RESUMO

It has been reported that chitosan scaffolds, due to their physicochemical properties, stimulate cell proliferation in different tissues of the human body. This study aimed to determine the physicochemical, mechanical, and biological properties of chitosan scaffolds crosslinked with ammonium hydroxide, with different pH values, to better understand cell behavior depending on the pH of the biomaterial. Scaffolds were either neutralized with sodium hydroxide solution, washed with distilled water until reaching a neutral pH, or kept at alkaline pH. Physicochemical characterization included scanning electron microscopy (SEM), elemental composition (EDX), Fourier-transform infrared (FTIR) spectroscopy, Raman spectroscopy, thermogravimetric analysis (TGA), and mechanical testing. In vitro cytotoxicity was assessed via dental-pulp stem cells' (DPSCs') biocompatibility. The results revealed that the neutralized scaffolds exhibited better cell proliferation and morphology. It was concluded that the chitosan scaffolds' high pH (due to residual ammonium hydroxide) decreases DPSCs' cell viability.

The effect of bovine serum albumin (BSA) upon interaction between CdTe QD functionalized by 3-Mercaptopropionic Acid (CdTe-3-MPA QD) and two water soluble porphyrins: positively charged meso-tetra methyl pyridyl porphyrin (TMPyP) and negatively charged meso-tetrakis(p-sulfonato-phenyl) porphyrin (TPPS4), was studied in function of pH using the steady-state and time resolved optical absorption and fluorescence spectroscopies. It was shown that, depending on the charge state of the components, interaction with albumin could either prevent the formation of the QD…PPh complex, form a mixed QD…PPh…BSA complex or not affect PPh complexation with QD at all. The obtained results may be of interest for application in photomedicine.

A new betacoronavirus named SARS-CoV-2 has emerged as a new threat to global health and economy. A promising target for both diagnosis and therapeutics treatments of the new disease named COVID-19 is the coronavirus (CoV) spike (S) glycoprotein. By constant-pH Monte Carlo simulations and the PROCEEDpKa method, we have mapped the electrostatic epitopes for four monoclonal antibodies and the angiotensin-converting enzyme 2 (ACE2) on both SARS-CoV-1 and the new SARS-CoV-2 S receptor binding domain (RBD) proteins. We also calculated free energy of interactions and shown that the S RBD proteins from both SARS viruses binds to ACE2 with similar affinities. However, the affinity between the S RBD protein from the new SARS-CoV-2 and ACE2 is higher than for any studied antibody previously found complexed with SARS-CoV-1. Based on physical chemical analysis and free energies estimates, we can shed some light on the involved molecular recognition processes, their clinical aspects, the implications for drug developments, and suggest structural modifications on the CR3022 antibody that would improve its binding affinities for SARS-CoV-2 and contribute to address the ongoing international health crisis.

Several studies have been conducted worldwide to develop effective and affordable methods to degrade pharmaceuticals and their metabolites/intermediates/oxidation products found in surface water, wastewater and drinking water. In this work, acetaminophen and its transformation products were successfully degraded in surface water by electrochemical oxidation using stainless steel electrodes. The effect of pH and current density on the oxidation process was assessed and the oxidation kinetics and mechanisms involved were described. Additionally, the results were compared with those obtained in acetaminophen synthetic solutions. It was found that conducting the electrochemical oxidation at 16.3 mA/cm2 and pH 5, good performance of the process was achieved and not only acetaminophen, but also its transformation products were totally degraded in only 7.5 min; furthermore, small number of transformation products were generated. On the other hand, degradation rates of acetaminophen and its transformation products in surface water were much faster (more than 2.5 times) and the reaction times much shorter (more than 4.0 times) than in synthetic solutions at all current densities and pH values evaluated. At pH 3 and pH 5, greater soluble chlorine formation due to the higher HCl amount used to acidify the surface water solutions could enhance the degradation rates of acetaminophen and its transformation products. However, constituents of surface water (ions and solids) could also have an important role on the oxidation process because at pH 9 (non-acidified solutions) the degradation rates were also much greater and the reaction times were much shorter in surface water than in acetaminophen synthetic solutions.

Photosensitized oxidation of bovine serum albumin (BSA), by using perinaphtenone as a sensitizer, has been studied at pH 7.4 and 11. The selected sensitizer does not present ground-state complexation with BSA and ensures that the mechanism is mediated by O2 (1 △g ). Strong dependence between BSA-O2 (1 △g ) photo-oxidation and the pH of the medium has been found. The relative oxygen uptake rate (v- â–³ O2 ) and the total quenching rate constant (kt ) values are higher at pH 11 than pH 7.4. The enhancement in the alkaline condition is due to conformational changes in the protein and the reactivity of tyrosinate anion with O2 (1 △g ). Even when the tendency with the pH in the presence of sodium dodecyl sulfate (SDS) micelles is similar to that observed in homogeneous media, an increment on the kt value is detected. This effect may be attributable to the strong interaction of BSA-SDS, which leads to the protein unfolding and could leave more exposed photo-oxidizable amino acids. A protective effect against the O2 (1 △g )-mediated photo-oxidation was observed in reverse micelles (RMs) of sodium bis(2-ethylhexyl)sulfosuccinate (AOT) by comparing the kt values obtained at W = 10 with respect to the one obtain in homogeneous media. The latter could be mainly explained by the modification in the solvent polarity. Also, another important observation was found, the internal pH inside RMs of AOT sensed through tyrosine absorption was independent of the one used for the formation of the water pool. Hence, the kt values observed at both pH, are quite similar.

The non-process-elements (NPE's) in the pulp and paper production, in particular chloride (Cl-) and potassium (K+), eventually accumulate in a sodium sulfate recovery cycle of a paper plant, especially on boiler fly ashes (flue gas cleaning). This accumulation often leads to fouling and corrosion. Thus, in order to reduce such problems, a leaching unit is typically integrated aiming to remove Cl- and K+ and recover sodium sulfate (Na2SO4) from boiler fly ashes. In this context, this research is focused on investigating the effects of temperature, ash concentration and pH on the separation efficiency (Cl- and K+ removal and Na2SO4 recovery) of recovery boiler fly ashes using design of experiments in a devised laboratory setup. As expected from the literature, the results obtained reveals that concentration has the most significant effect on the response variables followed by temperature; whereas pH, not mentioned in this application so far, also has a significant effect. The optimum operating condition, which yields 84 wt% of Na2SO4 recovery and 97 wt% of Cl- and K+ removal, is achieved at 60 °C, 50 wt% of ash and pH 9.0. For practical applications, this optimal condition reduces the accumulation of NPE's and mitigates the risks of fouling and corrosion.

Gold nanoparticles (AuNPs) are currently under intense investigation for biomedical and biotechnology applications, thanks to their ease in preparation, stability, biocompatibility, multiple surface functionalities, and size-dependent optical properties. The most commonly used method for AuNP synthesis in aqueous solution is the reduction of tetrachloroauric acid (HAuCl4) with trisodium citrate. We have observed variations in the pH and in the concentration of the gold colloidal suspension synthesized under standard conditions, verifying a reduction in the reaction yield by around 46% from pH 5.3 (2.4 nM) to pH 4.7 (1.29 nM). Citrate-capped AuNPs were characterized by UV-visible spectroscopy, TEM, EDS, and zeta-potential measurements, revealing a linear correlation between pH and the concentration of the generated AuNPs. This result can be attributed to the adverse effect of protons both on citrate oxidation and on citrate adsorption onto the gold surface, which is required to form the stabilization layer. Overall, this study provides insight into the effect of the pH over the synthesis performance of the method, which would be of particular interest from the point of view of large-scale manufacturing processes.

Ferritin is a globular protein that consists of 24 subunits forming a hollow nanocage structure that naturally stores iron oxyhydroxides. Elimination of iron atoms to obtain the empty protein called apoferritin is the first step to use this organic shell as a nanoreactor for different nanotechnological applications. Different protocols have been reported for apoferritin formation, but some are time consuming, others are difficult to reproduce and protein recovery yields are seldom reported. Here we tested several protocols and performed a complete material characterization of the apoferritin products using size exclusion chromatography, UV-vis spectroscopy, inductively coupled plasma optical emission spectrometry and dynamic light scattering. Our best method removes more than 99% of the iron from loaded holoferritin, recovering 70-80% of the original protein as monomeric apoferritin nanocages. Our work shows that pH conditions of the reduction step and the presence and nature of chelating agents affect the efficiency of iron removal. Furthermore, process conditions also seem to have an influence on the monomer:aggregate proportion present in the product. We also demonstrate that iron contents markedly increase ferritin absorbance at 280nm. The influence of iron contents on absorbance at 280nm precludes using this simple spectrophotometric measure for protein determination in ferritin­iron complexes. Apoferritin produced following our protocol only requires readily-available, cheap and biocompatible reagents, which makes this process standardizable, scalable and applicable to be used for in vivo applications of ferritin derivatives as well as nanotechnological and biotechnological uses.

Sensitized photooxidation processes in the presence of natural pigments may provide an alternative to antibiotics degradation since these compounds are transparent to natural light irradiation, therefore, they can be degraded by the action of photosensitizers which absorb light and produce highly reactive species, especially those derived from molecular oxygen (ROS). Most antibiotics used currently belong to a group of pharmaceutical substances that have been considered a new type of contaminants due to their persistence and bioaccumulation in the environment. OBJECTIVE: In this context, we decided to investigate the kinetic and mechanistic aspects of Vancomycin (Vanco) photosensitized degradation in the presence of the natural pigment Riboflavin (Vitamin B2, Rf) and the artificial dye Rose Bengal (RB) for comparative purposes. METHODS: The study have been done by using Stationary photolysis, Laser flash photolysis, Time-resolved phosphorence detection of O2(1Δg) experiments and Bactericidal activity evaluation. The experiments were carried out in aqueous solution at different pH values in order to establish relationships between the structure of the compound and its susceptibility to ROS-mediated photooxidation. RESULTS: Experimental evidence indicates that in the presence of Rf there is considerable contribution of the radical-mediated mechanism, while in the presence of RB the photooxidation process occurs exclusively through O2(1Δg) and the reactivity to this excited species increases with increasing pH of the environment. DISCUSSION: The results obtained, have been shown that Rf can raise the photodegradation of Vanco by both the radical pathway and the O2(1Δg) mediated. Furthermore, the antibiotic is able to interact with the excited electronic states of Rf as well as O2(1Δg) generated by energy transfer between the excited triplet state of the photosensitizer and the oxygen ground state. The predominant mechanism for photodegradation of Vanco in the presence of the Rf is the radical via because of the considerable interaction with the excited triplet state of the photosensitizer demonstrated by laser flash photolysis experiments. Microbiological test on Staphylococcus aureus ATCC25923 showed that the bactericidal activity of the antibiotic on the strain studied was affected by the sensitized photodegradation process, suggesting that photoproducts generated eventually do not retain the bactericidal properties of the original antibiotic.

background: Lactic acid (LA) is a carboxylic acid widely used as preservative, acidulant, and/or flavouring in food industry; it is also used as a raw material for the production of lactate ester, propylene glycol, 2,3-pentanedione, propanoic acid, acrylic acid and acetaldehyde. In recent years, the demand for LA production has dramatically increased due to its application as a monomer for poly-lactic acid synthesis, a biodegradable polymer used as a plastic in many industrial applications. LA can be produced either by fermentation or chemical synthesis; the former route has received considerable interest, due to environmental concerns and the limited nature of petrochemical feedstocks; thus, 90% of LA produced worldwide is obtained by fermentation, this process comprises the bioconversion of a sugar solution (carbohydrates) into LA in the presence of a microorganism. Objectives: This work is aimed at studying the effect of pH control and culture media composition on the LA production using renewable sources from the agroindustry sector. Methods: A Lactobacillus brevis strain is used to perform lab scale experiments under aerobic and anaerobic conditions, using three different culture media compositions: a high nutritional content medium (MRS), as a reference, a low nutritional content medium with glucose as the only carbon source (GM), and a potential low nutritional content medium with cassava flour as carbon source (HY1). results: The higher LA production is accomplished under anaerobic conditions, 17.6 ± 0.1, 12.6 ± 0.2 y 13.6 ± 0.2 g LA/L, for MRS, GM and HY1 medium, respectively. The effect of pH on LA biosynthesis in a 5L bioreactor is also studied using the HY1 medium. For a fermentation time of 120 h, the highest LA concentration obtained was 24.3 ± 0.7g LA/L, productivity 0.20 g/L/h, YP/S 0.32g LA/g syrup, at pH 6.5...
