RESUMEN

The association between hypertension and obesity-induced cardiac damage is usually accepted. However, no studies have been focused on cardiac alterations in obesity, independently of blood pressure increase. It is well known that Cardiac TRH induces Left Ventricular Hypertrophy (LVH) and fibrosis, and its inhibition prevents the development of hypertrophy. Also, it has been described that the adiponectin leptin induces TRH expression. Thus, we hypothesized that in obesity, the increase in TRH induced by hyperleptinemia is responsible for LVH, until now mostly attributed to pressure load. We studied obese Agouti mice suffering from hypertension with hyperleptinemia and found a significant LVH development with increased TRH gene expression. Consequently, we found higher fibrotic (collagens and TGF-ß) and hypertrophic markers (BNP and ß-MHC) expression vs lean black controls. As pressure could explain these results, we treated obese mice with diuretic (hydrochlorothiazide 20 mg/kg/day) since weaning. Diuretic treatment was successful as the diuretic group was normotensive in contrast to control obese mice. Nevertheless, both groups showed LVH development, higher cardiac precursor TRH gene and peptide expressions and elevated fibrotic and hypertrophic markers expression, pointing out that obesity-induced LVH is not due to hypertension. In addition, we performed Cardiac TRH inhibition by specific siRNA injection compared to control siRNA treatment and evaluated cardiac damage. As expected, expressions and protein increase in hypertrophic and fibrotic markers observed in the AG mouse with the native cTRH system were not seen in the AG mouse with the cTRH silencing. Indeed, the AG + TRH-siRNA group showed hypertrophic markers expression and fibrosis measurements similar to the lean BL mice. On the whole, these results point out that the novel Leptin-Cardiac TRH pathway is responsible for the cardiac alterations present in hyperleptinemic obesity, independent of blood pressure, and cTRH long-term silencing since early stages totally prevent LVH development and cardiac fibrosis.

The hypothalamic neuropeptides linked to appetite and satiety were investigated in obese mice treated with cotadutide (a dual receptor agonist of glucagon-like peptide 1 (GLP-1R)/Glucagon (GCGR)). Twelve-week-old male C57BL/6 mice were fed a control diet (C group, n = 20) or a high-fat diet (HF group, n = 20) for ten weeks. Each group was further divided, adding cotadutide treatment and forming groups C, CC, HF, and HFC for four additional weeks. The hypothalamic arcuate neurons were labeled by immunofluorescence, and protein expressions (Western blotting) for neuropeptide Y (NPY), proopiomelanocortin (POMC), agouti-related protein (AgRP), and cocaine- and amphetamine-regulated transcript (CART). Cotadutide enhanced POMC and CART neuropeptides and depressed NPY and AGRP neuropeptides. In addition, gene expressions (RT-qPCR) determined that Lepr (leptin receptor) and Calcr (calcitonin receptor) were diminished in HF compared to C but enhanced in CC compared to C and HFC compared to HF. Besides, Socs3 (suppressor of cytokine signaling 3) was decreased in HFC compared to HF, while Sst (somatostatin) was higher in HFC compared to HF; Tac1 (tachykinin 1) and Mc4r (melanocortin-4-receptor) were lower in HF compared to C but increased in HFC compared to HF. Also, Glp1r and Gcgr were higher in HFC compared to HF. In conclusion, the findings are compelling, demonstrating the effects of cotadutide on hypothalamic neuropeptides and hormone receptors of obese mice. Cotadutide modulates energy balance through the gut-brain axis and its associated signaling pathways. The study provides insights into the mechanisms underlying cotadutide's anti-obesity effects and its possible implications for obesity treatment.

Hypothalamic mTORC1 signaling is involved in nutrient sensing. Neurons that express the agouti-related protein (AgRP) are activated by food restriction and integrate interoceptive and exteroceptive signals to control food intake, energy expenditure, and other metabolic responses. To determine whether mTORC1 signaling in AgRP neurons is necessary for regulating energy and glucose homeostasis, especially in situations of negative energy balance, mice carrying ablation of the Raptor gene exclusively in AgRP-expressing cells were generated. AgRPΔRaptor mice showed no differences in body weight, fat mass, food intake, or energy expenditure; however, a slight improvement in glucose homeostasis was observed compared to the control group. When subjected to 5 days of food restriction (40% basal intake), AgRPΔRaptor female mice lost less lean body mass and showed a blunted reduction in energy expenditure, whereas AgRPΔRaptor male mice maintained a higher energy expenditure compared to control mice during the food restriction and 5 days of refeeding period. AgRPΔRaptor female mice did not exhibit the food restriction-induced increase in serum corticosterone levels. Finally, although hypothalamic fasting- or refeeding-induced Fos expression showed no differences between the groups, AgRPΔRaptor mice displayed increased hyperphagia during refeeding. Thus, some metabolic and neuroendocrine responses to food restriction are disturbed in AgRPΔRaptor mice.

Thirteen black agouti (Dasyprocta fuliginosa) male fetuses, acquired in two areas of the Amazon Forest, were used for the purpose of morphologically describing the fetal male urogenital organs with a focus on addressing histological and macroscopic aspects. The organs of interest were dissected and photographed in situ and ex situ. Fragments were collected and subjected to routine histological processing for inclusion in paraffin, which was cut and stained by haematoxylin and eosin and Gomori's Trichrome methods and subsequently analysed in a light microscope. The results showed that the urinary tract is like that of domestic animals and is composed of smooth unilobed kidneys covered by a dense capsule of connective tissue and divided into two well-defined regions, cortical and medullary. Ureters, urethra and urinary bladder also showed macroscopic and microscopic characteristics similar to those of domestic animals. The penis of these animals has fibroelastic characteristics, with numerous keratinized structures at its apex. In the middle third of the penis, a "U" shaped penile flexure was seen; the glans penis is covered by a keratinized epidermis containing horny spicules. The presence of a penile bone in an endochondral ossification process was observed, being more developed in gestational ages greater than 76 days. The annex glands were not observed, probably because they were fetuses; only the ampulla of the ductus deferens was identified.

A structural variant (SV) in the agouti signaling protein gene (ASIP), namely ASIP-SV1, has been found to strongly correlate with the darkness of hair coat in specific regions of the body of bulls from the zebu (Bos indicus) Nellore breed. Here we visually analyzed the whole-genome sequence of zebu and taurine (Bos taurus) cattle to elucidate the extent of spread of ASIP-SV1 in different cattle populations. Of 216 sequences analyzed, 63 zebu (45.9%) and five taurine (6.3%) animals had at least one copy of ASIP-SV1. Four of the taurine animals presenting the SV were Romagnola cattle, a breed with history of zebu introgression. The remaining taurine animal was a Simmental, a breed frequently used in crossbreeding. These data provide evidence that ASIP-SV1 is commonly found in zebu populations, in addition to taurine animals with zebu admixture.

Anthelmintic drugs have been used strategically in livestock reared in the tropics. These drugs have been used in the treatment of endoparasitism which have resulted in an increase in the animals performance. The agouti (Dasyprocta leporina) is a neo-tropical rodent with the potential for domestication and has been farmed intensively in Trinidad. However, the objective of this research was to investigate the effect of anthelmintic use of the growth performance of the agouti. In searching the literature, it was found that this type of study on the agouti has not been done. In this experiment fourteen weaned agoutis weighing 1kg were divided into two groups randomly. The first group (T1) was not given any anthelmintic treatment but the second group (T2) was treated with Endovet Ces® subcutaneously every three months. There were no significant differences (p > 0.05) between the two groups in the carcass weight, weight gain, dressing percentage (hot and cold), heart, lungs, skin, head and feet. However, a significant difference (p < 0.05) was seen between groups in the weight of liver and pluck. To the authors knowledge this is the first time that carcass parameters has been presented in literature. The live weight of the animals at the end of the experiment ranged from 2.4 kg to 2.6 kg and animals had a dressing percentage of 57% to 55%. The results are suggestive that the use of anthelmintic drugs in agoutis reared intensively had no significant effect on weight gain and dressing percentage.(AU)

Porém, o objetivo desta pesquisa foi investigar o efeito do uso de anti-helmínticos no desempenho de crescimento da cutia. Neste experimento, quatorze cutias desmamadas pesando 1kg foram divididas em dois gruposale atoriamente. In teleost, as in other vertebrates, stress affects reproduction. A key component of the stress response is the pituitary secretion of the adrenocorticotropic hormone (ACTH), which binds to the melanocortin 2 receptor (MC2R) in the adrenal glands and activates cortisol biosynthesis. In zebrafish, Mc2r was identified in male and female gonads, while ACTH has been shown to have a physiological role in modulating reproductive activity. In this study, the hypothesis that other melanocortins may also affect how the zebrafish gonadal function is explored, specifically steroid biosynthesis, given the presence of members of the melanocortin signaling system in zebrafish gonads. Using cell culture, expression analysis, and cellular localization of gene expression, our new observations demonstrated that melanocortin receptors, accessory proteins, antagonists, and agonists are expressed in both the ovary and testis of zebrafish (n = 4 each sex). Moreover, melanocortin peptides modulate both basal and gonadotropin-stimulated steroid release from zebrafish gonads (n = 15 for males and n = 50 for females). In situ hybridization in ovaries (n = 3) of zebrafish showed mc1r and mc4r in follicular cells and adjacent to cortical alveoli in the ooplasm of previtellogenic and vitellogenic oocytes. In zebrafish testes (n = 3), mc4r and mc1r were detected exclusively in germ cells, specifically in spermatogonia and spermatocytes. Our results suggest that melanocortins are, directly or indirectly, involved in the endocrine control of vitellogenesis in females, through modulation of estradiol synthesis via autocrine or paracrine actions in zebrafish ovaries. Adult zebrafish testes were sensitive to low doses of ACTH, eliciting testosterone production, which indicates a potential role of this peptide as a paracrine regulator of testicular function.

The morphology of the gastrointestinal tract (GI) is a strong indicator of a species' dietary habits. The objective of this study was to describe and quantitatively analyze the gross and microanatomy of the digestive tract of the orange/red-rumped agouti (Dasyprocta leporina) and relate it to the feeding habits of this animal. The digestive tracts of six adult males were used for this study. The results showed that the esophagus was thick (mean thickness of 1023.78 ± 28.97 µm) and lined by keratinized epithelium with scant esophageal glands. Mucosa-associated lymphocytic infiltration was robust throughout the GI tract. These findings suggest that the esophagus was well adapted to a coarse diet. The simple stomach with well-developed gastric glands in the fundus region (mean thickness of 605.39 ± 28.68 µm) was indicative of an adaptation to a carnivorous diet. The small intestine constituted approximately 80% of the length of the GI tract. The remarkable development of the jejunum with a greater villus length (mean thickness of 182.50 ± 27.38 µm) indicated a greater absorptive capacity in frugivorous and carnivorous diets. The long cecum and well-developed colon clearly indicated that the GI tract was well adapted to frugivorous and herbivorous diets. Overall, D. leporina showed that it is well adapted to an omnivorous diet. These results suggest that D. leporina can be fed a balanced omnivorous dry diet with a high protein content of plant or animal origin that is well suited to the architecture of the GI tract in captivity.

The aim of the study was to establish the pattern of the agouti pelvis by obtaining external and radiographic internal pelvimetric values. Forty-three agouti (Dasyprocta prymnolopha), females and males bred in under human care were used. The parameters measured were the external biiliac diameter; the external biischiatic diameter; right and left external ilioischiatic diameters and radiographic internal measurements (true conjugated, the diagonal conjugated; the vertical, the sacral, sagittal, coxal tuberosity, upper biiliac, lower biiliac, and biischiatic diameter. The inlet pelvic area and the outlet pelvic area were calculated, as well the height/width ratios of the entrance area of the pelvis and the pelvic outlet area were calculated. The mean values for each body measurement of females and males were: weight 1.91kg and 2.04kg, external biiliac diameter 6.32cm and 6.30cm, external biischiatic diameter 4.34cm and 4.28cm, right external ilioischiatic diameter 9.01cm and 9.33cm, left external ilioischiatic diameter 9.13cm and 9.30cm, true conjugated 3.90cm and 3.68cm, diagonal conjugated 7.13cm and 6.91cm, vertical diameter 2.59cm and 2.45cm, sacral diameter 2.63cm and 2.44cm, sagittal diameter 3.30cm and 3.09cm, coxal tuberosity diameter 2.52cm and 2.43cm, upper biiliac diameter 6.28cm and 6.24cm, lower biiliac diameter 2.98cm and 2.58cm, biischiatic diameter 2.60cm and 2.70cm, height/width ratio - vertical/ lower biiliac diameter 0.88cm and 0.95cm, sagital/coxal tuberosity diameter 1.32cm and 1.28cm, inlet pelvic area 82.38cm and 77.83cm and outlet pelvic area 24.76cm and 20.07cm. Agouti are dolichopelvic animals, demonstrating the existence of a discrete sexual dimorphism in adults and low intensity correlations between the external and internal measures studied.


O objetivo deste estudo foi estabelecer o padrão da pelve de cutia, masculina e feminina, por meio da obtenção dos valores médios da pelvimetria externa e interna radiográfica. Foram utilizadas 43 cutias (Dasyprocta prymnolopha), fêmeas e machos criadas sob cuidados humanos. Os parâmetros medidos foram o diâmetro biilíaco externo; o diâmetro biisquiático externo; diâmetros ilioisquiáticos externos direito e esquerdo e medidas internas radiográficas (diâmetros conjugado verdadeiro, diagonal conjugado, vertical, sacral, sagital, tuberosidade coxal, biilíaco superior, biilíaco inferior e diâmetro biisquiático). A área pélvica de entrada e a área pélvica de saída foram calculadas , assim como foram calculadas as razões altura/largura da área de entrada da pelve e da área de saída da pelve. Os valores médios para as medidas das fêmeas e dos machos foram, respectivamente: peso 1,91kg e 2,04kg, diâmetro biilíaco externo 6,32cm e 6,30 cm, diâmetro ilioisquiático externo 4,34cm e 4,28cm, diâmetro ilioisquiático externo direito 9,01cm e 9,33cm, diâmetro ilioisquiático externo esquerdo 9,13cm e 9,30cm, diâmetro conjugado verdadeiro 3,90cm e 3,68cm, diâmetro conjugado diagonal 7,13cm e 6,91cm, diâmetro vertical 2,59cm e 2,45cm, diâmetro sacral 2,63cm e 2,44cm, diâmetro sagital 3,30cm e 3,09cm, tuberosidade coxal diâmetro 2,52cm e 2,43cm, diâmetro biilíaco superior 6,28cm e 6,24cm, diâmetro biilíaco inferior 2,98cm e 2,58cm, diâmetro biisquiático 2,60cm e 2,70cm, relação altura/largura - vertical/diâmetro biilíaco inferior 0,88cm e 0,95cm, diâmetro sagital/coxal tuberosidade 1,32cm e 1,28cm, área pélvica de entrada 82,38cm e 77,83 cm e área pélvica de saída 24,76cm e 20,07cm. Ghrelin is a stomach-derived peptide hormone that acts via the growth hormone secretagogue receptor (GHSR) and displays a plethora of neuroendocrine, metabolic, autonomic and behavioral actions. It has been proposed that some actions of ghrelin are exerted via the vagus nerve, which provides a bidirectional communication between the central nervous system and peripheral systems. The vagus nerve comprises sensory fibers, which originate from neurons of the nodose and jugular ganglia, and motor fibers, which originate from neurons of the medulla. Many anatomical studies have mapped GHSR expression in vagal sensory or motor neurons. Also, numerous functional studies investigated the role of the vagus nerve mediating specific actions of ghrelin. Here, we critically review the topic and discuss the available evidence supporting, or not, a role for the vagus nerve mediating some specific actions of ghrelin. We conclude that studies using rats have provided the most congruent evidence indicating that the vagus nerve mediates some actions of ghrelin on the digestive and cardiovascular systems, whereas studies in mice resulted in conflicting observations. Even considering exclusively studies performed in rats, the putative role of the vagus nerve in mediating the orexigenic and growth hormone (GH) secretagogue properties of ghrelin remains debated. In humans, studies are still insufficient to draw definitive conclusions regarding the role of the vagus nerve mediating most of the actions of ghrelin. Thus, the extent to which the vagus nerve mediates ghrelin actions, particularly in humans, is still uncertain and likely one of the most intriguing unsolved aspects of the field.

This review aims at identifying cestodes that are present in hunted rodent species in the neo-tropical region. The rodent species that was investigated were the capybara (Hydrochoerus hydrochaeris, Linnaeus, 1766), lappe (Cuniculus paca, Linnaeus, 1766), agouti (Dasyprocta leporina, Linnaeus, 1758), chinchilla (Chinchilla chinchilla, Lichtenstein, 1829), Trinidad spiny rat (Proehimys trinitatus, Allen and Chapman, 1893), nutria (Myocastor coypus, Molina, 1782), and vizcacha (Lagostomus maximus, Desmarest, 1817). These rodent species are utilized for their meats in many rural communities in the Caribbean and South America. These rodents belong to the hystricomorphic group. Raillietina demerariensis Daniels, 1895 was commonly found in the gastrointestinal tract of D. leporina, C. paca and P. trinitatus. Similarly, in the liver, muscle and subcutaneous tissue the metacestodes on Echinococcus vogeli Daniels, 1895 and Echinococcus oligarthrus was found in the lappe and agouti. The capybara was found to have the most species of cestodes in its gastrointestinal tract when compared to the agouti and lappe. However, metacestodes were not recorded in the tissues of the capybara. This surprising feature shows the effect of the difference in feeding habits between the capybara and the agouti and lappe. The literature reviewed in this study includes scientific publications on cestodes and metacestodes of Hystricomorphic rodents. An exhaustive search was performed using the digital repositories in Google Scholar, Scielo, Redalyc, Scopus and Pubmed. Literature searched spanned the years 1970-2021. Cestodes of zoonotic significance were E. vogeli and E. oligarthrus, with humans becoming infected when consuming eggs of contaminated food and water. The agouti and lappe act as intermediate host in the life cycle of E. vogeli and E. oligarthrus, the definitive host (canids and felids) become infected by consuming of tissue infected with metacestodes. Humans become infected through the ingestion of eggs from the definitive host where cystic lesions develop in the liver, lungs and other abdominal organs.

This is the second of two literature reviews that focuses on the female reproductive anatomy and reproductive technologies used in selected neo-tropical hystricomorphic rodents. The rodents chosen included the agouti (Dasyprocta leporina), the capybara (Hydrochoerus hydrochaeris), and the paca (Cuniculus paca). Over seventy references were used, spanning the years 1919-2021. Knowledge of the reproductive tract is important in understanding any unique features, which may affect the use of reproductive technologies. Some unique characteristics common to these species included the presence of a vaginal closure membrane and a lobulated placenta with a vascular sub-placenta. The capybara had hyperpigmentation of the vagina that was unique to each individual, while the agouti and paca had accessory corpora lutea, in addition to the main one. Some reproductive technologies have been used, with attempts at estrous induction and synchronization taking place within the past five years. Even though most work has been done over the past twenty years, there is still a dearth of information.

Background: The species Dasyprocta prymnolopha is a wild rodent with a geographic distribution that extends from Mexico to South America, including Brazil. Agouti has been the subject of morphophysiological research, but data on the cardiovascular system remains limited. Therefore, the objective was to describe the macroscopic and microscopic morphology, including the study of the cardiac and coronary system syntopy of the species D. prymnolopha. Materials, Methods & Results: Twelve Dasyprocta primnolopha adults were used, 6 males and 6 females. Topographic analysis of the heart was evaluated in situ, with subsequent measurement, anatomovascular description and macroscopic study of cardiac and coronary vascularization. A microscopic investigation and identification of structural cardiac aspects were also carried out in adult agoutis, the biological samples of the heart were submitted to histological techniques and stained with Hematoxylin and Eosin and Masson's Trichrome. The heart is located between the end of the 2nd and the beginning of the 5th intercostal space, with the apex reaching the 6th space. It presents the presence of 2 ligaments: phrenopericardium and sternopericardium. The right atrioventricular valve is composed of 2 cusps, the parietal and the septal, with variations within the same species for 3 cusps. Projections that interconnect the papillary muscles with each other were observed. In the left ventricle there are variations in the number of papillary muscles (range 2 to 4). In the region of the aortic valve, 5 ostia were observed in the left aortic sinus in all animals. The coronary circulation has a predominantly left distribution. Histologically, the heart consists of 3 main layers: endocardium, myocardium, and epicardium. The cusp valves of the atrioventricular openings are composed of endocardial folds that contain a central plate of dense connective tissue, and inserted in this cardiac musculature was observed the cardiac skeleton, with its fibrous ring consisting of collagen and elastic fibers that surrounds the atrioventricular opening. Discussion: Cardiac assessment in wild animals is challenging, as in-depth knowledge of the morphology of the cardiovascular system is required for the use of diagnostic tools. In this first anatomical study of the heart, this organ presents syntopy with those of other rodents, but the topography may vary in individuals of the same species, which may be related to the accentuated vertebral curve. The phrenopericardial and sternopericardial ligaments were observed in this research, although there are no reports in other species of Dasyprocta sp. The arrangement of the arteries has, as a particularity, the larger left atrium in relation to the heart/atrium size ratio when compared to other domestic species, covering the left coronary sinus until it reaches the left atrial surface. In the agouti, it was observed that the vascularization is left, with the left coronary artery giving rise to both the paraconal and subsinuous interventricular branch, a fact found in animals such as ruminants, dogs. In the histological observations of the present study, the heart was similar to that of other mammals. Our data reveal morphological characteristics similar to those of other mammals, but with very expressive characteristics that differ even within the species. It is important to generate new information to elucidate cardiac and coronary diagnostic analyses, which can be extended to different species.

The objective of this experiment was to evaluate the effects of feeding different levels concentrations of dietary calcium propionate (CaPr) on lambs' growth performance; ruminal fermentation parameters; glucose-insulin concentration; and hypothalamic mRNA expression for neuropeptide Y (NPY), agouti-related peptide (AgRP), and proopiomelanocortin (POMC). Thirty-two individually fed lambs were randomly assigned to four treatments: (1) control diet (0 g/kg of CaPr), (2) low CaPr, (30 g/kg dry matter (DM)), (3) medium CaPr, (35 g/kg DM), and (4) high CaPr (40 g/kg DM). After 42 days of feeding, lambs were slaughtered for collecting samples of the hypothalamus. Data were analyzed as a complete randomized design, and means were separated using linear and quadratic polynomial contrast. Growth performance was not affected (p ≥ 0.11) by dietary CaPr inclusion. The ruminal concentration of total volatile fatty acids (VFA) increased linearly (p = 0.04) as dietary CaPr increased. Likewise, a linear increase in plasma insulin concentration (p = 0.03) as dietary CaPr concentration increased. The relative mRNA expression of NPY exhibited a quadratic effect (p < 0.01), but there were significant differences in the mRNA expression of AgRP and POMC (p ≥ 0.10). Dietary calcium propionate did not improve lamb growth performance in lambs feed with only forage diets. Intake was not correlated with feed intake with mRNA expression of neuropeptides.

Trichuriasis is the clinical disease of animals infected with the parasite of the genus Trichuris. This review attempts to present information on Trichuris spp. infestation in neo-tropical rodents that are utilized for meat consumption by humans. Neo-tropical rodents utilized for meat production can be divided into two categories: those that have been domesticated, which include the guinea pig (Cavia porcellus), and those that are on the verge of domestication, such as the capybara (Hydrochoerus hydrochaeris), lappe (Cuniculus paca/Agouti paca), and agouti (Dasyprocta leporina). This document reviews the literature on the species of Trichuris that affects the rodents mentioned above, as well as the clinical signs observed. The literature obtained spans over sixty years, from 1951 to 2020. Trichuris spp. was found in these neo-tropical rodents mentioned. However, there is a dearth of information on the species of Trichuris that parasitize these animals. The capybara was the only rodent where some molecular techniques were used to identify a new species named T. cutillasae. In most cases, Trichuris spp. was found in combination with other endoparasites, and was found at a low prevalence in the lappe and guinea pig. The presence of Trichuris spp. ranged from 4.62-53.85% in the agouti, 4.21-10.00% in the lappe, 50% in the capybaras, and 1-31% in guinea pigs. Further work must be done towards molecular identification of various Trichuris spp. present in these rodents, as well as the clinical effect of infection on the performance of agouti, lappe, capybara, and guinea pigs.

BACKGROUND: The placenta of hystricomorph rodents, lagomorphs and some primates includes an unusual structure, termed a subplacenta, which essentially consists of trophoblastic cells located deep to the central implantation site within the area of decidualization. It has been suggested that the subplacenta is functionally important, although considerable controversy remains on the issue. In this context, our objective was to compare the architecture and structure of the subplacentas of different hystricomorph species, to investigate the possibility that it is active in hormone synthesis. METHODS: In total, the placentas of 3 capybaras (Hydrochaeris hydrochaeris), 2 pacas (Agouti paca), 5 agoutis (Dasyprocta leporina), 5 rock cavies (Kerodon rupestris) and 3 guinea pigs (Cavia porcellus) at different stages of pregnancy (early, middle and near term) were used for gross and microscopic examination. This included the preparation of latex injection casts, immunohistochemistry for steroidogenic enzymes, scanning and transmission electron microscopy. Tissue steroid concentrations were also determined. RESULTS: The gross morphology and microvascular arrangement of the subplacentas were similar among the hystricomorphs studied including ultra-structural verification of cytotrophoblast and syncytiotrophoblast in all species. In guinea pigs, trophoblast cells exhibited characteristics consistent with intense metabolic and secretory activity in general. However, immuno-histochemical evidence also indicated that subplacental trophoblast expressed key steroidogenic enzymes, mainly in the chorionic villus region, consistent with tissue steroid concentrations. CONCLUSIONS: The subplacentas within placentas of hystricomorph rodent species are structurally similar and, in guinea pigs, have potential for steroid hormone secretion from, at least the early stages of pregnancy.

This review, which is the first of two, focuses on the male reproductive anatomy and reproductive technologies used in Neo-tropical hystricomorphic rodents with the potential for domestication, which are the agouti (Dasyprocta leporina), the capybara (Hydrochoerus hydrochaeris) and the paca (Cuniculus paca). We consider over seventy references spanning from 1965 to 2020, with the majority of work being done in the past twenty years. Knowledge of the reproductive tract and reproductive technologies is critical to the conservation and preservation of these species. Although all three animals had similarities in their anatomy, such as no overt scrotums and testes located intra-abdominally in the inguinal region, some had unique features-for example, the agouti and the paca had penile spines, and two lateral penile cartilages. High spermatogenic efficiency was noted in the agouti and the paca, making them good candidates for increasing their reproductive performance in conservation programs. A review of the literature has shown that there is increasing work taking place on the reproductive technologies used in these animals; however, a lot of work is still lacking, as, to the author's knowledge, standard protocols and artificial insemination procedures are yet to be established.
