Cadernos Técnicos de Medicina Veterinária e Zootecnia: Digitalização e disponibilização on-line da coleção integral nos 90 anos da Escola de Veterinária da UFMG pixabay.com das atividades bibliográficas e documentais necessárias ao desenvolvimento do ensino, da pesquisa, da extensão, da inovação e da gestão da universidade. A Biblioteca Professor Ivan Barbosa Machado Sampaio da Escola de Veterinária da UFMG5 é uma combinação orgânica de pessoas, recursos, coleções locais e virtuais, que tem como propósito o apoio aos usuários no processo de transformação da informação em conhecimento. Resumo Este estudo relata a estratégia para aprimorar o portfólio como instrumento de avaliação da aprendizagem em uma disciplina oferecida aos estudantes do quarto ano de graduação em Medicina, por meio do portfólio on-line. A disciplina é constituída de atividades práticas de atendimento integral à saúde da criança, do adolescente, do adulto e da mulher na atenção primária do município de Campinas. A utilização do portfólio permite que os estudantes reflitam sobre os atendimentos realizados em relação ao conhecimento, à habilidade clínica e ao vínculo médico-paciente na consulta. O portfólio on-line possibilitou rever os critérios de avaliação, agilizar e facilitar a gestão da correção, realizada por oitenta avaliadores de três especialidades médicas. O acesso às informações do portfólio na plataforma moodle permitiu a rápida tomada de decisão em aspectos que precisam ser adequados para a melhoria do processo ensino-aprendizagem.(AU)


Abstract This study reports on the strategy to improve the use of online portfolios as a learning assessment instrument of a discipline taught to fourth-year medical students. The discipline comprises practical activities of comprehensive care of children, teenagers, adults, and women in primary care in the Brazilian city of Campinas. Using portfolios, students are able to reflect upon the provided care as to the knowledge, clinical skills, and doctor-patient bond shown in the appointment. The online portfolio enabled to review the assessment criteria, and streamline and facilitate managing the assessment conducted by 80 evaluators from three medical specialties. Access to the portfolio's information in the Moodle platform enabled a fast decision-making process in aspects that need to be adequate to improve the teaching and learning process.(AU)


Resumen Este estudio relata la estrategia para perfeccionar el portafolio como instrumento de evaluación del aprendizaje en una disciplina ofrecida a los estudiantes del 4º. año de la graduación en medicina, por medio del portafolio online. La asignatura se compone de actividades prácticas de atención integral a la salud del niño, del adolescente, del adulto y de la mujer en la atención primaria del municipio de Campinas. La utilización del portafolio permite que los estudiantes reflexionen sobre las atenciones realizadas con relación al conocimiento, a la habilidad clínica y al vínculo médico-paciente en la consulta. El portafolio online posibilitó revisar los criterios de evaluación, agilizar y facilitar la gestión de la corrección realizada por 80 evaluadores de tres especialidades médicas. El acceso a las informaciones del portafolio en la plataforma Moodle permitió la rápida toma de decisiones en aspectos que tienen que adecuarse para la mejora del proceso enseñanza-aprendizaje.(AU)

A fully automated on-line system for monitoring the photocatalytic degradation of herbicides was developed using multisyringe flow injection analysis (MSFIA) coupled to a solid phase extraction (SPE) unit with UV detection. The calibration curves were linear in the concentration range of 100-1000 µg L(-1) for 3,6-dichloro-2-methoxybenzoic acid (dicamba) and 500-3000 µg L(-1) for 2,4-dichlorophenoxyacetic acid (2,4-D), while the detection limits were 30 and 135 µg L(-1) for dicamba and 2,4-D, respectively. The monitoring of the photocatalytic degradation (TiO2 anatase/UV 254 nm) of these two herbicides was performed by MSFIA-SPE system using a small sample volume (2 mL) in a fully automated approach. The degradation was assessed in ultrapure and drinking water with initial concentrations of 1000 and 2000 µg L(-1) for dicamba and 2,4-D, respectively. Degradation percentages of approximately 85% were obtained for both herbicides in ultrapure water after 45 min of photocatalytic treatment. A similar degradation efficiency in drinking water was observed for 2,4-D, whereas dicamba exhibited a lower degradation percentage (75%), which could be attributed to the presence of inorganic species in this kind of water.

A new automated, sensitive, and fast system for the simultaneous online isolation and preconcentration of lead and strontium by sorption on a microcolumn packed with Sr-resin using an inductively coupled plasma mass spectrometry (ICP-MS) detector was developed, hyphenating lab-on-valve (LOV) and multisyringe flow injection analysis (MSFIA). Pb and Sr are directly retained on the sorbent column and eluted with a solution of 0.05 mol L(-1) ammonium oxalate. The detection limits achieved were 0.04 ng for lead and 0.03 ng for strontium. Mass calibration curves were used since the proposed system allows the use of different sample volumes for preconcentration. Mass linear working ranges were between 0.13 and 50 ng and 0.1 and 50 ng for lead and strontium, respectively. The repeatability of the method, expressed as RSD, was 2.1% and 2.7% for Pb and Sr, respectively. Environmental samples such as rainwater and airborne particulate (PM10) filters as well as a certified reference material SLRS-4 (river water) were satisfactorily analyzed obtaining recoveries between 90 and 110% for both elements. The main features of the LOV-MSFIA-ICP-MS system proposed are the capability to renew solid phase extraction at will in a fully automated way, the remarkable stability of the column which can be reused up to 160 times, and the potential to perform isotopic analysis.

An hybrid mesoporous material synthesised in our laboratories for solid phase extraction (SPE) in flow through systems has been used for analytical purposes. The solid was obtained from mesoporous silica MCM-41 functionalized with 3-aminopropyltriethoxy silane by Sol-Gel methodology. In order to exploit the large sorption capacity of the material together with the possibility of modeling it for anions retention, a microcolumn (MC) filled with the solid was inserted in a flow system for preconcentration of Cr(VI) and its determination at ultratrace levels in natural waters. The analytical methodology involved a reverse flow injection system (rFI) holding a MC filled with the solid for the analyte extraction. Elution and colorimetric detection were carried out with 1-5 diphenylcarbazide (DPC) in sulfuric acid. DPC produced the reduction of Cr(VI) to Cr(III) together with the generation of a cationic red complex between Cr(III) and 1-5 diphenylcarbazone which was easily eluted and detected with a visible spectrophotometer. Moreover, the filling material got ready for the next sample loading remaining unspoiled for more than 300 cycles. The effect of several variables on the analytical signal as well as the influence of cationic and anionic interferences were discussed. Particular attention was given to sulfuric acid interference since it is the required media for the complex generation. Under optimal conditions, 99.8% of Cr(VI) recovery was obtained for a preconcentration time of 120s (sample and DPC flow rates=1 mL min(-1)) and an elution volume of 250 micro L. The limit of detection (3s) was found to be 0.09 microg L(-1) Cr(VI) with a relative standard deviation (n=10, 3 microg L(-1)) of 1.8. Since no Cr(III) was retained by the solid material and Cr(VI) was completely adsorbed, electrothermal atomic absorption spectrometry (ET AAS) determinations of Cr(III) were also performed by simply measuring its concentration at the end of the microcolumn after Cr(VI) retention by the mesoporous solid. Applications to the determination of Cr(VI) and Cr(III) in natural waters and the validation of the methodology were also studied.

Biomass is an important variable in biosurfactant production process. However, such bioprocess variable, usually, is collected by sampling and determined by off-line analysis, with significant time delay. Therefore, simple and reliable on-line biomass estimation procedures are highly desirable. An artificial neural network model (ANN) is presented for the on-line estimation of biomass concentration, in biosurfactant production by Candida lipolytica UCP 988, as a nonlinear function of pH and dissolved oxygen. Several configurations were evaluated while developing the optimal ANN model. The optimal ANN model consists of one hidden layer with four neurons. The performance of the ANN was checked using experimental data. The results obtained indicate a very good predictive capacity for the ANN-based software sensor with values of R2 of 0.969 and RMSE of 0.021 for biomass concentration. Estimated biomass using the ANN was proved to be a simple, robust and accurate method.

Calorimetry and other on-line techniques are used for the first time as complement to the traditional off-line methods in order to follow the growth of the green Chlorella vulgaris microalgae. A 2-L photo-bio-reactor was adapted from a commercial calorimeter used previously to study heterotrophic microbial growth. An external source of light was added to favor the photosynthesis of the autotrophic cells. Heterotrophic growth was also tested with external glucose in the broth. A third mode, mixotrophic, allowed faster autotrophic plus heterotrophic growth. Calorimetric measurements were performed considering the corresponding calibrations in order to consider only the energy involved during the microalgal growth. The three different modes of Chlorella cultures were energetically characterized. Besides calorimetry, the weight of diluted nitric acid added to maintain the pH of the culture was correlated with the cellular growth and the nitrogen composition of the algae. Additionally, the on-line infrared spectroscopy proved to be an efficient technique to follow the composition of the broth in glucose, nitrates, and phosphates. These results were compared and complemented with some classic off-line techniques used to track this kind of cultures.

A novel strategy for exploiting ion exchange in sequential injection systems is proposed. The procedure is based on the selection of a defined volume of a resin suspension, which is introduced and packed in the analytical path, establishing a resin mini-column in the system. The passage of a selected sample volume through the resin mini-column leads to the retention of the analyte, while the sample matrix is discarded. The analyte is eluted during the passage of the eluant/reagent by the packed beads, being the analytical signal monitored (absorbance) in the liquid phase. The beads are then aspirated back to the holding coil and directed to a recovery flask, linked at the selection valve; then the system is ready to begin a new cycle. With the proposed strategy, the main characteristics of the sequential injection system are kept as any new artifact is added to the manifold and system reconfiguration is not required. The feasibility of the approach is demonstrated by the phytic acid determination in food samples. For this specific application, AG1-X8 was selected as ion exchanger, and a solution containing Cl- and Fe(III)-salicylate complex was used as eluant and spectrophotometric reagent.

Sistema de detección en línea de Escherichia coli O157: H7: Un nuevo apoyo para la Salud Pública.Introducción. Motivado por los casos innumerables de infecciones de diarrea que frecuentemente ocurren, en los países en vías de desarrollo, se calibró un biosensor a fibra optica con intensidad modulada usando como ejemplar Escherichia coli O157: H7. Material y métodos. La sensibilidad del dispositivo se ha calibrado para las unidades formadoras de colonia (CFU) de 10 a 800 rindiendo un error más pequeño que 11 por ciento.Resultados. Se obtuvo una correlación entre la respuesta óptica y el número instantáneo de bacterias. El signo del rendimiento óptico fue 0.016 (ñ 0.001) dB por hora por bacteria. En todo los casos la detección empezó después de 270 ñ 4 minutos, 5-10 veces más rápido que las técnicas bacteriológicas convencionales.Discusión. Los resultados obtenidos pueden contribuir al problema de monitoreo de patógenos que representen problemas en el campo de la salud pública.

A system for on-line spike detection and analysis based on an IBM PC/AT compatible computer, written in TURBO PASCAL 6.0 and using commercially available analog-to-digital hardware is described here. Spikes are detected by an adaptive threshold which varies as a function of signal mean and its variability. Since the threshold value is determined automatically by the signal-to-noise ratio analysis, the user is not actively involved in controlling its level. This program has been reliably used for the detection and analysis of the spike discharge of vestibular system afferent neurons. It generates the interval-joint distribution graph, the interval histogram, the autocorrelation function, the autocorrelation histogram, and phase-space graphs, thus, providing a complete set of graphical and statistical data for the characterization of the dynamics of neuronal spike activity. Data can be exported to other software such as Excel, Sigmaplot and MatLab, for example.

Methods for the analysis of phenformin and its metabolite by high-performance liquid chromatography (HPLC), capillary electrophoresis (CE) and high-performance liquid chromatography-electrospray ionization mass spectrometry (HPLC-ESIMS) are developed. The effects of pH, buffer concentration and proportion of organic modifier on the retention of the compounds in HPLC have been studied. The optimum condition was used for the separation and identification of phenformin and its metabolite in microsomal metabolism by HPLC-ESIMS. A simple CE method is also described for the separation of these compounds. Optimum incubation conditions and cofactor requirements for the formation of 4-hydroxyphenformin by microsomal preparations of rat liver were determined. A linear response in the formation of product was found with increasing concentrations of protein and up to 15 min incubation. High concentrations of phenformin inhibited its metabolite formation, and K(m) was 4 microM.
