RESUMO

A novel solid-phase extraction methodology followed by UHPLC-MS/MS has been developed for Ochratoxin A (OTA) analysis in herbal infusions. For this purpose, a commercial polyurethane foam (PUF) was used as sorbent, and the experimental conditions were fully optimized. The strategy was satisfactory for reducing the matrix effect and allowed for OTA quantification in black tea and herbal infusions, with suitable recoveries and quantitation limits in agreement with those required by the maximum levels allowed by current regulations. The achieved results demonstrated the unprecedented use of polyurethane foam as an effective alternative for OTA retention and quantification in herbal infusions with the advantages of simple preparation, time saving, sustainability, and low cost for routine analysis.

The aim of this work is to evaluate the changes in compression properties of a bio-based polyurethane foam after exposure to 90 °C for different periods of time, and to propose a method to extrapolate these results and use a numerical approach to predict the compression behaviour after degradation for untested conditions at different degradation times and temperatures. Bio-based polymers are an important sustainable alternative to oil-based materials. This is explained by the foaming process and the density along the material as it was possible to see in a digital image correlation analysis. After 60 days, stiffness was approximately decreased by half in both directions. The decrease in yield stress due to thermo-oxidative degradation had a minor effect in the foaming directions, changing from 352 kPa to 220 kPa after 60 days, and the transverse property was harshly impacted changing from 530 kPa to 265 kPa. The energy absorption efficiency was slightly affected by degradation. The simulation of the compression stress-strain curves were in accordance to the experimental data and made it possible to predict the changes in mechanical properties for intermediate periods of degradation time. The plateau stress for the unaged foam transverse to the foaming direction presented experimental and numerical values of 450 kPa and 470 kPa, respectively. In addition, the plateau stresses in specimens degraded for 40 days present very similar experimental and numerical results in the same direction, at 310 kPa and 300 kPa, respectively. Therefore, this paper presents important information regarding the life-span and degradation of a green PUF. It provides insights into how compression properties vary along degradation time as function of material operation temperature, according to the Arrhenius degradation equation.

The nutrient biological removal from sewage, especially from anaerobic reactor effluents, still represents a major challenge in conventional sewage treatment plants. In this work, the nitrogen and phosphorus removal from anaerobic pre-treated domestic sewage in an up-flow anaerobic sludge blanket (UASB) reactor was assessed in a structured fixed bed reactor (SFBR) operated in a continuous and in a batch mode using polyurethane foam as material support for biomass and fermented glycerol as the exogenous carbon source. The SFBR was operated as a sequencing batch reactor with cycles of 90, 120, and 150 min under anaerobic, oxic, and anoxic conditions, respectively, reaching average efficiencies for total nitrogen and phosphorus removal of 88% and 56%, respectively. Fermented glycerol was added during the non-aerated periods. Under continuous feeding, the SFBR was operated with aeration/non-aeration periods of 2/1 (h) and 3/1 (h), hydraulic retention time of 12 h, and a recirculation ratio of 3. Without fermented glycerol addition, the maximum removal of total nitrogen (TN) reached 42%, while adding glycerol in the non-aerated period improved TN removal to 64.9% (2/1 h) and 69.5% (3/1 h). During continuous operation, no phosphorus removal was observed, which was released during the non-aerated period, remaining in the effluent. Optical microscopy analyses confirmed the presence of polyphosphate granules and of the phosphorus accumulating organisms in the reactor biofilm. It was concluded that the batch feeding method was determinant for phosphorus removal. The structured fixed bed reactor with polyurethane foam proved to be feasible in the removal of organic matter and nutrients remaining in the UASB reactor effluent.

The use of lignocellulosic fillers in rigid polyurethane foams (RPUFs) has been receiving great attention due to their good mechanical and insulation properties and the high sustainable appeal of the obtained cellular polymers, although high water uptakes are found in most of these systems. To mitigate this detrimental effect, RPUFs filled with wood flour (2.5% wt) were fabricated with the addition of furfuryl alcohol (FA) to create a polymer grafted with the wood filler. Two concentrations of FA (10 wt% and 15 wt%) were investigated in relation to the wood flour, and the RPUFs were characterized for cell morphology, density, compressive properties, thermal stability, and water uptake. The introduction of wood flour as a filler decreased the cell size and increased the anisotropy index of the RPUFs and, in addition to that, the FA grafting increased these effects even more. In general, there were no significant changes in both mechanical and thermal properties ascribed to the incorporation of the fillers. On the other hand, a reduction of up to 200% in water uptake was ascribed to the FA-treated fillers.

The relationship between lipopeptide and primary metabolite production by Bacillus spp. in solid-state fermentation (SSF) and submerged fermentation (SmF) was evaluated. Four wild-type strains and one mutant strain (unable to develop biofilm) were assessed in SSF and SmF, using a defined medium and polyurethane foam as inert support for SSF. Strain ATCC 21,332 in SSF presented the highest lipopeptide production. The wild-type strains revealed higher lipopeptide and biomass production and lower synthesis of primary metabolites in SSF than in SmF. However, the mutant strain showed a slightly higher production of primary metabolites in SSF than in SmF. Carbon balance analysis showed that the carbon flux was mainly directed to lipopeptides in SSF, whereas in SmF, it was directed to the production of primary metabolites and the carbon flux to lipopeptides is inversely related to primary metabolites in both types of cultures.

Counter-diffusional biofilms are efficient in the removal of nitrogen from low strength wastewaters. Although counter-diffusion is usually established using expensive gas-permeable membranes, a polyurethane sheet is used to separate the aerobic and anoxic environments in the novel foam aerated biofilm reactor (FABR). Foam sheets with thicknesses of 10, 5 and 2 mm and synthetic wastewater with COD/N ratios of 5 and 2.5 were evaluated. The 2 mm thick foam reactor did not show good biomass adherence and, therefore, did not show N removal efficiency. The 5 and 10 mm reactors, in both COD/N ratios, showed similar total nitrogen and COD removal performance, up to 60% and 80%, respectively. The denitrification efficiency was close to 100% throughout the experimental period. Nitrification efficiency decreased with microbial growth, which was recovered after removal of excessive biomass. Lower values of polyurethane foam thickness and COD/N ratio did not provide a higher nitrification rate, as expected. The increase in resistance to mass transfer was associated with the growth of biomass attached to the foam rather than to its thickness and resulted in specialization of the microbial communities as revealed by 16S amplicon sequencing. FABR reveals as a promising alternative for simultaneous removal of nitrogen and COD from low COD/N ratio wastewaters.

Abstract Two polyurethane foam-based sorbents (PUF) were synthesized by imprinting and grafting techniques and examined for selective separation and preconcentration of caffeine (CAF) in some pharmaceutical products and in black tea. Molecularly imprinted PUF was synthesized based on hydrogen-bonding interactions between CAF and alizarin yellow G (AYG) and subsequent polymerization into PUF. The static experiments indicated optimum sorption conditions at pH=6.5 and 5.5 for imprinted PUF (AY-IPUF) and grafted PUF (AY-GPUF), respectively. In the online experiments, the suitable preconcentration time was found to be 40 and 20s for (AY-IPUF) and (AY-GPUF), respectively, at a flow rate of 1.75 mL.min-1. Desorption of CAF has been affected by passing 500 µL of 0.05, 0.01 mol.L−1 HCl eluent onto (AY-IPUF) and (AY-GPUF), respectively. The online methods have provided satisfactory enrichment factors of 8.4 and 10.5 for (AY-IPUF) and (AY-GPUF), respectively. The time consumed for preconcentartion, elution and determination steps was 1.48 and 1.05 min, thus, the throughput was 42 and 57 h-1, for (AY-IPUF) and (AY-GPUF), respectively. The developed sorbents were studied for the determination of CAF in pharmaceutical samples which will be helpful to minimize caffeinism. Finally, in silico bioactivity, ADMET and drug-likeness predictive computational studies of caffeine were also carried out

ABSTRACT BACKGROUND: Surgical treatment of recurrent anal fistulas can lead to numerous complications, including fecal incontinence. Therefore, sphincter preserving techniques are gaining more popularity. OBJECTIVE: The aim of the study was to assess effectiveness of platelet-rich plasma (PRP) therapy in the patients with recurrent cryptoglandular anal fistulas. METHODS: A cohort of 18 patients with anal fistulas was enrolled into a preliminary and prospective trial. They were divided into two groups consisting of eight and ten patients respectively. PRP was injected locally in all patients, however in the group II it was applied after 7 days drainage of fistulas with polyurethane foam or negative pressure wound therapy. On average, three doses of PRP were administered, but with the opportunity to double the number of applications if it was clinically justified. The patients were evaluated in an out-patient department after fortnight and then in 1, 6, and 12 months following the last PRP application. RESULTS: Anal fistulas were closed in 4 (50%) patients from the group I and in 7 (70%) patients form the group II. Although, the difference between both groups was not statistically significant, PRP therapy should be preceded with fistulous tract drainage in all patients. Summarizing, that successful result was achieved in 11 (60%) patients from the entire group of 18 participants. CONCLUSION: The rate of recurrent cryptoglandular anal fistulas closure reaching 60%, after topical treatment with PRP, exceeds the results of other sphincter-saving methods of treatment. Therefore, it might become a novel method of anal fistulas therapy.


RESUMO CONTEXTO: O tratamento cirúrgico de fístulas anais recorrentes pode levar a inúmeras complicações, incluindo incontinência fecal. Portanto, as técnicas de preservação do esfíncter estão ganhando mais popularidade. OBJETIVO: O objetivo do estudo foi avaliar a eficácia da terapia de plasma rico em plaquetas (PRP) nos pacientes com fístulas anais criptoglandulares recorrentes. MÉTODOS: Uma coorte de 18 pacientes com fístulas anais foi inscrita em ensaio preliminar e prospectivo. Eles foram divididos em dois grupos compostos por 8 e 10 pacientes, respectivamente. PRP foi injetado localmente em todos os pacientes, porém no grupo II foi aplicado espuma de poliuretano ou terapia de feridas por pressão negativa após 7 dias de drenagem de fístulas. Em média, foram administradas três doses de PRP, mas com a oportunidade de dobrar o número de aplicações se fosse clinicamente justificado. Os pacientes foram avaliados em ambulatório após quinze dias e depois em 1, 6 e 12 meses após a última aplicação do PRP. RESULTADOS: As fístulas anais foram fechadas em 4 (50%) pacientes do grupo I e em 7 (70%) pacientes do grupo II. Embora a diferença entre ambos os grupos não tenha sido estatisticamente significante, a terapia PRP deve ser precedida de drenagem do trato fístulo em todos os pacientes. Resumindo, esse resultado bem-sucedido foi alcançado em 11 (60%) pacientes de todo o grupo de 18 participantes. CONCLUSÃO: A taxa de fechamento recorrente de fístulas anais criptoglandulares chegando a 60%, após tratamento tópico com PRP, excede os resultados de outros métodos de tratamento que preservam o esfíncter. Portanto, pode se tornar um novo método de terapia das fístulas anais.

Abstract This study evaluated an intermittently aerated, fixed-bed, single-batch reactor, with mini BioBob© as biofilm media support, as an alternative treatment of craft brewery wastewater. In order to remove chemical oxygen demand (COD) and total nitrogen (TN), seven conditions were performed in a central composite experimental design (CCD) with different aeration times (1, 2 and 3 h in a 4 h cycle) and hydraulic retention times (HRT) (12, 16 and 20 h). The results showed that the removal of COD and TN were positively affected by increased aeration time and HRT. The condition that presented the best quality effluent was Condition No. 1 (20 h HRT and 3 h aeration), with 209 ± 28 mg COD L-1; 3.00 ± 0.15 mg TKN L-1 ; and 0.67 ± 0.11 mg NO3-N L-1. Kinetic assays showed that the highest values for the substrate removal rate constant, kCOD = 0.1774 h-1 were obtained with the longest aeration time (3 h). The most probable number (MPN) test showed a higher concentration of denitrifying bacteria (heterotrophic), 3.3 x 106, than for AOB and NOB bacteria (autotrophic), which were 4.9 x 103 and 2.7 x 103, respectively. Moreover, it was possible to verify that correcting the influent alkalinity with 7.14 mg CaCO3 for each 1 mg of TKN resulted in better process efficiency. It was concluded that COD and TN can be removed from craft brewery wastewater using an intermittently aerated, fixed-bed, single-batch reactor with mini Biobob© as biofilm media support.

The acid mine drainage (AMD) can causes negative impacts to the environment. Physico-chemical methods to treat AMD can have high operational costs. Through passive biological methods, such as anaerobic reactors, sulfate reduction, and recovery of metals are promoted. This study evaluated the performance of a horizontal anaerobic immobilized biomass (HAIB) reactor for the treatment of synthetic AMD using polyurethane foam as support material, and anaerobic sludge as inoculum. Ethanol was used as an electron donor for sulfate reduction, resulting in an influent chemical oxygen demand (COD) in the range of 500-1,500 mg/L and COD/sulfate ratio at 1. A gradual increase of sulfate and COD concentration was applied that resulted in COD removal efficiencies higher than 78%, and sulfate removal efficiencies of 80%. Higher sulfate and COD concentrations associated with higher hydraulic retention times (36 h) proved to be a better strategy for sulfate removal. The HAIB reactor was able to accommodate an increase in the SLR up to 2.25 g SO42-/L d-1 which achieved the greatest performance on the entire process. Moreover, the reactor proved a suitable alternative for reaching high levels of metal removal (86.95 for Zn, 98.79% for Fe, and 99.59% for Cu).

To determine the growth kinetic parameters, substrate consumption and spore yields for Bacillus thuringiensis, liquid fermentation (SmF) and solid-state fermentation (SSF), on polyurethane foam (PUF), were analysed comprising strictly the same media. The analysis included three medium concentrations, maintaining the same C/N ratio, with initial glucose at 12.5, 25, and 50 g L-1 (1X, 2X and 4X, respectively). SSF at 2X and 4X produced higher amounts of total biomass, vegetative growth and even early sporulation. Notably, at all glucose concentrations, sporulation was not inhibited in SSF as seen partially in SmF at 2X, and totally at 4X. Micrographs from PUF cultures showed thin layers of bacteria forming large horizontal aggregates, associated with the higher biomass yields and the early cell differentiation. This is the first work showing that SSF improves spore yields of B. thuringiensis in media with high substrate concentrations, using PUF as a research tool for comparative analysis with application in new production systems including biofilm-forming microorganisms.

A novel bio-based polyurethane/chitosan foam (PU/chitosan) was synthesized using a polyol derived from castor oil and applied to remove Food Red 17 dye (FR17) from aqueous solutions. PU/chitosan foam presented better characteristics and adsorption potential than polyurethane foam (PU). PU/chitosan foam showed a semi-crystalline structure, with several functional groups, high porosity and good mechanical properties. These characteristics are adequate for adsorptive separations. Using identical adsorption conditions, PU/chitosan was able to remove >98% of FR17 dye from the solution, while, PU removed only 40%. The adsorption of FR17 on PU/chitosan composite foam was favored at pH 2. Pseudo-second order model was the most adequate to represent the kinetic data. The equilibrium data followed the Sips model, with a maximum adsorption capacity of 267.24 mg g-1. The adsorption process was spontaneous, favorable and endothermic. The results showed that polyurethane foams are capable to support chitosan, generating an adsorbent with better mechanical characteristics and high potential to remove anionic dyes from aqueous media.

The use of green sources for materials synthesis has gained popularity in recent years. This work investigated the immobilization of lipase NS-40116 (Thermomyces lanuginosus lipase) in polyurethane foam (PUF) using a biopolyol obtained through the enzymatic glycerolysis between castor oil and glycerol, catalyzed by the commercial lipase Novozym 435 for the PUF formation. The reaction was performed to obtain biopolyol resulting in the conversion of 64% in mono- and diacylglycerol, promoting the efficient use of the reaction product as biopolyol to obtain polyurethane foam. The enzymatic derivative with immobilized lipase NS-40116 presented apparent density of 0.19 ± 0.03 g/cm3 and an immobilization yield was 94 ± 4%. Free and immobilized lipase NS-40116 were characterized in different solvents (methanol, ethanol, and propanol), temperatures (20, 40, 60 and 80 °C), pH (3, 5, 7, 9 and 11) and presence of ions Na+, Mg++, and Ca++. The support provided higher stability to the enzyme, mainly when subjected to acid pH (free lipase lost 80% of relative activity after 360 h of contact, when the enzymatic derivative lost around 22%) and high-temperature free lipase lost 50% of relative activity, while the immobilized remained 95%. The enzymatic derivative was also used for esterification reactions and conversions around 66% in fatty acid methyl esters, using abdominal chicken fat as feedstock, were obtained in the first use, maintaining this high conversion until the fourth reuse, proving that the support obtained using environmentally friendly techniques is applicable.

We aimed to use physical methods of microalgal biomass rupture to study saccharification strategies using free and immobilized amylolytic enzymes. The biomass of Spirulina platensis, which consists of 50-60% carbohydrates, was exposed to physical cell rupture treatments, with better results obtained using freeze/thaw cycles following by gelatinization. In saccharification tests, it was possible to hydrolyze Spirulina biomass with hydrolysis efficiencies above 99% and 83%, respectively, using 1% (v/v) of free enzymes or 1% (m/v) of amylolytic enzymes immobilized together. The use of free and immobilized enzymes yielded high levels of conversion of polysaccharides to simple sugars in Spirulina biomass, showing that these processes are promising for the advancement of bioethanol production using microalgal biomass.

This work reports a study about the adsorption of the herbicides diquat and difenzoquat from aqueous medium employing polyurethane foam (PUF) as the adsorbent and sodium dodecylsulfate (SDS) as the counter ion. The adsorption efficiency was shown to be dependent on the concentration of SDS in solution, since the formation of an ion-associate between cationic herbicides (diquat and difenzoquat) and anionic dodecylsulfate is a fundamental step of the process. A computational study was carried out to identify the possible structure of the ion-associates that are formed in solution. They are probably formed by three units of dodecylsulfate bound to one unit of diquat, and two units of dodecylsulfate bound to one unit of difenzoquat. The results obtained also showed that 95% of both herbicides present in 45mL of a solution containing 5.5mgL-1 could be retained by 300mg of PUF. The experimental data were well adjusted to the Freundlich isotherm (r2 ≥ 0.95) and to the pseudo-second-order kinetic equation. Also, the application of Morris-Weber and Reichenberg equations indicated that an intraparticle diffusion process is active in the control of adsorption kinetics.

Polyurethane foam - air partition coefficients (KPUF-air) for 9 polycyclic aromatic hydrocarbons (PAHs), 10 alkyl-substituted PAHs, 4 organochlorine pesticides (OCPs) and dibenzothiophene were measured as a function of temperature over the range 5 °C-35 °C, using a generator column approach. Enthalpies of PUF-to-air transfer (ΔHPUF-air, kJ/mol) were determined from the slopes of log KPUF-air versus 1000/T (K), and have an average value of 81.2 ± 7.03 kJ/mol. The log KPUF-air values at 22 °C ranged from 4.99 to 7.25. A relationship for log KPUF-air versus log KOA was shown to agree with a previous relationship based on only polychlorinated biphenyls (PCBs) and derived from long-term indoor uptake study experiments. The results also confirm that the existing KOA-based model for predicting log KPUF-air values is accurate. This new information is important in the derivation of uptake profiles and effective air sampling volumes for PUF disk samplers so that results can be reported in units of concentration in air.

The present study describes the influence of the concentration of lignin when used as a filler in polyurethane foam for crude oil sorption. The foams (lignin 0-20wt%) were characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), thermogravimetric analysis, contact angle and density. The FTIR analysis confirmed urethane linkage formation, showing that the chemical structure of the polymer was preserved, despite the addition of different lignin concentrations. Thermogravimetric analysis showed that the presence of lignin has altered the onset temperature (Tonset) of the foams, decreasing as the concentration of lignin is increased. The contact angle analysis showed a decrease in the hydrophobicity of the foams with increasing lignin concentration. All modified foams showed an improvement in the oil sorption capacity in a PUF/oil/water system, and the PUF-10 showed an improvement of about 35.5% compared to the PUF-blank. The Langmuir isotherm showed a better fit to the data and predicted a maximum oil adsorption of 28.9gg-1 by the PUF-10. The ΔG° value of -4.4kJmol-1 indicated that crude oil adsorption process by PUF-10 was spontaneous. The results of reuse of the PUF-10 showed that oil removal efficiency remained greater than 95% after five consecutive cycles.

The cationic herbicides paraquat, diquat and difenzoquat are largely used in different cultures worldwide. With this, there is an intrinsic risk of environmental contamination when these herbicides achieve natural waters. The goal of this work was to propose a novel and low-cost sorbent for the removal of the cited herbicides from aqueous medium. The proposed sorbent was prepared by loading polyurethane foam with sodium dodecylsulfate. The influence of several parameters (SDS concentration, HCl concentration and shaking time) on the loading process was investigated. The results obtained in this work demonstrated that all studied variables influenced the loading process, having significant effect on the extraction efficiency of the resulted PUF-SDS. At optimized conditions, the PUF was loaded by shaking 200mg of crushed foam with 200mL of a solution containing 5.0×10(-3)molL(-1) SDS and 0.25molL(-1) HCl, for 30min. The obtained PUF-SDS was efficient for removing the three herbicides from aqueous medium, achieving extraction percentages higher than 90%. The sorption process followed a pseudo second-order kinetics, which presented excellent predictive capacity of the amount of herbicide retained with time.

In this work, the speciation analysis of inorganic antimony (Sb(III) and Sb(V)) is proposed using polyurethane foam loaded with bromopyrogallol red (PUF-BPR) as a selective sorbent for Sb(III). The quantification of Sb in the solutions was performed by graphite furnace atomic absorption spectrometry (GF AAS), and several variables that could affect the performance of the method were carefully evaluated. The best conditions for the sorption of Sb(III) were achieved by shaking 50mg of PUF-BPR with the solutions containing Sb(III) at pH 4.0 for 90 min. In this condition, the retention of Sb(V) was not significant, whereas the removal of Sb(III) from the solution was higher than 95%. The desorption of Sb(III) from loaded PUF-BPR was possible by shaking the loaded foam with 7.5 mL of a 2.5 mol L(-1) HNO3 solution for 30 min. No interference of metal cations was observed on the solid-phase extraction of Sb(III) by PUF-BPR. The developed method was applied in the speciation analysis of antimony in river water samples fortified with different concentrations of Sb(III) and Sb(V). Also, we performed the determination of Sb(III) in a sample of a Sb-based drug utilized in the treatment of leishmaniasis. The results show that Sb(III) can be separated from high concentrations of Sb(V), since recoveries in the range of 81-110% were obtained in the analysis of the samples. The method presented limits of detection and quantification of 0.6 and 2 µg L(-1), respectively, for Sb(III) and 1 and 3 µg L(-1) for Sb(V), respectively.

Pyrethroid pesticide cypermethrin is a environmental pollutant because of its widespread use, toxicity and persistence. Biodegradation of such chemicals by microorganisms may provide an cost-effective method for their detoxification. We have investigated the degradation of cypermethrin by immobilized cells of Micrococcus sp. strain CPN 1 in various matrices such as, polyurethane foam (PUF), polyacrylamide, sodium alginate and agar. The optimum temperature and pH for the degradation of cypermethrin by immobilized cells of Micrococcus sp. were found to be 30 °C and 7.0, respectively. The rate of degradation of 10 and 20 mM of cypermethrin by freely suspended cells were compared with that of immobilized cells in batches and semi-continuous with shaken cultures. PUF-immobilized cells showed higher degradation of cypermethrin (10 mM and 20 mM) than freely suspended cells and cells immobilized in other matrices. The PUF-immobilized cells of Micrococcus sp. strain CPN 1 were retain their degradation capacity. Thus, they can be reused for more than 32 cycles, without losing their degradation capacity. Hence, the PUF-immobilized cells of Micrococcus sp. could potentially be used in the bioremediation of cypermethrin contaminated water.
