RESUMO

Carotenoids are determinants of reproductive fitness and egg quality. Here we studied the accumulation of astaxanthin (AX), canthaxanthin (CA) zeaxanthin (ZX), lutein (LU), retinol (RX) and dehydroretinol (DR) during vitellogenesis comparing previtellogenic and vitellogenic pikeperch (Sander lucioperca) eggs (n = 5 each), as well as selected tissues (liver, fat and muscles) in first süawning females (1176-1450 g). Futhermore, we compared egg batches with high (8899% hatching rate, n = 5) or low (4067% hatching rate, n= 5) egg quality. Vitellogenic follicles revealed higher concentrations of DR, RX, ZX and LU compared to previtellogenic follicles. Neither CA nor AX was detectable. In parallel, DR and RX were mobilized in the liver. In adipose and muscle tissue, comparing previtellogenic and vitellogenic females, no significant differences in carotenoid/retinoid content were observed. In high quality egg batches, both DR and RX were increased. LU was lower in high quality than in low quality eggs. In a conclusion, the amount of retinoids seems suboptimal in low quality egg batches and increased DR and RX are desirable in pikeperch. Since hypervitaminosis of retinoids can be problematic though, supplementation of the food with carotenoids, which can serve as precursors for retinoids, has to be carried out carefully.(AU)

RESUMO

Marine angelfish (family: Pomacanthidae) are among the most sought-after fish species in the saltwater aquarium trade. However, there is a lack of information in the literature on their early ontogeny. The objective of this study was to describe the embryonic and early larval development of two dwarf angelfish, the bicolour angelfish, Centropyge bicolor and the coral beauty angelfish, Centropyge bispinosa. The eggs of these two species were obtained from spontaneous spawning of the broodstock fish in captivity and incubated at 26.0 ± 0.2°C throughout the study. Fertilized eggs (n = 15) of both species are transparent, pelagic and spherical; the mean diameters of the eggs were measured at 703.6 ± 7.8 µm for C. bicolor and 627.6 ± 7.8 µm for C. bispinosa. The eggs of both species possessed a narrow perivitelline space, smooth and thin chorion, a homogenous and non-segmented yolk as well as a single oil globule. Overall, the observed embryonic development pattern of C. bicolor and C. bispinosa was very similar, and the main difference was the embryonic pigmentation pattern, which only became evident close to hatching. Larvae of both species started hatching at 13 h 30 min after fertilization, and the larval characteristics of both species also showed high levels of similarities. However, the mouth opening time for C. bicolor was 72 h after hatching (AH) and 96 AH for C. bispinosa. In general, the observed early ontogeny of C. bicolor and C. bispinosa also resembled that of other Centropyge species documented in the literature.

RESUMO

The flameback pygmy angelfish Centropyge aurantonotus, highly appreciated and valued by the aquarium market, is heavily harvested and traded. Temperature is one of the abiotic factors that has the most influence on fish development, especially in the early stages of life. For captive production, it is essential to know the appropriate environmental parameters for each species. In this sense, this study aimed to evaluate the influence of temperature on the embryonic development and hatching rates of C. aurantonotus incubated at six different temperatures (20, 22, 24, 26, 28, 30°C). Embryonic development events were very similar in terms of morphological and chronological characteristics compared with other species of the genus Centropyge. Incubation time was inversely proportional to temperature. The treatment at 22°C required twice the time of that required by 30°C treatment for hatching to occur. The best incubation temperature range was 24-28°C. Values below 22°C and at 30°C showed lower hatching rates compared with other treatments. Based on these results, the recommended temperature at which to incubate C. aurantonotus eggs is between 24-28°C.

RESUMO

The totoaba, Totoaba macdonaldi, is an endangered fish of the Gulf of California with high economic and ecological potential. Therefore, our purpose was to characterize the Primordial Germ Cells (PGCs) of this Sciaenid with two objectives: (1) to provide the basis for PGCs cryopreservation to preserve the genetic resources and (2) to take the first step to know the gonadal genesis and sex differentiation of totoaba. Immunofluorescence analysis performed from 2-cell stage to 8-day after hatch (DAH) shows that Vasa protein is specific for PGCs. These cells were first observed in the peripheral and dorsal regions of the blastodisc at approximately the 50%-epiboly stage and migrated to both sides of embryo body during the development. Finally, at 7 DAH the PGCs of the hatching embryo reached the place where the gonad will be developed. Histology analysis of larvae showed a genital ridge with enclosed PGCs on the dorsal side of the peritoneum at 9 DAH, gonadal primordium growth was observed at 11 DAH as a result of the interaction between PGCs and somatic cells derived from the peritoneum. Results of qPCR showed that vasa expression was restricted to the embryonic and early larval development, highest values were observed in 2-cell and mid-blastula stage suggesting the maternal inheritance of vasa mRNA. These findings support the hypothesis of preformation in T. macdonaldi PGCs. The migration pattern of PGCs allow us to recommend the isolation and subsequent cryopreservation of these cells before 7 DAH when the embryonic and larval development is given at 21 °C.

This study aimed to evaluate different extenders in refrigerated storage of dusky grouper sperm. Initially, motility rate, motility time, and sperm density were analyzed to characterize the fresh sperm quality. Different diluents were used in the refrigerated storage sperm at 4ºC in normal atmosphere and modified atmosphere (100% oxygen). The fresh sperm has a spermatozoa density of 3.1±0.2 x 109 spermatozoa mL-1, motility rate 90% and motility time of 3,060 seconds. In the experiment of refrigerated storage, the motility rate and the motility time were maintained appropriate for 144 hours for the extender A (70%; 3,100 seconds) in normal atmosphere. In the modified atmosphere, the sperm quality decreased drastically during the first 24 hours, independently of the diluent used, not propitiating advantages. The refrigerated sperm of dusky grouper is a viable alternative for the short-term storage, being possible, to maintain for up to 144 hours an appropriate sperm viability.(AU)

RESUMO

In pelagic species such as Seriola lalandi, survival of both the eggs and embryos depends on yolk processing during oocyte maturation and embryo development. The main enzymes involved in these processes are the cathepsins, which are essential for the hydration process, acquiring buoyancy and nutrition of the embryo before hatching. This study aimed to investigate the mRNA expression profiles of cathepsins B, D and L (catb, catd and catl) and the activity of these enzymes during early development in S. lalandi. We included previtellogenic oocytes (PO). All three enzymes were highly expressed in PO, but the expression was reduced throughout development. Between PO and recently spawned eggs (E1) the transcript to catb and catd decreased, unlike catl. Cathepsin B activity, showed stable levels between PO until blastula stage (E4). High activities levels of cathepsins D and L were observed in E1 in comparison with later developmental stages. Cathepsin L activity remained constant until E1, consistent with observations in other pelagic spawners, where its participation in a second protolithic cleavage of the yolk proteins, has been proposed for this enzyme. Their profiles of both mRNA expression and enzymatic activity indicate the importance of these enzymes during early development and suggest different roles in egg yolk processing for the hydration process and nutrition in early embryos in this species.

RESUMO

Overfishing of king angelfish Holacanthus Passer reduced populations in the Gulf of California. This study aimed to determine the fatty acid and proximate composition of male and female gonads, and the E2 concentration in plasma of wild organisms with different stages of gonadal development captured during early and mid-summer at El Datil, near Kino Bay, Sonora, Mexico, to help establish basic reproductive aspects and possibly to help develop the aquaculture of this ornamental species. Out of the 45 adult specimens analyzed, 66.7% were females and 33.3% were males; 16.7% of the females had ripe gonads while 83.3% had spent gonads, and 20% of males had ripe gonads while 80% had spent gonads, Indicating they were reproducing earlier, likely in the spring. Females had smaller size but numerically greater gonadosomatic index (GSI) and condition factor (K) than males. Ripe and spent females showed no statistical differences in weight, length or K; however, ripe females showed significantly higher GSI (P = 0.0005) than spent females, 3.43 vs. 0.87%. Lipid content was higher in ripe gonads, with higher content of fatty acids. In both sexes, DHA was quantitatively the major fatty acid, (10.83-16.28 mg of DHA g-1 of gonad wet weight). The n-3/n-6 ratios varied from 1.99 to 3.54, lower for ripe organisms due to a higher content of ARA and n-6 derivatives. Gonad DHA content changed in relation to its developmental stage and it might serve as an additional maturation index. Information on the chemical composition and fatty acid profile of the gonads can be used as indicators of dietary lipid and essential fatty acid requirements of broodfish, which may contribute to the formulation of a balanced diet for the culture of this ornamental species.(AU)

RESUMO

Temperature is an environmental factor that influences the development of fish, and when changed abruptly can lead to high mortality. Some species of fish are influenced by this factor, exhibiting a longer time for embryonic development and time to first feeding. This study aims to evaluate the effect of water temperature on embryonic and larval development up to first feeding, to describe the time in hours post fertilization (hpf) of the emergence of different structures and to determine the best hatching rate and survival of animals under different treatments. Five different egg incubation temperatures were used (24, 26, 28, 30 or 32°C, respectively). The eggs were observed at regular intervals of 30 min up to 24 h, every 2 h until 48 h and every 4 h until the display of first feeding in all treatments. Embryonic development was longer for eggs incubated at 24°C and the best results for hatching rate and survival of spawning efficiency were at 28°C. We recommend that incubation of Trichogaster leeri eggs is carried out at 28°C up to the first feeding of larvae.

RESUMO

Abstract Lutjanid species exhibit multiple spawning behaviour during an extended spawning season in warm months, asynchronous oocyte development and indeterminate fecundity. Although early studies have contributed to knowledge of the reproductive cycle of many species within the group, they have not considered aspects about the number of cortical alveoli oocyte stage throughout maturity phases along spawning season. The latter aspect is also considered very important to confirm indeterminate fecundity hypothesis. In the present study, were analyzed 154 Brazilian snapper Lutjanus alexandrei female gonads obtained from artisanal fisheries in Pernambuco State (Brazil) between October 2010 and March 2011. Were measured oocyte size frequency distribution for maturity phases (developing, spawning capable and actively spawning), and oocyte development stage (unyolked oocytes, cortical alveoli, primary, secondary and tertiary vitellogenic oocytes and hydrated oocytes), and also the oocyte stage frequency during spawning season. The frequency of cortical alveoli oocyte stage was constantly found in the spawning period (>37%), showing slight variation throughout maturity phases. The absence of gap in the oocyte size frequency distribution between primary and secondary oocyte growth stages during spawning season is a strong indicator of continuous oocyte recruitment from reserve stocks. In addition, co-occurrence of tertiary vitellogenic oocytes, hydrated oocytes, post-ovulatory follicles and yellow-brown bodies in the histological sections of ovaries reinforce indeterminate fecundity hypothesis.(AU)

The 2010 Deepwater Horizon oil spill resulted in the accidental release of millions barrels of crude oil into the Gulf of Mexico. Photoinduced toxicity following coexposure to ultraviolet (UV) radiation is one mechanism by which polycyclic aromatic hydrocarbons (PAHs) from oil spills may exert toxicity. Mahi-mahi (Coryphaena hippurus), an important fishery resource, have positively buoyant, transparent eggs. These characteristics may result in mahi-mahi embryos being at particular risk from photoinduced toxicity. The goal of this study was to determine whether exposure to ultraviolet radiation as natural sunlight enhances the toxicity of crude oil to embryonic mahi-mahi. Mahi-mahi embryos were exposed to several dilutions of water accommodated fractions (WAF) from slick oil collected during the 2010 spill and gradations of natural sunlight in a fully factorial design. Here, we report that coexposure to natural sunlight and WAF significantly reduced percent hatch in mahi-mahi embryos. Effect concentrations of PAH in WAF were within the range of surface PAH concentrations reported in the Gulf of Mexico during the Deepwater Horizon spill. These data suggest that laboratory toxicity tests that do not include UV may underestimate the toxicity of oil spills to early lifestage fish species.

RESUMO

The spotted rose snapper (Lutjanus guttatus) is an important commercial species in Mexico with good culture potential. The osteological study at early stages in this species is an important tool to confirm normal bone structure and for the detection of malformations that may occur during early development. This study was carried out in order to evaluate and describe the normal osteological development of the vertebral column and caudal complex of this species grown under controlled conditions. For this, a total of 540 larvae of L. guttatus, between 2.1 and 17.5 mm of total length (TL), were cultured during 36 days; culture conditions were 28 degrees C, 5.74 mg/L oxygen and 32.2 ups salinity with standard feeding rates. To detect growth changes, a sample of 15 organisms was daily taken from day one until day 36 of post-hatch (DPH). Samples were processed following standard techniques of clearing, and cartilage (alcian blue) and bone staining (alizarin red). Results showed that the vertebral column is composed of ten vertebrae in the abdominal region, and 14 vertebrae including the urostyle in the caudal region. The development of the axial skeleton starts with the neural arches and haemal arches at 3.8 mm TL. Caudal elements such as the hypurals and parahypural began to develop at 4.1 mm TL. Pre-flexion and flexion of the notochord and the formation of all hypurals were observed between 5.3 and 5.8 mm TL. Ossification of the vertebrae in the abdominal region and in some neural arches initiated at 9.5mm TL. In the caudal region, all the neural and haemal arches ossified at 10.2 mm TL. All the abdominal vertebrae and their respective neural arches and parapophyses ossified at 11.2 mm TL, while the elements of the caudal complex that ossified were the hypurals, parahypurals and modified haemal spines. All caudal fm rays, 12 neural spines and 3 haemal arches were ossified by 15.5 mm. The complete ossification process of this specie under laboratory culture conditions was observed when larvae reached 17.3 mm TL on 36 DPH. Detailed analysis of the osteological structures will allow a reference description to evaluate and detect malformations that may occur during the larval culture of the spotted rose snapper.

Betta splendens is a very important ornamental species. The current paper describes the embryonic and larval development of B. splendens under stereomicroscopy and scanning electron microscopy. Eggs and larvae from natural spawning were collected at different developmental stages at previously established intervals and analysed. The eggs of B. splendens are yellowish, clear, spherical, demersal, translucent and telolecithal with a large amount of yolk. Between 0-2 h post-initial collection (hpIC), the eggs were at the egg cell, first cleavage and morula stages. The blastula stage was identified at 2-3 hpIC and the early gastrula phase was observed at 3-4 hpIC with 20% epiboly, which was finalized after 13-18 hpIC. When the pre-larvae were ready to hatch, the appearance of somites and the free tail were observed, at 23-25 hpIC. At 29 hpIC, the majority of larvae had already hatched at an average temperature of 28.4 ± 0.2°C. The newly hatched larvae measured 2.47 ± 0.044 mm total length. The mouth opened at 23 h post-hatching (hPH) and the yolk sac was totally absorbed at 73 hPH. After 156 hPH, the heart was pumping blood throughout the entire larval body. The caudal fin, operculum and eyes were well developed at 264 hPH. When metamorphosis was complete at 768 hPH, the larvae became juveniles. The current study presents the first results about early development of B. splendens and provides relevant information for its reproduction, rearing and biology.

Aquatic re-circulation systems (SAR) are being used more and more in agriculture, especially in the north of Chile where the environment conditions are suitable for breeding in SAR (aquatic re-circulation system) of species such as Bonito (Sarda chiliensis chiliensis), whose embryo or larvae origins are unknown, which complicates the evaluation of a technical and economical possibility of developing its breeding in SAR. Due to this, bases for its breeding were determined in January 2013, for 45 days. The characterized embryo periods were 31 in 71, 83 hours, defined in 5 stages: Morula, Blastula, Gastrula, Neurulation and Metamery. From the larvae development 35 periods in 519 hours were observed, a pre-juvenile stage observed at 591 hours and a juvenile one. The eggs were translucent of a telolecitic type of a slow growing in relation to Sarda sarda and Sarda ch. lineolata, since it took them almost three days to hatch. The surviving and hatching rates were high (86,316±9,808% and 89,596±11,683% respectively), so the conditions for their development would be appropriate. Meanwhile, the larvae presented more similarities with Sarda sarda in the first periods of development. The crucial period was between the 81 and 89 hours of life, when the oily drop was re-absorbed; besides, this was a distinguishing feature with the other two species because it was the only one that presented just one oily drop in the larvae period. The possibility of a Sarda ch. chiliensis hatchery is inferred, due to the similarities with Sarda sarda, and because of the success obtained in this breeding. From the morphometric characters, future monitoring is suggested, the ones that represent the most variables in the stages of development such as length and height of the head, and length and height of the eye.

The Colorado snapper (Lutjanus colorado) is one of the most commercially important fish species in México and it is considered a suitable candidate for culture. Until now, no research has been carried out on its reproduction, larviculture and fattening in captivity. This study is the first description of embryonic development of this species under controlled conditions. Fertilized eggs of Colorado snapper are pelagic, spherical and transparent and contain one drop of oil. Eggs measured 0.77±0.09 mm and the drop of oil 0.14±0.01 mm. First cell division occurred at 0.05 h post-fertilization (HPF), hatching at 17.22 HPF under the above described conditions. Larvae total length (LT) was 1.8±0.1 mm. Embryonic development of this species was similar to other lutjanidae species. These results provide basic information for developing the necessary biotechnology for commercial seed production of the Colorado snapper.

RESUMO

The goals of this study are to provide a technical foundation for the production of the red head goby Elacatinus puncticulatus by evaluating its reproductive behavior and its embryonic and early larval development. Five pairs were kept under controlled conditions for thirty days. Courtship behavior, spawning period and the number of eggs produced were recorded. For the evaluation of embryo development, eggs were sampled at 12, 18, 24, 48, 72, 96, 120, 144 and 168h post-fertilization(HPF). To test the influence of the incubation period on larval total length and height, eggs with six days (6D) of incubation and with seven days of incubation (7D) were subjected to flashlight illumination for 30min to induce larval hatching. Another experiment evaluated the difference in larval survival with three different diets: Euplotes sp. (EU); rotifers Brachionus rotundiformis and Brachionus plicatilis and Paramecium sp. (BP); plankton collected from the wild (WP). The males displayed a gray head and pale yellow and black body coloration. Females exhibited strong red and black colors until three days before spawning, which occurred at intervals of 7 to 10 days. The hatching rate was 98-99%. The larvae total mean lengths and heights were 3.05 and 2.95mm (p>0.05) and 0.37 and 0.48mm (p<0.05) for treatments 6D and 7D, respectively. However, both groups exhibited high mortality at 5 days post-hatch (DPH). No larvae from the EU group survived after 5 DPH. At 8 DPH, 4% survivorship was found in treatment BP and 2% in treatment WP.

RESUMO

BACKGROUND: During fish oocyte maturation, specific molecules are expressed and accumulated within oocyte until fertilization and embryo development. Special attention have been paid in members of the transforming growth factor (TGF-ß) superfamily; growth differentiation factor 9 (GDF9/gdf9) and bone morphogenetic protein 15 (BMP15/bmp15), which exert regulatory functions during oocyte maturation and follicle development. However, little attention has been paid to the involvement of these molecules during embryogenesis considering its importance for the formation of a good quality egg and subsequent embryo survival. The purpose of this study was to analyze the expression of gdf9 and bmp15 in previtellogenic oocytes and during early embryonic development in Seriola lalandi, a pelagic fish with increasing prospect for its aquaculture development, which however, show high mortality at embryo and larval stages. RESULTS: Through RT-qPCR it was found that gdf9 expression was higher in previtellogenic oocytes decreasing after ovulation. This expression profile agrees with its participation in early stages of the follicular development. The transcripts for bmp15 also showed the highest levels in previtellogenic oocytes, however this expression was lower than obtained with gdf9. Conversely, in recently spawned oocytes mRNA bmp15 levels were highest than observed to gdf9. This, is consequent with the main role proposed for this growth factor at the final fish oocyte maturation: avoid the ovulation of an immature oocyte. During embryo development, low levels of mRNA were detected to gdf9, with an increase in 48 H post-fertilization embryos. The bmp15 expression did not change throughout development and was higher than gdf9 at 16 cells, blastula and appearance embryos stages. CONCLUSIONS: Both (gdf9 and bmp15) expression profiles in previtellogenic oocytes and newly spawned eggs are consistent with the described functions for these growth factors in vertebrate ovarian physiology in early and late stages of the follicular development. So, these genes could be considered as quality biomarkers at these stages. However, further studies of these proteins throughout folliculogenesis, are necessaries to fully understand their functions during the oocyte formation. In addition, the persistent expression of these growth factors during development, allows us to speculate possible roles in embryonic processes, which must also be addressed.
