RESUMEN

Objective To analyze the incidence trend of hand-foot-mouth disease (HFMD) and its correlation with meteorological factors in Children in Kaizhou District, Chongqing from 2018 to 2021, and to provide a theoretical basis for the diagnosis and treatment of HFMD in children. Methods The HFMD epidemic information was collected from 2018 to 2021 in Kaizhou District of Chongqing by using the China Disease Surveillance Information and Report Management System. The epidemiological characteristics of HFMD were descriptively analyzed, and the correlation between HFMD incidence and meteorological factors was analyzed by multiple regression. Results A total of 5 121 HFMD cases were reported in Kaizhou District of Chongqing from 2018 to 2021, with an average annual incidence of 143.30/100 000. The incidence of HFMD fluctuated from 120.87/100,000 to 159.78/100,000 from 2018 to 2021, showing a downward trend year by year. There were 2929 males and 2192 females with HFMD. The incidence of HFMD was the highest in early childhood (70.13/100 000), followed by pre-school age (43.06/100 000). There was significant difference in the incidence of HFMD among different age groups (χ2=53.497, P<0.05). The cases were mainly scattered children (3127 cases, 61.06%). The second was nursery children (1627 cases, 31.77%). In addition, there were 289 cases of students (5.64%). There were 1084 laboratory-confirmed cases in Kaizhou District of Chongqing from 2018 to 2021, including 269 (24.82%) children with EV71 infection, 178 (16.42%) children with Cox A16 infection, and 637 (58.76%) children with other enterovirus infections. There were significant differences in pathogen composition among different years (Z=32.75, P<0.05). From 2018 to 2021, the proportion of EV71 increased year by year, while COX16 and other enterovirus infections showed a downward trend year by year. Average daily temperature (OR=1.873) and average daily pressure (OR=-1.498) were independent risk factors for HFMD in Kaizhou District of Chongqing (P<0.05). Conclusion The reported incidence of HFMD in Kaizhou District of Chongqing shows a decreasing trend, and the incidence is closely related to temperature and atmospheric pressure. It is still necessary to strictly implement the prevention and control measures in key population in the season of high incidence. The main virus is EV71, which can be vaccinated with EV71 HFMD vaccine to reduce the occurrence of severe cases.

Objective To investigate the death prognosis and risk factors of extensively drug-resistant Acinetobacter baumannii in hospitalized elderly patients with hematological infection, so as to facilitate clinical prevention and treatment. Methods The elderly (> 80 years old) hospitalized patients with hematological infection in our hospital from 2015 to 2021 were selected for analysis. Firstly, 314 patients with extensively drug-resistant Acinetobacter baumannii hematological infection were distinguished by etiological analysis. A total of 98 cases of death were detected during hospitalization (later referred to as the observation group). By comparing with the surviving patients (216 cases) (later referred to as the control group), the general data of patients with XDRAB infection were collected, and the risk of death and its influencing factors were analyzed. Results In the study, the proportion of patients in the observation group who used catheters was higher, the catheter retention time was longer, the acute physiology and chronic health status II scores were higher, and the proportion of patients who lost self-care ability was also higher. Conclusion The death of blood borne infection of extensively drug-resistant Acinetobacter baumannii in elderly patients is affected by many factors. Among them, patients who use catheters for a long time, have poor self-care ability and lose self-care ability have a higher risk of death, which is worthy of clinical attention.

Near-surface air temperature (Ta) is an important parameter in agricultural production and climate change. Satellite remote sensing data provide an effective way to estimate regional-scale air temperature. Therefore, taking Gansu section of the upper Weihe River Basin as the study area, using the filtered reconstructed high-quality long-time series normalized difference vegetation index (NDVI), interpolated reconstructed land surface temperature (LST), surface albedo, and digital elevation model (DEM) as the input data, the back-propagation artificial neural network algorithm (BP-ANN) was combined with a multiple linear regression method to estimate regional air temperature, and the influencing factors of air temperature estimation were analyzed. This method effectively compensates for the fact that air temperature data provided by a single station cannot represent regional air temperature information. The result shows that the temperature estimation accuracy is high. In terms of interannual variation, the air temperature in the study area showed a slightly increasing trend, with an average annual increase of 0.047°C. The calculation results of the interannual variation rate of temperature showed that the area with increased air temperature accounted for 75.8% of the total area. In terms of seasonal variation, compared with that in summer and winter, the air temperature rising trend in autumn was obvious, and the air temperature in the middle of the study area decreased in spring, which is prone to frost disasters. LST and NDVI in the study area were positively correlated with air temperature, and their positive correlation distribution areas accounted for 93.62% and 94.34% of the total study area, respectively. NDVI, LST and DEM influence the temperature change in the study area. The results show that there is a significant positive correlation between NDVI and air temperature, and the change of NDVI has a positive effect on the spatiotemporal variation of air temperature. The correlation coefficient between LST and air temperature in the southeast of the study area is negative, and there is a difference. In addition, the correlation coefficient between LST and air temperature in other areas of the study area is positive. The air temperature decreased with elevation, air temperature decreases by 0.27°C every hundred meters.

Tubulin vinca-domain ligands can inhibit microtubule polymerization, causing cell death in mitosis, and their potential against multiple cancer types has been demonstrated. However, due to drug resistance and toxicities, development of novel vinca-domain ligands is still needed. In this study, we determined the high-resolution crystal structures of vinorelbine, YXD, and Phomopsin A in complex with tubulin at 2.5 Å. Additionally, we recapitulated all previously published high-resolution crystal structures of the vinca binding site to reveal critical residues and the molecular mechanism of vinca-domain ligands interacting with tubulin. Furthermore, we designed putatively novel triazolopyrimidine derivatives by introducing secondary amine groups to establish salt-bridge and H-bond interactions with Asp179ß1 and Asn329α2 . Our studies provided the structural basis for designing novel tubulin vinca-domain ligands.

RESUMEN

The RBD (receptor binding domain) of the SARS-CoV-2 virus S (spike) protein mediates the viral cell attachment and serves as a promising target for therapeutics development. Mutations on the S-RBD may alter its affinity to cell receptor and affect the potency of vaccines and antibodies. Here we used an in-silico approach to predict how mutations on RBD affect its binding affinity to hACE2 (human angiotensin-converting enzyme2). The effect of all single point mutations on the interface was predicted. SPR assay result shows that 6 out of 9 selected mutations can strengthen binding affinity. Our prediction has reasonable agreement with the previous deep mutational scan results and recently reported mutants. Our work demonstrated in silico method as a powerful tool to forecast more powerful virus mutants, which will significantly benefit for the development of broadly neutralizing vaccine and antibody.

Objective To compare the efficacy of Letrozol ( LE) regimen for ovulation induction in elderly and younger patients with polycystic ovary syndrome (PCOS).Methods A total of 67 ovulation induction cycle by LE regimen in patients elderly than 35 years old or younger than 28 years old with PCOS were studied .The endometrial thickness,morphology,number of dominant follicles,ovulation on the day of injection HCG and pregnancy outcome were recorded.Results The endometrial thickness were significantly different between the two groups [(7.9 ±1.7) mm vs (9.4 ±3.2)mm,t=2.648,P0.05].Conclusion LE regimen for ovulation induction is effective for PCOS patients of all ages ,although the elderly patients with endometrial thick-ness is less than that in younger ones .The number of dominant follicles ,endometrial morphology and ovulation is no different,don′t reduce pregnancy rates .

BACKGROUND: Many studies have investigated the association between Glutathione S-Transferase M 1 (GSTM1) null genotype and risk of male infertility, but the impact of GSTM1 null genotype on infertility risk is unclear owing to the obvious inconsistency among those studies. This study aimed to quantify the strength of association between GSTM1 null genotype and risk of male infertility. METHODS: We searched the PubMed, Embase and Wangfang databases for studies investigating the association between GSTM1 null genotype and risk of male infertility. We estimated pooled odds ratio (OR) with its 95 % confidence interval (95 % CI) to assess this possible association. RESULTS: Twelve case-control studies with 1, 589 infertility cases and 1, 537 controls were included. Meta-analysis of total 12 studies showed that GSTM1 null genotype was associated with increased risk of male infertility (OR = 1.34, 95%CI 1.02-1.77, P = 0.036). In subgroup analysis of Caucasians, there was also an obvious association between GSTM1 null genotype and increased risk of male infertility (OR = 1.51, 95%CI 1.11-2.05, P = 0.006). Sensitivity analyses by sequential omission of individual studies or omitting studies without high quality did not significantly alter the overall pooled OR. Cumulative meta-analysis further showed a trend of more obvious association as information accumulated. No evidence of publication bias was observed. CONCLUSION: Meta-analyses of available data suggest that GSTM1 null genotype contributes to increased risk of male infertility.
