RESUMO

BACKGROUND: Genetically modified pigs are considered ideal models for studying human diseases and potential sources for xenotransplantation research. However, the somatic cell nuclear transfer (SCNT) technique utilized to generate these cloned pig models has low efficiency, and fetal development is limited due to placental abnormalities. RESULTS: In this study, we unprecedentedly established putative porcine trophoblast stem cells (TSCs) using SCNT and in vitro-fertilized (IVF) blastocysts through the activation of Wing-less/Integrated (Wnt) and epidermal growth factor (EGF) pathways, inhibition of transforming growth factor-ß (TGFß) and Rho-associated protein kinase (ROCK) pathways, and supplementation with ascorbic acid. We also compared the transcripts of putative TSCs originating from SCNT and IVF embryos and their differentiated lineages. A total of 19 porcine TSCs exhibiting typical characteristics were established from SCNT and IVF blastocysts (TSCsNT and TSCsIVF). Compared with the TSCsIVF, TSCsNT showed distinct expression patterns suggesting unique TSCsNT characteristics, including decreased mRNA expression of genes related to apposition, steroid hormone biosynthesis, angiopoiesis, and RNA stability. CONCLUSION: This study provides valuable information and a powerful model for studying the abnormal development and dysfunction of trophoblasts and placentas in cloned pigs.

Advances in Artificial Reproductive Technologies (ARTs) in bovine embryos to produce cloned pregnancies have been developed in the last years, however high pregnancy losses rates still present. Those rates are associated to placental morphology alterations that are majorly focused on extracellular matrix (ECM) alterations and consequently placentome hyperplasia, increased trophoblast cell migration and vascular defects. Herein, we aimed to search, at protein level, pathways altered by ART that can modify the placental development harmony. For this, we used 4-month-old control (n = 3), SDS-decellularized (n = 3) and cloned (n = 3) cotyledons for proteomic analysis. Samples were grouped by condition and were washed, lysed, urea-reduced, acetone-precipitated, DTT-educed, iodoacetamide-alkylated, trypsin digested, and C-18 column purified. At the end, 3 µg protein were loaded in Orbitrap Fusion Lumos spectrometer (ThermoScientific). Generated spectra were exported to MaxQuant software (v1.6.10.43) to produce the protein list of each sample, and the LFQ intensity were statistically analyzed by Inferno software (v.1.1.6970). After this, proteins related to ECM and cellular junction ontologies were filtered and manually annotated using DAVID Bioinformatics Resources 6.8. From 2577 identified protein sequences by MaxQuant software, 165 (7.1%) were filtered by selected ontologies. We found 10 proteins (B2M, COL6A6, FERMT3, LGALS3BP, NIBAN2, PDLIM5, PON1, PRP9, RASIP1 and SPARC) upregulated in clone, when compared to control condition. The ten pathways that enriched more proteins were: focal adhesion, ECM-receptor interaction, PI3K-Akt signaling pathway, protein digestion and absorption, amoebiasis, pathways in cancer, small cell lung cancer, platelet activation, regulation of actin cytoskeleton, and proteoglycans in cancer. Functionally, detected proteins, signaling pathways and ontologies are orchestrated to permit the binucleated trophoblastic cells migration and blood vessels modelling. In conclusion, the cloned condition presents the same mechanisms as control one, however overexpression of some specific ECM proteins could be responsible to exacerbate those mechanisms and can explain all morphophysiological alterations presented in cloned pregnancies associated to high pregnancies losses rates in this condition.

ABSTRACT: This study evaluated the viability of Nellore cloned calves derived from somatic cell nuclear transfer (SCNT) and compare their viability with animals of the same breed derived from in vitro fertilization (IVF). Thus, two groups were formed. Group I (GI) consisted of 10 calves derived from SCNT and group II (GII) consisted of 10 calves derived from IVF. The differences detected between the groups were in the physical examination of the respiratory tract in GI, which represented the most common clinical-pathological disturbances. The Apgar index score indicated that 80% of GI animals were depressed and all had pale mucous membranes. Thus, anemia was reported in GI. In GII, this started at 12 h of life and was probably caused by an iron deficiency. Moreover, total calcium and ionized calcium levels were higher in GI immediately after birth. These alterations probably resulted in a high incidence of mortality in GI, reaching 90% of the calves, whereas mortality was only 20% for the calves in GII. In conclusion, cloned calves, which were derived from SCNT, had physiological and metabolic alterations after delivery, leading to a higher mortality rate during the perinatal period.


RESUMO: O objetivo deste trabalho foi avaliar a viabilidade de bezerros da raça Nelore oriundos da técnica da transferência nuclear de células somáticas (TNCS), no período pós natal imediato, comparando-a com animais desta mesma raça, oriundos de fertilização in vitro (FIV). Para tanto, os animais foram alocados em dois grupos, a saber: Grupo I (GI) - 10 animais frutos de TNCS; e, Grupo II (GII) - 10 animais oriundos de FIV. Nos respectivos bezerros, todos obtidos por cesariana, foram realizadas as avaliações físicas, escore de APGAR, bem como coleta de amostras de sangue nos momentos 0 (ao nascimento), às 2, 4, 6 e 12 horas de vida, a fim de avaliar os resultados de eritrograma, análises bioquímicas e hormonais, comparando-os entre os grupos e momentos. Nos animais que vieram a óbito foi realizada a necropsia para investigar a causa mortis. As diferenças observadas foram em relação aos achados clínico-patológicos, envolvendo, principalmente, o sistema respiratório caracterizado por bradpneia associada à dispneia, e a presença de edema e atelectasia pulmonar observadas no GI. Ademais, após a colostragem notou-se que 80% dos animais avaliados não foram capazes de manter a glicemia sendo mais evidentes nos animais do GII, possivelmente devido à hiperinsulinemia que se manifestou neste grupo ao longo de todo o período experimental. A anemia após o nascimento foi evidente no grupo de bezerros clonados ao longo de todo período de avaliação ao contrário dos bezerros oriundos de GII, a qual foi observada somente às 12 horas de vida, sendo, possivelmente, de origem ferropriva. Casos de hipercalcemia foram denotados nos animais do GI ao nascimento, sendo possivelmente associados à asfixia perinatal. Estas alterações, em conjunto, levaram a uma taxa de mortalidade de 90% dos animais do GI e de 20% dos bezerros do GII. Conclui-se que os animais oriundos da TNCS apresentam alterações fisiológicas e metabólicas após o nascimento, responsáveis, em grande parte, pela maior taxa de óbitos dentro do período perinatal.

Licensure of a vaccine to protect against aerosolized Venezuelan equine encephalitis virus (VEEV) requires use of the U.S. Food and Drug Administration (FDA) Animal Rule to assess vaccine efficacy as human studies are not feasible or ethical. An approach to selecting VEEV challenge strains for use under the Animal Rule was developed, taking into account Department of Defense (DOD) vaccine requirements, FDA Animal Rule guidelines, strain availability, and lessons learned from the generation of filovirus challenge agents within the Filovirus Animal Nonclinical Group (FANG). Initial down-selection to VEEV IAB and IC epizootic varieties was based on the DOD objective for vaccine protection in a bioterrorism event. The subsequent down-selection of VEEV IAB and IC isolates was based on isolate availability, origin, virulence, culture and animal passage history, known disease progression in animal models, relevancy to human disease, and ability to generate sufficient challenge material. Methods for the propagation of viral stocks (use of uncloned (wild-type), plaque-cloned, versus cDNA-cloned virus) to minimize variability in the potency of the resulting challenge materials were also reviewed. The presented processes for VEEV strain selection and the propagation of viral stocks may serve as a template for animal model development product testing under the Animal Rule to other viral vaccine programs. This manuscript is based on the culmination of work presented at the "Alphavirus Workshop" organized and hosted by the Joint Vaccine Acquisition Program (JVAP) on 15 December 2014 at Fort Detrick, Maryland, USA.

A progesterona é um hormônio fundamental na manutenção da gestação nos mamíferos. Em bovinos clonados, perdas gestacionais significativas ocorrem devido à deficiência placentária. Tendo em vista esse problema, o objetivo do trabalho foi analisar em diferentes idades gestacionais a imunolocalização para receptores de progesterona em placentas de animais não clonados e clonados. Tecidos placentomais de bovinos não clonados foram obtidos da linha de abate no Matadouro Mantiqueira, enquanto os tecidos placentomais dos animais clonados com idades gestacionais de 68, 135, 225 e 270 dias de gestação foram obtidos em empresas especializadas em clonagem animal. Os tecidos foram processados e embebidos em paraplast para localização de receptores de progesterona, utilizando-se a técnica de imuno-histoquímica pelo anticorpo monoclonal (Progesterone receptor SER 294). Pela imuno-histoquímica, comparando-se a concentração de receptores de progesterona na placenta de animais clonados com as de animais não clonados, em diferentes idades gestacionais, observou-se que as células dos placentomas dos animais clonados apresentaram uma marcação mais intensa de receptor de progesterona nas idades de 135 e 270 dias de gestação em relação aos não clonados. Tal fato sugere que a presença de progesterona no final da gestação pode ser mais uma das possíveis causas de gestações prolongadas.


To progesterone is a fundamental hormone in the maintenance of the gestation in the mammals. In bovine they cloned, losses gestational significant, occur due to the placental deficiency. Having in mind that problem, the objective of the work was analyze in different gestational ages to immune stained for receivers of progesterone in placentas of normal animals and cloned. Placentomal tissues of bovine normal were obtained of the line of counts on in the Slaughterhouse Mantiqueira, while the placentomal tissues of the animals cloned with gestational ages of 68, 135, 225 and 270 days of gestation were obtained in companies specialized in animal cloning. The fabrics were prosecuted and absorbed in paraplast for progesterone receivers location, utilizing itself to technical of immunohistochemistry by the antibody monoclonal (Progesterone receiver BE 294). By the immunohistochemistry comparing the progesterone receivers concentration in the placenta of animals cloned, with the of animals done not manipulate, in different gestational ages we observe that the cells of the placentomal tissues of the animals manipulated (cloned) presented a more intense marking of receiver of progesterone in the ages of 135 and 270 days of gestation regarding the done not manipulate. Suggesting that the presence of progesterone in the end of the gestation can be more one of the possible cause of gestations prolonged.

Apesar dos esforços realizados nos últimos quatro anos, as taxas de mortalidade dos bezerros clonados da raça que chegam a termo são ainda altas, cerca de 50%. Demonstrou-se a ocorrência de graves distúrbios cardiorrespiratórios caracterizados por hiperfonese, presença de sopros cardíacos na 1ª e 2ª bulha associadosa dispneias, respiração rude e estertores. Em consequência ao nãofechamento do Forâmen de Botal e do Ducto Arterioso, há mistura de sangue arterial e venoso comprometendo a capacidade de oxigenação do sangue dos bezerros clonados. Observou-se ainda a ocorrência de macrossomia, hipoglicemia, hipotermia, anomaliasdas estruturas umbilicais, anemia e alopecia.


Despite all efforts during the last four years to improve cloned newborn care, the mortality rate of calf after term is still high, around 50%. Clinical symptoms observed in these cloned calf were related to severe cardiopulmonary disorders like hyperphonese, diastolic and systolic cardiac murmurs associated to dyspnea and crackling lung sounds. Due to the patency of the Foramen Ovale and Ductus Arteriosus, in which causes the mix of arterial and venous blood, the blood oxygenation in these cloned calf is compromised. In addition, cloned calf could also present increasedbirth weight, hypoglycemia, hypothermia, umbilical cord abnormalities, anemia, and alopecia.

Background: The advent of animal cloning from embryos reconstructed with nuclei from cells at different stages of differentiation has been responsible for the appearance of several anomalies in calves, with some alterations diagnosed during pregnancy and others after the birth of cloned animals. Despite efforts over the past five years, the rate of mortality after the birth of cloned calves is still high, at about 50%. Below is reported the research experience gained at the University of São Paulo in the period from 2005 to 2010, related to the birth of cloned calves from Nellore. Review: Research showed that cloned calves present cardiopulmonary disorders characterized by tachycardia with episodes of arrhythmia with bradycardia, increase in the 1st heart sound, presence or absence of heart murmurs in the 1st and 2nd heart sound associated with dyspnea, breathing harsh and crackling dry, moist and crackling. During the first days of life of cloned calves that died, ultrasound examination demonstrated the presence of concentric hypertrophy congenital cardiac and turbulent flow at the foramen of botal towards the right heart to left heart, indicating reversal of blood flow to the pattern observed in fetal life. As a result, mixture of arterial and venous blood occurs, compromising the ability of oxygen to the blood of cloned calves. The occurrence of macrosomia was observed in 20.9% of Nellore calves. It is believed that this syndrome is associated with disorders of carbohydrate metabolism in the placenta and/or fetus, leading to fetal gigantism, which mimicks excessive fetal growth seen in pregnant women with diabetes. We observed the occurrence of moderate to severe normocytic and normochromic anemia, with the anemia gradually appearing from 12 h of life onwards, reaching its maximum intensity at the end of the first week, to start a gradual recovery to normal values from the 15th day of life. The anemia observed in cloned calves was ferropriva, as such animals showed a significant decrease in serum iron levels associated with decreased transferrin saturation index (STI). Umbilical anomalies occurred in most clones. There was an increase in the thickness of the umbilical cord, which hindered its spontaneous rupture at birth. The umbilical arteries did not undergo retraction into the abdominal cavity, leaving them exposed in the remnant of the umbilical cord. In the first three days after birth, strong pulse of these arteries was noted in umbilical cord, making it necessary to use clamps on the arteries in order to prevent bleeding. There were also intra-abdominal hematomas involving the arteries and the urachus. Between 15 and 20 days of life, it was observed the occurrence of alopecia in about 75.0% of the calves, whose origin may be related to disturbances in the synthesis and absorption of vitamins, since the supplementation of calves with ADE vitamin complex decreased symptoms. Conclusion: The results confirm the occurrence of serious cardiopulmonary disorders characterized by tachycardia, hyperphonesis, presence of heart murmurs in the 1st and 2nd heart sounds, episodes of arrhythmia and bradycardia associated with dyspnea, rales, and coarse breath. As a result of the non-closure of the foramen of Botal and the Ductus Arteriosus, arterial and venous blood a mix, compromising the oxygen carrying capacity in the blood of cloned calves. There was also the occurrence of macrosomia, hypoglycemia, hypothermia, abnormal umbilical structures, anemia and alopecia

RESUMO

Apesar dos esforços realizados nos últimos quatro anos, as taxas de mortalidade dos bezerros clonados da raça que chegam a termo são ainda altas, cerca de 50%. Demonstrou-se a ocorrência de graves distúrbios cardiorrespiratórios caracterizados por hiperfonese, presença de sopros cardíacos na 1ª e 2ª bulha associadosa dispneias, respiração rude e estertores. Em consequência ao nãofechamento do Forâmen de Botal e do Ducto Arterioso, há mistura de sangue arterial e venoso comprometendo a capacidade de oxigenação do sangue dos bezerros clonados. Observou-se ainda a ocorrência de macrossomia, hipoglicemia, hipotermia, anomaliasdas estruturas umbilicais, anemia e alopecia.(AU)


Despite all efforts during the last four years to improve cloned newborn care, the mortality rate of calf after term is still high, around 50%. Clinical symptoms observed in these cloned calf were related to severe cardiopulmonary disorders like hyperphonese, diastolic and systolic cardiac murmurs associated to dyspnea and crackling lung sounds. Due to the patency of the Foramen Ovale and Ductus Arteriosus, in which causes the mix of arterial and venous blood, the blood oxygenation in these cloned calf is compromised. In addition, cloned calf could also present increasedbirth weight, hypoglycemia, hypothermia, umbilical cord abnormalities, anemia, and alopecia.(AU)

Os mecanismos de morte em animais clonados a partir de células somáticas são poucos elucidados. Malformações de órgãos, alterações de tamanho e peso desses animais foram anomalias já descritas, porém em casos isolados. Desse modo, estudos nos níveis anatomo e histopatológicos são de suma importância para ajudar a compreensão dos fatores responsáveis pelos altos índices de insucessos com a utilização da TNCS (transferência nuclear de células somáticas). Assim, o presente trabalho foi realizado com o objetivo de descrever, por meio de exames anatomopatológicos, as alterações presentes em um grupo de animais gerados por TNCS, que vieram a óbito entre 1 e 19 dias de vida. Esse experimento foi realizado entre os anos de 2004 e 2008 na fazenda Tambaú, cidade de Tambaú, Estado de São Paulo. No total foram gerados 21 animais da raça Nelore (Bos indicus), sendo que 11 vieram a óbito e dez apresentaram boa evolução clínica no período perinatal. Foram realizadas as necropsias dos animais e posterior análise histopatológica de tecidos alterados. Esse estudo mostrou alta prevalência de alterações cardiopulmonares, que provavelmente foram determinantes nos mecanismos de morte dos animais. Dentre essas alterações a hipertensão pulmonar e modificações hemodinâmicas diagnosticadas através do exame histopatológico foram as mais frequentes. (AU)


The death mechanisms in cloned animals are not well elucidated. There are few cases describing organ malformation, size and weight alterations in these animals. Pathologic studies are necessary to understand the high level of unsuccessful results using the NTSC (nuclear transfer somatic cell) technique. The present study was carried out to describe alterations present in a group of animals generated by NTSC technique that deceased between 0 to 19 days of life, using anatomopathologic examination. This experiment was conducted at Tambaú Farm in São Paulo State between years 2004 and 2008. Initially there were twenty one Nelore breed Bos indicus animals, but 11 of them deceased and ten survived and showed satisfactory clinical evolution in perinatal period. Animals necropsies and histopathologic analyses where performed. The results revealed a high prevalence of cardiopulmonary alterations, which were predominantly lung hypertension and hemodynamic profile changes, revealed by histopathologic analyses. (AU)

O comprimento das carúnculas foi de 1,55±0,57; 2,45±0,55; 4,66±2,00 e 5,72±1,90cm para os Grupos I, II, III e IV, respectivamente. Quanto à altura, as carúnculas apresentaram um crescimento linear durante a gestação e foram de 0,40±0,15; 0;57±0,21; 1,00±0,48 e 1,80±0,91cm, para os respectivos Grupos I, II, III e IV. A largura das carúnculas foi semelhante entre os Grupos I e II (0,97±0,30 e 1,42±0,71cm) e os Grupos III e IV (2,68±1,22 e 3,52±1,16cm). Quando o Grupo V foi comparado ao Grupo IV, as carúnculas do Grupo V apresentaram maior comprimento (5,72±1,90 vs. 7,88±2,13cm) e largura (3,52±1,16 vs. 4,93±1,46cm), porém foram semelhantes em altura (1,80±0,91 e 2,25±0,67cm). Verificou-se que em gestações de CC, as carúnculas apresentaram maior desenvolvimento que em gestações de CNC. As carúnculas fusionadas apresentaram medidas estatisticamente semelhantes às isoladas em todos os parâmetros e grupos. Sob microscopia de luz, observou-se a formação de um eixo estromal, da base da carúncula ao ápice da fissura fusional, de constituição histológica semelhante ao estroma endometrial. The objective of the study was to compare the characteristics of the caruncular fusion in gestations of non-cloned and cloned conceptuses. The non-cloned conceptuses were divided according to the gestation period: Group I (2 to 3 months; n=9), II (4 to 6; n=9); III (7 to 8; n=10) and IV (9 n=7). The cloned conceptuses formed the Group V: 9 months; n=4. The caruncles were observed macroscopically (number and dimensions: length, width and height), microscopically and submitted to statistical analysis (5 percent of significance). We observed three types of macroscopic caruncular fusions: oval (morphologically normal); two united adjacent caruncles and the lobulated type, characterized by regions with several united caruncles presenting a false fusion or deformation of the caruncular parenchyma. The length of the caruncles was 1.55±0.57; 2.45±0.55; 4.66±2.0 and 5.72±1.90cm for the groups I, II, III, IV respectively. As for the height, the caruncles presented a lineal growth during the gestation: 0.40±0.15; 0.57±0.21; 1.0±0.48 and 1.80±0.91cm, for the respective groups I, II, III and IV. The width of the caruncles was similar between the groups I and II (0.97±0.30 e 1.42±0.71cm) and the groups III and IV (2.68±1.22 and 3.52±1.16cm). When the group V was compared to the IV, the caruncles of the group V presented a larger length (5.72±1.90 vs. 7.88±.13cm) and width (3.52±1.16 vs. 4.93±1.46cm), however they were similar in height (1.80±0.91 and 2.25±0.67cm). We verified that in gestations of cloned conceptuses the caruncles presented a larger development than in gestations of non-cloned conceptuses.
