RESUMO

Hand hygiene interventions are critical for reducing farmworker hand contamination and preventing the spread of produce-associated illness. Hand hygiene effectiveness may be produce-commodity specific, which could influence implementation strategies. This study's goal was to determine if produce commodity influences the ability of handwashing with soap and water or two-step alcohol-based hand sanitizer (ABHS) interventions to reduce soil and bacteria on farmworker hands. Farmworkers (n = 326) harvested produce (cantaloupe, jalapeño, and tomato) for 30 to 90 minutes before engaging in handwashing, two-step ABHS (jalapeño and cantaloupe), or no hand hygiene. Hands were rinsed to measure amounts of soil (absorbance at 600 nm) and indicator bacteria (coliforms, Enterococcus sp., generic Escherichia coli, and Bacteroidales universal [AllBac] and human-specific [BFD] 16S rRNA gene markers). Without hand hygiene, bacterial concentrations (0.88 to 5.1 log10 CFU/hand) on hands significantly differed by the produce commodity harvested. Moderate significant correlations (ρ = -0.41 to 0.56) between soil load and bacterial concentrations were observed. There were significant produce-commodity-specific differences in the ability of handwashing and two-step ABHS interventions to reduce soil (P < 0.0001), coliforms (P = 0.002), and Enterococcus sp. (P = 0.003), but not the Bacteroidales markers AllBac (P = 0.4) or BFD (P = 0.3). Contamination on hands of farmworkers who harvested cantaloupe was more difficult to remove. Overall, we found that a two-step ABHS intervention was similar to handwashing with soap and water at reducing bacteria on farmworker hands. In summary, produce commodity type should be considered when developing hand hygiene interventions on farms.IMPORTANCE This study demonstrated that the type of produce commodity handled influences the ability of handwashing with soap and water or a two-step alcohol-based hand sanitizer (ABHS) intervention to reduce soil and bacterial hand contamination. Handwashing with soap and water, as recommended by the FDA's Produce Safety Rule, when tested in three agricultural environments, does not always reduce bacterial loads. Consistent with past results, we found that the two-step ABHS method performed similarly to handwashing with soap and water but also does not always reduce bacterial loads in these contexts. Given the ease of use of the two-step ABHS method, which may increase compliance, the two-step ABHS method should be further evaluated and possibly considered for implementation in the agricultural environment. Taken together, these results provide important information on hand hygiene effectiveness in three agricultural contexts.

OBJECTIVE: In developing countries, rural communities often face the lack of potable water infrastructure and must rely on untreated sources for drinking, which are often contaminated with waterborne pathogens. The use of home water treatment devices is seen as one means of reducing the risk of exposure to waterborne pathogens. The aim of this study was to evaluate the microbiological and physicochemical performance of a simple in-home point-of-use device based on gravity ultrafiltration through an ultrafilter membrane. METHODS: Twenty-five randomly selected households from 2 rural communities in Culiacán, Mexico, were enrolled. Water samples were collected before and after treatment and during storage for a period of 8 weeks. Heterotrophic bacteria, total coliforms, fecal coliforms, Escherichia coli, and Giardia spp were quantified, as well as various physicochemical parameters. RESULTS: All of the untreated water samples contained high levels of indicator bacteria, but none were detected in the treated water fulfilling the requirements set by the Mexican Norm (NOM-127-SSA1-1994) and the World Health Organization guidelines for drinking water. However, indicator bacteria (fecal coliforms and E coli) were detected in every sample from water stored 24 hours after treatment. CONCLUSION: This study demonstrated that point-of-use filters using gravity-fed ultrafilters are a low-cost, effective water treatment technology for water of poor microbial quality. However, further identification of the sources and mechanisms by which water is contaminated when stored after treatment will help with designing and implementing better strategies for keeping water safe for domestic use.

Microbiological pollution of recreational waters is a major problem for public health as it may transmit waterborne diseases. To assess water quality, current legislation only requires limits for bacterial indicators; however, these organisms do not accurately predict the presence of parasites. Small number of parasites is usually present in water and although they are capable of causing disease, they may not be high enough to be detected. Detection therefore requires water samples to be concentrated. In this work three recreational aquatic environments located in the province of Salta were monitored over one year. For parasite quantification, water samples were collected every three months and concentrated by ultrafiltration. Detection was performed by microscopy. In addition, monthly monitoring was carried out in each aquatic environment: physicochemical variables were measured in situ and bacteriological counts were determined by traditional microbiological techniques. Of 14 parasites identified, at least nine were detected in each aquatic environment sampled. While bacteriological contamination decreased in most cases during winter (76-99%), parasites were present year-round, becoming a continual threat to public health. Thus, we here propose that it is necessary to use specific parasitological indicators to prevent waterborne disease transmission. Our results suggest that Entamoeba would be a suitable indicator as it was found in all environments and showed minimal seasonal variation. The results obtained in this study have epidemiological relevance and will allow decision-makers to propose solutions for water protection in order to care for population health.

La contaminación microbiológica de aguas recreativas es un problema preocupante, ya que las personas que las utilizan pueden contraer enfermedades que podrían afectar su bienestar general. Para evaluar la calidad del agua, las legislaciones existentes solo establecen límites de indicadores bacterianos, los cuales no predicen con exactitud la presencia de parásitos. Además, la cantidad de parásitos presentes en el agua, aunque suficiente para producir enfermedad, suele ser pequeña, por lo que, se necesita una etapa previa de concentración para poder detectarlos. En este trabajo se monitorearon trimestralmente durante un año tres ambientes acuáticos de usos recreativos de la provincia de Salta, realizando la concentración de las muestras y la posterior preparación para la búsqueda de elementos parasitarios por microscopía. Adicionalmente, en cada ambiente se midieron mensualmente variables fisicoquímicas in situ y variables bacteriológicas por técnicas microbiológicas tradicionales. En cada ambiente se encontraron como mínimo 9 de los 14 parásitos detectados en conjunto. La presencia de los elementos parasitarios no presentó correlación con indicadores bacterianos en ningún ambiente ni en ninguna de las estaciones (p > 0,05). Mientras que en invierno la contaminación bacteriológica disminuyó entre un 76 % y un 99 %, los elementos parasitarios no presentaron disminución estacional. Los resultados permiten sugerir al género Entamoeba como indicador anual de contaminación parasitaria, ya que este fue encontrado en todos los ambientes con mínimas variaciones estacionales. Estos resultados poseen relevancia epidemiológica, dado que permitirán a los tomadores de decisiones proponer medidas para mejorar el bienestar de la población.(AU)

O leite de cabra é produzido em pequena escala, o que torna necessária sua estocagem, permitida pela legislação brasileira. O presente trabalho teve por objetivo avaliar a ação do resfriamento e congelamento sobre as características físico-químicas e microbiológicas do leite de cabra. Não foi observada variabilidade nos valores médios de acidez, estabilidade ao álcool, densidade, gordura, proteína, sólidos totais e lactose após armazenamento sob frio. Não foi observada instabilidade ao calor em nenhuma das amostras analisadas. Verificou-se tendência à diminuição na contagem de células somáticas (CSS) no leite congelado em relação ao fresco e ao refrigerado. A quantificação (NMP) de coliformes totais e termotolerantes variou de < 3 (ausência de crescimento) à > 1.100 UFC/mL, independentemente do armazenamento. Entretanto, determinou-se contagem de coliformes totais e termotolerantes significativamente maior (p < 0,0001) nas amostras mantidas sob refrigeração. Na prova de lactofermentação, observaram-se coágulos digeridos, floculosos e sulcados sem estabelecer correlação entre estes e a contagem bacteriana. A CCS e a contagem microbiana são critérios adotados no pagamento pela qualidade no leite caprino, e a temperatura de armazenamento pode influir sobre elas.(AU)

However, no variability was verified in average acidity, alcohol stability, density, fat protein, total solids and lactose values after cold storage. Heat instability was not observed in any of the analyzed samples. There was a decreasing tendency in the somatic cell count (SCC) of frozen milk in comparison to fresh and refrigerated milk. The quantification (NMP) of total and thermo-tolerant coliforms varies from < 3 (no growth) to > 1,100 UFC/mL, regardless of the storage method. However, a slightly higher count (p < 0.0001) was observed for total and thermo-tolerant coliforms in the samples kept under refrigeration. In the lacto-fermentation test, digested, flocculated and grooved clots were observed without establishing a correlation between them and the bacterial count. SCC and microbial count are the criteria adopted when paying for the quality of goat milk, and the storage temperature may affect these criteria.(AU)
