This study determined the effect of pelagic Sargassum on in vitro dry matter and organic matter degradation, total gas production (TGP), and protozoa population. The treatments were different levels of Sargassum inclusion on a basal substrate (Stargrass hay; Cynodon nlemfuensis) as follows: T0 (control treatment based on Stargrass hay), T10 (90% Stargrass hay + 10% Sargassum), T20 (80% Stargrass hay + 20% Sargassum), and T30 (70% Stargrass hay + 30% Sargassum). Ruminal fermentation kinetics and protozoa population were determined during 72 h of in vitro incubations. Compared to control, dry matter degradability at 48 and 72 h and organic matter degradability at 24 and 48 h were higher in Sargassum treatments. TGP was lower with T20 at 48 h. The total population of protozoa and the concentration of Entodinium spp. were lower at T20 at 48 h and T30 at 72 h. Cl, S, Ca, K, and Zn (103, 5.97, 88.73, 285.70 g/kg, and 15,900 mg/kg) were high in Sargassum, reaching twice or even nine times higher than the contents in Stargrass (11.37, 1.60, 43.53, 87.73 g/kg, and 866.67 mg/kg). Overall, up to 30% pelagic Sargassum could be included in hay-based substrates from tropical grasses without negative effects on in vitro dry matter and organic matter degradability.

The present study aimed to evaluate the potential of the laminarin-based formulation Vacciplant to protect and induce resistance in wheat against Zymoseptoria tritici, a major pathogen on this crop. Under greenhouse conditions, a single foliar spraying of the product 2 days before inoculation with Z. tritici reduced disease severity and pycnidium density by 42 and 45%, respectively. Vacciplant exhibited a direct antifungal activity on Z. tritici conidial germination both in vitro and in planta. Moreover, it reduced in planta substomatal colonization as well as pycnidium formation on treated leaves. Molecular investigations revealed that Vacciplant elicits but did not prime the expression of several wheat genes related to defense pathways, including phenylpropanoids (phenylalanine ammonia-lyase and chalcone synthase), octadecanoids (lipoxygenase and allene oxide synthase), and pathogenesis-related proteins (ß-1,3-endoglucanase and chitinase). By contrast, it did not modulate the expression of oxalate oxidase gene involved in the reactive oxygen species metabolism. Ultrahigh-performance liquid chromatography-mass spectrometry analysis indicated limited changes in leaf metabolome after product application in both noninoculated and inoculated conditions, suggesting a low metabolic cost associated with induction of plant resistance. This study provides evidence that the laminarin-based formulation confers protection to wheat against Z. tritici through direct antifungal activity and elicitation of plant defense-associated genes.

BACKGROUND Sargassum liebmannii is widely distributed throughout rocky, coastal upwelling areas in the tropical Mexican Pacific. This brown algae is of great environmental and industrial importance. However, no information is available that documents the genetic or phenotypic variability of the species, which is needed to determine how it may react to environmental variation related to climate change. In this study, S. liebmannii specimens were collected from the coast of Jalisco, Mexico, and molecular and morphological characterization was conducted. Intraspecific variability was estimated according to the study areas. RESULTS The inter-simple sequence repeat (ISSR) markers indicated a polymorphism percentage of 95%. The Shannon index and Nei index showed relatively low values among the populations (0.3569 and 0.081, respectively). On the other hand, the genetic differentiation coefficient indicated inter- and intrapopulation values of 36.69% and 63.31%, respectively. The Jaccard similarity coefficient was used to determine the degree of similarity among individuals by geographical area. The morphological characteristics and environmental variables that were used to correlate phenotypes and genotypes indicated that S. liebmannii showed low genetic flow because of the presence of geographical barriers due to substrate that was not optimal for algal development. CONCLUSIONS The ISSR markers were useful for detecting genetic differences among S. liebmannii individuals. The results indicate that a coupled genotypic-phenotypic study is beneficial for documenting the variation present in the little-studied algal species. These studies may be used in future research to clarify taxonomic controversies while generating additional genomic information

Solar radiation effects on the ecophysiology and biochemical responses of the brown macroalga Macrocystis pyrifera (L.) C. Agardh were evaluated using a mesocosm approach in Southern Chile. Treatments with different radiation attenuations were simulated with three vertical attenuation coefficients: (1) total (Kd = 0.8 m-1), (2) attenuated (Kd = 1.2 m-1), and (3) low (Kd = 1.6 m-1) radiation levels. Nutrient concentration and temperature did not show differences under the three light conditions. Photosynthetic activity was estimated by in vivo chlorophyll a (Chla) fluorescence under the three light treatments as an isolated physical factor in both in situ solar radiation in the field. This was achieved using a pulse amplitude-modulated (PAM) fluorometera-Diving PAM (in situ). Photosynthetic activity and biochemical composition were measured in winter during two daily cycles (1DC and 2DC) in different parts of the thalli of the plant: (1) canopy zone, (2) middle zone, and (3) down zone, associated with different depths in the mesocosm system. Nevertheless, the in situ electron transport rate (ETR in situ ) was higher in the exposed thalli of the canopy zone, independent of the light treatment conditions. The concentration of phenolic compounds (PC) increases in the down zone in the first daily cycle, and it was higher in the middle zone in the second daily cycle. The Chla increased in the morning time under total and attenuated radiation in the first daily cycle. Solar radiation increasing at midday prompted the photoinhibition of photosynthesis in the canopy zone but also an increase in productivity and phenol content. Therefore, light attenuation in the water column drove key differences in the photo-physiological responses of M. pyrifera, with the highest productivity occurring in thalli positioned in the canopy zone when exposed to solar irradiance.

The residue derived from the alginate extraction from S. filipendula was applied for the biosorption of aluminum from aqueous medium. The adsorptive capacity of the residue (RES) was completely evaluated in batch mode. The effect of pH, contact time, initial concentration, and temperature was assessed through kinetic, equilibrium, and thermodynamic studies. The biosorbent was characterized prior and post-Al biosorption by N2 physisorption, Hg porosimetry, He pycnometry, and thermogravimetry analyses. Equilibrium was achieved in 60 min. Kinetics obeys pseudo-second-order model at aluminum higher concentrations. Isotherms followed Freundlich model at low temperature (293.15 K) and D-R or Langmuir model at higher temperatures (303 and 313 K). Data modeling indicated the occurrence of both chemical and physical interactions in the aluminum adsorption mechanism using RES. The maximum adsorption capacity obtained was 1.431 mmol/g at 293 K. The biosorption showed a spontaneous, favorable, and exotherm character. A simplified batch design was performed, indicating that the residue is a viable biosorbent, achieving high percentages of removal using low biomass dosage.

Leishmaniasis is a group of diseases that have limited and high toxic therapeutic options. Herein, we evaluated the antileishmanial potential and cytotoxicity of hexanic extract obtained from the Antarctic brown alga Ascoseira mirabilis using bioguided fractionation against Leishmania amazonensis and murine macrophages, which was fractionated by SPE, yielding seven fractions (F1-F7). The fraction F6 showed good anti-amastigote activity (IC50 = 73.4 ± 0.4 µg mL-1) and low cytotoxicity (CC50 > 100 µg mL-1). Thus, in order to identify the bioactive constituent(s) of F6, the fraction was separated in a semipreparative HPLC, yielding four fractions (F6.1-F6.4). F6.2 was the most bioactive fraction (IC50 = 66.5 ± 4.5 µg mL-1) and GC-MS analyses revealed that the compounds octadecane, propanoic acid, 1-monomyristin and azelaic acid correspond to 61% of its composition. These data show for the first time the antileishmanial potential of the Antarctic alga A. mirabilis.

Macroalgae produce a large range of primary and secondary metabolites with ecological and economical importance. Studies on antioxidants from marine algae have increased notably, focusing on searching new sources of natural compounds for different applications, in which tropical species have been shown to have high potential, often improved by stressful environmental conditions during the tide cycle with periods of emersion and submersion. Therefore, in order to characterize the antioxidant activity and relate it to local environmental tide exposure, three species of brown marine algae, Canistrocarpus cervicornis, Dictyopteris delicatula, and Lobophora variegata from two beaches on the northeast coast of Brazil were studied. Dichloromethane:methanol (DCM:M) and aqueous extracts were tested for ferric reducing antioxidant power, 2,2-diphenyl-1-picrylhydrazyl, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) antioxidant assays and total phenolic compounds. Aqueous extracts of C. cervicornis showed up to 10 times major antioxidant activity and phenolic compounds than DCM:M extracts. Different characteristic of antioxidant activity were identified among the beaches, species, and extracts, in which aqueous extracts from C. cervicornis and L. variegata showed the most promising matrices for future prospection of natural antioxidants.

Undaria pinnatifida is a brown macroalga considered a high quality natural food because of its numerous health benefits. The aim of this paper is to provide seasonal information on the chemical content of blades and sporophylls of U. pinnatifida from San Jorge Gulf (SJG, Chubut, Argentina) in order to evaluate their different uses. Moreover, samples of algae deposited on the beach are also studied. A multi-elemental analysis is made by Total Reflection X-ray Fluorescence (TXRF). Sixteen elements are quantified: As, Br, Ca, Cr, Cu, Fe, K, Mn, Ni, P, Pb, Rb, S, Sr, V and Zn. The results reveal that the mineral content in blades of U. pinnatifida is high, especially in autumn. Some elements show an important seasonal variation, such as: K (14-54.8 g kg-1), P (2.7-7.0 g kg-1), Sr (361-569 mg kg-1), Fe (62-140 mg kg-1), Zn (8-103 mg kg-1), Br (45-94 mg kg-1) and Rb (4-24 mg kg-1). In the case of potentially toxic elements, a variation was seen mainly in arsenic, with higher values during summer and autumn. The concentrations of nickel and lead are below the limit of detection (0.9 mg kg-1). Sporophylls contain high concentrations of macro and micronutrients, with maximum values in spring. Besides, reproductive structures showed higher total arsenic values than blades. This could indicate that arsenic is mainly accumulated in sporophylls. Algae deposited on the beach are considered a waste; but they show a similar elemental composition to the samples extracted from the sea. We concluded that all samples analyzed could be used as food or fertilizers by local populations.

Absorption spectra are indicative of biological sample chemical composition and can be used as a basis for the construction of descriptive and predictive models for biotechnological screening or assays. In marine algae, chemical composition can vary due to species-specific differences in biochemistry, as well as intra-specific responses to unique environmental variables. Different indices (UVCi, UVB+Ai and PARi) were proposed and calculated to evaluate how photoprotective compounds vary in 18 species of Phaeophyceae. In addition, they were correlated to abiotic factors. Through this technique, seven main peaks were detected in the absorbing spectra of marine brown algal extracts. The highest photoprotective indices values were found in species collected in tropical areas, where higher solar radiation is observed compared to the southern Brazilian coast. Considering additional abiotic factors, water temperature and nitrate concentration were negatively correlated with UV indices. PARi's indices were positively affected by nitrate. All species collected on the Brazilian coast have absorption peaks in the region of phenolic compounds and carotenoids, suggesting that tropical marine brown macroalgae may have developed an effective antioxidant defense system, suggesting adaptation to environments characterized by high solar radiation. UVR/PAR indices congregated essential information to possible future biotechnological screening, facilitating selection of high priority species or sites, fostering actions to enhance alternative sustainable management strategies of coastal environments.

ABSTRACT Alginate is a major component of brown algae, but it cannot be utilized for ethanol fermentation by industrial microorganisms. A natural alginate degrading and ethanol producing strain was obtained in our previous research. However, the research on the ethanol metabolism process of the natural alginate fermentation strain is lacked. In this research, the key enzyme and metabolic process of ethanol fermentation were studied. Three kinds of key enzyme including alginate lyase, pyruvate dehydrogenase and ethanol dehydrogenase were determined. The enzyme activity in the metabolic process was relatively high at 60-96 h which was the most important period during the fermentation. Meanwhile the concentration change of the important substances including soluble sugar, reducing sugar, acidity, pyruvic acid and ethanol were tracked and analyzed. Total soluble sugar and reducing sugar change tendency during the fermentation was similar. In the whole fermentation process, the fermentation broth was acidic. The value of pyruvic acid content reached highest at 72 h. During 48-96 h, the growth of ethanol concentration was very obvious. The alginate metabolic process in natural alginate fermentation strain was to generate extracellular alginate lyase to degrade alginate to produce reducing sugar, and then some intermediate metabolites formed such as pyruvic acid. Finally under the effect of pyruvate dehydrogenase and ethanol dehydrogenase, ethanol was produced.

Approximately half of the Padina (Dictyotales, Phaeophyceae) species mineralize aragonite needles over the adaxial thallus surface, where mineral bands are interspersed with nonmineralized regions along the thallus from the apical to basal end. However, this calcification pattern and the related algal properties are not well understood. Therefore, this work was performed to elucidate a potential role of cell walls in the inhibition/induction of mineralization in the brown alga Padina gymnospora. In a comparison of specific thallus regions, differences were identified in the cellulose distribution, microfibrils arrangement and thickness, distribution and abundance of phenolic substances, and physical differences among the surfaces of the thallus (deformation, adhesion, topography, and nano-rugosity). In vitro mineralization assays indicated that phenolic substances are strong modulators of calcium carbonate crystals growth. In addition, de novo mineralization assays over cell wall surfaces that were used as templates, even without cellular activity, indicated that the cell wall remains a key factor in the induction/inhibition of mineralization. Overall, the current findings indicate a strong correlation between the physico-chemical and structural properties of the cell wall and the alternation pattern of the mineralization bands over the thallus of P. gymnospora.

Phytomyxea are obligate endoparasites of angiosperm plants and Stramenopiles characterised by a complex life cycle. Here Maullinia braseltonii sp. nov., an obligate parasite infecting the bull kelp Durvillaea (Phaeophyceae, Fucales) from the South-Eastern Pacific (Central Chile and Chiloe Island) and South-Western Atlantic (Falkland Islands, UK) is described. M. braseltonii causes distinct hypertrophies (galls) on the host thalli making it easily identifiable in the field. Sequence comparisons based on the partial 18S and the partial 18S-5.8S-28S regions confirmed its placement within the order Phagomyxida (Phytomyxea, Rhizaria), as a sister species of the marine parasite Maullinia ectocarpii, which is also a parasite of brown algae. The development of resting spores in M. braseltonii is described by light and electron microscopy and confirmed by FISH experiments, which visually showed the differential expression of the 28S non-coding gene, strongly in early plasmodia and weakly in late cysts. M. braseltonii is, so far, the only phytomyxean parasite of brown algae for which the formation of resting spores has been reported, and which is widely distributed in Durvillaea stocks from the Southeastern Pacific and Southwestern Atlantic.

Brown algae biomass has been shown to be a highly important industrial source for the production of alginates and different nutraceutical products. The characterization of this biomass is necessary in order to allocate its use to specific applications according to the chemical and biological characteristics of this highly variable resource. The methods commonly used for algae characterization require a long time for the analysis and rigorous pretreatments of samples. In this work, nondestructive and fast analyses of different morphological structures from Lessonia spicata and Macrocystis pyrifera, which were collected during different seasons, were performed using Fourier transform infrared (FT-IR) techniques in combination with chemometric methods. Mid-infrared (IR) and near-infrared (NIR) spectral ranges were tested to evaluate the spectral differences between the species, seasons, and morphological structures of algae using a principal component analysis (PCA). Quantitative analyses of the polyphenol and alginate contents and the anti-oxidant capacity of the samples were performed using partial least squares (PLS) with both spectral ranges in order to build a predictive model for the rapid quantification of these parameters with industrial purposes. The PCA mainly showed differences in the samples based on seasonal sampling, where changes were observed in the bands corresponding to polysaccharides, proteins, and lipids. The obtained PLS models had high correlation coefficients (r) for the polyphenol content and anti-oxidant capacity (r > 0.9) and lower values for the alginate determination (0.7 < r < 0.8). Fourier transform infrared-based techniques were suitable tools for the rapid characterization of algae biomass, in which high variability in the samples was incorporated for the qualitative and quantitative analyses, and have the potential to be used on an industrial scale.

Background: In recent years, Antarctica has become a key source of biotechnological resources. Native microorganisms have developed a wide range of survival strategies to adapt to the harsh Antarctic environment, including the formation of biofilms. Alginate is the principal component of the exopolysaccharide matrix in biofilms produced by Pseudomonas, and this component is highly demanded for the production of a wide variety of commercial products. There is a constant search for efficient alginate-producing organisms. Results: In this study, a novel strain of Pseudomonas mandelii isolated from Antarctica was characterized and found to overproduce alginate compared with other good alginate producers such as Pseudomonas aeruginosa and Pseudomonas fluorescens. Alginate production and expression levels of the alginate operon were highest at 4°C. It is probable that this alginate-overproducing phenotype was the result of downregulated MucA, an anti-sigma factor of AlgU. Conclusion: Because biofilm formation is an efficient bacterial strategy to overcome stressful conditions, alginate overproduction might represent the best solution for the successful adaptation of P. mandelii to the extreme temperatures of the Antarctic. Through additional research, it is possible that this novel P. mandelii strain could become an additional source for biotechnological alginate production.

A novel field transplantation technique, in which seaweed material is incorporated into dialysis tubing, was used to investigate intra-specific responses to metals in the model brown alga Ectocarpus siliculosus. Metal accumulation in the two strains was similar, with higher concentrations in material deployed to the metal-contaminated site (Ventanas, Chile) than the pristine site (Quintay, Chile). However, the oxidative responses differed. At Ventanas, strain Es147 (from low-polluted site) underwent oxidative damage whereas Es524 (from highly polluted site) was not affected. Concentrations of reduced ascorbate (ASC) and reduced glutathione (GSH) were significantly higher in Es524. Activities of the antioxidant enzymes superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT), and glutathione reductase (GR) all increased in Es524, whereas only SOD increased in Es147. For the first time, employing a field transplantation technique, we provide unambiguous evidence of inter-population variation of metal-tolerance in brown algae and establish that antioxidant defences are, in part, responsible.

Some populations of brown seaweed species inhabit metal-polluted environments and can develop tolerance to metal stress, but the mechanisms by which this is accomplished are still to be elucidated. To address this, the responses of two strains of the model brown alga Ectocarpus siliculosus isolated from sites with different histories of metal contamination exposed to total copper (CuT) concentrations ranging between 0 and 2.4 µM for 10 days were investigated. The synthesis of the metal-chelator phytochelatin (PCs) and relative levels of transcripts encoding the enzymes γ-glutamylcysteine synthetase (γ-GCS), glutathione synthase (GS) and phytochelatin synthase (PCS) that participate in the PC biosynthetic pathway were measured, along with the effects on growth, and adsorption and uptake of Cu. Growth of strain LIA, from a pristine site in Scotland, was inhibited to a greater extent, and at lower concentrations, than that of Es524, isolated from a Cu-contaminated site in Chile. Concentrations of intra-cellular Cu were higher and the exchangeable fraction was lower in LIA than Es524, especially at the highest exposure levels. Total glutathione concentrations increased in both strains with Cu exposure, whereas total PCs levels were higher in Es524 than LIA; PC2 and PC3 were detected in Es524 but PC2 only was found in LIA. The greater production and levels of polymerisation of PCs in Es524 can be explained by the up-regulation of genes encoding for key enzymes involved in the synthesis of PCs. In Es524 there was an increase in the transcripts of γ-GCS, GS and PCS, particularly under high Cu exposure, whereas in LIA4 transcripts of γ-GCS1 increased only slightly, γ-GCS2 and GS decreased and PCS did not change. The consequences of higher intra-cellular concentrations of Cu, lower production of PCs, and lower expression of enzymes involved in GSH-PCs synthesis may be contributing to an induced oxidative stress condition in LIA, which explains, at least in part, the observed sensitivity of LIA to Cu. Therefore, responses to Cu exposure in E. siliculosus relate to the contamination histories of the locations from where the strains were isolated and differences in Cu exclusion and PCs production are in part responsible for the development of intra-specific resistance.

Background: The increased industrial activity has resulted in the discharge of large amount of pollutants including non-degradable metals into the environment. Chromium is produced in several industrial processes and it can be found in the environment in two stable oxidation states, Cr(VI) and Cr(III). Cr(VI) is more hazardous due to its carcinogenic and mutagenic effects on living organisms. Although much less toxic, Cr(III) can also exert genotoxic effects under prolonged or severe exposure. It can be separated from the solution by precipitation but biosorption using brown algae seems to be an effective and sustainable treatment technique owing to its cost-effectiveness and environmental friendly characteristics. Macrocystis pyrifera and Undaria pinnatifida are two marine brown macroalgae with high capability of removing heavy metals including Cr(III) in batch mode of operation. In this work packed bed biosorption of Cr(III) by M. pyrifera and U. pinnatifida biomasses was evaluated. Results: The shapes of the breakthrough curves were rather different for each biomaterial. Parameters like the breakthrough time (t b) andzone mass transfer (MTZ) showed that U. pinnatifida has greater affinity for Cr(III). The maximum adsorption capacity at the exhaustion operating time (t e) demonstrated that M. pyrifera has higher retention capacity of Cr(III). The experimental data were fitted to Thomas, Yoon-Nelson and Dose-Response models. The best correlation coefficient (0.94 or 0.96) was obtained with Dose-Response that accurately describes the uptake behaviour of Cr(III) on the seaweed biomasses under different experimental conditions. The FT-IR spectra evidenced that Cr(III) adsorption occurred mainly by interaction between metal and carboxylate groups present on both the seaweed surfaces. Conclusions: M. pyrifera and U. pinnatifida biomasses are efficient biosorbents for Cr(III) adsorption under a continuous mode of operation although differences between uptake capacities suggest different mechanisms involved in the biosorption.

RESUMO

HIV-1 reverse transcriptase (HIV-1 RT) is a therapeutic target for the treatment of HIV-positive individuals or those already showing AIDS symptoms. In this perspective, the identification of new inhibitors for this enzyme is of great importance in view of the growing viral resistance to the existing treatments. This resistance has compromised the quality of life of those infected with multidrug-resistant strains, whose treatment options are already limited, putting at risk these individuals lives. The literature has recognized marine organisms and their products as natural sources for the identification of new therapeutic options for different pathologies. In this brief review, we consider the structure of HIV-1 RT and its most common inhibitors, as well as some marine diterpenes originally reported as HIV-1 RT inhibitors to encourage the identification and development of new marine antiviral prototypes.

The ischemic disorders, in which platelet aggregation and blood coagulation are involved, represent a major cause of disability and death worldwide. The antithrombotic therapy has unsatisfactory performance and may produce side effects. So, there is a need to seek molecules with antithrombotic properties. Marine organisms produce substances with different well defined ecological functions. Moreover, some of these molecules also exhibit pharmacological properties such as antiviral, anticancer, antiophidic and anticoagulant properties. The aim of this study was to evaluate, through in vitro tests, the effect of two extracts of brown algae and ten marine sponges from Brazil on platelet aggregation and blood coagulation. Our results revealed that most of the extracts were capable of inhibiting platelet aggregation and clotting measured by plasma recalcification tests, prothrombin time, activated partial thromboplastin time, and fibrinogenolytic activity. On the other hand, five of ten species of sponges induced platelet aggregation. Thus, the marine organisms studied here may have molecules with antithrombotic properties, presenting biotechnological potential to antithrombotic therapy. Further chemical investigation should be conducted on the active species to discover useful molecules for the development of new drugs to treat clotting disorders.
