RESUMEN

The feed additives (prebiotics and probiotics) are used to stabilize the healthy gut microbiome by supporting beneficial microorganisms, thereby improving the animal growth rate. Thirty growing lambs, with around 20.50 ± 0.65 kg live weight were placed into five equal groups (6 animals each). The concentrate feed mixture (CFM) + roughage was given to the control groups. The treatments (T) of T1, T2, and T3 treatments were fed the control ration with three levels of prebiotic supplementation: 0.50, 1.00, and 1.50 g/kg CFM of mannan oligosaccharids + beta glucan, respectively. The T4 received the control ration and was supplemented with 1.0 g/kg CFM probiotic (3.0 × 108 CFU/g, Bacillus amyloliquefaciens). The roughage was provided ad libitum, and the animals were supplemented with CFM at 2.00% of the body weight. A digestibility trial was conducted at the end of the 150-day feeding trial. The results demonstrated that increasing the prebiotic to 0.15% enhanced average daily gain and feed efficiency (P < 0.05) when compared to the control group. Although daily gain and feed efficiency in probiotic-fed animals were higher (P < 0.05) than in the control group, they were lower in prebiotic-fed lambs. The blood parameters were within normal range. The animals that received 0.10% prebiotic had the highest economic feed efficiency when compared to the other groups. Prebiotic treatment improved nutrient digestibility and nutritive values; however, the results for control and probiotic treatment were practically identical. Additionally, further research is needed to investigate the effects of prebiotics, probiotics and synbiotics as feed additives on productive and reproductive performance in ruminants.

RESUMEN

A middle aged COVID-19 male patient presented 2 weeks after discharge with new onset of dyspnoea and desaturation. Radiological studies revealed right side pneumothorax and lower lobe cystic air space. Chest drain was inserted and on a later date the patient underwent thoracoscopic surgery where a large pneumatocele was identified. Deroofing and closure of sources of air leak were done. Histopathological examination demonstrated extensive fibrosis, intra-alveolar Haemorrhage and pneumocytes hyperplasia.

BACKGROUND: Pleural collection is a common medical problem. For decades, the chest tube of different designs was the commonly used toll for pleural drainage. Over the past few years, small-bore catheter (SBC) has gained more popularity. We present our experience of using SBCs for the drainage of pleural collection of different etiologies. PATIENTS AND METHODS: A total of 398 small-bore pleural catheters were inserted in 369 patients with pleural collection during the period from January 2013 to October 2019. Data were collected regarding the efficacy of drainage, experienced chest pain, duration of drainage, and the occurrence of complications. RESULTS: Malignant associated (59.24%) and parapneumonic (19.57%) effusions constituted the most common causes. The drainage was successful in 382/398 (95.98%) occasions; six cases had incomplete fluid evacuation that required decortications; five cases (1.26%) had nonexpendable lung. Catheter reinsertion was needed due to dislodgment in 2 (0.50%) cases and obstruction in 3 (0.75%) cases. Sixty-two cases (15.58%) experienced chest pain. No patient developed empyema or cellulites at the site of catheter insertion. The duration of drainage ranged from 2 to 7 days, with an average of 3.5 days. CONCLUSIONS: SBC is equivalent to conventional chest tube for the drainage of pleural collection. Moreover, it has the advantages of less associated pain, versatility of insertion site, and relative safety of the technique in some risky and difficult situations.

Sex-specific differences in mitochondrial function and free radical homeostasis are reported in the context of aging but not well-established in pathogeneses occurring early in life. Here, we examine if sex disparity in mitochondria function, morphology, and redox status starts early and hence can be implicated in sexual dimorphism in cardiac as well as neurological disorders prevalent at young age. Although mitochondrial activity in the heart did not significantly vary between sexes, female brain exhibited enhanced respiration and higher reserve capacity. This was associated with lower H2O2 production in female cardiac and brain tissues. Using transmission electron microscopy, we found that the number of female cardiac mitochondria is moderately greater (117 ± 3%, P = 0.049, N = 4) than male's, which increased significantly for cortical mitochondria (134 ± 4%, P = 0.001, N = 4). However, male's cardiac mitochondria exhibited fragmented, circular, and smaller mitochondria relative to female's mitochondria, while no morphologic sex-dependent differences were observed in cortical mitochondria. No sex differences were detected in Nox2 and Nox4 proteins or O2-consuming/H2O2-producing activities in brain homogenate or synaptosomes. However, a strong trend of increased EPR-detected NOX superoxide in male synaptosomes hinted at higher superoxide dismutase activity in female brains, which was confirmed by two independent protocols. We also provide direct evidence that respiring mitochondria generally produce an order-of-magnitude lower reactive oxygen species (ROS) proportions than currently estimated. Our results indicate that sex differences in mitochondrial biogenesis, bioenergetics, and morphology may start at young age and that sex-dependent SOD capacity may be responsible for differences in ROS homeostasis in heart and brain.

Treatment with doxorubicin (DOX) is a common treatment for different types of cancer. DOX-NP is one of a well established marketed liposomal formulation for DOX. It has advantages over free DOX in reducing the cardiac toxicity and increasing the efficacy. Gold nanoparticles (GNPs), have been widely used in biomedical applications such as medical imaging and biosensors. Mice bearing Ehrlich tumor were injected with saline, free doxorubicin (DOX) in solution, gold nanoparticles loaded liposomes and commercial liposomal encapsulated doxorubicin (DOX-NP). The results showed that GNPs loaded liposomes could enhance the antitumor activity of commercial liposomal formulation (DOX-NP) and displayed significantly decreased systemic toxicity compared with free DOX and commercial liposomal formulation (DOX-NP) at the equivalent dose. So the injection of GNPs and DOX-NP is expected to increase the cell killing and make it a promising approach to cancer treatment.

Ehrlich tumors were grown in female balb mice by subcutaneous injection of Ehrlich ascites carcinoma cells. Mice bearing Ehrlich tumor were injected with saline, DOX in solution or DOX encapsulated within liposomes prepared from DMPC/CHOL/DPPG/PEG-PE (100:100:60:4) in molar ratio. Cytotoxicity assay showed that the IC50 of liposomes containing DOX was greater than that DOX only. Tumor growth inhibition curves in terms of mean tumor size (cm(3)) were presented. All the DOX formulations were effective in preventing tumor growth compared to saline. Treatment with DOX loaded liposomes displayed a pronounced inhibition in tumor growth than treatment with DOX only. Histopathological examination of the entire tumor sections for the various groups revealed marked differences in cellular features accompanied by varying degrees in necrosis percentage ranging from 12% for saline treated mice to 70% for DOX loaded liposome treated mice. The proposed liposomal formulation can efficiently deliver the drug into the tumor cells by endocytosis (or passive diffusion) and lead to a high concentration of DOX in the tumor cells. The study showed that the formulation of liposomal doxorubicin improved the therapeutic index of DOX and had increased anti-tumor activity against Ehrlich tumor models.

Date palm fruits of the species Phoenix dactylifera exhibit a great diversity in sensory attributes including skin color, smell and taste. This study attempts to elucidate the primary and secondary metabolite profiles of 21 date varieties from Egypt. A major difficulty is sugar rich matrix and skin-fibers embedding the secondary metabolites. A total of 49 metabolites extracted from the fruit skin were evaluated in a UPLC/PDA/ESI-qTOF-MS based metabolomics study. The total phenolic contents of the varieties varied from 233 to 1897mg/100g (2.3-19g/kg) dry weight. The predominant flavones were glycosides of luteolin and apigenin, quercetin conjugates were the principal flavonols, whereas caffeoyl shikimic acid was the main hydroxycinnamic acid conjugate. Aside from these major phenolic classes, a group of sphingolipids, fatty and other organic acids was also identified. The total non-fatty organic acid content correlates with reported shelf lives. GC-MS was further utilized to localize primary metabolites in fruits (i.e. sugars and organic acids) with glucose and fructose accounting for up to 95% of TIC among most varieties. Principal component and clustering analyses reveal that flavonols and sugars, both contribute the most to variety classification. This study provides the most complete map for polyphenol & sugar distribution in Egyptian date fruit varieties. By describing the metabolite profiles in a diverse dataset of P. dactylifera, this study provides the basis for future investigations of date fruit nutritional value beyond providing energy and its potential for nutraceutical enhancement or storability.

RESUMEN

A 6-month old baby referred to our department because of recurrent attacks of respiratory distress with chest infection. Chest radiology revealed reduction of the right hemithorax with mediastinal shift to the right. Multidetector computed tomography showed hypoplasia of the right lung and right pulmonary artery, systemic arterial supply to the lung from the abdominal aorta, and and absence of right venous drainage to the left atrium. This picture is consistent with congenital pulmonary venolobar syndrome. The patient underwent right pneumonectomy; the postoperative course was uneventful.

Gold nanoparticles were prepared and loaded into the bilayer of dipalmitoylphosphatidylcholine (DPPC) liposomes, named as gold-loaded liposomes. Biophysical characterization of gold-loaded liposomes was studied by transmission electron microscopy (TEM) and Fourier transform infrared (FTIR) spectroscopy as well as turbidity and rheological measurements. FTIR measurements showed that gold nanoparticles made significant changes in the frequency of the CH(2) stretching bands, revealing that gold nanoparticles increased the number of gauche conformers and create a conformational change within the acyl chains of phospholipids. The transmission electron micrographs (TEM) revealed that gold nanoparticles were loaded in the liposomal bilayer. The zeta potential of DPPC liposomes had a more negative value after incorporating of Au NPs into liposomal membranes. Turbidity studies revealed that the loading of gold nanoparticles into DPPC liposomes results in shifting the temperature of the main phase transition to a lower value. The membrane fluidity of DPPC bilayer was increased by loading the gold nanoparticles as shown from rheological measurements. Knowledge gained in this study may open the door to pursuing liposomes as a viable strategy for Au NPs delivery in many diagnostic and therapeutic applications.

BACKGROUND: One particularly exciting field of research involves the use of gold nanoparticles (GNPs) in the detection and treatment of cancer cells in the liver. The detection and treatment of cancer is an area in which the light absorption and emission characteristics of GNPs have become useful. Currently, there are no data available regarding the fluorescence spectra or in vivo accumulation of nanoparticles (NPs) in rat liver after repeated administration. In an attempt to characterise the potential toxicity or hazards of GNPs in therapeutic or diagnostic use, the present study measured fluorescence spectra, bioaccumulation and toxic effects of GNPs at 3 and 7 days following intraperitoneal administration of a 50 µl/day dose of 10, 20 or 50 nm GNPs in rats. METHODS: The experimental rats were divided into one normal group (Ng) and six experimental groups (G1A, G1B, G2A, G2B, G3A and G3B; G1: 20 nm; G2: 10 nm; G3: 50 nm; A: infusion of GNPs for 3 days; B: infusion of GNPs for 7 days). A 50 µl dose of GNPs (0.1% Au by volume) was administered to the animals via intraperitoneal injection, and fluorescence measurements were used to identify the toxicity and tissue distribution of GNPs in vivo. Seventy healthy male Wistar-Kyoto rats were exposed to GNPs, and tissue distribution and toxicity were evaluated after 3 or 7 days of repeated exposure. RESULTS: After administration of 10 and 20 nm GNPs into the experimental rats, two fluorescence peaks were observed at 438 nm and 487 nm in the digested liver tissue. The fluorescence intensity for 10 and 20 nm GNPs (both first and second peaks) increased with the infusion time of GNPs in test rats compared to normal rats. The position of the first peak was similar for G1A, G2A, G1B, G2B, G3B and the normal (438 nm); that for G3A was shifted to a longer wavelength (444 nm) compared to the normal. The position of the second peak was similar for G1A, G1B, G2A, G2B and the control (487 nm), while it was shifted to a shorter wavelength for G3A (483 nm) and G3B (483 nm). The fluorescence intensity of the first and second peaks increased for G1A, G2A, G1B and G2B, while it decreased for G3A and G3B compared to the control. CONCLUSIONS: The fluorescence intensity of GNPs varied with the number, size and shape of particles and with the ratio of surface area to volume in a given sample. Fluorescence intensity changes during infusion depended on the size and shape of GNPs, with smaller particles experiencing larger changes during the infusion time in addition to the quenching produced by the larger GNPs. It is likely that smaller particles, which have a much higher ratio of surface area to volume compared to larger particles, are more prone to aggregation and surface interaction with biological components. This study suggests that fluorescence intensity can be used to evaluate bioaccumulation and the toxicity of gold nanoparticles in rats.
