Abstract Melon production in the Brazilian semi-arid region is subject to the use of marginal waters with high salinity. However, the use of regulators and bioactivators in seed treatment can mitigate the harmful effects of salts in irrigation water. In this context, the objective was to evaluate the effect of pre-germination treatments with plant regulators and bioactivator in melon seeds for the production of seedlings irrigated with biosaline water from fish farming effluent. For this, two trials with the Goldex and Grand Prix hybrids were carried out separately. A completely randomized design was used in a 4 × 3 factorial scheme (pre-germination treatments × water dilutions). In addition to the control, the seeds were treated with salicylic and gibberellic acids and thiamethoxam. The waters used for irrigation were local-supply water, fish farming effluent (biosaline water) and these diluted to 50%. Physiological and biochemical analyses were performed for fourteen days. Biosaline water (5.0 dS m-1) did not affect the emergence of Goldex melon seedlings, but compromised the establishment of the Grand Prix cultivar. Seed pre-treatments with salicylic and gibberellic acids attenuate the effects of water salinity and promote growth modulations, resulting in more vigorous melon seedlings.


Resumo A produção de meloeiro no semiárido brasileiro está sujeita a utilização de águas marginais com salinidade elevada. Entretanto, a utilização de reguladores e bioativadores no tratamento de sementes podem mitigar os efeitos nocivos dos sais na água de irrigação. Nesse sentido, objetivou-se avaliar o efeito de tratamentos pré-germinativos com fitorreguladores e bioativador em sementes de melão para a produção de mudas irrigadas com água biossalina de efluente de piscicultura. Para isso, dois ensaios com os híbridos Goldex e Grand Prix foram realizados separadamente. Utilizou-se delineamento inteiramente casualizado em esquema fatorial 4 × 3 (tratamentos pré-germinativos × diluições de água). Além do controle, as sementes foram tratadas com os ácidos salicílico e giberélico, e tiametoxam. As águas utilizadas para irrigação foram a de abastecimento local, efluente de piscicultura (água biossalina) e estas diluídas a 50%. Durante quatorze dias foram realizadas as análises fisiológicas e bioquímicas. A água biossalina (5,0 dS m-1) não afetou a emergência de plântulas de meloeiro Goldex, mas prejudicou o estabelecimento da cultivar Grand Prix. Os pré-tratamentos de sementes com os ácidos salicílico e giberélico atenuam os efeitos da salinidade da água e promovem modulações no crescimento, proporcionando mudas de meloeiro mais vigorosas.

Huanglongbing (HLB), the most serious disease in citriculture, is caused by the bacteria Candidatus Liberibacter spp., which is transmitted by the Asian citrus psyllid (ACP) Diaphorina citri. HLB is mainly controlled with insecticides, necessitating the development of alternative methods, e.g., the use of trap plants such as curry leaf Bergera koenigii, which is highly attractive to the ACP. We evaluated the effects of the main systemic insecticides used by citrus growers, applied via drench to adults of D. citri on the curry leaf tree. We tested the persistence of three pesticides: thiamethoxam, thiamethoxam + chlorantraniliprole, and imidacloprid in protected cultivation and the field condition at 7, 14, 28, 42, 56, 70, 98, and 154 days after the application. Different concentrations of insecticides containing the active ingredient thiamethoxam were tested on adults to determine the LC10 and LC50. Finally, we assessed the sublethal effects on the oviposition and development of D. citri. The insecticides controlled the adults for long periods. However, in the field experiment, from 42 days after application there was a decrease in mortality caused by pesticides applied via drench, while in the protected cultivation, mortality did not decline until the last day of evaluation. The median lethal concentration (LC50) for thiamethoxam was 0.031 g of active ingredient per plant, and for thiamethoxam in a mixture, the LC50 was 0.028 g a.i. per plant. In the experiment with sublethal doses, D. citri did not oviposit on the treated plants. Our findings suggest that the attract-and-kill system using the curry leaf tree and systemic insecticides is effective for the control of D. citri and contributes to the integrated management of HLB.

During sowing using pneumatic machinery, dust may be abraded from pesticide-treated seed and contaminate adjacent bee-attractive off-crop areas. This study quantified the risk to native bees of dust released during sowing of Brazilian crop seeds treated with a thiamethoxam formulation (Cruiser 350FS). To address toxicity to native bees, adult acute contact LD50 data for thiamethoxam were collated from the literature, a species sensitivity distribution generated, and the HD5 calculated. The LD50 HD5 was used to refine the default safety factor applied to the honeybee acute contact LD50 from 10 to 5.45 for thiamethoxam. Crop-specific abraded dust data (Heubach dust and Heubach AI) were generated for seeds treated with Cruiser 350FS sourced from on-farm and industrial facilities. The mean Heubach dust levels was ranked as cotton = maize > sunflower = soybean > drybean. There was no correlation between the measured residues of thiamethoxam (Heubach AI) and those estimated in dust based on the thiamethoxam content of Cruiser 350FS. A hazard quotient (HQ) for each crop (based on application rate, the default dust deposition factor, and the honeybee contact LD50/10) identified risks during sowing for all crops. Refinement of the application rate with the measured 90th percentile Heubach dust (assuming 100% thiamethoxam) resulted in sowing of industrially treated soybean and on-farm treated cotton being identified as risks. Further refinement using either the measured 90th percentile Heubach AI or the acute contact LD50 (HD5 ) resulted in sowing of all crops treated with Cruiser 350FS as being identified as low risk. Similar high quality seed treatment should be demonstrated for other formulations containing insecticides with high toxicity to bees. Data on dust drift from machinery and crops more representative of those in Brazil may allow further refinement of the default dust deposition value of 17% used in this study. Thiamethoxam (TMX) is a systemic neonicotinoid that acts as a partial agonist of the nicotinic acetylcholine receptors (nAChRs). However, target species have shown resistance to formulations based on such neonicotinoids, which can also be expected for non-target insects. This research aimed to study the effects of a formulation based on TMX [Cruiser® 350 FS (CRZ)] on the life traits of Chironomus xanthus filial generation (F1) and compare it with the parental generation (P). Environmentally relevant concentrations of CRZ significantly decreased larvae growth P generation , also slowing and decreasing their emergence. Larvae of the F1 generation were less sensitive than their parents, suggesting that the progeny were able to thrive and perform basic physiological functions better than the parental generation. Our results highlight that insect resistance to neonicotinoids may be associated with the better performance of the filial generation, which is related to the change in affinities of the active ingredient for the sub-units constituting the nAChRs subtypes of F1 organisms, inherited from P organisms that were able to survive and reproduce. Moreover, further studies using biochemical and omics tools should be performed to disentangle the specific changes occurring at the nAChRs throughout insect development.

Whitefly, Bemisia tabaci cause important losses for many crops especially tomato, therefore farmers often use many chemical insecticides which had dangerous side effects on human beings and environment, thus there are need to determine the residues of these chemicals in plant . The dissipation of thiamethoxam in tomato leaves and fruits followed the first order kinetics; Half-lives were 2.91 and 3.15 days for fruits and leaves, respectively. The residues of thiamethoxam were determined using a QuEChERS method with HPLC-DAD and it was 0.14 mg/ kg in fruits after 7 days of treatment and less than the maximum residue limit (MRL 0.2 mg/kg. The residue in leaves was more than in fruits. Seedlings produced from treated seeds were more resistant to different stages of the whitefly population and protect tomato plants from whitefly infestation for 6-10 weeks after transplanting. The three sprays of thiamethoxam increased the reduction percentage of B. tabaci stages and the highest increase was recorded after the third spray in egg, nymph, and adult stages. It could be recommend that Thiamethoxam is considered an unconventional insecticide appropriate for B. tabaci control and safe for humans and the environment.


Bemisia tabaci, também conhecida como "Mosca-branca" causam perdas importantes para muitas culturas, especialmente tomate, por isso, os agricultores costumam usar muitos inseticidas químicos que têm efeitos colaterais prejudiciais aos seres humanos e ao meio ambiente. Nesse sentido, observa-se a necessidade de determinar os resíduos desses produtos químicos na planta. A dissipação do tiametoxam nas folhas e frutos do tomateiro seguiu a cinética de primeira ordem: As meias-vidas foram de 2,91 e 3,15 dias para frutos e folhas, respectivamente. Os resíduos de tiametoxam foram determinados pelo método QuEChERS com HPLC-DAD e foi de 0,14 mg/kg nos frutos após 7 dias de tratamento e inferior ao limite máximo de resíduos (LMR 0,2 mg/kg). O resíduo nas folhas foi maior do que nas frutos. Mudas produzidas a partir de sementes tratadas mostraram-se mais resistentes a diferentes estágios da população de mosca-branca e protegeram os tomateiros da infestação por mosca-branca por 6-10 semanas após o transplante. As 3 pulverizações de tiametoxam aumentaram a porcentagem de redução dos estágios de B. tabaci e o maior aumento foi registrado após a terceira pulverização nos estágios de ovo, ninfa e adulto, assim, é possível considerar o tiametoxam como um inseticida não convencional apropriado para o controle de B. tabaci e seguro para humanos e meio ambiente.

The study aimed to develop an electrochemical sensor based on glassy carbon, mixed oxide (SiO2/TiO2/Sb2O5), and carbon black. The material was synthesized, characterized, and used to determine thiamethoxam in raw honey and water. The morphologic structure and electrochemical performance of the sensor was characterized by scanning electron microscopy and cyclic voltammetry. Differential pulse voltammetry with a concentration of 0.1 mol L-1 of Britton-Robinson buffer at pH 7.0 allowed the generation of a method to determine thiamethoxam in a linear range of 0.25 to 100.5 µmol L-1 and with a limit of detection of 0.012 µmol L-1. The system efficiently quantified traces of thiamethoxam in raw honey and tap water samples. The modified sensor did not present interferences of K+, Na+, Ca2+, Mg2+, glyphosate, imidacloprid, and carbendazim. In addition, the device showed good recovery values for thiamethoxam when applied directly to honey and water samples without any treatment, presenting an electrochemical sensor to monitor real-time hazardous substances in environmental and food matrices.

In the present study, a method for the determination of residues of the neonicotinoid insecticides thiamethoxam and imidacloprid in the stingless bee Melipona scutellaris Latreille (1811) was optimized through a factorial design, tested using green metrics, and then applied to exposed bees. It combines the extraction with a modified quick, easy, cheap, effective, rugged, and safe method and the determination by liquid chromatography-tandem mass spectrometry analysis. Different parameters such as the mass of the sample, dispersive sorbents, and elution solvents were assessed. Method validation parameters were checked and include sensitivity, specificity, and linearity. The limit of quantification of 0.0025 µg g-1 was obtained for both insecticides, where accuracy was 94%-100% with satisfactory intraday and interday precisions (relative standard deviation <10%). The qualified method was applied to orally and topically exposed bee samples, and the results indicated that it is suitable for the determination and quantification of neonicotinoid pesticide residues in this species. Moreover, green analytical metrics like the National Environmental Methods Index, Eco Scale score, high-performance liquid chromatography with an environmental assessment tool (HPLC-EAT), waste generation, and amount of sample were compared with methods described in the literature involving neonicotinoid analysis in honeybees. As a result, the present study displayed the highest Eco Scale score and HPLC-EAT score and the second smallest amount of sample and waste generated. Thus, the method meets green analytical metrics more than other methods. In this sense, besides the application, the multicriteria decision analysis tool employed suggests that this is a good option as a green analytical method. Exposure to sublethal concentrations can have adverse effects on certain individuals, but, can also favor survival and reproduction of others. This study aimed to evaluate the effects of exposing Drosophila suzukii (Matsumura, 1931) (Diptera: Drosophilidae) adults to LC50 of spinetoram and lambda-cyhalothrin and field rate of thiamethoxam on their mating, reproductive output and longevity. The LC50 for lambda-cyhalothrin and for spinetoram were 7.83 and 32.91 ng a.i./cm2, respectively. Thiamethoxam, in the concentrations tested, was not toxic to D. suzukii, therefore, we use the recommended concentration for strawberry, 231.25 ng a.i./cm2, in the sublethal tests. Insects exposed to LC50 of lambda-cyhalothrin were more likely to mate, but presented shorter copula. Sublethal exposure to thiamethoxam lead to an increase in total fecundity. Among the insecticides tested, spinetoram presented the least expressive effects. Longevity was not affected by any of the tested insecticides. Results presented in this study are relevant to D. suzukii management since the stimulatory effects shown on this pest species can collaborate to outbreaks and insecticide resistance.

Sand was studied as a solid support in ultrasound-assisted matrix solid-phase dispersion (UA-MSPD) for the extraction of different pesticide classes, including organophosphates, carbamates, triazoles and pyrethroids from fruits and vegetables, with determination by GC-MS and LC-MS/MS. The performance of sand was compared with that of different types of classic solid supports and alternative natural materials from renewable sources. The best results were obtained using 0.5 g sample, 1 g sand as a solid support, 20 mg activated charcoal and 5 mL ethyl acetate as elution solvent. Recoveries ranged from 55 to 140% with an RSD ≤ 20%. LOQs varied from 0.005 to 0.5 mg kg-1 for all analytes. Thiamethoxam, captan, chlorpyrifos, dimethoate and pyrimethanil were found in strawberry samples at concentrations from 0.01 to 0.06 mg kg-1. Acephate and tebuconazole were found in a tomato sample at concentrations of 0.45 and 0.30 mg kg-1, respectively. The method developed was efficient, simple, cheap, robust, and environmentally friendly.

With the aim of developing tools for simultaneously managing Rhynchophorus palmarum and the coconut stem bleeding disease, we evaluated the effects of the insecticide thiamethoxam and the fungicide cyproconazole on R. palmarum larvae under laboratory conditions. Early-instar and intermediate-instar larvae were fed on an artificial diet containing 0.1% concentration of insecticide, fungicide or a mixture of both. Larval mortality was assessed daily during the 48 hours after the application of the treatments. Mean mortality values were subjected to analysis of variance and compared by Tukey's test (p ≤ 0.05). Thiamethoxam caused mortality of 100% of early-instar and 90% of intermediate-instar larvae. In contrast, mortality was significantly lower in the cyproconazole treatment (60% for early-instar and 0% for intermediate-instar larvae) and the control (0% mortality for both treatments). The insecticide/fungicide mixture was equally effective (100% for early-instar and 86.67% for intermediate-instar larvae) to the insecticide only treatment. Differences in mortality between early-instar and intermediate-instar larvae were significant only for the thiamethoxam and cyproconazole treatments. These results indicate that, for the doses used here, thiamethoxam is toxic to both early-instar and intermediate-instar larvae, while cyproconazole is toxic just to early-instar larvae. Moreover, cyproconazole does not increase the toxicity of thiamethoxam. Early-instar larvae are more sensitive to thiamethoxam and cyproconazole than intermediate-instar larvae. Com o objetivo de desenvolver ferramentas para o manejo simultâneo de Rhynchophorus palmarum e da resinose do coqueiro, avaliou-se o efeito do inseticida tiametoxam e do fungicida ciproconazol sobre larvas de R. palmarum em laboratório. Larvas de ínstares iniciais e intermediários foram alimentadas com dieta artificial contendo 0,1% do inseticida, do fungicida, ou da mistura de ambos. A mortalidade larval foi avaliada diariamente, durante 48 horas, e as médias foram submetidas à análise de variância e comparadas entre si pelo teste de Tukey (p ≤ 0,05). O tiametoxam causou mortalidade de 100% às larvas de ínstares iniciais e de 90% às de intermediários. Em contraste, a mortalidade foi significativamente menor no tratamento ciproconazol (60% para larvas de ínstares iniciais e 0% para as de intermediários) e no controle (0% de mortalidade para ambos). A mistura inseticida/fungicida foi igualmente eficiente (100% para larvas de ínstares iniciais e 86,67% para as de intermediários) ao inseticida sozinho. Diferenças na mortalidade entre as larvas de ínstares iniciais e intermediários foram significativas somente para os tratamentos tiametoxam e ciproconazol. Tais resultados indicam que, nas doses avaliadas, o tiametoxam é tóxico às larvas de ínstares iniciais e intermediários, enquanto o ciproconazol é tóxico somente às de ínstares iniciais. Além disso, o ciproconazol não aumenta a toxicidade do tiametoxam. Larvas de ínstares iniciais são mais sensíveis a tiametoxam e ciproconazol que as de ínstares intermediários. Conclui-se que a mistura tiametoxam + ciproconazol pode ser eficiente no manejo de R. palmarum e de doenças fúngicas associadas a essa praga em coqueiros.(AU)

Com o objetivo de desenvolver ferramentas para o manejo simultâneo de Rhynchophorus palmarum e da resinose do coqueiro, avaliou-se o efeito do inseticida tiametoxam e do fungicida ciproconazol sobre larvas de R. palmarum em laboratório. Larvas de ínstares iniciais e intermediários foram alimentadas com dieta artificial contendo 0,1% do inseticida, do fungicida, ou da mistura de ambos. A mortalidade larval foi avaliada diariamente, durante 48 horas, e as médias foram submetidas à análise de variância e comparadas entre si pelo teste de Tukey (p ≤ 0,05). O tiametoxam causou mortalidade de 100% às larvas de ínstares iniciais e de 90% às de intermediários. Em contraste, a mortalidade foi significativamente menor no tratamento ciproconazol (60% para larvas de ínstares iniciais e 0% para as de intermediários) e no controle (0% de mortalidade para ambos). A mistura inseticida/fungicida foi igualmente eficiente (100% para larvas de ínstares iniciais e 86,67% para as de intermediários) ao inseticida sozinho. Diferenças na mortalidade entre as larvas de ínstares iniciais e intermediários foram significativas somente para os tratamentos tiametoxam e ciproconazol. Tais resultados indicam que, nas doses avaliadas, o tiametoxam é tóxico às larvas de ínstares iniciais e intermediários, enquanto o ciproconazol é tóxico somente às de ínstares iniciais. Além disso, o ciproconazol não aumenta a toxicidade do tiametoxam. Larvas de ínstares iniciais são mais sensíveis a tiametoxam e ciproconazol que as de ínstares intermediários. Conclui-se que a mistura tiametoxam + ciproconazol pode ser eficiente no manejo de R. palmarum e de doenças fúngicas associadas a essa praga em coqueiros.(AU)

The beehive as a quantitative monitor of pesticide residues applied over a soybean crop was studied through a semi field experiment of controlled exposure of honey bees to pesticides in macro tunnels. The distribution within exposed beehives of pesticides commonly used in soybean plantation, was assessed. Residue levels of insecticides in soybean leaves, honey bees, wax, honey and pollen were analyzed. The transference from pesticides present in the environment into the beehive was evidenced. The obtained results allow relating pesticide concentrations present in the environment with traces found in foraging bees. Therefore, pesticide transference ratios could be calculated for each detected compound (acetamiprid, imidacloprid and thiamethoxam) which showed a linear inverse trend with their 1-octanol/water partition coefficient (Kow). The least transferred pesticide to the hive (acetamiprid) has the highest vapor pressure (Vp). This study gives new insights on the usefulness of monitoring the environment through beehives aiming to evaluate if agroecosystems remain sustainable. It also contributes to generate valuable information for model building aiming to predict environmental quality through beehive's analysis.

The development of effective baits to control the Argentine ant, Linepithema humile (Mayr), has been problematic because foragers prefer sweet liquids, while many toxicants are insoluble in water and liquid baits are generally difficult to deliver. The incorporation of thiamethoxam and sucrose solutions into a water-absorbing polyacrylamide hydrogel provides a unique and novel carrier and method of application for liquid baits. Formulations of thiamethoxam affected the size of the hydrogels, and sucrose solutions containing 0.0003% technical thiamethoxam provided hydrogels as large as those made with 25% sucrose solution or deionized water. Concentrations of thiamethoxam as low as 0.000075% in the hydrogels provided 50% kill of workers within 3 d in a laboratory setting. In small colony studies, baiting with 0.00015 and 0.000075% thiamethoxam hydrogels provided 100% mortality of workers and queens within 8 d. An enzyme-linked immunosorbent assay indicated that thiamethoxam was absorbed into the interior of the polyacrylamide matrix. The water loss rates of the hydrogels were dependent upon the relative humidity. Polyacrylamide hydrogels with >50% water loss were less attractive to ants. Field studies in highly infested areas indicated that concentrations of 0.0006 or 0.0018% thiamethoxam were more effective than 0.00015%. Hydrogels may provide a cost-effective alternative to providing aqueous baits to control Argentine ants.

O objetivo do trabalho foi avaliar o efeito de quatro tratamentos de sementes: água (T1), carbendazim + thiram (T2), fipronil + tiofanato metílico + piraclostrobina (T3) e abamectina + thiamethoxam + fludioxonil + mefenoxan + thiabendazole (T4) sobre parâmetros de crescimento de plantas de soja submetidas a dois regimes hídricos (presença e ausência de estresse hídrico) estabelecidos logo após a emergência das plântulas. O experimento foi conduzido em casa de vegetação e campo. Na casa de vegetação, nos tratamentos sem estresse hídrico, foram feitas quatro irrigações por dia e, nos tratamentos com estresse hídrico, foi realizada uma irrigação a cada três dias. Neste ensaio, foi avaliada a estatura de plantas, comprimento radicular, volume radicular, massa seca da parte aérea, massa seca radicular e área foliar. No ensaio de campo, o estresse hídrico foi estabelecido pela construção de túneis baixos de polietileno que impediram o fornecimento de água pela irrigação e chuva. No campo, foram avaliados estatura de planta, teor relativo de clorofila, emergência, ataque de lagarta-elasmo e rendimento de grãos. Nos dois ambientes, as plantas foram mantidas sob estresse até 28 dias após a emergência. Em casa de vegetação, todos os parâmetros foram influenciados pelo tratamento de sementes na presença e ausência de estresse hídrico. No campo, o tratamento com fipronil + tiofanato metílico + piraclostrobina (T3) e abamectina + thiamethoxam + fludioxonil + mefenoxan + thiabendazole (T4) proporcionaram maior estatura de planta e teor relativo de clorofila em ambos os regimes hídricos e maior emergência e rendimento de grãos em plantas sem e com restrição hídrica, respectivamente. Conclui-se que o tratamento de sementes com esses produtos promove alterações benéficas nas plantas, aumentando sua tolerância ao estresse hídrico, com efeito positivo sobre o rendimento de grãos da soja.

It was concluded that seed treatment with these products promote benefit changes in plant, increasing its tolerance to hydric stress, with positive effect on soybean grain yield.

The efficacy of the Metarhizium anisopliae strain ARSEF 6911 was determined in the laboratory and field against two sugarcane pests, Microtermes obesi Holmgren and Odontotermes obesus Rambur (Termitidae: Isoptera). The susceptibility of both termite species to different conidial suspensions (1 × 10(10), 1 × 10(8), 1 × 10(6) and 1 × 10(4) conidia/ml) was determined in laboratory. All conidial suspensions were able to induce mortality. Termite mortality caused by the fungal suspensions was dose dependent. There were no significant differences in the LT50 values between species. Field evaluation of M. anisopliae alone or in combination with diesel oil and thiamethoxam was carried out in two growing seasons (autumn 2005 and spring 2006) at two sites located in Punjab, Pakistan. Dipping the sugarcane setts in these suspensions was tried to determine their effects on germination and percentage of bud damage to sugarcane setts. All treatments significantly reduced termite infestation compared to the untreated control. The combined treatment of M. anisopliae and diesel oil significantly reduced insect damage by attaining higher germination > 55 percent and lower bud damage < 5.50 percent at both sites in both seasons. The results suggest that the application of M. anisopliae and diesel oil in combination might be a useful treatment option for the management of termites in sugarcane.
