RESUMO

The increasing water contamination by toxic heavy metals, particularly hexavalent chromium, has become a significant environmental concern. This study explores the pyrolysis of termite-processed biomass, specifically Pinus elliottii particleboard and its termite droppings (TDs), to produce biochar and its application for chromium (VI) adsorption. Termite droppings, rich in lignin, and particleboard, rich in cellulose, were pyrolyzed at various temperatures to assess the effect of biomass composition on biochar properties. The study found that lignin-rich termite droppings produced biochar with higher fixed carbon content and specific surface area than cellulose-rich particleboard biochar. FTIR and Raman spectroscopy revealed significant molecular structure changes during pyrolysis, which influenced the adsorption capabilities of the biochar. Adsorption experiments demonstrated that TD biochar exhibited significantly higher chromium (VI) adsorption capacity, attributed to its distinct chemical composition and enhanced surface properties due to higher lignin content. These findings underscore the crucial role of lignin in producing efficient biochar for heavy metal adsorption, highlighting the practical applicability of termite-processed biomass in water purification technologies.

RESUMO

Termite nest repairs are considered a defensive conduct as they reduce the colony's exposure to the external environment. Repair activities are carried out by worker castes that can be polymorphic, representing a relationship between polymorphism and divisions of functions that can enhance task completion. Repairs are influenced by the extent of damage, nest volume, and the population dynamics of the building species, which regulate the recruitment of individuals for this activity. Our objective was to verify the performances (recruitment for repair) of dimorphic workers of Constrictotermes cyphergaster (Silvestri, 1901) during the damage repair activities performed on the external walls of termite nests of different sizes. We found a significant difference in the presence of dimorphic workers that performed repairs, with greater recruitment of the small morphotype, and observed an alternation of morphotypes between initial and final repair activities, with no influence of morphotype on the replacement pattern. Our results also showed that the total number of recruited workers decreased with increasing nest volume. These results help to better understand the social organization of a Nasutitermitinae termite species and the strategies adopted to protect its colonies.

RESUMO

Reticulitermes flavipes is the most invasive species in its genus and is responsible for causing significant damage to human structures in areas where it has been introduced. Although it has already become established in Chile and Uruguay, it had not previously been reported in Argentina. In this study, we report the first detection of this species in Buenos Aires, the capital of Argentina. The colony was already producing alates, and species identification was confirmed through both morphology and mitochondrial gene 16S rRNA analysis. Our results, while not conclusive, suggest that this introduction was independent from the one that occurred in Chile and Uruguay, and potentially originated from the United States. The detection of R. flavipes in Argentina is significant because it highlights the potential for this species to establish itself in new regions and underscores the need for future research on and control of R. flavipes in this country.

RESUMO

Behavioral lab bioassays involving termites must be promptly performed to allow intended observations prior to death from dissecation, typical of these soft-bodied insects. To this end, topic markers have been proposed as an alternative to histological stains which, while not always toxic are inevitably lengthy to apply. Among recommended topic markers, gouache is easy to apply, dries out quickly, but it is known affect termites in the long run, being suitable only to short-term bioassays. Its alternative, colored glue, is also easy to apply, but it takes long to dry and it is too dense and heavy, being thus prone to affect termite walking patterns. Here we tested a mix of gouache and colored glue aiming to combine the qualities of both into a suitable topical marker for Cornitermes cumulans termites. Similar patterns of survival presented by marked and unmarked termites ruled out concerns about toxicity of this mixture. Such results were consistent across distinct group densities evidencing that the mixture does not interfere with, nor it is affected by, crowding effects. Because crowding regulates interindividual interactions and these underlie most behaviors, the mixture can be thought to be suitable to behavioral studies. We argue that this 1:2 glue:gouache mixture is an excellent alternative to mark termites for lab bioassays. Being atoxic, cheap, easy to apply, and non-invasive, this mixture may happen to be useful not only for termites but also in bioassaying other similarly soft-bodied insects.

Abstract Tropical grasslands have been systematically neglected worldwide in maps, conservation policies, and ecological studies. After eradicating invasive pine trees from a Cerrado reserve in southeastern Brazil, an unprecedented grassy ecosystem arose in recent satellite images. In the field, we confirmed the first record of wet grasslands with termite mounds - locally named campo com murundus ˗ beyond 21°S in the São Paulo state. Besides reporting this occurrence, we sampled the two plant communities forming this peculiar vegetation type (the mounds and the waterlogged matrix around them) to investigate if they are floristically and functionally distinct. We also explored how these two communities relate to those of the surrounding open vegetation types (savanna, dry and wet grassland). Woody plants were recorded on the mounds but not in the matrix, although the two communities share some ground layer species. Compared to the adjacent vegetation types, the mounds were floristically distinct and functionally more balanced in growth forms, dispersal syndrome, and tolerance to waterlogging. We hope this borderline record of campo com murundus can stimulate the search for other unnoticed remnants out of their known occurrence region, triggering efforts for their conservation and studies to improve comprehension of these iconic ecosystems.

Insects and fungi are abundant in many environments, in which facultative and/or obligate associations involving these groups have been established during evolution. In termites, mutualism with fungi is well reported for some termite lineages (e.g., Macrotermitinae). Within some subterranean termite species (Rhinotermitidae), egg-mimicking fungi, also referred to as "termite "balls", are often harbored inside the nest, mixed to the egg piles. Such interaction seems to be advantageous for both partners since the fungi are protected inside the nest while they may serve as an additional food source and also provide cellulases which may be incorporated into the termite digestive process. Although such mutualism has been reported for seven species of Reticulitermes and Coptotermes formosanus, all the samplings were restricted to temperate regions. Here, we provide the first Neotropical record of this termite-fungus association, and the first report for Coptotermes gestroi. The morphological characters of the "termite balls" observed in a C. gestroi nest resemble those already reported for Reticulitermes spp. and the congeneric species C. formosanus. They include a color ranging from light to dark brown, spherical shape, and a reduced diameter (0.23-0.34 mm). Our findings provide new insights into the geographical distribution of the association between termites and sclerotium-forming fungi. Future genetic analyses will be valuable aiming to identify the egg-mimicking fungi associated with C. gestroi and shed light on the evolution of this fascinating symbiosis.

RESUMO

Abstract We report the opportunistic predation of Roadside Hawk (Rupornis magnirostris - Accipitridae) on subterranean termite (Coptotermes testaceus - Rhinotermitidae). This predatory behaviour was observed during an avifauna survey in the municipality of Candeias do Jamari, Rondônia, Brazil, in the southwestern Brazilian Amazon. There was a great abundance of C. testaceus alates because of heavy rainfall. This is the season when C. testaceus colonies produce alates in large numbers to found new colonies. The Roadside Hawk stayed on its perch and captured winged termites with little energy expenditure. The bird in question appeared to take advantage of the termite mating flight to feed opportunistically on the abundant alates. Although feeding on insects by R. magnirostris is well known, this is the first documentation of feeding on termite alates.

Most animals move intermittently, pausing or slowing down for short moments and short moves, and darting away towards a new location where to hover again. This pattern occurs at a range of spatial and temporal scales (thence, resembling 'scale-free'), from the quick inspection of local areas to the sum of all movements performed from birth to death. While this pattern has been extensively described, its proximate drivers remain open to debate. A current hypothesis states that underlying proximate generative mechanisms of intermittent movement of animals are linked to external stimuli, e.g., interactions with the abiotic environment, resources, and other individuals. Here we investigated a prediction from this hypothesis, using termites as a biological model. We verified whether the social background in which a termite is inserted will modify the parameters of its intermittent scale-free spatial displacement. This relates to the hypothesis because it inspects how do external stimuli coming from intraspecifics can affect this type of movement. We tracked the trajectories of a focal termite confined along its nestmates in experimental clueless arenas, every 0.5 s along about six hours. Arenas varied in group size and comprised 22 distinct caste compositions, yielding 75 trackings (> 400 h) and more than two million Cartesian coordinates. Most of these trajectories (96% or 72/75) were classified as intermittent scale free (Lévy-like), indicating prevalence of this type of movement over non-Lévy-like ones (P=1.62×10-15). Moreover, intermittent scale-free movements performed by the focal termite (i) did arise outside the foraging-searching framework, and (ii) were modified by the social context while remaining within the limits of Lévy-walk realms. That is, some limits seem to exist along with the necessary plasticity to provide room for natural selection. Moreover, by arising outside the foraging framework, Lévy-like movements are shown to have broader relevance. Studies on processes that depend on social context and movement (e.g., collective behaviour, the spread of diseases) may, hence, profit from such concepts.

RESUMO

Asymmetrical snapping mandibles have evolved several times in termites. In the Neotropics, asymmetrical snapping mandibles are found in soldiers of four genera: Neocapritermes, Planicapritermes, Cornicapritermes and Dihoplotermes. Here, I describe Schievitermesglobicornis, new genus and species, from French Guiana. This genus is characterized by an absence of a frontal prominence and slightly asymmetrical mandibles in the soldier caste. The morphology and anatomy of the worker reveal a wood-based diet, and suggest that Schievitermes, Planicapritermes and Neocapritermes constitute a monophyletic group, which is consistent with mtDNA data.

RESUMO

The aim of the present study was to assess the biochemical and molecular structural characteristics of a novel alkali-thermostable GH10 xylanase (Xyl10B) identified in a termite gut microbiome by a shotgun metagenomic approach. This endoxylanase candidate was amplified, cloned, heterologously expressed in Escherichia coli and purified. The recombinant enzyme was active at a broad range of temperatures (37-60 ºC) and pH values (4-10), with optimal activity at 50 ºC and pH 9. Moreover, its activity remained at more than 80% of its maximum at 50 °C for 8 h. In addition, Xyl10B was found to be stable in the presence of salt and several ions and chemical reagents frequently used in the industry. These characteristics make this enzyme an interesting candidate for pulp and paper bleaching industries, since this process requires enzymes without cellulase activity and resistant to high temperatures and alkaline pH (thermo-alkaliphilic enzymes). The products of xylan hydrolysis by Xyl10B (short xylooligosaccharides, xylose and xylobiose) could be suitable for application as prebiotics and in the production of bioethanol.

RESUMO

Juvenile hormone (JH) controls insect reproduction and development through an intracellular receptor complex comprising two bHLH-PAS proteins, the JH-binding Methoprene-tolerant (Met) and its partner Taiman (Tai). Many hemimetabolous insects including cockroaches strictly depend on JH for stimulation of vitellogenesis. In termites, the eusocial hemimetabolans, JH also regulates the development of caste polyphenism. Studies addressing the agonist ligand binding to recombinant JH receptors currently include three species belonging to two holometabolous insect orders, but none that would represent any of the hemimetabolous orders. Here, we examined JH receptors in two representatives of Blattodea, the cockroach Blattella germanica and the termite Prorhinotermes simplex. To test the JH-binding capacity of Met proteins from these species, we performed chemical synthesis and tritium labeling of the natural blattodean JH homolog, JH III. Our improved protocol increased the yield and specific activity of [10-3H]JH III relative to formerly available preparations. Met proteins from both species specifically bound [3H]JH III with high affinity, whereas Met variants mutated at a critical position within the ligand-binding domain were incapable of such binding. Furthermore, JH III and the synthetic JH mimic fenoxycarb stimulated dimerization between Met and Tai components of the respective JH receptors of both species. These data present primary evidence for agonist binding by JH receptors in any hemimetabolous species and provide a molecular basis for JH action in cockroaches and termites.

RESUMO

Mound structures that soil termites build have diverse morphologies. Previous observational studies documented that mounds are built to provide regulated environments for the termites that live within them and their structures are formed in ways to support this purpose under the influence of the mounds' immediate environment. The objective of this study is to provide a methodology and a predictive computational model to investigate the reason behind the different but systematic shapes of termite mounds, considering all the relevant forces imposed on them and their thermoregulatory and gas-exchange functions. The gas-exchange function accounts for the capacity of the mound to diffuse metabolic gases generated in the mound's underground nest, while the thermoregulatory function satisfies the connection between the underground nest and deep ground temperatures. The proposed predictive model is based on the principles of heat transfer and thermodynamics and allows optimized mechanically stable structures to freely emerge. The results indicate that, while the model is free to generate any mechanically stable structure, under the relevant environmental and metabolic conditions, it produces structures with forms and geometrical characteristics similar to those of natural mounds. Investigation of the connection between the local environment and the mound shapes indicated that the Sun and wind play an important role in the mound structural form. Mounds exposed to stronger solar irradiance exhibit cone-shaped structures that are pointed towards the Sun, while shaded mounds are observed to be vertical domes. The local wind is observed to affect the external shape of the mound by preventing them to grow tall while controlling the features of the internal structure. By investigating the similarities between structures in different regions (i.e., India, Namibia, and Brazil), it is revealed that, unlike mounds with a strong need for gas-exchange, mounds with a significant demand for thermoregulation exhibit deeper nests, thicker external walls, and well-defined cone- (as opposed to the dome-) shaped structures. Overall, the form of termite mounds is strongly correlated to both regulatory functions and local environments, and the resulting mound shape arises as a combination of these factors.

RESUMO

BACKGROUND: The production of agricultural wastes still growing as a consequence of the population growing. However, the majority of these residues are under-utilized due their chemical composition, which is mainly composed by cellulose. Actually, the search of cellulases with high efficiency to degrade this carbohydrate remains as the challenge. In the present experiment, two genes encoding an endoglucanase (EC 3.2.1.4) and ß-glucosidase (EC 3.2.1.21) were overexpressed in Escherichia coli and their recombinant enzymes (egl-FZYE and cel-FZYE, respectively) characterized. Those genes were found in Trabulsiella odontermitis which was isolated from the gut of termite Heterotermes sp. Additionally, the capability to release sugars from agricultural wastes was evaluated in both enzymes, alone and in combination. RESULTS: The results have shown that optimal pH was 6.0 and 6.5, reaching an activity of 1051.65 ± 47.78 and 607.80 ± 10.19 U/mg at 39 °C, for egl-FZYE and cel-FZYE, respectively. The Km and Vmax for egl-FZYE using CMC as substrate were 11.25 mg/mL and 3921.57 U/mg, respectively, whereas using Avicel were 15.39 mg/mL and 2314.81 U/mg, respectively. The Km and Vmax for cel-FZYE using Avicel as substrate were 11.49 mg/mL and 2105.26 U/mg, respectively, whereas using CMC the enzyme did not had activity. Both enzymes had effect on agricultural wastes, and their effect was improved when they were combined reaching an activity of 955.1 ± 116.1, 4016.8 ± 332 and 1124.2 ± 241 U/mg on corn stover, sorghum stover and pine sawdust, respectively. CONCLUSIONS: Both enzymes were capable of degrading agricultural wastes, and their effectiveness was improved up to 60% of glucose released when combined. In summary, the results of the study demonstrate that the recombinant enzymes exhibit characteristics that indicate their value as potential feed additives and that the enzymes could be used to enhance the degradation of cellulose in the poor-quality forage generally used in ruminant feedstuffs.

RESUMO

This study assessed the potential of termite gut inhabiting bacteria towards bioconversion of cellulosic waste into biofuel. Total seven bacterial isolates from the gut of Heterotermes indicola were isolated. Among all the isolates, HI-1 produced the largest zone upon primary screening. Untreated paper had more cellulose content (73.03%) than acid (0.5%) treated paper that was used as a lignocellulosic substrate for saccharification. Among all the isolates tested, glucose yield (1.08mg/mL) was high for HI-1 isolate. Several factors were considered for optimizing augmented glucose yield (8.57mg/mL) and growth (8.07×108cfu/mL), such as temperature 37°C, pH 4.5, 5% (w/v) substrate concentration, 6 % bacterial inoculum size, agitation 150 rpm with PEG 0.25 % and Ca2+ ions 0.002 g/L. Overall 8-fold increase in glucose yield was achieved. Enzyme activity of HI-1 showed higher endoglucanase 0.29 ± 0.01 (U/mL/min) and exoglucanase 0.15±0.01 (U/mL/min) activity under optimum conditions, mentioned above. temperature 37°C, pH 4.5, substrate concentration 5%, inoculum size 6%, surfactants PEG 0.01%, ions Ca2+(0.002g/L) and agitation (120 rpm). Simultaneous saccharification and fermentation (SSF) of hydrolyzed office paper yielded 5.43mg/mL bioethanol. According to 16S rRNA sequence homology, the bacterial isolate H1 was identified as Alcaligenes faecalis. Bioethanol production from office paper untreated waste proved an effective strategy. Bacteria having natural tendency towards cellulosic waste consumption are promising for bioconversion of cellulosic waste to valuable products.

RESUMO

The minimum set of parameters that can be used to assess the adsorption capacity of activated carbon (AC) produced from termite bio-waste was determined. Three types of AC were prepared: AC600 at 600 °C, MAC600 at the same temperature and impregnated with FeCl3, and AC800 at 800 °C. The influence of the solution pH on the adsorption, adsorption kinetics, isotherms and thermodynamic parameters was considered to characterize the amoxicillin (AMX) adsorption process. The AC materials had surface areas (m2 g-1) of approximately 248.8 for AC600, 501.6 for AC800 and 269.5 for MAC600, with point of zero charge (pHPZC) values of 8.3, 7.5 and 1.7, respectively. A time period of 30 min was chosen for the adsorption kinetics, which was best represented by the pseudo-first-order model for AC600, the intraparticle diffusion model for AC800 and the pseudo-second-order model for MAC600. Regarding the isotherms, a maximum adsorption of 23.4 mg g-1 was found for AC800. In general, the thermodynamic parameters demonstrated a non-spontaneous process. It seems that the medium conditions, the adsorbate and adsorbent characteristics, and the Gibbs free energy are the most important parameters to be considered in a preliminary assessment of the adsorption efficiency of specific adsorbent/adsorbate pairs.

RESUMO

Food scarcity or abundance are factors regulating termites' foraging behavior in general. The aim of the present study is to evaluate the influence of four amounts of Eucalyptus grandis on foraging behavior events shown by worker and soldier of Nasutitermes corniger during laboratory tests. The tests were carried out with adult and active N. corniger colonies found in nests collected in the field, which were stored in glass cubes connected to the test arenas. Four different amounts of wood blocks were used in the tests and each amount concerned a treatment and defined a different experimental group: 1, 2, 3 and 4 blocks/arenas, with 5 repetitions. Each test lasted 60 minutes and consisted in observing, or not, the occurrence of behavioral events shown by foragers when they had contact with the treatment. The duration of each event was recorded, whenever it was observed. The number of recruited foragers and the number of workers consuming the blocks were recorded at the end of each test applied to each treatment. Nasutitermes corniger presented the three behavioral events in all treatments; however, there was not significant difference between treatments in the occurrence of the two first events, in the time taken from test start to the occurrence of a new event, in the number of recruited termites and in the number of gnawing workers. Only workers' mass recruiting was influenced by the amount of wood available. The occurrence of this event was significantly higher in treatments with greater amounts of wood. Thus, N. corniger adjusts its mass recruitment behavior in response to available food amount, which should be considered when developing baiting system for its control. (AU)

The Pyrearinus pumilus species group from South America includes luminescent click beetles (Agrypninae: Pyrophorini) associated with the phenomena of "luminescent termite mounds" and "luminous canga caves". The latter was recently reported in the state of Pará, Brazil. This group includes six species based on the morphology of adults, of which two have immature stages already described. In this work we present the morphology and biological aspects of mature larva and pupa of Pyrearinus pumilus (Candèze, 1863), from the canga caves. Moreover, we provide a key and illustrations for identification of male adults and the known larvae. Our study shows that: (1) morphological characters of immatures support the close relationship of P. pumilus with their allies in the P. pumilus species group; (2) the traits of the known larvae of the P. pumilus species group are reliable for species identification.
