Ultraviolet radiation (UVR) is widely known as deleterious for many organisms since it can cause damage to biomolecules either directly or indirectly via the formation of reactive oxygen species. The goal of this study was to analyze the capacity of high-mountain Espeletia hartwegiana plant phyllosphere microorganisms to survive UVR and to identify genes related to resistance strategies. A strain of Deinococcus swuensis showed a high survival rate of up to 60% after UVR treatment at 800J/m2 and was used for differential expression analysis using RNA-seq after exposing cells to 400J/m2 of UVR (with >95% survival rate). Differentially expressed genes were identified using the R-Bioconductor package NOISeq and compared with other reported resistance strategies reported for this genus. Genes identified as being overexpressed included transcriptional regulators and genes involved in protection against damage by UVR. Non-coding (nc)RNAs were also differentially expressed, some of which have not been previously implicated. This study characterized the immediate radiation response of D. swuensis and indicates the involvement of ncRNAs in the adaptation to extreme environmental conditions.

Bacteria under stress increase the proportion of dormant cells to ensure their survival. Cold and osmotic stress are similar, because in both the availability of water is reduced. Glycine betaine (GB) is one of the most common osmoprotectants in bacteria and possesses cryoprotectant properties. Our aim was to determine whether GB modifies the proportion of dormant Deinococcus sp. UDEC-P1 and Psychrobacter sp. UDEC-A5 cells exposed to osmotic stress. Both bacterial strains were incubated in the presence of up to 1 M NaCl with or without GB. Active and dormant cells were evaluated by both spectrophotometric and flow cytometry analysis. Without GB, Deinococcus sp. UDEC-P1 grew in the presence of 0.05 M NaCl, but with 5 mM GB grew at 0.1 M NaCl. Psychrobacter sp. UDEC-A5 grew in the presence of up to 0.25 M NaCl, but with 5 mM GB grew at 0.5 M NaCl. Under osmotic stress, the proportion of dormant cells of Deinococcus sp. UDEC-P1 and Psychrobacter sp. UDEC-A5 increased significantly (about eightfold and fivefold, respectively). The addition of GB (5 mM) exerted a different effect on the two strains, since it avoided the entrance into the dormancy of Psychrobacter sp. UDEC-A5 cells, but not of Deinococcus sp. UDEC-P1 cells. Our results suggest that the effect of GB on bacterial metabolism is strain dependent. For bacteria in which GB avoids dormancy, such as Psychrobacter sp. UDEC-A5, it could be a "double-edged sword" by reducing the "seed bank" available to recover the active population when favorable conditions return.

Kepler-96 is an active solar-type star harboring a Super-Earth planet in close orbit. Its age of 2.3 gigayears is the same as the Sun when there was a considerable increase of oxygen in Earth's atmosphere due to micro-organisms living in the ocean. We present the analysis of superflares seen on the transit light curves of Kepler-96b. The model used here simulates the planetary transit in a flaring star. By fitting the observational data with this model, it is possible to infer the physical properties of the flares, such as their duration and the energy released. We found three flares within the energy range of superflares, where the biggest superflare observed was found to have an energy of 1.81 × 1029 J (1.81 × 1035 ergs). The goal is to analyze the biological impact of these superflares on a hypothetical Earth in the habitable zone of Kepler-96, assuming this planet has protection through different scenarios: an Archean and present-day atmospheres. Also, we compute the attenuation of the flare ultraviolet (UV) radiation through an Archean ocean. The conclusion is that considering the increase in the UV flux by the strongest superflare emission, Escherichia coli and Deinococcus radiodurans could survive on the surface of the planet only if there was an ozone layer present on the planet atmosphere. However, they could escape from the hazardous UV effects at a depth of 28 and 12 m below the ocean surface, respectively. For smaller superflares contribution, D. radiodurans could survive in the surface even in an Archean atmosphere with no ozone.

Genus Deinococcus is characterized by an increased resistance toward reactive oxygen species (ROS). The chromosome of five strains belonging to this genus has been sequenced and the presence of a luxS-like gene was deduced from their genome sequences. The aim of this study was to assess if a complete QS circuit is present in Deinococcus sp. and if this QS is associated with ROS. Primers for searching luxS-like gene and the putative receptor gene, namely ai2R, were designed. AI-2 signal production was evaluated by luminescence analysis using Vibrio harveyi BB170 as reporter strain. AI-2 signal was also evaluated by competitive assays using cinnamaldehyde, ascorbic acid, and 3-mercaptopropionic acid as interfering molecules. Potassium tellurite and metronidazole were used as oxidative stressors. A luxS-like gene as well as an ai2R gene was detected in strain UDEC-P1 by PCR. Cell-free supernatant of strain UDEC-P1 culture induced luminescence in V. harveyi BB170, and this property was inhibited with the three interfering molecules. The oxidative stressors metronidazole and potassium tellurite decreased Deinococcus sp. viability, but increased luminescence of the reporter strain. The results demonstrate that both a functional luxS-like gene and a putative receptor for AI-2 signal are present in Deinococcus sp. strain UDEC-P1. This finding also suggests that a complete QS circuit is present in this genus, which could be related to oxidative stress.

Three ionizing-radiation-resistant bacterial strains (designated KR-196, KR-198 and KR-200(T)) were isolated from a sample of arid soil collected from a coastal desert in Chile. The soil sample was irradiated before serial dilution plating was performed using one-tenth-strength plate count agar. Phylogenetic analysis of the 16S rRNA gene sequences showed these organisms to represent a novel species of the genus Deinococcus, having sequence similarities of 87.3-90.8 % with respect to recognized Deinococcus species. Strains KR-196, KR-198 and KR-200(T) were aerobic and showed optimum growth at 30 degrees C and pH 6.5-8.0. The major respiratory menaquinone was MK-8. The predominant fatty acids in these strains were 16 : 1 omega 7c, 16 : 0, 15 : 1 omega 6c, 17 : 0 and 18 : 0. The DNA G+C content of strain KR-200(T) was 63.9 mol%. Strains KR-196, KR-198 and KR-200(T) were found to be resistant to >10 kGy gamma radiation. On the basis of the phylogenetic, chemotaxonomic and phenotypic data, strain KR-200(T) represents a novel species of the genus Deinococcus, for which the name Deinococcus peraridilitoris sp. nov. is proposed. The type strain is KR-200(T) (=LMG 22246(T)=CIP 109416(T)).
