (19.4%), Amblyomma americanum and R. sanguineus sensu lato (9.7% each). Cattle (70%) and dogs (13%) were the main target animal species. Regarding the application of EOs/EOCs formulations, 74% of the studies were conducted with topical application (spray, pour-on, foam, drop) and 26% with environmental treatment (spray). Efficacy results are difficult to evaluate because of the lack of information on the methodology and standardization. The nanotechnology and combination with synthetic acaricides were reported as an alternative to enhance the efficacy of EOs/EOCs. No adverse reactions were observed in 86.6% of the studies evaluating EOs/EOCs clinical safety. Studies regarding toxicity in non-target species and residues are scarce. CONCLUSIONS: This article provides a comprehensive review on the use of EOs and EOCs to reduce tick infestations, in both the hosts and the environment. As future directions, we recommend the chemical characterization of EOs, methodology standardization, combination of EOs/EOCs with potential synergists, nanotechnology for new formulations and safety studies for target and non-target organisms, also considering the environmental friendliness.

The agriculture is extremely important for the life of human beings. Therefore, it is needed to control the enemies that destroy vast areas of crops causing great economic losses. Chemical pesticides were the option for many decades, but the damage that they cause to environment and human health led to the idea of changing the use of these for more sustainable options such as biopesticides as a biological control. Among microbial pesticides, Serratia species have been found as suitable options to apply against several pests or pathogens. Serratia species produce a wide range of secondary metabolites with several biological activities such as antifungal, antibacterial, and pesticides which can be used in sustainable agriculture. It has been reported that several Serratia species are able to suppress some crop diseases caused by Fusarium oxysporum, Rhizoctonia solani, Phytophthora parasitica, Sclerotinia sclerotiorum, Verticillium dahlia, and Phytophthora capsici among others. Therefore, they have been used as biocontrol agents in agriculture. In this review, we summarized the genus Serratia describing its history and development and the metabolites it secretes, which are responsible for their antibacterial and antifungal activity. We have analyzed the insecticide capacity of several Serratia species as well antifungal properties of Serratia species against most important crops' pathogens. In conclusion, the use of Serratia as a biological control agent against plant pathogens can be a good option for a sustainable agriculture. More work is needed to assess the safety of the isolated new strains and their effectiveness against pathogens in in vivo conditions.

In Brazil, the production of beneficial microorganisms by growers exclusively for their own use is a practice known as "on-farm production". Regarding on-farm bioinsecticides, they were initially deployed for pests of perennial and semi-perennial crops in the 1970s but, since 2013, their use has extended to pests of annual crops such as maize, cotton, and soybean. Millions of hectares are currently being treated with these on-farm preparations. Local production reduces costs, meets local needs, and reduces inputs of environmentally damaging chemical pesticides, facilitating establishment of more sustainable agroecosystems. Critics argue that without implementation of stringent quality control measures there is the risk that the on-farm preparations: (1) are contaminated with microbes which may include human pathogens or (2) contain very little active ingredient, impacting on field efficacy. The on-farm fermentation of bacterial insecticides predominates, especially that of Bacillus thuringiensis targeting lepidopteran pests. However, there has been a rapid growth in the past 5 years in the production of entomopathogenic fungi, mostly for the control of sap-sucking insects such as whitefly (Bemisia tabaci (Gennadius)) and the corn leafhopper (Dalbulus maidis (DeLong and Wolcott)). In contrast, on-farm production of insect viruses has seen limited growth. Most of the ca. 5 million rural producers in Brazil own small or medium size properties and, although the vast majority still do not practice on-farm production of biopesticides, the topic has aroused interest among them. Many growers who adopt this practice usually use non-sterile containers as fermenters, resulting in poor-quality preparations, and cases of failure have been reported. On the other hand, some informal reports suggest on-farm preparations may be efficacious even when contaminated, what could be explained, at least partially, by the insecticidal secondary metabolites secreted by the pool of microorganisms in the liquid culture media. Indeed, there is insufficient information on efficacy and mode of action of these microbial biopesticides. It is usually the large farms, some with > 20,000 ha of continuous cultivated lands, that produce biopesticides with low levels of contamination, as many of them possess advanced production facilities and have access to specialized knowledge and trained staff. Uptake of on-farm biopesticides is expected to continue but the rate of adoption will depend on factors such as the selection of safe, virulent microbial strains and implementation of sound quality control measures (compliance with emerging Brazilian regulations and international standards). The challenges and opportunities of on-farm bioinsecticides are presented and discussed.

Fungal pathogens are one of the most important agents affecting crop production and food safety, and agrochemical application is one of the main approaches to reduce phytopathogenic fungi contamination in agricultural products. However, excessive and inadequate use can cause environmental damage, human and animal hazard, and increased phytopathogen resistance to fungicides. Biological control using lactic acid bacteria (LAB) and Bacillus spp. is an environmentally friendly strategy for phytopathogenic fungi management. Several molecules produced by these bacteria indeed affect fungal growth and viability in different plant crops. In this article, the activity spectra are reviewed along with the antifungal effect and antifungal compounds produced by LAB (e.g. organic acids, peptides, cyclic dipeptides, fatty acids, and volatile compounds) and Bacillus spp. (e.g. peptides, enzymes, and volatile compounds).

BACKGROUND: Freshwater organisms are facing increasing salinity levels, not only due to natural environmental processes, but also human activities, which can cause several physiological adaptations to osmotic stress. Additionally, these organisms might also have to deal with contamination by microbial insecticides. Our main goal was to use Chironomus xanthus to assess the chronic effects of increasing the salinity and commercial formulations of the microbial insecticides based on Bacillus thuringiensis subs. kurstaki (Btk) and Beauveria bassiana (Bb) as active ingredients, respectively. RESULTS: A significant interaction of growth was observed between the biopesticide based on Bb and NaCl on the larvae of C. xanthus. Single exposure to NaCl and each one of the formulations demonstrated deleterious impacts not only on larval development, but also on the emergence success and emergence time of this nontarget insect, with potential consequences for freshwater ecosystems due to cascading effects. CONCLUSION: The chronic effects induced by both bioinsecticides show that these formulations can have environmental impacts on nontarget freshwater insects. © 2023 Society of Chemical Industry.

The use of microbial insecticides and their toxins in biological control and transgenic plants has increased their presence in the environment. Although they are natural products, the main concerns are related to the potential impacts on the environment and human health. Several assays have been performed worldwide to investigate the toxicity or adverse effects of these microbial products or their individual toxins. This overview examines the published data concerning the knowledge obtained about the ecotoxicity and environmental risks of these natural pesticides. The data presented show that many results are difficult to compare due to the diversity of measurement units used in the different research data. Even so, the products and toxins tested present low toxicity and low risk when compared to the concentrations used for pesticide purposes. Complementary studies should be carried out to assess possible effects on human health.

The spotted wing drosophila (Drosophila suzukii) is one of the main invasive pests of small fruits in the world. Thus, 19 essential oils (EOs) were selected to analyze the effects through toxicity and repellency on oviposition and D. suzukii adults. In addition, their lethal and sublethal effects on the pupal endoparasitoid Trichopria anastrephae were evaluated. The EOs of C. flexuosus and Mentha spp. had the highest toxicity observed in the topical application bioassay for D. suzukii. In contrast, the EOs of C. verum, C. citratus QT citratus, and C. winterianus showed the highest toxicity in the ingestion bioassay for D. suzukii. The dry residues of C. verum and C. citratus QT citratus reduced the oviposition of D. suzukii. In the repellency bioassays, the 19 EOs analyzed repelled â 90% of the D. suzukii females. All EOs evaluated using the LC90 values of the products provided mortality of less than 20% of T. anastrephae adults and did not cause a reduction in the parasitism of surviving T. anastrephae females. We conclude that the EOs evaluated have the potential to be used in the management of D. suzukii. They can also serve as selective active ingredients for the formulation and synthesis of new biopesticides.

The use of fungal-based biopesticides to reduce pest damage and protect crop quality is often considered a low-risk control strategy. Nevertheless, risk assessment of mycopesticides is still needed since pests and beneficial insects, such as pollinators, co-exist in the same agroecosystem where mass use of this strategy occurs. In this context, we evaluated the effect of five concentrations of three commercial entomopathogenic fungi, Beauveria bassiana, Metarhizium anisopliae, and Cordyceps fumosorosea, by direct contact and ingestion, on the tropical stingless bees Scaptotrigona depilis and Tetragonisca angustula, temperate bee species, the honey bee Apis mellifera, and the bumble bee Bombus terrestris, at the individual level. Furthermore, we studied the potential of two infection routes, either by direct contact or ingestion. In general, all three fungi caused considerable mortalities in the four bee species, which differed in their response to the different fungal species. Scaptotrigona depilis and B. terrestris were more susceptible to B. bassiana than the other fungi when exposed topically, and B. terrestris and A. mellifera were more susceptible to M. anisopliae when exposed orally. Interestingly, increased positive concentration responses were not observed for all fungal species and application methods. For example, B. terrestris mortalities were similar at the lowest and highest fungal concentrations for both exposure methods. This study demonstrates that under laboratory conditions, the three fungal species can potentially reduce the survival of social bees at the individual level. However, further colony and field studies are needed to elucidate the susceptibility of these fungi towards social bees to fully assess the ecological risks.
RESUMO
Bacillus sp. strain FSQ1 was isolated from the common bean (Phaseolus vulgaris L.). The genome of this strain presented 3,598,499 bp; 43.0% G + C content; 925,913 bp N50; 2 L50; 33 contigs; 97 RNAs and 3,908 predicted coding DNA sequences (CDS) distributed in 315 subsystems. Based on genome mining, the biological control activity of strains FSQ1 could be associated with the biosynthesis of rhizocticin A and bacillibactin. Thus, this strain is a promising active ingredient for the formulation of biopesticides.

AIMS: The leaf-feeding pest Cerotoma arcuata tingomariana (Bechyné) (Coleoptera: Chrysomelidae) produces huge economic losses in different crops. This study aimed to produce conidia by semisolid-state fermentation and to establish the insecticidal activity of two formulation prototypes based on a native Beauveria bassiana isolate for controlling this pest. METHODS AND RESULTS: A novel fabric-based semisolid-state fermentation strategy for quick and large-scale conidia production was performed and characterized. Conidia were formulated as an emulsifiable concentrate (EC) and a water-dispersible granulate (WG). Afterwards, the mortality of C. a. tingomariana adults was assessed. A conidia concentration of 2.9 × 109 conidia cm-2 was obtained after 9 days-course fermentation and a yield of 33.4 g kg-1 dry-substrate. CONCLUSIONS: The polyester fabric-based fermentation is an efficient technique for producing and collecting B. bassiana spores. Regarding LC90 , the potency analysis showed that the EC was 21-fold more potent than the non-formulated conidia, and ~ 2.6-fold more potent than the WG. SIGNIFICANCE AND IMPACT OF STUDY: A high throughput fermentation based on polyester fabric as support for B. bassiana conidia production and subsequent formulation as an EC comprises a promising strategy for obtaining a bioproduct to control adults of C. a. tingomariana and other Chrysomelidae pests.

The essential oils (EOs) from bioactive species can provide an alternative tool for the management of stored grain insects that is less environmentally damaging than synthetic chemicals. The aim of this study was to assess the repellent action and toxicity of EOs obtained from phellandrene-rich Lippia origanoides, carvone-rich Lippia alba, citral-rich L. alba, and Pogostemon cablin aerial parts on adults of Ulomoides dermestoides. These EOs were isolated by hydrodistillation and characterized by gas chromatography coupled to mass spectrometry (GC-MS). The repellency assay was carried out using the area preference method, and the toxicity evaluated utilizing a filter paper contact test. The major components (>10%) of the studied EOs were sabinene (16.9%), trans-ß-caryophyllene (18.6%) and α-humulene (10.1%) for phellandrene-rich L. origanoides EO; limonene (40.1%) and carvone (37.7%) for carvone-rich L. alba EO; geranial (24.5%), geraniol (19.0%), and neral (11.9%) for citral-rich L. alba EO; and α-guaiene (13.3%), α-bulnesene (15.7%), and patchoulol (35.3%) for P. cablin EO. All EOs displayed 100% repellency at a concentration of 16 µL/mL, with lower toxicity than that elicited by the commercial repellent DEET. EO concentrations up to 8 µL/mL did not induce any mortality on the beetle. These findings show that the EOs provide active and safe molecules for natural repellent formulations to prevent and control insect infestations of stored products.

Pests and diseases are responsible for most of the losses related to agricultural crops, either in the field or in storage. Moreover, due to indiscriminate use of synthetic pesticides over the years, several issues have come along, such as pest resistance and contamination of important planet sources, such as water, air and soil. Therefore, in order to improve efficiency of crop production and reduce food crisis in a sustainable manner, while preserving consumer's health, plant-derived pesticides may be a green alternative to synthetic ones. They are cheap, biodegradable, ecofriendly and act by several mechanisms of action in a more specific way, suggesting that they are less of a hazard to humans and the environment. Natural plant products with bioactivity toward insects include several classes of molecules, for example: terpenes, flavonoids, alkaloids, polyphenols, cyanogenic glucosides, quinones, amides, aldehydes, thiophenes, amino acids, saccharides and polyketides (which is not an exhaustive list of insecticidal substances). In general, those compounds have important ecological activities in nature, such as: antifeedant, attractant, nematicide, fungicide, repellent, insecticide, insect growth regulator and allelopathic agents, acting as a promising source for novel pest control agents or biopesticides. However, several factors appear to limit their commercialization. In this critical review, a compilation of plant-derived metabolites, along with their corresponding toxicology and mechanisms of action, will be approached, as well as the different strategies developed in order to meet the required commercial standards through more efficient methods.

Nanoencapsulation of biopesticides is an important strategy to increase the efficiency of these compounds, reducing losses and adverse effects on non-target organisms. This study describes the preparation and characterisation of zein nanoparticles containing the botanical compounds limonene and carvacrol, responsive to proteolytic enzymes present in the insects guts. The spherical nanoparticles, prepared by the anti-solvent precipitation method, presented in the nanoparticle tracking analysis (NTA) a concentration of 4.7 × 1012 ± 1.3 × 1011 particles.mL-1 and an average size of 125 ± 2 nm. The formulations showed stability over time, in addition to not being phytotoxic to Phaseolus vulgaris plants. In vivo tests demonstrated that formulations of zein nanoparticles containing botanical compounds showed higher mortality to Spodoptera frugiperda larvae. In addition, the FTIC probe (fluorescein isothiocyanate) showed wide distribution in the larvae midgut, as well as being identified in the feces. The trypsin enzyme, as well as the enzymatic extract from insects midgut, was effective in the degradation of nanoparticles containing the mixture of botanical compounds, significantly reducing the concentration of nanoparticles and the changes in size distribution. The zein degradation was confirmed by the disappearance of the protein band in the electrophoresis gel, by the formation of the lower molecular weight fragments and also by the greater release of FTIC after enzymes incubation. In this context, the synthesis of responsive nanoparticles has great potential for application in pest management, increasing the selectivity and specificity of the system and contributing to a more sustainable agriculture.

Poor storage conditions provide favorable environment to stored grain pests for their growth. The bio-pesticides are the best alternatives to synthetic pesticides. Present study was conducted to compare toxicity of Rubus fruticosus and Valeriana jatamansi against granary weevil, Sitophilus granarius and subsequent changes in enzyme activity responsible for grain damage. In current research 5 g of R. fruticosus fruit and V. jatamansi rhizome powders were tested separately against S. granarius, in 50 g wheat whole grains for seven days in comparison with the control. The enzymatic activity of malate dehydrogenase and alpha-amylase was observed in the cellular extracts of S. granarius. The insects were crushed and homogenized in phosphate-buffer solution and centrifuged at 10000 rpm for 5 minutes. For the enzymatic measurement supernatant was tested; the spectrophotometer was adjusted at 340 nm. The reagents were mixed and incubated at 25 °C for five minutes. The cuvettes were placed in the experimental and reference sites of spectrophotometer and recorded the change in absorbance for 3-4 minutes. There was 5.60% and 14.92% reduction in the activity of malate dehydrogenase in R. fruticosus and V. jatamansi, treated insects, respectively. The alpha amylase enzyme activity was 6.82% reduced and 63.63% increase in R. fruticosus and V. jatamansi, treated insects, respectively. Present study addresses that both plant powders are effective against granary weevil by altering enzyme activities so both the plant powders can be used as bio-pesticides against the stored grains pests.(AU)
As más condições de armazenamento proporcionam um ambiente favorável às pragas armazenadas para o crescimento. Os biopesticidas são as melhores alternativas aos pesticidas sintéticos. O presente estudo foi conduzido para comparar a toxicidade de Rubus fruticosus e Valeriana jatamansi contra gorgulhos, Sitophilus granarius e subsequentes alterações na atividade enzimática responsáveis por danos aos grãos. Na pesquisa atual, 5 g de frutos de R. fruticosus e pós de rizoma de V. jatamansi foram testados separadamente contra S. granarius, em 50 g de grãos integrais de trigo por sete dias, em comparação com o controle. A atividade enzimática da malato desidrogenase e alfa-amilase foi observada nos extratos celulares de S. granarius. Os insetos foram esmagados e homogeneizados em solução tampão fosfato e centrifugados a 10000 rpm por 5 minutos. Para a medição enzimática, o sobrenadante foi testado; o espectrofotômetro foi ajustado a 340 nm. Os reagentes foram misturados e incubados a 25 °C por cinco minutos. As cubetas foram colocadas nos locais experimentais e de referência do espectrofotômetro e registradas as alterações na absorbância por 3-4 minutos. Houve redução de 5,60% e 14,92% na atividade da malato desidrogenase em R. fruticosus e V. jatamansi, insetos tratados, respectivamente. A atividade da enzima alfa amilase foi reduzida em 6,82% e aumento de 63,63% em R. fruticosus e V. jatamansi, insetos tratados, respectivamente. O presente estudo aborda que ambos os pós de plantas são eficazes contra o gorgulho do celeiro, alterando as atividades enzimáticas, de modo que ambos os pós de plantas possam ser usados como biopesticidas contra pragas de grãos armazenados.(AU)

Interest in chitin-degrading enzymes has grown over the years, and microbial chitinases are the most attractive and promising candidates for the control of plant pests (fungi and insects). Currently, there are many studies on chitinases produced by cultivable microorganisms; however, almost none of them have achieved acceptable applicability as a biopesticide in the field. Approximately 99% of the microorganisms from soil cannot be isolated by conventional culture-dependent methods, thus having an enormous biotechnological/genetic potential to be explored. On the basis of this, the present paper aims to provide a brief overview of the metagenomic opportunities that have been emerging and allowing access to the biochemical potential of uncultivable microorganisms through the direct mining of DNA sequences recovered from the environment. This work also shortly discussed the future perspectives of functional and sequence-based metagenomic approaches for the identification of new chitinase-coding genes with potential for applications in several agricultural and biotechnological industries, especially in biological control.

Biosurfactants can be widely used in industries as pharmaceutical agents, for microbial enhanced oil recovery, crop biostimulation, among others. Surfactin and rhamnolipids are well-known biosurfactants. These compounds have several advantages over chemical surfactants, however they are not economically competitive, since their production cost is up to 12 times higher than chemical surfactants. In this sense, an interesting approach is to replace synthetic culture medium, which represents ≈ 30% of the production cost by agro-industrial wastes. In addition, biosurfactant productivity can be easily enhanced by inductor supplementation into culture medium that triggers biosurfactant metabolism. Biosurfactant inducers are mainly a pool of hydrophobic molecules (e.g. olive oil-saturated and unsaturated fatty acids, proteins and vitamins). Nevertheless, there is little information on inducer effects of specific molecules (e.g. oleic acid). In general, hydrophobic inducers lead to higher fatty acid chain lengths (biosurfactant chemical structure). Therefore, the aim of this review was to critically discuss the current state of the art and future trends on biosurfactant production, in particular biosurfactant inducers. Taking into account the last 10 years, there is a clear lack of information on correlation between "inducers" or "hydrophobic inducers" AND "biosurfactants", since only 13 documents were found (Scopus database). Thus, it is essential to deeply investigate all inducer effects on biosurfactant production, mainly yield and chemical structure.

During the past years, microorganisms have been the cause of many problems for human's health. However, today with the development of many techniques of microbiology, the researchers have studied several roles of microorganisms which may help the society. Microbial-based products are expected to play important role in agriculture-enhancing plant production and therefore increasing crop's yieldeswani et al. . Microorganisms can act by several action mechanisms including antibiosis or mechanisms in plant-microbe interactions underlining the dual function of microbial strains toward plant nutrition and protection. The market has increased with the development of microbial-based products. Currently, it is normal to think that microorganisms help us in agriculture by applying them as biological control. In this mini review, we collect the last findings about this topic including very recent literature. KEY POINTS: ⢠Microorganisms play a beneficial role in agriculture by different mechanisms. ⢠One of these mechanisms is the secretion of chemical compounds with different activities.

The use of intensive non-sustainable agricultural practices for satisfying global food demand is degrading the agro-ecosystems, leading to their inability to produce efficient and equitable sources of calories. Microbial communities play an important role in the improvement of soil fertility and plant development; thus, the genetic and metabolic diversity of microbiota in agro-ecosystems is a promising alternative for designing microbial inoculants to not only produce enough food but also mitigates the economic, health, social, and environmental issues caused by conventional agriculture. This Special Issue has been launched to compile and inspire high-impact recent advancements on bioprospecting beneficial microorganisms as a sustainable strategy to warranty global food security.
