RESUMEN

Garlic oil (GO) and cinnamaldehyde (CIN) have shown potential to modify rumen fermentation. The aim of this study was to assess the effects of GO and CIN on rumen fermentation, microbial protein synthesis (MPS), and microbial populations in Rusitec fermenters fed a mixed diet (50:50 forage/concentrate), as well as whether these effects were maintained over time. Six fermenters were used in two 15-day incubation runs. Within each run, two fermenters received no additive, 180 mg/L of GO, or 180 mg/L of CIN. Rumen fermentation parameters were assessed in two periods (P1 and P2), and microbial populations were studied after each of these periods. Garlic oil reduced the acetate/propionate ratio and methane production (p < 0.001) in P1 and P2 and decreased protozoal DNA concentration and the relative abundance of fungi and archaea after P1 (p < 0.05). Cinnamaldehyde increased bacterial diversity (p < 0.01) and modified the structure of bacterial communities after P1, decreased bacterial DNA concentration after P2 (p < 0.05), and increased MPS (p < 0.001). The results of this study indicate that 180 mg/L of GO and CIN promoted a more efficient rumen fermentation and increased the protein supply to the animal, respectively, although an apparent adaptive response of microbial populations to GO was observed.

The objective of this study was to assess the impact of increasing levels of heat-treated soybean in the diet of crossbred cattle during the finishing phase on nutrient intake and digestibility, ruminal parameters, digesta passage rate, nitrogen balance, and microbial protein synthesis. Five steers, crossbred 7/8 Jersey x Zebu, fitted with rumen cannulas and with an average weight of 350 ± 50 kg, were utilized. The experimental treatments consisted of 0, 7, 14, 21, and 28% inclusion of heat-treated soybean in the total diet dry matter. The animals were randomly allocated in a 5 × 5 Latin square design. Evaluation of the animals took place over five experimental periods, each lasting 20 days. During each experimental period, the first 15 days were allocated for animal adaptation to the experimental diets, followed by five days of data collection. No significant differences were observed among the diets in terms of dry matter intake (average of 6.57 kg day-1; P = 0.615) and organic matter intake (average of 6.23 kg day-1; P = 0.832). However, heat-treated soybean had a significant impact on the digestibility of dry matter (P = 0.02), organic matter (P = 0.01), crude protein (P < 0.01), and neutral detergent fiber (P < 0.01). There was no observed change on microbial protein synthesis (average of 409.6 g day-1) in animals with the inclusion of heat-treated soybean in the diets. With each 1% inclusion of heat-treated soybean in the cattle diet, there was an increase of 0.00754 units in ruminal pH values and a reduction of 0.75839 mg dL-1 in ruminal ammoniacal nitrogen values. This study suggests that heat-treated soybean can be included in up to 15% of the dry matter in diets for finishing feedlot cattle.

Rumen microbial protein synthesis (MPS) provides at least half of the amino acids for the synthesis of milk and meat protein in ruminants. As such, it is fundamental to global food protein security. Estimating microbial protein is central to diet formulation, maximising nitrogen (N)-use efficiency and reducing N losses to the environment. Whilst factors influencing MPS are well established in vitro, techniques for in vivo estimates, including older techniques with cannulated animals and the more recent technique based on urinary purine derivative (UPD) excretion, are subject to large experimental errors. Consequently, models of MPS used in protein rationing are imprecise, resulting in wasted feed protein and unnecessary N losses to the environment. Newer 'omics' techniques are used to characterise microbial communities, their genes and resultant proteins and metabolites. An analysis of microbial communities and genes has recently been used successfully to model complex rumen-related traits, including feed conversion efficiency and methane emissions. Since microbial proteins are more directly related to microbial genes, we expect a strong relationship between rumen metataxonomics/metagenomics and MPS. The main aims of this review are to gauge the understanding of factors affecting MPS, including the use of the UPD technique, and explore whether omics-focused studies could improve the predictability of MPS, with a focus on beef cattle.

Objective: Estimating microbial protein synthesis (MPS) in the rumen of growing lambs based on the urinary excretion of purine derivatives (PDs) and forage to concentrate (F/C) ratio. Materials and Methods: 36 similar-growing male lambs (weight 32.53 ± 1.90 kg; age 93 ± 6.63 days) were used in a completely randomized design with four groups: a) the 20-80 F/C ratio (dry hay 10% + wheat straw 10%), b) the 20-80 F/C ratio (dry hay 0% + wheat straw 20%), c) the 10-90 F/C ratio (dry hay 5% + wheat straw 5%), d) and the 10-90 F/C ratio (dry hay 0% + wheat straw 10%) with nine replicates. Results: Total PD and rumen MPS synthesized increased (10.98 vs. 13.25 mmol/day and 59.45 vs. 71.80 gm/day) in group d compared to group a. Dry organic matter intake (0.869 kg/day), fermentable dry organic matter (0.563 kg/day), and microbial nitrogen (N) yield (11.48 gm/day) of group d were at the maximum, but in terms of gN/kg dry organic matter (22.37 gm/kg), the mean of group c was higher than others. Conclusion: Increasing the level of food concentrate and the gradual removal of alfalfa from the diet increased the excretion of PD and MPS in the rumen. It was also found that urinary PD monitoring is an accurate indicator for the estimation of MPS.

Despite the increasing importance of goat production in response to high demand for their products and their relative robustness to environmental stressors, and in contrast to other ruminant species, little data is available on how tannin extract feeding affects their feed intake, nutrient digestion and nitrogen (N) metabolism. Therefore, a trial in Oman investigated the respective variables by using a commercial tannin feed additive. In a 4 (treatments) x 3 (periods) x 2 (animals) Youden square, two weaned Batinah bucks each were fed a high or low protein diet of Rhodes grass hay and crushed barley grain, with or without the addition of a chestnut and quebracho tannin extract at 2 g/kg metabolic weight. Feed offered, feed refused and faeces and urine excreted were quantified to determine diet digestibility, total N excretion, N retention and rumen microbial protein synthesis (MPS). Due to their young age and low live weight, feed intake of goats was relatively low. Crude protein level and tannin addition had no statistically significant effect on dry matter (DM) and N intake, DM digestibility, N excretion in faeces and urine, as well as MPS. In consequence, no benefit of tannin feeding could be confirmed for the goats' N retention, irrespective of diet composition. These results indicate, on one hand, an effective neutralisation of the tested tannin extract along the gastrointestinal tract of goats, but on the other hand, that stimulation of MPS or N retention by tannins cannot be evidenced when diet components are present that simultaneously release energy and protein, as is the case with barley.

The review focused on (I) impact of the amides I and II, and the alpha-helix and beta-sheet-structure ratios in food and feeds; (II) Current research progress and update in synchrotron technique and application in feed and food molecular structure studies that are associated with nutrition delivery; (III) Impact of thermal processing- steam pressure toasting condition on feed and food; (IV). Impact of the microbial protein synthesis and methodology on feed and food; and (V). Impact on performance and production of ruminants with Faba beans.

Branched-chain amino acids are deaminated by amylolytic bacteria to branched-chain volatile fatty acids (BCVFA), which are growth factors for cellulolytic bacteria. Our objective was to determine the dietary conditions that would increase the uptake of BCVFA by rumen bacteria. We hypothesized that increased forage would increase cellulolytic bacterial abundance and incorporation of BCVFA into their structure. Supplemental polyunsaturated fatty acids, supplied via corn oil (CO), should inhibit cellulolytic bacteria growth, but we hypothesized that additional BCVFA would alleviate that inhibition. Further, supplemental BCVFA should increase neutral detergent fiber degradation and efficiency of bacterial protein synthesis more with the high forage and low polyunsaturated fatty acid dietary combination. The study was an incomplete block design with 8 dual-flow continuous cultures used in 4 periods with 8 treatments (n = 4 per treatment) arranged as a 2 × 2 × 2 factorial. The factors were: high forage (HF) or low forage (LF; 67 or 33%), without or with supplemental CO (3% dry matter), and without or with 2.15 mmol/d (which included 5 mg/d of 13C each of BCVFA isovalerate, isobutyrate, and 2-methylbutyrate). The isonitrogenous diets consisted of 33:67 alfalfa:orchardgrass pellet, and was replaced with a concentrate pellet that mainly consisted of ground corn, soybean meal, and soybean hulls for the LF diet. The main effect of supplementing BCVFA increased neutral detergent fiber (NDF) degradability by 7.6%, and CO increased NDF degradability only in LF diets. Supplemental BCVFA increased bacterial N by 1.5 g/kg organic matter truly degraded (6.6%) and 0.05 g/g truly degraded N (6.5%). The relative sequence abundance decreased with LF for Fibrobacter succinogenes, Ruminococcus flavefaciens, and genus Butyrivibrio compared with HF. Recovery of the total 13C dose in bacterial pellets decreased from 144 µg/ mg with HF to 98.9 µg/ mg with LF. Although isotope recovery in bacteria was greater with HF, BCVFA supplementation increased NDF degradability and efficiency of microbial protein synthesis under all dietary conditions. Therefore, supplemental BCVFA has potential to improve feed efficiency in dairy cows even with dietary conditions that might otherwise inhibit cellulolytic bacteria.

Nutrient synchronisation of protein and carbohydrates is a promising practice to improve ruminal nutrient utilisation. However, dietary sources supplying these nutrients can vary in ruminal nutrient availability due to differing degradation rates, therefore potentially affecting utilisation of nitrogen (N). The effects of the addition of non-fibre carbohydrates (NFCs) with different rumen degradation rates in high-forage diets on ruminal fermentation, efficiency and microbial flow were investigated in vitro using the Rumen Simulation Technique (RUSITEC). Four diets were tested: control with 100% ryegrass silage (GRS) and substitution of 20% on a DM basis of ryegrass silage with corn grain (CORN), processed corn (OZ) or sucrose (SUC). The four diets were assigned to 16 vessels in two sets of RUSITEC apparatuses in a randomised block design over a 17 d experimental trial; 10 d consisted of adaptation and 7 d for sample collection. Rumen fluid was collected from four rumen-cannulated dry Holstein-Friesian dairy cows and was treated without mixing. Then, rumen fluid from each cow was used to inoculate four vessels, and diet treatments were randomly allocated to each one. This was repeated for all cows resulting in 16 vessels. The inclusion of SUC in ryegrass silage diets improved DM and organic matter digestibility. The only diet to significantly lower ammonia-N concentration compared with GRS was SUC. The outflows of non-ammonia-N, microbial-N, and efficiency of microbial protein synthesis were not affected by diet type. However, the efficiency of nitrogen utilisation was improved by SUC compared with GRS. This indicates that the inclusion of an energy source with a high rumen degradation rate in high-forage diets improves rumen fermentation, digestibility, and N utilisation. Specifically, this effect was observed for the more readily available energy source, SUC, compared with the more slowly degradable NFC sources, CORN and OZ.

The objective of this study was to investigate the effects of dietary rumen-degradable starch (RDS, g/kg of DM) to rumen-degradable protein (RDP, g/kg of DM) ratios (SPR) on in vitro rumen fermentation characteristics and microbial protein synthesis (MCPS). Treatments were eight diets with SPR of 1.9, 2.0, 2.1, 2.2, 2.3, 2.4, 2.5 and 2.6 and were formulated to be isoenergetic, isonitrogenous, and isostarch. Substrates were anaerobically incubated in sealed culture vials (100 mL) for 6, 24 or 48 h. Three incubation runs were conducted within two consecutive weeks. With the increase of the dietary SPR, the gas production (GP), in vitro dry matter disappearance (IVDMD) and concentration of MCPS and total volatile fatty acids (TVFA) linearly increased after 6 h of incubation (p ≤ 0.01), whereas they quadratically increased and peaked at the SPR of 2.3 after 24 and 48 h of incubation (p < 0.05). In response to dietary SPR increasing, the in vitro neutral detergent fiber disappearance (IVNDFD) quadratically increased (p < 0.01), and the ammonia nitrogen (NH3-N) concentration linearly decreased (p < 0.01) after 6, 24 and 48 h of incubation. Based on the presented results, an SPR of 2.3 is recommended for formulating a diet due to its greatest IVDMD, IVNDFD, GP, TVFA and MCPS. However, as the results obtained are strictly dependent on the in vitro conditions, further in vivo studies are needed to verify our findings.

The objective of this study was to investigate the effects of heat treatment (toasting) and particle size alterations (grinding; rolling) on nutrient utilization, ruminal fermentation, and supply of metabolizable protein (MP), and to study the interaction between processing conditions of fava beans and forage type. Six Danish Holstein dairy cows fitted with ruminal, duodenal, and ileal cannulas were used in a 6 × 4 incomplete Latin square design with 4 periods of 21-d duration. Cows were fed ad libitum with 6 experimental diets: diets high in either grass-clover silage or corn silage were combined with ground untoasted, ground toasted, or rolled untoasted fava beans. Samples of ruminal fluid, digesta from duodenum and ileum, and feces were collected, and nutrient digestibility was estimated using Cr2O3 and TiO2 as flow markers. Diets high in corn silage resulted in higher ruminal pH and higher proportion of propionate in ruminal volatile fatty acids compared with diets high in grass-clover silage. Diets high in corn silage resulted in higher apparent total-tract digestibility of crude protein and starch but lower apparent ruminal and total-tract digestibility of neutral detergent fiber compared with diets high in grass-clover silage. Rolling of fava beans decreased the in situ small intestinal disappearance of rumen-undegradable protein corrected for particle losses. Compared with grinding, rolling of fava beans reduced apparent ruminal digestibility of starch, true ruminal digestibility of organic matter, crude protein, and AA, and small intestinal digestibility of AA and starch. Grinding of fava beans increased apparent ruminal digestibility of neutral detergent fiber and reduced the proportion of propionate in ruminal volatile fatty acids compared with rolling of fava beans. In addition, rolling of fava beans had no effect on MP supply. Toasting of fava beans had no effect on in vivo nutrient digestibility except for an interaction with forage source on apparent ruminal dry matter and organic matter digestibility. Toasting of fava beans did not affect small intestinal digestion of individual and total AA, and therefore failed to increase MP supply. In conclusion, neither replacing grass-clover silage with corn silage, nor toasting nor rolling of fava beans had an effect on supply of MP.

Objectives: The goal of this research was to figure out the effect of the local sorghum as silage on the performance of Bligon goats. Microbial protein synthesis, digestible nutrients, and average daily weight gain (ADWG) were measured to evaluate the goats' performance. Materials and Methods: The study was designed in a completely randomized design with a one-way pattern. Twelve female Bligon goats with 24.33 ± 2.83 kg (mean ± SEM) initial body weight were divided into three groups of total mixed ration (TMR) treatments. Group 1 received fresh Napier grass (FNG) as a control, group 2 received imported sorghum silage (ISS) of brown midrib resistance (BMR), and group 3 received local sorghum silage (LSS) of super-2. Analysis of variance was used to analyze the data on microbial protein synthesis and feed intake during the research. On the contrary, analysis of covariance was used to analyze ADWG with initial weight as a covariate. Results: Microbial protein synthesis, feed-intake, and ADWG of goats that received TMR based on silage of two varieties of sorghum, namely BMR (ISS) and super-2 (LSS), were lower (p < 0.05) than control. However, there was no significant difference between both TMR based on sorghum silages. ISS's feed conversion was better than LSS (p < 0.05), and FNG was the best. Sorghum silage as a basal ration in TMR had lower microbial protein synthesis but higher total digestible nutrient content than fresh forage, such as Napier grass. The sorghum varieties did not affect the microbial protein synthesis, digestible nutrients, and ADWG of Bligon goats. However, ISS treatment had higher feed efficiency than LSS. Conclusion: The local sorghum (super-2) silage can be used as ruminant feed as well as imported sorghum (BMR) offered as TMR. However, regardless of the cultivar, TMR based on sorghum silage cannot replace TMR based on fresh Napier grass.

The objective of the present study was to analyze the effects of yeast-fermented rubber seed kernels (YERSEK) on the feed intake, hematology, microbial protein synthesis, milk yield, and milk composition in dairy cows. Six crossbred Holstein Friesian (HF) × Thai lactating dairy cows with 110 ± 10 days in milk were randomly assigned to three different amounts of YERSEK at 0%, 10%, and 20% in a concentrate mixture using a 3 × 3 repeated Latin square design. Cows were fed with concentrate diets at a concentrate-to-milk yield ratio of 1:1.5, with rice straw fed ad libitum. The inclusion of YERSEK did not adversely affect feed intake, nutrient intake, or digestibility (p > 0.05), whereas ether extract intake and digestibility linearly increased in dairy cows receiving YERSEK (p < 0.01). Increasing YERSEK levels did not adversely affect blood urea nitrogen (BUN) levels, hematological parameters, or microbial protein synthesis (p > 0.05). Supplementation of YERSEK did not influence milk production, lactose, or protein levels (p > 0.05). However, milk fat and total solids decreased linearly (p < 0.05) with the addition of YERSEK. In conclusion, in a concentrate diet, YERSEK could be used as a protein source without negative effects on feed intake, digestibility, hematology, microbial protein synthesis, or milk yield. However, it reduced the milk fat and total solids of tropical lactating dairy cows.

The aim was to analyze the effects of two cereal grains differing in nutritional composition and starch degradation characteristics and the timing of their supplementation on feed intake, rumen microbial protein synthesis (MPS), performance, and nitrogen use of lactating dairy cows grazing an alfalfa-ryegrass sward. Four dietary treatments were tested in 24 lactating Brown Swiss cows in an incomplete 4 × 3 Latin square design. Cows were supplemented with 3.5 kg/d (as-fed basis) of a corn-based or an oat-based concentrate mixture (CM), of which either the majority (2.5 vs. 1.0 kg/d) was offered before or after grazing. Feed intake was similar across diets, but the interaction between type of CM and timing of supplementation affected eating time (p = 0.010), milk protein (p = 0.013) and energy-corrected milk yields (p = 0.025), efficiency of rumen MPS (p = 0.094), and nitrogen use efficiency (p = 0.081). Most of these variables were greater when the majority of the corn-based CM was offered after grazing and the oat-based CM before grazing. Supplementing slowly degradable starch sources after and rapidly degradable starch sources before grazing may improve the efficiency of rumen MPS, milk performance, and nitrogen use efficiency in dairy cows grazing alfalfa-ryegrass swards.

Background and Aim: Dairy ration formulations should consider the synchronization of the rumen degradable protein (RDP) to rumen undegradable protein (RUP) ratio (RDPR) with non-fiber carbohydrate (NFC) to achieve optimum microbial protein synthesis (MPS), reduce feed costs, and reduce N excretion to the environment. This study aimed to investigate the effect of RDPR and NFC synchronization on in vitro digestibility, fermentability, and MPS. Materials and Methods: The experiment used a 3×3 factorial randomized block design with four replications. The first factor was RDPR (RDPR1=50:50; RDPR2=55:45; RDPR3=60:40) and the second factor was NFC levels (NFC1=30%, NFC2=35%, NFC3=40%). The experimental diets were evaluated using a two-stage in vitro method. The examined parameters included rumen pH, NH3 concentration, total volatile fatty acid (VFA) concentration, the molar proportion of VFAs, rumen microbes (protozoa and total bacteria population), and MPS. Data were analyzed using ANOVA, followed by the Duncan test. Results: The results show that neither RDPR nor NFC affected rumen pH, NH3, total VFA, and the rumen microbe population. The interaction between RDPR and NFC affected the molar proportion of acetate, iso-butyrate, and n-valerate. The combination of RDPR1 and NFC1 produced a lower molar proportion of acetate (49.73%) than the other treatment combinations (>54%). The acetate to propionate ratio was influenced by the NFC levels, in which NFC2 and NFC3 produced the highest ratio (p<0.05). MPS was affected by RDPR and NFC, but not by their interaction. Treatments NFC2 and RDPR3 produced the highest MPS. NFC affected the dry matter and organic matter digestibility (DMD and OMD), with treatment NFC3 resulting in the highest DMD and OMD. Conclusion: The combination of a 60:40 RDPR with 35% NFC resulted in the best synchronization of protein and energy available for MPS and digestion activity in the rumen.

Shifting the cutting of grass from morning to afternoon has been shown to increase the concentration of nonstructural carbohydrates (NSC) in forages. We compared the effects of diets (66:34, forage:concentrate ratio) containing a mix (% of the diet dry matter) of baleages (46.5%) and silages (19.3%) harvested from timothy cut in the afternoon (p.m.-cut TIM diet) or morning (a.m.-cut TIM diet) on omasal flows of NSC and nitrogenous fractions, ruminal and total-tract digestibilities of nutrients, plasma concentration of AA, and milk yield and composition. Eight ruminally cannulated Holstein cows averaging (mean ± standard deviation) 31.4 ± 6.13 kg/d of milk, 136 ± 17.0 d in milk, and 611 ± 66.4 kg of body weight in the beginning of the experiment were used in a crossover design with 21-d periods (14 d for diet adaptation and 7 d for data and sample collection). Intake of total ethanol-soluble carbohydrates (TESC; +150 g/d), starch (+129 g/d), and total NSC (TESC plus starch = +278 g/d) was greater with feeding the p.m.- than the a.m.-cut TIM diet. Likewise, the apparent ruminal digestibilities of TESC (+149 g/d), starch (+167 g/d), and total NSC (+316 g/d) increased in the p.m.-cut TIM diet. Diets, however, had no effect on the omasal flows and apparent ruminal and total-tract digestibilities of dry matter and organic matter. Intake of N increased in cows fed the p.m.- versus the a.m.-cut TIM diet (562 and 528 g/d, respectively) despite no effect of diets on dry matter intake. Diets did not affect the omasal flows of total nonammonia N, total bacterial nonammonia N, nonammonia and nonbacterial N, and individual AA, and the efficiency of microbial protein synthesis in the rumen. Contrarily, supply of rumen-degradable protein increased (+9.2%) in cows fed the p.m.-cut TIM diet, with this response driven by the 6.4% increase in N intake. Plasma concentrations of essential and nonessential AA followed the omasal flow of AA and were not changed by diets. Feeding the p.m.- versus the a.m.-cut diet significantly increased yields of 4% fat-corrected milk and milk fat, and tended to increase energy-corrected milk, milk true protein, and milk lactose yields. Overall, feeding the p.m.-cut TIM diet to mid-lactation dairy cows did not improve microbial protein synthesis and omasal flow of AA, and these responses were in line with the lack of a treatment effect on dry matter intake.

BACKGROUND AND AIM: Feeding ruminants must notice the degradability of feed, especially protein. Microbial rumen requires ammonia from rumen degradable protein (RDP) beside that ruminant require bypass protein or rumen undegradable protein (RUP) and microbial crude protein. The aim of the study was to discover the best RDP:RUP ratio in beef cattle diets commonly used by Indonesian farmers using an in vitro methodology. MATERIALS AND METHODS: Samples of Pennisetum purpureum, Leucaena leucocephala, Indigofera zollingeriana, cassava, maize, palm kernel cake, rice bran, and tofu waste were formulated into dietary treatments (dry matter [DM] basis). All experiments were carried out using a 3×3×2 factorial, randomized block design with three replications. Treatments consisted of three protein levels (12%, 14%, and 16%), two energy levels (65% and 70%), and three RDP:RUP ratio levels (55:45, 60:40, and 65:35). The experimental diets were incubated in vitro using buffered rumen fluid for 48 h at 39°C. After incubation, the supernatants were analyzed to determine pH, ammonia concentration, total volatile fatty acid (VFA), and microbial protein synthesis. The residues were analyzed to determine DM, organic matter, protein, and RUP digestibility. RESULTS: Increased protein, energy, and RDP levels increased digestibility, ammonia concentrations, total VFAs, and microbial protein synthesis (p<0.05), while rations with 16% protein lowered these parameters (p<0.05). CONCLUSION: Increased dietary protein (from 12% to 14% DM), energy (from 65% to 70% DM), and RDP (from 55% to 65% crude protein [CP]) levels increased nutrient digestibility, ammonia concentration, total VFA levels, and microbial protein synthesis. The diet containing 14% DM dietary protein and 70% DM energy, which contained 55%, 60%, or 65% CP RDP optimally increased nutrient digestibility, ammonia concentration, total VFA levels, and microbial protein synthesis. Thus, feed based on these RDP:RUP ratios can optimize ruminant productivity.

The objective of this study was to evaluate the effect of rolled barley supplementation on microbial composition and omasal flows of bacterial, protozoal, and nonmicrobial AA in cows fed fresh perennial ryegrass (Lolium perenne L.; PRG). Ten ruminally cannulated multiparous Holstein cows averaging (mean ± standard deviation) 49 ± 23 d in milk and 513 ± 36 kg of body weight were assigned to 1 of 2 treatments in a switchback design. The treatment diets were PRG only or PRG plus 3.5 kg of dry matter rolled barley (G+RB). The study consisted of three 29-d periods where each period consisted of 21 d of diet adaptation and 8 d of data and sample collection. A double-marker system was used to quantify nutrient flow entering the omasal canal along with 15N-ammonium sulfate to label and measure the microbial and nonmicrobial omasal flow of AA. Overall, rolled barley supplementation had no effect on the AA composition of the omasal liquid-associated and particle-associated bacteria. Rolled barley supplementation affected the AA concentrations of omasal protozoa; however, the differences were nutritionally minor. Particle-associated bacteria AA flow was increased for all AA, except for Trp and Pro, in cows fed the G+RB diet. Rolled barley supplementation had no effect on protozoal AA flow. On average, protozoa accounted for 23% of the microbial essential AA flow, which ranged from 17 to 28% for Trp and Lys, respectively. The flow of all AA in omasal true digesta increased in cows fed the G+RB diet compared with the PRG-only diet, resulting in a 228 g/d increase in total AA flow in cows fed the G+RB diet. This increase in total AA flow in cows fed the G+RB diet was due to an increase in microbial AA flow. Rolled barley supplementation had no effect on nonmicrobial AA flow. The nonmicrobial AA flow modestly contributed to total AA flow, accounting for 15.6% on average. These results indicated that extensive ruminal degradation of PRG AA occurred (83.5%), and we demonstrated that cows consuming PRG-based diets exhibit a large dependence on microbial AA to support metabolizable AA supply. Rolled barley supplementation can increase the omasal flow of microbial AA in cows consuming PRG-based diets. However, further research is required to elucidate if this increased AA supply can support higher milk yield under such dietary conditions.

The aim was to study the effects of rumen N balance (RNB), dietary protein source, and their interaction on feed intake, N partitioning, and rumen microbial crude protein (MCP) synthesis in lactating dairy cows. Twenty-four lactating Holstein cows were included in a replicated 4 × 4 Latin square experimental design comprising four 20-d periods, each with 12 d of adaptation to the experimental diets and 8 d of sampling. The dietary treatments followed a 2 × 2 factorial arrangement (i.e., 4 treatments) with 2 main protein sources [faba bean grain (FB) and SoyPass (SP; Beweka Kraftfutterwerk GmbH, Heilbronn, Germany)] offered at 2 dietary RNB levels each [0 g/kg of dry matter, DM (RNB0) and -3.2 g/kg of DM (RNB-)]. The RNB was calculated as the difference between dietary crude protein (CP) intake and the rumen outflow of undegraded feed CP and MCP and divided by 6.25. Composition of concentrate mixtures was adjusted to create diets with desired RNB levels. Each of these protein sources supplied ≥35% of total dietary CP. Both diets for each protein source were isoenergetic but differed in CP concentrations. The DM intake (kg/d) was lower for RNB- than for RNB0 in diets containing FB, whereas no differences were seen between the RNB levels for SP diets. The RNB- decreased N intake and urinary N excretion but increased milk N use efficiency in both FB and SP diets, with greater differences between the RNB levels for FB diets than for SP diets. Similarly, duodenal MCP synthesis (g/kg of digestible organic matter intake) estimated from purine derivatives in the urine was lower for RNB- than for RNB0 in FB diets but similar between the RNB levels in diets containing SP. Low RNB of approximately -65 g/d (approximately -3.2 g/kg of DM) in diets reduced feed intake, N balance, and performance in high-yielding dairy cows with possibly more pronounced effects in diets containing rapidly degradable protein sources.
