RESUMO

In recent years, the monitoring of tropane alkaloids, specifically hyoscyamine and scopolamine, in food has become a pressing concern. This is due to increasing reports of food contamination with these compounds worldwide, raising awareness about the potential risks associated with their consumption. A novel method is proposed here for the determination of the sum of (+)-hyoscyamine, (-)-hyoscyamine, and (-)-scopolamine in buckwheat-based matrices, using solid-liquid extraction at low temperature and quantification by bidimensional chromatography coupled to tandem mass spectrometry. The validated method presented a linear response in the concentration range of 2.5-15 µg kg-1 (r > 0.99). The precision and accuracy were in the ranges from 0.8 to 11.0 % and from 96 to 103 %, respectively. The limit of quantification (LOQ) was 2.5 µg kg-1. No contamination was found at levels above the LOQ in any of the 18 samples analyzed (buckwheat flour, grains, and gluten-free mix).

In this work, buckwheat husks (Fagopyrum esculentum) were modified by acid treatment and posteriorly employed to remove the ketoprofen in batch adsorption. The characterization results indicated that a more irregular surface with new empty spaces was generated after acid treatment. The adsorptive process was favored at acidic pH = 3. The dosage of 0.85 g L-1 was fixed for the kinetic and isothermal tests, obtaining good removal and capacity indications. The kinetic studies were better represented by pseudo-second-order, obtaining an experimental capacity of 74.3 mg g-1 for 200 mg L-1 of ketoprofen. An increase in temperature negatively affected the adsorption isotherm curves, resulting in a maximum capacity of 194.1 mg g-1. Thermodynamic results confirmed the exothermic nature of the process with physical forces acting. The adsorbent presented high efficiency in treating a synthetic effluent containing different drugs and salts, 71.2%. Therefore, adsorbent development from buckwheat husks treated with a strong acid is an excellent alternative, given the good removal results and the low cost for its preparation.

This study describes the use of the Mixture Design for simultaneals to improve the physical properties and acceptability of gluten-free bread (GFB) based on whole pseudocereals flour, as well as to define dough and bread instrumental predictors of the sensory quality of GFB. Three simplex-centroid designs were used to study the effects of each pseudocereal flour (amaranth - AF, buckwheat - BF, and quinoa - QF) blended with rice flour (RF) and potato starch (PS) on dough and bread properties. A total of 30 GFB formulations were produced and evaluated. Results reveal relationships between dough Mixolab parameters, such as C3 and C4, related to gelatinization and starch stability, with crumb moisture and firmness of GFB formulation, in which higher values of these parameters related to higher acceptability scores (>7 on a 10 cm hydroid hedonic scale). However, higher values of the secondary parameter C3-C4 was related to lower loaf-specific volume, impairing appearance and texture acceptability, as well as overall liking. The interaction effects between pseudocereal flour and RF increases dough consistency, bread volume, softness, and acceptability. Blends of 50% AF, BF, or QF with 50% RF results in GFB with high acceptability (overall liking of 8). The maximum pseudocereal proportions to obtain acceptable GFB (scores ≥ 7 for appearance, color, odor, texture, flavor acceptability and overall liking) were 60% AF, 85% BF, and 82% QF blended with RF. The combination of instrumental and sensory methods was useful to identify parameters capable of predicting the GFB quality, which may be useful for food scientists and producers to face the challenges regarding the development of healthier and better quality GFB to meet consumer needs.

The objective of this work was to model and identify the best models for estimating the leaf area, determined by digital photos, of buckwheat (Fagopyrum esculentum Moench) of the cultivars IPR91-Baili and IPR92-Altar, as a function of length (L), width (W) or length x width product (LW) of the leaf blade. Ten uniformity trials (blank experiments) were carried out, five with IPR91-Baili cultivar and five with IPR92-Altar cultivar. The trials were performed on five sowing dates. In each trial and cultivar, expanded leaves were collected at random from the lower, middle and upper segments of the plants, totaling 1,815 leaves. In these 1,815 leaves, L and W were measured and the LW of the leaf blade was calculated, which were used as independent variables in the model. The leaf area of each leaf was determined using the digital photo method (Y), which was used as a dependent variable of the model. For each sowing date, cultivar and thirds of the plant, 80% of the leaves (1,452 leaves) were randomly separated for the generation of the models and 20% of the leaves (363 leaves) for the validation of the models of leaf area estimation as a function of linear dimensions. For buckwheat, IPR91-Baili and IPR92-Altar cultivars, the quadratic model (Ŷ = 0.5217 + 0.6581LW + 0.0004LW2, R2 = 0.9590), power model (Ŷ = 0.6809LW1.0037, R2 = 0.9587), linear model (Ŷ = 0.0653 + 0.6892LW, R2 = 0.9587) and linear model without intercept (Ŷ = 0.6907LW, R2 = 0.9587) are indicated for the estimation of leaf area determined by digital photos (Y) based on the LW of the leaf blade (x), and, preferably, the linear model without intercept can be used, due to its greater simplicity.


O objetivo deste trabalho foi modelar e identificar os melhores modelos para a estimação da área foliar, determinada por fotos digitais, de trigo mourisco (Fagopyrum esculentum Moench) das cultivares IPR91-Baili e IPR92-Altar, em função do comprimento (C), da largura (L) ou do produto comprimento vezes largura (CL) do limbo foliar. Foram conduzidos dez ensaios de uniformidade (experimentos em branco), sendo cinco com a cultivar IPR91-Baili e cinco com a cultivar IPR92-Altar. Os ensaios foram realizados em cinco datas de semeadura. Aluminum toxicity (Al) is one of the main constraints for plant growth on acid soils. While most plants are sensitive to Al, some species have developed strategies to cope with this metal. Fagopyrum esculentum, Moench., var Mancan (Polygonaceae), despite being an aluminum-tolerant plant, shows root inhibition as a seedling during the first hours of exposure to Al, whereas at later times, it fully recovers. In this study, we assessed whether abscisic acid (ABA) levels and the antioxidant system might be involved in the early tolerance mechanisms of F. esculentum. The results showed that seedlings exposed to 50 µM Al for 3, 6, 12, 24, and 48 h showed decreases in the relative root growth (RRG), and there was an accumulation of Al in the root apex from 3 to 24 h. In addition, reactive oxygen species (ROS) levels increased, and were detected early after Al exposure; endogenous ABA levels increased and antioxidant enzyme activity increased, including catalase (CAT, EC1.11.1.6), glutathione reductase (GR, EC 1.6.4.2), ascorbate peroxidase (APX, EC 1.11.1.11), and superoxide dismutase (SOD, EC 1.15.1.1) activity. Seedlings treated with exogenous ABA also showed increased ROS levels and CAT and APX activity. The results suggest that after the first 12 h of Al treatment, root growth declines while ROS levels increase due to the entrance of Al into the root. However, the enzyme antioxidant system is promoted, which may impact the recovery of the root growth at later times and increasing levels of ABA might mediate this effect.

This study aimed at assessing the effect of physicochemical properties and the particle size of different fractions of buckwheat and quinoa on the behaviour of gluten-free dough and bread quality. Quinoa and buckwheat grains were milled with a hammer mill and then separated in three fractions. These fractions where then re-milled with a cyclonic mill to obtain samples of similar sizes. Results showed that the chemical composition of these fractions was very different and played a major role on bread quality. Proteins, lipids and fibre negatively affected bread quality, whereas starch-rich fractions were more adequate for breadmaking. Re-milling quinoa and buckwheat fractions increased bread volume, although chemical composition still influenced bread properties. For hammer-milled fractions, both the finest fractions resulted in breads with higher technological quality, as well as a final product with more fibre, minerals and proteins.

BACKGROUND: Common buckwheat (Fagopyrum esculentum) is an important staple food crop in southwest China, where drought stress is one of the largest limiting factors that lead to decreased crop production. To reveal the molecular mechanism of common buckwheat in response to drought stress, we performed a comprehensive transcriptomics study to evaluate gene expression profiles of common buckwheat during PEG-mediated drought treatment. RESULTS: In total, 45 million clean reads were assembled into 53,404 unigenes with an average length of 749 bp and N50 length of 1296 bp. A total of 1329 differentially expressed genes (DEGs) were identified by comparing wellwatered and drought-treated plants, out of which 666 were upregulated and 663 were downregulated. Furthermore, we defined the functional characteristics of DEGs using GO and KEGG classifications. GO enrichment analysis showed that the DEGs were significantly overrepresented in four categories, namely, "oxidoreductase activity," "oxidation­reduction process," "xyloglucan:xyloglucosyl transferase activity," and "apoplast." Using KEGG pathway analysis, a large number of annotated genes were overrepresented in terms such as "plant hormone signal transduction," "phenylpropanoid biosynthesis," "photosynthesis," and "carbon metabolism." Conclusions: These results can be further exploited to investigate the molecular mechanism of common buckwheat in response to drought treatment and could supply with valuable molecular sources for abiotic-tolerant elite breeding programs in the future.

SUMMARY This trial compared forage production and quality of Pearl Millet and Buckwheat in three cutting age on irrigation conditions during dry season, evaluated in a randomized block split plot design with tree replications. Was determined content of Dry Matter (DM), Crude Protein (CP), Neutral Detergent Fiber (NDF), Acid Detergent Fiber (ADF), Mineral Matter (MM) and in vitro gas production. Forage production of Buckwheat was higher than Pearl Millet in all the cutting ages. Pearl Millet CP content was similar to all three cuts but Buckwheat CP content was higher in the first cut and reduced in the others. The NDF content of Buckwheat was higher in the first cut and reduced in the others, meanwhile Pearl Millet presented low NDF value in the first cut but increased in the second and third cut. The Buckwheat presented high ADF content than Pearl Millet in all the cuts. In vitro gas production from Pearl Millet was higher in the first cut but more high for Buckwheat in the second and third cuts. Buckwheat produced more forage with high quality than Pearl Millet during dry season.


RESUMO Objetivou-se com este trabalho comparar o valor forrageiro do milheto e trigo-mourisco, sob irrigação, em diferentes idades de corte. Utilizou-se delineamento experimental em blocos ao acaso, com os tratamentos arranjados em modelo de parcelas subdivididas. As plantas foram cultivadas durante a estação seca, sob irrigação e colhidas em três idades de corte, 47, 57 e 67 dias (tratamentos). Foram determinados os teores de matéria seca (MS), proteína bruta (PB), fibra em detergente neutro (FDN), fibra em detergente ácido (FDA), matéria mineral e produção de gases in vitro e a produção de matéria seca. A produção do trigo-mourisco (2301, 3144 e 4471 kg MS/ha) foi superior à do milheto (437, 592 e 2224 kg MS/ha) nas três idades de corte. Os teores de PB do milheto foram, em média, 22,3% nas três idades de corte, mas no trigo-mourisco reduziu de 23,8% no primeiro corte, para 14% nos demais cortes. O trigo mourisco apresentou maior teor de FDN no primeiro corte 57,6% contra 52,1% do milheto, entretanto diminuiu no segundo e terceiro cortes, 46,8% e 41,2%, respectivamente, enquanto que no milheto mantiveram-se em 55,1% e 52,7% no segundo e terceiro corte. Os teores de FDA forma mais elevados para o trigo mourisco em comparação com o milheto nas três idades de corte estudadas. A produção de gases foi maior para o trigo mourisco em comparação ao milheto, com exceção do primeiro corte, quando o trigo apresentou maiores taxas de fermentação. O trigo mourisco apresentou qualidade como forrageira, sendo mais produtivo e nutritivo do que o milheto durante o período seco, sob irrigação.

SUMMARY This trial compared forage production and quality of Pearl Millet and Buckwheat in three cutting age on irrigation conditions during dry season, evaluated in a randomized block split plot design with tree replications. Was determined content of Dry Matter (DM), Crude Protein (CP), Neutral Detergent Fiber (NDF), Acid Detergent Fiber (ADF), Mineral Matter (MM) and in vitro gas production. Forage production of Buckwheat was higher than Pearl Millet in all the cutting ages. Pearl Millet CP content was similar to all three cuts but Buckwheat CP content was higher in the first cut and reduced in the others. The NDF content of Buckwheat was higher in the first cut and reduced in the others, meanwhile Pearl Millet presented low NDF value in the first cut but increased in the second and third cut. The Buckwheat presented high ADF content than Pearl Millet in all the cuts. In vitro gas production from Pearl Millet was higher in the first cut but more high for Buckwheat in the second and third cuts. Buckwheat produced more forage with high quality than Pearl Millet during dry season.


RESUMO Objetivou-se com este trabalho comparar o valor forrageiro do milheto e trigo-mourisco, sob irrigação, em diferentes idades de corte. Utilizou-se delineamento experimental em blocos ao acaso, com os tratamentos arranjados em modelo de parcelas subdivididas. As plantas foram cultivadas durante a estação seca, sob irrigação e colhidas em três idades de corte, 47, 57 e 67 dias (tratamentos). Foram determinados os teores de matéria seca (MS), proteína bruta (PB), fibra em detergente neutro (FDN), fibra em detergente ácido (FDA), matéria mineral e produção de gases in vitro e a produção de matéria seca. A produção do trigo-mourisco (2301, 3144 e 4471 kg MS/ha) foi superior à do milheto (437, 592 e 2224 kg MS/ha) nas três idades de corte. Os teores de PB do milheto foram, em média, 22,3% nas três idades de corte, mas no trigo-mourisco reduziu de 23,8% no primeiro corte, para 14% nos demais cortes. O trigo mourisco apresentou maior teor de FDN no primeiro corte 57,6% contra 52,1% do milheto, entretanto diminuiu no segundo e terceiro cortes, 46,8% e 41,2%, respectivamente, enquanto que no milheto mantiveram-se em 55,1% e 52,7% no segundo e terceiro corte. Os teores de FDA forma mais elevados para o trigo mourisco em comparação com o milheto nas três idades de corte estudadas. A produção de gases foi maior para o trigo mourisco em comparação ao milheto, com exceção do primeiro corte, quando o trigo apresentou maiores taxas de fermentação. O trigo mourisco apresentou qualidade como forrageira, sendo mais produtivo e nutritivo do que o milheto durante o período seco, sob irrigação.

BACKGROUND: Effect of chlorocholine chloride (CCC) on phenolic acids composition and polyphenols accumulation in various anatomical parts (stems, leaves and inflorescences) of common buckwheat (Fagopyrum esculentum Moench) in the early stages of vegetation period were surveyed. RESULTS: Treatment of buckwheat seeds with 2% of CCC has been increased content of total phenolics in the stems, leaves and inflorescences. On analyzing the different parts of buckwheat plants, 9 different phenolic acids - vanilic acid, ferulic acid, trans-ferulic acid, chlorogenic acid, salycilic acid, cinamic acid, p-coumaric acid, p-anisic acid, methoxycinamic acid and catechins were identified. The levels of identified phenolic acids varied not only significantly among the plant organs but also between early stages of vegetation period. Same changes as in contents of chlorogenic acid, ferulic acid, trans-ferulic acid were found for content of salycilic acid. The content of these phenolic acids has been significant increased under effect of 2% CCC treatment at the phase I (formation of buds) in the stems and at the phase II (beginning of flowering) in the leaves and then inflorescences respectively. The content of catechins as potential buckwheat antioxidants has been increased at the early stages of vegetation period after treatment with 2% CCC. CONCLUSIONS: The obtained results suggest that influence of CCC on the phenolics composition can be a result of various mechanisms of CCC uptake, transforming and/or its translocation in the buckwheat seedlings.
