RESUMEN

Cancer stem cells (CSCs) play a pivotal role in the pathogenesis of human cancers. Previous studies have highlighted the role of long non-coding RNA (lncRNA) in modulating the stemness of CSCs. In our investigation, we identified an upregulation of lncRNA FOXD1-AS1 in CSCs. The enforced expression of lncRNA FOXD1-AS1 promotes tumorigenesis and self-renewal in pancreatic cancer CSCs. Conversely, the knockdown of lncRNA FOXD1-AS1 inhibits tumorigenesis and self-renewal in pancreatic cancer CSCs. Furthermore, our findings reveal that lncRNA FOXD1-AS1 enhances self-renewal and tumorigenesis in pancreatic cancer CSCs by up-regulating osteopontin/secreted phosphoprotein 1(SPP1) and acting as a ceRNA to sponge miR-570-3p in pancreatic cancer (PC) CSCs. Additionally, lncRNA FOXD1-AS1 depleted pancreatic cancer cells exhibit heightened sensitivity to 5-FU-indued cell growth inhibition and apoptosis. Analysis of patient-derived xenografts (PDX) indicates that a low level of lncRNA FOXD1-AS1 may serve as a predictor of 5-FU benefits in PC patients. Moreover, the introduction of SPP1 can reverse the sensitivity of lncRNA FOXD1-AS1-knockdown PC cells to 5-FU-induced cell apoptosis. Importantly, molecular studies have indicated that the elevated levels of lncRNAFOXD1-AS1 in PC are facilitated through METTL3 and YTHDF1-dependent m6A methylation. In summary, our results underscore the critical functions of lncRNA FOXD1-AS1 in the self-renewal and tumorigenesis of pancreatic cancer CSCs, positioning lncRNA FOXD1-AS1 as a promising therapeutic target for PC.

PURPOSE: This study was to evaluate clinical results of adhesives to sclerotic dentin in non-carious cervical lesions pretreated with ethylene diamine tetraacetic acid (EDTA). METHODS: Twelve patients who had at least one pair of teeth with typical cervical wedge-shaped defects and class III sclerotic dentin were selected. The teeth were randomly divided into experimental and control groups. In the experimental group, EDTA was used to pretreat dentin surface first and then self-etching resin adhesive; In the control group, only self-etching resin adhesive was used. Modified USPHS rating system was applied in baseline, 6 months and 12 months after treatment. SAS 8.02 software package was used for two-sample rank sum test. RESULTS: At the time point of 6-month and 12-month, the scores in the experimental group were significantly better than the control group (P<0.05). CONCLUSIONS: EDTA pretreatment of the sclerotic dentin of cervical wedge-shaped defects could improve the resin bonding strength, marginal discoloration, marginal form and surface quality.

OBJECTIVE: Evaluation of left ventricular (LV) function and dyssynchrony is of vital importance. A newly-developed three-dimensional (3D) speckle tracking echocardiography (STE) has the potential to circumvent limitations of two-dimensional STE. Our study was to evaluate LV function and dyssynchrony by 3D-STE. METHODS AND RESULTS: Fifty-two non-ischaemic dilated cardiomyopathy patients (DCM) with complete left bundle-branch block and 55 healthy subjects were enrolled. Of these patients, 24 underwent cardiac resynchronization therapy (CRT). 3D-STE was performed before and 1 month after CRT. We measured 3D (3DS), longitudinal (LS), circumferential (CS) and radial (RS) strain of the LV. LV dyssynchrony was evaluated by the standard deviation of time to peak negative value of 3D strain (3DS-SD) and time to reach the minimum regional volume of 16 segments (SDI) related to the heart cycle. DCM patients had significantly lower global 3DS (-17.12 +/- 6.70% vs -38.38 +/- 4.28%, P < 0.01), significantly greater 3DS-SD (11.11 +/- 5.14% vs 4.04 +/- 1.43%, P < 0.01) and SDI (9.69 +/- 4.82% vs 4.23 +/- 1.33%, P < 0.01) than normal volunteers. 3DS, LS, CS, RS had an excellent correlation with LV ejection fraction (LVEF) (r2 = -0.94, -0.91, -0.93, 0.89, respectively, P < 0.01 for all). 3DS-SD correlated well with SDI (ICC = 0.85). For mild, moderate and severe systolic dysfunction, 3DS-SDs were 4.56 +/- 1.53%, 9.87 +/- 2.58%, and 14.55 +/- 4.71%, respectively (P < 0.01) and SDIs were 4.46 +/- 1.27%, 8.19 +/- 2.35%, and 12.83 +/- 4.87%, respectively (P < 0.01), but independently of QRS width. After CRT therapy, global 3DS (-13.12 +/- 2.63% to -14.06 +/- 2.53%, P = 0.025), 3DS-SDs (12.99 +/- 3.92% to 11.53 +/- 4.53%, P = 0.015) and SDIs (10.85 +/- 3.74% to 9.50 +/- 4.63%, P = 0.013) were markedly improved. CONCLUSION: When image quality is optimal, 3D STE seems to be a promising approach assessing LV function and dyssynchrony.

RESUMEN

PURPOSE: To assess the efficacy of compound Chinese traditional medicine(CTM), which is composed of gallic acid, magnolol and polysaccharide of Bletilla, against apical periodontitis in dogs and cytotoxic assay. METHODS: A animal model of apical periodontitis was built, CTM was then used to disinfect the root canal. The effect of the restoration of periapical bone in dogs was investigated after regular root canal filling. SAS6.12 software package was used for statistical analysis, and MTT was used to test cell toxicity of CTM. RESULTS: CTM can cure inflammation effectively, and CTM had no cytotoxic effect on periodontal ligament cells at 5-week. CONCLUSIONS: The compound Chinese traditional medicine may be an effective disinfecting drug for root canal disinfection.

RESUMEN

In vivo tumor imaging technique method based on bioluminescence principle was established to evaluate the anti-tumor effect of paclitaxel mixed micelle (PMM). MDA-MB-231 tumor cells with luciferase reporter vectors were firstly implanted into nude mice, and subsequently the luciferase substrate was regularly injected during intraperitoneal administration of PMM. Then the tumor size, growth and the intensity of light signals were monitored with in vivo imaging technique. The method of luciferase tumor in vivo imaging could be real-time, reliable and exact in labeling and reflecting the growth of tumors, and the observed results were consistent with that by conventional method, so it would be a feasible approach to study anti-tumor effect of drugs. The anti-tumor effect of paclitaxel mixed micelle was observed by this method, and the results showed that this formulation could inhibit growth of tumor, and the anti-tumor rate of it was about 85%.
