RESUMO

Cyclodextrin glycosyltransferases (CGTases) are important enzymes in the biotechnology field because they catalyze starch conversion into cyclodextrins and linear oligosaccharides, which are used in food, pharmaceutical and cosmetic industries. The CGTases are classified according to their product specificity in α-, ß-, α/ß- and γ-CGTases. As molecular markers are the preferred tool for bacterial identification, we employed six molecular markers (16S rRNA, dnaK, gyrB, recA, rpoB and tufA) to test the identification of a CGTase-producing bacterial strain (DF 9R) in a phylogenetic context. In addition, we assessed the phylogenetic relationship of CGTases along bacterial evolution. The results obtained here allowed us to identify the strain DF 9R as Paenibacillus barengoltzii, and to unveil a complex origin for CGTase types during archaeal and bacterial evolution. We postulate that the α-CGTase activity represents the ancestral type, and that the γ-activity may have derived from ß-CGTases.

La glucosiltranferasa B es una enzima producida por Streptococcus mutans, que a partir de la sacarosa, cataliza la síntesis de glucanos insolubles los cuales dan soporte a la biopelícula, siendo uno de los principales factores de virulencia para la generación de la caries dental. Sin embargo, no se ha esclarecido su papel en los individuos libre de caries, portadores delmicroorganismo. El objetivo de este estudio fue determinar la producción de glucosiltransferasa B y la producción de glucanos por cepas de Streptococcus mutans aisladas de biopelícula de 30individuos libres de caries. Las cepas fueron cultivadas en caldo Todd Hewitt y las proteínas extracelulares fueron obtenidas por precipitación con sulfato de amonio las proteínas asociadas amembrana por extracción con urea. La presencia de GtfB fue determinada por peso molecular por SDS–PAGE, confirmada por Western Blot utilizando un anticuerpo específico y la producciónde polisacáridos por separación electroforética, incubación con sacarosa y coloración de Schiff. Los resultados muestran que el 96.7 por ciento de las cepas de Streptococcus mutans producen una banda a la altura del peso molecular correspondiente a las Gtf,de las cuales son reactivas por western blot el 63.4 por ciento El 93.3 por cientode las cepas producen polisacáridos. Conclusiones: la cepas de Streptococcus mutans aisladas de biopelícula de individuos sanos producen factores de virulencia asociados a la caries dental como glucosiltransferasa B y glucanos lo que indica que hay condiciones en la cavidad oral diferentes a estos factores que mantienen al individuo libre de caries dental, los cuales deben ser investigados en la búsqueda de estrategias para controlar la enfermedad.

Gangliosides are a subfamily of complex glycosphingolipids (GSLs) with important roles in many biological processes. In this study, we report the cDNA cloning, functional characterization, and the spatial and temporal expression of Xlcgt and Xlgd3 synthase during Xenopus laevis development. Xlcgt was expressed both maternally and zigotically persisting at least until stage 35. Maternal Xlgd3 synthase mRNA could not be detected and showed a steady-state expression from gastrula to late tailbud stage. Xlcgt is mainly present in involuted paraxial mesoderm, neural folds, and their derivatives. Xlgd3 synthase transcripts were detected in the dorsal blastoporal lip, in the presumptive neuroectoderm, and later in the head region, branchial arches, otic and optic primordia. We determined the effect of glycosphingolipid depletion with 1-phenyl-2-palmitoyl-3-morpholino-1-propanol (PPMP) in mesodermal layer. PPMP-injected embryos showed altered expression domains in the mesodermal markers. Our results suggest that GSL are involved in convergent-extension movements during early development in Xenopus.

Sucrose metabolism and the role of sucrose synthase were investigated in the fruit tissues (pericarp, perisperm, and endosperm) of Coffea arabica during development. Acid invertase, sucrose phosphate synthase, and sucrose synthase activities were monitored and compared with the levels of sucrose and reducing sugars. Among these enzymes, sucrose synthase showed the highest activities during the last stage of endosperm and pericarp development and this activity paralleled closely the accumulation of sucrose in these tissues at this stage. Carbon partitioning in fruits was studied by pulse-chase experiments with (14)C-sugars and revealed high rates of sucrose turnover in perisperm and endosperm tissues. Additional feeding experiments with (14)CO(2) showed that leaf photosynthesis contributed more to seed development than the pericarp in terms of photosynthate supply to the endosperm. Sugar analysis, feeding experiments, and histological studies indicated that the perisperm plays an important role in this downloading process. It was observed that the perisperm presents a transient accumulation of starch which is degraded as the seed develops. Two full-length cDNAs (CaSUS1 and CaSUS2) and the complete gene sequence of the latter were also isolated. They encode sucrose synthase isoforms that are phylogenetically distinct, indicating their involvement in different physiological functions during cherry development. Contrasting expression patterns were observed for CaSUS1 and CaSUS2 in perisperm, endosperm, and pericarp tissues: CaSUS1 mRNAs accumulated mainly during the early development of perisperm and endosperm, as well as during pericarp growing phases, whereas those of CaSUS2 paralleled sucrose synthase activity in the last weeks of pericarp and endosperm development. Taken together, these results indicate that sucrose synthase plays an important role in sugar metabolism during sucrose accumulation in the coffee fruit.
