A study was made of the lipid peroxidation in the microsomal fraction of human placenta tissue in the presence of flumecinol (Zixoryn, Gedeon Richter Pharmaceutical Works, Budapest, Hungary). The tissue samples were obtained from placenta in early pregnancy and also from placenta at term. Up to a concentration of 1 mumolar, flumecinol (3-trifluoromethyl-alpha-ethyl-benzhydrol), as an enzyme inductor, significantly enhanced the rate of lipid peroxidation but above 1 molar it scarcely influenced it. The in vitro study proved the enzyme-inducing effect of flumecinol on lipid peroxidation in early human placenta.

The response of LH release to exogenous estrogens was studied in three patients with androgen insensitivity syndrome (AIS) before and after gonadectomy and a prolonged treatment with a sequential pill. Estrogen provocation tests were performed in which 0.05 mg/kg body weight estradiol benzoate was administered intramuscularly and serum FSH and LH levels were assessed every 12 hours for 96 hours after gonadectomy, three months after a prolonged treatment with a sequential pill (16 cycles: 9 days mestranol 100 micrograms, 12 days mestranol 80 micrograms and chlormadinone acetate 2 mg) and in one patient before gonadectomy. The control group consisted of 10 normal females during the follicular phase. A positive feedback effect was induced in normal females and in AIS patients after prolonged treatment with a sequential pill.

A simple method was developed to measure the SHBG capacities of human serum, semen and sperm cells. After suitable dilution, disintegration and addition of labelled dihydrotestosterone-1,2-3 H or testosterone-1,2-3 H, the SHBG was precipitated by the addition of saturated ammonium sulphate in a final concentration of 42.3%. The precipitate was centrifuged and the activity of the non-bound, labelled steroid was counted in an aliquot of the supernatant. Subtraction of this result from the total activity yielded the SHBG-bound steroid in microgram/100 ml or nmol/l. Examination of 52 males gave normal values of means = 13.91 nmol/l (S.E.M. = 0,746) dihydrotestosterone binding globulin (DHTBG) and means = 11.67 nmol/l (S.E.M. = 0.555) testosterone binding globulin (TBG) in serum, while the concentrations in the seminal plasma were means = 10.89 nmol/l (S.E.M. = 0,723) DHTBG and means = 8.93 nmol/l (S.E.M. = 0.625) TBG. means = 5.57 ng/mg protein (S.E.M. = 0.516) DHTBG and means = 4.91 ng/mg protein (S.E.M. = 0.440) TBG were found in the disintegrated sperm cells.

In the presence of ethynodiol diacetate the ethynylestradiol concentration in serum and saliva after oral administration was measured by a rapid radioimmunoassay method developed by the authors. By means of the saliva/serum quotient, and from the concentration of ethynylestradiol in the saliva samples, it was possible to infer the ethynylestradiol content of the serum. Attention is called to the relation between the quotient and time, which should be taken into account when values are calculated. 2, 4 and 12 h after oral administration, the saliva/serum quotients for an ethynylestradiol dose of 0.05 mg were 0.27, 0.76 and 0.40, respectively.

It was found by radioimmunoassay that the ethynyl estradiol concentration of the cervical mucus was 8.8 times higher than that of the serum in the first third of the cycle, 14 times higher in the second and 7.3 times higher in the third. The ethynyl estradiol content of the cervical mucus in the middle of the cycle showed a marked increase even when expressed as protein ratio. The accumulation of ethynyl estradiol in the cervical mucus is best demonstrated by the cervical mucus/serum ethynyl estradiol quotient of 9.7 found 12 h after oral administration of the contraceptive Bisecurin.

A comparison was made between the stimulatory effects of luteinizing hormone-releasing hormone (LHRH) and long acting luteinizing-releasing hormone (I.a. LHRH) on the gonadotropin secretion of normally menstruating women. The luteinizing hormone (LH) and follicle stimulating hormone (FSH) concentrations were measured by RIA. The synthetic I.a. LHRH was found more potent than the parent hormone in terms of the magnitudes and durations of the LH and FSH responses. There was no close correlation between the initial gonadotropin values and the peak levels in normally menstruating women having basal LH and FSH values within the normal ranges.

Relationships between the spermiogram and sex hormones in 104 andrologic patients were studied. Endocrine connections in normal and in pathologic spermiogenesis have been studied. A rather close correlation was found between FSH and ICSH levels. ICSH increases parallel with the increase of FSH level. There is also a close relationship between sperm concentration and FSH level. Between ICSH and sperm count the correlation is not significant. Between steroid hormone level and sperm concentration no correlation ws found. It is only the plasma FSH level which show a close relationship with the disorder of spermiogenesis and it has diagnostic significance.
