This study aimed to propose and evaluate a drug susceptibility testing (DST) using the 2,3,5-triphenyl tetrazolium chloride (TTC) as a colorimetric indicator against Mycobacterium abscessus complex (MABC), M. avium complex (MAC), and M. kansasii strains, main nontuberculous mycobacteria (NTM) of clinical relevance. Our results demonstrated that the assay using TTC and the broth microdilution method recommended by the Clinical and Laboratory Standards Institute had essential agreement above 91%, 92%, and 100%, for drugs tested against MABC, MAC, and M. kansasii strains, respectively. Categorical agreement above 91% was obtained for most drugs tested against MABC, except to cefoxitin (76.5%). For drugs tested against MAC and M. kansasii, categorical agreement above 92% and 100% was observed, respectively. TTC showed to be a promising colorimetric indicator of growth to be used in DST for NTM, allowing an easier reading of results.

Background and aim: Prosopis strombulifera (Lam.) Benth is a rhizomatous shrub native from different zones of Argentine Republic. P. strombulifera aqueous extract (PsAE) has different effects and several biological activities have been reported. The goal of this study was to analyze the activity of PsAE on a murine model of cutaneous leishmaniasis caused by Leishmania amazonensis. Experimental procedure: PsAE was orally administered at 150 mg/animal/day on BALB/c mice infected in the right footpad (RFP) with 1 × 105 promastigotes of L. amazonensis. As a chemotherapeutic control of treatment, animals receive a commercial form of meglumine antimoniate (MA) (Glucantime®, Aventis, Paris, France). Results and conclusion: We observe that the size of RFP lesions of infected mice without treatment showed a grade of inflammation, ulceration and necrosis at the site of infection much greater than that observed with PsAE or MA treatment. Moreover, PsAE was capable of decreasing parasite burden and splenic index. Furthermore, PsAE treated mice showed a significant decrease in O.D. of total anti-Leishmania IgG antibody responses against L. amazonensis. This decrease was similar to those observed when the reference drug, MA, was used. This would indicate that PsAE treatment inhibits or delays disease progression in mice. In conclusion, our findings suggest that PsAE could be a potential candidate to be used, as a new therapeutic strategy, to treat cutaneous leishmaniasis caused by L. amazonensis.

Psoriasis is a chronic inflammatory skin disease, which does not have effective treatment options. However, olive oil has been suggested as an alternative to treat psoriasis, but no study has evaluated the mechanisms involved in the effects of olive oil on psoriasis. Thus, the current study investigated whether olive oil could ameliorate psoriasiform skin inflammation. To test this, mice received topical application of imiquimod to induce inflammation and were treated orally with olive oil. Human immortalized keratinocytes were also treated with imiquimod and olive oil. Epidermal thickness and keratinocyte proliferation were increased in imiquimod-induced lesions of olive-oil-treated animals. In both in vitro and in vivo studies, protein levels of nuclear factor erythroid 2-related factor 2 (Nrf2) were elevated following imiquimod and olive oil administration. Inhibition of Nrf2 abolished the increased proliferation of keratinocytes treated with imiquimod and olive oil, demonstrating the role of Nrf2 in olive oil-mediated exacerbation of psoriasiform skin inflammation. In addition, lower levels of linoleic acid and higher levels of oleic acid were observed in imiquimod- and olive-oil-treated animals, which may also contribute to the adverse effects of olive oil on psoriasis. In conclusion, dietary intake of olive oil aggravates the symptoms of psoriatic skin lesions through the overexpression of Nrf2 and an imbalance in oleic and linoleic acids levels, suggesting that a diet rich in olive oil may have significant negative effects on psoriasis.

Abstract The aim of this study was to investigate the effect of TiO2/N-succinyl-chitosan composite (TiO2/ NSCS) photodynamic therapy (PDT), while considering the effects of various light sources on the activation of photosensitizer. The methyl thiazolyl tetrazolium assay was used to examine the cell survival rate of the cells. The results showed that glioma cell strain (U251) was the most sensitive cancer cell strain to TiO2/NSCS. When the concentration of TiO2/NSCS was between 0 and 800 μg·mL-1, there was no obvious cytotoxicity to normal liver cells (HL-7702) and U251 cells. During the PDT process, the photokilling effect of TiO2/NSCS on U251 cells under ultraviolet-A (UVA) light irradiation was stronger than that of pure TiO2, and its killing effects were positively correlated with concentration and irradiation time. In addition, both UVA and visible light could excite TiO2/ NSCS, which had significant killing effect on U251 cells. The results of acridine orange/ethidium bromide fluorescent double staining and Annexin V/propidium iodide double staining indicated that TiO2/NSCS under UVA and visible light irradiation could kill U251 cells by inducing apoptosis, and the apoptosis rate of TiO2/NSCS treatment groups was higher than that of TiO2 treatment groups. Therefore, TiO2/NSCS might be used as a potential photosensitizer in PDT.

Aquaculture production has increased in the last decades, with crustacean production contributing with 9.8% of the total production. However, fisheries and aquaculture sectors present several challenges, such as fish stocks fished beyond biological sustainability, animal diseases, biosecurity, and environmental impact. It is important to improve shrimp production with healthy animals, avoiding environmental impacts, e.g. with the use of heterotrophic rearing system. It is known that the heterotrophic system can stimulate the activation of immune genes, but how it affects the shrimp immune system is unknown. To assess if a heterotrophic system influences the cellular immune response in shrimp, Litopenaeus vannamei shrimp were reared in heterotrophic and clear water systems. Cellular immune response parameters such as total and differential hemocyte counts, phagocytosis indices and the production of the superoxide anion were evaluated after 60, 120 and 180 days. After 60 days, total haemocyte counts were higher in shrimps reared in the clear water system, while after 120 days it was higher in shrimps reared in the heterotrophic system. No significant difference was observed after 180 days. Hyaline, granular and semi-granular cells showed similar behavior, peaking after 120 days in the heterotrophic system. By the 60th day, phagocytic capacity was higher in the heterotrophic system, while no differences were found for the 120th and 180th day. No differences were detected concerning the phagocytic index or superoxide anion production. The heterotrophic system can affect total and differential shrimp haemocyte counts and phagocytic capacity, depending on the period of time they were maintained in this system. However, the phagocytic index and superoxide anion production are not affected by the heterotrophic system at the time points evaluated herein.

Hydroxyapatite, chitosan, and carbon nanotube composite biomaterial were developed to improve bone healing. Previous studies suggested that a combination of biomaterials and mesenchymal stem cells (MSCs) can potentially help promote bone regeneration. In the present study, we first developed hydroxyapatite, chitosan, and carbon nanotube composite biomaterial. Then, the effect of different concentrations of the extract on the viability of Vero cells (ATCC CCL-81) and MSCs obtained from sheep bone marrow using methylthiazol tetrazolium (MTT) and propidium iodide (PI) assays were evaluated. The biomaterial group demonstrated an absence of cytotoxicity, similar to the control group. Samples with 50% and 10% biomaterial extract concentrations showed higher cell viability compared to samples from the control group (MTT assay). These results suggest that the presence of this composite biomaterial can be used with MSCs. This study also concluded that hydroxyapatite, chitosan, and carbon nanotube composite biomaterial were not cytotoxic. Therefore, these could be used for performing in vivo tests.(AU)


O compósito à base de hidroxiapatita, quitosana e nanotubo de carbono foi desenvolvido com o intuito de auxiliar na consolidação óssea. Estudos anteriores sugerem que a combinação de substitutos ósseos e células-tronco mesenquimais (CTM) podem auxiliar a potencializar e promover a regeneração óssea. No presente estudo, o biomaterial foi desenvolvido e a viabilidade e a citotoxicidade de células Vero (ATCC CCL-81) e CTM obtidas de medula óssea provenientes de ovinos utilizando ensaios metil-tiazol-tetrazólio, MTT e iodeto de propídeo (PI) foram avaliadas em diferentes concentrações de extrato desse compósito. O compósito demonstrou ausência de citotoxicidade com comportamento semelhante ao grupo controle. Amostras com 50% e 10% de concentração de extrato do compósito mostraram resultados maiores comparados ao grupo controle (ensaio MTT). Esses resultados também sugerem que a presença do biomaterial pode ser utilizada em associação a CTM. Assim, esse estudo conclui que o compósito apresentado de hidroxiapatita, quitosana e nanotubo de cabono não foi considerado citotóxico e pode ser utilizado em teste in vivo.(AU)

RESUMEN Amaranthus hybridus L., conocido como bledo, es una planta maleza en una gran variedad de cultivos en el mundo, que se caracteriza por producir alto número de semillas latentes. Para generar estrategias de manejo sostenibles en esta especie es necesario comprender mejor la fisiología de sus semillas. Por tanto, el objetivo fue evaluar el efecto de la luz, de las giberelinas y la profundidad de la siembra sobre la germinación de semillas de bledo. En un primer experimento, se evaluaron cuatro tratamientos de luz (rojo, azul, sin luz y luz natural «testigo¼); en el segundo, se imbibieron semillas en diferentes concentraciones de ácido giberélico (0, 200, 400 y 800mg L-1 de GA3) y, en un último experimento, se evaluaron ocho profundidades de siembra (10, 20, 30, 40, 50, 60, 70 y 80mm), utilizando turba como sustrato. Se encontró mayor porcentaje de germinación en oscuridad, indicando que son semillas fotoblásticas negativas. Para el experimento de giberelinas, el mejor resultado se obtuvo con 400 y 800mg L-1 de ácido giberélico. En el tercer experimento, se observó alto porcentaje de emergencia de plántulas en las primeras profundidades (10mm a 30mm); a mayor profundidad hubo menor emergencia. La respuesta diferencial a la luz, a la aplicación de las giberelinas y el alto porcentaje de semillas viables que no germinaron permite concluir que semillas de bledo poseen latencia fisiológica.


ABSTRACT Amaranthus hybridus L., known as bledo, is a weed in a wide variety of crops worldwide, characterized by producing high number of seeds with dormancy. To generate sustainable management strategies of this species, it is necessary to better understand the physiology of its seeds. Therefore, the objective was to evaluate the effect of light, gibberellins, and seed burying depth on seed germination of bledo. In the first experiment, four light conditions were evaluated (red, blue, without light and natural light (control)), in the second experiment the seeds were imbibed in different concentrations of gibberellic acid (0, 200, 400 and 800mg L-1 of GA3), and in the last experiment, 8 planting depths were evaluated (10, 20, 30, 40, 50, 60, 70 or 80mm) using peat as a substrate. A higher percentage of seed germination in darkness was found, indicating that this species had negative photoblastic seeds. For the gibberellin experiment, the best result was obtained with treatment with 400 and 800mg L-1 of gibberellic acid. In the third experiment, a high percentage of seedling emergence was observed from the first depths (10mm to 30mm); the greater was the depth, the less emergence was found. The differential responses to light, the application of gibberellins, and the high percentage of non-germinated viable seeds allowed to conclude that bledo seeds possess physiological dormancy.

Certain soil bacteria produce beneficial effects on the growth and health of plants; hence, their use is steadily increasing. Five strains of Bacillus with plant growth-promoting potential were selected in this study, which produced indole-3-acetic acid levels below 50 µg.mL-1. On the other hand, while only strains M8 and M15 dissolved phosphorus, the latter was the only strain that did not produce siderophores. Only strains M8 and M16 significantly inhibited the in vitro growth of Botrytis cinerea and Fusarium solani phytopathogens, whose inhibition ranges fluctuated between 60% and 63% for strains M8 and M16 against B. cinerea and between 40% and 53% for strains M8 and M16 against F. solani. Based on these results, the need to implement resistance induction against gray mold on pepper plants was determined using strains M8 and M16. In this case, strain M16 inhibited the propagation of the necrotic spot by approximately 70%, whereas strain M8 significantly reduced the superoxide dismutase activity in systemic leaves, which substantially increased in plants inoculated with strain M8 and infected with the pathogen. Accordingly, the use of native rhizobacteria may entail biotechnological progress for the integrated management of crops in agriculture industry.

OBJECTIVES: This systematic review aimed to compare the cytotoxicity of root canal filling materials (RCFMs) assessed using tetrazolium salt-based tests (TSBT), including the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay, with those obtained using other cell viability assays. METHODS: A search was performed on PubMed, Scopus, Web of Science and OpenGrey up to March 2019, followed by a manual search. According to the Participants, Exposure, Comparator and Outcomes (PECO) criteria, in vitro studies that evaluated the cytotoxic effect of RCFMss on animal and/or human cells through TSBT and at least one other viability assay were compared. The methodological quality of selected papers was assessed using ToxRTool® and SciRAP® . Data were analysed using Wilcoxon's signed-rank test for paired samples and linear weighting kappa. RESULTS: A total of 230 non-duplicated records were identified. After applying the eligibility criteria, 55 studies were selected for methodological evaluation, seven were selected by manual searching, 22 were excluded for methodological reasons, and 40 were included. A total of 410 comparisons were performed between TSBT and distinct cell viability tests (DCVT). MTT had moderate concordance with DCVT using human cells (n = 138 samples) (P = 0.507; k = 0.4225) and animal cells (n = 122 samples) (P = 0.124; k = 0.5775). XTT had good concordance using human (n = 110 samples) (P = 0.507; k = 0.6336) and animal cells (n = 12 samples) (P = 0.564; k = 0.6604). MTT, XTT, WST and MTS assays showed moderate concordance with DCVT (n = 410 samples) (P = 0.375; k = 0.5138) and complete agreement in 226 samples. DISCUSSION: The included studies had methodological heterogeneity that was minimized by the systematic review methodology. CONCLUSIONS: MTT and XTT do not cause over- or underestimation of cell viability during cytotoxicity screening of root canal filling materials, implying that these assays can be considered reliable for this purpose. Nonetheless, the development of protocols for the cytotoxic screening of these materials on 3D tissue-like cultures aiming to improve their predictability in the clinical scenario is suggested.

ABSTRACT Schizolobium parahyba is a fast-growing tropical tree, with various uses: forestry, agroforestry, landscaping, boat building, cabinetmaking and light packaging. Studies of morphometry and viability of forest species seeds are important for their identification, conservation and sustainable use. The objective of the study was to describe the morphometric and viability characteristics of the seeds of S. parahyba. Seeds of free pollination of plantations from three locations in Tierralta (Córdoba - Colombia) were used. In each plantation, five trees were randomly taken and, of each, 100 seeds to estimate the morphometric characteristics and seed weight. The morphological description was made with a sample of 10 seeds. The viability was determined by the tetrazolium test in an experiment under a completely randomized design, with six treatments and four replicates of 25 seeds each. The six treatments corresponded to the combinations of three concentrations of tetrazolium (0.5; 1.0 and 1.5%) and two staining times (2 and 3 hours). The width, length, and ratio width / length of the seed showed little variation, compared to the freshweight of a seed, freshweight of 100 seeds and number of seeds per kilogram. Tetrazolium and germination tests showed similar and reliable results. The concentration of 0.5% tetrazolium, with immersion of 2 hours, was sufficient to determine the viability of seeds of S. parahyba.


RESUMEN Schizolobium parahyba es un árbol tropical de crecimiento rápido, con diversos usos: forestería, agroforestería, paisajismo, construcción de embarcaciones, ebanistería y embalajes livianos. Los estudios de morfometría y viabilidad de semillas de especies forestales son importantes para la identificación, la conservación y el uso sostenible de éstas. El objetivo del estudio fue describir las características morfométricas y viabilidad de la semilla de S. parahyba. Se utilizó semilla sexual de polinizaciónlibre de plantaciones de tres localidades de Tierralta (Córdoba, Colombia). En cada plantación, se tomaron al azar cinco árboles y de cada uno 100 semillas, para la estimación de las características morfométricas y el peso de la semilla. La descripción morfológica, se hizo con una muestra de 10 semillas. La viabilidad, se determinó mediante la prueba de tetrazolio, en un experimento bajo un diseño completamente al azar, con seis tratamientos y cuatro repeticiones de 25 semillas cada una; los seis tratamientos correspondieron a las combinaciones de tres concentraciones de tetrazolio (0,5; 1,0 y 1,5%) y dos tiempos de tinción (2 y 3 horas). Las características de ancho, longitud y relación ancho/longitud de la semilla, acusaron poca variación, en comparación con el peso de una semilla, peso frescode 100 semillas y número de semilla por kilogramo. Las pruebas de tetrazolio y de germinaciónmostraron resultados similares y confiables. La concentración de 0,5% de tetrazolio, con inmersión de 2 horas, es suficiente para determinar la viabilidad de la semilla de S. parahyba.

Chemical management is the most widely adopted technique to control weeds in sugarcane crops. Purple nutsedge (Cyperus rotundus L.) is among the most difficult species to control. Herbicides need to be absorbed and translocated in a sufficient amount for the tuber chain to be effective. Thus, this study aimed to evaluate the effectiveness of the herbicides sulfentrazone, diclosulam, imazapic, imazapyr, halosulfuron, ethoxysulfuron, monosodium methyl arsenate (MSMA), and 2,4-dichlorophenoxyacetic acid (2,4-D) in controlling C. rotundus and reducing the viability of its tubers. The experiment was carried out in a greenhouse in a completely randomized design, with nine treatments and four replications. The herbicides sulfentrazone (800 g active ingredient ­ ai·ha?1), diclosulam (193.17 g ai·ha?1), imazapic (133 g ai·ha?1), and imazapyr (500 g acid equivalent ­ ae·ha?1) were applied in preemergence at five days after planting the tubers, while halosulfuron (112.5 g ai·ha?1), ethoxysulfuron (135 g ai·ha?1), MSMA (1,975 g ai·ha?1), and 2,4-D (1,340 g ae·ha?1) were applied in postemergence (4 to 5 leaves). The parameters visual control, shoot dry matter, number of tubers and bulbs, weight of tubers + bulbs, number of epigeal manifestations, and tuber viability were analyzed. The herbicides diclosulam, halosulfuron, and ethoxysulfuron provided 100% control of the shoot at 90 days after application (DAA). All herbicides reduced the number of bulbs, weight of tubers + bulbs, and shoot dry matter. The herbicides sulfentrazone, imazapic, halosulfuron, ethoxysulfuron, and MSMA provided the highest reduction in tuber viability.(AU)


O manejo químico é o mais adotado para o controle de plantas daninhas na cultura da cana-de-açúcar. Entre as espécies de difícil controle está a tiririca (Cyperus rotundus L.). Os herbicidas utilizados precisam ser absorvidos e transportados para a cadeia de tubérculos em quantidades suficientes para serem eficazes. Assim, objetivou-se com este estudo avaliar a eficácia dos herbicidas sulfentrazona, diclosulam, imazapique, imazapir, halossulfurom, etoxissulfurom, metano arseniato ácido monossodico (MSMA) e ácido diclorofenoxiacético (2,4-D) no controle de C. rotundus e na redução da viabilidade de seus tubérculos. O experimento foi realizado em casa de vegetação, em delineamento inteiramente randomizado, com nove tratamentos e quatro repetições. Os herbicidas sulfentrazone (800 g ingrediente ativo ­ i.a.·ha-1), diclosulam (193,17 g i.a.·ha-1), imazapique (133 g i.a.·ha-1) e imazapir (500 g equivalente ­ e.a.·ha-1) foram aplicados cinco dias após o plantio dos tubérculos em pré-emergência, enquanto que o halossulfurom (112,5 g i.a.·ha-1), etoxissulfurom (135 g i.a.·ha-1), MSMA (1.975 g i.a.·ha-1) e 2,4-D (1.340 g e.a.·ha-1) foram aplicados em pós-emergência (4 a 5 folhas). Foram feitas avaliações visuais de controle, massa seca da parte aérea, número de tubérculos e bulbos, peso de tubérculos + bulbos, número de manifestações epígeas e viabilidade de tubérculos. Aos 90 dias após a aplicação (DAA) os herbicidas diclosulam, halossulfurom e etoxissulfurom propiciaram 100% de controle da parte aérea. Todos os herbicidas reduziram o número de bulbos, peso de tubérculos + bulbos e massa seca da parte aérea. Sulfentrazona, imazapic, halossulfurom, etoxissulfurom e MSMA foram os herbicidas que mais reduziram a viabilidade dos tubérculos.(AU)

Chemical management is the most widely adopted technique to control weeds in sugarcane crops. Purple nutsedge (Cyperus rotundus L.) is among the most difficult species to control. Herbicides need to be absorbed and translocated in a sufficient amount for the tuber chain to be effective. Thus, this study aimed to evaluate the effectiveness of the herbicides sulfentrazone, diclosulam, imazapic, imazapyr, halosulfuron, ethoxysulfuron, monosodium methyl arsenate (MSMA), and 2,4-dichlorophenoxyacetic acid (2,4-D) in controlling C. rotundus and reducing the viability of its tubers. The experiment was carried out in a greenhouse in a completely randomized design, with nine treatments and four replications. The herbicides sulfentrazone (800 g active ingredient ­ ai·ha?1), diclosulam (193.17 g ai·ha?1), imazapic (133 g ai·ha?1), and imazapyr (500 g acid equivalent ­ ae·ha?1) were applied in preemergence at five days after planting the tubers, while halosulfuron (112.5 g ai·ha?1), ethoxysulfuron (135 g ai·ha?1), MSMA (1,975 g ai·ha?1), and 2,4-D (1,340 g ae·ha?1) were applied in postemergence (4 to 5 leaves). The parameters visual control, shoot dry matter, number of tubers and bulbs, weight of tubers + bulbs, number of epigeal manifestations, and tuber viability were analyzed. The herbicides diclosulam, halosulfuron, and ethoxysulfuron provided 100% control of the shoot at 90 days after application (DAA). All herbicides reduced the number of bulbs, weight of tubers + bulbs, and shoot dry matter. The herbicides sulfentrazone, imazapic, halosulfuron, ethoxysulfuron, and MSMA provided the highest reduction in tuber viability.(AU)


O manejo químico é o mais adotado para o controle de plantas daninhas na cultura da cana-de-açúcar. Entre as espécies de difícil controle está a tiririca (Cyperus rotundus L.). Os herbicidas utilizados precisam ser absorvidos e transportados para a cadeia de tubérculos em quantidades suficientes para serem eficazes. Assim, objetivou-se com este estudo avaliar a eficácia dos herbicidas sulfentrazona, diclosulam, imazapique, imazapir, halossulfurom, etoxissulfurom, metano arseniato ácido monossodico (MSMA) e ácido diclorofenoxiacético (2,4-D) no controle de C. rotundus e na redução da viabilidade de seus tubérculos. Reactive oxygen species (ROS) can be generated in mammalian cells via both enzymatic and non-enzymatic mechanisms. In sperm cells, while ROS may function as signalling molecules for some physiological pathways, the oxidative stress arising from the ubiquitous production of these compounds has been implicated in the pathogenesis of male infertility. In vitro studies have undoubtedly shown that spermatozoa are indeed susceptible to free radicals. However, many reports correlating ROS with sperm function impairment are based on an oxidative stress scenario created in vitro, lacking a more concrete observation of the real capacity of sperm in the production of ROS. Furthermore, sample contamination by leukocytes and the drawbacks of many dyes and techniques used to measure ROS also greatly impact the reliability of most studies in this field. Therefore, in addition to a careful scrutiny of the data already available, many aspects of the relationship between ROS and sperm physiopathology are still in need of further controlled and solid experiments before any definitive conclusions are drawn.

Um fator chave para a obtenção de altos rendimentos na lavoura de soja é a utilização de sementes com alta qualidade fisiológica, neste contexto é necessário que no momento da colheita os danos mecânicos sejam minimizados. O objetivo do trabalho foi avaliar a qualidade fisiológica de sementes de soja colhidas com máquinas equipadas com sistema de trilha radial e axial. O delineamento adotado foi inteiramente casualizado com dois tratamentos (Sistema de trilha axial e radial) e dez repetições, a cultivar utilizada foi a M6210, semeada com 16 plantas por metro. As colhedoras usadas no trabalho foram uma TC 5070 que estava regulada com 900 rpm de ventilação e velocidade do cilindro de 600 rpm, a velocidade de trabalho foi de 6 km h-1 e uma CR 5.85 com ventilação de 800 rpm e velocidade dos rotores de 750 rpm, a velocidade de colheita foi de 8 km h-1. As variáveis avaliadas foram os danos mecânicos, teste de tetrazólio, envelhecimento acelerado, viabilidade, germinação de plantas normais e anormais. As sementes colhidas com o sistema axial apresentaram 5% a menos de dano mecânico, 7% a mais de vigor pelo teste de tetrazólio, viabilidade 2,5% maior e 2,75% a mais de germinação.

O uso de probióticos a base de leveduras melhora o ganho de peso e o sistema imunológico de bovinos frente a grandes desafios imunológicos, no entanto permanece a dúvida se seu uso também apresentaria efeito em desafios mais tênues. Desta maneira, o presente trabalho verificou se o uso de probiótico a base de Enterococcus faecium e Saccaromyces cerevisiae via oral interfere na imunidade inata e no peso de bezerros sadios, em fase de crescimento, alimentados principalmente com silagem de milho. Para tanto, 12 bezerros da raça Jersey (120 kg ±50 kg e 4-7 meses de idade), foram alocados nos grupos controle (n=6) ou tratamento (n=6), de acordo com a oferta de probiótico (2g/dia, via oral) durante 45 dias. Nos dias 0, 15, 30 e 45, os animais foram submetidos a pesagens, exames clínicos, e colheita de sangue para realização de hemograma e mensuração do metabolismo oxidativo neutrofílico. Observou-se que, o probiótico promoveu pequeno e pontual incremento na função neutrofilica sanguínea (P=0,10, D15), incremento nos valores de hematócrito e hemoglobina (P=0,03 e 0,05, D45), além de maior ganho de peso nas fases iniciais da administração do suplemento (P=0,05), permitindo concluir que o suplemento é benéfico para animais em fase de adaptação de trocas de dieta e de manejo, mesmo quando estes desafios são tênues.(AU)


The use of yeast probiotics improves body weight gain and the immune system of cattle submitted to husbandry challenges, however the question remains whether the probiotics could have an effect on more tenuous challenges. In this way, the present study verified if the use of probiotic (Enterococcus faecium and Saccharomyces cerevisiae) provided orally, interferes with the innate immunity and body weight of calves fed mainly with corn silage. Twelve Jersey calves (120 kg ± 50 kg and 4-7 months of age) were allocated to the control (n = 6) or treatment (n = 6) group according to the probiotic supply (2 g / day, orally) for 45 days. On days 0, 15, 30 and 45, the animals were weighted, and submitted to clinical examinations, and blood collection for hemogram and measurement of neutrophil oxidative metabolism. It was observed that the probiotic promoted a slight and punctual increase in blood neutrophilic function (P = 0.10, D15), increased hematocrit and hemoglobin values P = 0.03 and 0.05, D45) and increased the body weight gain in the initial phases of supplementation (P = 0.08), allowing to conclude that the probiotic is beneficial for animals in the adaptation phase of diet changes and management, even when these challenges are mild.(AU)


El uso de probióticos de levadura mejora el aumento de peso corporal y el sistema inmunológico del ganado sometido a los desafíos de la cría, sin embargo, la pregunta sigue siendo si los probióticos podrían tener un efecto en desafíos más tenues. De esta manera, el presente estudio verificó si el uso de probióticos (Enterococcus faecium y Saccharomyces cerevisiae) proporcionado por vía oral interfiere con la inmunidad innata y el peso corporal de los terneros alimentados principalmente con ensilaje de maíz. Doce terneros de Jersey (120 kg ± 50 kg y 4-7 meses de edad) se asignaron al grupo control (n = 6) o tratamiento (n = 6) según el suministro de probióticos (2 g / día, por vía oral) para 45 dias. En los días 0, 15, 30 y 45, los animales se pesaron y se sometieron a exámenes clínicos y se recogieron muestras de sangre para el hemograma y la medición del metabolismo oxidativo de los neutrófilos. Se observó que el probiótico promovió un aumento leve y puntual de la función neutrofílica en sangre (P = 0,10, D15), aumento de los valores de hematocrito y hemoglobina (P = 0,03 y 0,05, D45) y aumento de la ganancia de peso corporal en las fases iniciales de la suplementación. (P = 0.08), lo que permite concluir que el probiótico es beneficioso para los animales en la fase de adaptación de los cambios y el manejo de la dieta, incluso cuando estos desafíos son leves.(AU)

Seeds with a good physiological quality are essential to high productivity. However, for some seeds, Phaseolus lunatus L., the viability and vigor analyze show deficiency in quality. This work aimed to evaluate the physiological quality of Phaseolus lunatus L. seeds using several vigor tests. The study was realized at the Laboratory of Seed Analysis of the Agricultural Sciences Center of the Federal University of Paraíba, in Areia ­ PB, Brazil. We used seeds from five cultivars of P. lunatus (Branca, Orelha de Vó, Cearense, Rosinha, and Roxinha). To determine the physiological quality, we measured the water content, the viability and vigor test (germination, emergence, electrical conductivity, tetrazolium, first germination count and emergence, seedling length and dry mass). We used a completely randomized design for seeds evaluation, and the field test was performed in randomized blocks. The water content of P. lunatus did not differ among the cultivars, indicating a uniformity in the percentage of moisture. Also, the percentage and speed of emergency and the dry mass of seedlings did not differ among the varieties. The tetrazolium and electrical conductivity tests indicated that Rosinha cultivar had the worst viability concerning the other cultivars, showing a low percentage of living tissue and lower electrical conductivity. The seeds of Roxinha cultivar had the better physiological quality, and the Cearense cultivar had the worst quality evaluation. We recommend the tetrazolium test but not the electrical conductivity test to evaluate the vigor of P. lunatus.


Em qualquer cultura, para se ter uma elevada produtividade é indispensável à aquisição de sementes com alta qualidade fisiológica, mas para sementes de Phaseolus lunatus L., ainda ocorre deficiência em análises de viabilidade e vigor. Dessa forma, o objetivo neste trabalho foi avaliar a qualidade fisiológica das sementes de cultivares de Phaseolus lunatus por diferentes testes de vigor. A pesquisa foi realizada no Laboratório de Análise de Sementes do Centro de Ciências Agrárias da Universidade Federal da Paraíba, em Areia - PB, com sementes de cinco cultivares de Phaseolus lunatus (Branca, Orelha de Vó, Cearense, Rosinha e Roxinha). Para avaliação da qualidade fisiológica das sementes determinou-se o teor de água e realizou-se testes de viabilidade e vigor (germinação, emergência, condutividade elétrica, tetrazólio, primeira contagem de germinação e emergência, comprimento e massa seca de plântulas). O delineamento utilizado foi o inteiramente casualizado, com exceção dos testes realizados em campo, que foi em blocos ao acaso. O teor de água das sementes dos cultivares de feijão-fava não diferiu estatisticamente entre si, indicando que houve uniformidade na porcentagem de umidade. Não houve diferença significativa entre as sementes dos cultivares de feijão-fava quando avaliou-se a porcentagem e velocidade de emergência, bem como a massa seca de plântulas. Os resultados dos testes de tetrazólio e condutividade elétrica constatou-se maior viabilidade para as sementes do cultivar Roxinha, Branca, Cearense e Orelha de Vó quando comparadas com as da Rosinha, uma vez que houve um maior percentual de tecidos vivos e uma menor condutividade elétrica, respectivamente. As sementes do cultivar Roxinha são de melhor qualidade fisiológica e, o cultivar Cearense de pior qualidade; o teste de tetrazólio é recomendado para avaliação do vigor de sementes de cultivares de Phaseolus lunatus, enquanto, o teste de condutividade elétrica não é recomendado para avaliar o vigor das sementes dos cultivares de P. lunatus.

ABSTRACT BACKGROUND: Gastric cancer is known as the fourth most common cancer. Current treatments for cancer have damaged the sensitive tissues of the healthy body, and in many cases, cancer will be recurrent. Therefore, need for treatments that are more effective is well felt. Researchers have recently shifted their attention towards antipsychotic dopamine antagonists to treat cancer. The anticancer activities of aripiprazole remain unknown. OBJECTIVE: This study aimed to evaluate the efficacy and safety of aripiprazole on gastric cancer and normal cell lines. METHODS: In this regard, the cytotoxicity and genotoxicity of aripiprazole were investigated in MKN45 and NIH3T3 cell lines by methyl tetrazolium assay and on peripheral blood lymphocytes by micronucleus assay. For this purpose, cells were cultured in 96 wells plate. Stock solutions of aripiprazole and cisplatin were prepared. After cell incubation with different concentrations of aripiprazole (1, 10, 25, 50, 100 and 200 μL), methyl tetrazolium solution was added. For micronucleus assay fresh blood was added to RPMI culture medium 1640 supplemented, and different concentrations of aripiprazole (50, 100 and 200 μL) were added. RESULTS: The finding of present study showed that the IC50 of aripiprazole in the cancer cell line (21.36 μg/mL) was lower than that in the normal cell line (54.17 μg/mL). Moreover, the micronucleus assay showed that the frequency of micronuclei of aripiprazole at concentrations below 200 μM was much less than cisplatin. CONCLUSION: Aripiprazole can be a good cytotoxic compound and good candidate for further studies of cancer therapy.


RESUMO CONTEXTO: O câncer gástrico é conhecido como o quarto câncer mais comum. Os tratamentos atuais para o câncer danificaram os tecidos sensíveis do corpo saudável e, em muitos casos, o cancro será recorrente. Portanto, a necessidade de tratamentos que são mais eficazes é desejada. Recentemente, os pesquisadores mudaram sua atenção para os antagonistas antipsicóticos da dopamina para tratar o câncer. As atividades anticâncer de aripiprazol permanecem desconhecidas. OBJETIVO: Este estudo objetivou avaliar a eficácia e a segurança do aripiprazol no câncer gástrico e nas linhagens celulares normais. MÉTODOS: A este respeito, a citotoxicidade e a genotoxicidade do aripiprazol foram investigadas em linhas celulares MKN45 e NIH3T3 por ensaio de metil tetrazólio e em linfócitos periféricos de sangue por ensaio de micronúcleos. Para este efeito, as células foram cultivadas em 96 placas. As soluções de estoque de aripiprazol e cisplatina foram preparadas. Após incubação celular com diferentes concentrações de aripiprazol (1, 10, 25, 50, 100 e 200 μL), a solução de metil tetrazólio foi adicionada. Para o ensaio do micronúcleo o sangue fresco foi adicionado ao meio de cultura RPMI 1640 suplementado, e as concentrações diferentes de aripiprazole (50, 100 e 200 μL) foram adicionadas. RESULTADOS: O presente estudo mostrou que o IC50 de aripiprazol na linhagem celular cancerosa (21,36 μg/mL) foi menor do que na linha celular normal (54,17 μg/ mL). Além disso, o ensaio de micronúcleos demonstrou que a frequência de micronúcleos de aripiprazol em concentrações inferiores a 200 μM foi muito inferior à cisplatina. CONCLUSÃO: O aripiprazol pode ser um bom composto citotóxico e bom candidato para estudos adicionais da terapia do câncer.

Agribusiness has a great relevance in the world׳s economy. It generates a considerable impact in the gross national product of several nations. Hence, it is the major driver of many national economies. Nowadays, from each new planting to harvesting process it is mandatory and crucial to apply some kind of technology to optimize a given singular process, or even the entire cropping chain. For instance, digital image analysis joined with machine learning methods can be applied to obtain and guarantee a higher quality of the harvest, leading to not only a greater profit for producers, but also better products with lower cost to the final consumers. Thus, to provide this possibility this work describes a visual feature dataset from soybean seed images obtained from the tetrazolium test. This is a test capable to define how healthy a given seed is (e.g. how much the plant will produce, or if it is resistant to inclement weather, among others). To answer these questions we proposed this dataset which is the cornerstone to provide an effective classification of the soybean seed vigor (i.e. an extremely tiresome human visual inspection process). Besides, as one of the most prominent international commodity, the soybean production must follow rigid quality control process to be part of world trade. Hence, small mistakes in the seed vigor definition of a given seed lot can lead to huge losses.
