Homogenisation is a widely used technique in manufacturing powdered milk with a direct impact on product solubility, and the homogenisation pressure is a central attribute of this process. We aimed to understand the effect of increasing homogenisation pressures (0/0, 15/5, and 75/5 MPa, 1st/2nd stages) on particle-size distribution during homogenised whole milk powder manufacture and rehydration of the final product. The fluid milk was thermally treated, homogenised, concentrated by rotary evaporation, and then dried using a spray dryer. Particle size (Dv90) was monitored at all stages of the manufacturing process. The final product (milk powder) was analysed using particle-size distribution, electronic scanning microscopy, water activity, and isotherms. The results demonstrated that increasing the homogenisation pressure leads to milk powder with smaller particle size when rehydrated (Dv90 values: 6.08, 1.48 and 0.64 µm for 0, 20 and 80 MPa, respectively). Furthermore, the volume (%) of the particles in the 'sub-micro' region (smaller than 1.0 µm) presented an inversely proportional profile to the homogenisation pressure (homogenised fluid milk: 86.1, 29.3 and 2.4%; concentrated milk: 86.1, 26.5 and 5.7%, and reconstituted milk powder: 84.2, 31.8 and 10.9%). Surprisingly, this pattern was not observed in the SPAN value (which corresponds to the width or range of the size distribution based on the volume). Additionally, the increase in the homogenisation pressure did not affect the sorption isotherm pattern. These results demonstrate that increasing the homogenisation pressure decreases the particle size of the reconstituted powdered milk, indicating the potential for future studies on how this phenomenon affects its physicochemical and final product properties.

Extracellular matrix (ECM) alterations in visceral leishmaniasis are related mainly to collagen deposition (fibropoiesis). In canine visceral leishmaniasis (CVL), an intense fibrosis associated to chronic inflammation in organs such as kidneys is described. However, renal fibropoiesis has not been described in natural or experimental infections with L. (L.) infantum. We aimed to characterize renal nephropathies by histology and confocal microscopy comparing renal lesions in dogs naturally and experimentally infected with L. (L.) infantum. Sixty-two mixed-breed symptomatic dogs naturally infected with L. (L.) infantum, sixteen beagles experimentally infected with two strains of L. infantum (eleven dogs with the BH400 strain and five dogs with the BH401 strain), and five uninfected beagles (controls) were used. Samples were stained with hematoxylin & eosin for routine histology. Congo red was used to visualize amyloid protein deposits, periodic acid-Schiff to identify glomerular basal membrane anomalies, Masson's trichrome for collagen deposits, and Jones' methenamine silver to reveal membranous glomerulonephropathy. Immunohistochemistry was used to identify Leishmania amastigotes, and confocal microscopy was used for macrophage characterization (L1/calprotectin and CD163 antigen receptors). The most common lesions were chronic glomerular and interstitial nephritis, which was found in all naturally infected dogs and dogs experimentally infected with L. infantum strain BH401 but not with the BH400 strain. Glomeruloesclerosis was the main lesion presented in all BH401 group. Morphometric analysis revealed positive correlation of renal glomeruli tufts with cellular expression of L1/calprotectin and CD163 antigens. Leishmania infantum strain BH401 shows pathogenicity that may be sufficient to induce classic chronic visceral renal leishmaniasis.

Vibrio ordalii is an extracellular, Gram-negative bacterium that produces vibriosis in salmonids. While pathogenesis is not fully understood, this bacterium has numerous likely genes for adhesion, colonization, invasion factors and, as recently suggested, intracellular behaviour. Therefore, this study aimed to clarify possible intracellular behaviour for V. ordalii Vo-LM-18 and ATCC 33509T in the fish-cell lines SHK-1 and CHSE-214. Confocal microscopy revealed Vo-LM-18 and ATCC 33509T inside cytoplasm in both fish-cell lines at 4 hr post-inoculation (hpi). At 8 and 16 hpi, the proportion of fish cells invaded by both strains increased. Moreover, intracellular V. ordalii were observed after 8 hpi inside mouse embryonic fibroblasts (MEF), demonstrating that entry was not due to a cellular phagocytosis process. Flow cytometry confirmed immunocytochemistry results, with both V. ordalii evidencing statistically significant differences in the number of infected cells between 8 and 16 hpi. Interestingly, V. ordalii infection did not significantly damage fish cells, as determined by LDH liberation. Viable counts at 8 hpi detected, on average for both lines, 176 ± 47 CFU/ml of culturable intracellular Vo-LM-18 and ATCC 33509T cells. These in vitro findings support the facultative intracellular behaviour of V. ordalii and may be of importance for understanding pathogenicity and survival in aquatic environments.

Renal fibrosis is characterized by glomerulosclerosis and tubulointerstitial fibrosis and its pathogenesis is associated with the activity of mesenchymal cells (fibroblasts), being essentially characterized by a process of excessive accumulation resulting from the deposition of extracellular matrix components. The aim of this study was to characterize the morphological presentation of chronic and fibrotic lesions in the glomerular, tubular, interstitial, and vascular compartments in feline CKD, as well as the possible participation of myofibroblasts in renal fibrotic processes in this species. Cat kidneys were collected and processed according to the conventional techniques for light microscopy, circular polarization, immunohistochemistry, and electron microscopy. Fibrotic alterations were present in all compartments analyzed. The main findings in the glomerular compartment were different degrees of glomerular sclerosis, synechia formation, Bowman's capsule calcification, in addition to glomerular basement membrane thickening and pericapsular fibrosis. The tubulointerstitial compartment had intense tubular degeneration and the immunostaining in tubular cells for mesenchymal cell markers demonstrated the possibility of mesenchymal epithelial transition and consequent involvement of myofibroblasts in the development of interstitial tubule damage. Infiltration of inflammatory cells, added to vessel thickening and fibrosis, demonstrated the severity and role of inflammation in the development and perpetuation of damage. Thus, we may conclude that fibrotic lesions play a relevant role in feline CKD and the mechanism of perpetuation of these lesions need further elucidation regarding the origin and participation of myofibroblasts and consequent mesenchymal epithelial transition in this species.

Dactylogyridae is overwhelmingly the most abundant and diverse taxon among monogeneans in continental waters of South America. Their small body size requires considerable sampling effort and training for collecting and identifying the worms from the gills, skin, nasal cavities, and other microhabitats. Indeed, diagnostic characteristics as sclerites and male copulatory complex are generally less than 100-µm long and are essential for taxonomic description and identification of species. Here, a combination of simple and routine methods for three-dimensional morphological studies on hard structures is proposed for dactylogirids: SDS treatment for clarification of specimens and enzymatic digestion with proteinase K for freeing sclerotized structures, followed by laser confocal microscopy. This method is applicable to fresh or fixed specimens and does not require staining or dehydration. Indeed, stable autofluorescence emission is detectable at 500-530 nm for bars, anchors, and male copulatory complex when excited by argon laser. Advantages of this protocol over previous methodologies for taking laser confocal images are discussed. Open access software for image processing was used for three-dimensional reconstruction of sclerotized structures generating models and full 360° rotation videos.

Raillietina celebensis is a cestode that parasitizes the small intestine of rats and humans. Here, we detail the morphology and morphometry of R. celebensis based on specimens collected from Rattus norvegicus in the municipality of São Gonçalo, state of Rio de Janeiro, Brazil, by light and confocal scanning laser microscopies and also report the results of molecular phylogenetic analyses to determine its relationships within the family Davaineidae. Analysis of the number and size of testes, number and shape of rostellar hooks, cirrus sac length, capsules and eggs per capsule, and morphology of the mature proglottid allowed concluding that the present specimens constitute a new record of R. celebensis in South America. Our genetic and phylogenetic analyses, based on the partial small subunit 18S rRNA gene, revealed R. celebensis to be in the family Davaineidae within the genus Raillietina, in agreement with the morphological taxonomy. Phylogenetic trees obtained by neighbor-joining and maximum likelihood methods demonstrated R. celebensis as a unique taxonomic unit, and also demonstrated some taxonomic inconsistences. The incorporation of Brazilian R. celebensis sequences derived from mammals in the phylogeny of davaineids is consistent with the assertion that neither Raillietina nor Fuhrmannetta can be supported as distinct genera.

Pygidiopsis macrostomum and Ascocotyle (Phagicola) pindoramensis (Digenea: Heterophyidae) parasitize guppies as intermediate hosts and, respectively, fish-eating mammals or birds as definitive hosts. Heterophyids have zoonotic potential, and molecular studies associated with morphological and ecological aspects have helped to clarify their taxonomy and phylogeny. Poecilia vivipara naturally parasitized by metacercariae of both species (100% prevalence) exhibit no external signs of parasitism. In this work, four new sequences of P. macrostomum (18S rDNA, 28S rDNA and ITS2 rDNA) and one new sequence of A. (P.) pindoramensis (mtDNA cox-1) are presented. Phylogeny reconstructions linked P. macrostomum to other heterophyids, but the separation of the Heterophyidae and Opisthorchiidae remains unclear. Additionally, we used indirect immunocytochemistry and the phalloidin-fluorescence techniques allied with confocal laser scanning microscopy to describe muscular and neuronal structures of P. macrostomum. A complex arrangement of muscular fibres is associated with the tegument, suckers, gut and reproductive system. Radial fibres around the ventral sucker are thick, branched and extend to the body wall. High-resolution confocal imaging revealed a typical digenean muscular arrangement and important heterophyid morphological traits. These data will support future control measures to reduce the parasitism in guppies reared in fish farming systems, especially for aquarium and experimental purposes.

BACKGROUND: The process for obtaining monoclonal antibodies against a specific antigen is very laborious, involves sophisticated technologies and it is not available in most research laboratories. Considering that most cytokines remain partially conserved among species during evolution, the search for antibody cross-reactivity is an important strategy for immunological studies in veterinary medicine. In this context, the amino acid sequence from human and canine cytokines have demonstrated 49-96 % homology, suggesting high probability of cross-reactivity amongst monoclonal antibodies. For this, 17 commercially available anti-human monoclonal antibodies [IL-1α, IL-1ß, IL-2, IL-4, IL-5, IL-6, IL-8 (#1, #2), IL-10, IL-12, IL-13, IL-17A, IFN-γ (#1, #2), TNF-α (#1, #2) and TGF-ß], were evaluated in vitro for intracellular cytokine detection in a stimulated canine blood culture by flow cytometry and confocal microscopy. Lymphocytes from peripheral blood of healthy and two unhealthy dogs were analyzed. RESULTS: Eleven anti-human mAbs [IL-1α, IL-4, IL-5, IL-6, IL-8 (#1, #2), IL-12, IL-17A, TNF-α (#1, #2) and TGF-ß] cross-reacted against canine intracellular cytokines. The specificity of the assays was not affected after Fc-blocking. Three anti-human cytokine mAbs [IL-4, IL-8 (#2) and TGF-ß] when evaluated by confocal microscopy also cross-reacted with intracellular canine cytokines. The identification of human mAbs that cross-reacted with canine cytokines may support their use as immunological biomarkers in veterinary medicine studies. CONCLUSION: The identification of these 11 anti-human cytokine mAbs that cross-reacted with canine cytokines will be useful immunological biomarkers for pathological conditions by flow cytometry and fluorescence microscopy in dogs.

Our study performed qualitative and quantitative studies on the corneal ultrastructure of healthy female Merino sheep of ages 4 months and 6 years old from the Argentinean Pampa. The corneas were evaluated using ex vivo laser-scanning confocal microscopy, light microscopy and transmission electron microscopy. Those studies allowed us to obtain detailed images of the corneal layers as well as quantitative data of the cellular and sub-basal nerve densities in the cornea from sheep of different ages. The density of the corneal cells was significantly different in the anterior versus the posterior epithelium and stroma. Moreover, the density of the epithelial, stromal cells and endothelial cells, as well as the sub-basal nerve density were significantly lower in adult than in young animals. Our work provided a wide-ranging description of the corneal ultrastructure of healthy female Merino sheep, which adds to the current knowledge about the ophthalmological aspects of this species and undoubtedly benefits veterinarians.

The aims of this study were to assess in vivo fertility and in vitro sperm characteristics of different sires and to identify sperm variables important for the prediction of conception rate. Multiparous Nelore cows (n = 191) from a commercial farm underwent the same timed artificial insemination (timed-AI) protocol. Three batches of frozen semen from three Angus bulls were used (n = 9). A routine semen thawing protocol was performed in the laboratory to mimic field conditions. The following in vitro sperm analyses were performed: Computer Assisted Semen Analysis (CASA), Thermal Resistance Test (TRT), Hyposmotic Swelling Test (HOST), assessment of plasma and acrosomal membrane integrity, assessment of sperm plasma membrane stability and of lipid peroxidation by flow cytometry and assessment of sperm morphometry and chromatin structure by Toluidine Blue staining. For statistical analyses, Partial Least Squares (PLS) regression was used to explore the importance of various sperm variables in the prediction of conception rate. The following in vitro sperm variables were determined to be important predictors of conception rate: total motility (TM), progressive motility (PM), TM after 2 h of thermal incubation (TM_2 h), PM after 2 h of thermal incubation (PM_2 h), Beat Cross Frequency after 2 h of thermal incubation (BCF_2 h), percentage of rapidly moving cells after 2 h of thermal incubation (RAP_2 h), intact plasma membrane evaluated by HOST, intact plasma and acrosomal membranes evaluated by flow cytometry, intact plasma membrane suffering lipid peroxidation, major defects, total defects, morphometric width/length ratio, Fourier_0 and Fourier_2 and Chromatin Heterogeneity. We concluded that PLS regression is a suitable statistical method to identify in vitro sperm characteristics that have an important relationship with in vivo bull fertility.

Streptococcus phocae is a beta-haemolytic bacterium that causes systemic infections in Atlantic salmon, Salmo salar L., cultured in southern Chile and also in seals. In this study, the host-pathogen interaction between S. phocae and seven types of cell lines (fish and mammalian) was examined using an indirect fluorescent antibody and confocal microscopy (CM). Chinook salmon embryo (CHSE-214), epithelioma papulosum cyprini (EPC), salmon head kidney (SHK-1) and Atlantic salmon kidney were used as the fish cell lines, while human cervix epithelial adenocarcinoma (HeLa), African green monkey kidney fibroblast (Cos-7) and mouse leukaemic monocyte macrophage (Raw 264.7) were included as mammalian cell lines. Streptococcus phocae type strain ATCC 51973(T) and isolates LM-08-Sp and P23 were selected as representatives from the salmon and seal host, respectively. For the CM examination, monolayers seeded on round coverslips were studied at 2- and 20-h post-inoculation (pi). The results showed that there is no common infectivity pattern between the three S. phocae strains at 2-h pi and the cell lines tested, regardless of the source of isolation (seal or salmon). All S. phocae strains could internalize and were found inside the fish and mammalian cell cytoplasm after 20-h pi. Regardless of the cells studied (fish or mammal) and incubation (2 and 20 h), S. phocae was never observed inside the nuclei. Seal and salmon isolates showed the highest number of bacteria entering into the primate cell lines (HeLa and Cos-7) from 2-h pi, while ATCC 51973(T) was not found outside or inside the HeLa and Cos-7 cells.

A morfologia das papilas linguais da cabra doméstica (Capra hircus) foi estudada por Microscopia Eletrônica de Varredura (MEV) e Microscopia de luz. Papilasfiiliformes, cônicas e lentiformes possuem função mecânica, enquanto que as papilas fungiformes e as valadas possuem função gustativa. As papilasfiiliformes estavam distribuídas por toda a extensão da língua, nas superfícies dorsal e lateral. Possuem um formato cônico com a extremidade pontiaguda direcionada caudalmente. Possuem vários processos secundários, de tamanhos diferentes, com a mesma orientação. A histologia revelou abundante tecido queratinizado. As papilas fungiformes foram observadas nas superfícies dorsal e lateral do ápice e do corpo da língua. Possuem formato semelhante a cogumelos, com epitélio corni[1]icado espesso e botões gustativos distribuídos na superfície. As papilas lentiformes se distribuíam na linha mediana da parte mais dorsal do toro, com projeções elevadas além da superfície da língua. Algumas apresentavam o formato de pirâmide e outras um formato mais achatado. As papilas valadas foram encontradas na superfície lateral da parte mais caudal do toro. Possuem formato arredondado, envolvido por um sulco pouco profundo. Possuem botões gustativos na parede lateral que se abrem no sulco. As papilas cônicas foram observadas no toro da língua e possuem formato alongado, com base larga e ponta romba. Embora as principais características morfológicas e estruturais do epitélio lingual sejam específicas da espécie, o tipo de alimentação e os hábitos alimentares podem influenciar na sua estrutura. Assim, o presente trabalho fornece uma descrição histológica e ultraestrutural da língua de cabras domésticas submetidas à alimentação baseada em gramíneas e ração.


The morphology of the lingual papillae of domestic goat (Capra hircus) was studied by scanning electron microscopy (SEM) and light microscopy (LM). Filiform, conical and lentiform papillae exert mechanical function whereas fungiform and valate papillae have gustative function. Filliform papillae were distributed all over the lateral and dorsal surfaces of the tongue. They present a conical shape with the thin ends pointed caudally; and possess several secondary processes, of different sizes, with the same orientation. Histological examination of these papillae revealed abundant queratinized tissue. Fungiform papillae were observed on the dorsal and lateral surface of the apical region and body of the tongue. Present a fungi-like shape, with a thick cornified epithelium and taste buds distributed on the surface. Lentiform papillae were distributed over the median line of the dorsal torus, presenting projections above the surface of the tongue. Some displayed pyramidal shapes; other were more flattened. Valate papillae were observed on the lateral surface of the posterior part of the torus and present a rounding shape delimited by a deep groove. These papillae possess taste buds on their lateral wall that open up on the groove. Conical papillae were observed on the lingual torus and present an extended shape, with a wide base and a blunt tip. Although the main morphological and structural characteristics of the lingual epithelium are species-specific, some features may be influenced by the type of food and alimentary habits. Therefore, the present study provides a histological and ultrastructural description of the tongue of the domestic goat fed with mixed food (grass and concentrate).

A morfologia das papilas linguais da cabra doméstica (Capra hircus) foi estudada por Microscopia Eletrônica de Varredura (MEV) e Microscopia de luz. Papilasfiiliformes, cônicas e lentiformes possuem função mecânica, enquanto que as papilas fungiformes e as valadas possuem função gustativa. As papilasfiiliformes estavam distribuídas por toda a extensão da língua, nas superfícies dorsal e lateral. Possuem um formato cônico com a extremidade pontiaguda direcionada caudalmente. Possuem vários processos secundários, de tamanhos diferentes, com a mesma orientação. A histologia revelou abundante tecido queratinizado. As papilas fungiformes foram observadas nas superfícies dorsal e lateral do ápice e do corpo da língua. Possuem formato semelhante a cogumelos, com epitélio corni[1]icado espesso e botões gustativos distribuídos na superfície. As papilas lentiformes se distribuíam na linha mediana da parte mais dorsal do toro, com projeções elevadas além da superfície da língua. Algumas apresentavam o formato de pirâmide e outras um formato mais achatado. As papilas valadas foram encontradas na superfície lateral da parte mais caudal do toro. Possuem formato arredondado, envolvido por um sulco pouco profundo. Possuem botões gustativos na parede lateral que se abrem no sulco. As papilas cônicas foram observadas no toro da língua e possuem formato alongado, com base larga e ponta romba. Embora as principais características morfológicas e estruturais do epitélio lingual sejam específicas da espécie, o tipo de alimentação e os hábitos alimentares podem influenciar na sua estrutura. Assim, o presente trabalho fornece uma descrição histológica e ultraestrutural da língua de cabras domésticas submetidas à alimentação baseada em gramíneas e ração.(AU)


The morphology of the lingual papillae of domestic goat (Capra hircus) was studied by scanning electron microscopy (SEM) and light microscopy (LM). Filiform, conical and lentiform papillae exert mechanical function whereas fungiform and valate papillae have gustative function. Filliform papillae were distributed all over the lateral and dorsal surfaces of the tongue. They present a conical shape with the thin ends pointed caudally; and possess several secondary processes, of different sizes, with the same orientation. Histological examination of these papillae revealed abundant queratinized tissue. Fungiform papillae were observed on the dorsal and lateral surface of the apical region and body of the tongue. Present a fungi-like shape, with a thick cornified epithelium and taste buds distributed on the surface. Cucullanus mycteropercae n. sp. is described from the intestine of the black grouper, Mycteroperca bonaci Poey, from the northern coast of Yucatán, México. The new species is readily distinguishable from other Cucullanus species because it possesses an ellipsoidal papilla-like structure situated medially on the anterior cloacal lip of males. Other differentiating characters include the variable position of postcloacal pair 8 in males, the subventral position of phasmids, a slightly ventrally hooked posterior end of gubernaculum, and the presence of a large, cylindroconical sclerotized tail end in both sexes. This is the fourth record of a marine cucullanid off the Yucatán Peninsula in México belonging to Cucullanus Müller, 1777.

Serum amyloid A (SAA) is a family of acute-phase proteins, recognized as important effectors of innate immunity in higher vertebrates. Under pro-inflammatory conditions, up-regulation of saa transcripts occurs not only in the liver, but also in several extrahepatic tissues of a wide variety of vertebrates. SAA is also known as the precursor to amyloid A (AA), a major component of amyloid fibrils deposited in liver, kidney and spleen of humans suffering chronic inflammatory diseases. Here we show the up-regulation of saa transcription in lesions affecting skin, adipose tissue and skeletal muscle of rainbow trout naturally and experimentally infected with Flavobacterium psychrophilum, the causative agent of cold water disease (CWD). Using an antiserum against a trout acute SAA peptide that was previously shown to specifically recognize intact recombinant trout SAA and peptides derived from it, we showed by confocal microscopy analysis extensive colocalization of SAA and thioflavin T (ThT) staining in the skeletal muscle fibers of infected fish, suggesting for the first time the presence of AA-derived aggregates in the skeletal muscle of a lower vertebrate. These findings support the idea that SAA and/or its derivatives could constitute relevant markers for fish health and also for fish meat quality control.

Synthesium pontoporiae n. comb. is redescribed, together with Synthesium tursionis and Synthesium seymouri n. comb.; the parasites were obtained from stranded and accidentally caught cetaceans. The sucker ratio (ratio between widths of the oral and ventral suckers) in S. pontoporiae was 1:1.8-3.0 (mean 1:2.2); in S. tursionis was 1:0.8-1.2; and in S. seymouri was 1:0.5-0.7. Synthesium pontoporiae differed from its congeners by additional diagnostic characters, including: oval to lobed testes; small cirrus with pyriform proximal region and flexible, tubular distal region formed by evagination of ejaculatory duct; and vitellarium in small follicles extending from the level of the seminal vesicle to the posterior extremity of the body and not forming dendritic radial bunches. Data on the morphology of adult S. pontoporiae and S. tursionis were inferred from confocal laser microscopical observations.
