The aim of this work was to develop a liver tissue function index during the transition period of dairy cows managed in low-tropic conditions. In two farms, twenty crossbred and synthetic native cows during the peripartum period were selected, and blood samples were taken on days -30 and -15 prepartum, the calving day, and 7, 20, 35, 50, 65, 80 and 105 days postpartum for serum metabolic tests. On each measurement day, body condition scores (BCS) and parameters on nitrogen metabolism (total protein-TP, albumin-ALB, globulin-GLOB, urea), adipose tissue metabolism (cholesterol-COL, non-esterified fatty acids-NEFA) and two transaminases (alanine aminotransferase-ALT and aspartate aminotransferase-AST) were evaluated. Data analysis included the Spearman correlation, principal components, multiple linear regression and cluster analysis. Results showed that regarding the days after calving and BCS, a liver tissue function index can be constructed using the TP, urea, COL, ALT and NEFA. The estimated index generated three groupings, both by days after calving and BCS. In the former, the index discriminated the metabolic behavior in the prepartum, parturition and postpartum periods, while in the latter, the index discriminated between extreme (2.25, 2.50 and 4.25), slightly low (2.75 and 3.0) and slightly high (3.25 to 4) conditions. The results allow us to conclude that it is feasible to construct mathematical function indexes for liver function to monitor metabolic changes during highly demanding productive phases in dairy cows under tropical conditions.

Maternal status during the transition period can significantly impact the health and performance of Holstein dairy calves, with lasting effects on various variables. However, the relationship between maternal late gestation metabolic status, seasonality, and their impact on offspring remains unclear. This study aimed to assess the influence of maternal variables at calving on the performance, metabolism, and immunity of 28 dairy calves during their first month of life. Blood samples were collected from 28 Holstein cows at calving. Median results for maternal variables including non-esterified fatty acids (NEFA), ß-hydroxybutyrate (BHB), glucose, total protein (TP), albumin, triglycerides (TG), total cholesterol (TC), haptoglobin (Hp), body weight (BW), and body condition score (BCS) were determined. These median values served as a basis for categorizing the offspring into two groups based on their dams' high or low degree of each maternal variable. Additionally, calves were categorized by the season of birth (Spring vs. Winter), with 14 in each. Blood samples were collected from the calves at birth and on days 1, 7, 14, and 28 to assess IgG, biochemical parameters, and haptoglobin concentration. Reactive oxygen species (ROS) production by polymorphonuclear cells stimulated by various agents was also evaluated. Clinical assessments were conducted for diarrhea and bovine respiratory disease frequencies. Despite the overall health of the cows, differences were observed in the calves between maternal groups. Heavier cows with high maternal BCS tended to have larger offspring, while high maternal BCS was associated with increased diarrhea prevalence. Low maternal BCS resulted in a stronger innate immune response, indicated by higher ROS production. Calves from cows experiencing metabolic changes during calving displayed elevated Hp concentrations. Spring-born calves were larger but had lower serum IgG concentration and reduced innate immune response compared to winter-born calves. Additionally, spring-born calves exhibited higher Hp and increased diarrhea prevalence on day 28. These findings underscore the importance of the prenatal period in determining neonatal health and suggest further research to elucidate the long-term clinical implications of maternal effects on offspring health and growth. Investigating offspring constituents later in life can provide insight into the persistence of maternal effects over time.

This study aimed to evaluate the relationship between prepartum subclinical hypomagnesemia (pre-SHMg) and the occurrence of dystocia, metritis, clinical mastitis, lameness, and subclinical hypomagnesemia postpartum (post-SHMg) in pasture-based dairy cows. Also, the difference in means of prepartum magnesium (Mg) concentration by postpartum health events was evaluated. A total of 890 dairy cows from 32 commercial farms located in southern Chile were enrolled. Cows were examined twice, once between 30 and 3 days before and once between 3 and 30 days after calving. Blood samples were collected on both assessments, and cows were considered as having SHMg if serum total Mg < 0.65 mmol/L. On the postpartum visit, cows were evaluated for metritis and lameness. Information about clinical mastitis and dystocia was collected from on-farm records. Data were analyzed using multivariable mixed linear models and multivariable mixed logistic regression models. The overall prevalence of pre-SHMg was 9.9%, and its presence was associated with the occurrence of post-SHMg (odd ratio [OR] = 5.7; P < 0.0001) and metritis (OR = 3.1; P = 0.04). However, we did not detect an association between pre-SHMg and dystocia, clinical mastitis, or lameness after calving. Prepartum serum Mg concentrations were lower in cows that developed post-SHMg than those that did not (LSM ± SE = 0.75 ± 0.02 mmol/L vs. 0.83 ± 0.02 mmol/L; P < 0.0001). In conclusion, pre-SHMg was associated with a higher risk of post-SHMg and metritis in grazing dairy cows but not other postpartum health events.

Researchers have reported the benefits of feeding rumen-protected methionine (RPM) during the peripartum on the health parameters of dairy cows. Rumen-protected Met has reportedly improved milk yield, milk components, and liver health, but the literature is scarce on its effects in commercial herds. Therefore, we aimed to determine the effects of feeding RPM (Smartamine M, Adisseo Inc., Antony, France) prepartum (8 g/cow per day) and postpartum (15 g/cow per day) on performance, metabolic profile, and culling rate of Holstein cows in a commercial herd. One hundred sixty-six (n = 166) Holstein cows, 58 nulliparous and 108 parous, were randomly assigned to 1 of 2 dietary treatments, consisting of TMR top-dressed with RPM (RPMet; 2.35% and 2.24% Met of MP for close-up and fresh cows, respectively) or without (control [CON] 2.03% and 1.89% Met of MP for close-up and fresh cows, respectively), fed from 21 ± 6 d prepartum until 16 ± 5 d postpartum. From 17 DIM until dry-off, all cows received RPMet. Daily milk yield was recorded, and milk samples were collected in the first and second weeks after calving to determine their composition. Blood samples were collected before the morning feeding on -14, -7, +1, +7, and +14 d relative to calving. Mortality and morbidity were recorded during the first 60 DIM. Cows supplemented with RPMet had greater milk yield during the first 16 DIM (31.76 vs. 30.37 kg/d; SEM = 1.04, respectively), and had greater milk fat content (4.45 vs. 4.10%; SEM = 0.11, respectively), but not milk total protein (3.47 vs. 3.39%; SEM = 0.04, respectively) and casein contents (2.74 vs. 2.66%; SEM = 0.04, respectively) than CON cows. Cows in RPMet had increased plasma Met concentrations than cows in CON (24.9 vs. 21.0 µmol/L; SEM = 1.2, respectively). Although morbidity was similar between treatments, the culling rate from calving until 60 DIM was lower for RPMet cows than for CON cows (2.4% vs. 12.1%; SEM = 0.02). In conclusion, cows receiving RPMet have greater milk yield, improved milk fat content, and a lower culling rate at 60 DIM than CON cows.

BACKGROUND: The periparturient period in dairy cows is marked by immunosuppression which increases the likelihood of infectious disorders, particularly also mastitis. An in-depth understanding of peripartum leukocyte biology is vital for the implementation of highly successful post-partum disease prevention measures. Immune checkpoint molecules, such as programmed death 1 (PD-1) and cytotoxic T-lymphocyte antigen 4 (CTLA-4), are critical inhibitory receptors expressed on immune cells, particularly T cells, that drive immunosuppressive signaling pathways. However, the potential role of immune checkpoint molecules expression in T-cells on udder health has never been explored. Thus, the association between the occurrence of new postpartum intramammary infections (IMIs) and the expression of programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4) on blood T-cells during the peripartum period was investigated. RESULTS: In this study, the incidence of IMIs by any pathogen in early lactation was not associated with a higher expression of PD-1 and CTLA-4 in the periparturient period. However, the incidence of IMIs by major pathogens throughout the first month of lactation was significantly associated with higher expression of PD-1 at 14 days before calving (P = 0.03) and CTLA-4 at parturition (P = 0.03) by blood T-cells. Also, the expression of CTLA-4 at D0 (P = 0.012) by T-cells was associated with the occurrence of persistent IMIs during the first month of lactation. CONCLUSIONS: To our knowledge, this is the first report to investigate the expression of PD-1 and CTLA-4 by blood T-lymphocytes during the periparturient period in dairy cows and to explore their relationship with the incidence of new IMIs in the postpartum period. Thus, a comprehensive understanding of leukocyte biology during peripartum would appear to be a prerequisite for the identification of resilient dairy cows or targets innovative (immunological) non-antibiotic approaches in the transition period.

The aims of this study were to estimate the genetic parameters for fat-to-protein ratio (F:P) within the first 90 days of lactation and to examine their genetic associations with daily milk yield (MY), somatic cell score (SCS), and calving interval between the first and second calving (IFSC) and between the second and third calving (ISTC) during the first three lactations of Holstein cows. We utilized 200,626 production-related data officially recorded from 77,436 cows milked two or three times a day from 2012 to 2022, sourced from the Holstein Cattle Breeders Association of Paraná State, Brazil. The (co)variance components were estimated using animal models, adopting the restricted maximum likelihood (REML) method with single-trait analysis (for heritability and repeatability) and two-trait analysis (for genetic and phenotypic correlations), per lactation. Regardless of lactation number, heritability estimates were relatively low, ranging from 0.08 ± 0.005 to 0.10 ± 0.003 for F:P; 0.08 ± 0.01 to 0.18 ± 0.005 for MY; 0.04 ± 0.01 to 0.07 ± 0.004 for SCS; and 0.03 ± 0.01 for both IFSC and ISTC. Repeatability estimates within the same lactation were low for F:P (ranging from 0.17 ± 0.002 to 0.19 ± 0.03), high for MY (between 0.50 ± 0.003 and 0.53 ± 0.002), and moderate to high for SCS (between 0.39 ± 0.003 and 0.44 ± 0.004). Genetic correlations between F:P and MY ranged from -0.26 ± 0.03 to -0.15 ± 0.02; F:P and SCS, from -0.06 ± 0.03 to -0.03 ± 0.08; F:P and IFSC, 0.31 ± 0.01; F:P and ISTC, 0.20 ± 0.01; MY and IFSC, 0.24 ± 0.05; and MY and ISTC, 0.13 ± 0.08. The fat-to-protein ratio during early lactation showed low genetic variability, regardless of lactation number. Furthermore, it was genetically correlated with MY, IFSC, and ISTC, although there is an antagonistic and unfavorable correlation between traits that can limit genetic progress.

In the present work we aimed to study the effects of parenteral vitamin and mineral supplementation on hepatic fatty acid metabolism as well as on the oxidative stress biomarkers in biological samples of transition cows. The supplemented group (SG, n = 11) received a subcutaneous injection of 5 mL of vitamin A palmitate 35 mg/mL, vitamin E acetate 50 mg/mL plus other injection of 5 mL of copper edetate 10 mg/mL, zinc edetate 40 mg/mL, manganese edetate 10 mg/mL, and sodium selenite 5 mg/mL on days - 60, - 30, and 7 (± 3) relative to calving. The control group (CG, n = 11) received two subcutaneous injections of 5 mL of 9 mg/mL sodium chloride at the same times of the SG. Blood, urine, and liver biopsies were sampled 21 (± 3) days before the expected calving date and 7 and 21 (± 3) days after calving. Results revealed that supplemented animals had higher glutation peroxidase (GSH-Px) activity, lower and higher concentration of 3-nitrotyrosine (3-NT) in the liver and plasma, respectively, higher expression of the mitochondrial beta-oxidation enzyme carnitine palmitoyltransferase 1 in the liver, and lower content of hepatic triacylglycerol, mirroring plasma liver function parameters. No differences between groups were found in the superoxide dismutase activity, MDA concentrations, the protein abundance of peroxisomal acyl-CoA oxidase 1, diacylglycerol O-acyltransferase 1, and peroxisome proliferator-activated receptor alpha. These results suggest that the vitamin and mineral supplementation provided to dairy cows had a beneficial effect on GSH-Px activity, hepatic 3-NT concentration, and on the metabolic adaptation during the peripartum period.

This study aimed to evaluate the effects of parity, body condition score (BCS) at calving, and milk yield on the metabolic profile of Gyr (Zebu) cows. Healthy cows in late pregnancy were grouped according to parity (primiparous, biparous, and multiparous); to BCS scale at calving (high-HBCS and normal-NBCS); and to milk yield (high-HP and moderate-MP production). BCS was assessed, and blood samples were collected on -21, -7, 0, 7, 21, and 42 days relative to parturition. The concentrations of non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHB), cholesterol, glucose, total protein (TP), albumin, total calcium (Ca), phosphorus (P), and magnesium (Mg); and activities of aspartate aminotransferase and gamma-glutamyltransferase were measured. Data were analyzed by two-way repeated measures ANOVA. The frequencies of high lipomobilization, subclinical ketosis, subclinical hypocalcemia (SCH), and the occurrence of diseases during early lactation were established. Regardless of grouping, NEFA, BHB, and cholesterol increased during early lactation; glucose showed higher values at calving; TP and albumin were higher at 21 and 42 DIM; and Ca, P, and Mg were lower at calving. Parity had little effect on the metabolic profile, HBCS did not differ from NBCS cows, and HP did not differ from MP cows in most metabolites. High lipomobilization in early lactation and SCH at calving were the most common imbalances but were not related to postpartum diseases. High-yielding Gyr cows have a balanced metabolic profile during the transition period, with few biologically relevant effects of parity, BCS at parturition, or milk yielded.

This study aimed to analyze the possible relationship between days to conception and different oxidative stress (OS) biomarkers and liver functional parameters in multiparous dairy cows. Besides, a fast reliable method for the accurate measurement of malondialdehyde (MDA) by liquid chromatography-tandem mass spectrometry was developed in several matrices. During lactation, the days to conception of 28 cows were determined for a retrospective study. According to this parameter, cows were divided into two groups: high and low days to conception (HDC and LDC, respectively). Blood, urine and liver biopsies were sampled 21 days before the expected calving date, and 7 and 21 days after calving. The method developed for MDA was validated according to international requirements. The lower limit of quantification was 0.25 µmol/L for plasma and urine and 10.00 µmol/L for liver tissue. No differences between groups were observed in the systemic concentration of non-esterified fatty acids, ß-hydroxybutyric acid and liver triacylglycerol content (P > 0.05). Cholesterol concentration was higher in the LDC than in the HDC group (P < 0.05). Plasma 3-nitrotyrosine (3-NT) concentration was lower in the LDC than in the HDC group on day 21 post-calving (P < 0.05). Superoxide dismutase activity was higher in the LDC than in the HDC group (P < 0.05). Particularly, in the liver, 3-NT and MDA concentrations were lower in the LDC than in the HDC group (P < 0.05). These results allow inferring that the amelioration of OS biomarkers in plasma and liver could be related to a better reproductive performance of dairy cows.

This study aimed to evaluate the impact of body condition score (BCS) at calving, parity, and the calving season on the performance of dairy cows and their offspring. Data from 521 Holstein cows that calved a female calf and had their BCS evaluated at calving from a single commercial farm located in Southern Brazil were used. Cows were categorized into five BCS classes: class 1: <3.0 (n = 19), class 2: 3.0-3.25 (n = 134), class 3: 3.5-3.75 (n = 160), class 4: 4.0-4.25 (n = 142), and class 5: >4.25 (n = 66). Data were also categorized by calving order (primiparous and multiparous dams) and by calving season. The study was designed as a prospective cohort study. Variables with normal distribution were analyzed by the MIXED procedure of SAS, while binary outcomes were analyzed by the GLIMMIX procedure of SAS. Daughters from primiparous dams were born lighter (39.1 ± 0.42 vs. 41.4 ± 0.29 kg, p < 0.01), but they had the same weights as the daughters from multiparous cows at weaning (121.5 ± 1.67 vs. 120.4 ± 1.58 kg, p = 0.20). As expected, primiparous cows showed lower (p < 0.01) 305-day milk yields than multiparous ones: 8633 ± 363 vs. 10,761 ± 249 kg, respectively. Regarding the calving season, cows that calved in the winter were the most productive ones, and those that calved in the fall had lower milk yields (p = 0.01). Calves born in the winter were heavier at birth (p < 0.01), calved younger (p = 0.04), and produced more milk at first lactation (p = 0.03). The BCS class had an impact (p < 0.01) on calf birth weights; daughters from Class 1 cows (BCS < 3.0) were lighter (38.0 ± 1.0 kg) than the calves from Class 5 cows with a BCS > 4.25 (41.9 ± 0.57 kg). Calves from dams with a BCS < 3 (Class 1) had a 31.8% culling rate until weaning, while calves from cows with a BCS of 3.0-3.25 (Class 2) had a 9.6% culling rate (p = 0.12). These results suggest that maternal and environmental factors, such as calving season and parity, in addition to the dams' body condition score at calving, are associated with different offspring performances.

The present study aimed to verify the occurrence of displacement of the abomasum (DA) in dairy herds from a high-yielding dairy region of Southern Brazil. Data on breed, age, lactation number, days after calving, breeding system, number of animals, and number of lactating cows in the herd were obtained from 135 cases of DA. A total of 39 herds, and 6,454 cows, including 2,987 lactating cows from the municipality of Palmeira, Paraná State, were included in this study. The overall prevalence of DA was 2.09%, and occurrence of DA during lactation was 4.42%. Left displacement was more prevalent, with 94.07% of the cases. The mean number of lactations and age of the cows were 2.5±1.16 lactations and 50.9±18.5 months, respectively. DA occurred predominantly in the semi-intensive breeding system (68.1% of cases), during the first 4 weeks postpartum (84.4% of cases), in Black and White Holstein-Friesian cows (94.07% of cases), and during the winter (31.1% of cases). Cows with DA in high-yielding dairy farms in Southern Brazil were similar to cows from herds of high-milk-yielding regions of North America and Europe. Multiparous Black and White Holstein-Friesian cows showed the highest prevalence of DA during the transition period, mostly on the left side.


O presente estudo objetivou verificar a ocorrência do deslocamento de abomaso (DA) em rebanhos leiteiros de uma bacia leiteira de alta produção, na região sul do Brasil. Dados sobre raça, idade, números de lactação, número de dias após o parto, sistema de criação, número total de animais e de vacas em lactação foram obtidos de 135 casos de DA. Trinta e nove rebanhos, 6.454 vacas, sendo 2.987 vacas em lactação da cidade de Palmeira, Estado do Paraná, foram incluídos neste estudo. A frequência de DA foi de 2,09%, sendo que em vacas lactantes esta frequência foi de 4,42%. O deslocamento à esquerda foi o mais prevalente, com 94,07% dos casos. O número médio de lactações e a idade das vacas acometidas por DA foram 2,5±1,16 lactações e 50,9±18,5 meses, respectivamente. O DA ocorreu predominantemente em animais criados no sistema semi-intensivo (68,1% dos casos), durante as primeiras quatro semanas pós-parto (84,4% dos casos), em vacas Holandesa branco e preta (94,07% dos casos) e durante o inverno (31,1% dos casos). As vacas com DA em rebanhos leiteiros de alta produção do Sul do Brasil apresentaram similaridades com vacas de rebanhos de alta produção localizados em outros países da América do Norte e Europa. Vacas multíparas, Holandesa Preto e Branca e durante o período de transição tiveram a maior ocorrência de DA, sendo a maioria para o lado esquerdo.

O trabalho foi realizado em uma fazenda de exploração leiteira em Castrolanda, no município de Castro ­ PR. O sistema de manejo é free-stall, com 220 vacas da raça Holandesa com RHA305 de 6.740 litros. Foram coletadas amostras de sangue de 18 vacas de pré-parto, 4 dias antes da data prevista para o parto; e nos dias 4, 7 e 12 pós-parto, mediante punção venosa coccígea, utilizando-se SnapTest digital Ketovet®, constituindo 72 amostras, no período de fevereiro a maio de 2020. Em 4 vacas o BHB do sangue total apresentou-se acima do limite para cetose subclínica no quarto dia após o parto e 17 apresentaram-se acima do limite no sétimo dia, declinando em seguida, principalmente devido às intervenções clínicas. As médias e desvios-padrão foram: D - 4: 0,89 ± 0,257061; D 4: 1,05 ± 0,283279; D 7: 1,81 ± 0,456131; e D 12: 1,19 ± 0,437762. The study was carried out on a dairy farm in Castrolanda, in the county of Castro - PR. The management system is free stall, with 220 Holstein cows with RHA305 of 6,740 liters. Blood samples were collected from 18 pre-calving cows, 4 days before the expected date of calving; and on days 4, 7 and 12 postpartum, by means of coccygeal venipuncture, using SnapTest digital Ketovet®, constituting 72 samples, from February to May 2020. In 4 cows the BHB of whole blood was shown above the limit for subclinical ketosis on the fourth day after delivery and 17 presented above the limit on the seventh day, then declining mainly due to clinical interventions. The means and standard deviations were D -4: 0.89 ± 0.257061; D 4: 1.05 ± 0.283279; D 7: 1.81 ± 0.456131; and D 12: 1.19 ± 0.437762. The benefit of monitoring BHB was immediate clinical intervention, avoiding clinical ketosis and concomitant illnesses.(AU)


El estudio se realizó en una explotación lechera de Castrolanda, en la comarca de Castro - PR. El sistema de manejo es estabulación libre, con 220 vacas Holstein con RHA305 de 6.740 litros. Se recogieron muestras de sangre de 18 vacas pre- parto, 4 días antes de la fecha prevista de parto; y en los días 4, 7 y 12 postparto, mediante venopunción coccígea, utilizando SnapTest digital Ketovet®, constituyendo 72 muestras, desde febrero a mayo de 2020. En 4 vacas la BHB de sangre total se mostró por encima del límite para cetosis subclínica en el cuarto día después del parto y 17 presentaron por encima del límite en el séptimo día, disminuyendo después debido principalmente a intervenciones clínicas. Las medias y desviaciones estándar fueron D -4: 0,89 ± 0,257061; D 4: 1,05 ± 0,283279; D 7: 1,81 ± 0,456131; y D 12: 1,19 ± 0,437762. El beneficio de monitorizar la BHB fue la intervención clínica inmediata, evitando la cetosis clínica y las enfermedades concomitantes.(AU)

ABSTRACT: This study evaluated milk fat to protein ratio (FPR) in the first test-day after calving of dairy cows in Paraná State. Data from 257,847 first monthly test-days after calving of 114,162 cows were submitted to analysis after the data edition. Procedures MIXED, CORR and STEPWISE from SAS were used in the data analysis. In order to validate FPR, a herd with regular postpartum monitoring of blood β-hydroxybutyrate (BHB) was used to estimate the correlation with FPR. There was a strong positive correlation between FPR and fat content (r = 0.85; P < 0.01) and a weak negative correlation between FPR and protein content (r = - 0.23; P < 0.01). The regression equation that best fitted FPR was 1.1806 + 0.3304*%F - 0.3877*%P (R2 = 0.98), where the variable with the greatest influence was milk fat content (partial R2 = 0.72). Animals of 4th and 5th or more lactations had higher (P < 0.01) FPR, followed by animals of third, first, and second lactations. Jersey cows had higher (P < 0.01) FPR than Holsteins and Brown Swiss animals. Cows milked twice daily had higher (P < 0.01) FPR than animals milked three times daily. There were small positive correlations between milk FPR and blood BHB on days 4, 7 and 12 after calving (0.07, 0.13, and 0.14, respectively). In conclusion, milk fat content was reported to be the most important variable affecting FPR changes, but the milk FPR has limited value to evaluate hyperketonemia incidence during the transition period.


RESUMO: Este estudo teve como objetivo avaliar a relação gordura:proteína (RGP) no primeiro controle leiteiro no pós-parto de vacas no estado do Paraná. Dados de 257.847 controles de 114.162 vacas leiteiras, realizados no primeiro controle mensal após o parto foram submetidos à análise estatística após edição dos dados. Os procedimentos MIXED, CORR e STEPWISE do programa estatístico SAS 9.4 foram utilizados. A fim de validar a RGP, um rebanho com monitoramento regular de β-hidroxibutirato (BHB) no sangue foi utilizado para estimar a correlação com RGP. Observou-se uma correlação forte e positiva entre a RGP e teor de gordura no leite (r = 0,85; P < 0,01) e uma correlação fraca e negativa entre a RGP e o teor de proteína (r = -0,23; P < 0,01). A equação de regressão que melhor se ajusta à RGP foi 1,1806 + 0,3304*%G - 0,3877*%P (R2 = 0,98), na qual a variável com maior influência foi a porcentagem de gordura (R2 = 0,72). Animais de 4ª e 5ª lactações ou mais, apresentaram maior (P < 0,01) RGP, seguidos dos animais de terceira, primeira e segunda lactações, com menores RGP. Vacas da raça Jersey apresentaram maior (P < 0,01) RGP que vacas da raça Holandesa e Pardo-Suíça. Animais com duas ordenhas diárias apresentaram maior (P < 0,01) RGP que os animais ordenhados três vezes ao dia. The transition period in taurine dairy cows is widely reported in the literature. However, little is known about the metabolism of zebu animals and their crossbreeding with taurine breeds during this phase. Considering the importance of these breeds in tropical and subtropical regions, this study aimed to evaluate the feed intake, milk production and hepatic metabolism in Holstein, Gyr and Girolando-F1 (½ Holstein × ½ Gyr) heifers presenting high body condition score (BCS) during the transition period (prepartum weeks -2 and -1 and postpartum weeks +1 and + 3). Twelve heifers of each genetic group were used, totaling 36 animals. Variables considered were blood metabolites related to liver function, dry matter intake (DMI), body condition score (BCS), body weight (BW), milk yield (MY), and fat and protein concentrations in milk. Gyr heifers had the lowest concentrations of paraoxonase in weeks -2 (43.13 U/mL), -1 (62.10 U/mL) and + 3 (77.89 U/mL), albumin in week -1 (3.07 g/dL), and the highest concentration of non-esterified fatty acids (NEFA) in weeks -2 (1.35 mmol/L) and -1 (1.19 mmol/L). ß-hydroxybutyrate (BHB) and NEFA values were negatively correlated with prepartum DMI (-0.82 and -0.57, respectively), while paraoxonase was positively correlated to DMI (0.54). Gyr cows were more susceptible to inflammation despite having intermediate BCS and lower milk yield. Girolando-F1 animals showed the highest BCS among groups, although their hepatic metabolism had better results than Gyr cows. Holstein animals had lower lipomobilization and higher DMI and MY between breeds. These results suggest that the metabolism of zebu and crossbred cows does not react equally to the metabolism of Holstein cows concerning stress factors such as transition period and obesity. Therefore, the present study addresses an emerging theme that highlights the need for differentiated management during the transition period between the different breeds studied in order to ensure the maximum health and welfare of these animals.

The present study sought to evaluate the expression of PD-1 and CTLA-4 in blood T lymphocytes during the periparturient period and their relationship with uterine health in dairy cows, as determined by endometrial cytology and serum concentrations of ß-hydroxybutyrate (BHB) and non-esterified fatty acids (NEFAs), which are indicators of a negative energy balance. The second objective of this study was to investigate whether the expression of PD-1 and CTLA-4 in T lymphocytes is associated with the serum acute phase-protein haptoglobin concentration during the periparturient period. To address these objectives, 26 clinically healthy dairy cows were used. Peripheral blood was collected 14 days prepartum (T-14), at calving (T0), and 30 days postpartum (T30) to measure the expression of PD-1 and CTLA-4 in blood T lymphocytes by flow cytometry. In addition, we collected blood at T0, 10 days after parturition (T10), and T30 to obtain serum and determine the serum concentrations of NEFA, BHB, and Hp. Endometrial cytology was performed at T10, 20 days after parturition (T20), and T30. In the present study, we observed higher expression of CTLA-4 and PD-1 in T lymphocytes at parturition and in the prepartum period, which could indicate a relationship between these immune checkpoints and immunological tolerance during gestation in dairy cattle. In addition, a negative association between the expression of these immune checkpoints prepartum or at parturition and endometrial cytology at T20 and T30 was observed, indicating the negative implications of these immune response regulators in susceptibility to infections. This finding was further corroborated by the relationship between the serum concentration of haptoglobin and the expression of CTLA-4 and PD-1 by T lymphocytes. However, we did not observe a relationship between the indicators of negative energy balance, evaluated by the serum concentrations of BHB and NEFA, and the expression of the immune checkpoint markers studied. Thus, our findings represent an initial step that paves the way for the development of new therapeutic alternatives directed by the host with the objective of increasing the resistance of dairy cattle to infections in this critical period of life.

Our objective was to determine the effects of feeding 25-hydroxyvitamin D3 [25(OH)D3], or vitamin D3 (cholecalciferol) on plasma, mineral, and metabolite concentrations, mineral balance, mineral excretion, rumination, energy balance, and milk production of dairy cows. We hypothesized that supplementing 3 mg/d of 25(OH)D3 during the prepartum period would be more effective than supplementing vitamin D3 at the National Research Council (2001) levels to minimize calcium imbalance during the transition period and improve milk production of dairy cows. Forty multiparous, pregnant nonlactating-Holstein cows were enrolled in this study. Body weight, body condition score, parity, and milk yield in the previous lactation (mean ± standard deviation) were 661 ± 59.2, 3.46 ± 0.35, 1.79 ± 0.87, and 33.2 ± 6.43 kg/d, respectively. Cows were enrolled into the blocks (n = 20 for each treatment) at 30 d of the expected day of calving to receive an acidogenic diet (373 g/kg of neutral detergent fiber and 136 g/kg of crude protein, dry matter basis; -110 mEq/kg) associated with the treatments: (1) control (CTRL), vitamin D3 at 0.625 mg/d (equivalent to 25,000 IU of vitamin D3/d) or (2) 25(OH)D3 at 3 mg/d (equivalent to 120,000 IU of vitamin D3/d). All cows were fed with the base ration for 49 d after calving. Blood samples were taken on d 7, 0, 1, 2, 21, and 42, relative to calving. No effect of treatment was observed for prepartum dry matter intake or body condition score. A trend for increase of ionized Ca was observed for the cows fed 25(OH)D3, compared with the CTRL, but no effect of treatment was detected for total Ca or total P. Feeding 25(OH)D3 increased colostrum yield. The plasmatic concentration of 25-hydroxyvitamin D3 was increased with 25(OH)D3 supplementation. 25-Hydroxyvitamin D3 supplementation increased plasma glucose concentration at parturition. The postpartum dry matter intake was not influenced by treatments. Feeding 25(OH)D3 increases milk yield, 3.5% fat-corrected milk, and energy-corrected milk and improves milk yield components in early lactation. Overall, these findings suggest that 25(OH)D3 at 3 mg/d can improve the energy metabolism and lactation performance, compared with the current-feeding practice of supplementing vitamin D3 at 0.625 mg/d.

This study aimed to examine the effects of selenium (Se) and vitamin E (vitE) supplementation on blood cell counts and blood metabolite concentrations in goats and their kids. Fifteen Saanen goats (average age 6 years of age; average initial body weight of 70 ± 10 kg) and 21 ½ Saanen × ½ Pardo Alpine crossbred goat kids (average body weight of 3.70 ± 0.64 kg) were used. Animals were distributed in a completely randomized design with five replicates per diet for mother goats and seven for goat kids and randomly assigned into three groups in the following diets: CON, control basal diet; Se, inclusion of 3.2 mg of Se/kg DM; SevitE, inclusion of 3.2 mg Se/kg DM and 1145 IU/day vitE/kg DM. Effects of time were observed on red blood cells, hemoglobin, hematocrit, mean corpuscular volume, and mean corpuscular hemoglobin in goats and goat kids. Effects of time were observed on differential counts of leucocytes, lymphocytes, and monocytes in goat kids. Interaction was observed for high-density lipoprotein and total protein in goats and for triglycerides, beta-hydroxybutyrate (BHBA), and gamma-glutamyltransferase (GGT) in goat kids. Effects of time were observed on low-density lipoprotein, triglycerides, glucose, lactate, BHBA, non-esterified fatty acids (NEFA), creatinine, aspartate-aminotransferase, and GGT in goats and all blood metabolites in goat kids. Selenium, vitE, or association in the evaluated levels are not sufficient to change blood cell counts when supplied in diets for goats or goat kids. However, the effect of time or interaction between time and diets change the blood metabolite concentrations in the animals.

This study attempted to determine the associations between metabolic imbalances and lameness or hoof injuries in high-producing Holstein × Gir cows, and to determine whether the metabolic profile affects the occurrence of lameness. Eighty cows were followed from -60 to 60 days relative to calving and hoof injuries were reported on days -60, 7 and 60. Locomotion score (LS), body condition score (BCS), the concentrations of non-esterified fatty acids, ß-hydroxybutyrate, glucose, cholesterol, albumin, total protein, blood urea nitrogen (BUN), calcium, phosphorus, and magnesium, and the activity of aspartate aminotransferase were determined at days -42, -21, -7, 0, 7, 21 and 42. The McNemar and Chi-square tests were used to compare frequencies of lameness and hoof injuries over time and to verify the associations between lameness, BCS, hoof injuries, and metabolic disorders. Two-way repeated measures ANOVA was used considering groups (non-lame × lame cows) and variations of BCS and metabolites over time. Lameness and hoof injuries increased between days -60 (20% and 66.3%) and 60 (44.7% and 98.6%). Excessive postpartum loss of BCS (P=0.017) and subclinical hypocalcemia (P=0.012) were associated with lameness on day 60. In general, the metabolic profile did not differ between lame and non-lame cows but cholesterol, albumin, BUN and magnesium concentrations were higher in non-lame cows. The postpartum decrease in BCS can affect the occurrence of lameness, and the metabolic profile of lame cows shows little difference from that of non-lame cows.


Este estudo objetivou verificar as associações entre desequilíbrios metabólicos e claudicação ou lesões nos cascos em vacas mestiças Holandesa × Gir de alta produção e determinar se o perfil metabólico afeta a ocorrência de claudicação. Oitenta vacas foram acompanhadas de -60 a 60 dias em relação ao parto e as lesões nos cascos foram avaliadas nos dias -60, 7 e 60. O escore de locomoção (EL), o escore de condição corporal (ECC), as concentrações de ácidos graxos não esterificados, ß-hidroxibutirato, glicose, colesterol, albumina, proteína total, nitrogênio ureico no sangue (BUN), cálcio, fósforo e magnésio e a atividade da aspartato aminotransferase foram determinados nos dias -42, -21, -7, 0, 7, 21 e 42. Os testes de McNemar e de Qui-quadrado foram empregados para comparar as frequências de claudicação e de lesões nos cascos ao longo do tempo e para verificar as associações entre claudicação, ECC, lesões nos cascos e distúrbios metabólicos. A análise de variâncias de medidas repetidas bifatorial foi usada considerando-se grupos (vacas com claudicação × vacas sem claudicação) e variações de BCS e de metabólitos ao longo do tempo. A claudicação e as lesões nos cascos aumentaram entre os dias -60 (20% e 66,3%) e 60 (44,7% e 98,6%). A perda excessiva de ECC no pós-parto (P=0,017) e a hipocalcemia subclínica ao parto (P=0,012) foram associadas com claudicação no dia 60. Em geral, o perfil metabólico não diferiu entre vacas com e sem claudicação, mas as concentrações de colesterol, albumina, BUN e magnésio foram maiores em vacas sem claudicação. A redução do ECC no periodo pós-parto pode afetar a ocorrência de claudicação, e o perfil metabólico das vacas claudicantes apresenta pouca diferença em relação ao das vacas não claudicantes.

Cows are affected by environmental factors associated with warm weather conditions; however, little is known about the effect of shade access especially during the prepartum period of dairy cows in temperate regions. This study assessed the effect of shade on the behavior (lying, rumination, feeding, and drinking), body fat mobilization, and health status of outdoor-housed dairy cows during the prepartum period under temperate summer conditions. During the 3 weeks prior to calving, 24 multiparous Holstein cows were grouped (4 cows/group) and assigned to either an open corral without shade or with access to shade until calving. We daily measured shade use, lying, rumination, feeding, and drinking behavior. Weekly, prepartum non-esterified fatty acids (NEFA) and postpartum b-hydroxybutyrate (BHB) concentrations were measured. Clinical examination was periodically performed individually until 21 postpartum days. Shade use averaged 45.6, 46.0, and 19.8% during the hottest hours of the day (11-18 h) in weeks 3, 2, and 1 prior to calving, respectively. Shaded cows had higher values for rumination time and feeding time during the morning but spent less time drinking during the warmest hours than unshaded cows. NEFA and BHB concentrations and clinical diseases were similar between both treatments. These findings suggest that under temperate summer conditions the access to an artificial shade is an important resource, observing beneficial effects mainly on behavioral variables.
