Caseous lymphadenitis (CLA), an infectious disease caused by Corynebacterium pseudotuberculosis in small ruminants, is highly prevalent worldwide. Economic losses have already been associated with the disease, and little is known about the host-pathogen relationship associated with the disease. The present study aimed to perform a metabolomic study of the C. pseudotuberculosis infection in goats. Serum samples were collected from a herd of 173 goats. The animals were classified as controls (not infected), asymptomatic (seropositives but without detectable CLA clinical signs), and symptomatic (seropositive animals presenting CLA lesions), according to microbiological isolation and immunodiagnosis. The serum samples were analyzed using nuclear magnetic resonance (1H-NMR), nuclear Overhauser effect spectroscopy (NOESY), and Carr-Purcell-Meiboom-Gill (CPMG) sequences. The NMR data were analyzed using chemometrics, and principal component analysis (PCA) and partial least square discriminant analysis (PLS-DA) were performed to discover specific biomarkers responsible for discrimination between the groups. A high dissemination of the infection by C. pseudotuberculosis was observed, being 74.57% asymptomatic and 11.56% symptomatic. In the evaluation of 62 serum samples by NMR, the techniques were satisfactory in the discrimination of the groups, being also complementary and mutually confirming, demonstrating possible biomarkers for the infection by the bacterium. Twenty metabolites of interest were identified by NOESY and 29 by CPMG, such as tryptophan, polyunsaturated fatty acids, formic acid, NAD+, and 3-hydroxybutyrate, opening promising possibilities for the use of these results in new therapeutic, immunodiagnosis, and immunoprophylactic tools, as well as for studies of the immune response against C. pseudotuberculosis. KEY POINTS: • Sixty-two samples from healthy, CLA asymptomatic, and symptomatic goats were screened • Twenty metabolites of interest were identified by NOESY and 29 by CPMG • 1H-NMR NOESY and CPMG were complementary and mutually confirming.

This study aimed to characterize the production and management systems of white Morada Novasheep herds and to present data on the occurrence of caseous lymphadenitis (CLA) and clinical mastitis (CM) in a population maintained in a semi-arid region. Blood samples were collected from adult females from herds of the Brazilian northeastern region. The first venous blood sample was used for a complete blood examination. The second blood sample was used to detect the presence of anti-C. pseudotuberculosis specific antibodies. The production and management systems of the herds were characterized using questionnaires, divided into three blocks of indicators: Facilities; Characterization of the production system; Health management. Body weight and hematological results were submitted to an analysis of variance, on which the fixed effect was clinical diagnosis, and the principal component analysis was performed to determine the main variables. For CLA, three herds stood out as having the highest rates of seropositivity (85.71, 79.41 and 60.45%). The same herds also had the highest occurrences of CM (9.30, 8.82 and 5.71%). The main variables that showed differences according the diagnosis of both diseases were blood granulocytes and lymphocytes. The study made it possible to observe the relationship between the best sanitary practices and the lowest occurrence rates of both CLA and CM. Este estudo teve como objetivo caracterizar os sistemas de produção e manejo de rebanhos de ovelhas Morada Nova da variedade branca e apresentar dados sobre a ocorrência de linfadenite caseosa (CLA) e mastite clínica (MC) em uma população mantida em região semiárida. Amostras de sangue foram coletadas de fêmeas adultas de rebanhos da região nordeste do Brasil. A primeira amostra de sangue venoso foi utilizada para um exame de sangue completo. A segunda amostra de sangue foi utilizada para detectar a presença de anticorpos anti-C. anticorpos específicos de pseudotuberculosis. Os sistemas de produção e manejo dos rebanhos foram caracterizados por meio de questionários, divididos em três blocos de indicadores: Instalações; Caracterização do sistema de produção; Gestão de saúde do rebanho. O peso corporal e os resultados hematológicos foram submetidos a uma análise de variância, na qual o efeito fixo foi o diagnóstico clínico, e a análise de componentes principais foi realizada para determinar as variáveis principais. Para o CLA, três rebanhos se destacaram com os maiores índices de soropositividade (85,71, 79,41 e 60,45%). Os mesmos rebanhos também apresentaram as maiores ocorrências de MC (9,30, 8,82 e 5,71%). As principais variáveis que apresentaram diferenças de acordo com o diagnóstico de ambas as doenças foram granulócitos e linfócitos. O estudo possibilitou observar a relação entre as melhores práticas sanitárias e as menores taxas de ocorrência tanto de CLA quanto de MC. As doenças apresentam ocorrência considerável nas populações de ovinos

Caseous lymphadenitis (CLA), an infectious disease caused by Corynebacterium pseudotuberculosis in goats and sheep, is highly prevalent worldwide and is characterized by economic losses in small ruminant production. Currently available techniques for clinical and laboratory diagnosis of the disease lack market availability and/or sensitivity, and therefore, infected animals can remain in the herd, serving as a source of infection for other animals. The present study aimed to verify hematological and clinical biochemistry changes in goats naturally infected by C. pseudotuberculosis. One hundred seventy-three Canindé goats were included in this study, from which blood samples and caseous lesions were collected. The animals were classified as uninfected, asymptomatic, and symptomatic according to microbiological isolation and serological assays. A high dissemination of the infection was observed in the herd, with 86.13% of positive animals, being 74.57% asymptomatic and 11.56% symptomatic. In the hemogram and clinical biochemistry analyses, the only statistical difference found was a higher level of serum urea in asymptomatic individuals than in non-infected animals. In addition, this study points to the possibility of chronic CLA being potentially reflected in hepatic and renal biochemical markers.

Corynebacterium pseudotuberculosis is a bacillus that causes caseous lymphadenitis in small ruminants, leading to great losses to rural producers; thus, an efficient diagnosis is necessary for using disease control measures. This study aimed to evaluate the antigenic potential of four C. pseudotuberculosis recombinant proteins (rSodC, rPknG, rNanH, and rSpaC) against sera of goat and sheep experimentally infected with one of three different C. pseudotuberculosis strains. Goats were infected with CAP76 or CAP21 strain (n = 10), sheep with VD57 strain (n = 6), and a group of not-infected animals (goats and sheep) were kept as a healthy control (healthy n = 12). Sera were collected at 0, 14, 60, 90, 180, or 190 days after inoculation for antigenicity testing using Western blotting and enzyme-linked immunosorbent assay (ELISA) techniques. Cross-reactivity tests with recombinant proteins were performed in goat serum experimentally vaccinated with Nocardia sp. or Rhodococcus equi bacterin. The rSodC protein showed discriminatory antigenic reactivity with a statistically significant difference against three different C. pseudotuberculosis strains evaluated in goats and sheep samples, while rPknG showed statistical significance only against two C. pseudotuberculosis strains evaluated in goats. rSodC was proved to be a strong candidate as a tool for diagnosis of C. pseudotuberculosis infection, once it was able to recognize antibodies against all strains evaluated in goats and sheep.

Background: Mycobacteriosis is caused by bacteria belonging to the genus Mycobacterium, with considerable zoonoticpotential and risk to public health. Infection in dogs is rare and is usually associated with immunosuppression, resultingfrom eating meat or contact with contaminated soil or fomites. Dogs are also known as potential sources for the spread ofatypical tuberculosis in humans and other animals. This paper aims to describe the clinical, cytological, histopathological,and molecular findings of a male canine seen at University Veterinary Hospital of Cuiabá, Mato Grosso, with generalizedlymphadenomegaly associated Mycobacterium intracellulare infection.Case: A 2-year-old male Lhasa Apso dog was referred to the University Veterinary Hospital in Cuiabá city, located in theMidwest region of Brazil. The patient had a history of intermittent claudication of the left pelvic limb for approximately6 months and lymphadenomegaly with progression for approximately 2 months. The dog had wheezing and generalizedlymphadenopathy (submandibular, axillary, and popliteal lymph nodes); cryptorchidism was also observed. A completeblood count revealed nonspecific results, and in the serum biochemical profile, the values of urea, creatinine, albumin, andalanine aminotransferase were within the reference range. No changes were observed on the radiography of the femurotibiopatellar joints. Considering the generalised lymphadenopathy, fine needle aspiration cytology and histopathologicalexamination through biopsy of the lymph nodes was performed. On the cytology and histopathology, numerous negativeimages of moderately refringent bacillary structures distending the cytoplasm from the macrophages was found. Background: Caseous Lymphadenitis (CL) is a chronic infectious disease caused by the bacterium Corynebacteriumpseudotuberculosis, which is considered the main agent responsible for abscess lesions. In the visceral form it can affect theinternal organs of sheep and goats, which could negatively affect animal health and cause large economic losses for producers.Case: This study aims to report a case of intestinal CL in sheep, with suspected diagnosis during physical examinationand identification during the performance of the oophorectomy procedure, adopted as a management approach. It is amixed breed sheep, aged over 5 years; weight 28 kg; emaciated on physical examination; with pale pink and moist eyelidmucosa; heart and respiratory rate: 81 beats/min and 22 movements/min, respectively; body temperature 39.2°C; ruminalmovements at 1 movement/min; without identification of lymphadenomegaly on palpation, however, it was observed thatthe right submandibular lymph node presented tissue retraction compatible with the healing process. For the surgical procedure, an 18-h fast was used and pre-anesthetic medication with 2% xylazine (0.1 mg/kg), 10% ketamine (5 mg/kg) and50 mg/mL tramadol (2 mg/kg) administrated intramuscularly. The animal was placed in the left lateral decubitus position,then was performed trichotomy and epidural administration of 2% lidocaine (4 mg/kg) and maintenance with propofol10 mg/mL intravenous dose-effect and oxygen mask 3 L/min, antibiotic prophylaxis was performed with 10% enrofloxacin(2.5 mg/kg). O objetivo deste trabalho foi avaliar a eficácia do florfenicol na dose usualmente empregada em equinos de 22 mg/kg pelas vias intravenosa, intramuscular e oral para o tratamento de adenite equina por Streptococcus equi. subsp. equi, usando a modelagem farmacocinética/farmacodinâmica (PK/PD ­ Pharmacokinetic/Pharmacodynamic) e a simulação de Monte Carlo. Foi realizada uma simulação de Monte Carlo a partir dos parâmetros PK, logo depois, efetuou-se a modelagem PK/PD para determinar as taxas de eficácia do antimicrobiano para o tratamento dessa infecção bacteriana, de acordo com o valor da concentração inibitória mínima (CIM), em um intervalo de CIM de 0,125 ­ 4 µg/mL. Pela via intravenosa, a probabilidade de erradicação bacteriana foi de 100% para CIM até 0,5 µg/mL e efeito bacteriostático com probabilidades de 99% e 80% para CIMs de 2 e 4 µg/mL, respectivamente. Já pelas vias intramuscular e oral a probabilidade de se atingir o índice de erradicação bacteriológica foi de 100% para CIM de até 0,5 µg/mL, contudo, atinge valores de 80% e 81%, respectivamente, para CIM de 1 µg/mL considerando o efeito bactericida (p<0,01). Portanto, através desse estudo é evidenciado a eficácia do florfenicol até a CIM de 0,5 µg/mL para as três vias de administração citadas, entretanto, para CIMs superiores a esse valor, é imprescindível o ajuste da dose farmacológica, evitando falhas na terapêutica e possível resistência microbiana.


The objective of this study was to evaluate the efficacy of florfenicol at the dose usually used in horses of 22 mg/kg by intravenous, intramuscular and oral routes for the treatment of equine adenitis caused by Streptococcus equi. subsp. equi, using Pharmacokinetic/Pharmacodynamic (PK/PD) modeling and Monte Carlo simulation. A Monte Carlo simulation was performed from the PK parameters, then PK/PD modeling was performed to determine the antimicrobial efficacy rates for the treatment of this bacterial infection, according to the minimum inhibitory concentration (MIC) value, in a MIC range of 0.125 - 4 µg/mL. Intravenously, the probability of bacterial eradication was 100% for MICs up to 0.5 µg/mL, and the bacteriostatic effect was 99% and 80% for MICs of 2 and 4 µg/mL, respectively. However, for the intramuscular and oral routes, the probability of reaching the bacteriologic eradication index was 100% for MICs of up to 0.5 µg/mL, however, it reaches values of 80% and 81%, respectively, for MICs of 1 µg/mL considering the bactericidal effect (p<0.01). Therefore, through this study the efficacy of florfenicol is evidenced up to the MIC of 0.5 µg/mL for the three routes of administration cited, however, for MICs higher than this value, it is essential to adjust the pharmacological dose, avoiding failures in therapy and possible microbial resistance.

Caseous lymphadenitis (CLA) is an infectious chronic disease responsible for economic losses in sheep and goat breeding worldwide. CLA has no effective treatment, evidencing the vaccination schedule as the best control strategy. Although some commercial vaccines have been available, none of them provides total protection, which is sometimes insufficient and does not reach the same efficiency when compared in sheep and goats. They also have questionable safety levels and side effects. In light of this, several experimental vaccines are in development in order to improve safety, reproducibility, and protective immune response against the etiologic agent of CLA, Corynebacterium pseudotuberculosis. In this review, we discussed aspects as antigen, adjuvant, routes of administration, protection level, and animal models used in CLA vaccine development, as well the challenges and future perspectives. KEY POINTS: Caseous lymphadenitis (CLA) does not have an appropriate commercial vaccine. Different experimental vaccines are in development aiming to protect against Corynebacterium pseudotuberculosis. An ideal vaccine for CLA is necessary for the disease control.

Caseous lymphadenitis (CL) in sheep is a chronic contagious disease caused by Corynebacterium pseudotuberculosis, commonly characterized by abscess formation in peripheral lymph nodes and disseminated infections. Nonetheless, other microorganisms, including with zoonotic relevance, can be isolated from CL-resembling lymph nodes. Currently, mycobacteria have been reported in visceral granulomatous lesions in small ruminants, a fact that poses a public health issue, particularly in slaughtered sheep intended for human consumption. Cytology using fine needle aspiration and microbiological culturing are suitable tests for routine diagnostic, whereas present drawbacks and molecular methods have been confirmatory. Data about the occurrence of mycobacteria in both lymph nodes with aspect of CL and apparently healthy visceral nodes of sheep slaughtered for human consumption are scarce. In this study, 197 visceral lymph nodes of sheep showed lymphadenitis and 202 healthy visceral lymph nodes of slaughtered sheep intended for human consumption were submitted to conventional bacteriological diagnosis, mycobacteria culturing, and cytological evaluation. Compatible Corynebacterium isolates were subjected to multiplex PCR targeting 16S rRNA, rpoB, and pld genes to detect C. pseudotuberculosis. Based on microbiological identification, C. pseudotuberculosis (86/197; 43.7%), streptococci γ-hemolytic (17/197; 8.6%), and Trueperella pyogenes (12/197; 6.1%) were prevalent in lymph nodes with abscesses, as opposed to staphylococci (53/202; 26.2%) in apparently healthy lymph nodes. No mycobacteria were isolated. Cytology identified 49.2% (97/197) Gram-positive pleomorphic organisms (coryneform aspect). Multiplex PCR confirmed genetic material of C. pseudotuberculosis in 74.4% (64/86) of the samples with C. pseudotuberculosis isolation and 66% (64/97) samples with cytological coryneform aspect (κ = 86.78%; 95% CI = 79.87-93.68%). These findings emphasize the prevalence of C. pseudotuberculosis in abscess formation among peripheral lymph nodes of sheep. Other bacteria were also identified in lymph nodes sampled that resembling C. pseudotuberculosis-induced infections that may difficult the diagnosis. Multiplex PCR revealed a valuable assay to detect C. pseudotuberculosis, in addition to routine methods applied to CL-diagnosis. No mycobacteria were identified in lymph nodes sampled, with and without apparent lesions. Nonetheless, due to public health impacts, this pathogen should be considered as a differential diagnosis of C. pseudotuberculosis-induced infections during inspection procedures of slaughtered sheep intended for human consumption.

O objetivo deste trabalho foi avaliar a eficácia do florfenicol na dose usualmente empregada em equinos de 22 mg/kg pelas vias intravenosa, intramuscular e oral para o tratamento de adenite equina por Streptococcus equi. subsp. equi, usando a modelagem farmacocinética/farmacodinâmica (PK/PD Pharmacokinetic/Pharmacodynamic) e a simulação de Monte Carlo. Foi realizada uma simulação de Monte Carlo a partir dos parâmetros PK, logo depois, efetuou-se a modelagem PK/PD para determinar as taxas de eficácia do antimicrobiano para o tratamento dessa infecção bacteriana, de acordo com o valor da concentração inibitória mínima (CIM), em um intervalo de CIM de 0,125 4 μg/mL. Pela via intravenosa, a probabilidade de erradicação bacteriana foi de 100% para CIM até 0,5 μg/mL e efeito bacteriostático com probabilidades de 99% e 80% para CIMs de 2 e 4 μg/mL, respectivamente. Já pelas vias intramuscular e oral a probabilidade de se atingir o índice de erradicação bacteriológica foi de 100% para CIM de até 0,5 μg/mL, contudo, atinge valores de 80% e 81%, respectivamente, para CIM de 1 μg/mL considerando o efeito bactericida (p<0,01). Portanto, através desse estudo é evidenciado a eficácia do florfenicol até a CIM de 0,5 μg/mL para as três vias de administração citadas, entretanto, para CIMs superiores a esse valor, é imprescindível o ajuste da dose farmacológica, evitando falhas na terapêutica e possível resistência microbiana.(AU)


The objective of this study was to evaluate the efficacy of florfenicol at the dose usually used in horses of 22 mg/kg by intravenous, intramuscular and oral routes for the treatment of equine adenitis caused by Streptococcus equi. subsp. equi, using Pharmacokinetic/Pharmacodynamic (PK/PD) modeling and Monte Carlo simulation. A Monte Carlo simulation was performed from the PK parameters, then PK/PD modeling was performed to determine the antimicrobial efficacy rates for the treatment of this bacterial infection, according to the minimum inhibitory concentration (MIC) value, in a MIC range of 0.125 - 4 μg/mL. Intravenously, the probability of bacterial eradication was 100% for MICs up to 0.5 μg/mL, and the bacteriostatic effect was 99% and 80% for MICs of 2 and 4 μg/mL, respectively. However, for the intramuscular and oral routes, the probability of reaching the bacteriologic eradication index was 100% for MICs of up to 0.5 μg/mL, however, it reaches values of 80% and 81%, respectively, for MICs of 1 μg/mL considering the bactericidal effect (p<0.01). Therefore, through this study the efficacy of florfenicol is evidenced up to the MIC of 0.5 μg/mL for the three routes of administration cited, however, for MICs higher than this value, it is essential to adjust the pharmacological dose, avoiding failures in therapy and possible microbial resistance.(AU)

Caseous lymphadenitis (CLA) caused by the pathogenic bacterium, Corynebacterium pseudotuberculosis, results in economic loss in goat farming. Its prevalence in Brazilian herds varies from 5 to 50%, generating expenses with treatment, loss in production, carcass and organ condemnation, and reduced price of the hide. This study aimed to describe the distribution of lesions and quantify the loss associated with CLA in goat breeding due to condemnation in a slaughterhouse located in a semiarid region. The study was conducted at the municipal slaughterhouse of the city of Patos, State of Paraíba, Northeastern Brazil, where goats from this intermediate geographical region are slaughtered. In 2017, 3,662 animals were slaughtered, an average of 305 per month. During the study period, from March to August 2017, 304 goats of both sexes and different ages of undefined breed were assessed. In the ante-mortem examination, inspection and palpation of the superficial lymph nodes was done; in the post-mortem examination, organ and viscera were assessed. The caseous material collected was sent for microbiological analysis. Of the 304 individuals, 227 [74.67% (95% CI: 69.50-79.23)] did not have any lesions, while 77 [25.33% (95% CI: 20.77-30.50)] showed abscesses suggestive of CLA, which was confirmed in 65 goats [84.41% (95% CI: 76.30-92.50)]. Sex (P = 0.044) and age (P = 0.002) were associated with infection. While the sex of the animals affected carcass and viscera weights and carcass yield, their age affected live weight, carcass, organ, and viscera weights; conversely, the incidence of CLA did not affect these variables. The average price in Real (R$) of a kilogram (kg) of goat meat cuts and “offal” was used to calculate the economic loss. Considering the average carcass weight (11,485 kg), organ weight (1,085 kg), and viscera weight (2,013 kg), 4,433.232 kg of meat was produced, resulting in R$ 72,945.43. The condemnations resulted in the loss of 269.894 kg, which is equivalent to R$ 4,540.33. CLA is responsible for a negative impact of 6.09% on production. Implementing control measures for this disease is important to increase the financial return in goat farming.(AU)


Linfadenite Caseosa (LC) causada pela bactéria patogênica Corynebacterium pseudotuberculosis resulta em perda econômica na caprinocultura. No rebanho brasileiro sua prevalência varia de 5 a 50%, gerando gastos com tratamento, queda de produção, condenação de carcaça e órgãos, e desvalorização da pele. O objetivo deste trabalho foi quantificar a perda associada à LC na caprinocultura de corte por condenação em abatedouro do semiárido, bem como caracterizar a distribuição das lesões. O trabalho foi realizado no Abatedouro Municipal da cidade de Patos, Estado da Paraíba, Nordeste do Brasil, onde ocorre abate de caprinos provenientes desta região geográfica intermediária. No ano de 2017 foram abatidos 3.662 animais, uma média de 305 por mês. Durante o período de estudo, março a agosto do mesmo ano, avaliou-se 304 caprinos sem padrão racial definido, ambos os sexos e idades diversas. No exame ante mortem foram submetidos à inspeção e palpação de linfonodos superficiais; post mortem, órgãos e vísceras. O material caseoso coletado foi submetido à análise microbiológica para diagnóstico. Dos 304 indivíduos, 227 [74,67% (IC 95%: 69,50-79,23)] não apresentaram qualquer lesão, enquanto 77 [25,33% (IC 95%: 20,77-30,50)] evidenciaram abscessos sugestivos de LC, com confirmação em 65 [84,41% (IC 95%: 76,30-92,50)]. Constatou-se associação de sexo (P = 0,044) e idade (P = 0,002) à infecção. Foi verificado efeito do sexo dos animais nas variáveis peso de carcaça, peso de vísceras e rendimento de carcaça, bem como houve efeito da idade nas variáveis peso vivo, peso de carcaça, peso de órgãos e peso de vísceras; no entanto, não houve efeito da ocorrência de LC nessas variáveis. Para cálculo de perda econômica adotou-se o preço médio em Real (R$) do quilograma (kg) dos cortes de carne caprina, e “miúdos”. Considerando o peso médio de carcaça (11,485 kg), órgãos (1,085 kg) e vísceras (2,013 kg), houve produção de 4.433,232 kg, resultando em R$ 72.945,43. As condenações subtraíram 269,894 kg, equivalente a R$ 4.540,33. Atribui-se à LC um impacto negativo de 6,09% na produção. Implementar medidas de controle para essa doença é importante para que se amplie o retorno financeiro à atividade.(AU)

A linfadenite caseosa (LC) é uma enfermidade que acomete pequenos ruminantes, caracterizada clinicamente por lesões abscedantes em linfonodos, pele e órgãos, causada principalmente pela bactéria Corynebacterium pseudotuberculosis. O presente estudo investigou a prevalência da LC em criatórios de pequenos ruminantes do estado do Amazonas, bem como a sensibilidade microbiana in vitro de C. pseudotuberculosis aos antimicrobianos. Foi realizado exame clínico em 1.280 animais, dos quais 4,53% (58/1.280) dos animais apresentavam abscessos e/ou cicatrizes na pele e/ou linfonodos. No perfil de sensibilidade in vitro aos antimicrobianos, 100% dos isolados de C. pseudotuberculosis obtidos de ovinos foram sensíveis à ampicilina (8/8), amoxicilina/ácido clavulânico (8/8), cefotaxima (8/8), ciprofloxacina (8/8), norfloxacino (8/8), sulfazotrim e tetraciclina (8/8). Nos isolados de C. pseudotuberculosis obtidos de caprinos, verificou-se 100% de sensibilidade in vitro diante da amoxicilina/ácido clavulânico (5/5), cefotaxima (5/5), ciprofloxacina (5/5) e norfloxacino (5/5). Ratifica-se a importância do primeiro isolamento do agente em pequenos ruminantes no estado, bem como o primeiro estudo epidemiológico na Região Norte, constatando que a enfermidade possui baixa prevalência em ovinos e caprinos no Amazonas.(AU)

El presente estudio investigó la prevalencia de LC en pequeñas granjas de rumiantes en el estado de Amazona, así como la sensibilidad microbiana in vitro de C. pseudotuberculosis a los antimicrobianos. Se realizó un examen clínico en 1.280 animales evaluados, de los cuales 4.53% (58 / 1,280) de los animales tenían abscesos y / o cicatrices en la piel y / o ganglios linfáticos. En el perfil de sensibilidad in vitro a los antimicrobianos, el 100% de los aislados de C. pseudotuberculosis obtenidos de ovejas eran sensibles a ampicilina (8/8), amoxicilina / ácido clavulánico (8/8), cefotaxima (8/8), ciprofloxacina (8 / 8), norfloxacino (8/8), sulfazotrim y tetraciclina(8/8). Caseous lymphadenitis (CL) is one of the main infectious diseases in sheep, responsible for great economic losses, mainly in Northeast region of Brazil, which has the largest sheep flock in the country. Thus, the aim of this survey was to determine the factors associated with the seroprevalence of this disease in sheep from Northeastern Brazil using a planned sample of flocks and animals. Samples were collected from 2312 adult and young sheep of both sex from 196 farms, located in 51 municipalities in five Northeastern Brazilian states (Ceará, Paraíba, Piauí, Rio Grande do Norte, and Sergipe). Serological diagnosis was performed using the indirect ELISA technique. Factors associated with seropositivity were identified by variables extracted from epidemiological questionnaires administered to breeders, using univariable and multivariable analyses. Factors associated with prevalence were purebred sheep (Prevalence ratio - PR = 1.189; P = 0.017), ram acquisition from animal expositions (PR = 1.192; P = 0.020), offspring is separated from ewes (PR = 1.132; P = 0.048), water supplied to sheep from ponds (PR = 1.365; P = 0.002), and delayed disposal of infected animals (PR = 1.263; P = 0.027). Flock-level and animal-level prevalences were 93.88 % (95 % CI = 89.60-96.46%) and 37.46 % (95 % CI = 35.51-39.45%), respectively. In view of the high prevalence found here and based on the associated factors, the implementation of an efficient CL control programme in Northeastern Brazil is recommended.

Caseous lymphadenitis (CLA) is an infectious disease caused by Corynebacterium pseudotuberculosis in small ruminants and is characterized by the development of granulomas in the lymph nodes, spleen, liver, and lungs. Although little is known about the host-pathogen relationship of this bacterium, it was previously reported that the pathogen's lipids are important for its taxonomic classification and survival inside macrophages. However, there are no studies regarding the composition of these molecules. In this study, cell wall glycolipids from two C. pseudotuberculosis strains presenting different virulence profiles were purified and its composition was characterized. A difference was observed between the electrophoretic and chromatogram profiles for cell wall components from the two strains, mainly among molecules with low molecular weights. IgM from sheep with acute CLA recognized antigens with an estimated molecular weight of 11 kDa of the low-pathogenicity strain, while low-molecular weight antigens from the high-pathogenicity strain presented a lower recognition by these antibodies. Mass spectrometry analysis showed that the cell wall of the high-pathogenicity strain contained glycolipids with high amounts of unsaturated fatty acids and glycerophosphoinositols, which may contribute to the capacity of this strain to cause severe disease. In conclusion, it is indicated that cell wall non-protein antigens can play a key role in C. pseudotuberculosis virulence.

Corynebacterium pseudotuberculosis is a Gram-positive bacterium that causes caseous lymphadenitis, a disease that predominantly affects sheep, goat, cattle, buffalo, and horses, but has also been recognized in other animals. This bacterium generates a severe economic impact on countries producing meat. Gene expression studies using RNA-Seq are one of the most commonly used techniques to perform transcriptional experiments. Computational analysis of such data through reverse-engineering algorithms leads to a better understanding of the genome-wide complexity of gene interactomes, enabling the identification of genes having the most significant functions inferred by the activated stress response pathways. In this study, we identified the influential or causal genes from four RNA-Seq datasets from different stress conditions (high iron, low iron, acid, osmosis, and PH) in C. pseudotuberculosis, using a consensus-based network inference algorithm called miRsigand next identified the causal genes in the network using the miRinfluence tool, which is based on the influence diffusion model. We found that over 50% of the genes identified as influential had some essential cellular functions in the genomes. In the strains analyzed, most of the causal genes had crucial roles or participated in processes associated with the response to extracellular stresses, pathogenicity, membrane components, and essential genes. This research brings new insight into the understanding of virulence and infection by C. pseudotuberculosis.

In the in vitro sensitivity profile to antimicrobials, 100% of C. pseudotuberculosis isolates obtained from sheep were sensitive to ampicillin (8/8), amoxicillin / clavulanic acid (8/8),cefotaxime (8/8), ciprofloxacin (8 / 8), norfloxacin (8/8), sulfazotrim and tetracycline (8/8). In isolates of C. pseudotuberculosis obtained from goats, there was 100% sensitivity in vitro to amoxicillin / clavulanic acid (5/5), cefotaxime (5/5), ciprofloxacin (5/5) and norfloxacin (5/5). The importance of the first isolation of the agent in small ruminants in the state is ratified, as well as the first epidemiological study in the North Region, confirming that the disease has a low prevalence in sheep and goats in Amazonas.


La linfadenitis caseosa (LC) es una enfermedad que afecta a pequeños rumiantes, caracterizada clínicamente por lesiones de absceso en los ganglios linfáticos, la piely los órganos, causada principalmente por la bacteria Corynebacterium pseudotuberculosis. El presente estudio investigó la prevalencia de LC en pequeñas granjas de rumiantes en el estado de Amazona, así como la sensibilidad microbiana in vitro de C. pseudotuberculosis a los antimicrobianos. Se realizó un examen clínico en 1.280 animales evaluados, de los cuales 4.53% (58 / 1,280) de los animales tenían abscesos y / o cicatrices en la piel y / o ganglios linfáticos. En el perfil de sensibilidad in vitro a los antimicrobianos, el 100% de los aislados de C. pseudotuberculosis obtenidos de ovejas eran sensibles a ampicilina (8/8), amoxicilina / ácido clavulánico (8/8), cefotaxima (8/8), ciprofloxacina (8 / 8), norfloxacino (8/8), sulfazotrim y tetraciclina(8/8). En los aislados de C. pseudotuberculosis obtenidos de cabras, 100% de sensibilidad in vitro a amoxicilina / ácido clavulánico (5/5), cefotaxima (5/5), ciprofloxacina (5/5) y norfloxacino (5/5). Se ratifica la importancia del primer aislamiento del agente en pequeños rumiantes en el estado, así como el primer estudio epidemiológico en la Región Norte, lo que confirma que la enfermedad tiene una baja prevalencia en ovejas y cabras en Amazona.

The objective of this study was to determine the frequency of seropositivity Corynebacterium pseudotuberculosis in sheep in five states of northeastern Brazil, using an indirect enzyme-linked immunosorbent assay (i-ELISA). Young and adult sheep of both sexes were used. Blood samples were collected from 2638 sheep from 223 herds across all states. For the i-ELISA, antigens produced from the strain of C. pseudotuberculosis BRM 029971, a bacterial isolate from the Northeast region of Brazil, were used. Sensitivity and specificity indexes were calculated for the validation of the test, using as reference 49 and 134 serum samples from sheep known to be positive and negative, respectively. The i-ELISA presented four false-negative and four false-positive results, showing a specificity of 97.01%, a sensitivity of 91.84%, and an accuracy of 95.63%. These results were calculated based on an optical density (OD) cutoff point = 0.138. Of the 2638 sheep tested, 996 (37.76%, 95% CI = 35.93-39.62%) were seropositive, and of the 223 evaluated herds, 210 (94.17%, 95% CI = 90.28-96.56%) seropositive. The i-ELISA showed adequate sensitivity and specificity, proving to be a useful tool in the diagnosis of caseous lymphadenitis (CLA) in sheep. Infection by C. pseudotuberculosis, determined by serology, is disseminated in the sheep herds in the states of Northeast Brazil. Thus, there is a need to implement effective control measures that prevent the spread of infectious agents.
