In the present study, the biochemical composition of some Echinodermata (Holothuroidea, Echinoidea) from the Red Sea, Egypt was investigated. The results showed that the highest percentage of saturated fatty acids (SFA) was in Holothuria nobilis and the lowest percentage was recorded in Holothuria scabra. The highest percentage of monounsaturated fatty acids (MUFA) was recorded in Pearsonothuria graeffei while the lowest percentage was recorded in Holothuria atra. The highest percentage of polyunsaturated fatty acids (PUFA) was recorded in Holothuria nobilis and the lowest percentage was recorded in Holothuria atra. The highest percentage of omega-3 fatty acids was recorded in Stichopus hermanni however, the lowest percentage was recorded in Diadema setosum. The highest percentage of omega-6 fatty acids was recorded in Bohadschia vitiensis where, the lowest percentage was recorded in Holothuria atra. In addition, nonessential amino acid (NEAA) glycine were the highest in all species and serine was the lowest while Essential amino acid (EAA) arginine was the highest and cysteine was the lowest. All of the NEAA recorded highest concentrations in S. hermanni except proline. The recorded essential amino acids were histidine, isoleucine, phenylalanine, and leucine. The latter was high in Pearsonothuria graeffei. Tyrosine, valine, threonine, and arginine were high in Stichopus hermanni. The present study proved that sea cucumbers has great nutritional value.(AU)

Edible red sea urchin (Loxechinus albus) is an endemic echinoderm species of the Chilean coasts. The worldwide demand for high-quality gonads of this species has addressed the depletion of its natural populations. Studies on this sea urchin are limited, and genomic information is almost nonexistent. Hence, generate a transcriptome is crucial information that will considerably enrich molecular data and promote future findings for the L. albus aquaculture. Here, we obtained transcriptomic data of the edible red sea urchin by Illumina platform. Total RNA was extracted from gonads, intestines, and coelomocytes of juvenile urchins, and samples were sequenced using MiSeq Illumina technology. A total of 91,119,300 paired-end reads were de novo assembled, 185,239 transcripts produced, and a reference transcriptome created with 38.8% GC content and an N50 of 1769 bp. Gene ontology analysis revealed notable differences in the expression profiles between gonads, intestines, and coelomocytes, allowing the detection of transcripts associated with specific biological processes and KEGG pathways. These data were validated using 12 candidate transcripts by real-time qPCR. This dataset will provide a valuable molecular resource for L. albus and other species of sea urchins.

In echinoderms, the immune system plays a relevant role in defense against infection by pathogens. Particularly, in sea urchins, the immune system has been shown to be complex, especially in terms of the variety of immune genes and molecules described. A key component of the response to external pathogens are the Toll-like receptors (TLRs), which are a well-characterized class of pattern recognition receptors (PRRs) that participate in the recognition of pathogen-associated molecular patterns (PAMPs). Despite the fact that TLRs have been described in several sea urchin species, for the red sea urchin (Loxechinus albus), which is one of the most important sea urchins across the world in terms of fisheries, limited information on the TLR-mediated immune response exists. In the present study, for the first time, we evaluated the effect of thermal stress, LPS and poly I:C treatment on the coelomocyte immune response of Loxechinus albus to determine how these factors modulate TLR and strongylocin (antimicrobial peptides of echinoderms) responses. We show that the tlr3-like, tlr4-like, tlr6-like and tlr8-like transcripts are modulated by poly I:C, while LPS only modulates the tlr4-like response; there was no effect of temperature on TLR expression, as evaluated by RT-qPCR. Additionally, we showed that strongylocin-1 and strongylocin-2 are modulated in response to simulated viral infection with poly I:C, providing the first evidence of strongylocin expression in L. albus. Finally, we determined that temperature and LPS modify the viability of coelomocytes, while poly I:C treatment did not affect the viability of these cells. This study contributes to the knowledge of immune responses in sea urchins to improve the understanding of the role of TLRs and strongylocins in echinoderms.

Histological examination of the gonads confirmed the process of sexual transformation in this fish species, wherein individuals mature first as female, and then change sex to male (protogynous hermaphroditism). Histological sections also showed that the sexual maturation of males of L. lenjtan comprised three main stages, while the sexual development of females could be classified into four main stages. Extended spawning in the form of batches released during different months throughout the year were recorded for this fish species, with the main spawning season in February and March, and an additional, shorter spawning season in September.(AU)

Seaweeds constitutes an abundant marine reserve that can be harnessed as source of new pharmaceutical agents. Sargassum binderi Sonder ex J. Agardh is a brown seaweed that is predominantly available from December to March in the Red Sea, Jazan, Kingdom of Saudi Arabia (KSA). In this study, three extracts were isolated using three different techniques, and were subjected to antibacterial assay. The petroleum ether extract of Sargassum binderi was more effective against selected human pathogenic bacteria than the other extracts. Therefore, further studies were focused on developing oleic acid vesicles entrapped with the petroleum ether extract of Sargassum binderi, with the aim of enhancing its penetration property. Oleic acid vesicles were prepared by entrapping petroleum ether extract of Sargassum binderi using film hydration technique. The formulated vesicles were in nanoscale, and so were termed phyto-nanovesicles (PNVs). The spectrum of antibacterial activity of PNVs showed that it is a promising formulation against S. aureus, S. pyogenes, B. subtilis, E. coli, K. pneumoniae and P. aeruginosa. The microbial sensitivities to the PNVs was in the order E.coli > B. subtilis > S. aureus > S. pyogenes > K. pneumoniae > P. aeruginosa. Thus, the PNV formulation possesses promising and effective antimicrobial potential against human pathogenic bacteria

Some species of Eunice might reach giant size, often being longer than 2m, and they are known from tropical and temperate seas. Despite their large size and recent internet notoriety, there remain some taxonomic problems in large-sized eunicids, especially since original descriptions were brief and type materials are often missing. As a mean to encourage the solution of this situation, we review the historical progress in the taxonomy of the group, including some comments on generic and specific delineation, and recommend some critical steps to solve the current confusion. These ideally would include collecting in type localities, evaluate ontogenetic morphological changes, and generate some molecular analysis to complement the morphological approach. Rev. Biol. Algunas species de Eunice pueden alcanzar tamaño gigantesco, a menudo sobrepasan los 2m de largo, y se conocen de mares tropicales y templados. A pesar de su gran tamaño y de su reciente notoriedad en Internet, todavía hay problemas taxonómicos entre los eunícidos gigantes, especialmente dado que las descripciones originales fueron breves y a menudo se carece de materiales tipo. Para incentivar la solución del problema, revisamos el desarrollo histórico de la taxonomía del grupo y se incluyen algunos comentarios sobre la delineación de los géneros y de las especies y recomendamos algunos pasos críticos para alcanzar este fin. Idealmente, esto incluiría recolectar en las localidades tipo, evaluar cambios morfológicos durante la ontogenia y generar algunos análisis moleculares para complementar el enfoque morfológico.
