In this study, twelve campesterol derivatives (2-13) were prepared by esterification reaction at the hydroxy group in C-3 and catalytic hydrogenation at the carbon-carbon double bond in C-5(6). All obtained compounds were characterized by IR, 1 H-NMR, 13 C-NMR, and MS spectra. Campesterol (1) and its derivatives (2-13) were evaluated in vitro against Staphylococcus aureus (ATCC 6538), Streptococcus mutans (ATCC 0046), Escherichia coli (ATCC 10536), Pseudomonas aeruginosa (ATCC 15442), and Klebsiella pneumoniae (ATCC 10031) using the microdilution method. Among tested compounds, 4, 6, 9, 11, 12, and 13 displayed the best antibacterial activity. Moreover, to support the antibacterial activity experiments, the investigation of molecular interactions of more active compounds, and also compound 1 and neomycin, used as starting material and positive control, respectively, at the binding site of the target proteins was performed using molecular docking simulations. Four compounds (7, 9, 10 and 11) are herein described for the first time.

Phase shift is characterized by an abrupt change in the structure of a community in response to a disturbance that can break its resistance, displacing it from its natural variation. This phenomenon has been recognized in several ecosystems and often points to human activities as the main cause. However, reactions of shifted communities to anthropogenic impacts have been less studied. In recent decades, heatwaves resulting from climate change have strongly affected coral reefs. Mass coral bleaching events are recognized as the main cause of coral reef phase shifts on a global scale. In 2019, an unprecedented heatwave hit the southwest Atlantic Ocean causing mass coral bleaching in non-degraded and phase-shifted reefs of Todos os Santos Bay, at an intensity never recorded in a 34-year historical series. We analyzed the effects of this event on the resistance of phase-shifted reefs, dominated by the zoantharian Palythoa cf. variabilis. Using benthic coverage data from 2003, 2007, 2011, 2017, and 2019, we analyzed three non-degraded reefs and three phase-shifted reefs. We estimated the coverage and bleaching of corals and P. cf. variabilis on each reef. There was a reduction in coral coverage in non-degraded reefs before the 2019 mass bleaching event (i.e., heatwave). However, there was no significant coral coverage variation after the event and the structure of non-degraded reef communities did not change. In phase-shifted reefs the coverage of zoantharians did not change significantly before the 2019 event, however, after the mass bleaching, there was a significant reduction in the coverage of these organisms. Here we revealed that the resistance of the shifted community was broken, and its structure was altered, indicating that reefs in this condition were more susceptible to bleaching disturbance than non-degraded reefs.

Abstract Introduction: The coral-associated bacteria with antimicrobial activity may be important to promote the health of their host through various interactions, and may be explored as a source of new bioactive compounds. Objective: To analyze the antimicrobial activity of bacteria associated with the zoanthid Palythoa caribaeorum from the coral reefs of Carapibus, Paraiba state, Brazil. Methods: The phylogenetic analysis of the bacteria was conducted based on partial sequences of the 16S rRNA gene using molecular and bioinformatics tools. The antimicrobial activity of the 49 isolates was tested against four bacterial strains and one yeast strain: Bacillus cereus (CCT0198), Escherichia coli (ATCC 25922), Staphylococcus aureus (ATCC 25923), Pseudomonas aeruginosa and Candida albicans (ATCC 10231). The antibiosis and antibiogram assays were conducted and the Minimal Inhibitory Concentration (MIC) was determined by the microdilution method. Results: The bacterial isolates belonged to Firmicutes phylum (84 % of the isolates) and the Proteobacteria phylum (16 % of the isolates). Among the 49 isolates five genera were found, with the Bacillus genus being the most abundant (82 % of the isolates), followed by Vibrio (10 %), Pseudomonas (4 %), Staphylococcus (2 %) and Alteromonas (2 %). Antibiosis test revealed that 16 isolates (33 %) showed antimicrobial activity against one or more of five tested reference strains. The highest number of antagonistic bacteria were found in the Bacillus genus (12 isolates), followed by Vibrio (three isolates) and Pseudomonas (one isolate) genera. The B. subtilis NC8 was the only isolate that inhibited all tested strains in the antibiosis assay. However, antibiogram test with post-culture cell-free supernatant of NC8 isolate showed the inhibition of only B. cereus, S. aureus and C. albicans, and the lyophilized and dialyzed material of this isolate inhibited only B. cereus. The lyophilized material showed bacteriostatic activity against B. cereus, with a MIC value of 125 μg/μl, and in the cytotoxicity assay, the hemolysis value was of 4.8 %, indicating its low cytotoxicity. Conclusions: The results show the antimicrobial potential of some bacterial isolates associated with the P. caribaeourum tissue, especially those belonged to Bacillus genus.


Resumen Introducción: La actividad antimicrobiana realizada por las bacterias asociadas con los corales, además de promover la salud de su huésped, representa una fuente para obtener nuevos compuestos bioactivos. Objetivo: Analizar la actividad antimicrobiana de las bacterias asociadas con el zoantario Palythoa caribaeorum de los arrecifes de Carapibus, Paraíba, Brasil. Metodología: El análisis filogenético de la bacterias se realizó con base en secuencias parciales del gen RNAr 16S utilizando herramientas moleculares y de bioinformática. La actividad antimicrobiana de las cepas se probó contra cuatro cepas bacterianas y una cepa de levadura: Bacillus cereus (CCT0198), Escherichia coli (ATCC 25922), Staphylococcus aureus (ATCC 25923), Pseudomonas aeruginosa y Candida albicans (ATCC 10231), utilizando ensayos antibiosis y antibiograma, y la concentración inhibitoria mínima (CIM) que se determinó por el método de microdilución. Resultados: Las cepas bacterianas pertenecían a Firmicutes (84 %) y Gammaproteobacteria (16 %). Entre 49 cepas se encontraron cinco géneros de bacterias: Bacillus, Vibrio, Pseudomonas, Staphylococcus y Alteromonas. Un total de 19 cepas exhibieron actividad antimicrobiana, siendo el género Bacillus el responsable del mayor número de bacterias antagonistas, con 12 cepas positivas en el ensayo de antibiosis y cuatro en la prueba de antibiograma. El mayor número de bacterias antagonistas se encontró en Bacillus (12 aislamientos), seguido por Vibrio (tres aislamientos) y Pseudomonas (un aisladmiento). El NC8, clasificado como Bacillus subtilis, inhibió todas las cepas estándar en el ensayo de antibiosis y las cepas de B. cereus, S. aureus y C. albicans en la prueba de antibiograma. El material liofilizado del B. subtilis NC8 mostró acción bacteriostática contra B. cereus, con un valor de CIM de 125 μg/μl. En la prueba de citotoxicidad, el grado de hemólisis fue del 4.8 % para el material liofilizado a las concentraciones probadas, lo que indica su baja citotoxicidad. Conclusión: Los resultados muestran el potencial antimicrobiano de algunos aislamientos bacterianos asociados al P. caribaeourum, especialmente los pertenecientes al género Bacillus.

Marine isolates such as palytoxin (PTX) are of concern within the Caribbean region due to their toxicity. PTX for example has been described as a one of the most known potent marine toxins, known to prevent predation from larger species (e.g. vertebrates) as well as the prevention of being overgrown from other coral species. PTX is a polyhydroxylated polyether toxin with a very large and complex chemical structure that possesses both hydrophilic and lipophilic properties. Previous acute toxicity tests using brine shrimp (Artemia salina) and PTX extract had shown it to be moderately toxic. In humans, PTX has been credited to be responsible for extreme symptoms such anaphylactic shock, rapid cardiac failure and eventual death occurring within minutes. Extrapolation for human dose ranges has therefore been suggested to be between 2.3 and 31.5 µg. This study isolates a potentially PTX-enriched extract from Palythoa caribaeorum and examines its organic extract toxicity from a biogeography perspective from a within-colony to a variety of reef sites around Trinidad and Tobago that are popular for marine visitors. This research represents an acute study with a high level of crude organic extract toxicity on A. salina whilst postulating potential factors which may contribute to its extreme toxicity and the risk posed to users of these environments.

BACKGROUND: Marine invertebrate-associated microbial communities are interesting examples of complex symbiotic systems and are a potential source of biotechnological products. RESULTS: In this work, pyrosequencing-based assessment from bacterial community structures of sediments, two sponges, and one zoanthid collected in the Mexican Caribbean was performed. The results suggest that the bacterial diversity at the species level is higher in the sediments than in the animal samples. Analysis of bacterial communities' structure showed that about two thirds of the bacterial diversity in all the samples belongs to the phyla Acidobacteria and Proteobacteria. The genus Acidobacteriumappears to dominate the bacterial community in all the samples, reaching almost 80% in the sponge Hyrtios. CONCLUSIONS: Our evidence suggests that the sympatric location of these benthonic species may lead to common bacterial structure features among their bacterial communities. The results may serve as a first insight to formulate hypotheses that lead to more extensive studies of sessile marine organisms' microbiomes from the Mexican Caribbean.

This work aimed to test the influence of Palythoa caribeorum, a widely distributed zoanthid in the Atlantic, on site-attached reef fish in a subtropical rocky shore. Density, richness and vertical distribution of reef fish inside (ID) and outside (OD) previously chosen P. caribaeorum dominance patches were compared through stationary visual censuses along three different periods. Fishes were grouped in different trophic guilds to evidence differences in resources uses in both treatments. A complexity index was estimated by the chain link method and percentage covering of benthic organisms was obtained analyzing random points from replicated photo-quadrats. We observed thirty-eight species of fishes, belonging to twenty-five families. Reef fish communities between studied patches were similar,both in terms of species composition and vertical distribution. Considering only the most site-attached fishes, which were the most frequent and abundant species, data showed that ID sustains higher diversity and abundance than OD. Results showed that benthic composition differ significantly among patches whereas complexity remained without differences. Otherwise, results indicated that these areas might play an important role in space limitation, structuring neighboring benthic community and consequently reef fish assemblages.


Este estudo visou testar a influência de Palythoa caribeorum, um zoantídeo amplamente distribuído no Atlântico, na estruturação da comunidade de peixes recifais associados a um costão rochoso de uma região subtropical. A densidade, a riqueza e a distribuição vertical de peixes recifais em áreas previamente selecionadas com e sem a dominância de Palythoa caribaeorum foram comparadas através de censos visuais estacionários em três períodos distintos de tempo. Os peixes foram agrupados em guildas tróficas a fim de evidenciar diferenças nos usos dos recursos nas diferentes áreas analisadas. Foram analisados também índices de complexidade estrutural através do método da corrente e os percentuais de cobertura bentônica através de fotos quadracts replicados. Foram registradas trinta e oito espécies de peixes recifais de vinte e cinco famílias diferentes. Acomunidade de peixes entre as áreas estudadas foi similar tanto em composição de espécies quanto em distribuição vertical. Considerando apenas as espécies mais associadas ao substrato, que foram as mais freqüentes e abundantes, os dados mostraram que as áreas com dominância de P. caribaeorum sustentam maior diversidade e abundância do que as áreas sem a dominância de P. caribaeorum. Foram encontradas ainda diferenças significativas na composição bentônica entre os diferentes tratamentos estudados, mas não foram verificadas diferenças entre a complexidade estrutural entre estas áreas. No entanto, os resultados sugerem que as áreas com dominância de P. caribaeorum podem desempenhar papel importante na limitação de espaços, estruturando as comunidades bentônicas e, conseqüentemente, afetando a comunidade de peixes recifais.
