RESUMO

Melanoma is the most aggressive and lethal type of skin cancer, characterized by therapeutic resistance. In this context, the present study aimed to investigate the cytotoxic potential of manool, a diterpene from Salvia officinalis L., in human (A375) and murine (B16F10) melanoma cell lines. The analysis of cytotoxicity using the XTT assay showed the lowest IC50 after 48 h of treatment with the manool, being 17.6 and 18.2 µg/ml for A375 and B16F10, respectively. A selective antiproliferative effect of manool was observed on the A375 cells based on the colony formation assay, showing an IC50 equivalent to 5.6 µg/ml. The manool treatments led to 43.5% inhibition of the A375 cell migration at a concentration of 5.0 µg/ml. However, it did not affect cell migration in the B16F10 cells. Cell cycle analysis revealed that the manool interfered in the cell cycle of the A375 cells, blocking the G2/M phase. No changes in the cell cycle were observed in the B16F10 cells. Interestingly, manool did not induce apoptosis in the A375 cells, but apoptosis was observed after treatment of the B16F10 cells. Additionally, manool showed an antimelanoma effect in a reconstructed human skin model. Furthermore, in silico studies, showed that manool is stabilized in the active sites of the tubulin dimer with comparable energy concerning taxol, indicating that both structures can inhibit the proliferation of cancer cells. Altogether, it is concluded that manool, through the modulation of the cell cycle, presents a selective antiproliferative activity and a potential antimelanoma effect.

RESUMO

The objective of this work was to evaluate the influence of different growth regulators on the mineral and total phenolic contents of Salvia officinalis. The samples received the applications of salicylic acid (AS); gibberellic acid (GA3); abscisic acid (ABA) and solution without regulators (control). The exploratory evaluation of the samples was carried out through the Principal Component Analysis (PCA). In addition, has been used supervised learning methods with support vector machine (SVM) algorithms to classify the samples. The phenolic and total flavonoid contents were higher in the plants treated with the regulators. The element found in the highest concentration in Salvia officinalis was N. Plants sprayed with ABA showed higher concentrations of N, K, and Mn; Fe and Al were higher with ABA and gibberellin application, while the application of AS provided the highest accumulation of P. The application of plant regulators improves the nutraceutical properties of Salvia officinalis.

RESUMO

SUMMARY: In Saudi Arabia, it is widely believed that women with reproductive problems can use the extract of the sage plant as a tea drink. This study was conducted to investigate the effects of this herb on the fertility of female rats and embryo implantation. Forty-eight Wistar virgin female rats were divided into four groups at random, with 12 rats in each group. The control group received distilled water orally. The three treatment groups received different concentrations of sage extract: 15, 60, or 100 mg/kg for 14 days before mating, then mated with a male and sacrificed on the 7th day of gestation, the uterine horns removed, and photographed. The total body weight of mothers, weight of uteri and ovaries and number of fetuses were determined. Ovarian and uteri tissues were cut into 5 µ sections and stained with hematoxylin and eosin. Serum FSH, LH were determined by the ELISA method. The present study showed that low dose of sage (15 mg/kg) have no effects on serum concentration levels of FSH and LH hormones, also has no effect on the number of growing follicles. The present study showed a significant differences (P≤0.05) in body weight, ovary and uterus weight in the groups treated with high doses of Salvia officinalis as compared to control group. Also a significant differences (P≤0.05) found in FSH, LH hormones. Histological study showed overall histomorphological structural configurations including growing and matured graafian follicular countable changes, besides a number of corpora lutea and regressed follicles in the treated groups with high doses of Salvia officinalis as compared to control group. The researchers concluded that the extract of the sage plant with high doses can stimulate the growth graafian follicles and improve fertility in female rats.

This study aimed to evaluate the antioxidant activities (AA) of lyophilized rosemary extract and lyophilized sage extract, and their effects on the oxidative stability of poultry pátê. For this purpose, four poultry pátê formulations with rosemary, sage, sodium erythorbate and a control (without antioxidants) were produced. The rosemary and sage were characterized according to total phenolic compounds (TPC) and AA by several methods. The poultry pátês stored at 4 °C were evaluated by the lipid oxidation. High concentrations of TPC were detected in the rosemary extract and sage extract (46.48 and 41.61 mg GAE/g (Gallic acid equivalent), respectively). The AA of the rosemary and sage extracts determined by free radical-scavenging were 4745.72 and 2462.82 µmol TE/g (Trolox equivalents), respectively. The high concentrations of catechin, rutin, myricetin and p-coumaric acids in these extracts may be responsible for the strong inhibitory action against food pathogens. Besides this, these compounds can be responsible for the best performance in inhibiting lipid oxidation in poultry pátês during storage. This study suggests that rosemary and sage extracts may be used as a natural antioxidant in meat products.

RESUMO

Manool, a diterpene isolated from Salvia officinalis, was evaluated by the XTT colorimetric assay for cytotoxicity and selectivity against different cancer cell lines: B16F10 (murine melanoma), MCF-7 (human breast adenocarcinoma), HeLa (human cervical adenocarcinoma), HepG2 (human hepatocellular carcinoma), and MO59J, U343 and U251 (human glioblastoma). A normal cell line (V79, Chinese hamster lung fibroblasts) was used to compare the selectivity of the test substance. Manool exhibited higher cytotoxic activity against HeLa (IC50 = 6.7 ± 1.1 µg/mL) and U343 (IC50 = 6.7 ± 1.2 µg/mL) cells. In addition, in the used experimental protocols, the treatment with manool was significantly more cytotoxic for different tumor cell lines than for the normal cell line V79 (IC50 = 49.3 ± 3.3 µg/mL), and showed high selectivity. These results suggest that manool may be used to treat cancer without affecting normal cells.

ABSTRACT Peptic ulcers are caused by an unbalance among endogenous, exogenous damaging agents and protective agents. In this study we evaluated the in vivo antiulcerogenic activity for the aqueous extract of Salvia officinalis (EAS). After treatments (n = 6; v.o) with control (C) water; pantoprazole (30 mg/kg) and EAS (100, 250 and 400 mg/kg, respectively) the rats received a 0.3 M HCl solution in ethanol 60% (v.o) and it were performed morphological evaluations of the stomachs through the use of a table of injury scores (ANOVA, one way and Tukey). The EAS (400 mg/kg) showed an antiulcerogenic activity (32.53%). Similar to the pantoprazole (29.91%), in comparison with (p<0.0001) the group C. The phytochemical analysis through high performance liquid chromatography revealed high acid rosmarinic (3.53%) for the EAS. The flavonoids (0.25%) and the total phenolics (GAE 309.0) were quantified by spectrophotometry UV/Vis. The EAS and the rosmarinic acid exhibited elevated antioxidant activities by DPPH (Effective Concentration 50% -EC50 9.2 and 0.7 mg/mL, respectively,) at least partially explaining the antiulcerogenic effect of the extract.

RESUMO

Salvia officinalis (sage) is a perennial woody subshrub native to the Mediterranean region that is commonly used as a condiment and as an anti-inflammatory, antioxidant and antimicrobial agent due to its biological activities. Manool is the most abundant micro-metabolite found in Salvia officinalis essential oils and extracts. We therefore decided to evaluate the cytotoxic, genotoxic and antigenotoxic potential of manool in Chinese hamster lung fibroblasts (V79) and human hepatoma cells (HepG2). Cytotoxicity was assessed by the colony-forming assay in V79 cells and toxic effects were observed at concentrations of up to 8.0 µg/mL. The micronucleus test was used to evaluate the genotoxicity and antigenotoxicity of manool in V79 and HepG2 cells at concentrations of 0.5-6.0 µg/mL and 0.5-8.0 µg/mL, respectively. For evaluation of antigenotoxicity, the concentrations of manool were combined with methyl methanesulfonate (MMS, 44 µg/mL). The results showed a significant increase in the frequency of micronuclei in cultures of both cell lines treated with the highest concentration tested, demonstrating a genotoxic effect. On the other hand, manool exhibited a protective effect against chromosome damage induced by MMS in HepG2 cells, but not in V79 cells. These data suggest that some manool metabolite may be responsible for the antigenotoxic effect observed in HepG2 cells.

RESUMO

In this paper we screened the dichloromethane extract from the aerial parts of Salvia officinalis L., Lamiaceae, against a representative panel of microorganisms that cause caries, conducted a bioassay-guided fractionation to establish themselves the most active metabolite (manool) and determined the Salvia officinalis fraction with the manool highest concentration to be used to activate an ingredient in oral care products such as toothpastes and mouthwashes. Both manool and S. officinalis extract showed very promising minimal inhibitory concentration values (between 6.24 and 31.36 µg.ml-1) and time kill curves against the primary causative agents of dental caries (Streptococcus mutans) revealed that, at twice its minimal bactericidal concentration (12.48 µg.ml-1), manool required 6 h to completely kill the bacteria. Salvia officinalis extract at twice its minimal bactericidal concentration (31.36 µg.ml-1 ) needed 12 h. The results achieved with Salvia officinalis extract motivated us to develop and validate an analytical RP-HPLC method to detect and determine manool in this extract. The validation parameters were satisfactorily met and evaluated allows us to consider the developed method suitable for use in different labs. In conclusion, our results evidenced that the manool-rich S. officinalis extract can be considered an analytically validated alternative to develop novel and effective antimicrobial agents against the main bacteria responsible for dental caries.

The objective was to evaluate the conservation of strawberries with coatings based on xanthan gum and, or, sage essential oil. The strawberries were minimally processed, added with coatings, dried under ventilation, packaged in trays of polyethylene terephthalate and stored at 4 °C for 12 days. Analyses of fungal decay, weight loss, firmness, color, soluble solids and pH were carried out, and at the end of storage, fungi and yeasts count as well. Regardless of the coating, mass loss of strawberries was reduced during the refrigerated storage period. However, the coatings based on xanthan gum indicated to be more efficient, by providing greater fruit firmness, while those containing xanthan gum and sage essential oil were the ones which provided the lowest incidence of fungi. In general, the coatings did not affect the color and pH of the strawberries.

RESUMO

Melanin is a photoprotective skin pigment, and pathologies characterized by hypo or hyperpigmentation are common. New compounds that regulate melanogenesis are, therefore, opportune, and many natural products with this property, as polyphenols, have been described. Salvia officinalis L., Lamiaceae, is a widely used food spice that contains high amounts of phenol derivates, including rosmarinic acid. The aim of this work was to evaluate the contribution of rosmarinic acid in the melanogenic activity of sage extracts. Fluid and aqueous extracts of sage and purified rosmarinic acid were assayed for B16F10 cytotoxicity and, then, evaluated on melanin production and tyrosinase activity. While sage extracts showed a concentration-dependent ability to significantly increase melanin production without necessarily changing the enzymatic activity, rosmarinic acid showed a dual behavior on melanogenesis, increasing melanin biosynthesis and tyrosinase activity at low concentrations and decreasing it at higher levels. Rosmarinic acid may collaborate with sage extracts activity on melanogenesis, although other compounds may be involved. This is the first time that a dual action of rosmarinic acid on melanogenesis is reported, which may be useful in further studies for therapeutic formulations to treat skin pigmentation disorders.

RESUMO

Salvia officinalis (L.), or common sage, is an aromatic herb that has been used in medicine and cooking since ancient times and has been investigated for the treatment of various diseases, especially infections and skin inflammation. We conducted phytochemical prospecting and quality control with hydroalcoholic extracts of dried sage, to identify active compounds in the plant. The aim was to assess antibacterial and antifungal activity against Staphylococcus aureus, Streptococcus agalactiae, Candida albicans and Candida tropicalis. Antimicrobial susceptibility was investigated in vitro by agar-overlay and well-diffusion techniques, in which disc and well were used. Salvia officinalis (L.) was not effective against Streptococcus agalactiae, Candida albicans or Candida tropicalis, but best results were observed for antibacterial activity against Staphylococcus aureus. Considering the results of the inhibition tests presented here, we suggest that cosmetic formulations containing Salvia officinalis (L.) could contribute to inhibitor of pathogens in the skin microbiota.

The aim of this work was to investigate the effects of four different light intensities on the growth characteristics, physiological parameters and leaf photosynthetic pigments of Salvia officinalis L. The plant's dry mass, number of the leaves and physiological parameters indicated a strong positive correlation with the light intensity. On the other hand, the plant's height and leaf photosynthetic pigments were increased at low light treated plants. These results suggest that the aromatic herb Salvia officinalis L. is adaptable to different light environments.

Background: Gingivitis is the most prevalent periodontal problem in people, so we need to find new alternatives for treatment and prevention. Aim: To evaluate the effects of Salvia officinalis in the declining modified gingival index (GI) in dentistry students of the Universidad del Desarrollo diagnosed with gingivitis, year 2010. Subjects and Methods: This was a quantitative study, blinded, experimental type of placebo-controlled clinical trials on the action of extract of S. officinalis in toothpaste and mouthwash for plaque-induced gingivitis in 33 dental students from Universidad del Desarrollo, Concepción; 13 women and 20 men between 21 and 29 years old, were divided into three groups: placebo, mouthwash and toothpaste. After the first appointment we call the patients every 7, 14 and 28 days, we ended the treatment at day 14. Results: In the control group there was no significant difference (p=0.061) the first week, while groups with mouthwash and toothpaste, there were significant differences (p=0.00). The last week, as we remove the placebo, the GI was maintained. On the other hand, in groups with S. officinalis the drop did not continue. The decrease of GI was higher in groups with mouthwash and toothpaste than in the control group. Discussion: The use of S. officinalis is effective in reducing the GI. The mouthwash and toothpaste decrease more rapidly the IG. The mouthwash achieved the greatest reduction of IG.

RESUMO

The biosynthesis and the accumulation of the chemical composites that have allelopathy characteristics in Salvia officinalis L. (Lamiaceae) are resulting from the secondary plant metabolism. The present work had as objective to analyze the allelopathic effects of aqueous extracts of fresh leaves of Sálvia collected during the seasons, obtained by decoction, static infusion and grinding, on the germination and development of seedlings of lettuce (Lactuca sativa L.). The experiment had 20 treatments (concentrations 0; 7.5; 15; 22.5 and 30%). The results had been submitted to the Tukey test, to the level at 5% probability. The appraised variables, germination percentage, the germination speed index (GSI), the growth of shoot and roots and abnormality formation of seedlings did not present a pattern of allelopathy in the different seasons, indicating a possible alteration in the plant metabolism in function of the seasonal variation in the plant ambient.

The use of medicinal plants, associated to infectious processes, has stimulated a number of chemical and pharmacological studies to obtain new compounds with therapeutic properties. Nowadays, microorganisms which cause damages to human health have resistance to most of the antimicrobial drugs. Many technological steps are suggested for solving the bacterial resistance matter, and one of them is the search for new antimicrobial drugs from vegetal species. Therefore, this article assesses the antibacterial activity from different extracts of Rosmarinus officinalis, Origanum majorana, Salvia officinalis, Bidens pilosa, Ocimum gratissimum, Cymbopogon citratus, Sida rhombifolia and Leonotis nepetaefolia tested against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa by using the disk diffusion method. The results of the test showed that extracts from R. officinalis have inhibited the activity against S. aureus, B. pilosa, and O. majorana plants acted upon E. coli and P. aeruginosa, and S. officinalis upon P. aeruginosa. The other plants were ineffective to inhibit the bacteria tested. From these tests, it was verified that Rosmarinus, Bidens, Origanum and Salvia may be sources of antimicrobial agents.
