RESUMO

Anammox bacteria are widely applied worldwide for denitrification of urban wastewater. Differently, their application in the case of industrial effluents has been more limited. Those frequently present high loads of contaminants, demanding an individual evaluation of their treatability by anammox technologies. Bioreactors setting up and recovery after contaminants-derived perturbations are slow. Also, toxicity is frequently not acute but cumulative, which causes negative macroscopic effects to appear only after medium or long-term operations. All these particularities lead to relevant economic and time losses. We hypothesized that contaminants cause changes at anammox proteome level before perturbations in the engineered systems are detectable by macroscopic analyses. In this study, we explored the usefulness of short-batch tests combined with environmental proteomics for the early detection of those changes. Copper was used as a model of stressor contaminant, and anammox granules were exposed to increasing copper concentrations including previously reported IC50 values. The proteomic results revealed that specific anammox proteins involved in stress response (bacterioferritin, universal stress protein, or superoxide dismutase) were overexpressed in as short a time as 28 h at the higher copper concentrations. Consequently, EPS production was also increased, as indicated by the alginate export family protein, polysaccharide biosynthesis protein, and sulfotransferase increased expression. The described workflow can be applied to detect early-stage stress biomarkers of the negative effect of other metals, organics, or even changes in physical-chemical parameters such as pH or temperature on anammox-engineered systems. On an industrial level, it can be of great value for decision-making, especially before dealing with new effluents on facilities, deriving important economic and time savings.

When captured, free-living crocodilians respond by hyperstimulation of the hypothalamic-pituitary-adrenal (HPA) axis, which triggers a cascade of downstream events of physiological stress. We examined these responses in two unstressed, and stressed Amazonian caimans, Caiman crocodilus and Melanosuchus niger. Plasma corticosterone levels increased in both stressed caiman species. In M. niger, the levels of this hormone increased 5.2-fold compared with the basal range values, while in C. crocodilus this was only 1.7-fold. After stress, M. niger needed more than 6 h to return its corticosterone levels to basal range values, whereas in C. crocodilus just 0.5 h was enough. Downstream events were characterized by an increase in glucose levels, which is associated with corticosterone increments. Excessive muscle activity resulted in increased plasma lactate content in both species. Lactate levels were also related to plasma calcium concentration, possibly due to the buffering capacity for preventing lactic acidosis. Clearance of excessive lactate load was faster in M. niger (0.5 h) than in C. crocodilus (more than 6 h). Although both caiman species respond in the same way to capture, the amplitude and duration of activation of the HPA axis are different. M. niger may be potentially more sensitive to acute stress than C. crocodilus. On the other hand, C. crocodilus needs more time to recover from the lactic acid load. Our experiment provides a useful diagnostic tool for management and conservation programs, as well as evaluating the impacts of tourism and recreational capture on caimans in the Amazon.

Heat shock proteins (HSP) are synthesized in living tissues exposed to transient increase in temperature and play a central role in the protective response against heat and other stresses. In fruits, this response to heat treatment provides resistance to a physiological alteration known as chilling injury. Despite the physiological importance of this group of proteins, publications comparing different methodological alternatives for their analysis are rather scarce. In the present paper, we conducted a comparative study using different electrophoretic and immunological techniques to evaluate the HSP response in fruits. Proteins were extracted from tomato fruit exposed to an HSP-inducing temperature (38 °C) for different times (0, 3, 20, and 27 h). Different alternatives of analysis (SDS-PAGE, SDS-PAGE followed by IEF, Western blot, and dot blot) were performed, and their potential application discussed. The study was complemented with a practical application, in which tomatoes were subjected to heat and anaerobic treatments and then stored in a chill-inducing temperature. This application evidences the relevance of knowing the level of proteins attained by stress treatments which correlates with the acquired tolerance.

Since mechanical stress in structures affects issues such as strength, expected operational life and dimensional stability, a continuous stress monitoring scheme is necessary for a complete integrity assessment. Consequently, this paper proposes a stress monitoring scheme for cylindrical specimens, which are widely used in structures such as pipelines, wind turbines or bridges. The approach consists of tracking guided wave variations due to load changes, by comparing wave statistical patterns via Principal Component Analysis (PCA). Each load scenario is projected to the PCA space by means of a baseline model and represented using the Q-statistical indices. Experimental validation of the proposed methodology is conducted on two specimens: (i) a 12.7 mm ( 1 / 2 " ) diameter, 0.4 m length, AISI 1020 steel rod, and (ii) a 25.4 mm ( 1 " ) diameter, 6m length, schedule 40, A-106, hollow cylinder. Specimen 1 was subjected to axial loads, meanwhile specimen 2 to flexion. In both cases, simultaneous longitudinal and flexural guided waves were generated via piezoelectric devices (PZTs) in a pitch-catch configuration. Experimental results show the feasibility of the approach and its potential use as in-situ continuous stress monitoring application.

Social species in captivity may face allostatic overload due to artificial grouping and other social constraints. In rescue centres, groups of psittacines are constantly mixed due to the arrival and/or release of individuals; this procedure is potentially harmful to animal welfare. This study aimed at evaluating the possible impacts of mate replacement on the stress levels of captive blue-fronted amazon parrots (Amazona aestiva). During five weeks, we recorded agonistic interactions and dropping-glucocorticoid metabolites (GCM) concentrations of individuals allocated in a group whose members were kept constant and in a group subjected to frequent member replacement. In both groups, non-linear hierarchies developed, without sex differences regarding aggression or hierarchical positions. The replacement of individuals had no effect on the number of agonistic interactions or on the animals' stress levels. In both groups, higher-ranking individuals had higher stress loads than subordinates. Our study, the first to investigate the social dynamics of A. aestiva, indicated that introducing or removing individuals in captive groups does not seem to affect the welfare of the birds in the short term. This information favours release and reintroduction programs and is relevant for conservation management of this, and possibly other parrot species with similar environmental requirements.
