RESUMO

The objective of this study was to evaluate the effects of combining immunocastration and ß-adrenergic agonists (ß-AA) on blood metabolites and their correlations with performance and carcass traits of Nellore cattle. Ninety-six Nellore males were distributed in a randomized block design with a 2×3 factorial arrangement. The factors were two sexual conditions (immunocastrated and non-castrated), and three diets (control, with no inclusion of ß-AA; RH, with 300 mg of ractopamine hydrochloride/d; and ZH, with 80 mg of zilpaterol hydrochloride/animal/d). The trial was conducted during 100 d, during which animals were fed ZH and RH for the last 30 and 33 d, respectively. Blood metabolites related to lipid and protein metabolism were assessed at the baseline (0 d of ß-AA supplementation) and after 13 and 30 d of ß-AA supplementation. No effect of sexual condition × ß-AA supplementation × time on feed interaction was observed. Combining immunocastration with ß-AA supplementation modified cholesterol and non-esterified fatty acids concentrations without affecting protein metabolism. Immunocastration enhances lipogenesis and reduces skeletal muscle accretion by increasing high-density lipoprotein and triglycerides concentrations and decreasing creatinine and creatine kinase concentrations, respectively. Zilpaterol hydrochloride enhances skeletal muscle accretion by decreasing urea and total protein concentrations and increases creatinine and creatine kinase concentrations without modifying lipid metabolism.(AU)

Ractopamine hydrochloride is a commercial beta-adrenergic agonist commonly used as a dietary supplement in cattle production for improved feed efficiency and growth promotion. Currently, regulatory target tissues (as approved in the New Animal Drug Application with Food and Drug Administration) for ractopamine residue testing are muscle and liver. However, other tissues have recently been subjected to testing in some export markets for U.S. beef, a clear disregard for scientific maximum residue limits associated with specific tissues. The overall goal of this study was to develop and validate an LC-MS/MS assay to determine whether detectable and quantifiable levels of ractopamine in digestive tract-derived edible offal items (i.e., abomasum, omasum, small intestine, and reticulum) of cattle resulted from tissue residues or residual ingesta contamination of exposed surfaces of tissues (rinsates). Tissue samples and corresponding rinsates from 10 animals were analyzed for parent and total ractopamine (tissue samples only). The lower limit of quantitation was between 0.03 and 0.66 ppb depending on the tissue type, and all tissue and rinsate samples tested had quantifiable concentrations of ractopamine. The highest concentrations of tissue-specific ractopamine metabolism (represented by higher total vs. parent ractopamine levels) were observed in liver and small intestine. Contamination from residual ingesta (represented by detectable ractopamine in rinsate samples) was only detected in small intestine, with a measured mean concentration of 19.72 ppb (±12.24 ppb). Taken together, these results underscore the importance of the production process and suggest that improvements may be needed to reduce the likelihood of contamination from residual ractopamine in digestive tract-derived edible offal tissues for market.

The objective of this study was to evaluate the effects of compensatory gain associated with the use of 10ppm ractopamine after a period of feed restriction in finishing pigs on performance, carcass and meat quality. Twenty castrated males and 20 females, at 110 days of age and 66.137±6.13kg live weight, were submitted to four treatments using a 2 x 2 factorial design (fed ad libitum or with 20% restriction between 0(21 days of age and fed with or without 10ppm ractopamine for 22(42 days of experimentation), with 10 replicates (animals). There was no interaction between the factors for any of the evaluated parameters. Animals treated with ractopamine presented better weight gain (1.083 versus 1.259kg), feed conversion (2.910 versus 2.577), warm and cold carcass weight (86.08 versus 89.00 and 83.46 versus 87.20kg, respectively), loin depth (63.02 versus 68.40mm), loin eye area (41.43 versus 46.59mm2) and muscle fiber diameter (27.48 versus 35.85µm). Animals submitted to feed restriction followed by ad libitum feed presented compensatory gain without losses to carcass and meat characteristics, but with a reduction in the ethereal extract (2.19 versus 1.64%) and lower water loss due to thawing in the meat (11.35 versus 9.42%). The effects of compensatory gain after food restriction and ractopamine are independent of the parameters evaluated.(AU)


Objetivou-se avaliar os efeitos do ganho compensatório associado ao uso de 10ppm de ractopamina após um período de restrição alimentar, em suínos em terminação, sobre características de desempenho, carcaça e qualidade de carne. Foram utilizados 20 machos castrados e 20 fêmeas, com 110 dias de idade e 66,137±6,13kg de peso vivo, submetidos a quatro tratamentos, fatorial 2 x 2 (alimentação à vontade ou com 20% de restrição entre zero e 21 dias de experimentação; e alimentação à vontade, sem ou com 10ppm de ractopamina, durante 22 a 42 dias de experimentação), com 10 repetições, sendo o animal a repetição. Não houve interação entre os fatores para nenhum dos parâmetros avaliados. Animais tratados com ractopamina apresentaram melhor ganho de peso (1,083 versus 1,259kg), conversão alimentar (2,910 versus 2,577), peso da carcaça quente e fria (86,08 versus 89,00 e 83,46 versus 87,20kg, respectivamente), profundidade do lombo (63,02 versus 68,40mm), área de olho de lombo (41,43 versus 46,59mm2) e diâmetro de fibras musculares (27,48 versus 35,85µm). Animais submetidos à restrição alimentar seguida de arraçoamento ad libitum apresentaram ganho compensatório sem prejuízos às características de carcaça e à carne, mas com redução do extrato etéreo (2,19 versus 1,64%) e menor perda de água por descongelamento na carne (11,35 versus 9,42%) Os efeitos do ganho compensatório após a restrição alimentar e da ractopamina mostram-se independentes sobre os parâmetros avaliados.(AU)

O exercício aeróbio promove efeitos benéficos na prevenção e tratamento de doenças como hipertensão arterial, aterosclerose, insuficiência venosa e doença arterial periférica. Os receptores β-adrenérgicos estão presentes em várias células. No sistema cardiovascular, promovem inotropismo e cronotropismo positivo cardíaco e relaxamento vascular. Embora os efeitos do exercício tenham sido investigados em receptores cardíacos, estudos focados nos vasos são escassos e controversos. Esta revisão abordará os efeitos do exercício físico sobre os receptores β-adrenérgicos vasculares em modelos animais e humanos e os mecanismos celulares envolvidos na resposta relaxante. Em geral, os estudos mostram resultantes conflitantes, onde observam diminuição, aumento ou nenhum efeito do exercício físico sobre a resposta relaxante. Assim, os efeitos do exercício na sensibilidade β-adrenérgica vascular merecem maior atenção, e os resultados mostram que a área de fisiopatologia vascular é um campo aberto para a descoberta de novos compostos e avanços na prática clínica.


Aerobic exercise promotes beneficial effects on the prevention and treatment of diseases such as arterial hypertension, atherosclerosis, venous insufficiency, and peripheral arterial disease. β-adrenergic receptors are present in a variety of cells. In the cardiovascular system, β-adrenergic receptors promote positive inotropic and chronotropic response and vasorelaxation. Although the effect of exercise training has been largely studied in the cardiac tissue, studies focused on the vascular tissue are rare and controversial. This review examines the data from studies using animal and human models to determine the effect of physical exercise on the relaxing response mediated by β-adrenergic receptors as well as the cellular mechanisms involved in this response. Studies have shown reduction, increase, or no effect of physical exercise on the relaxing response mediated by β-adrenergic receptors. Thus, the effects of exercise on the vascular β-adrenergic sensitivity should be more deeply investigated. Furthermore, the physiopathology of the vascular system is an open field for the discovery of new compounds and advances in the clinical practice.

A series of cationic drug-like substances with distinct basicity, hydrogen-bonding ability, and hydrophobicity, including three catecholamines, two beta-agonists, and thirteen beta-blockers, was successfully detected in a capillary electrophoresis system using an end-capillary coupled potentiometric sensor consisting of a PVC-based liquid membrane deposited directly on a 100 mum diameter copper rod. The electrophoretic separation was performed on a 72 cm x 75 microm id uncoated fused-silica capillary with an acidic background electrolyte containing phosphoric acid in a water-acetonitrile mixture, pH* 2.8. Samples were injected electrokinetically at 5.0 kV for 10 s and a running voltage of 19.5 kV was applied. Excluding the bufuralol/practolol pair, baseline separation of all substances was achieved in the developed CE system within 9 minutes. A linear relationship (R(2) 0.8752) between the sensitivity of the applied potentiometric detector and the parameter log P characterising the hydrophobicity of the analytes was demonstrated. The best observable limits of detection (LODs) were obtained for the highly hydrophobic substances, i. e. bufuralol (8.10 x 10(-8) M injected concentration, S/N = 3), propranolol, alprenolol, and clenbuterol (ca. 1.10 x 10(-7) M). In the case of hydrophilic catecholamines and carbuterol their LODs with potentiometric detection were lowered by a factor of almost one thousand, reaching a value of 6.6 x 10(-5) M.

Validation studies were carried out on a multi-residue screening method for anilinic type beta-agonists (clenbuterol, mabuterol, brombuterol, cimaterol, cimbuterol, mapenterol, clenpenterol) and a method for the phenolic type beta-agonist, salbutamol, in bovine liver. The validation was performed according to the European Union Commission Decision 2002/657/EC (European Commission 2002), which establishes criteria and procedures for the determination of parameters such as the detection capability (CCbeta), specificity, stability of standard solutions and stability of the analyte in matrix. CCbeta values for the eight target compounds were between 0.25 and 0.5 microg kg(-1). The stability of standard solutions and analytes in matrix and the specificity of the antibody were characterized. The methods are applicable for qualitative screening of beta-agonists for regulatory programmes according to European Union performance requirements, or as a semi-quantitative research tool for known target compounds.

This paper describes the separation and quantification of salbutamol in pharmaceutical products (salbutamol syrups) by capillary electrophoresis (CE) with contactless conductivity detection (C(4)D). The system was studied by micellar electrokinetic capillary chromatography (MEKC) and free solution capillary electrophoresis (FSCE), being the latter chosen in function of best resolution and sensitivity in comparison with the MEKC method. CE-C(4)D was applied to analysis of salbutamol in syrups utilizing 1.0 x 10(-2) molL(-1) acetic acid/sodium acetate buffer (pH 4.9) as running electrolyte. Tetraethylammonium (TEA) solution was used as internal standard. The results obtained include a linear dynamic range from 7. x 10(-5) to 3.0 x 10(-4) molL(-1) and good repeatability (R.S.D.=4.7% for n=10 for a 7.0 x 10(-5) molL(-1) salbutamol solution). The detection limit was calculated as 1.0 x 10(-5) molL(-1) and the limit of quantification was estimated as 3.3 x 10(-5) molL(-1). For syrups analysis the reproducibility presented deviations between 1.5% and 2.5% (three different days) obtained for measurements in triplicate.

In this work a simple and sensitive fluorimetric method for determination of salbutamol (4-[2-(tert-butylamino)-1-hydroxyethyl]-2-(hydroxymethyl) phenol) using an Eu enhanced signal was developed. The employed methodology is based on the formation of a ternary complex formed with Eu, salbutamol and trioctylphosphine oxide (TOPO). Intermolecular transfer of energy from the excited organic molecule to the lanthanide followed by lanthanide emission is responsible for excitation of the lanthanide ion in complex solutions and fluorescent enhancement. The luminescence properties of the ternary complex formed with TOPO and optimum formation conditions were investigated. The calibration curve is linear in the range between 6.92-180 microg l(-1) of salbutamol. The detection limit was 2.31 microg l(-1). Common excipients for these formulations were not found to interfere. A proposed method for the assay in commercial aerosols and nebulizer solutions containing salbutamol was applied with very good precision.
