RESUMEN

To solve the inherent problems of conductive hydrogels, such as relatively low mechanical properties and fatigue resistance, inability to work after water loss, and difficulty weaving. In this study, the borax-crosslinked polyvinyl alcohol/k-carrageenan (kC) conducting hydrogels (BPKKOH) were prepared by a simple one-pot method, and KOH treatment was used instead of the cumbersome and time-consuming freeze-thaw cycle to improve the comprehensive properties. The KOH treatment increased the hydrogel hydrogen bonding content by 7.72 % and synergized with the induction of kC by K+ to enhance the tensile and compressive strengths by 8.12 and 34.6 times, respectively. Meanwhile, the BPKKOH hydrogel's fatigue resistance and shape recovery after water loss were improved. Additionally, the BPKKOH hydrogels can be monitored for finger bending, showing clear and stable differences in electrical signals. BPKKOH hydrogels combined with Morse code realize applications in information transmission and encryption/decryption. Notably, introducing KOH ensures the molding and preparation of BPKKOH hydrogel fibers while having good signal responsiveness and monitoring ability. More importantly, it can be woven into fabrics that can be loaded with heavy weights, which has the potential to be directly applied in smart wearables. This work provides new ideas for applying flexible sensors and wearable smart textiles.

RESUMEN

Bio-aerogels have attracted much attention owing to their remarkable properties, but their brittle and poor elasticity has limited their further applications. Here, we propose a strategy of in-situ silanization crosslinking combined with unidirectional freeze casting (SUFC) to prepare superelastic sodium alginate (SA) aerogels. The resulting aerogel was ultra-light (0.048 g/cm3), high porosity (96.86 %), and self-extinguishing from fire. Aerogels exhibited anisotropic properties, such as low-temperature elasticity (500 g compression at -70 °C 10-cycle, 99.6 % recovery), exceptional fatigue resistance (100-cycle at 50 % strain), and excellent thermal insulation (0.0696 W·m-1·K-1). Thus, the SUFC strategy provides considerable freedom for constructing multi-material, lamellar/honeycomb structured alginate-based aerogels, which pave the way to thermal insulation development at low temperatures.

In the current study we investigated the inhibitory effect of rucaparib (Rubraca®) on human ovarian cancer SKOV3 and A2780 cells and its possible mechanism. Cancer cells and human normal ovarian epithelial IOSE80 cells were treated with Rubraca® at different concentrations. Cell viability was measured by MTT assay. Necrotizing apoptosis was detected by Annexin V-FITC/PI double staining combined with flow cytometry. Reactive oxygen species were measured by 2',7'-dichlorofluorescent yellow diacetate (DCFH-DA) fluorescent probe. The expression of receptor-interacting protein kinase 1 (RIP1) and RIP3 protein was determined by Western Blot. Our data showed that Rubraca® inhibited the proliferation of ovarian cancer SKOV3 and A2780 cells in a dose-and time-dependent manner. After Rubraca® treatment, the apoptotic rate of SKOV3 and A2780 cells (Annexin V+/PI-cells) did not change significantly, but the proportion of necrotic cells (PI+cells or Annexin V+/PI+cells) increased significantly, which was different from the control group. Furthermore, Rubraca® could significantly induce SKOV3 and A2780 cells to produce excessive reactive oxygen species and significantly upregulate the expression of RIP1 and RIP3. When pretreated with reactive oxygen species inhibitor N-acetyl-L-cysteine (NAC) or RIP1 inhibitor (Nec-1), the necrosis apoptotic rate of SKOV3 and A2780 cells decreased significantly. In summary, Rubraca® could significantly inhibit the proliferation of ovarian cancer SKOV3 and A2780 cells, which may be partially achieved via upregulating the expression of RIP1 and RIP3 proteins, and activating the process of necrotic apoptosis.

BACKGROUND: Loss of heterozygosity (LOH) on chromosome arm 3p, where the gene of thyroid hormone receptor beta (THRB) is located, has been reported in breast cancer. Although some studies performed in vitro have suggested that THRB could act as a tumor suppressor in breast cancer development, there is still no unequivocal evidence to support this. METHODS: To determine the role of LOH in breast tumor development, the LOH of THRB and its proximal microsatellite markers D3S1293, D3S3659, D3S3700, D3S2307 and D3S2336 was investigated in a genomic region spanning ~3.3 Mb in tumor specimens and in corresponding normal tissues of 74 invasive breast cancer patients. The association was analyzed between LOH in microsatellite markers and clinicopathological characteristics. RESULTS: LOH was detected in D3S1293 (36.7%), THRB (59.4%), D3S3659 (37.5%) and D3S3700 (55.2%) among the informative cases, while LOH was not detected in D3S2307 and D3S2336. Cases exhibited LOH of 52.8%-71.4% if any 2 markers were combined and analyzed out of the first 4 microsatellite markers. LOH in THRB was associated with negative estrogen receptor (ER), negative progesterone receptor (PR), both negative estrogen receptor and progesterone receptor (HR) and human epidermal growth factor receptor-2 (HER2) and lymph node metastasis (p = 0.0001, p = 0.005, p = 0.001 and p = 0.018). The association with negative PR in LOH in THRB and/or D3S1293 was pronounced (p<0.0001). LOH in D3S3700 showed an association with lymph node metastasis (p = 0.014). This association was enhanced if D3S3700 was combined with THRB or D3S3659 (p = 0.0004, p = 0.0002). CONCLUSIONS: LOH in THRB and its proximal microsatellite markers may play a role in tumorigenesis and development in invasive breast cancer.

Infrared spectroscopy (IR) is an important means of seeing the characteristics of the structural properties. Fourier transform infrared spectroscopy (FTIR) was applied to analyze the structural properties of biochar from different materials with different methods. The results showed that: the biochars have IR absorption peaks of hydroxyls group, aromatic group and containing organic group with the activated? charcoal, but in other absorption peaks, with a significant difference. The high temperature can make -OH, -CH3, -CH2-, -C=O to be associated or loss, and promotes the formation of aromatic groups during Carbonization of corn straw. At the different carbonization mode, the heating and microwave carbonization, has a carbonize mechanism of biochar, heating method may make -OH in alcohol and phenol to combinative with each other or loss, and to form benzene ring group and an aromatic group, Aromatic group in microwave method was so preventing to participate in the hot reaction, to form the more benzene substances. These results show that the Infrared spectrum can well analysis the structural characteristics of biochar, and showed that it comprises -OH, the aromatic group and other active groups.

This study concerns the cytogenetic stability of in vitro human bone marrow-derived mesenchymal stem cells (MSCs) in primary culture and after passaging. Bone marrow samples were collected from seven brain malfunction patients involved in autologous MSC transplantation trials. Chromosome preparations from primary MSC cultures and after 3 passages were analyzed by conventional staining and G-banding techniques. All MSCs showed normal diploid karyotypes, 46 XY or 46 XX, without aneuploidy or polyploidy; chromosome structural abnormalities were not detected. The results indicate that the in vitro cultured MSCs retained normal cytogenetics before being transplanted back into the patients.
