Resveratrol, a natural polyphenol found in tempeh, has not been investigated especially in vitro as a neuroprotective agent against 2-methoxyethanol (2-ME)-induced beta-amyloid cytotoxicity. Beta amyloid peptides (Aβ) could initiate neurotoxic events and neuron-inflammatory response via microglial activation. However, it remains unknown whether the neurotoxic effect of beta-amyloid and/or associated with the potential of 2-ME to induce neurotoxic effects on primary culture of nerve cells induced by 2-ME. This study investigated potential neuroprotective of trans-resveratrol a promising agent tempeh and soybean seed coats-derived against beta amyloid cytotoxicity on primary culture of nerve cells induced by 2-methoxyethanol. Biotium and MTT assays were used to analyze neurons, which were isolated from the cerebral cortex of fetal mice at gestation day 19 (GD-19). A standard solution of 2-methoxyethanol was dosed at 10 μL. The cultured cells were randomly divided into the following groups: (1) 2-ME group + resveratrol standard, (2) 2-ME group + resveratrol isolated from tempeh, (3) 2-ME group + resveratrol isolated from soybean seed coats, and (4) the control group, without the addition of either 2-ME or resveratrol. Exposure of the primary cortical neuron cells to beta-amyloid monoclonal antibody pre-incubated for 24 h with 10 µL of 4.2 µg/mL resveratrol and 7.5 mmol/l 2-methoxy-ethanol additions. Here, we report that the addition of 2-ME and resveratrol (standard and isolated from tempeh) of cell culture at concentrations of 1.4, 2.8 and 4.2 µg/mL showed that the majority of neurons grew well. In contrast, after exposure to 2-ME and Beta-amyloid, showed that glial activated. These findings demonstrate a role for resveratrol in neuroprotective-neurorescuing action.(AU)


O resveratrol, um polifenol natural encontrado em tempê, não foi investigado apenas in vitro como agente neuroprotetor contra a citotoxicidade beta-amiloide induzida por 2-metoxietanol (2-ME). Os peptídeos beta amiloides (Aβ) podem iniciar eventos neurotóxicos e resposta inflamatória dos neurônios via ativação microglial. No entanto, permanece desconhecido se o efeito neurotóxico do peptídeo beta-amiloide associado ao potencial do 2-ME causa efeitos neurotóxicos na cultura primária de células nervosas induzidas pelo 2-ME. Este estudo investigou o potencial neuroprotetor do agente trans-resveratrol em cascas de sementes de soja e tempê derivadas da citotoxicidade beta-amiloide na cultura primária de células nervosas induzidas pelo 2-metoxietanol. Ensaios de biotium e MTT foram utilizados para analisar os neurônios isolados do córtex cerebral de camundongos fetais no dia da gestação 19 (GD-19). As células cultivadas foram divididas aleatoriamente nos seguintes grupos: (1) grupo 2-ME + padrão de resveratrol; (2) grupo 2-ME + resveratrol isolado de tempê; (3) grupo 2-ME + resveratrol isolado de cascas de sementes de soja; e (4) grupo controle, sem a adição de 2-ME ou resveratrol. Houve exposição das células primárias dos neurônios corticais ao anticorpo monoclonal beta-amiloide pré-incubado por 24 horas, com 10 µL de 4,2 µg/mL de resveratrol, e adições de 7,5 mmol/l de 2-metoxietanol. A adição de 2-ME e resveratrol (padrão e isolado do tempê) da cultura de células nas concentrações de 1,4, 2,8 e 4,2 µg/mL mostrou que a maioria dos neurônios cresceu bem. Por outro lado, após a exposição ao 2-ME e beta-amiloide, a glia foi ativada. Esses achados demonstram um papel do resveratrol na ação neuroprotetora e de neurorresgate.(AU)

This study aimed to investigate the effect of supplementing 300 mg/kg tea polyphenols (TP) on yolk cholesterol content and production performance of laying hens during the egg-laying period. A total of 600 Roman laying hens aged 24 weeks were randomly divided into two dietary treatment groups. The feeding experiment lasted for 48 weeks. Layers fed basal diet supplemented with 0 (control group) and 300mg/kg TP (TP group) diet, respectively. The yolk cholesterol content, laying performance, and egg quality were determined at 28, 38, 48, 58, and 68 weeks of age. The yolk cholesterol content in the TP group was significantly decreased at 28-68 weeks of age (p<0.01), compared to the control group. There was a significant increase in laying rate in the TP group at 38 weeks of age (p<0.05), compared to the control group, while no significant differences during the other laying periods were obtained (p>0.05). The FCR significantly decreased in the TP group at 38 weeks of age whereas AEW significantly increased in the TP group at 58 weeks of age (p<0.05). Similarly, the eggshell thickness and eggshell strength in the TP group significantly increased (p<0.05), compared with the control group at 38 weeks of age. The albumen height and Haugh unit significantly increased at 28 weeks of age (p<0.05). In conclusion, the results showed that the diet supplemented with 300 mg/kg TP had positive effects on production performance of layers during the egg-laying period, and could lessen yolk cholesterol content significantly at 28-68 weeks of age.(AU)

OBJECTIVES: To test the hypotheses that estimated mean pulmonary arterial pressure (MPAP) decreases and pulmonary vascular maturation, assessed by the ratio of pulmonary arterial flow acceleration time to ejection time (AT/ET ratio), increases after reversal of fetal ductus arteriosus constriction by reducing maternal intake of the causal agent (prostaglandin inhibitors, such as polyphenol-rich foods or non-steroidal anti-inflammatory drugs), and that these effects are independent of gestational age, which are inferences not yet demonstrated in the clinical setting. METHODS: This was a prospective cohort study comparing Doppler echocardiographic ductal flow dynamics, MPAP and pulmonary arterial flow AT/ET ratio in third-trimester fetuses (≥ 28 weeks' gestation) with ductus arteriosus constriction, at the time of diagnosis and after 2 weeks of reduced maternal intake of prostaglandin inhibitors either by suspending the use of pharmacological agents with potential for prostaglandin inhibition or by restricting the consumption of polyphenol-rich foods. MPAP was estimated using the Dabestani equation (MPAP = 90 - (0.62 × AT)), and pulmonary vascular maturity was assessed using the AT/ET ratio, according to reported validation studies. Student's t-test was used for comparison of variables at diagnosis with those after reversal of ductal constriction. Change in MPAP and pulmonary AT/ET ratio between the two assessments was compared with the expected change in the same gestational period in normal fetuses based on reference curves of MPAP and pulmonary AT/ET ratio constructed in normal fetuses from healthy pregnant women at 19-37 weeks' gestation, encompassing the same gestational age range as the study group (28-37 weeks). RESULTS: Seventy pregnancies with fetal ductus arteriosus constriction were included in the study. After 2 weeks of reduced maternal intake of prostaglandin inhibitors, normalization of mean systolic (change from 1.86 ± 0.34 m/s at diagnosis to 1.38 ± 0.41 m/s; P < 0.001) and diastolic (change from 0.41 ± 0.11 m/s to 0.21 ± 0.065 m/s; P < 0.001) ductal velocities and of mean pulsatility index (change from 1.99 ± 0.20 to 2.55 ± 0.42; P < 0.001) was demonstrated. MPAP decreased between the assessments (change from 66.7 ± 6.90 mmHg at diagnosis to 54.5 ± 6.70 mmHg after 2 weeks; P < 0.001) and mean pulmonary AT/ET ratio increased (change from 0.20 ± 0.06 to 0.33 ± 0.07; P < 0.001). Change in MPAP between diagnosis and after 2 weeks of reduced maternal intake of prostaglandin inhibitors was -12.2 ± 0.30 mmHg, which was 5.3-times higher than that in 305 normal fetuses over 2 weeks during the same gestational period (-2.3 ± 0.19 mmHg) (P < 0.001), and change in pulmonary AT/ET ratio between the two assessments was 0.13 ± 0.08, which was 8.7-times higher than that in normal fetuses in the same gestational period (0.015 ± 0.08) (P < 0.001). CONCLUSIONS: Resolution of fetal ductal constriction is followed by a fall in MPAP and by an increase in pulmonary vascular maturity, to a significantly greater degree than is observed in normal fetuses in the same gestational-age period. © 2021 International Society of Ultrasound in Obstetrics and Gynecology.

The objective was to quantify the flow of intestinal nutrients and nitrogen excretion and retention in sheep receiving isoproteic diets. Eight Texel x Lacaune wethers (average body weight = 25±2.5 kg) were fitted with duodenal cannula and housed in metabolic cages. Wethers were assigned to the treatments in a crossover design with two periods of 20 days each, and all feces and urine produced by the wethers were collected. The treatments consisted of two isoproteic (160 g kg−1 of crude protein on dry matter basis) diets composed of red clover (RC) or lucerne (LU; Medicago sativa) silages plus corn silage and concentrate feed. The digestible organic matter and metabolizable energy intake did not differ between treatments. The intestinal non-ammonia N (NAN) flow was 5.9 g day−1 (37%) higher in RC wethers than in those of the LU treatment. This result was a consequence of both an increase in the efficiency of microbial protein synthesis (12.7% higher) and a decrease in ruminal degradable protein (RDP) content (20% lower) of the diet. However, the increase in the intestinal NAN flow was accompanied by a reduction in intestinal digestibility of N, resulting in similar daily N retention between treatments. The reduction of RDP content was probably the main reason for reductions in N urinary excretion in RC wethers compared with those in the LU treatment, showing that RC silage may be a tool for mitigating the impact of N excretion in ovine production systems, without changes in N retention.(AU)

This study aims to investigate whether resveratrol (RES) could inhibit NF-κB activity and protect in vitro cultured chicken lymphocytes from apoptosis induced by continuous LPS stimulation. Blood lymphocytes of chickens were collected and cultured. LPS was added to the samples of treatment group, while the same volume of normal saline (NS) was supplemented to those of the control group. In the treatment groups, four different concentrations of RES (0, 20, 40, and 80 µg/ml) were admixed. Then, the following indicators were tested: lymphocyte apoptotic rate, lymphocyte viability, NF-κB activity, TNF-a level, reactive oxygen species (ROS), and expression of apoptosis-related proteins (Fas/FasL and Caspase-8). The results indicated that the application of different concentrations of RES could significantly increase the viability of lymphocytes and decrease the apoptotic rate (p<0.05) after continuous stimulations by LPS. The activity of NF-κB levels of TNF-a and ROS in the RES treatment subgroups was significantly decreased in comparison with that in the RES-blank subgroup (p<0.05). The cells in the treatment group that had been treated with 20, 40 and 80 μl/ml of RES exhibited a significantly lower Fas and Caspase-8 expression level than that in the RES-blank subgroup (p<0.05). These findings revealed that RES could substantially diminish the concentrations of ROS and TNF-a, down-regulate NF-κB activity, decrease the expression of Fas and Caspase-8, stimulate the activity of peripheral lymphocytes, and lower the rate of their apoptosis, induced by continuous LPS stimulation in chicken lymphocyte cultures.(AU)

CONTEXT: Propolis has promising biological activities. Propolis samples from the Northeast of Bahia, Brazil - sample A from Ribeira do Pombal and B, from Tucano - were investigated, with new information regarding their biological activities. OBJECTIVE: This paper describes the chemical profile, antioxidant, anti-glycation and cytotoxic activities of these propolis samples. MATERIAL AND METHODS: Ethanol extracts of these propolis samples (EEP) and their fractions were analyzed to determine total phenolic content (TPC); antioxidant capacity through DPPH•, FRAP and lipid peroxidation; anti-glycation activity, by an in vitro glucose (10 mg/mL) bovine serum albumine (1 mg/mL) assay, during 7 d; cytotoxic activity on cancer (SF295, HCT-116, OVCAR-8, MDA-MB435, MX-1, MCF7, HL60, JURKAT, MOLT-4, K562, PC3, DU145) and normal cell lines (V79) at 0.04-25 µg/mL concentrations, for 72 h. The determination of primary phenols by ultra high-pressure liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS) and volatile organic compounds content by gas chromatography-mass spectrometry (GC-MS) were also performed. RESULTS: The EEP polar fractions exhibited up to 90% protection against lipid peroxidation. The IC50 value for anti-glycation activity of EEP was between 16.5 and 19.2 µg/mL, close to aminoguanidine (IC50 = 7.7 µg/mL). The use of UHPLC-MS/MS and GC-MS allowed the identification of 12 bioactive phenols in the EEP and 24 volatile compounds, all already reported. CONCLUSIONS: The samples present good antioxidant/anti-glycation/cytotoxic activities and a plethora of biologically active compounds. These results suggest a potential role of propolis in targeting ageing and diseases associated with oxidative and carbonylic stress, aggregating value to them.

This review aimed to investigate possible protective or deleterious effects of polyphenol-rich foods (PRF) on chronic diseases, e.g. cardiovascular, and in pregnant women, along with their antioxidant and anti-inflammatory action. A great variety of foods and beverages, such as herbal teas, grape and orange derivatives, dark chocolate, and many others contain high concentrations of flavonoids and are freely consumed by the general population. In humans, PRF consumption reduces lipid peroxidation, and several studies have shown a positive correlation between an increased consumption of PRF and a decrease in the incidence of cardiovascular disease. On the other hand, current studies have suggested that maternal ingestion of PRF, especially during the third trimester of pregnancy, could be associated to fetal ductal constriction (DC). Fetuses exposed to this type of diet show higher ductal velocities and lower pulsatility indexes, as well as larger right ventricles than those exposed to minimal amounts of these substances. The underlying mechanism involved in these conditions has not been entirely elucidated, but it seems to be a result of the antioxidant and anti-inflammatory effects of polyphenols by some pathway. Furthermore, taking into account the deleterious effect in late-pregnancy against the numerous positive effects associated to polyphenols, this dual behavior deserves attention particularly to control the dietary ingestion of PRF during gestation. In this line, same PRF, natural constituents of human diet, may represent risk to fetal in late pregnancy compared to the use of nonsteroidal anti-inflammatory drugs.

O câncer de mama é o segundo tipo de neoplasia mais prevalente no mundo e o mais comum entre as mulheres. É descrito que o padrão de consumo alimentar materno e paterno está relacionado à suscetibilidade da prole ao desenvolvimento de doenças crônicas não transmissíveis, inclusive o câncer. A amora-preta é uma das frutas com maior conteúdo antioxidante e seus compostos bioativos possuem atividade antioxidante, anticarcinogênica e anti-inflamatória. Sendo assim, o presente trabalho propõe avaliar os efeitos do consumo materno e/ou paterno de extrato de amora-preta (Rubus spp.) na suscetibilidade da prole feminina ao desenvolvimento de neoplasias mamárias quimicamente induzidas. Para tanto, camundongos da linhagem C57BL/6 foram divididos aleatoriamente em 4 grupos: pai amora (PA), mãe amora (MA), pai e mãe amora (PMA) e controle (CTRL). Os pais receberam extrato de amora-preta logo após o desmame durante 8 semanas e as mães receberam o extrato durante a gestação e lactação. O extrato de amora-preta foi administrado na água de beber (0.84g de antocianinas/L) ad libitum. Os pais tratados com extrato de amora apresentaram redução na atividade enzimática da superóxido dismutase (SOD) e da catalase (CAT) no testículo (p<0.05 e p<0.001, respectivamente), aumento na capacidade antioxidante plasmática, na porcentagem de espermatozoides normais e na produção diária de espermatozóides em relação ao grupo controle (p<0.001 para todos). Além disso, os grupos PA, MA e PMA apresentaram aumento na taxa de prenhez (p<0.05) e redução da mortalidade perinatal (p<0.01, p<0.05 e p<0.001, respectivamente). Em relação à prole feminina não submetida à carcinogênese foi observada redução na capacidade antioxidante plasmática nos grupos PA (p<0.001) e MA (p<0.01), enquanto o grupo PMA apresentou aumento nesse parâmetro (p<0.001). No desenvolvimento da glândula mamária, houve aumento do desenvolvimento epitelial nos grupos PA, MA e PMA (p<0.001 para todos), de diferenciação nos grupos MA e PMA (p<0.01 para ambos) e da taxa de apoptose nos grupos MA e PMA (p<0.05), além de redução no número de TEBs nos grupos PA, MA e PMA (p<0.01, p<0.001 e p<0.001, respectivamente). Não foram observadas alterações significativas nas filhas submetidas à indução química da carcinogênese mamária por DMBA. Assim, é possível concluir que apesar de ter alterado o desenvolvimento da glândula mamária, o consumo materno e/ou paterno de extrato de amora-preta não foi capaz de impactar sobre a suscetibilidade da prole feminina à carcinogênese mamária quimicamente induzida


Breast cancer is the second most prevalent type of cancer in the world and the most common among women. It is known that maternal and paternal food intake pattern are related to offspring susceptibility to non-communicable diseases, including cancer. Blackberry is one of the fruits with high antioxidant content and its compounds have antioxidant, anticarcinogenic and anti-inflammatory properties. So, the aim of the present study was evaluate the effects of maternal and/or paternal blackberry extract consumption on female offspring susceptibility to chemically-induced breast carcinogenesis. Thus, C57BL/6 mice were divided into 4 groups: father blackberry (FB), mother blackberry (MB), father and mother blackberry (FMB) and control (CTRL). Fathers received blackberry extract from weaning during 8 weeks and the mothers were treated during gestation and lactation. Blackberry extract was given in the drink water (0.84g anthocyanins/L) ad libitum. Fathers treated with blackberry had a reduction on superoxide dismutase (SOD) and catalase (CAT) activities in the testis (p<0.05 and p<0.001, respectivelly), an increase on plasmatic antioxidant capacity, percentage of normal sperm and daily sperm production in relation to control group (p<0.001 for all comparisons). Moreover, FB, MB and FMB groups had an increase of pregnancy rate (p<0.05) and a decrease of perinatal mortality (p<0.01, p<0.05 and p<0.001, respectively). Female offspring had a reduction of plasmatic antioxidant capacitity in FB (p<0.001) and MB (p<0.01) groups, while FMB group showed an increase in this parameter (p<0.001). On mammary gland development, it was observed higher epithelial development in FB, MB and FMB groups (p<0.001 for all comparisons), increased differentiation in MB and FMB groups (p<0.01 for both) and higher apoptosis rate in MB and FMB groups (p<0.05 for both), besides decreased TEBs number in FB, MB and FMB groups (p<0.01, p<0.001 and p<0.001, respectively). It was not found significant differences in the female offspring submitted to chemically-induced breast carcinogenesis. So, it is possible to conclude that in spite of maternal and/or paternal blackberry extract consumption changed the mammary gland development, it was not able to change the female offspring susceptibility to chemically-induced breast carcinogenesis

OBJECTIVE: Because we have previously demonstrated the relation between polyphenol-rich foods (PRF) consumption and ductus arteriosus constriction, in this work, pregnant sheep were submitted to oral PRF intake for 14 days to understand how this process occurs. Fetal Doppler echocardiography, oxidative and inflammatory biomarkers and total polyphenol excretion were evaluated. RESULTS: The high polyphenol intake induced ductus arteriosus constriction by 71.6% increase in systolic (P = 0.001) and 57.8% in diastolic velocities (P = 0.002), and 18.9% decrease in pulsatility index (P = 0.033), along with 1.7-fold increase in total polyphenol excretion, 2.3-fold decrease in inflammatory mediator nitric oxide and following redox status changes (mean ± standard deviation): higher protein carbonyls (1.09 ± 0.09 and 1.49 ± 0.31), catalase (0.69 ± 0.39 and 1.44 ± 0.33) and glutathione peroxidase (37.23 ± 11.19 and 62.96 ± 15.03) in addition to lower lipid damage (17.22 ± 2.05 and 12.53 ± 2.11) and nonprotein thiols (0.11 ± 0.04 and 0.04 ± 0.01) found before and after treatment, respectively. Ductal parameters correlated to NOx , catalase, glutathione peroxidase and protein carbonyl. CONCLUSION: Our results highlight the need to reduce maternal PRF intake in late pregnancy to prevent fetal duct constriction through NO-mediated vasoconstrictive action of polyphenols.
