Imposex is a phenomenon widely associated with environmental exposure to organotin compounds which were quite common components of antifouling paints applied on boats and ship hulls. Here we study the incidence of imposex in neogastropods and its relation with water quality and maritime traffic in the coastal strip of Santa Marta, Colombia. Imposex was determined via specialized indexes and related to the organisms' size, somatic conditions, variables of water quality and maritime traffic, in a space-time assessment. There was evidence of imposex in five species Plicopurpura patula, Vasula deltoidea, Stramonita haemastoma, S. floridana, and Gemophos auritulus. Purpura patula and Vasula deltoidea species were found in all sampling sites. The results have proved that imposex is highly influenced by the maritime traffic variable, with greater prevalence during the dry season, and with P. patula being more sensitive than V. deltoidea.

Resumo Imposex é o desenvolvimento de características sexuais masculinas causadas por poluentes tóxicos de alguns produtos químicos que atuam como desreguladores endócrinos. Tintas anti-incrustantes são as que contêm estes produtos químicos. Cartagena carece de estudos para indicar a extensão do imposex nas suas águas costeiras. O objetivo deste estudo foi determinar a prevalência de imposex no gastrópode Stramonita haemastoma em Cartagena, Colômbia. Os espécimes foram coletados durante 2013 de locais de alta e baixa influência da atividade portuária. Foram registradas as medidas morfométricas e a frequência da ocorrência do imposex. A comparação entre as variáveis morfométricas mostrou diferenças estatisticamente significantes entre os dois locais estudados. Além disso, as fêmeas da espécie S. haemastomaapresentaram uma frequência de imposex de 93,1% na Ilha das Aves, Baía das Cartagena, em comparação com 31,8% em La Bocana. O índice do comprimento relativo do pênis ou RPLI (10,145 e 3,231) e o índice da sequência do vaso deferente ou VDSI (2,83 e 1,16), mostraram possível contaminação por compostos organoestânicos em ambos os locais.

Imposex is the superimposition of non-functional male sex organs in gastropod females. This syndrome is a hormonal imbalance induced by tributyltin (TBT) which have been used in antifouling paints formulation. The present study aimed to perform an integrated environmental assessment of imposex and butyltin (BT) contamination using surface sediments and tissues of Thaisella chocolata (an edible gastropod) from northern Chile. The results showed imposex incidence in 11 out of 12 sites. In the most contaminated sites, which are areas under the influence of maritime activities, and also used for fishing and aquaculture, RPLI were over 60 and VDSI over 4 (high incidence of sterile females). Exceptionally high contamination levels and evidences of fresh inputs of tributyltin (TBT) were detected along the studied area. TBT levels above 300 and 90ngSng-1, respectively, were recorded in sediments and edible gastropod tissues of 6 sites. Thus, a daily ingestion of 90 to 173g of T. chocolata foot (4 to 8 organisms) from the most contaminated sites will certainly lead to the consumption of BT exceeding the tolerable daily intake recommended by European Food Safety Authority. It is reasonable to consider that human risk is even higher if daily consumption of additional seafood is considered. Moreover, some contaminated sites were located within the marine reserve "Isla Grande Atacama", indicating that even marine protected areas are under the influence of TBT contamination. These findings suggest that current levels of TBT in the studied area are sufficient to induce harmful effects on the environment and constitutes a potential threat to seafood consumers. Thus, national regulatory actions toward environmental protection and food safety of local populations are still mandatory, even after 8years of the TBT global ban by IMO.

Abstract This study investigated the effects of tributyltin (TBT) on the morphology of the genital system of the gastropod Stramonita rustica in southern Bahia, Brazil. For this, 330 specimens were collected during the summer of 2014 at eight sampling points to ascertain whether male sex organs had developed in addition to the complete female genital tract in females (= imposex). The analyses were made under a stereoscopic microscope. Imposex and their associated indexes, and the sterile females, exhibited the highest rates in harbors and shipyards areas. Despite the total ban of TBT in anti-fouling paints on a global scale since 2003, the results of this and other studies indicate the continued use of those paints on the Brazilian coast. This shows the inefficiency of existing legislation and the need to strengthen enforcement of the ban.


Resumo Este estudo investigou os efeitos de tributil-estanho (TBT) sobre a morfologia do sistema genital do gastrópode Stramonita rustica (Lamarck, 1822) no Litoral Sul da Bahia, Brasil. Para isso, 330 espécimens foram coletados durante o verão de 2014 em oito pontos de amostragem, para verificar se os órgãos sexuais masculinos tinham se desenvolvido para além do trato genital feminino completo (= imposex). As análises foram feitas sob um microscópio estereoscópico. O imposex e seus índices associados, assim como as fêmeas estéreis, apresentaram as maiores taxas em áreas portuárias e estaleiros. Apesar da proibição total do TBT em tintas anti-incrustantes em escala global desde 2003, os resultados deste e de outros estudos indicam a continuidade de uso dessas tintas na costa brasileira. Isso mostra a ineficácia da legislação vigente e reforça a necessidade de se cumprir integralmente o banimento.

A spatiotemporal evaluation of butyltin contamination was performed between 2010 and 2012 along Todos os Santos Bay (Northeast Brazil) using surface sediments, bivalve tissues (Anomalocardia brasiliana and Mytella guyanensis), and imposex occurrence (Stramonita rustica). The spatial study detected high tributyltin (TBT) levels (maximum values of 262 ng Sn g (-1) - 21,833 ng Sn g(-1) of total organic carbon - for surface sediments and 421 ng Sn g(-1) for bivalve tissues) in the innermost part of the bay. The TBT levels detected in M. guyanensis tissues might cause human health risk since local population consumes these organisms. These high concentrations observed in the bivalves might result in ingestions higher than the safe limits established by European Food Safety Authority (250 ng TBT kg(-1) day(-1)). Considering the temporal evaluation, no difference (p > 0.05) was observed between TBT concentrations in sediments obtained during the two sampling campaigns (2010/2011 and 2012). However, the increasing predominance of TBT metabolites (butyltin degradation index (BDI) >1) in more recent sediments indicates further degradation of old TBT inputs. In spite of that, recent inputs are still evident at this region. Nevertheless, a reduction of imposex parameters in S. rustica over the last decade suggests an overall decline in the TBT contamination, at least in the outermost and possible less impacted region of the bay. The TBT contamination is probably reducing due to the national and international legislative restrictions on the use of TBT as antifouling biocide. The contamination levels, however, are still relevant especially in the inner part of Todos os Santos Bay since they are above those that are likely to cause toxicity to the biota.

Pulp and paper mill effluents (PPMEs) have been shown to increase gonad size, cause early maturation, and disrupt hormone functions in native and non-native Chilean fish. In this study, we assessed reproductive (plasma vitellogenin; VTG, gonad development) and metabolic (ethoxyresorufin-O-deethylase activity; EROD) end points, relative liver size (LSI) and condition factor (K) of juvenile female and male rainbow trout exposed to effluents. Unlike previous studies, which have focus either on the specific effects of effluent on fish in laboratory exposures or biotic population statuses downstream of discharge sites, we simultaneously assessed the impacts of PPMES on trout using two approaches: (1) laboratory exposures of tertiary treated PPME produced from processing Eucalyptus globulus or Pinus radiata; and (2) in situ bioassay downstream of the combined discharge of the same pulp mill. Despite an increase in the average gonadosomatic index (GSI) in exposed fish, no statistical differences in gonad size between exposed and unexposed individuals was detected. However, both female and male fish exposed to effluents showed significantly higher concentrations of plasma VTG, so more in fish exposed to Eucalyptus-based effluent when compared to Pinus PPME. In addition, male fish showed intersex characteristics in all exposure assays (Eucaliptus and Pinus) and, despite the low concentration of effluent in the river (<1% [v/v]), similar responses were observed in the caged fish. Finally, EROD activity was induced in both in situ exposures and laboratory assays at the higher PPME concentration (60-85% PPME). This study confirms estrogenic effects in Chilean fish exposed to PPME and the necessity for biological effects monitoring in addition to the assessment of physical-chemical endpoints as required in current government regulations.

Voluta musica is a dioecious marine gastropod endemic of the South Caribbean. Tributyltin (TBT) and copper (Cu) are potential inducers of imposex, an endocrine disorder by which females develop a penis and/or vas deferens. The goal of this work was to determine the imposex incidence in V. musica populations from Northeastern Peninsula de Araya. For this, we selected three sites (Isla Caribe, Isla Lobos and Bajo Cuspe) and made monthly samplings of 15 snails in each site, during one year, and determined: (1) sizes; (2) sex and imposex incidence and (3) the Relative Penis Length Index (RPLI). We also performed histological analysis of the gonads, and measured TBT and Cu concentrations in sediments from the studied localities. Our results showed that the total number of sampled females affected by imposex was 24.5% at Isla Caribe, 12% at Isla Lobos, and none at Bajo Cuspe. In sediments, Cu was detected mostly in Isla Lobos. The female gonads with imposex did not show any development of male cells in any of the sampled sites. The higher percentage of females with imposex matched with the higher boat traffic locality, and higher TBT level (Isla Caribe). No esterilization was evident in this work, nevertheless, the presence of TBT and Cu in the sediments and females with imposex were considered as a potential threat to V. musica populations in this region. In Venezuela there is no control over this particular issue, possibly because of the lack of information and research in this topic, but certainly, this information will be useful in biodiversity conservation policies.

Voluta musica es un gasterópodo dioico endémico del Caribe sur. El TBT y el Cu, son potenciales causantes del imposex, fenómeno donde las hembras desarrollan un pene y/o vaso deferente. El objetivo fue determinar la incidencia de imposex en V. musica en el noreste de la Península de Araya. Se seleccionaron tres localidades y se captura-ron mensualmente 15 individuos durante un año para determinar: (1) talla de los individuos; (2) sexo y presencia de imposex; (3) índice Largo Relativo del Pene (RPLI). Se realizó histología de la gónada de los individuos. Se determinó TBT y Cu en el sedimento de cada localidad. En Isla Caribe, el 24.5% de las hembras presentó imposex, y se halló 3.9ngSn/g de TBT; en Isla Lobos, el 12% de las hembras desarrollaron imposex; en Bajo del Cuspe no se observó imposex. Se halló Cu en mayor concentración en Isla Lobos. Las gónadas femeninas con imposex no demos-traron masculinización. El mayor porcentaje de imposex coincide con la localidad de mayor tráfico de embarcacio-nes y con mayor nivel de TBT (Isla Caribe). No se eviden-ció esterilización, sin embargo la presencia de TBT, Cu e imposex son potenciales amenazas para las poblaciones de V. musica en la región. Hasta ahora, en Venezuela no se está tomando ninguna medida de control sobre este tema en particular, posiblemente por la escasez de información y orientación de las investigaciones hacia este tema, pero que sin duda se debería tomar en cuenta en las políticas para la conservación de la biodiversidad.

Environmental agencies have identified a growing number of environmental contaminants that have endocrine disrupting activity, and these can become a major public health problem. It is suggested that endocrine disruptors could account for the higher-than-expected increase in the prevalence of some non-communicable diseases, such as obesity, diabetes, thyroid diseases, and some cancers. Several endocrine Disrupting Chemicals (EDCs), such as pesticides, bisphenol A, phthalates, dioxins, and phytoestrogens, can interact with the female reproductive system and lead to endocrine disruption. Initially, it was assumed that EDCs exert their effects by binding to hormone receptors and transcription factors, but it is currently known that they may also alter the expression of enzymes involved in the synthesis or catabolism of steroids. Biomonitoring studies have identified these compounds in adults, children, pregnant women, and fetuses. Among the diseases of the female reproductive tract associated with EDCs exposure are the following: precocious puberty, polycystic ovary syndrome, and premature ovarian failure. The different populations of the world are exposed to a great number of chemicals through different routes of infection; despite the various available studies, there is still much doubt regarding the additive effect of a mixture of EDCs with similar mechanisms of action.

As diversas agências de controle ambiental têm identificado um crescente número de contaminantes ambientais que apresentam atividade de desregulador endócrino e estes poderão se tornar um dos maiores problemas de saúde pública. Sugere-se que os Desreguladores Endócrinos (EDCs) poderiam justificar o aumento na prevalência de algumas doenças não transmissíveis acima do esperado, como, por exemplo, obesidade, diabetes, doenças tireoidianas e alguns tipos de cânceres. Vários EDCs, como pesticidas, bisfenol A, ftalatos, dioxinas e fitoestrógenos, podem interagir com o sistema reprodutivo feminino e levar à desregulação endócrina. Inicialmente, supunha-se que os EDCs exercessem seus efeitos através da ligação com receptores hormonais e fatores de transcrição, mas, atualmente, sabe-se que também podem alterar a expressão de enzimas envolvidas na síntese ou no catabolismo dos esteroides. Estudos de biomonitoramento têm identificado esses compostos em adultos, crianças, gestantes e em fetos. Entre as patologias do trato reprodutor feminino associadas à exposição aos EDCs, destacam-se: puberdade precoce, síndrome dos ovários policísticos e falência ovariana prematura. As diversas populações estão expostas a um grande número de substâncias químicas, através de diferentes vias de contaminação. Apesar dos diferentes estudos disponíveis, ainda permanece uma grande dúvida sobre o efeito aditivo de uma mistura de EDCs com similar mecanismo de ação.

The objective of the present work was to study the effect of a low dose of bisphenol A (BPA), on the reproductive axis of prepuberal male rats exposed to the endocrine disruptor (ED) during gestation and lactation period. Wistar-mated rats were treated with either 0.1 % ethanol or BPA in their drinking water until their offspring were weaned at the age of 21 days. The estimated average dose of exposure to dams was approximately 3 µg/kg/day of BPA. The pups were sacrificed on the 35th day of life. Body weight was measured during the development and at the moment of the sacrifice; testicular and seminal vesicles weight and their respective relative weights were also measured. LH, FSH and testosterone were determined and histological studies of testicular tissue were also performed. Body weight at the moment of the sacrifice was significantly higher in the group exposed to BPA; testicular weight decreased significantly; seminal vesicles weight and relative weights of testes and seminal vesicles were not modified by treatment. LH and FSH serum levels increased significantly after treatment, meanwhile testosterone showed no significant changes. Histological studies showed the lumen of seminal tubes reduced by the presence of immature cells of the spermatic lineage. Our results suggest that pre- and early postnatal exposure to a low dose of BPA disrupts the normal function of the reproductive axis in prepuberal male rats. The effects of the ED may be exerted at different levels of the axis and may be dependent on the dose, manner of administration, and the moment of exposure to the disruptor.

This is the first report on imposex occurrence in Pugilina morio. Imposex levels in P. morio from Ceará River Estuary were assessed by a non-lethal method. The obtained imposex parameters were: percentage = 37.1 %, female penis length index (FPLI) = 0.93 mm, relative penis length index (RPLI) = 6.5 %, and vas deferens sequence index (VDSI) = 1.0. Because imposex is induced by tributyltin, and P. morio is widely distributed on the East coast of South America, the species can be used as a tributyltin contamination sentinel in these estuaries. Additionally, a non-lethal method of evaluating imposex minimized the impact on the population of P. morio from sample collection, as all organisms were returned to the environment.

The imposex incidence was appraised in South American gastropods, considering the scenario before and after the global ban of tributyltin (TBT). A statistically significant reduction in imposex indexes was observed in Stramonita haemastoma collected in 2006 and 2010 from areas under the influence of four coastal harbors from southern Brazil. This reduction may be because of the effectiveness of the global ban issued by the International Maritime Organization, although the restrictions on TBT-based antifouling paints in Brazil might also have helped. Even so, a residual organotin contamination was still detected in female tissues (levels ranged from 7.6 to 164.9 ng Sn/g for TBT; from <2 to 214.5 ng Sn/g for dibutyltin; from <3.5 to 178.8 ng Sn/g for monobutyltin; and from <1.5 to 53 ng Sn/g for triphenyltin). Thus, although a reduction in imposex and environmental levels of organotins is expected in every ocean worldwide soon after the implementation of national and international restriction regulations, this will depend on the effectiveness of the global TBT ban; the effectiveness of local restrictions on producing, selling, and using TBT-based antifouling paints; and specific characteristics of local sediments, because metabolization rates and sorption/desorption of TBT previously deposited might affect its environmental bioavailability. Therefore, the reduction trend detected in the present study cannot be extrapolated to other Brazilian or South American coastal areas.

The first appraisal of butyltin (BT) contamination linked with imposex incidence in Peruvian coastal areas was performed in the present study. Imposex occurrence and BT levels in female bodies were analyzed in the rock snail Thais chocolata distributed along 10 sites in areas under the influence of Callao and Chimbote Harbors. Imposex levels associated to high tributyltin (TBT) concentrations were observed in six of 10 studied sites, and the highest imposex levels were seen in gastropods collected near the harbor terminals. Tributyltin concentrations were the highest reported for gastropod mollusks (up to 662 ng Sn g(-1)) in recent studies. Although TBT-based antifouling paints have been banned globally (International Maritime Organization, 2008), the combination of high imposex and TBTs level with butyltin degradation index values indicated fresh TBT inputs in these Peruvian coastal areas. Thus, the need to implement local regulatory strategies related to the use of TBT-free antifouling paints is urgent, as is a thorough assessment of the effectiveness of the TBT global ban. Moreover, the wide distribution of T. chocolata combined with its capability to elicit imposex and bioaccumulate BTs renders this a good sentinel species for TBT contamination along the western South American coast.

The first appraisal of butyltins (BT) contamination linked with imposex incidence in the Pacific coast of South America was performed in the current study. Imposex occurrence was analyzed in three muricid species (Thais biserialis, T. brevidentata, and T. kiosquiformis) distributed along 14 sites from the coastal areas of Ecuador. All studied muricids species showed imposex, which was observed in 11 out of 14 sites. The highest imposex levels were observed in T. kiosquiformis collected inside the Gulf of Guayaquil [relative penis length index (RPLI) 9.0-40.7], whereas RPLI values for T. biserialis and T. brevidentata ranged from 4.0 to 26.1 and 0.0 to 1.9, respectively. Additionally, BT compounds [tributyltin (TBT), dibutylin (DBT), and monobutylin (MBT)] were determined in surface sediments collected in 6 of the sites (main harbor areas), and values ranged between 12.7 and 99.5 ng Sn g(-1) (dry weight) for TBT, 1.8 and 54.4 ng Sn g(-1) for DBT, and 44 and 340 ng Sn g(-1) for MBT. Although BT degradation indexes suggested an aged input of TBT, the high imposex levels detected in the current study suggest that international restrictions on TBT based antifouling paints were still not effective in Ecuador. Considering that the southern coast, especially nearby harbors inside the Gulf of Guayaquil, have shown to be contaminated by BTs, a combination of T. kiosquiformis and T. biserialis (the most sensitive species) can be used for future biomonitoring of Ecuador coastal areas.

Organotin compounds (OTs) were used in antifouling paints for more than four decades. However, due to their widespread intensive use and high toxicity, undesirable effects in non-target marine organisms have been detected since the early 1980s. Consequently, the International Maritime Organization banned new maritime applications of these products on January 1, 2003 and their presence on ship hulls from January 1, 2008. Although extensively studied in Europe, North America, Oceania, and Asia, environmental levels and effects of organotin contamination are still poorly known for South America. Thus, the current review aimed to present the actual status of this problem in South America by summarizing and comparing the available data in the literature. An overview of the OTs concentrations in sediment and biota and their effects, mainly imposex in marine gastropods, are presented. This work showed that in Atlantic coastal areas of South America there are "hot spots" of OTs contamination, similar to that observed in industrialized countries of Northern Hemisphere. On the other hand, the number of accomplished studies in the Pacific coast is extremely low. Despite the limitation on studies about OTs environmental levels and their related effects, the available data pointed out for a widespread TBT contamination along the South American coastal areas. Therefore, the establishment of baselines of organotin contamination in the Pacific coast and the implementation of temporal trend studies in the South American coastal areas is crucial to verify the effectiveness of local regulations and OTs global ban, and to map the most sensitive areas related to present and future antifouling impacts.

The aim of this study was to assess the health indicators and reproductive endpoints in a wild population of lambari Astyanax fasciatus, a small characid fish widely distributed in South America. A range of biomarkers, from the molecular to population levels, was evaluated in adult fish sampled from five sites from the Furnas Reservoir, Grande River. At the sites that receive untreated agriculture and industrial residues, fish showed reduced body size and delayed gonadal maturation. Fish from the sites located immediately downstream from the municipal wastewater discharges exhibited feminisation, intersex and evidence of contamination by xenoestrogens. Elevated levels of zona radiata proteins were found by Western blot in the livers of male fish from three contaminated sites. Overall, this study provides the first evidence of endocrine disruption in a wild fish population inhabiting the Furnas Reservoir, and suggests water contamination by oestrogenic compounds and agricultural residues with a possible risk to human health and wildlife.

OBJECTIVE: This study investigates the effects of prenatal lipopolysaccharide (LPS) exposure on the maternal behavior of pregnant rats and the physical development and sexual behavior of their male offspring in adulthood. METHODS: For two experiments, pregnant rats were injected with LPS (250 microg/kg, i.p.) on gestation day (GD) 21. In the first experiment, the maternal behavior (postnatal day, PND, 6) and the dam's open-field general activity (PND7) were evaluated. In the second experiment, the maternal pre- and postnatal parameters, the pup's development, the offspring's sexual behavior in adulthood, and the pup's organ weights were assessed. RESULTS: Compared to the control group, the LPS-treated dams presented reduced maternal behavior, decreased general activity, a smaller body weight difference between GD21 and PND1, a greater number of perinatal deaths, and smaller litters. For the male pups, LPS treatment resulted in a decreased body weight on PND2, whereas the anogenital distance and the day of testis descent were not modified. The male sexual behavior was impaired by prenatal LPS. Particularly the number of ejaculating animals was reduced. The testis weight was also lower in the prenatally LPS-treated rats than in the control rats. CONCLUSION: We propose that prenatal LPS exposure on GD21 acts as an imprinting factor that interferes with the programming of brain sexual determination in offspring.
