Household water contamination at point of use depends on human, animal and environmental factors embodying all aspects of a One Health approach. This study investigated the association between household factors, the presence of thermotolerant coliform, and the presence of antibiotic resistant bacteria in drinking water among 314 households with children under 5 in Cajamarca, Peru. This study analysed data from a baseline sampling of a randomized controlled trial, including household surveys covering household water management and factors such as household animals, as well as microbiological data from samples collected from drinking water. Data were analysed using generalized linear models. Drinking water samples collected from narrow-mouthed containers were less likely to be contaminated than samples collected from the faucet (OR = 0.55, p = 0.030) or wide mouthed containers. The presence of thermotolerant coliform was associated with owning farm birds, which increased the proportion of contamination from 42.2% to 59.1% (OR = 1.98, p = 0.017) and with animal waste observed in the kitchen area, which increased the prevalence of contamination from 51.4% to 65.6% (OR = 1.80, p = 0.024). Resistance to any antibiotic was higher among pig owners at 60%, relative to non-pig owners at 36.4% (OR = 1.97, p = 0.012) as well as households with free-roaming animals in the kitchen area at 59.6% compared to households without free-roaming animals at 39.7% (OR = 2.24, p = 0.035). Recent child antibiotic use increased the prevalence of trimethoprim-sulfamethoxazole resistance among E. coli isolates to 22.3% relative to 16.7% (OR = 3.00, p = 0.037). Overall, these findings suggest that water storage in a secure container to protect from in-home contamination is likely to be important in providing safe drinking water at point of use. In addition, transmission of thermotolerant coliform and AMR between domestic animals and human drinking water supplies is likely. Further research should explore transmission pathways and methods to support safe drinking water access in multi-species households.

The aimed of the study was to evaluate the quality of raw milk from some suppliers in rural and urban areas and to test the hypothesis that it is unsafe for direct consumption. The study was conducted with suppliers in rural and urban areas in Presidente Médici municipality, Rondônia state. The method of the Ministério da Agricultura, Pecuária e Abastecimento (MAPA), was used, which consists of titrating a portion of the sample with a sodium hydroxide solution of known concentration, using phenolphthalein as an indicator. According to Brazilian legislation and literature, stability of Alizarol, foreign substances, Chloride, Hypochlorite, Hydrogen Peroxide and Colostrum were analyzed. Microbiological analysis, standard plate count (SPC) and quantification of thermotolerant coliforms were also performed using the most probable number (MPN) technique. Values above the limits established by legislation were found, mainly for temperature and titleable acidity, in addition to stability results against Alizarol and the presence of colostrum. There were high microbial contents with regard to SPC, and, in this case, the presence of pathogenic microorganisms cannot be disregarded. The presence of thermotolerant coliforms may indicate probable contact of milk with feces. Therefore, it is considered that the samples analyzed here are not safe for ingestion in the raw form.(AU)


O objetivo do estudo foi avaliar a qualidade do leite cru de alguns fornecedores da zona rural e urbana e tes-tar a hipótese de que não são seguros para o consumo direto. O estudo foi conduzido em fornecedores da zona rural e urbana do município de Presidente Médici, estado de Rondônia. Empregou-se o método do Ministério da Agricultura Pecuária e Abastecimento (MAPA), que consiste na titulação de uma porção da amostra por uma solução hidróxido de sódio de concentração conhecida, utilizando como indicador a fenolftaleína. Conforme a legislação brasileira e a lite-ratura, foram analisados estabilidade do Alizarol, substâncias estranhas, Cloreto, Hipoclorito, peróxido de Hidrogênio e Colostro. Foram realizadas também análises microbiológicas, contagem Padrão em Placa (CPP) e quantificação dos coliformes termotolerantes através da técnica do número mais provável (NMP). Foram encontrados valores acima dos limites estabelecidos pela legislação, principalmente para temperatura e acidez titulável, além dos resultados de estabili-dade frente ao Alizarol e da presença de colostro. Houve altos teores microbianos no que se refere à CPP, e, nesse caso, a presença de microrganismos patogênicos não pode ser desconsiderada. A presença de coliformes termotolerantes pode indicar provável contato do leite com fezes. Portanto, considera-se que as amostras aqui analisadas não são seguras para ingestão na forma crua.(AU)

Abstract Foodborne diseases are common illnesses caused by the consumption of food contaminated with microorganisms, such as viruses, fungi, bacteria, and protozoa. Every year, 600 million people become ill and 420,000 people die as a result of consuming contaminated food. Therefore, food safety is an important issue. In this study, samples of homemade spiced mayonnaise and self-serve acai sold in the city of Araguaína, Tocantins, Brazil were analyzed for microbiological contaminants. Acai was collected from 10 stores, one sample from each store, and tested for mold, yeast, and coliforms, as well as coliform identification and total and thermotolerant coliform counts. Mayonnaise was collected from 20 snack bars, one sample from each. These samples were inoculated on MacConkey and Salmonella Shigella agar plates, and the plates were analyzed for growth. Salmonella spp. were detected in some Mayonnaise samples, and coliforms were detected in all acai samples; 60% of samples had thermotolerant coliforms, and only 40% were within the limits established by ANVISA. The collected samples of mayonnaise and acai were contaminated with molds and yeasts above the established limit of 103 CFU/g. Thus, the analyzed mayonnaise and acai samples were contaminated and unfit for consumption, demonstrating the importance of hygienic-sanitary measures in food handling.


Resumo As doenças transmitidas por alimentos são doenças comuns causadas pelo consumo de alimentos contaminados com microrganismos, como vírus, fungos, bactérias e protozoários. Todos os anos, 600 milhões de pessoas ficam doentes e 420.000 pessoas morrem como resultado do consumo de alimentos contaminados. Portanto, a segurança alimentar é uma questão importante. Neste estudo, amostras de maionese temperada caseira e açaí self-service vendidas na cidade de Araguaína, Tocantins, Brasil foram analisadas quanto a contaminantes microbiológicos. O açaí foi coletado em 10 lojas, uma amostra de cada loja, e testado para mofo, levedura e coliformes, bem como identificação de coliformes e contagem de coliformes totais e termotolerantes. A maionese foi coletada de 20 lanchonetes, sendo uma amostra de cada. Essas amostras foram inoculadas em placas de ágar MacConkey e Salmonella Shigella, e as placas foram analisadas quanto ao crescimento. Salmonela spp. foram detectados em algumas amostras de maionese e coliformes foram detectados em todas as amostras de açaí; 60% das amostras apresentaram coliformes termotolerantes, e apenas 40% estavam dentro dos limites estabelecidos pela ANVISA. As amostras coletadas de maionese e açaí estavam contaminadas com bolores e leveduras acima do limite estabelecido de 103 UFC/g. Assim, as amostras de maionese e açaí analisadas estavam contaminadas e impróprias para o consumo, demonstrando a importância das medidas higiênico-sanitárias na manipulação de alimentos.

Lipases are enzymes that hydrolyze triglycerides to fatty acids and glycerol. A typical element in lipases is a conserved motif of five amino acids (the pentapeptide), most commonly G-X-S-X-G. Lipases with the pentapeptide A-X-S-X-G are present in species of Bacillus, Paucimonas lemoignei, and the yeast Trichosporon asahii; they are usually thermotolerant and solvent resistant. Recently, while searching for true lipases in the Trichoderma harzianum genome, one lipase containing the pentapeptide AHSMG was identified. In this study, we cloned from T. harzianum strain B13-1 the lipase ID135964, renamed here as ThaL, which is 97.65% identical with the reference. We found that ThaL is a lid-containing true lipase of cluster III that belongs to a large family comprising highly conserved proteins in filamentous fungi in the orders Hypocreales and Glomerellales, in which predominantly pathogenic fungi are found. ThaL was expressed in conidia, as well as in T. harzianum mycelium, where it was cultured in liquid minimal medium. These results-together with the amino acid composition, absence of a signal peptide, mitochondrial sorting prediction, disordered regions in the protein, and lineage-specific phylogenetic distribution of its homologs-suggest that ThaL is a non-canonical effector. In summary, AHSMG-lipase is a novel lipase family in filamentous fungi, and is probably involved in pathogenicity.

Changes in fauna and abiotic factors in estuaries are a consequence of their exploitation; thus, bivalve mollusks, as they filter, are widely used as environmental bioindicators. The aim of this study was to analyze the existence of seasonal variation in the concentration of total coliforms (TC) and thermotolerant coliforms (Ct), in addition to correlating the data obtained with the salinity and temperature in collection areas of Mytella falcata and the soft tissue cultivated in the Estuarine Complex of Cananéia. Two biweekly samples of water and tissue were taken, for 12 consecutive months, in nine cultivation areas of M. falcata. The analysis of the water samples showed that the Fisheries Institute, Mosquiteiro, and Itapitangui have high TC densities, with respective annual averages of 772.22, 592.67, and 563.75 most probable number (MPN).100 ml-1 . The highest concentrations of TC and Ct occurred in the summer, in most areas. There was a decrease in both TC and Ct with increasing salinity. However, there was an increase in Ct with increasing temperature. The soft tissue showed TC densities from 9 to 26 MPN.100 ml-1 and Ct densities from 6 to 14 MPN.100 ml-1 , displaying the highest counts in the summer. Places where coliforms were found in high concentrations could indicate contamination, because Ct do not support higher salinities for a long time. PRACTITIONER POINTS: The general mean was 77.64 (total coliforms) and 39.35 (thermotolerant) MPN.100 ml-1 . Annual average (thermotolerant) exceeded the permitted Brazilian limit in five points. Higher concentrations of total and thermotolerant coliforms occurred in the summer. In mollusk's tissue, concentrations of thermotolerant were 6.30 to 26.16 MPN.100 g-1 . Coliforms' MPN varied with increasing salinity and temperature.

Anthropic activities performed in a river basin, especially those that promote changes in the soil surface, have great effect on the quality of the waters in the basin. Thus, the objective of this study was to evaluate water quality by applying the Water Quality Index (WQI) in springs and watercourses located in an area with different land use and occupation characteristics, as well as to compare the analyzed variables with the standards established for class 2 of the National Environmental Council (CONAMA, for its acronym in Portuguese) Resolution No. 357/2005. Seven sampling points located in the rural area of the municipality of Jacutinga, Minas Gerais State (MG), were monitored. The water samples were collected from October 2018 to July 2019. Comparison with the resolution showed that the quality condition was lower than the standard established for class 2 in most campaigns conducted. Regarding the mean WQI, the water quality at points S1, WC1, WC2, and WC3 was classified as "average," while the water quality at points S2, S3, and S4 was classified as "good." There was seasonal variation in the WQI only at point WC1. Overall, thermotolerant coliforms (TC), nitrate, and phosphorus are the main factors deteriorating water quality, and it may be directly related to the agricultural activities practiced in the monitored areas.

BACKGROUND: Argentina's geothermal areas are niches of a rich microbial diversity. In 2020, species of Bacillus cytotoxicus were isolated for the first time from these types of pristine natural areas. Bacillus cytotoxicus strains demonstrated the capability to grow and degrade chicken feathers with the concomitant production of proteases with keratinolytic activity, enzymes that have multitude of industrial applications. The aim of this research was to study the production of the proteolytic enzymes and its characterization. Also, feather protein hydrolysates produced during fermentation were characterized. RESULTS: Among the thermotolerant strains isolated from the Domuyo geothermal area (Neuquén province, Argentina), Bacillus cytotoxicus LT-1 and Oll-15 were selected and put through submerged cultures using feather wastes as sole carbon, nitrogen, and energy source in order to obtain proteolytic enzymes and protein hydrolysates. Complete degradation of feathers was achieved after 48 h. Zymograms demonstrated the presence of several proteolytic enzymes with an estimated molecular weight between 50 and > 120 kDa. Optimum pH and temperatures of Bacillus cytotoxicus LT-1 crude extract were 7.0 and 40 °C, meanwhile for Oll-15 were 7.0 and 50 °C. Crude extracts were inhibited by EDTA and 1,10 phenanthroline indicating the presence of metalloproteases. Feather protein hydrolysates showed an interesting antioxidant potential measured through radical-scavenging and Fe3+-reducing activities. CONCLUSION: This work represents an initial approach on the study of the biotechnological potential of proteases produced by Bacillus cytotoxicus. The results demonstrated the importance of continuous search for new biocatalysts with new characteristics and enzymes to be able to cope with the demands in the market.

Ice cream is susceptible to contamination by handling and bad hygiene conditions during both the storage process and the fractioning for sale, and once contaminated, it can cause diseases. The purpose of this survey was to evaluate the microbiological quality of ice cream sold in bulk, of pasty and soft types, offered for consuming. Thirty samples of pasty ice cream sold in bulk, and thirty samples of soft ice cream were analyzed through the counting of thermotolerant coliforms, coagulase-positive Staphylococcus spp., and searching for the presence of Salmonella spp. During the study, a total of ten (33%) samples of pasty ice cream and five (16%) samples of soft ice cream were found to be beyond the limits established by the Brazilian law. Salmonella spp. was found in four samples (6.7%). These results are an alert for the need of greater attention to the microbiological quality of ice cream in order to ensure the safety of its consumers.(AU)

RESUMO Este estudo objetivou verificar a qualidade microbiológica da água e dos mexilhões cultivados pela Associação dos Maricultores de Piúma (AMPI), Espírito Santo, Brasil. Foram realizadas sete coletas de água e mexilhões, mensalmente, entre outubro de 2016 e maio de 2017. Os mexilhões foram coletados nos long lines da AMPI, e em cada mês foram coletados 40 mexilhões Perna perna e 100 mL de água do local. O material coletado foi destinado ao laboratório para a realização das análises microbiológicas em duplicata, número mais provável de coliformes totais (CT) e termotolerantes (Ctt), presença ou ausência de Salmonella sp e número de unidades formadoras de colônias de Staphylococcus aureus. Os resultados mostraram que o número de Ctt nas amostras de água no mês de janeiro estavam acima do permitido pela Resolução n° 357 do Conselho Nacional do Meio Ambiente (CONAMA). Já os níveis de Ctt e Staphylococcus aureus na carne dos mexilhões mostraram-se dentro do limite aceitável pela RDC n° 12 da Agência Nacional de Vigilância Sanitária (ANVISA). Entretanto, foram encontradas bactérias com características do gênero Salmonella sp nos meses de dezembro e março nos mexilhões, impossibilitando sua comercialização e seu consumo. Durante esses meses, a cidade tem alto fluxo de turistas. Por fim, recomenda-se a realização das análises microbiológicas continuamente, principalmente no período do verão, época que tem grande fluxo de turistas no município de Piúma e que registrou presença de Salmonella na carne dos mexilhões e níveis de Ctt na água acima do permitido pelas legislações vigentes.


ABSTRACT This study aimed to verify the microbiological quality of water and mussels cultivated by the Piúma Farmers Association (Associação dos Maricultores de Piúma — AMPI), Espírito Santo, Brazil. Seven samples of water and mussels were collected monthly from October 2016 to May 2017. The mussels were collected from the long lines of the AMPI, and in each month 40 mussels Perna perna and 100 mL of water were sampled from the site. The collected material was sent to the laboratory for duplicate microbiological analysis, Most Probable Number of Total (CT) and Thermotolerant (Ctt) coliforms, presence or absence of Salmonella sp and number of Staphylococcus aureus colony forming units. The results showed that the number of Ctt in the water samples in January was higher than that allowed by Resolution 357 of the National Environment Council (Conselho Nacional do Meio Ambiente — CONAMA). The levels of Ctt and Staphylococcus aureus in mussel meat were within the acceptable range by the Brazilian National Environment Council (Agência Nacional de Vigilância Sanitária — ANVISA) Resolution RDC No. 12. However, colonies with characteristics of Salmonella sp were found in December and March in the mussels, making it impossible to sell and consume. This period coincides with a high flow of tourists in the municipality. Finally, it is recommended to perform microbiological analyzes continuously, especially in the summer, where there are a lot of tourists in the city of Piúma, period that showed the presence of Salmonella in the meat of mussels and Ctt levels in water above the allowed current legislation.

Monitoring water quality is a fundamental process to ensure proper anthropogenic usage and environmental protection of this resource. This study collected monthly measurements of 9 parameters (pH, Temperature, BOD, Total Solids, Thermotolerant Coliforms, Dissolved Oxygen, Total Nitrogen and Total Phosphorus) in 5 sampling stations along the Marrecas water stream, during a 1-year period. Temporal and seasonal variations were analyzed and interpreted for each element, explaining how specific geographical and anthropogenic factors affected the water body. Principal Component Analysis (PCA) was applied to evaluate each element's correlation and to reduce the number of parameters, easing the assessment of water quality for each location. Results were followed by the creation of an improved index for the region, which could better estimate the quality of water, only considering 4 of the original parameters. It was also recognized that each water body possesses several subtleties that impact on how its water quality should be measured and indexed into a single value, which validates the case for the creation of regional WQI's.

BACKGROUND: The use of thermotolerant yeast strains can improve the efficiency of ethanol fermentation, allowing fermentation to occur at temperatures higher than 40 °C. This characteristic could benefit traditional bio-ethanol production and allow simultaneous saccharification and fermentation (SSF) of starch or lignocellulosic biomass. RESULTS: We identified and characterized the physiology of a new thermotolerant strain (LBGA-01) able to ferment at 40 °C, which is more resistant to stressors as sucrose, furfural and ethanol than CAT-1 industrial strain. Furthermore, this strain showed similar CAT-1 resistance to acetic acid and lactic acid, and it was also able to change the pattern of genes involved in sucrose assimilation (SUC2 and AGT1). Genes related to the production of proteins involved in secondary products of fermentation were also differentially regulated at 40 °C, with reduced expression of genes involved in the formation of glycerol (GPD2), acetate (ALD6 and ALD4), and acetyl-coenzyme A synthetase 2 (ACS2). Fermentation tests using chemostats showed that LBGA-01 had an excellent performance in ethanol production in high temperature. CONCLUSION: The thermotolerant LBGA-01 strain modulates the production of key genes, changing metabolic pathways during high-temperature fermentation, and increasing its resistance to high concentration of ethanol, sugar, lactic acid, acetic acid, and furfural. Results indicate that this strain can be used to improve first- and second-generation ethanol production in Brazil.

RESUMO O objetivo deste estudo foi investigar as águas superficiais e os níveis foliares dos elementos-traço (Cu, Zn, Cd e Pb) da Avicennia schaueriana na área de proteção ambiental (APA) Tinharé-Boipeba, litoral sul da Bahia, com ênfase na relação entre a qualidade ambiental e o gerenciamento costeiro. Mensuraram-se in situ variáveis físico-químicas nas águas superficiais, e tomaram-se alíquotas para análises microbiológicas. Também foram coletadas folhas de árvores de mangue em nove estações amostrais. Realizou-se a leitura dos elementos-traço por espectrometria de absorção atômica com chama (F-AAS). As análises físico-químicas e microbiológicas das águas superficiais, ainda que preliminares, indicaram perda de qualidade e desafios ao saneamento, os quais merecem a atenção de autoridades sanitárias ou de saúde pública e consulta à comunidade para elaboração de soluções técnicas compatíveis com os usos da natureza e modos de vida tradicionais na APA. Os níveis foliares dos metais foram normais e não tóxicos, sendo menores do que em áreas costeiras impactadas. Os bosques de mangue encontram-se em bom estado de conservação e servem de área de referência, recomendando-se o monitoramento dos elementos-traço nas folhas ou sedimentos de mangue e qualidade da água. Esta pesquisa tem relevância à conservação dos manguezais, aos usos culturais da natureza e ao gerenciamento costeiro.


ABSTRACT The objective of this study was to investigate the surface waters and leaf levels of trace elements (Cu, Zn, Cd, and Pb) of the Avicennia schaueriana in the Tinharé-Boipeba Environmental Protection Area (APA), South Coast of Bahia, with emphasis to the relationship between environmental quality and coastal management. Physical-chemical variables were measured in situ in surface waters and aliquots were taken for microbiological analysis. Also, leaves of mangrove trees were collected at nine sampling stations. The trace element reading was performed by F-AAS. Physical-chemical and microbiological analyzes of surface water, although preliminary, indicated that there is a loss of quality and challenges to sanitation, which deserve the attention of sanitary authorities, and of public health, and consultation with the community to elaborate technical solutions compatible with the uses of nature and traditional lifestyles in APA. Leaf metal levels were normal and nontoxic, being lower than in impacted coastal areas. Mangrove forests are in good state of conservation and serve as a reference area, recommending the monitoring of trace elements in mangrove leaves or sediments and water quality. This research has relevance to the conservation of mangroves, cultural uses of nature and coastal management.

Abstract This study evaluated the production of endoxylanases by Streptomyces malaysiensis AMT-3 in submerged fermentation using by-products of the food industry at 28ºC. In shake-flasks experiments, the highest endoxylanase activity of 45.8 U.mL-1 was observed within 6 days in a medium containing (w/v) 2.5% wheat bran and 1.2% corn steep liquor. The same culture conditions were used to evaluate the enzyme production in a 2 L stirred tank reactor under different agitation (300, 450 and 600 rev.min-1) and aeration (30 and 60 L.h-1) conditions. The use of 450 rev.min-1 coupled to an aeration of 90 L.h-1 resulted on 81.3 U.mL-1 endoxylanase activity within 5 days. The effect of temperature and pH on endoxylanase activity and stability showed the highest activity at 60 ºC and pH 6.0. Zymography showed the presence of three xylanolytic bands with molecular masses of 690, 180 and 142 kDa. The results showed that the thermotolerant actinobacterial endoxylanase can be produced in high titers using by-product of the food industry.

Esta pesquisa teve por objetivo avaliar a qualidade microbiológica de amostras da água e do tecido de Mytella guyanensis (Mollusca: Bivalva) espécie de relevante interesse comercial e que faz parte direta da renda e alimentação das comunidades tradicionais em regiões litorâneas. Este estudo foi realizado em duas comunidades do Estuário de Cananeia conhecidos como Retiro e Itanhoapina. Durante o período de 2017, onde a espécie é coletada para consumo humano, as amostras de água e do bivalve foram coletadas nas localidades a fim de determinar as concentrações de coliformes totais e termotolerantes. As Amostras de água e de M. guyanensis foram analisadas para determinar o Número Mais Provável de Coliformes (NMP), baseado na Técnica de Tubo Múltiplo. Os resultados das analises, demonstraram baixas concentrações de coliformes nas amostras de água, tendo ocorrido resultado semelhante para as amostras de tecidos dos indivíduos coletados. A análise da água apresentou média geométrica de 34,81 coliformes totais e 20,70 MPN 100 mL-1 de coliformes termotolerantes. As médias de coliformes nos tecidos moles de M. guyanensis foram de 190 MPN g-1 para coliformes totais e 174 MPN g-1 para coliformes termotolerantes. As concentrações de coliformes no tecido dos espécimes coletados, apresentaram correlação positiva com a temperatura e negativa com a salinidade. Houve diferenças significativas entre a sazonalidade em relação à concentração de coliformes totais e termotolerantes no tecido. O verão período de chuvas, apresentou a maior média sazonal de coliformes em M. guyanensis. Todas os valores obtidos para NMP de coliformes estiveram abaixo do limite estabelecido pela legislação.


This study aimed to evaluate the microbiological quality of water and tissue samples of Mytella guyanensis (Mollusk: Bivalve), a species of relevant commercial interest and is a direct part of the income and food of traditional communities in coastal regions. This study was carried out in two communities in the Cananeia Estuary known as Retiro and Itanhoapina. During the period of 2017, where the species is collected for human consumption, water and bivalve samples were collected in the localities in order to determine the concentrations of total and thermotolerant coliforms. The samples of water and M. guyanensis were analyzed to determine the Most Probable Number (MPN) of Coliforms, based on the Multiple Tube Technique. The results of the analyses showed low concentrations of coliforms in the water samples, with a similar result for the tissue samples of the individuals collected. The water analysis showed a geometric mean of 34.81 total coliforms and 20.70 MPN 100 mL-1 of thermotolerant coliforms. The averages of coliforms in the soft tissues of M. guyanensis were 190 MPN g -1 for total coliforms and 174 MPN g -1 for thermotolerant coliforms. The concentrations of coliforms in the tissue of the collected specimens showed a positive correlation with temperature and a negative correlation with salinity. There were significant differences between seasonality in relation to the concentration of total and thermotolerant coliforms in the tissue. The summer rainy season showed the highest seasonal average of coliforms in M. guyanensis. All values obtained for MPN of coliforms were below the limit established by the legislation.

Abstract The isolation of Escherichia coli from food is a major concern. Pathogenic strains of these bacteria cause diseases which range from diarrhea to hemolytic-uremic syndrome. Therefore the virulence genes in E. coli isolates from the mussel ( Mytella guyanensis) commercialized in Cachoeira, Bahia, Brazil were investigated. Samples were purchased from four vendors: two from supermarkets and two from fair outlets. They were conditioned into isothermal boxes with reusable ice and transported to the laboratory for analysis. E. coli strains were isolated in eosin methylene blue agar, preserved in brain-heart infusion medium with 15% glycerol and stored at -20 °C, after microbiological analysis. Virulence genes in the isolated strains were identified by specific primers, with Polymerase Chain Reaction. Twenty-four isolates were obtained, with a prevalence of elt gene, typical from enterotoxigenic infection, in 75% of the isolates. The stx and bfpA genes, prevalent in enterohemorragic and enteropathogenic E. coli, respectively, were not detected. The occurrence of elt virulence-related gene in the E. coli isolates of Mytella guyanensis reveals urgent improvement in food processing, including good handling practices, adequate storage and cooking before consumption, to ensure consumer's health.


Resumo O isolamento de Escherichia coli a partir de alimentos é uma grande preocupação, pois cepas patogênicas desta bactéria podem causar desde diarreia até síndrome hemolítico-urêmica. Diante do exposto, o objetivo do trabalho foi pesquisar genes de virulência em isolados de Escherichia coli provenientes do sururu Mytella guyanensis comercializado na cidade de Cachoeira, Bahia, Brasil. As amostras foram adquiridas de quatro comerciantes, sendo duas de mercados e duas em pontos de venda na feira livre da cidade de Cachoeira, acondicionadas em caixas isotérmicas com gelo reutilizável e transportadas até o laboratório para a análise. Após a análise microbiológica, as cepas de Escherichia coli foram isoladas em ágar Eosina Azul de Metileno e preservadas em caldo Brian Heart Infusion e glicerol a 15% e mantidas a - 20° C. A identificação dos genes de virulência nas cepas isoladas foi realizada utilizando primers específicos, por meio da Reação em Cadeia da Polimerase. Foram obtidos 24 isolados de Escherichia coli, destes a prevalência do gene elt , característico de Escherichia coli enterotoxigênica, foi de 75% dos isolados. Não houve a detecção dos genes stx e bfpA nos isolados, os quais são prevalentes nas cepas de Escherichia coli enterohemorrágica e Escherichia coli enteropatogênica, respectivamente. A presença do gene elt relacionado à virulência de Escherichia coli nos isolados de Mytella guyanensis revela a necessidade da melhoria no processamento, incluindo boas práticas de manipulação, armazenamento adequado e cocção previa ao consumo, visando a garantia da saúde do consumidor.

This study was conducted to evaluate the microbiological quality of a mangrove estuary in the Vitória Bay region, Espírito Santo, Brazil. We analyzed the presence and concentration of enteric viruses and thermotolerant coliforms in water, mussels (Mytella charruana and Mytella guyanensis), and oysters (Crassostrea rhizophorae), collected over a 13-month period. Human adenovirus, rotavirus A (RVA), and norovirus genogroup II were analyzed by quantitative PCR. The highest viral load was found in RVA-positive samples with a concentration of 3.0 × 104 genome copies (GC) L-1 in water samples and 1.3 × 105 GC g-1 in bivalves. RVA was the most prevalent virus in all matrices. Thermotolerant coliforms were quantified as colony-forming units (CFU) by the membrane filtration method. The concentration of these bacteria in water was in accordance with the Brazilian standard for recreational waters (< 250 CFU 100 mL-1) during most of the monitoring period (12 out of 13 months). However, thermotolerant coliform concentrations of 3.0, 3.1, and 2.6 log CFU 100 g-1 were detected in M. charruana, M. guyanensis, and C. rhizophorae, respectively. The presence of human-specific viruses in water and bivalves reflects the strong anthropogenic impact on the mangrove and serves as an early warning of waterborne and foodborne disease outbreaks resulting from the consumption of shellfish and the practice of water recreational activities in the region.

The objective of this meta-analysis was to summarize available information on the prevalence of thermotolerant Campylobacter (TC) in different food-producing animals worldwide. Databases (i.e., PubMed, ScienceDirect, Scopus) were searched from 1980 to 2017 unrestricted by language. The inclusion criteria were as follows: prevalence or incidence studies, published in peer-reviewed journals, and they must have reported the total number of animal samples studied and the number of samples that were positive for the presence of TC. When the identification of Campylobacter species was available, this information was included in the analysis. Multilevel random-effect meta-analysis models were fitted to estimate mean occurrence rate of TC and to compare them among different factors potentially associated with the outcome. The mean occurrence rate of TC in food-producing animals was 0.424 (95% CI: 0.394-0.455), and the mean occurrence rate of Campylobacter jejuni and Campylobacter coli were 0.214 and 0.133, respectively. Pigs and poultry showed the highest prevalence of TC; however, there were differences in the prevalence of each Campylobacter species. Campylobacter jejuni was observed in broilers (0.322; 95% CI: 0.273-0.377) and hens (0.395; 95% CI: 0.265-0.542), while C. coli was restricted essentially in pigs (0.553; 95% CI: 0.541-0.650). The prevalence of C. jejuni in intensively bred cattle was higher (0.302; 95% CI: 0.227-0.389) than the prevalence in extensively bred cattle (0.172; 95% CI: 0.119-0.242) while the prevalence of C. coli was similar (0.051; 95% CI: 0.028-0.091 vs. 0.050; 95% CI: 0.027-0.091) in both production systems. Agar with or without blood used for the isolation of TC did not affect the prevalence observed. The method of species identification did not seem to generate differences in the prevalence of Campylobacter species. The prevalence of Campylobacter in primary food production has a strong impact on the entire agri-food chain. National authorities must monitor the situation with the aim to establish the appropriate risk management measures.
