RESUMO

Este trabalho tem como objetivo avaliar o efeito do horário de aplicação da gonadotrofina coriônica humana (hCG), sobre os parâmetros reprodutivos de 40 machos das variedades de tilápias do Nilo GIFT e UFLA. O experimento foi conduzido no Laboratório de Recirculação de Água da Estação de Piscicultura, do Departamento de Zootecnia da Universidade Federal de Lavras - MG. O delineamento experimental foi inteiramente casualizado, em esquema fatorial 2 x 4 (duas variedades e quatro horários de aplicação do hCG) com cinco repetições. Foram utilizados 20 machos da variedade GIFT e 20 machos da variedade UFLA, microchipados e alojados em um sistema de recirculação de água. Os horários foram 6, 12, 18 e 24 horas. A dosagem de hCG foi realizada em dose única. Foram analisadas as variáveis: taxa e duração da motilidade espermática, morfologia espermática, volume de sêmen e a concentração de espermatozoides do sêmen. Os dados obtidos foram submetidos à análise da variância e as médias foram comparadas pelo teste SNK a 5% de significância. O horário de aplicação e a variedade utilizada nao influenciaram as variáveis analisadas, porém a variedade UFLA apresentou um maior número de animais que espermiaram. A concentração apresentou uma media de 5,73 x 105 espermatozoide/mL para a variedade UFLA e 4,71 x 105 espermatozoide/mL para a variedade GIFT. O volume de sêmen encontrado para a GIFT foi de 0,9 mL enquanto para a UFLA foi de 0,6 mL. A motilidade espermática encontrada foi de 96,5% com duração de 4,33 minutos para a variedade UFLA e 88,4% com duração de 4,8 minutos para variedade GIFT. A quantidade de células espermáticas normais apresentou uma média de 86,35% para a variedade UFLA e 86,46% para a variedade GIFT. A indução com hCG em diferentes horários de aplicação não apresentou diferença significativa sobre os parâmetros de qualidade espermática dos machos de tilápia.(AU)


This study evaluated the effect of time of application of human chorionic gonadotropin (hCG) on the reproductive performance of 40 males of of Nile tilapia males from varieties GIFT and UFLA. The experiment was conducted at the Water Recirculation Laboratory of the Fish Farm Station, Department of Animal Science, Federal University of Lavras - MG. The experiment was completely randomized design, in a 2 x 4 factorial scheme (two varieties and four hCG application periods) with five replicates. Twenty males of the GIFT variety and 20 males of the UFLA variety, with microchips, were housed in a water recirculation system. The application periods were at 6 a.m. 12 a.m., 6 p.m. and 12 p.m.. The hCG dosage was performed in a single dose. The following variables were analyzed: sperm motility rate and duration, sperm morphology, semen volume and semen sperm concentration. The obtained data were submitted to variance analyses and the averages were compared by the SNK test at 5% of significance. The period of application and the variety used did not influence the analyzed variables, however the UFLA variety showed a larger number of animals that spermized. The concentration presented an average of 5.73 x 105 sperm/mL for the UFLA variety and 4.71 x 105 sperm/mL for the GIFT variety. The volume of semen found for GIFT was 0.9 mL whereas for UFLA it was 0.6 mL. The sperm motility was 96.5% with a duration of 4.33 minutes for the UFLA variety and 88.4% with a duration of 4.8 minutes for the GIFT variety. The number of normal sperm cells presented an average of 86.35% for the UFLA variety and 86.46% for the GIFT variety. Sixty six indigenous Saccharomyces cerevisiae strains were evaluated in stressful conditions (temperature, osmolarity, sulphite and ethanol tolerance) and also ability to flocculate. Eighteen strains showed tolerant characteristics to these stressful conditions, growing at 42 ºC, in 0.04% sulphite, 1 mol L-1 NaCl and 12% ethanol. No flocculent characteristics were observed. These strains were evaluated according to their fermentative performance in sugar cane juice. The conversion factors of substrates into ethanol (Yp/s), glycerol (Yg/s) and acetic acid (Yac/s), were calculated. The highest values of Yp/s in sugar cane juice fermentation were obtained by four strains, one isolated from fruit (0.46) and the others from sugar cane (0.45, 0.44 and 0.43). These values were higher than the value obtained using traditional yeast (0.38) currently employed in the Brazilian bioethanol industry. The parameters Yg/s and Yac/s were low for all strains. The UFLA FW221 presented the higher values for parameter related to bioethanol production. Thus, it was tested in co-culture with Lactobacillus fermentum. Besides this, a 20-L vessel for five consecutive batches of fermentation was performed. This strain was genetically stable and remained viable during all batches, producing high amounts of ethanol. The UFLA FW221 isolated from fruit was suitable to produce bioethanol in sugar cane juice. Therefore, the study of the biodiversity of yeasts from different environmental can reveal strains with desired characteristics to industrial applications.
