RESUMO

The aim of this study was to evaluate the effects of increasing doses of putrescine injected in ovo on hatchability, intestinal morphology and pre-starter performance of broilers. For this purpose, 720 eggs from broiler breeders were separated into a negative control (no injection) and injection treatments with increasing doses of putrescine (0.05; 0.1; 0.15 and 0.2%), totalling five treatments of 144 eggs each. Eggs were distributed in a completely randomized design inside the setter and the injection of solutions occurred at 17 days of incubation. After hatch, 330 birds were housed in mixed lots following the original treatments, totalling 5 treatments of 6 replicates with 11 birds each. Six birds per treatment were weighed and euthanized by cervical dislocation to collect the liver, intestine and breast 24 hr after injection, at hatch and 24 hr after hatch. At 2 days of age, intestines were collected from 4 animals per treatment to analyse histomorphology. The effects of putrescine levels were evaluated by polynomial regression models, ANOVA and Tukey test at 5% probability. The hatchability decreased linearly in response to increased doses of putrescine. The percentage of residual yolk was lower in animals that received putrescine compared to the control. After injection, the percentage of breast increased linearly, and the percentage of intestine had a quadratic response to increased doses of putrescine. However, 24 hr after hatch, the percentage of intestine linearly decreased, and the percentage of liver linearly increased in response to increased doses of putrescine. Villus height increased quadratically, crypt depth decreased linearly, and goblet cells increased linearly in response to the putrescine dose. FI and BWG were not affected in the pre-starter phase; however, FCR increased in response to increased levels of putrescine. Due to putrescine effects on embryos, it is recommended that the doses injected in ovo not exceed 0.1%.

The intestinal physiology and mechanisms involved in nutrient transport are not well established in quails (Coturnix coturnix japonica). The present study assessed the growth performance, morphological development, duodenal density and the expression of Sglt1 and Glut2 of female Japanese quails from 1 to 49 days of age. The three small intestine segments were sampled weekly from 1 to 49 days of age to evaluate villus height, crypt depth and villus: crypt ratio, and goblet cell counts. Scanning electronic microscopy was used to determine duodenal villus density, and real-time polymerase chain reaction (qPCR) was used to study the sodium/glucose cotransporter-1 Sglt1 and glucose transporter Glut2 in the jejunum. Villus height and crypt depth in the duodenum, jejunum and ileum increased with age until 42 and 49 days of age (P < 0.001), and regression analysis evidenced a quadratic effect (P < 0.0001), indicating increasing values to a maximum and then a decrease afterwards. Goblet cell counts increased (P < 0.001) in duodenum, jejunum and ileum from 1 to 42 days, decreasing at 49 days, which was also corroborated by the regression analysis. Villus density in the duodenum was greater in the first week, decreased with age and increased again at 42 days, probably due to the proximity with egg production onset. The expression of Sglt1 and Glut2 mRNA in the jejunum varied with age. In conclusion, the intestinal mucosa of female Japanese quail developed morphologically until 42days and functionally until earlier ages, indicating an adaptation to the exogenous diet during the first weeks of life.

The capacity of the chicken intestinal tract to digest and absorb nutrients is not fully developed on the day of hatch. The aim of this study was to investigate the role of betaine on the intestinal development of broiler chickens, including brush border enzyme activities indices and morphological changes during the first week of growth. Fiftyone-day-old Ross 308 broiler chickens were randomly distributed into five groups: control (C), low methionine(LM), low methionine plus betaine (LM+B), betaine 1 (B1) and betaine 2 (B2). Betaine was dissolved with a concentration of 500 mg/L in the drinking water of LM+B and B1groups and a concentration of 1000 mg/L in the B2 group. Histological assessment showed lower duodenal crypt depth and villi height/crypt depth ratio in B1 as compared to C. Jejunal villi height of B2birds was significantly higher than that of C birds, whereas jejunal crypt depth values of both B2 and LM birds were higher relative to LM+B and C birds. Betaine-fed groups had significantly a thicker ileal muscularis mucosa layer than C and LM groups. Duodenal and jejunal g-glutamyl transferase activity was significantly higher in B2birds in LM birds. Alkaline phosphatase activity in all three segments of the small intestine was significantly higher in the LM birds than in other groups. Jejunal alanine aminopeptidase activity in LM birds was significantly higher than in other groups, particularly in B1 and B2. The results suggest that betaine may be a promising agent to promote the intestinal development of broilers fed low-methionine or standard diets.

Isa Brown® layers with 48 weeks of age were distributed in a completely randomized experimental design with a 3 x 2 + 1 factorial arrangement, resulting in seven treatments with seven replicates of eight birds each. At the end of the fourth experimental period (28 days each), birds were 64 weeks of age and were randomly chosen (two birds per replicate, totaling 14 birds per treatment), weighed and sacrificed by neck dislocation. Their intestine was dissected and the duodenum, jejunum and ileum were collected for subsequent analysis of intestinal morphometry. Treatments consisted of diets containing three different fiber sources (cottonseed hulls, soybean hulls or rice husks) and two crude protein levels (12% or 16%). Soybean hulls and 16% crude protein level promoted, in general, an increase in villus height and crypt depth in the three intestinal segments. In the duodenum, the control diet resulted in higher villus height and crypt depth relative to the diets containing fiber. In the jejunum, higher crypt depth values. In the ileum, dietary fiber increased villus height as compared to the control diet.
