Studies have shown that gut dysbiosis is associated with the steatotic liver disease associated with metabolic dysfunction (MALSD) and its severity. This study evaluated the effects of two commercially available prebiotics fructooligosaccharides (FOS) and galactooligosaccharides(GOS) on hepatic adipogenesis, inflammation, and gut microbiota in high-fat diet-induced MALSD. The results indicated that FOS and GOS effectively reduced insulin resistance, hyperglycaemia, triglyceridemia, cholesterolaemia, and IL-1ß serum levels. Moreover, FOS and GOS modulated the lipogenic (SREBP-1c, ACC, and FAS) and lipolytic (ATGL) signalling pathways, and reduced inflammatory markers such as p-NFκB-65, IL-6, iNOS, COX-2, TNF-α, IL-1ß, and nitrotyrosine. FOS and GOS also enhanced the abundance of acetate producers' bacteria Bacteroides acidifaciens and Bacteroides dorei. FOS and GOS also induced positive POMC/GPR43 neurons at the arcuate nucleus, indicating hypothalamic signalling modulation. Our results suggest that FOS and GOS attenuated MALSD by reducing the hepatic lipogenic pathways and intestinal permeability through the gut microbiota-brain axis.

The gut microbiome plays a critical role to all animals and humans health. Methods based on ex vivo cultures are time and cost-effective solutions for rapid evaluation of probiotic effects on microbiomes. In this study, we assessed whether the protein secretome from the potential probiotic Enterococcus durans LAB18S grown on fructoligosaccharides (FOS) and galactoligosaccharides (GOS) had specific effects on ex vivo cultured intestinal microbiome obtained from a healthy individual. Metaproteomics was used to evaluate changes in microbial communities of the human intestinal microbiome. Hierarchical clustering analysis revealed 654 differentially abundant proteins from the metaproteome samples, showing that gut microbial protein expression varied on the presence of different E. durans secretomes. Increased amount of Bacteroidetes phylum was observed in treatments with secretomes from E. durans cultures on FOS, GOS and albumin, resulting in a decrease of the Firmicutes to Bacteroidetes (F/B) ratio. The most functionally abundant bacterial taxa were Roseburia, Bacteroides, Alistipes and Faecalibacterium. The results suggest that the secretome of E. durans may have favorable effects on the intestinal microbial composition, stimulating growth and different protein expression of beneficial bacteria. These findings suggest that proteins secreted by E. durans growing on FOS and GOS have different effects on the modulation of gut microbiota functional activities during cultivation.

Introducción: Los GOS son prebióticos naturales presentes en la leche materna que pue-den obtenerse enzimáticamente a partir de la lactosa de leche de vaca durante la fabricación de yogur. El producto lácteo resultante será reducido en lactosa y contendrá prebióticos y bacterias potencialmente probióticas. Sin embargo, mantendrá la baja relación Ca/Pi que aporta la leche de vaca, lo que podría alterar el remodelamiento óseo y la mineralización. Objetivo: comparar si un yogur reducido en lactosa que contiene GOS (YE) ofrece ventajas adicionales respecto de un yogur regular sin GOS (YR) sobre las absorciones (Abs) de Ca y Pi, retención y calidad ósea durante el crecimiento normal. Al destete, ratas machos fueron divididas en 3 grupos alimentados con AIN Ì93-G (C), YE o YR durante 28 días. Resultados: YE mostró el mayor aumento de lactobacilos fecales; producción de ácidos grasos de cadena corta especialmente p, profundidad de las criptas colónicas y menor pH cecal. El %AbsCa y %AbsPi aumentó en el siguiente órden: YE> YR> C (p < 0,05). El contenido de Ca y Pi en fémur, la densidad y contenido mineral óseos y los parámetros biomecánicos fueron similares en YE y C, mientras que YR mostró valores significativa-mente menores (p < 0,05). Conclusiones: YE aumentó las Abs y biodisponibilidad de minerales, alcanzando la retención y calidad ósea de C. El aumento en las Abs observado en YR no logró obtener la retención y calidad ósea de C. Conclusión: YE habría contrarrestado el efecto negativo del mayor aporte de Pi de la leche de vaca y sería una buena estrategia para lograr el pico de masa ósea y calidad del hueso adecuados, especialmente en individuos intolerantes a la lactosa. Breast milk contains an optimal calcium/phosphate (Ca/Pi) ratio and GOS. These natural prebiotics can be enzymatically produced via cow's milk lactose inyogurt manufacture. This milk product is low in lactose and contains prebiotics and potentially probiotic bacteria but maintains a low Ca/Pi ratio that could alter bone remodeling and mineralization. We evaluated if a lactose-reduced yogurt containing GOS (YE) offers additional advantages over regular yogurt without GOS (YR) on Ca and Pi absorption (Abs), bone retention and quality during normal growth. Weaning male rats were divided into 3 groups fed AIN'93-G (C), YE or YR for 28 days. Results: YE showed the highest increase in fecal lactobacilli; short-chain fatty acids production, especially propionate and butyrate; intestine crypt depth, and the lowest cecal pH. AbsCa% and AbsPi% increased in this order: YE> YR> C (p <0.05). Ca and Pi content in femur, bone density and mineral content, and biomechanical parameters were similar in YE and C, while YR showed the significantly lowest value (p < 0.05). Conclusions: YE increased mineral Abs reaching the retention and bone quality of C. Although YR increased Abs, bone retention and quality did not achieve C values. Seemingly, YE compensated for the negative effect of the higher Pi supply and would be a good strategy to achieve adequate peak bone mass and bone quality, especially in lactose intolerant individuals. Despite the nutritional properties of alfalfa, its production is mainly for animal feed and it is undervalued as a food source. In this study, the valorization of alfalfa as a potential source of bioactive carbohydrates [inositols, α-galactooligosaccharides (α-GOS)] is presented. A Box-Behnken experimental design was used to optimize the extraction of these carbohydrates from leaves, stems, and seeds of alfalfa by solid-liquid extraction (SLE) and microwave-assisted extraction (MAE). Optimal extraction temperatures were similar for both treatments (40 °C leaves, 80 °C seeds); however, SLE required longer times (32.5 and 60 min vs. 5 min). In general, under similar extraction conditions, MAE provided higher yields of inositols (up to twice) and α-GOS (up to 7 times); hence, MAE was selected for their extraction from 13 alfalfa samples. Pinitol was the most abundant inositol of leaves and stems (24.2-31.0 mg·g-1 and 15.5-22.5 mg·g-1, respectively) while seed extracts were rich in α-GOS, mainly in stachyose (48.8-84.7 mg·g-1). In addition, inositols and α-GOS concentrations of lyophilized MAE extracts were stable for up to 26 days at 50 °C. These findings demonstrate that alfalfa is a valuable source of bioactive carbohydrates and MAE a promising alternative technique to obtain functional extracts.

Simultaneous synthesis and purification (SSP) of galacto-oligosaccharides (GOS) from lactose was conducted using a combi-biocatalyst formed by crosslinked enzyme aggregates of Aspergillus oryzae ß-galactosidase and Saccharomyces cerevisiae cells co-immobilized by entrapment in calcium alginate gel particles. Product yield obtained with the combi-biocatalyst was similar than obtained with the soluble enzyme (23.3%), having a final purity of 25.7%. During the simultaneous process, ethyl-ß-galactoside was produced from the ethanol generated as a metabolic product of yeast cells, but ethyl-ß-galactoside was considerably decreased at high aeration (4 vvm). The combi-biocatalyst can be recovered and reused but its performance is limited by the reduction of the metabolic capacity of the cells. In this way, a process was developed for the SSP of GOS from lactose, obtaining a comparable product yield and higher specific productivity than in a conventional synthesis.

The Gaucher Outcome Survey (GOS) is an international disease-specific registry established in 2010 for patients with a confirmed diagnosis of Gaucher disease (GD), regardless of GD type or treatment status. Historically, there has been a limited understanding of type 3 GD (GD3) and its natural history in patients irrespective of their treatment status. Here, we describe the disease characteristics of patients with GD3 enrolled in GOS. As of October 2015, 1002 patients had been enrolled, 26 of whom were reported as GD3. The majority of patients with GD3 were from the US (13; 50.0%), seven (26.9%) were from the UK, three (11.5%) from Israel, and three (11.5%) from Brazil. No patients were of Ashkenazi Jewish origin. Median age of symptom onset was 1.4 (interquartile range: 0.5-2.0) years. The most common GBA1 mutation genotype was L444P/L444P, occurring in 16 (69.6%) of 23 patients who had genotyping information available. Nine patients reported a family history of GD (any type). Of 21 patients with treatment status information, 20 (95.2%) had received GD-specific treatment at any time, primarily imiglucerase (14 patients) and/or velaglucerase alfa (13 patients). Hemoglobin concentrations and platelet counts at GOS entry were within normal ranges for most patients, and there were no reports of severe hepatomegaly or of splenomegaly in non-splenectomized patients, most likely indicative of the effects of treatment received prior to GOS entry. This analysis provides information on the characteristics of patients with GD3 that could be used as the baseline for longitudinal follow-up of these patients.

Probiotics and prebiotics, mainly commercialised as food ingredients and also as supplements, are considered highly profitable niche markets. However, in recent years, the food industry has suffered from a series of health claim restrictions on probiotics and prebiotics in many parts of the world, including those made by the European Food Safety Authority. Therefore, we reviewed the core benefits of probiotic and prebiotic consumption on health. A number of studies have examined the prevention and/or management of intestinal infections, respiratory tract infections, CVD, osteoporosis, urogenital infections, cavities, periodontal disease and halitosis, allergic reactions, inflammatory bowel disease and irritable bowel syndrome and Helicobacter pylori gastric infections. In fact, a deeper understanding of the mechanisms involved in human microbiota and immune system modulation by probiotics and prebiotics relies on continuous efforts to establish suitable biomarkers of health and diseases risk factors for the design of clinical trials required for health claim approval. In spite of the promising results, the performance of large, long-term, well-planned, well-aligned clinical studies is crucial to provide more reliability and a more solid basis for the outcomes achieved and to support the potential use of probiotics and prebiotics in clinical practice.

Fed-batch synthesis of galacto-oligosaccharides (GOS) from lactose with ß-galactosidase from Aspergillus oryzae was evaluated experimentally and reaction yield was maximized via optimal control technique. The optimal lactose and enzyme feed flow rate profiles were determined using a model for GOS synthesis previously reported by the authors. Experimentally it was found that fed-batch synthesis allowed an increase on the maximum total GOS concentration from 115 (batch synthesis) to 218 g L(-1) as consequence of the increase in total sugars concentration from 40 to 58% w/w. Such high concentration of total sugars was not attainable in batch operation because of the low solubility of lactose at the reaction temperature (40°C). Simulations predicted a GOS yield of 32.5 g g(-1) in fed-batch synthesis under optimal conditions, while experimentally the same yield as in batch synthesis was obtained (28 g g(-1) ). Besides, an enrichment of total oligosaccharides in GOS with a high polymerization degree (GOS-5 and GOS-6) was observed in the fed-batch synthesis. Experimental profiles for all sugars were similar to the ones predicted by simulation, which supports the use of this methodology for the optimization of GOS synthesis.

Environmental differences occur between lentic and lotic communities' establishment, especially because of water time residence. Here we used a combination of field measurements and statistical analysis to evaluate the influent factors on the aquatic macroinvertebrates' composition and structure in 15 anthropogenic lakes from Alto Iguaçu's region. Macroinvertebrates were sampled from the marginal vegetation (with or without macrophytes) and sediment. Chemical parameters ranged among the anthropogenic lakes, mainly those which are connected to the Iguaçu River, beyond the presence or absence of macrophytes, interfering on the community's structure. Lake's morphometric data were measured in each lake and its relation to community was tested. The community structure was not related to the margin development index (Ds), macrophytes and altitude. Higher equitability was associated to the macrophytes presence in more than 30% of water surface and also where the riparian vegetation was more complex. Chironomids and oligochaetes densities were associated to lakes with the water surface completely covered by macrophytes and complex riparian vegetation. We observed a close association between productivity (phosphorous and nitrogen) and the community equitability (R = 0.3; p < 0.05) and Oligochaeta dominance (R = 0.32; p < 0.05), and not to Ostracoda's (R = 0.33; p > 0.05) and Chironomidae's (R = 0.34; p > 0.05) dominances, although we had not weighty difference among analyzed biological indexes. The obtained values from the BMWP' and EPT indexes resulted in acceptable, doubtful or critical water quality, what corroborates with the macroinvertebrate's structure of the composition and with the environmental variables observed in field.
Diferenças ambientais ocorrem entre o estabelecimento de comunidades de ambientes lênticos e lóticos, especialmente por causa do tempo de residência da água. Foi usada uma combinação de métricas de campo e análise estatística para avaliar os fatores influentes sobre a composição e estrutura de macroinvertebrados aquáticos em 15 lagos antropogênicos da região do Alto Iguaçu. Os macroinvertebrados foram amostrados da vegetação marginal (com ou sem macrófitas) e sedimento. Parâmetros químicos variaram entre os lagos antrópicos, principalmente aqueles conectados ao Rio Iguaçu, além da presença ou ausência das macrófitas, interferindo na estrutura da comunidade. Dados de morfometria de lagos foram mensurados e sua relação com a comunidade foi testada. A estrutura da comunidade não foi relacionada ao índice de desenvolvimento da margem (Ds), macrófitas e altitude. Maior equitabilidade foi associada à presença de macrófitas em mais de 30% da superfície da água e também onde a vegetação ripária foi mais complexa. Densidades de quironomídeos e oligoquetas foram associadas a lagos com a superfície completamente coberta por macrófitas e complexa vegetação ripária. Foi observada associação entre a produtividade (fósforo e nitrogênio) e a equitabilidade da comunidade (R = 0.3; p < 0.05) e dominância de Oligochaeta (R = 0.32; p < 0.05) e não para a dominância de Ostracoda (R = 0.33; p > 0,05) e Chironomidae (R = 0.34; p > 0.05), apesar de não haver diferença entre os índices biológicos analisados. Os valores obtidos através do Índice BMWP' e EPT/C resultaram em ambientes de qualidade aceitável, duvidosa ou crítica, o que corrobora com a estrutura da composição de invertebrados e com as características ambientais observadas em campo.

Poultry processing facilities are known for using a great amount of water, which is mainly used on carcasses chilling stage. In Brazil, meat regulations state that each chiller tank must be emptied, cleaned and sanitized every 8 working hours. The aim of the current study was to assess the microbial load of chiller water used in poultry immersion chilling system after 8, 16 and 24 working hours in order to evaluate the reduction of water changes and chiller sanitization. Conventional physicochemical and microbiological assays were done in water supply samples (n=69) to suppress interferences caused by freshwater addition; pre chilled (n=345) post chilled carcasses (n=345) and chiller water samples of the last stage (n=69). The results showed no significant differences on microbial load samples between the three shifts suggesting that the proposed reduction may be secure and reduces the volume of wastewater that would impact the environment, besides improving the rational use of processing time.

Parte desse volume utilizado se faz necessário, em cumprimento à legislação que determina que cada tanque do sistema de pré-resfriadores contínuos por imersão deve ser completamente esvaziado, limpo e desinfetado no final de cada período de trabalho (oito horas). O objetivo deste estudo foi comparar a carga microbiana das águas do sistema de resfriamento e das carcaças de frango ao final de oito, dezesseis e vinte e quatro horas de trabalho do abatedouro, para possível redução do número de vezes do completo esvaziamento dos tanques do sistema de resfriamento. Foram avaliadas, por meio de métodos convencionais microbiológicos e físico-químicos, amostras da água de abastecimento (n=69), visando a evitar possível interferência nas contagens das águas do sistema de resfriamento, amostras de carcaças de frango antes (n=345) e após (n=345) sua passagem pelo sistema de resfriamento e amostras de águas do último estágio do sistema de resfriamento de carcaças (n=69). Os resultados obtidos demonstraram que não houve diferença significativa na carga microbiana das amostras entre as três jornadas de trabalho do estabelecimento, sugerindo que a redução é segura, diminuindo assim o volume de águas residuais e seu impacto no meio ambiente, bem como melhorando o uso racional do tempo de processamento.

Introducción: La principal causa de muerte en Colombia son las muertes violentas y de éstas entre 49 por ciento y 70 por ciento corresponden a trauma craneoencefalico. Existen publicaciones en Colombia que indican la epidemiología de esta catastrofe nacional, pero no realizan un seguimiento para conocer el estado neurológico-funcional posterior de estos pacientes. Objetivos: Conocer el estado funcional al a±o en pacientes con trauma craneoencefalico. Métodos: Se realizó seguimiento a una cohorte de personas que ingresaron al Hospital Universitario del Valle, Cali, Colombia, con trauma craneoencefalico entre julio de 2003 y junio de 2004. Ademas de recoger información sociodemogrßfica y clínica, se aplicó la escala Glasgow Outcome Score (GOS) al egreso, al mes y al año de sufrido el trauma. Resultados: Se incluyeron 2,049 pacientes de los cuales 83 por ciento eran hombres. Por escala de Glasgow 53 por ciento con trauma leve, 31 por ciento moderado y 16 por ciento severo. La mortalidad hospitalaria fue de 13 por ciento (10 por ciento en urgencias y según la severidad, 0.3 por ciento, 1.4 por ciento y 8 por ciento, respectivamente). Al año del trauma la mortalidad fue 14 por ciento y el 85 por ciento se encontraba entre un GOS de 4 a 5. Conclusiones: La incidencia de las variables evaluadas en los pacientes con traumatismo craneoencefalico en el presente estudio, son muy semejantes a las series descritas en la literatura mundial. En cuanto al seguimiento a 12 meses el grado de funcionalidad y mortalidad son también semejantes a la de países con alta tecnología y desarrollo.
Introduction: The main cause of death in Colombia is the violence, in which 49 percent to 70 percent correspond to traumatic brain injury (TBI). There are publications in Colombia that expose the epidemiology of this national catastrophe, but there are a few studies that follow the neurological-functional state after the head injury on this patient. Objectives: To know the functional state after one year following a traumatic brain injury on patients. Methods: A cohort of patients that were hospitalized on the Hospital Universitario del Valle, Cali, Colombia, with traumatic brain injury between July 2003 and June of 2004. The Glasgow Outcome Score (GOS) scale was apply when the patient leave the hospital, and at the first and twelve month after the brain injury. Results: 2049 patients were include on the study. 83 percent were men. 53 percent of them were classified as mild TBI, 31 percent moderate and 16 percent severe by the Glasgow Score Scale. The mortality was 13 percent intrahospital (0.3 percent, 1.4 percent y 8 percent of mortality en mild, moderate and severe respectly), and after a year of TBI the mortality was 14 percent, and 85 percent of the patients was on GOS of 4 and 5. Conclusions: The incidences of the variables evaluated on the TBI patients on the present study are similar to the world literature series. After 12 months, the followed up of functional state and the mortality of TBI patients were similar to the data of countries of high technology and developed.
