The biotransformation of (4R)-(-)-carvone by Mentha pulegium (pennyroyal) leaves and its endophytic bacteria was performed in order to search for novel biocatalysts with enoate reductase activity. The obtained results clearly indicated that endophytes play an important role in the biotransformation of (4R)-(-)-carvone with pennyroyal plant tissues. The best activity was associated to the endophytic bacteria Pseudomonas proteolytica FM18Mci1 and Bacillus sp. FM18civ1. Enoate reductase activity for the reduction of (4R)-(-)-carvone and (4S)-(+)-carvone as model substrates was evaluated for each strain. Finally, both isolated strains were evaluated for the kinetic resolution of racemic carvone. The two bacteria gave (1R, 4R) or (1R, 4S)-dihydrocarvone as major products. P. proteolytica FM18Mci1 had preference for the 4S-(-)-carvone, reaching a conversion 95% in 24 h. In contrast, Bacillus sp. FM18civ1 had preference for (4R)-(-)-carvone. The results obtained in the kinetic resolution of carvone indicated that the Bacillus strain could be useful for resolving a racemic mixture of carvone.

RESUMO

Mentha pulegium (Lamiaceae) tea has been used as a traditional medicine; however, the modulatory effect of M. pulegium extracts on damage to human erythrocytes associated to t-butyl hydroperoxide (t-BHP) exposure remains to be investigated. Accordingly, we perform this study in order to test the hypothesis that aqueous and ethanolic extracts of M. pulegium could modulate the hemolysis associated to t-BHP exposure, non-protein thiol (NPSH) oxidation and lipid peroxidation (measured as thiobarbituric acid reactive substances - TBARS) in human erythrocytes. Samples were co-incubated with t-BHP (4 mmol/L) and/or aqueous or ethanolic extracts (10-1000 mg/mL) during 120 min to further analysis. We found that both extracts, when associated to t-BHP, potentiate NPSH oxidation and hemolysis. Moreover, both extracts significantly prevents against t-BHP-induced TBARS production. A significant correlation among hemolysis and NPSH levels was found. Taking together, our data points that the association of M. pulegium extracts with t-BHP culminates in toxic effect to exposed erythrocytes, besides its protective effect against t-BHP-induced TBARS production. So, we infer that the use of this extract may exert negative effect during painful crisis in sickle cell anemia. However, more studies are still necessary to better investigate/understand the mechanism(s) involved in the toxic effect resultant from this association.

RESUMO

The objective of this study was to determine the chemical composition, apparent metabolizable energy (AME), AME corrected by nitrogen retention (AMEn) and metabolization coefficients (MC and MCn) of different feeds for broilers from the western region of Paraná State. The experiment was conducted at the Physiology of Birds Laboratory at UNIOESTE. Two hundred and sixty male chicks of the Cobb 500 strain, at 21 days old, were distributed in a randomized design with 12 treatments and four replicates of five birds per experimental unit. The feed ingredients studied were ten different wheat middlings, one triticale, one wheat grain and a reference diet. Values for dry matter, nitrogen, gross energy, neutral detergent fiber, acid detergent fiber, mineral matter, calcium, phosphorus, sodium, potassium and magnesium composition were determined as well as the geometric mean diameter of the feed particles. To determine the apparent metabolizable energy (AME) and AME corrected for nitrogen balance (AMEn) the total excreta collection method was used with growing birds. The chemical composition of the feeds was variable. The values for AME and AMEn ranged from 1. 819-2. 958 and 1. 734-2. 882 kcal. kg-1, respectively. Based on these energetic values and the values for gross energy, the coefficients of metabolizability were calculated. The values ranged from 43. 26-75. 24% for MC and from 42. 28-73. 31% for MCn.(AU)

The aim of this study was to compare the yield, chemical composition and antifungal activity of the essential oil that is obtained from Mentha pulegium in different developmental stages and cultivated under controlled conditions in southern Brazil. The hydrodistillation of fresh leaves that were collected at 60, 70 and 85 days resulted inessential oil yields of 0.17 %, 0.23 % and 0.17 %, respectively. All of the essential oil samples showed antifungal activity against Cladosporium herbarum. The gas chromatograph (GC) and GC/MS analysis revealed eleven constituents: seven (pulegone, piperitenone, menthone, isomenthone, neoisomenthol, piperitone and 1.1-dimethoxy-2-nonyne) were common to three samples, while menthofuran and myrtenal were detected only in samples of the first and second stages. Pulegone was the main constituent of the essential oil samples from the first and second stages (26.65 %), followed by piperitenone (20.41; 12.60 %). The pulegone concentration increased to 31.05 % in the last collection, while the major constituent was piperitenone (36.32 %). In conclusion, the results demonstrated that M. pulegium essential oil presents potential as an antifungal agent, and its chemical composition depends on the stage of development during which it was extracted.(AU)

RESUMO

OBJECTIVES: We evaluated the relaxant activity of the essential oil of Mentha pulegium L. (EOMP) and pulegone in rat isolated tracheal and bladder smooth muscles. METHODS: ISOMETRIC contractions of isolated tracheal and bladder strips from male Wistar rats were induced by KCl (K60; 60 mm) or acetylcholine (ACh; 10 µm). EOMP and its majory compound pulegone were incubated, after contracting agent, with the tissues in cumulating concentrations. KEY FINDINGS: EOMP (3-300 µg/ml) inhibited the contractions induced by ACh and K60 in both tissues, but was more effective against the contractions induced by K60 in trachea (IC50 = 40.47 ± 3.27 µg/ml) compared with ACh. Its relaxant action rules out ganglia and NO participation. Pulegone (10(-7) to 10(-3 ) m) inhibited the contractions induced by ACh and K60 in both tissues. EOMP concentration-dependently inhibited the contractions evoked by addition of CaCl(2) in depolarised trachea, suggesting inhibition of extracellular calcium entry. CONCLUSIONS: These findings suggests that EOMP induced relaxant responses in pre-contracted smooth muscles of rat trachea and bladder, which are likely to be mediated via inhibition of calcium entry, mainly by its major compound, pulegone. These effects are coherent with the popular use of EOMP as an antispasmodic agent.

RESUMO

The aim of the present work was to determine the in vitro effect of Mentha piperita and Mentha pulegium essential oils against Echinococcus granulosus and to compare the effectiveness of both oils according to the exposure time and concentration. Although both treatments had a protoscolicidal effect, M. pulegium had a considerably stronger effect than M. piperita. Essential oil of M. pulegium produced dose- and time-dependent effects. Maximal protoscolicidal effect was observed after 12 days of incubation and reached 0% after 18 days. This lack of viability was proved during the determination of infectivity into mice. Essential oil of M. piperita produced only a time-dependent effect. At 24 days p.i., the viability of protoscoleces decreased to approximately 50%. Scanning and transmission electron microscopy (SEM and TEM) demonstrated the drug-induced ultrastructural damage. On the other hand, a loss of turgidity was detected in all M. pulegium-treated cysts respective of the drug concentration. There was a correlation between the intensity of damage and the concentration of the essential oil assayed. Studies by SEM revealed that the germinal layer of treated cysts lost the feature multicellular structure. M. pulegium essential oil showed piperitone oxide as main compound in their composition, and we suggest that this component could be responsible of the markedly anthelmintic effect detected. Our data suggest that essential oils of Mentha spp. can be a promising source of potential protoscolicidal agents. The isolation of active anthelmintic constituents is in progress and may lead to the discovery of compounds with improved therapeutic value.

This work investigated the chemical composition of the essential oils of two medicinal plants grown in Ilhéus Municipality, Bahia State, Brazil: Plectranthus amboinicus (Lour.) Spreng (Mexican mint) and Mentha pulegium L. (pennyroyal). The essential oils were extracted from fresh leaves by hydrodistillation and analyzed by GC-FID and GC-MS, in the spring and in the winter. In both seasons, P. amboinicus yielded 0.10 percent oil and had thymol as major component. M. pulegium yielded 0.20 percent and 0.09 percent oil in the spring and winter, respectively. The major components were pulegone and trans-caryophyllene, whereas borneol, menthol and piperitone were identified at small quantities. Higher menthol and isomenthol levels were detected in the winter. In addition, neoisomenthyl, menthyl and isomenthyl acetates were only observed in the winter. This work contributes to the knowledge of plant species grown in southern Bahia.

RESUMO

In a rural population of Bulnes, Bio-Bio Region of Chile, it was assessed the daily consumption of medicinal plants and drugs. Through 168 interviews the consumption of 5 medicinal plants was evaluated, considering their application, used plant parts, type of consumption, preparation, age range of people consuming mainly medicinal plants, use of other pharmacological treatments, main diseases present in the population under study and the type of drugs consumed. The obtained results demonstrate that higher consumed medicinal plants were peppermint (Mentha piperita) chamomile (Matricaria recutita), rute (Ruta graveolens), oregano (Origanum vulgare) and pennyroyal (Mentha pulegium). The consumption of medicinal plants was mainly complementary. The therapeutic use of medicinal plants in the rural population of Bulnes, is mainly digestive and it is not necessarily related to those diseases with more prevalence or most commonly observed in the population. It is important to consider future studies to determine if medicinal plants are more likely used to reduce the digestive discomfort which arises as a lateral damage of pharmaceutical drugs.

Mentha pulegium is common known as "poleo" and used for the treatment of diarrhea, headache and cough in Mexican traditional medicine. Organic extracts from aerial parts were evaluated to determine their spasmolytic action on rat isolated ileum test. Hexanic (HEMp), dichloromethanic (DEMp) and methanolic (MEMp) extracts induced a concentration-dependent (0.97 to 1000 microg/mL) antispasmodic effect on spontaneous contractions. DEMp was the most active extract; therefore, spasmolytic mechanism was investigated. This extract (200 microg/mL) induced a significant depression on cumulative concentration-response curve for carbachol and serotonin (P<0.05). Besides, extract decreased and shifted to the right KCl- and CaCl2-induced contraction curves. Moreover, pre-incubation with chlorpromazine (0.001 mM) shifted to the left the relaxant curve. Pre-treatment with L-NAME (1 mM), papaverine (0.01 mM), teophylline (0.01 mM), TEA (1 mM) and glybenclamide (0.1 mM) did not produced any changed of the relaxant curves of DEMp. Findings indicate that dichloromethanic extract of M. pulegium induced its spasmolytic effect through Ca2+-influx blockade, which may explain its traditional use against diarrhea.

RESUMO

The repeated use of permethrin and other insecticides for the control of head lice, Pediculus humanus capitis De Geer (Anoplura: Pediculidae), during past decades has resulted in the development of marked levels of resistance. Thus, new alternative insecticides are needed for the control of head lice. We studied the fumigant and repellent properties of essential oils from 16 native and exotic plants in Argentina, and 21 chemical components against permethrin-resistant head lice from Argentina. With a direct vapor-exposure bioassay, the most effective oil was from the native Myrcianthes cisplatensis Cambess (Myrtaceae) with a time to 50% knockdown (KT50) of 1.3 min, followed by exotic species, Eucalyptus cinerea F.V. Muell., Eucalyptus viminalis Labill., and Eucalyptus saligna Smith. with KT50 values of 12.0, 14.9, and 17.4 min, respectively. The most effective components were 1,8-cineole and anisole, with KT50 values of 11.1 and 12.7 min, respectively. Regression analysis of KT50 values and vapor pressures and water-partition coefficients for the essential oil components revealed that the most effective fumigants were among the more volatile components. Repellency assays indicated that the essential oil from Mentha pulegium L. and its benzyl alcohol component were the most effective repellents, having repellency indices of 75.5 and 57.8%, respectively. Thus, some Argentinean plants contain essential oils and components that function as fumigants or as repellents and thereby show potential for development of new control products for head lice.

RESUMO

The effects of the essential oil of Mentha pulegium L. (EOMP), a plant commonly known as "pennyroyal" or "poejo" that is used in folk medicine as an abortifaceant, were assessed on the isolated rat myometrium. Myometrial strips were stimulated with 10 nM oxytocin or 10 microM PGF (2alpha). EOMP (10 - 300 microg/mL) concentration-dependently and reversibly inhibited the amplitude of oscillatory contractions, being approximately 3-fold more active against contractions stimulated by oxytocin than those by PGF (2alpha) (IC (50) values of 45.7 +/- 5.6 microg/mL and 160.9 +/- 5.9 microg/mL , respectively), although the maximal inhibitory effect occurred at the same concentration (300 microg/mL ) in both cases. This action was shared by pulegone (30 - 300 microM), the principal component of the essential oil (IC (50) values of 21.8 +/- 2.1 microg/mL and 12.7 +/- 4.6 microg/mL , respectively). Nifedipine (30 nM - 30 microM) also abolished agonist-stimulated contractions, and was approximately twice and 12 times as potent as EOMP in inhibiting oxytocin- and prostaglandin F (2alpha) (PGF (2alpha))-stimulated contractions, respectively. In conclusion, our results show that the essential oil of the abortifaceant plant Mentha pulegium exerts an inhibitory effect on the contractile activity of the isolated rat myometrium. This oil shares a common effect with the voltage-dependent calcium channel (VDCC) blocker nifedipine, although ostensibly acting via a different mechanism. It thus appears that EOMP and pulegone do not exert direct toxic effects on the myometrium per se that would cause abortion, and other possibilities such as systemic metabolism of plant constituents may rather underlie the abusive use of Mentha pulegium in popular medicine.
