Wide-ranging species are seldom considered conservation priorities, yet they have the potential to harbour genetically deeply differentiated units across environments or ecological barriers, including some that warrant taxonomic recognition. Documenting such cryptic genetic diversity is especially important for wide-ranging species that are in decline, as they may comprise a set of even more endangered lineages or species with small distributions. However, studies of wide-ranging species, particularly when they cross political borders, are extremely challenging. One approach to overcoming these challenges is to conduct detailed local analyses in combination with less detailed, range-wide studies. We used this approach with the red-footed tortoise (Chelonoidis carbonarius), a threatened species likely to contain cryptic diversity given its vast range and the distinctive ecoregions that it inhabits. Previous single-gene molecular studies indicated the presence of at least five lineages, two of which occur in different ecoregions separated by the Andes within Colombia. We used a comprehensive genomic analysis to test the hypothesis of cryptic diversity within the single jurisdiction of Colombia. We used a combination of restriction-site-associated DNA sequencing and environmental niche modelling to provide three independent lines of evidence that support the presence of important cryptic diversity that may deserve taxonomic recognition: allopatric reproductive isolation, local adaptation and ecological divergence. We also provide a fine-scale genetic map with the distribution of conservation units in Colombia. As we complete ongoing range-wide analyses and make taxonomic adjustments, we recommend that the two lineages in Colombia be treated as separate units for conservation purposes.


Las especies con distribuciones amplias rara vez son consideradas prioridades de conservación, sin embargo, tienen el potencial de albergar unidades genéticamente diferenciadas que en algunos casos justifican reconocimiento taxonómico. Documentar dicha diversidad genética críptica es especialmente importante para las especies de rangos amplios que ya están en peligro de extinción, pues pueden comprender un conjunto de linajes o especies aún más amenazadas y con distribuciones más pequeñas. Sin embargo, los estudios de especies de rangos amplios, particularmente cuando cruzan fronteras políticas, son extremadamente desafiantes. Un enfoque para superar estos desafíos es realizar análisis locales detallados en combinación con estudios en todo el rango de distribución menos detallados. Nosotros usamos este enfoque con la tortuga de patas rojas (Chelonoidis carbonarius), una especie amenazada que probablemente contiene diversidad genética críptica dada su amplia distribución y las distintas ecorregiones en las que habita. Estudios moleculares previos de un solo gen indicaron la presencia de al menos cinco linajes, dos de los cuales ocurren en diferentes ecorregiones separadas por los Andes en Colombia. En este estudio utilizamos una combinación de secuenciación de ADN asociada a sitios de restricción (RADseq) y modelamiento de nicho ecológico para proporcionar tres líneas independientes de evidencia que respaldan la presencia de diversidad críptica importante que puede merecer reconocimiento taxonómico: aislamiento reproductivo alopátrico, adaptación local y divergencia ecológica. También proporcionamos un mapa genético a escala fina con la distribución de unidades de conservación en Colombia. Mientras completamos análisis genómicos en todo el rango de distribución y hacemos ajustes taxonómicos, recomendamos que los dos linajes en Colombia se traten como unidades independientes para fines de conservación.

We report new occurrences of Testudinidae and Chelidae from 11 different fossil sites in the Huayquerías and Tunuyán formations (late Miocene-Pliocene) in the Huayquerías del Este, Mendoza, Argentina. A total of 36 specimens were collected, representing at least three taxa from two different clades: 80.6% Testudinidae and 19.4% Chelidae. Testudinids are present throughout the stratigraphic succession, in alluvial, fluvial, and interdune sedimentary environments, represented by dorsal and plastron carapace plates, with some remains of the appendicular skeleton. A complete articulated plastron with some peripheral plates of the carapace was also recognized and is assigned to the extant species Chelonoidis chilensis, representing the oldest record for this taxon (late Miocene, Messinian). Another possible morphotype was recognized in the form of a large scapular girdle and femur that likely corresponded to an individual with a dorsal carapace of about 80 cm. This assemblage of small and large tortoise species is recognized for first time in the Messinian Age/Stage of Mendoza. Chelids are mostly restricted to fluvial deposits in the upper part of the Huayquerías Formation, except for two specimens which appear in the lower part. One specimen is represented by a right half plastron of an indeterminate Chelidae species which could correspond to a carapace of up to 24 cm in length with plastral lobes narrower than those observed in Phrynops species. Another specimen is characterized by a large costovertebral tunnel in the carapace, peripheral plates of the posterior margin of the carapace with ornamentation of fine grooves forming irregular polygons.

Tortoises of the genus Gopherus evolved in North America and have survived major environmental challenges in the past 40 million years. However, this genus now faces multiple anthropogenic threats, such as the introduction of invasive plant species. Buffelgrass (Cenchrus ciliaris) is considered one of the greatest threats to arid and tropical ecosystems, where gopher tortoises inhabit, because the grass displaces native flora and fauna. Modification of the environment as a result of this invasive plant portends an alteration of the available thermal landscape. The aim of this paper is twofold: 1) to evaluate the thermal quality of the primary habitat of Gopherus evgoodei (tropical deciduous forest [TDF], and 2) determine the potential thermal changes due to habitat modification by buffelgrass. First, we obtained data on body temperature of active tortoises in semi-captivity. Second, we measured the operative environmental temperature during 5 years at three sites south of Sonora, Mexico that support G. evgoodei: a) a pristine TDF (Conserved-TDF); b) a forest patch surrounded by introduced buffelgrass pasture (Partial-TDF); and c) an introduced buffelgrass pasture area (Buffel-Pasture). Our results demonstrate that the intact microhabitats within the TDF provide G. evgoodei with high thermal quality at both spatial and temporal scales. However modified habitat by buffelgrass had higher operative temperatures for G. evgoodei than TDF. The thermal quality of the sites disturbed with buffelgrass can exceed the thermal requirements of G. evgoodei by up to 25 °C. Finally, we discussed potential collateral effects of habitat modification by invasion by buffelgrass.

Chelonians are considered good bioindicators of environmental quality. The assessment of the health status of chelonian populations in the Amazon is important because they are traditionally consumed in large numbers in riverine communities and sustainable use reserves. The present study aimed to evaluate the health of Podocnemis unifilis (Testudines, Podocnemididae) in an environmental protection area in the Amazon region in Brazil. We analyzed the biomarkers lipoperoxidation, carbonylation of proteins, occurrence of micronuclei and erythrocytic nuclear abnormalities, quantified metallothioneins, and evaluated mercury bioaccumulation. We generated pioneering data regarding biomarkers in wild Amazonian freshwater turtles. All biomarker responses did not vary significantly between the sexes. The occurrence of oxidative and genotoxic damage, as well as concentrations of metallothioneins was low compared to other studies. In addition, the bioaccumulation of mercury in the muscle of the animals was below the limit recommended for human consumption by the World Health Organization. Our results provide baseline data on mercury bioaccumulation and biomarker responses that can be useful for future comparisons with other freshwater turtles. The data also provide evidence that the sustainable exploitation of these turtles in the study area (Piagaçu-Purus Sustainable Development Reserve) does not pose a risk to local riverine communities, as the detected mercury concentrations are safe for human consumption. In this sense, our results highlight the importance of protected areas for the conservation of healthy turtle populations, at the same time ensuring the health of the human populations that use them as a food resource.(AU)


Quelônios são considerados bons bioindicadores da qualidade ambiental. A avaliação do estado de saúde de populações de quelônios na Amazônia também é importante porque estes animais são tradicionalmente consumidos em grandes quantidades em comunidades ribeirinhas e em reservas de uso sustentável. O presente estudo avaliou a saúde de Podocnemis unifilis (Testudines, Podocnemididae) em uma área de proteção ambiental da Amazônia no Brasil. Analisamos lipoperoxidação, carbonilação de proteínas, ocorrência de micronúcleos e anormalidades nucleares eritrocíticas, quantificamos metalotioneínas, e avaliamos a bioacumulação de mercúrio. Geramos dados pioneiros sobre biomarcadores em quelônios de água doce amazônicos silvestres. Todas as respostas aos biomarcadores não variaram significativamente entre os sexos. A ocorrência de danos oxidativos e genotóxicos, bem como as concentrações de metalotioneínas foram baixas em comparação a outros estudos. Adicionalmente, a bioacumulação de mercúrio no músculo dos animais ficou abaixo dos limites recomendados para consumo humano pela Organização Mundial de Saúde. Nossos resultados constituem um conjunto de dados de referência para bioacumulação de mercúrio e respostas de biomarcadores que podem ser úteis para futuras comparações com outros quelônios de água doce. Os dados também fornecem evidências de que a exploração sustentável desses quelônios na área de estudo (Reserva de Desenvolvimento Sustentável Piagaçu-Purus) não apresenta risco às comunidades ribeirinhas locais, já que as concentrações de mercúrio detectadas são seguras para o consumo humano. Nesse sentido, nossos resultados evidenciam a importância das áreas protegidas para a conservação de populações saudáveis de quelônios, concomitantemente assegurando a saúde das populações humanas que os utilizam como fonte alimentar.(AU)

Atractis thapari Petter, 1966, an atractid nematode, was collected parasitizing the large intestine of tortoises of the species Chelonoidis carbonarius (Spix, 1824) (Cc) and C. denticulatus (Linnaeus, 1766) (Cd) (Testudinidae) in the Zoobotanical Park of the municipality of Teresina, state of Piauí, Brazil. Taxonomic identification was based on morphological and morphometrical features, and using bright-field and scanning electron microscopy. The present study adds new observations on the morphology of A. thapari, mainly relating to mouth papillae, the excretory pore, deirids, and male and female posterior ends. The parasitic indices of prevalence (P), mean intensity (MI), mean abundance (MA) and range of infection (RI) for A. thapari in these two tortoise species were: P = 100%, MI = 154,667, MA = 154,667, RI = 5,500-588,500 (Cc); P = 100%, MI = 93,639, MA = 93,639, RI = 1,000-224,500 (Cd). This report confirms the occurrence of A. thapari in Neotropical region, South America, Brazil, and extends its occurrences to a new host, the tortoise C. carbonarius. Adjustment of host management with the aim of improving hygiene and health conditions is suggested.(AU)


Atractis thapari Petter, 1966, um nematoide atractídeo, foi coletado parasitando o intestino grosso de jabutis, Chelonoidis carbonarius (Spix, 1824) (Cc) e C. denticulatus (Linnaeus, 1766) (Cd) no Parque Zoobotânico, município de Teresina, estado do Piauí, Brasil. A identificação taxonômica foi baseada nos caracteres morfológicos e morfométricos usando microscopias de campo claro e eletrônica de varredura. O presente estudo adiciona novas observações na morfologia, principalmente relacionadas as papilas bucais, poro excretor, deirídeos, e extremidade posterior de machos e fêmeas. Os índices parasitários de prevalência (P), intensidade média (IM), abundância média (AM) e amplitude de variação de infecção (AI) de A. thapari em ambos jabutis foram P = 100%, IM = 154.667, AM = 154.667, AI = 5.500-588.500 (Cc); P = 100%, I M= 93.639, AM = 93.639, AI = 1.000-224.500 (Cd). Este registro confirma a ocorrência de A. thapari na região Neotropical, América do Sul, Brasil, e expande a um novo hospedeiro, o jabuti C. carbonarius. Insular giant tortoise diversity has been depleted by Late Quaternary extinctions, but the taxonomic status of many extinct populations remains poorly understood due to limited available fossil or subfossil material, hindering our ability to reconstruct Quaternary island biotas and environments. Giant tortoises are absent from current-day insular Caribbean ecosystems, but tortoise remains from Quaternary deposits indicate the former widespread occurrence of these animals across the northern Caribbean. We report new Quaternary giant tortoise material from several cave sites in Pedernales Province, southern Dominican Republic, Hispaniola, representing at least seven individuals, which we describe as Chelonoidis marcanoi sp. nov. Although giant tortoise material was first reported from the Quaternary record of Hispaniola almost 35 years ago, tortoises are absent from most Quaternary deposits on the island, which has been studied extensively over the past century. The surprising abundance of giant tortoise remains in both vertical and horizontal caves in Hispaniola's semi-arid ecoregion may indicate that this species was adapted to open dry habitats and became restricted to a habitat refugium in southeastern Hispaniola following climatic-driven environmental change at the Pleistocene-Holocene boundary. Hispaniola's dry forest ecosystem may therefore have been shaped by giant tortoises for much of its evolutionary history.

Desert tortoises (Testudines; Testudinidae; Gopherus agassizii group) have an extensive distribution throughout the Mojave, Colorado, and Sonoran desert regions. Not surprisingly, they exhibit a tremendous amount of ecological, behavioral, morphological and genetic variation. Gopherus agassizii was considered a single species for almost 150 years but recently the species was split into the nominate form and Morafka's desert tortoise, Gopherus morafkai, the latter occurring south and east of the Colorado River. Whereas a large body of literature focuses on tortoises in the United States, a dearth of investigations exists for Mexican animals. Notwithstanding, Mexican populations of desert tortoises in the southern part of the range of Gopherus morafkai are distinct, particularly where the tortoises occur in tropical thornscrub and tropical deciduous forest. Recent studies have shed light on the ecology, morphology and genetics of these southern 'desert' tortoises. All evidence warrants recognition of this clade as a distinctive taxon and herein we describe it as Gopherus evgoodei sp. n. The description of the new species significantly reduces and limits the distribution of Gopherus morafkai to desertscrub habitat only. By contrast, Gopherus evgoodei sp. n. occurs in thornscrub and tropical deciduous forests only and this leaves it with the smallest range of the three sister species. We present conservation implications for the newly described Gopherus evgoodei, which already faces impending threats.

A pneumonia é uma doença respiratória comum na clínica de répteis. Agentes infecciosos são capazes de causar pneumonia primária em répteis mantidos em cativeiro, porém na maioria dos casos, são secundárias a problemas de manejo, higiene e nutricionais. O objetivo desse trabalho foi relatar a ocorrência de pneumonia bacteriana em jabuti-piranga (Chelonoidis carbonaria), e descrever o diagnóstico clínico, microbiológico, radiográfico e a conduta terapêutica. O animal apresentava sinais de distúrbios respiratórios e foi descrito durante a anamnese que houve um diagnostico anterior de pneumonia. Os achados radiográficos foram sugestivos de pneumonia/edema pulmonar. Baseado nos exames radiográficos e sinais clínicos apresentados iniciou-se o tratamento com administração de Cloranfenicol (40mg/kg/SID/IM) por 10 dias. Foram isoladas Klebsiella spp. e Citrobacter spp. da cultura bacteriana realizada da coleta de lavado endotraqueal. Ambas com perfil de resistência múltipla aos antibióticos testados. Instituiu-se protocolo terapêutico utilizando Gentamicina (5mg/kg/IM), em sete aplicações com intervalos de 72h. Após o segundo protocolo terapêutico notou-se melhora dos sinais clínicos do animal, porém foi observada a persistência de secreção nasal. Foi realizado novo exame radiográfico, demonstrando discreta diminuição na opacidade do campo pulmonar direito e nenhuma alteração significativa no campo pulmonar esquerdo na projeção craniocaudal. Devido à permanência do sinal clínico apresentado, nova coleta de material endotraqueal foi realizada, e houve isolamento de Citrobacter spp. e Enterobacter spp. A partir dos resultados obtidos no antibiograma, instituiu-se novo protocolo com uso de amicacina (2,5mg/kg/IM), em sete aplicações com intervalos de 72h. Após antibioticoterapia, outro exame radiológico foi realizado, e demonstrou redução satisfatória do quadro pulmonar, e sinais clínicos.(AU)


Pneumonia is a common respiratory disease in clinical of reptiles. Infectious agents are capable of causing primary pneumonia in reptiles maintained in captivity, but in most cases are secondary to problems of management, hygiene and nutrition. The aim of this study was to report the occurrence of bacterial pneumonia in red-footed tortoise (Chelonoidis carbonaria), and describe the clinical, microbiologic, radiographic and therapeutic management. The animal showed signs of respiratory disorders and has been described in the clinical history before diagnosis of pneumonia. The radiographic findings were suggestive of pneumonia/pulmonary edema. Based on the displayed radiographic examination and clinical signs began treatment with administration of chloramphenicol (40mg/kg/SID/IM) for ten days. Were isolated Klebsiella spp. and Citrobacter spp. bacterial culture done collecting endotracheal lavage. Both with multiple antibiotic resistance profile tested. Treatment protocol was instituted using gentamicin (5mg/kg/IM) applications into seven intervals of 72h. There was improvement in clinical signs of the animal, but the presence of nasal secretion was still observed. New radiographic examination, demonstrating slight decrease in the opacity of the right lung field and no significant change in the left lung field in craniocaudal projection was performed. Because of the persistence of clinical signs presented new collection endotracheal material was performed, and there was isolation of Citrobacter spp. and Enterobacter spp. From the results obtained in the antibiogram, was instituted new protocol with the use of amikacin (2.5mg/kg/IM) applications into seven intervals of 72h. After antibiotic therapy, other radiological examination was performed, and showed satisfactory reduction in pulmonary function and clinical signs.(AU)

The aim of this study was to report the occurrence of bacterial pneumonia in red-footed tortoise (Chelonoidis carbonaria), and describe the clinical, microbiologic, radiographic and therapeutic management. The animal showed signs of respiratory disorders and has been described in the clinical history before diagnosis of pneumonia. The radiographic findings were suggestive of pneumonia/pulmonary edema. Based on the displayed radiographic examination and clinical signs began treatment with administration of chloramphenicol (40mg/kg/SID/IM) for ten days. Were isolated Klebsiella spp. and Citrobacter spp. bacterial culture done collecting endotracheal lavage. Both with multiple antibiotic resistance profile tested. Treatment protocol was instituted using gentamicin (5mg/kg/IM) applications into seven intervals of 72h. There was improvement in clinical signs of the animal, but the presence of nasal secretion was still observed. New radiographic examination, demonstrating slight decrease in the opacity of the right lung field and no significant change in the left lung field in craniocaudal projection was performed. Because of the persistence of clinical signs presented new collection endotracheal material was performed, and there was isolation of Citrobacter spp. and Enterobacter spp. From the results obtained in the antibiogram, was instituted new protocol with the use of amikacin (2.5mg/kg/IM) applications into seven intervals of 72h. After antibiotic therapy, other radiological examination was performed, and showed satisfactory reduction in pulmonary function and clinical signs...
