RESUMO

Enhancing the initial stages of plant growth by using polymeric gels for seed priming presents a significant challenge. This study aimed to investigate a microgel derived from polyetheramine-poly(propylene oxide) (PPO) and a bisepoxide (referred to as micro-PPO) as a promising alternative to optimize the seed germination process. The micro-PPO integrated with an iron micronutrient showed a positive impact on seed germination compared with control (Fe solutions) in which the root length yield improved up to 39%. Therefore, the element map by synchrotron-based X-ray fluorescence shows that the Fe intensities in the seed primers with the micro-PPO-Fe gel are about 3-fold higher than those in the control group, leading to a gradual distribution of Fe species through most internal embryo tissues. The use of micro-PPO for seed priming underscores their potential for industrial applications due to the nontoxicity results in zebrafish assays and environmentally friendly synthesis of the water-dispersible monomers employed.

RESUMO

Spent mushroom substrate (SMS) is a promising alternative for supplementing oyster mushroom substrate, replacing conventional cereal bran. Therefore, the objective was to evaluate the production of Pleurotus ostreatus supplemented with Lentinula edodes' SMS, through the nutritional analysis of the substrate. Wheat straw was used as substrate and supplemented with rice bran (RB) or SMS in 0%, 7%,15% and 30%. Ca, K, Mg, Mn, Zn, Cu and Fe contents of the cultivation substrates (before and after harvest) were determined through atomic absorption spectrophotometry. Mycelial growth (cm²/day), mycelial time colonization (days), number of clusters, number of pileus, average clusters weight (g), pileus lenght (cm) and width (cm), productivity (1st, 2nd and 3rd flush) (%), biological efficiency (%) of mushrooms were evaluated. Results indicated mycelial growth was higher (0.87 cm²/day compared to the Control) when the substrate was supplemented regardless of the source. The proportions of 15% of SMS achieved the highest biological efficiency (107% - 15% SMS versus 66% - Control). The only nutrients that showed differences in absorption were Ca, K and Mn, in which substrates supplemented with SMS had greater absorption of Ca (5.37 g.kg- 1 versus 1.94 g.kg- 1 in Control) while substrates supplemented with RB absorbed more K (6.56 g.kg- 1 versus 3.74 g.kg- 1 in Control). The mineral composition of the substrate has a direct impact on the growth and yield of P. ostreatus, highlighting the potential of SMS as a alternative to traditional bran supplementation.

RESUMO

Phosphorus (P) availability is important for metabolic process, tillering and formation of a vigorous root system in sugarcane, but sugarcane varieties differ in P uptake efficiency. This study evaluated the enzymatic, nutritional, and biometric parameters of two sugarcane varieties under two conditions of P availability by monitoring the initial development of plants grown in nutrient solution. The experiment was performed using randomized complete block design (RCBD) with five replicates and included two varieties, RB966928 (high nutritional requirements) and RB867515 (low nutritional requirements), and two concentrations of P in the nutrient solution: low (2 mg L-1) and suitable (16 mg L-1). Carbohydrate concentrations and partitioning, leaf nutrient concentrations, enzymatic activity, and shoot and root biometric parameters were analyzed. Regardless of sugarcane variety and the part of the plant, reducing sugar were approximately 32.5% higher in RB867515 and 38.5% higher in RB966928 under suitable P compared with low P. Sucrose concentrations were significantly higher in both varieties under suitable P than in low P. According to PCA, the relationship between reducing sugars and sucrose was closer in RB966928 than in RB867515. Under low P, soluble protein content decreased, and the activities of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX) and the concentrations of hydrogen peroxide (H2O2), and malondialdehyde (MDA). The variety RB966928 under suitable P appears to have a high capacity for proline (120%) upregulation under abiotic stress compared with RB867515 (54%), and thus higher biomass accumulation of this RB966928 variety; however, RB867515 had superior results compared to RB966928 under low P. Suitable P increased leaf concentrations of N, P, Mg, B, and Mg and decreased leaf Zn content. Root and shoot dry matter, root length, plant height, and root and stalk diameter increased by suitable P. Regardless of variety, both nutritional and biometric parameters were directly influenced by P levels, including sugarcane yield. In relation of sugarcane dry matter, RB966928 was less sensitive to low P levels and more responsive to P supply than RB867515 and thus may be more suitable for environments in which P is limiting.

RESUMO

Studying the composition of a certain food is not enough to predict its health benefits. Research over the past decades has decisively strengthened the notion that any putative health benefit is best related to the fraction of compounds transferred from ingested foods into the body since the absorption may be incomplete after oral consumption. In other words, the bioavailability of food components is crucial information. Therefore, a variety of in vitro models have been developed to predict their bioaccessibility and bioavailability in the most diverse food matrices and food products. These models can also be applied to study the impact of several endogenous or exogenous factors on the bioaccessibility and bioavailability of nutrients and bioactive compounds, guiding nutrition and food scientists, technologists, and engineers towards the development of strategies to optimize the positive impact of the diet on well-being and quality of life. While bioavailability is ideally examined in human volunteers, in vitro digestion methods, as well as intestinal absorption and microphysiological models, simulate human physiological conditions. Additionally, in vitro methods are alternatives to offset ethical, economical, and experimental limitations associated with in vivo studies conducted either with individuals or animals. This graphical review draws parallels between in vitro models mimicking digestion processes, uptake, absorption, metabolism, and distribution of dietary compounds and human physiology.

RESUMO

The effect of in ovo threonine (Thr) supplementation on the ileal expression of glucose, peptide and amino acid transporters was assessed in Salmonella Enteritidis-challenged broiler chicks. At 17.5 days of incubation, fertile eggs were supplemented in the amniotic fluid with sterile saline or 3.5% threonine. Hatchlings were individually weighed, and Salmonella Enteritidis negative status was confirmed. At 2 days of age, half of the birds of each group were inoculated with sterile nutrient broth or Salmonella Enteritidis inoculum. Relative expression of sodium-dependent glucose transporter 1 (SGLT1), glucose transporter 2 (GLUT2), di- and tri-peptide transporter 1 (PepT1) and alanine, serine, cysteine, threonine transporter (ASCT1) was assessed at hatch, 2 and 9 days of age, i.e., before inoculation and 7 days post-inoculation (dpi). At 9 days of age (7dpi), threonine increased SGLT1 and GLUT2 expression, whereas GLUT2 expression decreased in Salmonella-challenged birds. There was a significant interaction between threonine and Salmonella for PepT1 and ASCT1. Threonine increased PepT1 expression only in non-challenged birds. In addition, in ovo supplementation increased expression of ASCT1 regardless of post-hatch inoculation; Salmonella inoculation resulted in decreased expression of ASCT1 only in supplemented birds. The results suggest that while intra-amniotic threonine administration in broiler embryos increases the expression of genes related to the absorption of monosaccharides and amino acids, Salmonella challenge may negatively affect the expression of protein related transporters in the ileum of broilers.

RESUMO

It is important to consider the nutritional demand among genotypes of the same species to achieve high yields. Thus, the objective of this study was to determine the concentration of nutrients in leaves, flowers, grains, and fruit straw in conilon coffee genotypes. The experiment was carried out under a randomized block design, with three replications and the evaluation of nine genotypes. Leaf collections were carried out every three months, from August 2019 to May 2020. Flowers were collected at flowering in July 2019 and fruits in June 2020, upon complete maturation of the genotypes. The materials were dried in an oven and sent for laboratory analysis to determine the nutritional content. Data were subjected to analysis of variance followed by a comparison of means and estimates of genetic parameters and clustering using the hierarchical method (UPGMA). The nutrients found in the highest concentrations in the evaluated plant organs were N and K for macronutrients and Fe for micronutrients. For the leaves, the concentrations of the main nutrients were high in the first and reduced in the last evaluated periods, possibly due to mobilization to the fruits. Considering all the plant tissues evaluated, the order of concentration of macronutrients and micronutrients was N > K > Ca > P = Mg = S and Fe > B > Mn > Cu > Zn, respectively. For a nutritional diagnosis, it is important to take comparisons of the genetic diversity and evaluation periods into consideration.

RESUMO

This work deepens our understanding of starch digestion and the consequent absorption of hydrolytic products generated in the human small intestine. Gelatinized starch dispersions were digested with α-amylase in an in vitro intestinal digestion system (i-IDS) based on a dialysis membrane process. This study innovates with respect to the existing literature, because it considers the impact of simultaneous digestion and absorption processes occurring during the intestinal digestion of starchy foods and adopts phenomenological models that deal in a more realistic manner with the behavior found in the small intestine. Operating the i-IDS at different flow/dialysate flow ratios resulted in distinct generation and transfer curves of reducing sugars mass. This indicates that the operating conditions affected the mass transfer by diffusion and convection. However, the transfer process was also affected by membrane fouling, a dynamic phenomenon that occurred in the i-IDS. The experimental results were extrapolated to the human small intestine, where the times reached to transfer the hydrolytic products ranged between 30 and 64 min, according to the flow ratio used. We consider that the i-IDS is a versatile system that can be used for assessing and/or comparing digestion and absorption behaviors of different starch-based food matrices as found in the human small intestine, but the formation and interpretation of membrane fouling requires further studies for a better understanding at physiological level. In addition, further studies with the i-IDS are required if food matrices based on fat, proteins or more complex carbohydrates are of interest for testing. Moreover, a next improvement step of the i-IDS must include the simulation of some physiological events (e.g., electrolytes addition, enzyme activities, bile, dilution and pH) occurring in the human small intestine, in order to improve the comparison with in vivo data.

RESUMO

Flying mammals present unique intestinal adaptations, such as lower intestinal surface area than nonflying mammals, and they compensate for this with higher paracellular absorption of glucose. There is no consensus about the mechanistic bases for this physiological phenomenon. The surface area of the small intestine is a key determinant of the absorptive capacity by both the transcellular and the paracellular pathways; thus, information about intestinal surface area and micro-anatomical structure can help explain differences among species in absorptive capacity. In order to elucidate a possible mechanism for the high paracellular nutrient absorption in bats, we performed a comparative analysis of intestinal villi architecture and enterocyte size and number in microchiropterans and rodents. We collected data from intestines of six bat species and five rodent species using hematoxylin and eosin staining and histological measurements. For the analysis we added measurements from published studies employing similar methodology, making in total a comparison of nine species each of rodents and bats. Bats presented shorter intestines than rodents. After correction for body size differences, bats had ~41% less nominal surface area (NSA) than rodents. Villous enhancement of surface area (SEF) was ~64% greater in bats than in rodents, mainly because of longer villi and a greater density of villi in bat intestines. Both taxa exhibited similar enterocyte diameter. Bats exceeded rodents by ~103% in enterocyte density per cm2 NSA, but they do not significantly differ in total number of enterocytes per whole animal. In addition, there is a correlation between SEF and clearance per cm2 NSA of L-arabinose, a nonactively transported paracellular probe. We infer that an increased enterocyte density per cm2 NSA corresponds to increased density of tight junctions per cm2 NSA, which provides a partial mechanistic explanation for understanding the high paracellular absorption observed in bats compared to nonflying mammals.

RESUMO

The effects of in ovo feeding with threonine (Thr) on intestinal morphology, ileal gene expression and performance of broiler chicken between 1 and 21 d of age (d) were assessed. On day 17.5 of incubation, fertile eggs were randomly allotted to 5 treatments of Thr injection in the amniotic fluid (0; 1.75; 3.5; 5.25; 7%, corresponding to 17.5; 35; 52.5 and 70 mg Thr/mL). After hatch, chicks were given a commercial corn-soybean diet up to 21 d. Daily feed intake (FI), body weight (BW), and food conversion ratio (FCR) were measured from 1 to 7, 14, and 21 d of age. The ileal gene expression of mucin (MUC2), peptide transporter (PepT1), and aminopeptidase enzyme (APN) were evaluated on day of hatch and at 21 d, as well as intestinal morphometric traits. In ovo feeding with threonine significantly increased final weight (FI) and weight gain (WG) and decreased FCR in the period from 1 to 21 d. Threonine levels affected beneficially the villus height, vilo: crypt ratio and villus area on day of hatch and at 21 d. At hatch, all Thr levels increased the expression of MUC2 and PepT1 compared to the control group. APN expression also increased, but for the lowest and the highest threonine levels (1.75 and 7%). At 21 d, there was no effect of threonine on the expression of MUC2, PepT1, and APN. In conclusion, in ovo threonine feeding beneficially affected the morphological and functional development of the intestinal mucosa, which ensured improved performance of chicks at hatch and at 21 d.

The gut of insects is a less protected interphase against external agents. The identification of the molecular mechanisms involved in physiological-digestive processes allows one to find potential targets for insect control. The molecules involved in nutrient absorption, buffering and fluid fluxes in the midgut of the insect-model M. domestica (Diptera Cyclorrhapha) were identified. For this, gene expression along the midgut was analyzed; proteins were identified and annotated by bioinformatics; apical localization of proteins was confirmed by proteomics of purified microvillar membranes; the role of proteins was confirmed by in vivo experiments using different diets, dyes and inhibitors. Middle midgut acidification occurs by the action of an apical H+ V-ATPase with protons coming from carbonic anhydrase activity accompanied by chloride transported with potassium by a K+Cl- symporter. Potassium is recovered by a potassium channel, and cation homeostasis maintained by a basolateral Na+/K+-ATPase. Acidification is directly regulated by a Na+/H+ antiporter and indirectly by a copper homeostasis protein. Mucus protecting epithelium is neutralized with bicarbonate generated by a GPI-ancored carbonic anhydrase and transported by a Na+HCO3-/H+Cl- antiporter. Intracellular acidification is avoided by a basolateral Na+/H+ antiporter. Posterior midgut alkalization occurs by the action of an apical ammonia transporter and proton amino acid (and peptide) symporters. Intracellular acid is eliminated by a basolateral H+ V-ATPase. Fluid fluxes are generated by K+Cl- and Na+Cl-Cl- symporters helped by aquaporins. Inhibition of these symporters showed that the countercurrent flux of water allows trypsin recycling. Finally, posterior midgut is the main location of nutrient absorption, except for copper which is absorbed in the middle midgut

RESUMO

Physiological data showed that T. molitor midgut is buffered at pH 5.6 at the two anterior thirds and at 7.9 at the posterior third. Furthermore, water is absorbed and secreted at the anterior and posterior midgut, respectively, driving a midgut counter flux of fluid. To look for the molecular mechanisms underlying these phenomena and nutrient absorption as well, a transcriptomic approach was used. For this, 11 types of transporters were chosen from the midgut transcriptome obtained by pyrosequencing (Roche 454). After annotation with the aid of databanks and manual curation, the sequences were validated by RT-PCR. The expression level of each gene at anterior, middle and posterior midgut and carcass (larva less midgut) was evaluated by RNA-seq taking into account reference sequences based on 454 contigs and reads obtained by Illumina sequencing. The data showed that sugar and amino acid uniporters and symporters are expressed along the whole midgut. In the anterior midgut are found transporters for NH3 and NH4+ that with a chloride channel may be responsible for acidifying the lumen. At the posterior midgut, bicarbonate-Cl- antiporter with bicarbonate supplied by carbonic anhydrase may alkalinize the lumen. Water absorption caused mainly by an anterior Na+-K+-2Cl- symporter and water secretion caused by a posterior K+-Cl- may drive the midgut counter flux. Transporters that complement the action of those described were also found.

In the nutritional treatment of diarrhea, good results have been obtained using liquid formulas with chicken meat together with cereals and/or starchy vegetables, prepared at home or at the clinic. In a previous study, formulas prepared with chicken meat or isolated soybean protein were tested in the treatment of infantile diarrhea. In order to determine if the results obtained in children could be reproduced in rats, the objective of the present study was to evaluate the effectiveness of the same diets in the nutritional treatment of well-nourished as well as undernourished rats with lactose induced diarrhea. The results showed that all the rats with diarrhea absorbed and retained less of the dietary nutrients and consequently, grew less than their counterparts without diarrhea. However, the absorption and retention measured in the nourished as well as in the undernourished rats were similar, indicating that a short period of malnutrition previous to the diarrhea episode, did not affect rats' absorption capacity. In addition, similarly to the results previously obtained in children, the rats with lactose induced diarrhea utilised equally well the dietary nutrients present in the chicken as well as in the soybean protein based diets. These results confirmed the value of isolated soybean protein in the nutritional treatment of diarrhea and showed that lactose induced diarrhea in rats is an appropriate model to study the nutritional consequences of diarrhea.

Adequate amounts and balanced proportions of nutrient supply is fundamental for the forage productive process. For the fertilization handling, forage plant nutrient requiriment must be known and, consequently, the plant capacity to extract soil nutrients. Therefore, the research had as objective to evaluate the nutrient extraction by Brachiaria brizantha phytomass under nitrogen doses. A randomized complete block experimental design with three replications in a plot repeated in time was used. In the plots, a factorial 3 x 4 arrangement was used, being three cultivars of Brachiaria brizantha (MG-4, Marandu and Xaraés) and four nitrogen doses (0, 50, 100 and 150 mg.dm-3). Three cuts of evaluation were allocated in time, referring. to the time of cuts. The source of nitrogen used was urea. The sharpest treatment effects on phytomass production of Brachiaria brizantha cultivars and nutrient extraction were influenced by the highest nitrogen doses. Xaraés-grass showed greater productivity in relation to the other cultivars, being.considered more responsive to the nitrogen fertilization. The maximum macronutrient extraction by Brachiaria brizantha followed the decreasing.order for the macronutrients: K > N > P > Mg.> S and micronutrients: Mn > Faith > Zn > Cu, being. necessary to restore soil nutrients, with the increase of nitrogen.

RESUMO

Iron-defi ciency anemia affects many groups, with a prevalence of more than 50% among preschoolers in Brazil. One of the many possible approaches to defeat the problem is the regular and suffi cient offer of dietary iron, combined with absorption agonists in the same meal. The objective of the present work was testing the quality of the diet offered to preschoolers assisted by the National School Meal Program (PNAE û Programa Nacional de Alimentação Escolar) in Campinas, in the countryside of São Paulo state. Energy (Kcal), total iron (mg) and absorbable iron (mg) were measured. The sample was composed by 167 children aged 25 to 72 months in 2003. After quality evaluation, only the data from 154 subjects were analyzed. Results showed that iron consumption is higher among girls: 73% of the girls presented a consumption above the RDA value, against 51% of the boys (p<0.05). There were no statistical differences for energy (p = 0.069), total iron (p = 0.395) and absorbable iron (p = 0.353) consumption between the genders. Absorbable iron in preschoolers' meals is expected to be as much as an average 5% of the total iron intake. However, the main reason for this low absorption is not the poor frequency or the scarcity of dietary iron sources, but the limited offer of agonistic absorption factors (mainly meat and vitamin C) in the meals.

The experiment was carried out in a green house, at Embrapa Arroz e Feijão. It aimed to evaluate the offect of cutting period on dry mass production and bromatologic- chemical composition of Brachiaria brizantha cv. MG-5. The experimental design was completely randomized, with five replications. The treatments were constituted by four cutting intervals (15, 20, 30 and 60 days). After cuttings at a distant from soil, of ammonium sulfate (100 mg dm-3) was applied to all treatments. Four cuttings were carried out at 15 - day intervals, three at 20 day-intervals, two at 30 - day intervals and one at a 60 day-interval. Increases in cutting intervals caused dry mass production, neutral detergent fiber and acid detergent fiber rase. However, the opposite was observed for bromatologic composition and phosphorus, copper and iron concentrations.

RESUMO

Anato plant seeds (Bixa orellana var. Peruana) were germinated in a soil + organic matter (2:1) substrate that received triple superphosphate (1.5 g/l). After 115 days, plants were transplanted to a high fertility Dark Red Latosol. Spacing was 5m x 5m a top dressing with 5g of KCl and l0g urea per plant was applied at the 201 day. Starting on day 115 plants (4 per sampling) were collected at 60 day intervals until the 507 day. Samples were subdivided in leaves, branches, stem and reproductive parts. At the 507 day fruits were taken and divided into seeds and hulls. Anato plants grew slowly until the 267 day and improved growth rate from then on to the 447 day, when growth almost stopped. The macro and micronutrient extraction by a 400 plant per hectare crop at the 507 day were: N - 25.7 kg, P - 4.1 kg, K - 16.7 kg, Ca - 16.4 kg, Mg - 4.4 kg, S - 2.5 kg, B - 56.4 g, Cu - 16.2 g, Fe - 240.1 g, Mn - 130.3 g and Zn - 50.7 g.
