RESUMO

Caryocar brasiliense Cambess is a plant species typical of the Cerrado, a Brazilian biome. The fruit of this species is popularly known as pequi, and its oil is used in traditional medicine. However, an important factor hindering the use of pequi oil is its low yield when extracted from the pulp of this fruit. Therefore, in this study, with aim of developing a new herbal medicine, we an-alyzed the toxicity and anti-inflammatory activity of an extract of pequi pulp residue (EPPR), fol-lowing the mechanical extraction of the oil from its pulp. For this purpose, EPPR was prepared and encapsulated in chitosan. The nanoparticles were analyzed, and the cytotoxicity of the encapsu-lated EPPR was evaluated in vitro. After confirming the cytotoxicity of the encapsulated EPPR, the following evaluations were performed with non-encapsulated EPPR: in vitro anti-inflammatory activity, quantification of cytokines, and acute toxicity in vivo. Once the anti-inflammatory activity and absence of toxicity of EPPR were verified, a gel formulation of EPPR was developed for topical use and analyzed for its in vivo anti-inflammatory potential, ocular toxicity, and previous stability assessment. EPPR and the gel containing EPPR showed effective anti-inflammatory activity and lack of toxicity. The formulation was stable. Thus, a new herbal medicine with anti-inflammatory activity can be developed from discarded pequi residue.

RESUMO

The aim of this study was to evaluate the topical application of alcoholic extracts of Dipteryx alata Vogel almonds and bark in skin wound healing in mice. Fifty-four C57BL/6 mice were equally distributed into three groups: Control, Almond, and Bark. A 9 mm skin fragment was resected from the dorsal region of the animals' thorax. The wounds were submitted to topical application of base cream (vehicle), 10% hydroalcoholic almond extract, or bark extract twice a day. Macroscopic, histological, and immunohistochemical evaluations were conducted on the 7th, 14th, and 21st postoperative days. No significant difference was observed regarding skin wound area among groups, with the parameter presenting only a temporal effect on healing (p>0.05). The almond and control groups exhibited more intense collagenization than the bark group (p<0.05). Dipteryx alata Vogel showed to be inert in the wound healing process in mice.(AU)

In the search for the reduction of the side effects caused by allopathy, phytotherapeutic products are already on the veterinary market. This study aimed was to evaluate Elevated Plus-Maze and Hole Board test as an analytical methodology for quality control of anxiolytic medicines based on Passiflora incarnata. The efficacy of three brands commercial differents of Passiflora incarnata extracts was tested in the Elevated Plus-maze (for anxiolytic effect) and the Hole Board test (for locomotor and neophilia activity) in Wistar rats. Treatment with the P. incarnata brands revealed a significant 3-fold increase in the length of stay in the open arms in the LCE. In the ambulation, animals from bands 1 and 3 presented a significant increase, being brand 2 the only one that did not present this relevance. In the neophilia, Passiflora brand 1 presented a significant increase of 3 times and groups 2 and 3 an increase of approximately 2 times. The results showed that different brands of Passiflora have a satisfactory quality standard related to biological activity and showed that the use of Elevated plus-maze/Hole board test can be an effective tool in the quality control of herbal medicines with anxiolytic activity associated with the phytochemical analysis.

Cyperus articulatus L. is popularly known as priprioca. Its rhizomes are used as a medicine in the treatment of malaria in traditional medicine. Since priprioca oil is extracted for commercial purpose, we evaluated if the components from the priprioca residue can be a source of antiplasmodial active molecules. This study aimed to determine the in vitro antiplasmodial and cytotoxicity activities of the ethanolic extract of C. articulatus as an in vitro antiplasmodial agent. From the solid residue of the plant rhizomes, 40 g samples were removed and subjected to hot extraction using a Soxhlet extractor. The in vitro antiplasmodial activity was determined using the W2 and 3D7 strains of P. falciparum. The phytochemical study identified the following main compounds: corymbolone (14.25%), cyclocolorenone (9.75%), and cadalene (8.36%). The extract exhibited moderate IC50 (inhibitory concentration) against the two strains of P. falciparum: 1.21 ± 0.01 against the W2 strain and 1.10 ± 0.06 µg/mL against the 3D7 strain. Our results show the therapeutic potential of priprioca residue as a low-cost antiplasmodial agent.

RESUMO

Silymarin is a phytotherapeutic agent derived from the species Silybum marianum (Asteraceae), commonly is known as milk thistle, and traditionally used as a hepatoprotective; however, recent studies have proposed its use in order to promote lactogenesis, but there are few reports of its effects on the development of offspring. Thus, the objective of this study was to evaluate the effect of silymarin treatment during pregnancy and breastfeeding on the sensory-somatic-motor development and adult behavior of F1-generation Swiss mice. The pregnant females of the parental generation were distributed in four experimental groups and treated orally with doses of 100, 200 or 300 mg/kg of silymarin, with a control group receiving vehicle - vegetable oil (VEH), to obtain the F1-generation. At the end of lactation, the parental generation were submitted to euthanasia. Body mass evolution was determined in both generations. The sensory-motor development of the offspring (F1-generation) was evaluated, and one male pup from each litter was followed up for an analysis of adult behavior. In the F1 analysis, no differences between the groups were observed in initial development from the sensory-somatic-motor analysis performed during the 1st to 21st postnatal days. In the behavioral evaluation of adults from the F1 generation, all the groups from dams treated with silymarin in open field (OF) analysis showed a decrease in the time spent in the periphery and an increase in the time spent in the center, but the ambulation observed by the number of quadrant crossed showed no difference. In addition, during OF, the 100 and 200 mg/kg groups presented an increase in fecal bolus compared with the VEH group. There was a decrease in immobility time in the forced swimming test in the 300 mg/kg group compared to the VEH group. Regarding the memory and learning test, the groups did not differ in their recognition scores. The results of this study using an animal model indicate that treatment with silymarin during pregnancy and breastfeeding does not promote significant morpho-functional changes in the offspring in their initial development and adult behavior, indicating the safety of its use during gestation and lactation.

RESUMO

Sheep farming is an important economic activity from a commercial and subsistence perspective. However, helminth infections present a large sanitary problem for the production, especially haemonchosis caused by strains resistant to traditional anthelmintics. Here, we evaluated the effects of the stem and leaf of Tithonia diversifolia on the inhibition of Haemonchus contortus larval development. We performed a quantitative fecal culture test with feces of sheep inoculated with larvae tolerant to albendazole. Fecal cultures were treated with leaf or stem powder and were compared to the controls containing distilled water or levamisole. The T. diversifolia leaf and stem powders significantly reduced the production of H. contortus larvae. However, leaf powder was more effective, with 64.95% efficacy in the reduction of larvae production, representing a promising alternative in the integrated control of this nematode.

It was evaluated the effects of maternal treatment with the Trichilia catigua (ExTc) crude extract on the antibodies' production by their offspring. Female rats received ExTc or saline from the first day of pregnancy until the twenty-first day after the birth of the pups, when the pups were weaned. All pups were inoculated with two doses of 50 µg of IgY diluted in aluminium hydroxide/PBS on days 26 and 40 of life. Antibody levels were analysed by ELISA. Our results show an increase in levels of IgG1 and IgG2a anti-IgY in female offspring of mothers treated with ExTc compared to female offspring of untreated mothers. Furthermore, ExTc treatment suppressed the production of IgG2a anti-IgY antibodies in males. The data show that maternal exposure to ExTc can modulate the production of antibodies in the offspring.

ABSTRACT The species Mikania glomerata Sprengel, popularly known in Brazil as “guaco”, is widely used as an expectorant to treat respiratory diseases. Its pharmacological activity is mainly attributed to coumarin. The results showed that the method for determining coumarin presented linearity from 0.05 to 0.8 mg mL-1. It was considered selective, accurate, and precise according to the specific resolution from ANVISA, the Brazilian regulatory agency. The proposal of a rapid method for the determining coumarin in extracts of guaco is interesting for routine industrial quality control in order to obtain standardized, efficient, and safe phytotherapic medicines.

RESUMO

Momordica charantia, popularly known as bitter melon, is a plant widely used in ethnobotanical medicine. It has antibacterial, antifungal, anthelmintic, antidiabetic, antiviral, and antimalarial activities, among others. The goal of this study was to evaluate the genotoxic and/or antigenotoxic activity of the aqueous extracts obtained from the aerial parts and fruit of this plant by means of the Drosophila melanogaster wing spot test. Third-stage larvae that obtained standard (ST) cross and high bioactivation (HB) cross were treated with aqueous extracts of the aerial parts (IQA) and fruit (IQF) of M. charantia, following two protocols (genotoxicity and antigenotoxicity). The aqueous extracts are not genotoxic in lower concentrations. The frequencies of mutant spots observed in the descendants of the ST and HB crosses treated with doxorubicin (DXR) alone were 8.65 and 9.25, respectively, whereas in those cotreated with IQA and DXR, the frequencies ranged from 15.90 to 29 in the ST cross and from 15.05 to 24.78 in the HB cross. In cotreatment with IQF, the frequencies ranged from 30.10 to 30.65 in the ST cross and from 13.60 to 14.50 in the HB cross, whereas the frequencies obtained with DXR were 32.50 in the ST cross and 26.00 in the HB cross. In conclusion, the IQA has a synergistic effect, enhancing the genotoxicity of DXR in the ST cross and the HB cross, whereas the IQF has antigenotoxic effects in the HB cross.

RESUMO

Objective: To evaluate in vitro the pH, Brix and antibacterial activity of herbal tinctures of calendula (Calendula officinalis L.) and cashew tree (Anacardium occidentale L.) against the following bacterial strains: Streptococcus mutans (ATCC 25175), Streptococcus oralis (ATCC 10557), Streptococcus salivarius (ATCC 9758), Enterococcus faecalis (ATCC 29212) and Eikenella corrodens (ATCC 23834). Material and Method: The strains were reactivated in agar Brain Heart Infusion (BHI) and seeded in blood agar with BHI. Susceptibility tests were performed and the strains were incubated in microaerophilia (37 °C) for 48 h. Chlorhexidine at 0.12% was used as a positive control and distilled water as a negative control. Dilution of the tinctures was carried out from 1:1 (pure form) to 1:64 in alcohol 70%, which is found in their composition, in order to evaluate a possible influence of alcohol on the antibacterial activity of the tinctures. O Bx readings were made by refractometry using an Abbe refractometer and the pH was measured using a digital pH meter. Results: The calendula tincture had pH=5.43 and 18 °Brix and cashew tree tincture had pH=7.2 and 17 °Brix. The cashew tree tincture did not presentantibacterial activity against S. oralis, but it inhibited the growth of all other bacteria up to the 1:64 dilution. The calendula tincture presented antibacterial activity against S. oralis up to 1:16 dilution (6.5 mm halo diameter); against S. mutans and S. salivarius up to 1:32 dilution (7 and 7.5 mm halo diameter, respectively); and against E. faecalis and E. corrodens up to 1:64 dilution. Alcohol 70% did not show any antibacterial activity against the bacterial strains. Conclusion: The tinctures of calendula and cashew tree have soluble solids, have different pH values and present a significant antibacterial activity against several bacterial strains, except for the cashew tree tincture against S. oralis, which was ineffective...

RESUMO

The Rhipicephalus (Boophilus) microplus is the most important cattle tick in Brazil. For the ticks control, various studies on plants have been developed in an attempt to find extracts with acaricidal properties. This research was aimed at evaluating in vitro and in vivo effects of citronella (Cymbopogon winterianus Jowitt) oil on cattle ticks. Negative control group and eight concentrations of citronella oil and mineral oil (0.5; 1; 2; 5; 10; 20; 50; 100%), were used on in vitro trials with engorged female ticks. The efficacy against cattle ticks was 0; 34.5; 43; 48; 82; 99; 99; 100 and 100%, respectively, for citronella oil. For mineral oil contradictory results of acaricidal efficacy were observed. At the in vivo trial eighteen Holstein cows were allocated to three groups: negative control; citronella oil at 8.6% - level estimated by regression analysis, accounting for 95% efficacy against ticks on in vitro trial and amitraz at 0.025%. Engorged female ticks were counted before, mean of days -3, -2, -1, and after treatment, 1, 2, 3, 5, 7, 10, 14 and 21 days. On the 1st and 2nd milked after treatments, physiologic variables were evaluate (negative control x phytotherapic treatment). The efficacy against tick was 0; 91.3 and 60%, respectively, at 21 days after treatment. Similar results were found with physiologic variables between treatments.

The indiscriminate use of the plant Himatanthus drasticus (Janaúba) by small goat farmers to control nematode parasites in small ruminants was the aim of this phytochemical study of the plant. Barks were collected in the eastern mesoregion of the state of Maranhão, Brazil, and sent to the laboratory for botanical and phytochemical identification by the methodology of Preliminary Prospecting and TLC, testing for the various classes of secondary metabolites. From the ground, dehydrated bark, we conducted the preparation of the crude extract (CE). The material was placed in a water-alcohol mixture of EtOH:H2O (7:3 v:v) and subjected to sporadic mechanical agitation. The subextracts were obtained using the HACE process by liquid-liquid partition, ETOH:H2O (2:1, v:v). The mixtures were prepared with organic solvents of increasing polarity, such as hexane, ethyl acetate and butanol. The GC analysis showed the presence of groups of secondary metabolites in the extract and subextractes. The classes of secondary metabolites that showed greater expressiveness are the alkaloids and tannins, when analyzed by prospecting, and flavonoids and terpenes, when analyzed by TLC, and H. drasticus presents elements indicative of pharmacological action.

On the 1 st and 2nd milked after treatments, physiologic variables were evaluate (negative control x phytotherapic treatment). The efficacy against tick was 0; 91.3 and 60%, respectively, at 21 days after treatment. Similar results were found with physiologic variables between treatments.

RESUMO

This study aimed to evaluate and select different dermatological bases incorporated with propolis for veterinary use as well as to analyze the chemical compounds of the propolis hydroalcoholic extract by LC-MS/MS. Thus, formulations were submitted to accelerated stability tests under different temperatures and to mechanical stress, and evaluated for the appearance, color, odor, pH, viscosity, spreadability, and the mean size of the dispersed globules from the internal phase during a period of three months. The creamy gel formulation showed satisfactory results for all the evaluated items with an excellent capability to incorporate the hydroalcoholic extract of propolis associated to the maintenance of its physicochemical properties. The propolis used in this study had been shown to possess antibacterial and antifungal in vitro activity against the main microorganisms responsible for such diseases. Therefore, the propolis creamy gel described here could be a promising formulation for use in the veterinary medicine.
