1. Based on the hypothesis that 25-hydroxycholecalciferol (25-OH-D3) inclusion would optimise dietary mineral digestibility and ameliorate growth performance and bone mineralisation in available phosphorus (AvP) deficient-fed broilers, a trial was conducted to evaluate its effect on diets with different levels of AvP.2. Broilers aged 1-21 d were randomly assigned one of the eight treatments, consisting of four dietary levels of AvP (0.45%, 0.42%, 0.39%, and 0.36%) and with or without supplementation with 25-OH-D3 at 69 µg/kg of feed. All diets contained 100 µg/kg of vitamin D3 (cholecalciferol).3. The addition of 25-OH-D3 resulted in higher feed intake and body weight gain, and lower FCR (P < 0.05) compared to non-supplemented diets, whereas AvP levels had a quadratic effect only on feed intake. There were no interactions between treatment factors.4. Increasing AvP levels linearly reduced the ileal digestibility of Ca and P (P < 0.01) and supplementing 25-OH-D3 increased both Ca and P ileal digestibility (P < 0.05), without any interactions observed for ileal digestibility.5. There was an interaction, whereby 25-OH-D3 inclusion increased serum metabolites in broilers fed 0.36% to 0.42% AvP compared to the non-supplemented diets (P < 0.001), whereas, at 0.45% AvP, diets with or without 25-OH-D3 had similar results.6. The P content in bone linearly increased in line with AvP levels (P < 0.05) and supplementation of 25-OH-D3 increased ash bone content (P < 0.001).7. Broilers can benefit from 25-OH-D3 supplementation combined with cholecalciferol with regard to Ca and P utilisation and vitamin D status, allowing for a reduction of dietary AvP levels down to 0.36% without impairing growth performance or bone status.

To reduce P excretion and increase the sustainability of poultry farms, one needs to understand the mechanisms surrounding P metabolism and its close link with Ca metabolism to precisely predict the fate of dietary P and Ca and related requirements for birds. This study describes and evaluates a model developed to estimate the fate of Ca and P consumed by broilers. The Ca and P model relies on three modules: (1) digestion of Ca and P; (2) dynamics of Ca and P in soft tissue and feathers; and (3) dynamics of body ash. Exogenous phytase affects the availability of Ca and P; thus, to predict the absorption of those minerals, the model also accounts for the effect of phytase on Ca and P digestibility. We used a database to estimate the consequences of dietary Ca, P, and phytase over feed intake response. This study followed a four-step process: (1) Ca and P model development and its coupling with a growth broiler model; (2) model behavior assessment; (3) sensitivity analysis to identify the most influential parameters; and (4) external evaluation based on three databases. The proportion of P in body protein and the Ca to P ratio in bone are the most sensitive parameters of P deposition in soft tissue and bone, representing 91 and 99% of the total variation. The external evaluation results indicated that body water and protein had an overall mean square prediction error (rMSPE) of 7.22 and 12.3%, respectively. The prediction of body ash, Ca, and P had an rMSPE of 7.74, 11.0, and 6.56%, respectively, mostly errors of disturbances (72.5, 51.6, and 90.7%, respectively). The rMSPE for P balance was 13.3, 18.4, and 22.8%, respectively, for P retention, excretion, and retention coefficient, with respective errors due to disturbances of 69.1, 99.9, and 51.3%. We demonstrated a mechanistic model approach to predict the dietary effects of Ca and P on broiler chicken responses with low error, including detailed simulations to show the confidence level expected from the model outputs. Overall, this model predicts broilers' response to dietary Ca and P. The model could aid calculations to minimize P excretion and reduce the impact of broiler production on the environment. A model inversion is ongoing that will enable the calculation of Ca and P dietary quantities for a specific objective. This will simplify the use of the model and the feed formulation process.

Introducción: Los GOS son prebióticos naturales presentes en la leche materna que pue-den obtenerse enzimáticamente a partir de la lactosa de leche de vaca durante la fabricación de yogur. El producto lácteo resultante será reducido en lactosa y contendrá prebióticos y bacterias potencialmente probióticas. Sin embargo, mantendrá la baja relación Ca/Pi que aporta la leche de vaca, lo que podría alterar el remodelamiento óseo y la mineralización. Objetivo: comparar si un yogur reducido en lactosa que contiene GOS (YE) ofrece ventajas adicionales respecto de un yogur regular sin GOS (YR) sobre las absorciones (Abs) de Ca y Pi, retención y calidad ósea durante el crecimiento normal. Al destete, ratas machos fueron divididas en 3 grupos alimentados con AIN ́93-G (C), YE o YR durante 28 días. Resultados: YE mostró el mayor aumento de lactobacilos fecales; producción de ácidos grasos de cadena corta especialmente p, profundidad de las criptas colónicas y menor pH cecal. El %AbsCa y %AbsPi aumentó en el siguiente órden: YE> YR> C (p < 0,05). El contenido de Ca y Pi en fémur, la densidad y contenido mineral óseos y los parámetros biomecánicos fueron similares en YE y C, mientras que YR mostró valores significativa-mente menores (p < 0,05). Conclusiones: YE aumentó las Abs y biodisponibilidad de minerales, alcanzando la retención y calidad ósea de C. El aumento en las Abs observado en YR no logró obtener la retención y calidad ósea de C. Conclusión: YE habría contrarrestado el efecto negativo del mayor aporte de Pi de la leche de vaca y sería una buena estrategia para lograr el pico de masa ósea y calidad del hueso adecuados, especialmente en individuos intolerantes a la lactosa. Breast milk contains an optimal calcium/phosphate (Ca/Pi) ratio and GOS. These natural prebiotics can be enzymatically produced via cow's milk lactose inyogurt manufacture. This milk product is low in lactose and contains prebiotics and potentially probiotic bacteria but maintains a low Ca/Pi ratio that could alter bone remodeling and mineralization. We evaluated if a lactose-reduced yogurt containing GOS (YE) offers additional advantages over regular yogurt without GOS (YR) on Ca and Pi absorption (Abs), bone retention and quality during normal growth. Weaning male rats were divided into 3 groups fed AIN'93-G (C), YE or YR for 28 days. Results: YE showed the highest increase in fecal lactobacilli; short-chain fatty acids production, especially propionate and butyrate; intestine crypt depth, and the lowest cecal pH. AbsCa% and AbsPi% increased in this order: YE> YR> C (p <0.05). Ca and Pi content in femur, bone density and mineral content, and biomechanical parameters were similar in YE and C, while YR showed the significantly lowest value (p < 0.05). Conclusions: YE increased mineral Abs reaching the retention and bone quality of C. Although YR increased Abs, bone retention and quality did not achieve C values. Seemingly, YE compensated for the negative effect of the higher Pi supply and would be a good strategy to achieve adequate peak bone mass and bone quality, especially in lactose intolerant individuals. The objective of this study was to evaluate the effect of increasing doses of bacterial phytase (RONOZYME HiPhos) on performance and carcass characteristics of growing and finishing pigs. The study included 120 castrated males with initial weight of 23.21 ± 1.91 kg and 68 days of age, distributed in a randomized block design with five treatments and eight replicates with three animals each. The pigs were fed five corn-soybean meal-based diets: positive control (PC), supplemented with inorganic phosphorus and calcium; negative control (NC), with 0.13% reduction in available phosphorus and 0.11% in calcium; and three NC diets supplemented with 1,000, 2,000, and 3,000 phytase units (FYT)/kg in the feed. Compared with the NC diets without phytase, diets with 1,000, 2,000, and 3,000 FYT/kg inclusion increased the daily weight gain by +12% (quadratic, p<0.05) during the growing I period; +2.9, +2.9, and +10.5% (linear, p<0.01), respectively, during the growing II period; and +4.1, +5.1, and +8.2% (linear, p<0.001), respectively, over the entire experimental period. The daily feed intake increased by 0, +2.8, and +4.3% (linear, p<0.05), respectively, considering the entire experimental period; and the final live weight increased by +3.2, +4.2, and +6.1% (linear, p<0.001), respectively. The phytase treatments did not influence feed conversion ratio, carcass weight and yield, backfat thickness, loin depth and carcass lean meat. According to the European Carcass Classification (SEUROP), however, the animals fed the PC diet and the three phytase levels had more carcasses classified as E (between 55-60% lean meat) when compared to carcasses of pigs fed the NC. Supplementing increasing levels of phytase to a corn- and soybean meal-based diet with inorganic P and Ca reduction improved daily weight gain and feed intake of growing pigs, and such effects were maintained until slaughter age.

Phosphorus (P) is an essential mineral for the development and maintenance of the vertebrate skeletal system. Modulation of P levels is believed to influence metabolism and the physiological responses of gene expression. In this study, we investigated the influence of dietary P on skeletal deformities and osteocalcin gene expression in zebrafish (Danio rerio), and sought to determine appropriate levels in a diet. We analyzed a total of 450 zebrafish within 31 days of hatching. Animals were distributed in a completely randomized experimental design that consisted of five replications. After an eight-week experiment, fish were diaphanized to evaluate cranial and spinal bone deformities. Increases in dietary phosphorus were inversely proportional to the occurrence of partial spine fusions, the absence of spine fusions, absence of parallelism between spines, intervertebral spacing, vertebral compression, scoliosis, lordosis, ankylosis, fin caudal insertion, and craniofacial deformities. Additionally, osteocalcin expression was inversely correlated to P levels, suggesting a physiological recovery response for bone mineralization deficiency. Our data showed that dietary P concentration was a critical factor in the occurrence of zebrafish skeletal abnormalities. We concluded that 1.55% P in the diet significantly reduces the appearance of skeletal deformities and favors adequate bone mineralization through the adjustment of osteocalcin expression.

Hyperphosphatemia is a common condition in patients with chronic kidney disease (CKD) and can lead to bone disease, vascular calcification, and increased risks of cardiovascular disease and mortality. Inorganic phosphate (Pi) is absorbed in the intestine, an important step in the maintenance of homeostasis. In CKD, it is not clear to what extent Pi absorption is modulated by dietary Pi. Thus, we investigated 5/6 nephrectomized (Nx) Wistar rats to test whether acute variations in dietary Pi concentration over 2 days would alter hormones involved in Pi metabolism, expression of sodium-phosphate cotransporters, apoptosis, and the expression of matrix extracellular phosphoglycoprotein (MEPE) in different segments of the small intestine. The animals were divided into groups receiving different levels of dietary phosphate: low (Nx/LPi), normal (Nx/NPi), and high (Nx/HPi). Serum phosphate, fractional excretion of phosphate, intact serum fibroblast growth factor 23 (FGF-23), and parathyroid hormone (PTH) were significantly higher and ionized calcium was significantly lower in the Nx/HPi group than in the Nx/LPi group. The expression levels of NaPi-IIb and PiT-1/2 were increased in the total jejunum mucosa of the Nx/LPi group compared with the Nx/HPi group. Modification of Pi concentration in the diet affected the apoptosis of enterocytes, particularly with Pi overload. MEPE expression was higher in the Nx/HPi group than in the Nx/NPi. These data reveal the importance of early control of Pi in uremia to prevent an increase in serum PTH and FGF-23. Uremia may be a determining factor that explains the expressional modulation of the cotransporters in the small intestine segments.

High phosphate intake is known to aggravate renal osteodystrophy along various pathogenetic pathways. Recent studies have raised the possibility that dysregulation of the osteocyte Wnt/ß-catenin signaling pathway is also involved in chronic kidney disease (CKD)-related bone disease. We investigated the role of dietary phosphate and its possible interaction with this pathway in an experimental model of adynamic bone disease (ABD) in association with CKD and hypoparathyroidism. Partial nephrectomy (Nx) and total parathyroidectomy (PTx) were performed in male Wistar rats. Control rats with normal kidney and parathyroid function underwent sham operations. Rats were divided into three groups and underwent pair-feeding for 8 weeks with diets containing either 0.6% or 1.2% phosphate: sham 0.6%, Nx+PTx 0.6%, and Nx+PTx 1.2%. In the two Nx+PTx groups, serum creatinine increased and blood ionized calcium decreased compared with sham control group. They also presented hyperphosphatemia and reduced serum parathyroid hormone (PTH) and fibroblast growth factor 23 (FGF23) levels. Fractional urinary excretion of phosphate increased in Nx+PTx 1.2% rats despite lower PTH and FGF23 levels than in sham group. These biochemical changes were accompanied by a decrease in bone formation rates. The Nx+PTx 1.2% group had lower bone volume (BV/TV), higher osteoblast and osteocyte apoptosis, and higher SOST and Dickkopf-1 gene expression than the Nx+PTx 0.6% group. Nx+PTx 0.6% rat had very low serum sclerostin levels, and Nx+PTx 1.2% had intermediate sclerostin levels compared with sham group. Finally, there was a negative correlation between BV/TV and serum sclerostin. These results suggest that high dietary phosphate intake decreases bone volume in an experimental model of CKD-ABD, possibly via changes in SOST expression through a PTH-independent mechanism. These findings could have relevance for the clinical setting of CKD-ABD in patients who low turnover bone disease might be attenuated by optimal control of phosphate intake and/or absorption.

RESUMO

Requirements for optimal nutrition, especially for micronutrients, are not well defined for premature infants. The "reference fetus," developed by Ziegler et al,(1) has served as a model to define nutritional needs and studies designed to determine nutrient requirements. Revision of nutrient requirements and provision of optimal nutrition may lead to improved outcomes in preterm infants. Appropriate provision of nutrients also may help prevent nutritional disorders, such as metabolic bone disease and anemia. In this review, we discuss calcium, phosphorus, magnesium, vitamin D, iron, and copper, and define optimal intakes based on the available published data.

Two experiments were conducted to determine the requirement of available P (aP) of pigs selected for high lean deposition during summer (maximum and minimum temperatures of 29.8 ± 3.6 and 21.3 ± 1.1°C, respectively; Exp. 1) and winter (maximum and minimum temperatures of 24.7 ± 2.1 and 9.1 ± 1.6°C, respectively; Exp. 2). Pigs (66.5 ± 2.8 kg, Exp. 1; and 61.1 ± 0.6 kg, Exp. 2) were randomly allotted to 5 dietary treatments. Exp. 1 had 5 pens/treatment and Exp. 2 had 6 pens/treatment, and all pens had 2 pigs. Treatments were composed of a corn-soybean meal basal diet and 4 diets with additional dicalcium phosphate to obtain 5 aP concentrations (0.122, 0.187, 0.252, 0.317, and 0.382% in Exp. 1, and 0.135, 0.200, 0.265, 0.330, and 0.395% in Exp. 2). Pigs were allowed ad libitum access to their respective diets for 30 d. In Exp. 1, increasing aP improved ADFI and ADG (quadratic, P < 0.05), with maximum responses obtained at 0.226 and 0.256% of aP, respectively. Increasing aP improved (P < 0.05) G:F. A broken-line analysis indicated that G:F was maximized (P < 0.05) at 0.295% of aP or 9.04 g of aP/d. Increasing aP also increased (quadratic, P < 0.05) P and Ca (g/kg) in the metacarpals with maximum responses obtained at 0.314 and 0.272% of aP, respectively. The percentage of ash in the metacarpals continued to increase (linear, P < 0.05) with increasing aP. In Exp. 2, ADG and G:F improved (linear and quadratic, P < 0.05) as aP increased in the diet. A broken-line analysis indicated that G:F was maximized (P < 0.05) at 0.316% of aP or 10.58 g of aP per day. However, increasing aP had no effect on ADFI. Percentages of ash, P, and Ca in the metacarpals increased (linear, P < 0.05) with increasing aP. In conclusion, results of this experiment indicated that finishing pigs (60 to 100 kg) kept in a thermoneutral environment require 0.316% aP in the diet (10.58 g/d) to maximize G:F, but the requirement is only 0.295% aP (9.04 g/d) when pigs are under heat stress.

BACKGROUND: Cardiac remodeling in uremia is characterized by left ventricular hypertrophy, interstitial fibrosis and microvascular disease. Cardiovascular disease is the leading cause of death in uremic patients, but coronary events alone are not the prevalent cause, sudden death and heart failure are. We studied the cardiac remodeling in experimental uremia, evaluating the isolated effect of parathyroid hormone (PTH) and phosphorus. METHODS: Wistar rats were submitted to parathyroidectomy (PTx) and 5/6 nephrectomy (Nx); they also received vehicle (V) and PTH at normal (nPTH) or high (hPTH) doses. They were fed with a poor-phosphorus (pP) or rich-phosphorus (rP) diet and were divided into the following groups: 'Sham': G1 (V+normal-phosphorus diet (np)) and 'Nx+PTx': G2 (nPTH+pP), G3 (nPTH+rP), G4 (hPTH+pP) and G5 (hPTH+rP). After 8 weeks, biochemical analysis, myocardium morphometry and arteriolar morphological analysis were performed. In addition, using immunohistochemical analysis, we evaluated angiotensin II, α-actin, transforming growth factor-beta (TGF-ß) and nitrotyrosine, as well as fibroblast growth factor-23 (FGF-23), fibroblast growth factor receptor-1 (FGFR-1) and runt-related transcription factor-2 (Runx-2) expression. RESULTS: Nx animals presented higher serum creatinine levels as well as arterial hypertension. Higher PTH levels were associated with myocardial hypertrophy and fibrosis as well as a higher coronary lesion score. High PTH animals also presented a higher myocardial expression of TGF-ß, angiotensin II, FGF-23 and nitrotyrosine and a lower expression of α-actin. Phosphorus overload was associated with higher serum FGF-23 levels and Runx-2, as well as myocardial hypertrophy. FGFR-1 was positive in the cardiomyocytes of all groups as well as in calcified coronaries of G4 and G5 whereas Runx-2 was positive in G3, G4 and G5. CONCLUSION: In uremia, PTH and phosphorus overload are both independently associated with major changes related to the cardiac remodeling process, emphasizing the need for a better control of these factors in chronic kidney disease.

RESUMO

The present study evaluated the nutritious potential of 18 fruits, all native of the Brazilian Cerrado biome. Mineral contents were determined by inductively coupled plasma atomic emission; phytic acid and tannin contents by a colorimetric and titrimetric method, respectively; and the potential mineral bioavailability by the molar ratio of phytic acid/mineral. Baru nut showed the highest zinc, copper, iron, phosphorus and magnesium content, and, together with macaúba, showed also the highest calcium content and caloric value. Macaúba, pitomba, ingá and murici fruits were classified as a source of iron. The jatoba and baru nut had the highest concentration of phytic acid and tannins. The [phytic acid]/[iron] and [phytic acid]/[zinc] molar ratios were higher than the critical values (14 and 10, respectively) only in the baru nuts, which suggests that iron and zinc bioavailability is low in this nut. The [phytic acid]/[calcium] molar ratios were lower than the critical value (1.56) in all analyzed fruits, which suggests that calcium is bioavailable in the fruits.

OBJECTIVE: To evaluate the effects of usual childhood dairy intake on adolescent bone health. STUDY DESIGN: Dietary data collected in the Framingham Children's Study over 12 years were used to evaluate usual dairy consumption and adolescent bone health. Each child's average Food Pyramid servings were estimated from yearly sets of 3-day diet records. Bone mineral content (BMC) and area (BA) for total body and 6 regions (arms, legs, trunk, ribs, pelvis, and spine) at ages 15 to 17 years were the primary outcomes. Analysis of covariance was used to adjust for potential confounding by sex and physical activity, as well as age, height, body mass index and percent body fat at the time of the bone scan. RESULTS: Consuming >or= 2 servings/day of dairy (versus less) was associated with significantly higher mean BMC and BA. Higher intakes of meats/other proteins (>or= 4 servings per/day) were also associated with higher mean BMC and BA values. Children with higher intakes of both dairy and meats/other proteins had the highest adjusted BMC (3090.1 g), and children consuming less of each had the lowest BMC (2740.2 g). CONCLUSIONS: These prospective data provide evidence for a beneficial effect of childhood dairy consumption on adolescent bone health.

Objetivo. Comparar el balance metabólico de calcio y fósforo de lactancia versus el de destete, así como determinar la producción de calcio y fósforo en la leche de mujeres de una comunidad rural de México a diferentes tiempos de lactancia. Métodos. Se estudiaron 36 mujeres con edades entre 18 y 36 años, peso de 49 ñ 3 kg y talla de 148 ñ 2 cm, divididas en seis grupos: cuatro grupos de lactancia (1º, 3º, 12º mes), uno de destete y uno no lactantes. El balance metabólico fue diez días, con duplicados de la dieta, orina de 24 horas (3 días, heces de 72 horas y muestras de leche de 24 horas (2 días). Se determinó calcio (Ca) por espectrofotometría de absorción atómica y fosforo por método colorimétrico con molibdato-metavanadato. Resultados y conclusiones. Al tercer mes de lactancia se observó el mayor contenido de Ca en leche (246 mg/día). En la lactancia se observaron balances negativos de Ca(789 ñ 165 mg/día); en los primeros meses fue menor de excreción urinaria, lo que sugiere un mecanismo de conservación de Ca. no se observaron diferencias en el contenido de fósforo en leche. Los grupos de mujeres no lactantes y del destete presentaron balances positivos de fósforo, mientras que en la lactancia se observaron balances negativos (115 a 475 mg/día). La excreción fecal de Ca y fósforo fue salta (~1300 mg/día) lo que contribuyó a la condición negativa del babance. La producción de Ca y fósforo en leche fue similar a lo referido para poblaciones rurales y urbanas de Africa, Asia, Europea y Estados Unidos

One thousand male Hubbard chicks were used in a 21-d study (10 birds per battery cage) to determine relative biological availability of phosphorus in seven samples of commercial dicalcium phosphate, expected to contain variable amounts of monocalcium phosphate. Five samples were from established producers in Brazil and two from the U.S. Pure calcium phosphate dibasic dihydrate was used as the reference standard. Phosphates were added to the corn-soybean basal diet (22.5% CP; 0.4% total phosphorus) to provide 0.1, 0.2, and 0.3% supplemental phosphorus. The calcium level was 1.0% for all diets. Left tibias were removed for bone ash (BA) and bone strength (BS) determination. Body weight, feed intake (FI), BA, BS, and plasma phosphorus increased (P < 0.01) and plasma calcium and alkaline phosphatase decreased (P < 0.01) with increasing dietary phosphorus regardless of source. The availability of phosphorus for each test phosphate was determined by slope ratio, with BW, BA, and BS regressed on phosphorus added within each phosphorus source. A relative biological value (RBV) was calculated based on BW, BA, and gain:feed ratio. Availability based on BW ranged from 97.07 to 110.41%. When BA was the criterion, values were 80.32 to 107.84% and for BS were 79.34 to 110.52%. The RBV ranged from 97.55 to 100.60%. Phosphate sources did not vary greatly in phosphorus availability. Overall phosphorus availability averages were higher for BW (103%) and RBV (99%) and lowest for BA (96%) and BS (94%).
