ABSTRACT
BACKGROUND:The establishment of animal models of Parkinson’s disease plays an important role in clinical and basic experiments of Parkinson’s disease, and its stability directly affected study results. OBJECTIVE:To evaluate the behavior and pathological changes of rats with Parkinson’s disease established by unilateral two-point injection with 6-hydroxydopamine. METHODS:A total of 62 Sprague-Dawley rats were randomly assigned into experimental group (n=50) and normal group (n=12). Using stereotatic technique, 6-hydroxydopamine was injected into the right substantia nigra RESULTS AND CONCLUSION:At 2 weeks, after induction with apomorphine, left rotation speed>7 r/min was observed in only 22 rats of the experimental group. No significant difference in the number of rotation between 2 and 4 weeks was detected (P>0.05). Tyrosine hydroxylase and dopamine content significantly reduced in the model group. Results indicated that unilateral two-point injection of 6-hydroxydopamine could be used to successful y establish rat models of Parkinson’s disease with similar behaviors and pathologies.
