Biopolymers such as carboxymethyl cellulose and hyaluronic acid are alternative substrates for conformable organic light-emitting diodes (OLEDs). However, drawbacks such as mechanical stress susceptibility can hinder the device's performance under stretched conditions. To overcome these limitations, herein, we developed a nanocomposite based on CMC/HA (carboxymethyl cellulose/hyaluronic acid) and synthetic Laponite, intending to improve the mechanical strength without compromising the film flexibility and transparency (transmittance >80%; 380-700 nm) as substrates for conformable OLEDs. From XRD, FTIR, CP-MAS NMR, and TGA/DTG characterization techniques, it was possible to conclude the presence of Laponite randomly dispersed between the polymer chains. CMC/HA with 5% (w/w) Laponite, CMC/HA 5, presented a higher tensile strength (370.6 MPa) and comparable Young's modulus (51.0 ± 1.2 MPa) in comparison to the nanocomposites and pristine films, indicating a better candidate for the device's substrates. To produce the OLED, the multilayer structure ITO/MoO3/NPB/TCTA:Ir(ppy)3/TPBi:Ir(ppy)3/BPhen/LiF was deposited onto the CMC/HA 5 substrate. The OLEDs fabricated using CMC/HA 5 substrates showed higher luminance (12 kcd/m2) and irradiance (0.9 mW/cm2) values when compared with those based on commercial bacterial cellulose. However, the same device presented a lower efficiency (3.2 cd/A) due to a higher current density. Moreover, the OLED fabricated onto the Laponite-modified biopolymer presented reproducible behavior when submitted to continuous bending stress. Thus, CMC/HA 5 demonstrates potential as a transparent conductor substrate for biopolymer-based OLEDs with comparable performance to commercial bacterial cellulose features.

Introduction: Cortical reaction is a secretory process that occurs after a spermatozoon fuses with the oocyte, avoiding the fusion of additional sperm. During this exocytic event, the cortical granule membrane fuses with the oocyte plasma membrane. We have identified several molecular components involved in this process and confirmed that SNARE proteins regulate membrane fusion during cortical reaction in mouse oocytes. In those studies, we microinjected different nonpermeable reagents to demonstrate the participation of a specific protein in the cortical reaction. However, the microinjection technique has several limitations. In this work, we aimed to assess the potential of cell-penetrating peptides (CPP) as biotechnological tools for delivering molecules into oocytes, and to evaluate the functionality of the permeable tetanus toxin (bound to CPP sequence) during cortical reaction. Methods: Arginine-rich cell-penetrating peptides have demonstrated the optimal internalization of small molecules in mammalian cells. Two arginine-rich CPP were used in the present study. One, labeled with 5-carboxyfluorescein, to characterize the factors that can modulate its internalization, and the other, the permeable light chain of tetanus toxin, that cleaves the SNAREs VAMP1 and VAMP3 expressed in mouse oocytes. Results: Results showed that fluorescent CPP was internalized into the oocyte cytoplasm and that internalization was dependent on the concentration, time, temperature, and maturation stage of the oocyte. Using our functional assay to study cortical reaction, the light chain of tetanus toxin bound to arginine-rich cell-penetrating peptide inhibited cortical granules exocytosis. Discussion: Results obtained from the use of permeable peptides demonstrate that this CPP is a promising biotechnological tool to study functional macromolecules in mouse oocytes.

Fluoroquinolones are important antimicrobial agents for the treatment of Pseudomonas infections. A total of 11 isolates of P. aeruginosa were collected from different clinical samples from different medical centers in the North West Bank-Palestine during 2017. In this study, resistance to fluoroquinolones and secretions of β-lactamases were detected by phenotypic methods, while presence of β-lactamase gene sequences and other virulence factors were detected by PCR technique. PCR product for gyrA, parC and parE genes were sequenced for further analyses. The phylogenetic analyses, population diversity indices and haplotypes determination were conducted using computer programs MEGA version 6, DnaSP 5.1001 and median-joining algorithm in the program Network 5, respectively. Results of this study showed that the MIC for ciprofloxacin and norfloxacin had a range of 32-256 µg/ml. In addition, all isolates carried either exoT or exoT and exoY genes, different β-lactamase genes and 82% of these isolates harbored class 1 integrons. Analyses of the gyrA, parC and parE sequences were found to be polymorphic, had high haplotype diversity (0.945-0.982), low nucleotide diversity (0.01225-0.02001) and number of haplotypes were 9 for each gyrA and parE genes and 10 haplotypes for parC gene. The founder haplotypes being Hap-1 (18%), Hap-2 (27.3%) and Hap-6 (9.1%) for gyrA, parC and parE genes, respectively. Two of ParE haplotypes were detected as indel haplotypes. The Median-joining- (MJ) networks constructed from haplotypes of these genes showed a star-like expansion. The neutrality tests (Tajima’s D test and Fu’s Fs test) for these genes showed negative values. Palestinian fluoroquinolone resistant P. aeruginosa strains showed high MIC level for fluoroquinolones, β-lactamase producers, carried type III secretion exotoxin-encoding genes, most of them [...].

Neste estudo, resistência a fluoroquinolonas e secreções de β-lactamases foram detectadas por métodos fenotípicos, enquanto a presença de sequências do gene β-lactamase e outros fatores de virulência foram detectados pela técnica de PCR (Proteína C-reativa). As análises moleculares mostraram que os haplótipos de P. aeruginosa resistentes à fluoroquinolona palestina não são geneticamente diferenciados; no entanto, mais mutações podem existir nessas cepas.

Helicobacter pylori is a spiral-shaped bacterium, which plays a role in the aetiology of gastric diseases in humans. Non-H. pylori Helicobacter (NHPH) species naturally colonise the stomach of animals and also induce gastric lesions in humans, highlighting their zoonotic importance. We evaluated the gastric bacterial colonisation density and gastric lesions and sought to identify the main phylogenetic groups of the Helicobacter spp. obtained from dogs in the central region of Rio Grande do Sul, Brazil, with this study aiming to investigate the occurrence of Helicobacter spp. in saliva and gastric samples from these dogs. This study included 35 dogs and used analysis such as cytology, histopathology, PCR, rapid urease testing, and phylogenetic analysis. Of the dogs, 94.3% were positive for Helicobacter spp., and these bacteria were present in the stomach of 32 dogs and saliva of eight. Respectively, eight, 15, and nine dogs had mild, moderate, and severe colonisation. Lymphocytic-plasmacytic infiltrate was the main gastric lesion. However, the presence of Helicobacter and the density appeared to be unrelated to the gastric lesions. The samples possessed a high nucleotide identity with remarkably similar sequences among some of the species of NHPH such as H. heilmannii s.s., H. salomonis, H. felis, and H. bizzozeronii. The saliva of domestic dogs, even of those who appear clinically healthy, can cause Helicobacter infection in humans and other animals, with, in these dogs, increased density, occurrence rate, and predominance of NHPH of zoonotic importance being found in the stomach with a lower occurrence of Helicobacter spp. in the saliva.

CONTEXT: Follicle-stimulating hormone (FSH) plays an essential role in gonadal function. Loss-of-function mutations in the follicle-stimulating hormone receptor (FSHR) are an infrequent cause of primary ovarian failure. OBJECTIVE: To analyze the molecular physiopathogenesis of a novel mutation in the FSHR identified in a woman with primary ovarian failure, employing in vitro and in silico approaches, and to compare the features of this dysfunctional receptor with those shown by the trafficking-defective D408Y FSHR mutant. METHODS: Sanger sequencing of the FSHR cDNA was applied to identify the novel mutation. FSH-stimulated cyclic adenosine monophosphate (cAMP) production, ERK1/2 phosphorylation, and desensitization were tested in HEK293 cells. Receptor expression was analyzed by immunoblotting, receptor-binding assays, and flow cytometry. Molecular dynamics simulations were performed to determine the in silico behavior of the mutant FSHRs. RESULTS: A novel missense mutation (I423T) in the second transmembrane domain of the FSHR was identified in a woman with normal pubertal development but primary amenorrhea. The I423T mutation slightly impaired plasma membrane expression of the mature form of the receptor and severely impacted on cAMP/protein kinase A signaling but much less on ß-arrestin-dependent ERK1/2 phosphorylation. Meanwhile, the D408Y mutation severely affected membrane expression, with most of the FSH receptor located intracellularly, and both signal readouts tested. Molecular dynamics simulations revealed important functional disruptions in both mutant FSHRs, mainly the loss of interhelical connectivity in the D408Y FSHR. CONCLUSIONS: Concurrently, these data indicate that conformational differences during the inactive and active states account for the distinct expression levels, differential signaling, and phenotypic expression of the I423T and D408Y mutant FSHRs.

Abstract The haemostatic efficacy of different extract types of Satureja thymbra L., Thymbra spicata L. (Lamiaceae) and Verbascum fruticulosum Post. (Scrophulariaceae) was evaluated in this study via the Prothrombin time (PT) and Activated partial thromboplastin time (aPTT) analysis. Aqueous, methanol and ethanol extracts of the examined plant species leaves were prepared to a final concentration 50 mg/mL. In vitro PT and aPTT assays were conducted on normal platelet poor plasma blood samples by a digital coagulation analyzer. The obtained results revealed anticoagulation activity of all investigated plant species with observed variations among them. The aqueous and ethanol extracts of T. spicata as well as the aqueous extract of S. thymbra prolonged PT values significantly (p < 0.05). While, all V. fruticulosum extract types have had no significant effect on the PT values. The recorded aPTT data showed that all aqueous extracts have had a significant effect on the blood haemostasis as they increased aPTT values in all plant species under study. Out of which, both the ethanol and methanol extracts of T. spicata and methanol extract of S. thymbra showed similar effect. Of great concern, it was clearly noticed that the aqueous and ethanol extract of T. spicata and the aqueous extract of S. thymbra possess the strongest anticoagulation effect as they increased both PT and aPTT values significantly relative to the control (p < 0.05). The variable anticoagulation bioactivity among the studied plant species could be referred to the various solvents degrees of solubility of different phyto-constituents. Thus, the efficacy of the plant species extracts evaluation as anticoagulants or coagulants were related to the plant species and to the solvent of extraction.

Introducción: Para Costa Rica, los estudios realizados en el Pacífico Tropical Este o PTE son importantes ya que se relacionan con el Corredor Marino del Pacífico Este Tropical, el cual es un esfuerzo de conservación multinacional. El presente estudio describe los parámetros físicos de temperatura del mar y salinidad del océano de las masas de agua que rodean a la Isla del Coco y su entorno, el PTE, a través del análisis de perfiles de CTD obtenidos durante campañas hidrográficas realizadas entre el año 2008 y el 2012; y comparar los resultados de los datos in-situ con los del reanálisis del modelo HYCOM-NCODA. Métodos: Se compilaron los resultados de 8 diferentes campañas científicas a la Isla del Coco de abril de 2008, marzo de 2009, abril de 2010, julio de 2011 y marzo de 2012. También se analizan las campañas de octubre de 2010 y marzo de 2011 en donde se realizó un transecto latitudinal y un transecto longitudinal en las aguas del Pacífico Tropical Este de Costa Rica. En julio de 2012 se realizaron 3 transectos latitudinales en las aguas en torno a la Isla del Coco. Las mediciones se hicieron por medio de un CTD, instrumento utilizado para determinar la conductividad, temperatura y presión de la columna de agua, entre otras variables. Resultados: Se observó la presencia de Agua Superficial Tropical en los primeros 50m de profundidad, con temperaturas superiores a 25°C y salinidades menores 33psu y Agua Sub-superficial Sub-tropical bajo los 60m con temperaturas menores a 25°C y salinidades alrededor de las 35ups. La zona de transición entre ellas se presentó entre los 50-60m. Los resultados de la temperatura del mar y la salinidad de los transectos analizados reflejan las condiciones climáticas y atmosféricas observadas, en donde se nota que las variaciones estacionales en el PTE están fuertemente influenciadas por la migración meridional de la Zona de Convergencia Inter-Tropical. Conclusiones: El estudio mostró que los datos del reanálisis del modelo de circulación general HYCOM-NCODA son comparables con los resultados de los perfiles del CTD en el PTE. La distribución de la temperatura de los muestreos tiende a ser similar a los datos del modelo oceánico y las principales diferencias se dan en la capa profunda para la salinidad, por lo que el modelo HYCOM es capaz de reproducir las características observadas en el PTE de temperatura y salinidad y puede ser usado para estudiar la dinámica tropical oceánica, en regiones más extensas y en otras épocas del año.


Introduction: In Costa Rica, studies done in the Eastern Tropical Pacific or ETP are important because of their interaction/relationship with the Eastern Tropical Pacific Seascape, a known multinational conservation initiative. The present study describes the physical parameters of sea temperature and salinity around Cocos Island and its environment, the ETP, using CTD profiles. Methods: Data were obtained in April 2008, March 2009, April 2010, July 2011 and March 2012 near this island. Latitudinal and longitudinal transects were also done during October 2010 and March 2011 in a broader Costa Rican ETP region. In July 2012, three latitudinal transects were done around Cocos Island. Records were obtained using a CTD, measuring conductivity, temperature and pressure, among other variables. Results: Tropical Surface Water was observed in the upper 50m, showing temperatures above 25°C and salinities below 33psu. Below the surface waters, Subsurface Subtropical Water was detected below a 60m depth, showing temperatures colder than 25°C and salinities around 35psu. The transition between these waters was around 50-60m in depth. Temperature and salinity results were related with the atmosphere conditions observed, in which ETP variability was strongly influenced by the Inter Tropical Convergence Zone migration. Conclusions: This study also showed that reanalysis data from the HYCOM-NCODA general circulation model are comparable and in a good agreement with the CTD profiles. Temperature and salinity HYCOM data also reproduced, in general terms, the main characteristics of CTD data, having some small differences in the deeper levels. This reanalysis data set could also be used to study the tropical ocean dynamics, in wider ETP regions and for different seasons.

RESUMO

Excellent quadratic non-linear optical (ONL-2) properties of the poly(2,5-bis(but-2-ynyloxy) benzoate, containing a polar diacetylene as a chromophore, were found. According with the Maker fringes method, oriented polymer films showing an order parameter of ∼0.23 can display outstanding and stable Second Harmonic Generation (SHG) effects under off-resonant conditions (SHG-532 nm). Also, the macroscopic non-linear optical (NLO)-coefficients were evaluated under the rod-like molecular approximation, obtaining: χzzz(2) and χzxx(2) in the order of 280 ± 10 and 100 ± 10 pm V-1, respectively. The mechanical and chemical properties, in addition to the large ONL-2 coefficients exhibited by this polymer, make it a promising organic material in the development of optoelectronic/photonic devices.

De grande preocupação, notou-se claramente que os extratos aquoso e etanólico de T. spicata e o extrato aquoso de S. thymbra apresentam efeito anticoagulante mais forte, aumentando os valores de TP e TTPa significativamente em relação ao controle (p 0,05). A variável bioatividade anticoagulante dentre as espécies vegetais estudadas pôde ser referida aos vários graus de solventes de solubilidade de diferentes fitoconstituintes. Assim, a eficácia da avaliação de extratos de espécies vegetais como anticoagulantes ou coagulantes foi relacionada às espécies vegetais e ao solvente de extração.

RESUMO

La pudrición carbonosa, causada por Macrophomina phaseolina, es una enfermedad importante de la caña de azúcar en México. Este estudio se realizó con el objetivo de caracterizar aislados de M. phaseolina obtenidos de caña de azúcar mediante análisis morfológicos y moleculares. La caracterización morfológica de 10 aislados se llevó a cabo con el uso de microscopia electrónica de barrido y microscopia de luz. Para confirmar la identificación, se extrajo el ADNr de 2 aislados representativos, y la región del espaciador interno transcrito (ITS) se amplificó mediante la reacción en cadena de la polimerasa y se secuenció usando los iniciadores específicos MpKF1 y MpKR1. Los aislados se identificaron como M. phaseolina con base en la morfología. El análisis de secuencias ITS mostró 100% de similitud con las secuencias de M. phaseolina depositadas en el GenBank. Para nuestro conocimiento, este es el primer estudio del mundo enfocado a caracterizar aislados de M. phaseolina obtenidos de caña de azúcar

Resumen Se estudió la dinámica de las mareas y el oleaje frente al Humedal Nacional Térraba-Sierpe (HNTS) y su relación con los procesos de erosión y la muerte del manglar en esta región. Se encuentra que un aumento relativo del nivel del mar, ya sea generado por el basculamiento de las costas, o por calentamiento global, es el responsable de crear una nueva plataforma para el oleaje y está modificando el perfil de la costa y cambiando el tipo de sedimento sobre el cual crece el manglar. Una mayor deposición de sedimentos sobre la región norte de este delta, mantiene el equilibrio de este sistema en esta zona, donde más bien hay una ganancia de sedimentos en la isla al frente de la desembocadura del Río Grande de Térraba. Un menor aporte de sedimentos del Rio Sierpe está generando una migración tierra adentro de este sistema estuarino frente a esta región, lo cual es un síntoma de un aumento relativo del nivel del mar. Los escenarios futuros sobre una mayor energía del oleaje, y de un continuo aumento del nivel del mar, no permitirán un equilibrio en la dinámica de los sedimentos, especialmente sobre la parte sur de este delta, por lo que continuará perdiendo la cobertura del manglar en este sistema estuarino.


Abstract The Térraba-Sierpe Reserve has the largest mangrove in Costa Rica but has suffered from changes in sea level. I used published data to analyze the area and found that sea level rise, generated by coast subsidence or global warming, created a new platform for waves and changed the beach profile and sediment type. Increased sediment deposition on the north maintains the balance of this system, with a sediment gain on the island in front of the Térraba river mouth. A lower sediment output from the Sierpe river is generating a landward delta migration, especially in the southern part, a symptom of local sea level rise. Future scenarios of greater wave energy and additional sea level rise will not allow a dynamic sediment balance, especially on the southern part of the delta, and will generate a loss mangrove cover. INTRODUCTION: Mosquito salivary proteins are able to induce an antibody response that reflects the level of human-vector contact. IgG antibodies against dengue virus (DENV-IgG) are indicators of previous exposure. The risk of DENV transmission is not only associated to mosquito or dengue factors, but also to socioeconomic factors that may play an important role in the disease epidemiology. OBJECTIVE: To determine the effect of the presence of Aedes aegypti mosquitos in different stages in households and the history of dengue exposure on vector-human contact determined by the level of anti-salivary protein antibodies in people living in a Colombian endemic area. MATERIALS AND METHODS: A pilot study of 58 households and 55 human subjects was conducted in Norte de Santander, Colombia. A questionnaire for socioeconomic factors was administered and houses were examined for the presence of Ae. aegypti specimens in the aquatic stages. The level of DENV-IgG antibodies (DENV-IgG), in addition to IgG and IgM anti- Ae. aegypti salivary gland extract (SGE) antibodies (SGE-IgG, SGE-IgM) were evaluated by ELISA using blood collected in filter paper. RESULTS: We found a significant higher level of SGE-IgG antibodies in subjects living in houses with Ae. aegypti in aquatic stages. We also found a higher concentration of SGE-IgG antibodies in people exposed to DENV, a positive correlation between IgM-SGE and IgG-DENV and a negative correlation with IgG-SGE. CONCLUSION: Anti-salivary proteins antibodies are consistent with the presence of Ae. aegypti aquatic stages inside houses and DENV-IgG antibodies concentrations.
