Isostichopus badionotus and Isostichopus sp. are two holothuroids exploited in the Caribbean region of Colombia. Until recently, they were considered a single species. During one year, 222 individuals of Isostichopus sp. and 114 of I. badionotus were collected in two bays of the Santa Marta region to study their reproductive biology and collect information on their size, weight and habitat. Both sea cucumber morphotypes showed an annual reproductive cycle, with a reproductive season from September to November, closely related to the increase in water temperature and rainfall. In both sea cucumbers the population structure exhibited a unimodal distribution composed of mature individuals and a sex ratio of 1:1. Isostichopus sp. had an average size and weight (193 ± 52 mm and 178 ± 69 g) and size and weight at first maturity (175 mm and 155 g) that was much lower than I. badionotus (respectively, 324 ± 70 mm and 628 ± 179 g; 220 mm and 348 g). While 98% of Isostichopus sp. individuals were collected in the upper 2.5 m, on rocky bottoms between cracks, 73% of I. badionotus individuals were found between 3 and 7.8 m depth, exposed on sandy bottoms. These differences imply that management measures (e.g. minimum catch size) should not be the same for both sea cucumbers morphotypes.

RESUMO

Regeneration of lost or injured organs is an intriguing process in which numerous cellular events take place to form the new structure. Studies of this process during reconstitution of the intestine have been performed in echinoderms, particularly in holothurians. Many cellular events triggered during regeneration have been described using the sea cucumber Holothuria glaberrima as a research model. More recent experiments have targeted the molecular mechanisms behind the process, a task that has been facilitated by the new sequencing technologies now available. In this review, we present studies involving cellular processes and the genes that have been identified to be associated with the early events of gut regeneration. We also present ongoing efforts to perform functional studies necessary to establish the role(s) of the identified genes. A synopsis of the studies is given with the course of the regenerative process established so far.

Assessments of the effectiveness of marine protected areas (MPAs) usually assume that fishing patterns change exclusively due to the implementation of an MPA. This assumption increases the risk of erroneous conclusions in assessing marine zoning, and consequently counter-productive management actions. Accordingly, it is important to understand how fishers respond to a combination of the implementation of no-take zones, and various climatic and human drivers of change. Those adaptive responses could influence the interpretation of assessment of no-take zone effectiveness, yet few studies have examined these aspects. Indeed, such analysis is often unfeasible in developing countries, due to the dominance of data-poor fisheries, which precludes full examination of the social-ecological outcomes of MPAs. In the Galapagos Marine Reserve (Ecuador), however, the availability of long-term spatially explicit fishery monitoring data (1997-2011) for the spiny lobster fishery allows such an analysis. Accordingly, we evaluated how the spatiotemporal allocation of fishing effort in this multiple-use MPA was affected by the interaction of diverse climatic and human drivers, before and after implementation of no-take zones. Geographic information system modelling techniques were used in combination with boosted regression models to identify how these drivers influenced fishers' behavior. Our results show that the boom-and-bust exploitation of the sea cucumber fishery and the global financial crisis 2007-09, rather than no-take zone implementation, were the most important drivers affecting the distribution of fishing effort across the archipelago. Both drivers triggered substantial macro-scale changes in fishing effort dynamics, which in turn altered the micro-scale dynamics of fishing patterns. Fishers' adaptive responses were identified, and their management implications analyzed. This leads to recommendations for more effective marine and fishery management in the Galapagos, based on improved assessment of the effectiveness of no-take zones.

Resumen Holothuria grisea, a pesar de no ser considerado un recurso comercial, ha sido objeto de extracciones ilegales en Colombia, hecho que se vuelve más grave debido al vació de información biológica de estos organismos. Por esta razón, se estimó la densidad poblacional y estructura de talla de H. grisea en tres sectores del sur del golfo de Morrosquillo de abril a noviembre 2015. Para ello, se ubicaron mensualmente tres transectos circulares en cada sector de muestreo abarcando un área de 300 m2, donde se contabilizaron y midieron in situ los individuos encontrados. La densidad media de H. grisea en el área estudiada fue 0.4 ind m-2, presentándose los mayores valores en el sector La Ahumadera (1.09 ± 0.11 ind m-2) y los menores valores en el Banco de Arena (0.02 ± 0.004 ind m-2). El análisis de varianza señalo una diferencia significativa en los valores de densidad y talla de H. grisea en los sectores estudiados. La especie presentó una distribución de tallas unimodal, registrándose una talla promedio de 13.5 ± 0.91 cm con una talla mínima de 2.5 y una máxima de 30 cm, en general las tallas intermedias presentaron una mayor representatividad a lo largo de este estudio (77.2 %). Los aspectos biológicos presentados en este documento son de gran importancia para la conservación de esta especie y corresponde a un avance en el conocimiento de la clase Holothuroidea en el Caribe Colombiano.


Abstract Holothuria grisea, despite not being considered a commercial resource has been the subject of illegal extractions in Colombia, a fact that becomes more serious due to the empty of biological information of these organisms. For this reason the density population and size structure of H. grisea were evaluated in three sectors south of the gulf of Morrosquillo from April to November 2015. To this end, three circular transects were located each month in each sampling site covering an area of 300 m2, where individuals were counted and measured in situ. The average density of H. grisea in the study area was 0.4 ind m-2, with the highest values in the La Ahumadera (1.09 ± 0.11 ind m-2) and the lowest values in Banco de Arena (0.02 ± 0.004 ind m-2). The analysis of variance showed a significance in the values of density and size of H. grisea in the studied sectors. The species presented a unimodal distribution of sizes with an average size of 13.5 ± 0.9 cm with a minimum size of 2.5 and a maximum of 30 cm, intermediate sizes generally showed a higher representativeness throughout this study (77.2 %). The biological aspects presented in this paper are of great importance for the conservation of this species and corresponds to an advance in the knowledge of the Holothuroidea class in the Colombian Caribbean. Melanotransferrin (MTf) is a protein associated with oncogenetic, developmental, and immune processes which function remains unclear. The MTf gene has been reported in numerous vertebrate and invertebrate species, including echinoderms. We now report the finding of four different MTfs in the transcriptome of the sea cucumber Holothuria glaberrima. Sequence studies and phylogenetic analyses were done to ascertain the similarities among the putative proteins and their relationship with other transferrin family members. The genes were shown to be differentially expressed in various holothurian organs and to respond differently when the animals were challenged with the immune system activator lipopolysaccharide (LPS). Moreover, the four genes were found to be highly overexpressed during the early stages of intestinal regeneration. The finding of four different genes in the holothurian is particularly surprising, because only one MTf gene has been reported in all other animal species sequenced to date. This finding, combined with the increase expression during intestinal regeneration, suggests a new possible function of MTf in organ regenerative processes.

Most species of Taeniogyrus Semper, 1867 are known from shallow water in the Indo-Pacific, with other records in Antarctica, Mediterranean Sea, and the Atlantic. A new species of Taeniogyrus is described and illustrated here from the continental slope of Campos Basin, southeast of Brazil. In this species, sigmoid hooks (336-405 µm) are much larger than in any other in the genus, bearing a long and conspicuous hook region. Wheels with six spokes (86-169 µm), inner margin with 60-125 continuous teeth, are confined to round papillae along each interradius. Polian vesicles are ventral, numerous (15-21), of different sizes, and tubular shaped with a terminal round region. This new species represents the deepest record of the genus Taeniogyrus. It increases to three the number of chiridotids in Brazilian waters, and the number of Taeniogyrus species in the Atlantic. Additionally, Taeniogyrus furcipraeditus (Salvini-Plawen, 1972) from the Mediterranean Sea and Taeniogyrus havelockensis (Rao, 1975) from the Andaman Sea are proposed as new combinations.

Abstract The sea cucumber Holothuria (Halodeima) grisea is a widely distributed and economically important species that has been harvested in Brazil for several years, where there are no studies on its reproduction. We studied a population near Guaratuba Bay in Southern Brazil. Four sampling campaigns were made, in February, April, June and October 2008. We examined the macroscopic and microscopic features of the gonads to describe the gametogenesis. In addition, we analyzed the weight change of body organs. A total of 94 individuals were examined and no evidence of external sexual dimorphism was found. In April there were more males than females. Biometrically the body wall and gonads are the most important organs. The body wall increases in the June and the gonads during October and February; there might be an exchange of energy between these organs. Mature gonads are elongated and thick (red in females, cream in males). There were mature individuals year round, with an increase of sexual activity in February when oocyte mature diameter ranges from 70 to 160 µm. Spawning was in winter (June). Resumen El pepino de mar Holothuria (Halodeima) grisea es una especie ampliamente distribuida y económicamente importante que se ha recolectado en Brasil desde hace varios años. No hay estudios previos realizados sobre la reproducción de esta especie en Brasil. Además, son aún poco conocidos los aspectos bio-ecológicos de esta especie. En este estudio, se investigaron algunos aspectos de la reproducción del pepino de mar Holothuria (Halodeima) grisea en una población cerca de la bahía de Guaratuba en el sur de Brasil. Se hicieron cuatro salidas de muestreo, en febrero, abril, junio y octubre de 2008, abarcando las cuatro estaciones. Examinamos las características macroscópicas y microscópicas de las gónadas para describir la gametogénesis, adicionalmente, se analizó el cambio de peso de cada componente de los órganos del cuerpo (pared corporal, gónadas, sistema haemal, tracto digestivo y árbol respiratorio). Se examinó un total de 94 individuos y no se encontraron evidencias de dimorfismo sexual externo. Los resultados del análisis estadístico de la prueba x² rechazan la hipótesis nula (H0) con una relación de sexos igual a 1, sin embargo, el análisis de cada mes, esta diferencia de género se observó sólo en el mes de abril, cuando el número de los males es mucho mayor que la de las hembras. Las medidas biométricas mostraron que la pared del cuerpo y las gónadas son los órganos más importantes, con aumentos de la pared corporal en el junio y de las gónadas durante octubre y febrero, que pueden indicar que hay un intercambio de energía entre estos órganos. Macroscópicamente, las gónadas maduras muestran túbulos largos, gruesos y ramificados siendo que las hembras presentan coloración rojiza e los machos tonos de crema. Sin embargo, durante el crecimiento y agotado, no se observaron diferencias morfológicas macroscópicas entre los sexos. Microscópicamente, se observó la presencia constante de individuos maduros entre meses, con un aumento de la actividad sexual en febrero, cuando los ovocitos maduros tuvieron un diámetro oscila entre 70 a 160 μ. El período de desove ocurrió en los meses de invierno (junio). El periodo de maduración y la variación del cuerpo de los componentes fueron similares entre los sexos. La similitud en las demandas metabólicas de la población podría estar confirmando una estrategia para maximizar la sincronía reproductiva.

Sulfated fucans, the complex polysaccharides, exhibit various biological activities. Herein, we purified two fucans from the sea cucumbers Holothuria edulis and Ludwigothurea grisea. Their structures were verified by means of HPGPC, FT-IR, GC-MS and NMR. As a result, a novel structural motif for this type of polymers is reported. The fucans have a unique structure composed of a central core of regular (1→2) and (1→3)-linked tetrasaccharide repeating units. Approximately 50% of the units from L. grisea (100% for H. edulis fucan) contain sides of oligosaccharides formed by nonsulfated fucose units linked to the O-4 position of the central core. Anticoagulant activity assays indicate that the sea cucumber fucans strongly inhibit human blood clotting through the intrinsic pathways of the coagulation cascade. Moreover, the mechanism of anticoagulant action of the fucans is selective inhibition of thrombin activity by heparin cofactor II. The distinctive tetrasaccharide repeating units contribute to the anticoagulant action. Additionally, unlike the fucans from marine alga, although the sea cucumber fucans have great molecular weights and affluent sulfates, they do not induce platelet aggregation. Overall, our results may be helpful in understanding the structure-function relationships of the well-defined polysaccharides from invertebrate as new types of safer anticoagulants.

RESUMO

This is a taxonomic work on the aspidochirotid sea cucumbers from shallow waters in Northeast Brazil. We studied 165 specimens, classified into 2 families, 3 genera, 7 subgenera and 9 species. Descriptions, illustrations and literature data are included for each species, in addition to ecological notes and information on their distribution in the northeast coast of Brazil. A taxonomic key is provided for the identification of the species.

Age and growth analysis is essential to fisheries management. Indirect methods to calculate growth are widely used; however, length frequency data analysis in sea cucumbers is complicated by high data variability caused by body wall elasticity. Here we calculated Isostichopus badionotus parameters of the von Bertalanffy growth function. In order to address bias produced by body wall elasticity, we compared the performance of four measurements and one compound index that combines different biometric parameters: the square root of the length-width product (SLW). Results showed that variability in length data due to body wall elasticity was controlled by using body length (Le) from the SLW compound index. Growth in I. badionotus follows a negative allometric tendency. Slow or zero growth periods were observed during October and November, when weather conditions were adverse.

Camamu Bay (Bahia, Brazil) is an Environmental Protected Area with mangroves of economic and ecological importance. The objective of this study was to elucidate, through the distribution of the benthic macrofauna and its relationships with trace metals and bioavailabilty, if the current quality of the mangrove sediments of Camamu Bay is acceptable for the protection of aquatic life. Seven sampling stations were selected as representative of the meso-littoral region of Camamu Bay. It was found 581 macrobenthic organisms distributed in 38 species. The highest values of metals occurred at stations located in areas that influenced by barite extraction, while the lowest concentrations were found in areas with relatively lower anthropogenic influence. Analysis of acid volatile sulphide and simultaneously extracted metals showed the occurrence of the ratio SEM/AVS >1, indicative of possible bioavailability of metals, at only one station. The results suggested that Camamu Bay is a nearly pristine area.
