RESUMO

Previous reports described the antidepressant-like action of the aqueous extract of pomegranate (Punica granatum: AEPG). Thus we evaluated the effect of AEPG and the main compounds found in the extract, punicalagin (PNCG) and ellagic acid (EA), on forced swimming test and the redox environment (reactive oxygen species [ROS] production, lipoperoxidation [LPX], and cellular function) in the brain of rats treated with 3 weeks post ovariectomy exposed ex vivo to pro-oxidants. Also, we selected PNCG and EA to study their antidepressant-like effects (0.001, 0.01, 0.1, 1.0, and 10 mg/kg) in the forced swimming test and their scavenging capacities in chemical combinatorial assays (expressed as IC50 values). We observed a 2-fold increase in the formation of ROS and LPX in the brain after exposure to FeSO4. However, these effects were significantly attenuated when rats were treated with AEPG, PNCG, and EA (1 mg/kg and 0.010 mg/kg for 14 days). AEPG and EA significantly increased the cellular function values of brains that had been affected by the effect of FeSO4 and with ONOO-. PNCG and EA significantly reduced immobility behavior at the lower doses used in this study. The capacity of scavenging compounds to eliminate radicals was for hydroxyl radical (⋅OH), superoxide anion (O2⋅⁣-), and peroxynitrite (ONOO-) as follows: AEPG > punicalagin > ellagic acid. In conclusion, the AEPG and their active compounds PNCG and EA promote antidepressant-like actions and antioxidant activity as they attenuate oxidative damage and prevent cellular dysfunction in ovariectomized rat brains.

The aim of this review is to provide overall information on Argentine propolis and to shed light on its potential, especially the one from the Monte region so as to support future research in the field. Around 1999, the Argentine propolis began to be chemically and functionally characterized to give it greater added value. Because Argentina has a wide plant biodiversity, it is expected that its propolis will have various botanical origins, and consequently, a different chemical composition. To date, five types have been defined. Based on their functionality, several products have been developed for use in human and veterinary medicine and in animal and human food. Because the Argentine propolis with the greatest potential is that of the Monte eco-region, this review will describe the findings of the last 20 years on this propolis, its botanical source (Zuccagnia punctata Cav.), its chemical composition, and a description of markers of chemical quality (chalcones) and functionality. Propolis can regulate the activity of various pro-inflammatory enzymes and carbohydrate and lipid metabolism enzymes, as well as remove reactive oxygen and nitrogen species. Consequently, it can modulate metabolic syndrome and could be used as a functional ingredient in food. Furthermore, hydroalcoholic extracts can act against human and animal pathogenic bacteria and human yeast, and mycelial pathogenic fungi. The ability to stop the growth of post-harvest pathogenic bacteria and fungi was also demonstrated. For this reason, Argentine propolis are natural products capable of protecting crops and increasing the lifespan of harvested fruit and vegetables. Several reports indicate the potential of Argentine propolis to be used in innovative products to improve health, food preservation, and packaging. However, there is still much to learn about these natural products to make a wholesome use of them.

Melon (Cucumis melo L.) has high economic value and in recent years, its production has increased; however, part of the fruit is wasted. Usually, inedible parts such as peel and seeds are discarded during processing and consumption. Extracts of melon residues were prepared and their phenolic compounds, antioxidants and antiproliferative activities were evaluated. Total phenolic compounds were found in hydroethanolic, hydromethanolic, and aqueous extracts, especially for melon peel (1.016 mg gallic acid equivalent/100 g). Flavonoids total content found for melon peel aqueous extract was 262 µg of catechin equivalent (CA)/100 g. In all extracts of melon peel significant amounts of gallic acid, catechin, and eugenol were found. For total antioxidant capacity, reported as ascorbic acid equivalent, the hydroethanolic and hydromethanolic extracts in peels and hydromethanolic in seeds were 89, 74, and 83 mg/g, respectively. Different extracts of melon showed iron and copper ions chelating activity at different concentrations, especially melon peel aqueous extract, reaching values of 61% for iron and 84% for copper. The hydroethanolic extract of melon peel presented a significant ability for hydroxyl radicals scavenging (68%). To assess the antiproliferative potential in human cancer cell lines, such as kidney carcinoma, colorectal carcinoma, cervical adenocarcinoma and cervical carcinoma, MTT assay was performed. The proliferation was inhibited by 20-85% at extracts concentrations of 0.1-1.0 mg/mL in all cancer cell lines. The results suggest that melon residues extracts display a high antioxidant activity in in vitro assays and have effective biological activity against the growth of human tumor cells.

Tithonia diversifolia, Asteraceae, a promising alternative for insect control such as leaf-cutting ants, have revealed repellent and insecticidal activity against Atta cephalotes. The different biological activities of T. diversifolia, are due to the existence of sesquiterpene lactones and phenolic compounds. Some phenolic compounds have antioxidant activity and curiously also exhibited tyrosinase inhibitory activity, key enzyme in insect metamorphosis. To expand the plant phytochemical knowledge, especially for isolation and identification of secondary metabolites such as phenolic compounds not yet discovered in the polar extracts, a new caffeic acid derivative was isolated from the leaves of Tithonia diversifolia. The structure was determined as (E)-3-(((3-(3,4-dihydroxyphenyl)acryloyl)oxy)methyl)-2-methyloxyrane-2-carboxylic acid (1). Additionally, three known phenolic acids, 4,5-dicaffeoylquinic acid (2), 3,4-dicaffeoylquinic acid (3) and 3,5-dicaffeoylquinic acid (4), were also isolated; 1, 2 and 3 are the first reports for this species. The structures of compounds 1, 2 and 3 were established based on LC-MS and one- and two-dimensional (1D)- and (2D)-NMR spectroscopic analyses. Spectroscopic data of compound 1 were compared with 4-O-caffeoyl-2-C-methyl-d-threonic acid (5). Antioxidant activity evaluation in 96-well plate format, showed caffeic acid derivatives as strong 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavengers and moderate ferric reducing antioxidant power (FRAP).

ABSTRACTThe chemical component and biological activity of propolis depend on flora area of bee collection and bee species. In the study, the propolis from three stingless bee species, Lepidotrigona ventralis Smith, Lepidotrigona terminata Smith, and Tetragonula pagdeni Schwarz, was collected in the same region of mangosteen garden from Thailand. Total phenolic content, alpha glucosidase inhibitory effect, and free-radical scavenging activity using FRAP, ABTS, DPPH assays were determined. The most potent activity of propolis extract was investigated for bioactive compounds and their quantity. The ethanol extract of T. pagdeni propolis had the highest total phenolic content 12.83 ± 0.72 g of gallic acid equivalents in 100 g of the extract, and the strongest alpha glucosidase inhibitory effect with the IC50 of 70.79 ± 6.44 µg/ml. The free-radical scavenging activity evaluated by FRAP, ABTS, DPPH assays showed the FRAP value of 279.70 ± 20.55 µmol FeSO4 equivalent/g extract and the IC50 of 59.52 ± 10.76 and 122.71 ± 11.76 µg/ml, respectively. Gamma- and alpha-mangostin from T. pagdeni propolis extract were isolated and determined for the biological activity. Gamma-mangostin exhibited the strongest activity for both alpha glucosidase inhibitory effect and free-radical scavenging activity. Using HPLC quantitative analysis method, the content of gamma- and alpha-mangostin in the extract was found to be 0.94 ± 0.01 and 2.77 ± 0.08% (w/w), respectively. These findings suggested that T. pagdeni propolis may be used as a more suitable raw material for nutraceutical and pharmaceutical products and these mangostin derivatives as markers.

The species of the Byrsonima genus (Malpighiaceae) have been used in folk medicine to treat gastrointestinal disorders, asthma and skin infections. In this study, the total phenolic content, antioxidant activity and phytochemical screening were carried out in polar extracts of Byrsonima coccolobifolia, Byrsonima verbascifolia and Byrsonima intermedia. The presence of tannins and flavonoids in the samples and high phenols content was observed. B. coccolobifolia and B. intermedia species showed the best results of antioxidant action. It was analysed the correlation between the methods, and there was a great correlation between phenolic content and antioxidant assay, showing that the phenolic substances contribute to the antioxidant activities of these plants.

The total phenol and flavonoid content, in addition to the antioxidant and cytotoxic activities, of extracts and fractions of Piptadenia moniliformis was determined. This honey plant species is commonly known as "catanduba" or "angico de bezerro". The aqueous fraction derived from the peels of the fruits exhibited the highest antioxidant activity, remaining comparable to the standard value, and there was a general correlation between this activity and the phenol and flavonoid content. The antioxidant potential of this species provides a basis for future developments in herbal medicines and cosmetics. Only the hydro alcoholic extract, the dichloromethane fractions and the ethyl acetate fractions showed moderate cytotoxicity.


Foram determinados o teor de fenóis e flavonoides, as atividades antioxidante e citotóxica dos extratos e frações de Piptadenia moniliformis. Essa é uma espécie melitófila comumente conhecida como "catanduba" ou "angico de bezerro". Das frações testadas, a fração aquosa das cascas dos frutos apresentou a maior atividade antioxidante, com valor comparável ao padrão, e no geral houve correlação do teor de fenóis e flavonoides com essa atividade. Os resultados do potencial antioxidante para essa espécie fornecem subsídios para futuros trabalhos que visem o desenvolvimento de fitoterápicos e cosméticos. Quanto à atividade citotóxica apenas o extrato hidroalcoólico e as frações diclorometano e acetato de etila demonstraram moderada citotoxicidade.

Ethanol extract of the leaves of Paullinia elegans Cambess., Sapindaceae, and its hexane, chloroform, ethyl acetate, and hydroethanol fractions were evaluated for their antiedematogenic and free radical scavenging activities. The ethanol extract and the hexane fraction produced statistically significant inhibition (74.4 and 76.0 percent, respectively) of the ear edema induced by croton oil in mice, observed at doses of 5 mg/ear. The ethyl acetate and hydroethanol fractions showed significant radical scavenging effect in the DPPH assay, with IC50 of 36.7 and 30.1 µg/mL, respectively. Fractionation of the extracts through chromatographic methods afforded epifriedelanol, oleanolic acid 3-O-acetyl, a mixture of stigmasterol 3-β-O-glucopyranoside and sitosterol 3-β-O-glucopyranoside, kaempferol 3,7-O-α-dirhamnopyranoside, kaempeferol-3-O-α-rhamnopyranoside and 2-O-methyl-chiro-inositol. The compounds were identified on the basis of their NMR spectral data and comparison with those of literature.


O extrato etanólico das folhas de Paullinia elegans Cambess., Sapindaceae, e as frações n-hexano, clorofórmio, acetato de etila e hidroetanólica, obtidas de seu fracionamento, foram avaliadas quanto às suas atividades anti-edematogênica e sequestradora de radicais livres. O extrato etanólico e a fração hexano produziram inibição significativa (74,4 e 76,0 por cento, respectivamente) do edema da orelha induzido pelo óleo de cróton em ratos, em doses de 5 mg/orelha. As frações acetato de etila e hidroetanólica mostraram atividade sequestradora de radicais livres no ensaio de DPPH, com IC50 de 36,7 e de 30,1 µg/mL, respectivamente. O fracionamento dos extratos pelo uso de métodos cromatográficos resultou no isolamento do epifriedelanol, ácido 3-O-acetil oleanólico, mistura do stigmasterol 3-β-O-glucopiranosídeo e sitosterol 3-β-O-glucopiranosídeo, canferol, canferol 3,7-O-α-diramnopiranosídeo, canferol 3-O-α-ramnopiranosídeo e 2-O-metil-chiro-inositol. Os compostos foram identificados com base na comparação de seus dados espectroscópicos de RMN com os da literatura.

Ethanolic extracts from 23 different dried herbs and spices commercialized in Brazil were investigated for their free radical scavenging properties using the stable free radicals 2,2'-diphenyl-β-picrylhydrazyl (DPPH•) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS•+), and Trolox as reference (TEAC) for both radicals. The kinetic curves for both radicals showed to follow the first order kinetics model and the decay rate constant (k obs) was calculated. For all the samples, the two methods showed a good linear TEAC correlation, indicating that the average reactivity of the compounds present in the ethanolic extracts was similar. Sage and rosemary extracts showed the highest free radical scavenging capacities, while onion showed negligible activity and colorifico, one of the most consumed spices in Brazil, showed low ABTS•+ scavenging activity. Three distinct situations were found for the extracts concerning the DPPH• scavenging capacities: (1) extracts, like rosemary and laurel, that presented the same efficient concentrations (EC50 ) but differed in the TEAC values and velocities of action (k obs), (2) extracts, such as garlic and basil, that showed similar EC50 and TEAC values, but different k obs values and (3) extracts that reacted at the same velocities but completely differed in the free radical scavenging capacities, like black pepper, savory, nutmeg, rosemary and sage. Similar considerations could be done for the ABTS•+ results. For the first time the ABTS•+ scavenging activity for allspice, basil, cardamom, chives, colorifico, cumin, dill, laurel, marjoram, parsley and tarragon was reported.


Extratos etanólicos de 23 ervas e condimentos desidratados comercializados no Brasil foram analisados quanto as suas propriedades antioxidantes utilizando os radicais 2,2'-difenil-β-picrilhidrazil (DPPH•) e ácido 2,2'-azino-bis(3-etilbenzotiazolina-6-sulfônico) (ABTS•+), Trolox foi usado como referência para ambos radicais. As curvas cinéticas seguiram o modelo cinético de primeira ordem, e a partir destas foram calculadas as constantes de velocidade de (k obs). Os extratos de sálvia e de alecrim apresentaram as maiores capacidades redutoras de radicais livres, enquanto o extrato de cebola praticamente não apresentou atividade e o colorífico, um dos condimentos mais consumidos no Brasil, apresentou baixa atividade de aprisionamento de radicaisABTS•+. Três situações distintas foram observadas em relação à capacidade de aprisionar radicais DPPH•: (1) extratos, como os de alecrim e de louro, que apresentaram as mesmas concentrações eficientes (EC50), porém, valores de TEAC e velocidade de ação (k obs) distintos ; (2) extratos, como os de alho e de manjericão, que apresentaram valores similares de EC50 e TEAC, com valores distintos de k obs e (3) extratos com a mesma velocidade de ação que diferiram totalmente na capacidade de aprisionamento de radicais livres, como de pimenta preta, segurelha, noz moscada, alecrim e sálvia. Considerações semelhantes podem ser feitas para os resultados de ABTS•+. Osdois métodos apresentaram boa correlação de TEAC, indicando que a reatividade dos compostos presentes nos extratos etanólicos foi similar. A atividade redutora de ABTS•+ para pimenta da Jamaica, manjericão, cardamomo, cebolinha, colorífico, cominho, dill, louro, manjerona, salsinha e estragão foi reportada pela primeira vez.

Se determinaron las actividades antioxidantes de los extractos crudos de una mezcla de polen apícola y de cada uno de los seis polen constituyentes que formaban esa mezcla. Las determinaciones se hicieron por el método de sustancias reactivas al ácido tiobarbitúrico (TBARS) en preparaciones microsomales de hígado y por el método del bloqueo del radical libre 2,2-difenil-1-picrilhidracilo (DPPH*). Las actividades se compararon con la composición de flavonoles y ácidos fenólicos y con los contenidos de flavonoles en el polen. Todos los extractos mostraron actividades antioxidantes. Éstas fueron diferentes para cada especie y no estuvieron claramente asociadas al contenido de flavonoles en el polen. El polen de Amarathus hybridus mostró una alta capacidad inhibidora de la oxidación lipídica. El de la mezcla entera y el de Tagetes sp. fueron efectivos bloqueadores de radicales libres.


The antioxidant activities of total extracts of a mixture of honeybee-collected pollen and its six constituent pollens were determined by lipid peroxidation assay by the thiobarbituric acid reactive substances (TBARS) test on hepatic microsomal preparations and by free radical scavenging (2,2-diphenyl-1-picrylhydrazyl; DPPH*) method. Activities were compared to the flavonol and phenolic acid compositions and flavonol contents in pollen. All extracts showed antioxidant activities as radical scavenger substances and as inhibitors of lipid peroxidation. Antioxidant activities were different for each species and were not clearly associated to the flavonol content in pollen. Pollen of Amaranthus hybridus was a potent lipid oxidation inhibitor, and that of Tagetes sp. and the whole mixture were effective antiradical substances.


Determinaram-se as atividades antioxidantes dos extratos crus de uma mescla de pólen apícola e de cada um dos seis polens constituintes que formavam essa mescla. As determinações se fizeram pelo método de substâncias reativas ao ácido tiobarbitúrico (TBARS) em preparações microssomais de fígado e pelo método do bloqueio do radical livre 2,2-difenil-1-picrilhidracilo (DPPH*). As atividades se compararam com a composição de flavonóis e ácidos fenólicos e com os conteúdos de flavonóis no pólen. Todos os extratos mostraram atividades antioxidantes. Estas foram diferentes para cada espécie e não estiveram claramente associadas ao conteúdo de flavonóis no pólen. O pólen de Amarathus hybridus mostrou uma alta capacidade inibidora da oxidação lipídica. O da mescla inteira e o de Tagetes sp. foram efetivos bloqueadores de radicais livres.
